During the extreme 1992–1997 El Niño drought event, widespread stem mortality, or tree dieback, of both mature and juvenile eucalypts occurred within the tropical savannas of northeast Australia. Most of the dieback occurred in individuals of the ironbark species complex (Eucalyptus crebra E. xanthoclada) while individuals of the bloodwood species Corymbia erythrophloia, exhibited significantly less stem mortality. Indicative of greater water stress, predawn and midday xylem water potentials of ironbark adults and saplings were significantly more negative than predawn values of bloodwoods. The very negative xylem water potentials in ironbarks suggest that stem mortality in both adult and juvenile ironbarks results from drought-induced embolism and that ironbarks perhaps have a shallower and less extensive root system than bloodwoods. Although predawn and midday water potentials for ironbark adults and saplings were similar, a census of mature and juvenile ironbark trees indicated that mortality was higher in adult trees. Cavitation vulnerability curves indicated that ironbark saplings may be better buffered against cavitation than adult trees. If they possess smaller root systems, saplings are more likely than adults to experience low xylem water potentials, even in non-drought years. Xylem conduits produced in adult trees during periods of normal rainfall, although perhaps more efficient in water conduction, may be more vulnerable to cavitation during infrequent severe droughts.  相似文献

Summary Xylem cavitation has been studied in Ricinus plants using vibration detection to examine its induction by different factors. These observations provide considerable circumstantial evidence in justification of the new technique as already described and further developed. In general cavitation is induced only when the tissue water balance is reduced hydrostatically. Thus cavitation is promoted by intense radiation which enhances transpiration, or alternatively by the blockage of xylem conduits by suspended particles carried in the transpiration stream. In contrast a reduction in radiation, or prevention of transpiration tends to restrict cavitation. Thus cavitation can be prevented by immersing a leaf in liquid paraffin. This technique has been used to see if radioactive bombardment would trigger its induction but no detectable effect has been observed even when exposed to intense radiation.An excised leaf, losing water in air, produces a click total. On restoration to full turgor by standing the petiole in water it recovers very slowly and subsequently its click total is much reduced. If however the newly wilted leaf is allowed to recover in water following gas evacuation treatment the cavitation total often approaches the original and the rate of recovery is extremely rapid. Apparently gas emboli develop rapidly in conduits which have cavitated, but they can be removed by vacuum injection: the conduits refill and conduction is restored.  相似文献

Summary A change occurs in the relationship between xylem water potential (x) measured with the pressure chamber and leaf water potential ((w)) when a period of post-excision water loss is allowed before measurement of (x) and (x). When this occurs, water stress is over-estimated by the pressure chamber measurement. Over the same period of water loss, cavitation vibrations have been detected acoustically in excised leaves. It is suggested that the measurement of (x) is affected by emptying of some of the xylem vessels due to cavitation. This would require that additional pressure be applied to a leaf in the pressure chamber in order to measure (x).  相似文献

