The influence of temperature and light on prepenetration development of single and mixed isolates of Puccinia graminis avenae and Puccinia coronata avenae was studied on 0–2% water agar and on leaves of three oat cultivars and on three non-cultivated species of Avena. Germination of uredospores of P. graminis avenae and P. coronata avenae occurred best at 10–30oC and at 20oC respectively. The optimum temperature for germ-tube growth and for appressorial formation was 20oC for both rusts. An inverse relationship was observed between light intensity and prepenetration development with maximal germination of uredospores, germ-tube growth and appressorial formation occurring in darkness. Under optimum conditions maximum percentage germination and appressorium formation of both rusts was attained within 4 and 12 h after inoculation respectively. The proportion of germinated uredospores of crown rust which gave rise to appressoria was about twice that observed for stem rust. No significant differences were observed in prepenetration development between the single and mixed race inocula of the two rusts. Although germination of uredospores was significantly greater on water agar than on oat leaves, there were no significant differences in prepenetration development of the rusts on the various oat cultivars and species examined. Consequently, the data failed to indicate the presence of resistance mechanisms operating during the prepenetration phase of the infection process on the cultivars and species examined.  相似文献

Yellow rust caused by Puccinia striiformis f. sp. hordei is an important disease of barley (Hordeum vulgare L.) in some parts of the world. We compared the effectiveness of different types of resistance in field plots at Ardabil Agricultural Research Station (Iran) during 2010–2011. Yield components along with slow rusting parameters including final rust severity (FRS), apparent infection rate (r), relative area under disease progress curve (rAUDPC) and coefficient of infection (CI) were evaluated for 25 barley cultivars. In all, two barley cultivars with race-specific resistance, 19 cultivars with different levels of slow rusting resistance and four susceptible cultivars were included in two experiments with and without fungicide protection under high disease pressure. Barley cultivars with slow rusting resistance displayed a range of severity responses indicating phenotypic diversity. Mean thousand kernels weight (TKW) losses for susceptible, race-specific and slow rusting genotypes were 31, 3 and 12%, respectively, and mean kernels per spike (KPS) losses for susceptible, race-specific and slow rusting genotypes were 19, 0.2 and 8%, respectively. Correlation coefficient of mean TKW and KPS losses with epidemiological parameters; rAUDPC, r, CI and FRS were highly significant. Slow rusting cultivars with low values of different parameters as well as genotypes with low yield component losses despite moderate disease levels were identified. Such genotypes can be used for breeding barely genotypes with high levels of resistance and negligible yield losses.  相似文献

The potential markers of juvenility (cyanidin 3-glucoside and cyanidin 3-rutinoside) in autumn leaves of seven Acer palmatum Thunb. cultivars were investigated. Three shoot positions were marked on each cultivar—crown shoot, middle shoot, and basal shoot—and the anthocyanins were analyzed using HPLC-MS. The results showed great differences in cyanidin 3-glucoside and cyanidin 3-rutinoside concentrations among seven cultivars; moreover, significant differences in cyanidin 3-glucoside content levels were also observed among three shoot positions regardless of the cultivar analyzed. The concentration decreased basipetally and reached levels up to 52 times higher in leaves obtained from crown shoots in comparison to basal shoot leaves. Therefore, the content level of cyanidin 3-glucoside can be defined as a quantitative marker of positional effect in all the Acer palmatum Thunb. cultivars analyzed. The content level of cyanidin 3-rutinoside did not express the same positional dependence.  相似文献

Five physiological populations of Gymnosporangium juniperi-virginianae Schw. were identified on the basis of pycnial and aecial lesion formation when seven apple cultivars grown in a glasshouse were inoculated with basidiospores of rust populations collected from six locations in the U.S.A. The relationship between cultivars and rust populations was explored quantitatively using the parameters N, D, and DR, where N was the highest number of pycnial lesions on a leaf/plant; D was the largestdiameter measurement of pycnial lesions on a leaf/plant; and DR, the disease rating, was proportional to the diseased area of the leaf and calculated as ND2. Statistically significant differences among cultivars over all rust populations for variables N, D, and DR were obtained, indicating the possible existence of race non-specific resistance in apple to cedar-apple rust. Moreover, significant differences among the rust populations over all cultivars for variable N and DR were obtained, confirming the existence of physiologically different rust populations of the pathogen. A change in the pathogenicities of these physiological populations must have occurred over time (10-year period). A significant interaction between cultivars and rust populations was observed for lesion size (D) only, indicating a probable existence of race specific resistance in apple as well.  相似文献

