In a bid to identify suitable microbial indicators of compost stability, the process evolution during windrow composting of poultry manure (PM), green waste (GW) and biowaste was studied. Treatments were monitored with regard to abiotic factors, respiration activity (determined using the SOUR test) and functional microflora. The composting process went through typical changes in temperature, moisture content and microbial properties, despite the inherent feedstock differences. Nitrobacter and pathogen indicators varied as a monotonous function of processing time. Some microbial groups have shown a potential to serve as fingerprints of the different process stages, but still they should be examined in context with respirometric tests and abiotic parameters. Respiration activity reflected well the process stage, verifying the value of respirometric tests to access compost stability. SOUR values below 1 mg O2/g VS/h were achieved for the PM and the GW compost.  相似文献

AIMS: Determination of the minimum requirements (time-temperature relationship and moisture content) that are needed for a sufficient eradication of an indicator organism. METHODS AND RESULTS: To determine the hygienic safety of composting processes, the indicator organism Salmonella enterica ssp. enterica serotype Senftenberg strain W 775 (further abbreviated as W 775) was artificially inoculated on a meat carrier and monitored subsequently. Different types of composting processes, e.g. composting in enclosed facilities, in open-air and in-vessel composting, were investigated. The waste feedstocks used in this work were either biowastes (i.e. vegetable, fruit and garden wastes; also called source-separated household wastes) or pure garden wastes. Beside these large-scale trials, we also conducted some lab experiments in order to determine the impact of temperature, moisture content and the presence of an indigenous microflora on the eradication of W 775. We found the temperature to be the most important parameter to eradicate W 775 from compost. When the temperature of the compost heap is 60 degrees C and the moisture content varies between 60-65%, W 775 (10(8) CFU g(-1)) will be inactivated within 10 h of composting. The moisture content is, beside temperature, a second parameter that influences the survival of W 775. When the water content of the composting materials or meat carriers is reduced, a higher survival rate of W 775 was observed (survival rate increases 0.5 log(10) unit when there is a reduction of 5% in moisture content). In addition, other parameters (such as microbial antagonism, toxic compounds, etc.) have an influence on the survival of W 775 as well. CONCLUSIONS: Our study demonstrates that all types of composting processes tested in this work were sufficient to eradicate W 775 providing that they are well managed in terms of temperature and moisture content. AIMS: The objectives of this study were to determine the changes of microbial properties of pig manure collected from pens with different management strategies and composted using different turning and moisture regimes; relate their association with humification parameters and compost temperature; and identify the most suitable microbial indicators of compost maturity. METHODS AND RESULTS: Six different microbial parameters, including total bacterial count, oxygen consumption rate, ATP content, dehydrogenase activity, and microbial biomass C and N, along with humification parameters [humic acid (HA), fulvic acid (FA) and HA : FA ratio] and compost temperature were monitored during composting. Significant positive correlations were found between temperature and microbial properties, including O2 consumption rate, ATP content, dehydrogenase activity, and microbial biomass N. The humification parameters also showed significant correlations with microbial properties of the manure compost. For instance, HA contents of pig manures was positively correlated with total aerobic heterotrophs, and microbial biomass N and C; and negatively correlated with O2 consumption rate, ATP content, and dehydrogenase activity. Among the six microbial parameters examined, dehydrogenase activity was the most important factor affecting compost temperature and humification parameters. Composting strategies employed in this study affected the speed of composting and time of maturation. If the moisture content is maintained weekly at 60% with a 4-day turning frequency, the pig manure will reach maturity in 56 days. CONCLUSIONS: The composting process went through predictable changes in temperature, microbial properties and chemical components despite differences in the initial pig manure and composting strategies used. Among the six microbial parameters used, dehydrogenase activity is the most suitable indicator of compost maturity. Compared with respiration rate, ATP content and microbial biomass procedures, dehydrogenase activity is the simplest, quickest, and cheapest method that can be used to monitor the stability and maturity of composts. SIGNIFICANCE AND IMPACT OF THE STUDY: The results presented here show that microbial parameters can be used in revealing differences between composts and compost maturity. The statistical relationship established between humification parameters and microbial parameters, particularly dehydrogenase activity, demonstrates that it is possible to monitor the composting process more easily and rapidly by avoiding longer and more expensive analytical procedures.  相似文献