Zusammenfassung Fruchtbarkeit und Wachstum von vier auf Ackerbohnen (Vicia faba) lebenden Aphiden-Arten (Aphis fabae, A. craccivora, Acyrthosiphon pisum, Megoura viciae) werden von den Sorten Schlanstedter und Rastatter in gegensätzlicher Weise gefördert bzw. gehemmt. Post-infektionelle Resistenzeffekte treten daher einerseits bei Rastatter gegen A. fabae, andererseits aber bei Schlanstedter Ackerbohnen gegen Erbsen- und Wickenläuse auf, während sich A. craccivora auf beiden etwa gleich gut entwickelt.
Summary Aphis fabae, Aphis craccivora, Acyrthosiphon pisum, and Megoura vicae (abbreviated Af, Ac, Ap and Mv, respectively) were allowed to develop under similar uncaged conditions on Rastatter (R) and Schlanstedter (S) field beans. Large, sometimes contradictory, differences in the fecundities and weights attained by adult apterous virginoparae on these plants were recoreded and ascribed to postinfestation resistance effects.Ap, which feed only on leaves, and Mv, which feed exclusively on internodes, weigh significantly more (17–18%) on R than on S. In contrast, Af, which feed on various plant parts, are 61% heavier on the leaves and 33% heavier on the stems of S than on the corresponding parts of R.However, Ac, which have a distribution on the plants similar to that of Af, are somewhat heavier (12%) on R-leaves and on S-stems than on the corresponding plant parts of the other variety. Such differences are greatly diminished if the aphids are reared on excised leaves and stems immersed in water. In this case growth of the aphids on excised parts of the resistant varieties (on which they previously grew poorly) closely approaches that on the intact satisfactory varieties (on which they grew well). Only small (Af, Ac) or no (Ap, Mv) improvement in growth results when the aphids are reared on excised parts of the satisfactory variety. Thus, the weights of apterous adults of Ap on excised S-leaves were only 17% greater than on intact S-leaves, whereas they were similar on excised and intact R-leaves. For Af, adult weights were 81% greater on excised R-leaves, 51% greater on excised S-leaves, and only 19% or no greater on excised R- and S-stems, respectively, than those on the corresponding intact plant parts.The possibility is discussed that these contradictory effects result from a mobilization and increase of proteinaceous constituents in the excised plant parts. Whereas the nutrition and growth of aphids on the resistant variety may thus become optimal, such an increase above an already optimal nutrient level on the satisfactory variety would not increase growth beyond the limit already achieved by a species. It is further deduced that postinfestational resistance to aphids of field beans depends on the nutrient value of the sap available to the aphids. However, since the nutritional demands of different aphid species may also differ, it is unlikely that a single plant species will be resistant to aphids in general.

Summary The effects of CaCl2, (2-chloroethyl) phosphonic acid (Ethephon) and ethylene on leaf abscission of debladed and intact bean plants (Phaseolus vulgaris L.) were studied. Ethephon (1000 g/l) and ethylene (8 l/l) induced abscission in debladed and intact plants in 24–72 h whereas IAA (10-5M), cycloheximide (10-5M) and CaCl2 (0.068M) delayed abscission in debladed plants. CaCl2 completely inhibited the abscission-enhancing effect of Ethephon in intact bean leaves. When CaCl2 and Ethephon were applied simultaneously to separate halves of the leaf blade, leaves with Ethephon applied closest to the pulvinus abscised rapidly; when CaCl2 was applied closest to the pulvinus, abscission was prevented. Summary Stomata of paper birch (Betula papyrifera Marsh.) seedlings were more open at high humidity than at low humidity and responded rapidly to changes in vapor pressure deficit. SO2 at 0.2 or 0.8 l l-1 caused partial stomatal closure. Seedlings fumigated with SO2 at 0.2 or 0.5 l l-1 for 30 h or 0.2 l l-1 for 75 h took up more SO2 at high than at low humidity. Differences in pollutant uptake could be explained by stomatal conductance with no need to invoke changes in mesophyll conductance. Betula seedlings were more sensitive to SO2 when fumigated at high humidity, as manifested in more leaf necrosis, increased leaf abscission, and greater growth inhibition compared to seedlings fumigated at low humidity. Maximum shoot induction from stem explants ofCanavalia lineata was obtained with an agar-solidified PC medium containing 10 M benzylaminopurine and 1 M naphthaleneacetic acid. Rooting of thesein vitro produced shoots was achieved with hormone-free PC medium. Canavanine was produced almost exclusively in the leaves and was not detected in the roots ofin vitro propagatedC. lineata. To exclude the possibility of imminent translocation of canavanine from the root to leaf, adventitious roots were induced from leaf explants in PC medium supplemented with 1 M kinetin and 20 M indole-3-acetic acid and subcultured in medium lacking growth regulators, and the roots excised from germinated seedlings were cultured in hormone-free PC medium. All the roots were incapable of accumulation of canavanine. These results suggest that leaves ofC. lineata are the possible site of canavanine synthesis.  相似文献