Abstract

The results of studying the types of resistance in 44 recognized and promising winter wheat cultivars for the North Caucasus region are presented. Relatively high levels of race-specific resistance at the germination stage were found in the cultivars Batko, Bystritsa, Krasota, Knyazhna, Polovchanka, Fisht and Yara; low race-specific resistance levels were reported for the most of cultivars studied. Under simulated infection conditions of wheat plants in fields, most cultivars had resistance similar to that one in adult plants. The cultivar Rufa demonstrated slow leaf rust progress, while the cultivar Yuna was highly susceptible to the North Caucasus leaf rust population. The analysis of leaf rust resistance in the cultivars tested makes it possible for agricultural producers to continuously improve their zoning structure according to preferable types of genetic resistance control.  相似文献

During a period of five years (1998‐2002), 26 strawberry cultivars and five selections were tested for resistance to crown rot. Cold stored plants inoculated with zoospores of Phytophthora cactorum were used in all experiments. The results showed that resistance to P. cactorum varies greatly between cultivars, and the most resistant ones were Senga Sengana, Induka, Melody, Glima and Bogota, while the most susceptible were Tamella, Inga, Evita and Jonsok. The results were compared to those obtained by other authors, and there were clear indications that the genetic background affects the degree of susceptibility to P. cactorum. Many of the most resistant cultivars descend from Senga Sengana (e.g. Bounty, Glima, Induka and Melody), and several of the most susceptible cultivars have common parentage (e.g. Inga, Tamella, Elsanta and Evita). In a resistance test of the progeny from two separate crosses between a resistant and a susceptible cultivar 61% and 65% of the offspring were intermediately to very susceptible, and 17% and 13% were resistant at the level of Senga Sengana. Hence, there is a high risk of losing offspring with high levels of resistance if selection for resistance to crown rot is not performed at an early stage in a breeding programme.  相似文献

Wheat plants with different resistance to stem rust were inoculated with urediospores of Puccinia graminis f. sp. tritici race 32. By means of isolated chloroplasts Hill activity and chlorophyll fluorescence were measured during the first 8–10 days of the infection. Highly resistant cultivars (infection type 0) showed no significant differences in Hill activity whereas resistant (infection type 1) and susceptible plants (infection type 4) exhibited a significant decrease in electron transport. This was associated with an apparent reduction of chlorophyll content. In both the incompatible as well as in the compatible interaction an additional factor had an effect on the inhibition of the Hill activity. Chlorophyll fluorescence was also affected in compatible and in incompatible interactions. However, the two interactions showed different patterns. Whereas the decline in variable fluorescence dominated in resistant varieties, susceptible cultivars showed an additional significant increase in basic fluorescence. The determination of Hill activity and chlorophyll fluorescence demonstrated that photosynthesis was inactivated in different ways in resistant and in susceptible cultivars.  相似文献

From two field samples of crown rust, isolates were selected for increased aggressiveness on four of 10 resistant perennial ryegrass clonal lines. Two lines which were formerly highly resistant became highly susceptible to the selected isolates but two which were formerly moderately resistant became only moderately susceptible. Thus, moderate resistance, although no less likely to be race-specific, was more stable than a high level of resistance. Cultures from highly resistant lines did not show increased aggressiveness on the varieties from which the resistant lines had been selected; hence individual genes of major effect were unimportant in determining varietal resistance. In contrast, cultures from moderately resistant lines were substantially more aggressive on their corresponding varieties. Three varieties contained many genes which tended to buffer changes in virulence of the rust.  相似文献

