Marine blue-green algae, Lyngbya Lagerheimii, Microcoleus chthonoplastes, Plectonema terebrans, Agmenellum quadruplicatum, and freshwater blue-green algae, Anacystis nidulans, Anabaena variabilis, Nostoc muscorura, and Oscillatoria sp. treated with lysozyme (muramidase) formed spheroplasts but not protoplasts. The time needed for spheroplast induction varied with the species. Approximate internal osmotic pressures of the blue-green algae were determined. The intracellular distribution of glycollate dehydrogenase EC 1.2.1.17 has been investigated in extracts of the cyanobacteria (blue-green algae) Anabaena cylindrica, Nostoc muscorum and Chlorogloea fritschii. Most of the enzyme activity was associated with a chlorophyll-containing cell-free pellet, which also exhibited Photosystem I and II activities. Sucrose density gradient centrifugation of this washed pellet resulted in the formation of a green band within which maximal chlorophyll concentration and enzymic glycollate oxidation coincided. Antiserum raised to this fraction obtained from A. cylindrica inhibited glycollate dehydrogenase and Photosystem II activity. The data indicate that most of the cyanobacterial glycollate dehydrogenase is associated with the thylakoids and thus provide evidence for the dual role of these membranes in photosynthetic and respiratory processes.Abbreviations DCMU 3-(3,4-dichlorophenyl)-1,1-dimethylurea - DCPIP 2,6-dichlorophenolindophenol - DPC diphenylcarbazide  相似文献

Several blue-green algae were surveyed for the occurrence of the hydrogenase which was assayed by the oxyhydrogen or Knallgas reaction in the intact organisms. In aerobically grown cultures, the reaction was detectable in Anabaena cylindrica, Nostoc muscorum and in two Anabaena variabilis species, whereas virtually no activity was observed in Anacystis nidulans and Cyanophora paradoxa. In these latter two algae, the reaction was, however, found after growth under molecular hydrogen for several days, which drastically increased the activity levels with all the algae tested. In the nitrogen fixing species, the activity of the Knallgas reaction was enhanced when all combined nitrogen was omitted from the media. H2 and hydrogenase could not significantly support the CO2-fixation in photoreduction experiments with all blue-green algae investigated here. Hydrogenase was assayed by the dithionite and methyl viologen dependent evolution of hydrogen and was found to be present with essentially the same specific activity levels in preparations of both heterocysts and vegetative cells from Anabaena cylindrica. Na2S2O4 as well as H2 supported the C2H2-reduction of the isolated heterocysts. The H2-dependent C2H2-reduction did not require the presence of oxygen but was strictly light-dependent where H2 served as an electron donor to photosystem I of these cells. Abstract Light-induced proton efflux has been investigated with intact cells of Anabaena, Nostoc, Anacystis , and Aphanocapsa . The proton efflux by filamentous blue-green algae is biphasic, strongly inhibited by ortho -vanadate and insensitive to cyanide. These data are taken as evidence for a proton-pumping ATP-hydrolase present on the cytoplasmic membrane of Anabaena and Nostoc .  相似文献

Summary Indole-3-acetic acid (IAA) stimulated the growth (increase in dry weight) of the blue-green algae Anacystis nidulans, Chlorogloea fritschii, Phormidium foveolarum, Nostoc muscorum, Anabaena cylindrica, and Tolypothrix tenuis and the green algae Chlorella pyrenoidosa, Ankistrodesmus falcatus and Scenedesmus obliquus growing under as sterile conditions as possible. The optimum concentration varied from species to species; in the blue-green algae it ranged from 10-5 to 10-9 M and in the green algae it was 10-3 M. These results are discussed in the light of present studies in this field.  相似文献