The effects of N-(2,4-dimethyl-5-(((trifluoromethyl)sulfonyl)amino)phenyl)acetamide (mefluidide) and 3,6-dichloro-o-anisic acid (dicamba) on in vitro growth and somatic embryogenesis ofDactylis glomerata L. (orchard grass) were studied using suspension cultures and explanted leaf bases. All experiments employed modified Schenk and Hildebrandt medium amended with concentrations of dicamba ranging from 15 to 120 M (SH-15 to SH-120) and of mefluidide ranging from 1 to 100 M. SH medium without either growth regulator was used for embryo germination. Embyro production in suspension cultures with SH-30 medium plus 3 g/L casein hydrolysate was significantly reduced by 1 M mefluidide. Only 15% of these embryos germinated and produced plants compared to 84% from controls. Growth, as measured by dry weight, was significantly reduced by 50 or 100 M mefluidide. The number of embryos formed on leaf sections was significantly reduced by 20 or 25 M mefluidide. Embryos that formed with 10 M or more mefluidide were callused on both SH-15 and SH-30 media. Shoot formation was inhibited from individual embryos and embryo/callus masses that developed on either SH-15 or SH-30 medium containing 5 M or more mefluidide. Radicle emergence was significantly reduced with 10 M mefluidide regardless of 15 or 30 M dicamba. Histological examination revealed that mefluidide inhibited both shoot and root meristem development with shoot development being the more sensitive. Inhibition of both was independent of dicamba concentrations. Shoot formation was also reduced from embryos that had developed on SH-30 medium without mefluidide when transferred to medium containing mefluidide without dicamba.  相似文献

Summary Midday closure of stomata of well-watered ( between-10 and-25 bar) or moderately stressed ( between-25 and-35 bar) Arbutus unedo plants occurs when midday leaf temperatures increase above 30°C and vapor pressure difference between leaf air spaces and the external air increases above approximately 30 mbar/bar. Moderate water stress decreases maximum conductance and may result in greater sensitivity to high leaf temperature and vapor pressure dificit, which results in earlier closure and later reopening of stomata. Severe water stress ( of-50 bar) changes the form of the daily pattern observed for leaf conductance. A single morning peak in conductance occurs followed by decrease in conductance over the remainder of the day. Morning fog in Portugal during the dry season may facilitate stomatal opening and may allow improvement of carbon balances of the leaves for short periods, but contributes little to improvement of plant water balances over the longer term.  相似文献

Summary In roots from which the root cap and quiescent center have been removed new apical tissues regenerated in line with the main axis of the root. Regeneration of these tissues occurred from the region of the proximal meristem, which extends for no more than 350 m from the cut surface. Accompanying the regeneration of new apical tissues is a change in the architecture of the root apex and initiation and enlargement of a new quiescent center. A possible role for the quiescent center in the establishment of pattern at the apex is considered. The effects of benzyladenine (BA) on the mesophyll functioning, such as osmotic potential (), the effect of the inhibitors of +-ATPase on the influx of 14C-sucrose, the direction of carbon metabolism, and the rate of dark respiration, were followed in the detached leaves of pumpkin (Cucurbita pepo L.) and broad beans (Vicia faba L.). BA elevated and established a gradient of (p) between the treated and untreated leaf regions. The inhibitors of H+-ATPase did not affect the BA-induced influx of 14C-sucrose. The changes were accompanied with the elevated synthesis of starch and other polymeric compounds and the diminished synthesis of the substances of relatively low molecular weight. The stimulation of dark respiration was short and inconsiderable. The author concludes that the BA-induced transport was a passive process related to a increase. Leaf expansion accompanied by the synthesis of high-molecular-weight substances essential for cell growth and by starch synthesis apparently increased the sink capacity of the BA-treated detached leaves. The diminished efflux from the leaf blade was probably related to a lowered level of the transportable carbon compounds restricting their entry into the phloem. The influx induction could result from the activation of growth and metabolic processes, the decline in the number of organic molecules per cell volume unit, and the development of p between the source and sink leaf regions.  相似文献

Axillary shoots from three selected white ash (Fraxinus americana L.) clones were harvested from in vitro shoot cultures. Roots were initiated by pulsing excised shoots for eight days in the dark in MS medium supplemented with 2% sucrose, 0.7% agar, 5 M NAA, and 1 M IBA. Pulsed shoots were transferred to a root elongation medium consisting of 25% MS macrosalts, full-strength microsalts and organics, 1% sucrose, 0.7% agar and no auxins. When roots were visible (6–10 days after transfer to root elongation medium), microplants were transferred to vessels containing the same minimal medium and tall fescue (Festuca elatior var. arundinacea (Schreb.) Wimm.) leaf extracts, leaf leachates, or soil leachates from plant boxes with and without tall fescue sod. After four weeks in vitro, primary adventitious and secondary root growth was reduced by extracts obtained from 5 and 10 g ground leaves per 100 ml of medium. Leachates obtained from 5 g soaked leaves per 100 ml of medium stimulated primary root growth. Soil leachates from bare soil also stimulated primary root growth. Variation was observed among the clones for root growth when plantlets were grown in extracts or leachates from tall fescue.  相似文献