【目的】向日葵白锈病、黑茎病是2种危害严重的新入侵病害,新疆地区新源县和特克斯县为这2种病害发生区。研究目前种植的油用型向日葵品种对向日葵白锈病和黑茎病的抗性差异,可为新疆伊犁地区向日葵品种选育及其病害综合防控提供理论依据。【方法】选择伊犁地区广泛种植的16种油用型向日葵种质资源为供试品种,试验地设在新源县和特克斯县田间,于2015年5—9月调查样地的病叶数,并计算病情指数,同时测定样地的平均产量,最后综合以上结果确定各品种的抗性水平。【结果】不同向日葵品种在同一供试条件下对白锈病和黑茎病存在明显的抗性差异,在新源县NX19012(西域朝阳)的病情指数分别为7.83和25.62,而品种KWS204的病情指数为21.04和39.50;部分品种在不同的供试地点表现出一致的抗性水平,品种新引S31感染白锈病后在新源县的病情指数为9.41,在特克斯县为9.38;另有部分品种在不同地点存在不同的抗性水平,如G101感染白锈病后在新源县和特克斯县的病情指数分别为9.16和11.78,而感染黑茎病后在新源县和特克斯县的病情指数分别为31.05和37.79。【结论】通过对白锈病和黑茎病病情指数、向日葵产量及当地气象数据的综合研究,发现NX19012(西域朝阳)最适宜在新疆伊犁地区种植,品种S606、TO12244、NX01025(西部骆驼)也可作为多样化种植的候选品种,而新葵杂系列品种中新葵杂5号可作为今后品种选育的试验材料。  相似文献

The inheritance of resistance in red raspberry (Rubus idaeus) to yellow rust (Phragmidium rubi-idaei) was studied in a diallel cross among the cultivars Boyne, Meeker, Mailing Jewel, Glen Prosen and Glen Clova. The progenies and clonally propagated parents were exposed initially in a rust-infected plantation where the incidence and severity of infection was assessed at the telial stage. The following spring leaves on new canes of the same plants were inoculated with urediniospores in a glasshouse at 18 ± 2 °C. The latent period and number of uredinia per cm2 of abaxial leaf surface were determined 13 and 18 days after inoculation. Cv. Boyne developed only chlorotic flecking on inoculated leaves in the glasshouse and no sporulation was observed. It is postulated that this reaction was determined by a major gene which is designated Yr. Some of the other parents showed incomplete resistance of the slow rusting type, and segregants of Boyne selfed that were susceptible also showed low levels of rust, which suggests that Boyne carries both major- and minor-gene resistance. Diallel analyses of both the field and glasshouse data from the susceptible segregates indicated that the degree of incomplete resistance present was determined predominantly by additive gene action, though small but significant non-additive effects also occurred: cv. Meeker was the most resistant parent and contributed the most resistance to the progenies. The Meeker progenies also segregated for gene H, which determines cane hairiness and is known to be associated with resistance to other diseases. Segregants without hairs (genotypes hh) were found to be the more resistant to rust.  相似文献

Crown rust (Puccinia coronata Corda f.sp. avenae) can devastate oats (Avena sativa). Oxidative stress is part of the resistance mechanism in several pathosystems, but in the oat–crown rust system, it is unclear, especially regarding partial resistance. We evaluated the effects of P. coronata on oxidative stress in oat cultivars: URS 21 (partially resistant), Leggett (race‐specific resistant), URS22 and Clintland 64 (susceptibles). Seedlings and plants were inoculated with P. coronata uredospores. Cultivars were assessed for antioxidant enzyme activity and the reactive oxygen species (ROS) hydrogen peroxide and superoxide. Due to the importance of the partial resistance of URS21, this cultivar and URS 22 were also appraised for total phenolics and the relative expression of oxidative stress genes. Postinoculation, Leggett and URS 21 showed no increased peroxide levels. The susceptible cultivars increased ROS and ascorbate peroxidase activity. Clintland 64 increased also catalase activity, whereas URS 22 increased glutathione reductase and the expression of genes encoding antioxidant enzymes. URS 21 showed almost no antioxidant enzyme induction. Shortly after inoculation, URS 21 showed increased expression of genes encoding lipoxygenase and peroxidase. Cultivars URS 21 and Leggett accumulated cell wall fluorescent compounds, phenolics being detected in the former. Oxidative stress appears not to cause the hypersensitive response in this pathosystem, but late ROS accumulation did occur in the susceptible cultivars. Cultivar URS 21 may, differently from other known mechanism to date, reduce ROS accumulation by increasing the level of phenolics, resulting in later pathogen and cell death, showing non‐specific resistance to races of the pathogen also at seedling stage.  相似文献