Blue-green algae as a soil-based inoculum was applied to short-duration Indica rice in combination with 72 kg N/ha and compared with just N fertilization applied as 144 kg N/ha. Fertilizer N was applied in two equal doses 25 days after transplanting and at mid-tillering stage. The algal inoculum, which containedAnabaena cylindrica, Anabaena oryzae, Nostoc muscorum andTolypothrix tenuis, was applied at 100 kg/ha fresh material (90% moisture) 5 days after transplanting. Five different combinations of microelements were sprayed as a foliar application simultaneously with fertilizer N. Plant performance was enhanced by inoculation with algae and microelements compared with complete N fertilization only. Natural infestation with the stem borer,Chilo agamemnon, and leaf miner,Hydrellia prosternalis, decreased significantly during growth and up to harvesting with application of algae, Endosulfan, and increased with application of microelements.  相似文献

Photochromic Pigments from Blue-Green Algae: Phycochromes a, b, and c   总被引:1,自引:0,他引:1  
Aqueous extracts of blue-green algae were fractionated by electrofocusing. In all algae investigated, fractions with iso-electric points at or near 4.6 showed photochromic behaviour analogous to that of phytochrome, although they were sensitive to light of shorter wavelength. Three main types of photochromic pigments were found: Phycochrome a (in Tolypothrix distorta, Phormidium luridum, Nostoc muscorum 1453/12, and Anacystis nidulans) has one form absorbing maximally at about 590 nm (formed under red light) and one absorbing maximally at about 630 nm (formed under green light). Phycochrome b (in Tolypothrix distorta) has one form absorbing maximally near 510 nm and one form absorbing maximally at 570 nm (formed in yellow-green and blue-green light, respectively). Phycochrome c (in Nostoc muscorum A and probably in Tolypothrix tenuis) has one form absorbing maximally at 650 nm (formed under green light) and one absorbing very weakly in the green region (formed under red light). The conversion of Phormidium phycochrome a from its red-absorbing form to its green-absorbing form causes the same spectral change as if an f-chromophore of phycocyanin were transformed into an s-chromophore. The quantum yield for this conversion is estimated to be 0.1, while the quantum yield for the reversion is estimated to be 0.4 on the assumption that the absorption coefficients are those of f- and s-chromophores. Phycochrome c is less light-sensitive than phycochromes a and b.  相似文献

Inoculation of the Japonica rice, Giza 171, with blue-green algae along with 36 kg N/ha or complete N fertilization by 108 kg N/ha were tested along with blast-controlling fungicides: Kitazin-17, Fuji-one and Beam. The algal inoculum, which containedAnabaena cylindrica, Anabaena oryzae, Nostoc muscorum and Tolypothrix tenuis, was applled as 100 kg soil-based inoculum (90% moisture)/ha 5 days after transplanting. Plant growth, yield, N content and fertilizer-N-use efficiency were increased and the incidence and severity of leaf and neck intection withPyricularia oryzae were decreased by treatment with the fungicides applied with the algae along with 36 kg N/ha inslead of 108 kg N/ha without algae.

Summary A model has been proposed to account for growth inhibition by L-histidine in a variant strain of Nostoc muscorum. This strain has been characterized for its response to 3-amino-1,2,4-triazole and 1,2,4-triazole-3-alanine known to act as false corepressors of the histidine biosynthesis genes. The histidine sensitive strain retained its sensitivity to triazole alanine while the inhibitory effects of aminotriazole were much reduced indicating a change in regulation of his genes. The probable interactions between nif and his genes in cyanobacteria (blue-green algae) have been discussed.  相似文献

The present study focused on the responses of six freshwater algal species (Anabaena ambigua, Anabaena subcylindrica, Nostoc commune, Nostoc muscorum, Spirogyra sp., and Spirulina sp.) to manganese and zinc. Laboratory experiments were conducted for the assessment of biochemical responses to manganese and zinc at various concentrations (0.1, 0.5, 1.0, 2.0, 3.0, 4.0, and 5.0 mg/L) for 15 days of exposure. After the incubation period, 10 ml of sample was centrifuged at 6000 rpm for 15 min and the pellets were used for measurement of the various experimental parameters. The toxicological study of manganese on algae showed that Anabaena ambigua was most sensitive algae. Regarding effects of manganese concentrations, chlorophyll, protein, carbohydrate, starch, and amino acid were inhibited 50% (IC50) at 3 mg/L, whereas the toxicological study of zinc on algae Anabaena subcylindrica showed most adverse effects. Regarding effects of zinc concentrations, chlorophyll, protein, carbohydrate, starch, and amino acid were inhibited 50% (IC50) at 1 mg/L. The inhibitory and stimulatory effects of either of the used heavy metals depend on concentration. Different organisms, however, have different sensitivities to the same metal, and the same organism may be more or less damaged by different metals.  相似文献