Various physical and chemical parameters were monitored to evaluate their influence on the microbial communities present in composting municipal sewage sludge. Temperature, moisture content, depth, pH, protein content, total nitrogen, total carbon, lipid phosphate biomass, and the rates of microbial incorporation of substrates into lipids were measured at several times throughout the 17- to 19-day composting runs. Temperature was found to have the most consistent and dramatic effect on microbial activity and biomass. When temperatures exceeded 55 to 60°C, microbial activity fell dramatically, usually by more than 1 order of magnitude. Microbial activity was generally greatest in samples taken from the 35 to 50°C areas of the composting piles. Changes in the composition of the compost over time included increased pH, increased protein content, and decreased total organic content. The changes in these parameters appeared to reflect the microbial activity and biomass present. The results of this study indicate that the rate of composting may best be optimized by controlling the composting temperatures, provided that the other parameters fall within reasonable limits in the starting material.  相似文献

水分是堆肥微生物生命活动的基础 ,也是堆肥中重要的工艺控制参数。弄清湿度对堆肥微生物及理化性质的影响 ,对于优化堆肥工艺参数、提高堆肥效率、降低投资和运行成本具有重要意义。综述了堆肥湿度研究的动态 ,指出了当前研究中存在的问题 ,并提出了未来的研究方向。大量的研究表明 ,湿度低于 4 5 %或高于 6 5 %都不利于堆肥处理。湿度太高会导致堆料的压实度增加、FAS减少、透气性能降低 ,从而导致堆体内氧气供应不足、堆肥升温困难、有机物降解速率降低、堆肥周期延长。湿度过低 ,水分会限制堆肥微生物的新陈代谢 ,导致微生物活性下降、堆肥腐熟困难。由于鼓风、散热、水蒸发等会使堆体内存在湿度的空间变异 ,也会降低堆肥效率和堆肥产品的质量。另外 ,堆肥湿度还影响堆肥的保肥能力。由各文献得出结论 ,堆肥的最佳湿度范围一般为 5 0 %~ 6 0 %左右  相似文献

The interactions between temperature and the microbial communities in composting municipal sewage sludge were studied to determine the optimal temperature range for efficient decomposition (stabilization) of the sludge. Information concerning thermophilic successions in such communities was also obtained. Samples were taken from several different temperature areas in a production-scale composting pile throughout the 19-day processing run. Optimum temperatures for microbial activity, determined as the rate of [14C]acetate incorporation into microbial lipids, were determined for each sample. Biomass was determined from the lipid phosphate content of the sample. Maximal activities were generally found in samples coming from lower-temperature areas (25 to 45 degrees C), whereas samples from high temperatures (55 to 74 degrees C) usually had relatively little activity. The temperature giving the optimum activity in samples incubated at a variety of temperatures during the assay tended to increase as the composting time progressed, but never exceeded about 50 degrees C. Many of these temperature response curves were similar in nature to curves reported for purified enzyme systems and pure cultures of bacteria. Comparisons of the apparent energies of activation calculated for different temperature ranges over time also indicated that the overall community was better adapted to higher temperatures during the latter part of the composting run. It was also found that the relationship between the apparent energies of activation and the apparent energies of inactivation (apparent heats of denaturation) consistently changed with sample temperature throughout the composting run, suggesting that the microbial communities from hotter samples were better adapted to high temperatures than those from cooler samples, and vice versa.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

Aims: The purpose of this study was to determine the effect of moisture on thermal inactivation of Salmonella spp. in poultry litter under optimal composting conditions. Methods and Results: Thermal inactivation of Salmonella was studied in fresh poultry compost by simulating early phase of composting process. A mixture of three Salmonella serotypes grown in Tryptic soy broth with rifampin (TSB‐R) was inoculated in fresh compost with 40 or 50% moisture at a final concentration of c. 7 log CFU g?1. The inoculated compost was kept in an environmental chamber which was programmed to rise from room temperature to target composting temperatures in 2 days. In poultry compost with optimal moisture content (50%), Salmonella spp. survived for 96, 72 and 24 h at 50, 55 and 60°C, respectively, as compared with 264, 144 and 72 h at 50, 55 and 60°C, respectively, in compost with suboptimal moisture (40%). Pathogen decline was faster during the come‐up time owing to higher ammonia volatilization. Conclusions: Our results demonstrated that Salmonella spp. survived longer in fresh poultry compost with suboptimal moisture of 40% than in compost with optimal moisture of 50% during thermophilic composting. High nitrogen content of the poultry compost is an additional factor contributing to Salmonella inactivation through ammonia volatilization during thermal exposure. Significance and Impact of the Study: This research validated the effectiveness of the current composting guidelines on Salmonella inactivation in fresh poultry compost. Both initial moisture level and ammonia volatilization are important factors affecting microbiological safety and quality of compost product.  相似文献