Components of partial resistance [disease incidence (DI), infection frequency (IF), latent period (LP), spores per lesion (SPL)] were assessed on glasshouse-grown barley seedlings following inoculation with spore suspensions of Rhynchosporium secalis at growth stage 12 (Zadoks, Chang & Konzak, 1974). Four experiments were carried out at different times during 1988. Three spring barley cultivars [two from Cyprus (cvs Kantara and Athenais) and one from the UK (cv. Triumph)] were used in the first three experiments. In the fourth experiment eight additional UK cultivars with NIAB resistance ratings ranging from 3 to 9 were used. Two races of R. secalis were used in the first three experiments and three in the fourth. The three cultivars, Kantara, Athenais and Triumph, were examined in all four experiments and significant differences detected for virtually all components of partial resistance in each. Differences, however, were often small and ranking of cultivars varied in different experiments. The greater susceptibility of cv. Kantara compared to cv. Athenais, observed under field conditions in Cyprus, would not be anticipated from the small differences in components of partial resistance observed in these experiments, but, for these cultivars, the possibility of a marked genotype x environment interaction cannot be discounted. Mean values for the components of partial resistance differed in the four experiments. LP was correlated with mean glasshouse temperature from inoculation to the onset of sporulation but differences in IF and SPL were not correlated with temperature. For these components, light quality and/or duration appeared to be more important. Overall, there were no differences between races but significant race X cultivar interactions were observed in two experiments. In the fourth experiment, examining 11 cultivars, there were significant differences between cultivars for all components of partial resistance. IF and LP were correlated but neither of these components was correlated with SPL indicating independent control of this latter component. Both IF and LP were correlated with field performance (NIAB ratings) but there was no correlation with SPL. However, combining IF with mean values of SPL restricted to the 5 days following the end of the LP, produced the best correlation (r= 0.92) with NIAB ratings. Problems of assessing components of partial resistance and possible means of improving assessments are discussed.  相似文献