The physiological and biochemical changes during the adaptation of Nostoc muscorum to salt are accompanied by specific structural changes. Cells of Nostoc muscorum exposed to saline medium vary in size and envelope organization. There are also drastic changes in the intracellular organization of the thylakoidal assembly. The heterocysts exhibit a preferential tolerance to NaCl rather than mannitol. These findings suggest that Nostoc muscorum is equipped with a specific physiological capacity for NaCl tolerance.  相似文献

Respiratory particles containing an aa3-type cytochrome oxidase were prepared from Anacystis nidulans, Synechocystis 6714, Synechococcus lividus, Anabaena variabilis, Nostoc sp. strain MAC, Nostoc muscorum, and Mastigocladus laminosus. Oxidation of c-type cytochromes by membrane preparations of the different blue-green algae was observed using purified cytochromes from horse heart, Candida krusei, tuna, Saccharomyces oviformis, Rhodospirillum rubrum, Rhodospirillum molischianum, Rhodopseudomonas palustris, Rhodocyclus purpureus, Paracoccus denitrificans, Anacystis nidulans, Anabaena variabilis, Euglena gracilis, and Scenedesmus obliquus. Rapid oxidations were consistently observed with the mitochondrial c-type cytochromes (horse heart cytochrome c reacts most rapidly) and with cytochromes c2 from Rhodopseudomonas palustris and Rhodocyclus purpureus; in contrast, the cytochrome c2 from Rhodospirillum rubrum and the plastidic cytochromes from E. gracilis and Scendesmus obliquus were inactive with all membrane preparations. All reactions were inhibited by low concentrations of KCN, NaN3, and CO, and they were activated by Tween 80, thus indicating participation of the terminal oxidase. The results are discussed in view of the spectral similarities between the terminal oxidase of blue-green algae and the mitochondrial aa3-type cytochrome oxidase of plants and other eukaryotes.  相似文献

Summary The efficiency of the inoculation of three cultures of N-fixing blue-green algaeviz. (i)Aulosira fertilissima (A1), (ii)Nostoc muscorum (A2) and (iii) their mixture (A3) in increasing the grain and straw yield of rice, nitrogen uptake in grain and nitrogen content in soil was studied in a green house experiment with an alluvial soil in presence or absence of urea nitrogen application. Inoculation significantly increased the grain and straw yield of rice and nitrogen uptake in grain, but the efficiency of inoculation gradually decreased with the increase in the levels of urea nitrogen application, the extent of decrease varying with the algal cultures inoculated. The nitrogen content in the soils after the crop harvest recorded a significant increase due to inoculation but after air drying the soil a marker decrease of the same was observed, which indicated that most of the nitrogen added to the soil by blue-green algae through fixation did not persist after air drying the soil.  相似文献

Electron microscopic studies of Sphagnum lindbergii (Schimp.) and S. riparium (Ångstr.) have revealed the presence of intracellular organisms such as blue-green algae, green algae, bacteria and fungi. Nitrogenase activities of these Sphagnum mosses were found to be related mainly to the presence of intracellular Nostoc filaments. The appearance of nitrogen-fixing blue-green algae within bryophytes is thus not restricted to liverworts. The association is likely to be of ecological importance as it seems to occur in very acid habitats generally lacking blue-green algae. Possible interrelations between the moss, the blue-green algae and different types of bacteria are discussed.  相似文献