TheN-linked carbohydrate chains of the-subunit of highly purified urinary human chorionic gonadotropin have been re-investigated. The oligosaccharides were released enzymatically by peptide-N 4-(N-acetyl--glucosaminyl)asparagine amidase-F, and fractionated by a combination of FPLC and HPLC. As a result of the application of improved fractionation methods, apart from the earlier reported carbohydrate chains, also small amounts of trisialo tri- and tri-antennary oligosaccharides were found. Summary Wheat seedlings were grown under a 14-hour photoperiod and the first leaves excised at the end of the eighth dark period. The effect of treatments causing wilting on the inhibitor- content of such leaves was studied.When leaves were rapidly wilted (i.e. to a 6% fresh weight loss) and extracted immediately, the amount of inhibitor- per leaf was found to be the same as in fresh turgid leaves. However, when the leaves were maintained in a wilted condition in darkness for a period of 110 minutes, there was a marked increase in inhibitor- content.The greater the degree of wilting (i.e. up to about a 9% loss in fresh weight) the greater the eventual inhibitor- content. Moreover, the increment in inhibitor- was shown to be temperature dependent.The time lapse requirement and the temperature dependency of the inhibitor- formation suggest an enzymic conversion from a precursor.If a similar phenomenon occurs during the wilting of intact plants then the increase in this growth inhibitor might play a role in some of the physiological changes which accompany water stress.  相似文献

A presumed XY chromosome pair is described fromt estis squashes from the mesopelagic deep-sea fish Bathylagus wesethi, whose 2N chromosome number was determined as 34-XY. Although the metacentric X-chromosome is the largest in the entire compliment, the Y is the smallest and only acrocentric element. The positive heteropycnosis of the sex elements was not easily distinguishable in the nuclei of first meiotic prophase. Tetraploid nuclei were observed in peripheral supporting cells of the testis. The acrylamide quenching of the intrinsic tryptophanyl fluorescence of normal and sickle apohemoglobins has been studied in 0.05 M potassium phosphate buffer,pH 7.5, at 5°C over a protein concentration range from 1 to 50M. Analysis of quenching dynamics revealed a strong dependence on acrylamide concentration for the intrinsic fluorescence of both normal and sickle apohemoglobins, suggesting that one tryptophanyl residue [presumably that at position 37(C3)], was more accessible to collisional quencher than the other tryptophanyl residue [15(A12)]. Additional studies, which altered viscosity and subunit assembly experimental parameters, supported the assignment of residue 37 as the more dynamically accessible residue. Finally, the quenching data were also found to be dependent on protein concentration, implying that this difference in the mobility between the two residues is a sensitive probe of self-aggregation. Extrapolated dynamic quenching constants at low concentration of acrylamide were used to estimate the dimer-monomer equilibrium dissociation constants of normal and sickle apohemoglobins, and were found to be 5.6 and 2.4M, respectively, thus demonstrating distinct self-association properties of A and S apohemoglobins.  相似文献

The lack of comparative reviews of the efficacy of EMG frontalis biofeedback versus alternative procedures for reduction of muscle tension prompted the present meta-analytic treatment of literature previously concluded to be equivocal. Twenty studies comparing EMG frontalis biofeedback with other tension-reduction procedures produced a total of 68 separate effect sizes suitable for meta-analysis. Differences between clinical and normal samples were nonsignificant, and data analyses revealed that EMG frontalis biofeedback was significantly superior to control (p<.05) but that alternative forms of muscle relaxation, while effective, did not reach statistical significance.  相似文献