Crown rust, caused by Puccinia coronata f. sp. lolii, is one of the most important diseases of temperate forage grasses, such as ryegrasses (Lolium spp.), affecting yield and nutritional quality. Therefore, resistance to crown rust is a major goal in ryegrass breeding programmes. In a two-way pseudo-testcross population consisting of 306 Lolium multiflorum individuals, multisite field evaluations as well as alternative methods based on artificial inoculation with natural inoculate in controlled environments were used to identify QTLs controlling resistance to crown rust. Disease scores obtained from glasshouse and leaf segment test (LST) evaluations were highly correlated with scores from a multisite field assessment (r = 0.66 and 0.79, P < 0.01, respectively) and thus confirmed suitability of these methods for crown rust investigations. Moreover, QTL mapping based on a linkage map consisting of 368 amplified fragment length polymorphism (AFLP) and simple sequence repeat (SSR) markers revealed similar results across different phenotyping methods. Two major QTLs were consistently detected on linkage group (LG) 1 and LG 2, explaining up to 56% of total phenotypic variance (V p). Nevertheless, differences between position and magnitude of QTLs were observed among individual field locations and suggested the existence of specific local pathogen populations. The present study not only compared QTL results among crown rust evaluation methods and environments, but also identified molecular markers closely linked to previously undescribed QTLs for crown rust resistance in Italian ryegrass with the potential to be applied in marker-assisted forage crop breeding. The wheat crop remains vulnerable to all three rust diseases (leaf rust, stem rust and yellow rust) caused by Puccinia spp. according to the prevalence of the pathogen in different wheat-growing areas worldwide. Stripe rust or yellow rust caused by Puccinia striiformis f. sp. tritici is the most significant rust pathogen which prefers cool, moist areas and highlands. The pathogen is recognised as responsible for huge production losses in wheat. Genetic variation in pathogen makes its control difficult. Therefore, resistance against all the races of the pathogen known as durable or race-non-specific resistance is preferred. The present study was carried out to identify durable resistance against stripe rust in selected wheat cultivars from Pakistan through seedling testing, field evaluation at adult stage, morphological marker studies and marker-assisted selection. Results revealed that 4% of the cultivars were resistant at the seedling stage while the rest were susceptible or intermediate. To confirm their field resistance, the same cultivars were evaluated under field conditions at Cereal Crops Research Institute Pirsabak (located in Khyber Pakhtunkhwa, KP) a hot spot of stripe rust in Pakistan. Observations exhibited that at the adult stage 4% of the cultivars were resistant, 70% intermediate or moderately resistant while the others were highly susceptible. Leaf tip necrosis was observed in 30% of the cultivars. Wheat cultivars showing susceptibility at the seedling stage were highly to moderately resistant at adult stage showing durable resistance. For further validation, morphological markers were also observed in cultivars indicating the presence of Yr18/Lr34 gene. Eleven cultivars (C-518, Mexipak, Kohinoor-83, Faisalabad-83, Zardana-93, Shahkar-95, Moomal-2002, Wattan-94, Pasban-90, Kiran-95, and Haider-2000) were identified, having durable or race non-specific resistance against stripe rust. The diamondback moth, Plutella xylostella (L.) (Lep.: Plutellidae) is one of the most important pests of cruciferous plants throughout the world. In recent years, it has been identified as a serious pest of the cauliflower fields in Tehran province. Resistance of P. xylostella to all main groups of insecticides has been recorded and it is ranked in the 20 most resistant pest species reported until now. According to many researchers, to solve the problem of pest resistance to chemical pesticides, an integrated pest management programme should be used. Despite this condition, it seems that the use of resistant cauliflower cultivars is an appropriate policy for integrated control of the pest in the field. In order to identify the most resistant cultivar in the field, eight cauliflower cultivars in a completely randomised design with five replicates were planted at the Shahed University research field (south of Tehran). Density of eggs, larvae and pupae of P. xylostella were measured every 10 days in these cultivars. The results showed that there is no significant difference between numbers of eggs per plant on different cultivars. But number of larvae and pupae per plant were significantly different among different cultivars. Smilla and Snow mystique cultivars had the highest number of larvae and pupae. On the other hand, Buris and Snow crown cultivars had the lowest number of pupae and Snow crown and SG cultivars had the lowest number of larvae per plant. According to the results, the Buris and Snow crown cultivars had the lowest infestation and had a kind of resistance to pest.  相似文献

It is known that few wheat cultivars maintain their resistance to rust diseases for a long period of time, particularly when crop populations become genetically more uniform. A number of genetically diverse, so far unexploited, sources of rust resistance in the natural as well as mutagenized population of wheat cultivars were identified. Several of these genes were placed in agronomically superior well-adapted backgrounds so that they could be used as pre-breeding stocks for introducing genetic diversity for resistance in a crop population. Some of these stocks when employed as parents in several cross combinations in a breeding programme have generated a number of promising cultivars with diversity for resistance.Many presently grown wheats in India, near-isogenic lines each with Lr14b, Lr14ab, Lr30 and certain international cultivars were identified as possessing diverse sources of adult plant resistance (APR) to leaf rust. Prolonged leaf rust resistance in some of the Indian cultivars was attributed to the likely presence of Lr34 either alone or in combination with other APR components. Tests of allelism carried out in certain cultivars that continue to show adequate levels of field resistance confirm the presence of Lr34, which explains the role that this gene has played in imparting durability for resistance to leaf rust. Also, Lr34 in combination with other APR components increases the levels of resistance, which suggests that combination of certain APR components should be another important strategy for breeding cultivars conferring durable and adequate levels of resistance. A new adult plant leaf rust resistance source that seems to be associated with durability in Arjun has been postulated. Likewise, cultivars possessing Sr2 in combination with certain other specific genes have maintained resistance to stem rust.Further, non-specific resistances that were transferred across widely different genotypes into two of the popular Indian wheats provided easily usable materials to the national breeding programmes for imparting durable resistance to stripe rust.  相似文献