The aim of this research was to study the influence of lignin content and composting temperature on the biodegradation of lignin-containing pulp and paper products in a controlled composting test (European standard prEN 14046). Lignin reduced the biodegradation of the samples, and there was a linear correlation between the lignin content and the biodegradation of pulp and paper products at 58°C. The influence of incubation temperature (35, 50 and 58°C) on biodegradation was studied using bleached kraft paper containing 0.2 wt% lignin and mechanical pulp (stone-ground wood) containing 24–27 wt% lignin. Mechanical pulp biodegraded better at lower temperatures, while kraft paper biodegraded well at all three temperatures. Microbial activity was evaluated by measuring CO2 evolution and the change in ATP content, and fungal biomass by measuring the ergosterol content during the composting experiments. Kraft paper strongly increased microbial activity during the controlled composting test, but the activity returned to the background level at the end of the composting test. The proportion of sample carbon converted to microbial biomass carbon was considerably higher at lower incubation temperatures. Changes in microbial community structure during biodegradation of mechanical pulp and kraft paper at 50°C were studied by the PCR-based technique denaturing gradient gel electrophoresis. 以澳大利亚南昆士兰州典型森林类型——湿地松、南洋杉和贝壳杉林为对象,开展土壤可溶性有机碳和氮(SOC和SON)、微生物生物量碳和氮(MBC和MBN),以及土壤酶活性的研究,剖析森林类型对土壤质量的影响.结果表明:不同林型土壤SOC、SON含量分别在552 ~1154 mg·kg-1和20.11~57.32mg·kg-1;MBC、MBN分别在42~149 mg·kg-1和7~35 mg·kg-1.MBC、MBN之间呈显著相关.土壤几丁质酶、酸性磷酸酶、碱性磷酸酶和β-葡萄糖苷酶的活性分别为2.96 ~7.63、16.5 ~29.6、0.79 ~ 3.42和3.71 ~9.93 μg ·g-1·h-1,亮氨酸氨肽酶活性为0.18~0.46 μg·g-1·d-1.不同林型土壤SOC含量,以及土壤几丁质酶和亮氨酸氨肽酶活性为湿地松林、南洋杉林、贝壳杉林依次降低;而SON含量为南洋杉林>贝壳杉林>湿地松林,且南洋杉林的SON含量显著(P<0.05)高于湿地松林;MBC和MBN以及碱性磷酸酶活性为贝壳杉林>湿地松林>南洋杉林;酸性磷酸酶和β-葡萄糖苷酶活性为湿地松林>贝壳杉林>南洋杉林.在土壤生物代谢因子中,MBC、MBN、SON和亮氨酸氨肽酶对不同森林类型土壤影响较大.  相似文献

Pilot composting experiments of swine manure with corncob were conducted to evaluate the performance of the aerated static bin composting system. Effects of temperature control (60 and 70 degrees C) and moisture content (70% and 80%) were monitored on the composting by measuring physical and chemical indexes. The results showed that (1) the composting system could destroy pathogens, converted nitrogen from unstable ammonia to stable organic forms, and reduced the volume of waste; (2) significant difference of NH(4)(+)-N (P(12) = 0.074), and (NO(3)(-) + NO(2)(-))-N (P(12) = 0.085) was found between the temperature control treatments; (3) anaerobic reaction in the treatment with 80% moisture content resulted in significant difference of pH (P(23) = 0.006), total organic matter (P(23) = 0.003), and germination index (P(23) = 0.040) between 70% and 80%. Therefore, the optimum initial moisture content was less than 80% with the composting of swine manure and corncob by using the composting system.  相似文献