Summary A variant strain of the nitrogen-fixing bluegreen alga Nostoc muscorum, displaying impairment in its elemental nitrogen dependent growth and complete inhibition of growth by L-histidine in otherwise nitrogen-free medium, has been isolated and characterized for its response to L-glutamic acid and L-glutamine in presence of other inorganic nitrogen sources. A model based on the possibility of nif-his interaction has been proposed to account for the observed behaviour of the strain. It is inferred that the two sets of genes may occupy neighbouring positions on the blue-green algal genome.  相似文献

Zea mays (Sakha 321 ) andPhaseolus vulagris (Giza 6) were irrigated with sewage wastewater treated withNostoc muscorum andAnabaena subcylindrica. The results showed that irrigation of plants by untreated or treated wastewater caused stimulation in the measured growth parameters of both plants. In addition pigmentation as well as protein and carbohydrate contents were stimulated. This stimulation was attributed to the presence of high levels of essential nutrients such as nitrogen, phosphorus, and organic matters in wastewater. On the other side, there was a slight inhibitory effect of wastewater on some measured growth parameters of the plants. Summary Nitrogen fixation (C2H2 reduction) by blue-green algae occurring on the juvenile lava field of Heimaey, Iceland was examined both in the laboratory (potential at 20° C and 39° C) and in the field, three and a half years after the volcanic eruption.Already at this early stage of colonization representatives of unicellular and filamentous heterocystous and non-heterocystous blue-green algae were commonly observed. The predominating algae were Nostoc sp. (20° C) and Schizothrix sp. — Microcoleus chthonoplastes, (39° C), the former often in association with the protonemata-rhizoids of moss plants.The potential for nitrogen fixation was recorded at an average rate of 109.2 (20° C) and 138.1 (39° C) ng N g-1 h-1 in soil collected from localities randomly distributed over the lava field.Tests for nitrogen fixation performed in situ revealed significant fixation activities in all the eleven localities subject to examination. The activities ranged from 2.8 to 63.4 (mean 21.5) ng N g-1 h-1 and 1.9 to 17.7 (mean 7.9) ng N cm-2 h-1.All the nitrogen fixation data noted imply that blue-green algae contribute a substantial part of the nitrogen input to the lava. Further, it was found that material incubated under micro-aerophilic conditions exhibited considerably enhanced nitrogenase activity.The role of nitrogen-fixing blue-green algae in general and Nostoc muscorum in particular in being suitable as pioneering organisms preparing the bare lava for ingress of other plants is also discussed.  相似文献

Summary 15N tracer was used to detect the extent to which nitrogen of appliedAzolla caroliniana, Anabaena variabilis andNostoc muscorum was available for assimilation by the growing rice plants in pots under 4 cm flood water for 60 days. The rate of release of nitrogen from the above biofertilizers, the amount of nitrogen remaining in the soils and the amount that was lost from the soils during this period were also examined. Previously15N-labelled biomass of Azolla, Anabaena and Nostoc to provide 40 mg N was mixed thoroughly with 0.5 kg silt loam Bangladesh soil (Sonatola series) in each of three pots used for a single treatment. Each pot received four 16 days old IR8 rice seedlings. A parallet set of experiments was conducted without rice plants.It was found that nitrogen uptake in the rice plants was increased by 91, 176 and 215% on using Azolla, Anabaena and Nostoc which resulted in increased total dry matter yields (shoot plus root) of 74, 105 and 125%, respectively. Of the total15N applied at the start, 26, 49 and 53% was released from Azolla, Anabaena and Nostoc; about 7, 14 and 13% was lost by denitrification and 74, 51 and 47% remained in the soils as the undecomposed part of the biofertilizers, respeciively, after 60 days. Of 15.76, 22.72 and 25.92 mg N assimilated by the rice plants, 48, 61 and 62% was supplied by Azolla, Anabaena and Nostoc, respectively. The rest was obtained from the soil used.In the absence of the rice plants 30, 43 and 45% of applied15N of Azolla, Anabaena and Nostoc was released, respectively, in 60 days of which 93–96% was lost as N2 through denitrification.  相似文献