Leaf rust, caused by the fungus Puccinia triticina, is considered one of the most important foliar diseases in durum wheat. Hypersensitive resistance (HR) may be rapidly overcome by the pathogen when resistant cultivars are grown on a large acreage or following changes in virulence in the pathogen population. Prolonging the durability of the resistance requires uses of other types of resistance such as partial resistance (PR). In this study, six durum wheat lines provided by the International Center for Corn and Wheat Improvement (CIMMYT) with a high level of PR to leaf rust were studied in monocyclic tests in a growth chamber. Inoculations were performed on both primary and fifth leaves using the Spanish race DGB/BN. UV fluorescence microscopy was employed to determine microscopic components of the resistance, such as the number of early aborted infection units not associated with plant cell necrosis (EA?) and relative colony size (RCS) of the established infection units. Macroscopic components of PR such as latency period, infection frequency and uredinium size were measured as well. All six resistant lines were characterized by a higher EA? and smaller RCS respect to the susceptible control ‘Don Rafael’. Line 3 showed the highest level of PR. It had 22% of EA? compared with 4% in the susceptible control, and the smallest RCS (17% respect to RCS of ‘Don Rafael’) at adult plant stage. Both EA? and RCS had a high heritability (more than 97%) and the correlation with macroscopic parameters (latency period and uredinium size) was also high (significant at 0.001 level). Hence, PR to leaf rust in these durum wheat genotypes has been revealed at microscopic level (higher EA? and smaller RCS).  相似文献

Two Iranian common wheat landraces AUS28183 and AUS28187 from the Watkins collection showed high levels of seedling resistance against Australian pathotypes of leaf rust and stripe rust pathogens. Chi-squared analyses of rust response segregation among F3 populations derived from crosses of AUS28183 and AUS28187 with a susceptible genotype AUS27229 revealed monogenic inheritance of leaf rust and stripe rust resistance. As both genotypes produced similar leaf rust and stripe rust infection types, they were assumed to carry the same genes. The genes were temporarily named as LrW1 and YrW1. Molecular mapping placed LrW1 and YrW1 in the short arm of chromosome 5B, about 10 and 15 cM proximal to the SSR marker gwm234, respectively, and the marker cfb309 mapped 8–12 cM proximal to YrW1. LrW1 mapped 3–6 cM distal to YrW1 in two F3 populations. AUS28183 corresponded to the accession V336 of the Watkins collection which was the original source of Lr52. Based on the genomic location and accession records, LrW1 was concluded to be Lr52. Because no other seedling stripe rust resistance gene has previously been mapped in chromosome 5BS, YrW1 was permanently named as Yr47. A combination of flanking markers gwm234 and cfb309 with phenotypic assays could be used to ascertain the presence of Lr52 and Yr47 in segregating populations. This investigation characterised a valuable source of dual leaf rust and stripe rust resistance for deployment in new wheat cultivars. Transfer of Lr52 and Yr47 into current Australian wheat backgrounds is in progress.  相似文献

Brown rust of barley (Puccinia hordei) is widespread in New Zealand and causes crop losses in some years. The cultivars presently grown lack adequate genetic resistance but a highly resistant line has been produced. Twelve different virulence combinations were identified in the pathogen population using the Welsh differential lines; there were differences between North Island and South Island isolates; and no virulence was observed for resistance conferred by factors Pa 3 and Pa 7.  相似文献

The extent to which resistance to leek rust (Puccinia allii) in commercially useful cultivars of leek (Allium ampeloprasum) could be developed by selection within current commercial cultivars or their recent predecessors was investigated. Differences in resistance among commercial cultivars was not great. However, a wide range of resistance levels was found in both half‐sib and inbred progeny derived from these cultivars. In a crop simulation, one of the breeding lines showed a reduction in the rate of new pustule appearance of 44% compared to cv. Wintra. It is concluded that selection within modern cultivar level material is a potentially productive strategy for the commercial development of leek rust resistance in the short to medium term.  相似文献