A volume of tissue of lettuce (Lactuca sativa L.) plants extending 2 mm basipetally from the apical meristem and including leaf primordia and young expanding leaves was surveyed using electron-probe microanalysis (EPMA) on both frozen-hydrated and freeze-dried samples. This analysis was carried out either 2 or 5 d following NaCl salinization of the medium from the 10 mol · m–-3 control level up to 80 mol · m–-3. The objective was the investigation of possible changes in the nutritional status of the apical meristem that might account for some aspects of salt-induced growth inhibition. Sodium and chloride increased significantly in tissues basal to the apical meristem, while both phosphorus and potassium decreased in the same region. These changes were evident in specimens collected just 2 d after the commencement of salinization (20 h after completion of the salinization) and were not exacerbated by an additional 3 d of treatment; they were present in tissue as close as 100 m to the meristem and extending down to 500 m. The apical 10–50 m were relatively protected from both the increase in sodium and chloride and the decrease in phosphorus and potassium that occurred in more basal regions. Young leaves (up to 1.5 mm in length) appear to control their own mineral nutrient levels when challenged by salinization of the medium, presumably because of altered growth. A decrease in the concentration of total Ca as a result of salinization was significant in cells 500 m basal to the meristem, but was evident as a tendency in the data even within the first 50 m. Using an improved automatic method for the analysis of calcium by EPMA, it was found that total Ca was reduced by salinization, especially in basal regions (500 m below the apex) and also in young leaves (1–1.5 mm in length). We suggest that the nutrition of the shoot apical meristem may be disturbed soon after salinization and that the shoot meristem might be the source of a signal to expanding leaves, as well as exerting its own direct influence over leaf emergence.Abbreviation EPMA electron-probe microanalysis This work was supported by U.S. Department of Agriculture grant 87-CRCR-1-2462.  相似文献

Summary Colonies of an Australian swarm founding polistine wasp,Ropalidia romandi, on the Atherton Tableland, Queensland, manage perennial nests by practicing seasonal colony activity. In summer nests, most cells were occupied by immatures of all developmental stages. The proportion of cells that did not contain immatures increased towards winter, when most nest cells did not contain immatures and half of these winter nests had rather large amounts of nectar deposited in empty cells. While the visits to tree flowers byR. romandi were not frequent in summer, nearly 60 % of insects collected on tree flowers in winter wereR. romandi. With such seasonal switching in the modes of colony reproductive activity and of nectar foraging behavior,R. romandi responds to its physical and biotic environment. In this way,R. romandi may be able to continue colonies throughout the year, with much reduced level of colony activity in winter, in areas with seasons that are unfavorable for reproductive activity of a colony.  相似文献

A catalytic fragment, 1-298, derived from limited chymotryptic digestion of phosphorylaseb kinase (Harris, W.R.et al., J. Biol. Chem., 265: 11740–11745, 1990), is reported to have about six-fold greater specific activity than does the subunit-calmodulin complex. To test whether there is an inhibitory domain located outside the catalytic core of the subunit, full-length wild-type and seven truncated forms of were expressed inE. coli. Recombinant proteins accumulate in the inclusion bodies and can be isolated, solubilized, renatured, and purified further by ammonium sulfate precipitation and Q-Sepharose column. Four out of seven truncated mutants show similar ( 1-353 and 1-341) or less ( 1-331 and 1-276) specific activity than does the full-length wild-type , 1-386. Three truncated forms, 1-316, 1-300, and 1-290 have molar specific activities approximately twice as great as those of the full-length wild-type and the nonactivated holoenzyme. All recombinant s exhibit similarK m values for both substrates, i.e., about 18M for phosphorylaseb and about 75 M for MgATP. Three truncated s, 1-316, 1-300, and 1-290, have a 1.9- to 2.5-fold greater catalytic efficiency (V max/K m) than that of the full-length wild-type and a 3.5- to 4.5-fold greater efficiency than that of the truncated 1-331. This evidence suggests that there is at least one inhibitory domain in the C-terminal region of , which is located at 301-331· 1-290, but not 1-276, which contains the highly conserved kinase domain, is the minimum sequence required for the subunit to exhibit phosphotransferase activity. Both 1-290 and 1-300 have several properties similar to full-length wild-type , including metal ion responses (activation by free Mg2+ and inhibition by free Mn2+) pH dependency, and substrate specificities.  相似文献