A three-strain mixture of Escherichia coli O157:H7 was inoculated into fresh dairy compost (ca. 10(7) CFU/g) with 40 or 50% moisture and was placed in an environmental chamber (ca. 70% humidity) that was programmed to ramp from room temperature to selected composting temperatures in 2 and 5 days to simulate the early composting phase. The surviving E. coli O157:H7 population was analyzed by direct plating and enrichment. Optimal and suboptimal compost mixes, with carbon/nitrogen (C/N) ratios of 25:1 and 16:1, respectively, were compared in this study. In the optimal compost mix, E. coli O157:H7 survived for 72, 48, and 24 h in compost with 40% moisture and for 72, 24, and 24 h with 50% moisture at 50, 55, and 60°C, respectively, following 2 days of come-up time (rate of heating up). However, in the suboptimal compost mix, the pathogen survived for 288, 72, and 48 h in compost with 40% moisture and for 240, 72, 24 h in compost with 50% moisture at the same temperatures, respectively. Pathogen survival was longer, with 5 days of come-up time compared with 2 days of come-up. Overall, E. coli O157:H7 was inactivated faster in the compost with 50% moisture than in the compost with 40% at 55 and 60°C. Both moisture and come-up time were significant factors affecting Weibull model parameters. Our results suggest that slow come-up time at the beginning of composting can extend pathogen survival during composting. Additionally, both the C/N ratio and the initial moisture level in the compost mix affect the rate of pathogen inactivation as well.  相似文献

In order to determine the conditions for the maximum performance of a fed-batch composting (FBC) reactor, polymerase chain reaction-denaturing gradient gel electrophoresis (PCR-DGGE) was used to analyze the microbial communities established under the confined conditions of moisture content and environmental temperature. To evaluate the effects of microbial community structures on the performance of FBC reactors, degradation experiments using small-scale reactors and model waste were conducted under confined environmental conditions. A high degradation rate was observed under a wide range of MC conditions (30-60%) and at higher than usual temperatures (30-50 degrees C). The microbial communities that formed in the experimental FBC reactors were analyzed by DGGE of PCR-amplified 16S rRNA genes. The DGGE banding patterns at the same level as the degradation rates were similar even if the environmental conditions were different. Sequence analysis of the DGGE bands revealed the primary microbes which act in the reactor.  相似文献

Large parts of the central highlands of Mexico are heavily eroded and the success of a planned reforestation program will greatly improve when the organic matter and nutrient content of the soil increases prior to the planting of the trees. This study investigated how the application of biosolids from a pharmaceutical company producing cephalosporines or third generation antibiotics could be used as a soil amendment and affect dynamics of C, P and N in soil. A sandy clay loam soil was sampled, amended with 24 g of dry biosolids kg(-1) dry soil or approximately 32 x 10(3) kg ha(-1) for the 0-10 cm layer, and incubated aerobically while production of carbon dioxide (CO(2)), dynamics of ammonium (NH(4)(+)),nitrite (NO(2)(-)), nitrate (NO(3)(-)), sodium bicarbonate (NaHCO(3)) extractable phosphorus (PO(4)(3-)), and microbial biomass carbon (C) were monitored. Results showed that the biosolid with pH 12, organic C content 162 g kg(-1), total N 21 g kg(-1), was of excellent quality considering its heavy metal content (USEPA) and a class "B" (USEPA) biosolid considering the amount of pathogens. No cephalosporines could be detected in the biosolid. Addition of biosolid to soil increased production of CO(2) 1.4 times and added >60 mg NH(4)(+) kg(-1). The application of biosolids did not significantly increase the concentration of NO(2)(-) which remained <2 mg N kg(-1) soil, but the concentration of NO(3)(-) did increase with 175 mg N kg(-1) soil. The microbial biomass C did not change when sewage biosolids was added and concentrations of extractable PO(4)(3-) only increased temporarily. Washing the biosolids reduced concentrations of NH(4)(+) and NO(3)(-), but also reduced pathogens and concentrations of chloride (Cl(-)), which might pose a treat to humans and the environment, respectively. Although the biosolid added valuable nutrients to the soil and did not inhibit C and N mineralization, further investigation into possible long-term environmental effects on soil processes and plant growth is necessary before this biosolid can be used in the field.  相似文献

Composting of fish processing wastes and biosolids with wood by-products and yard trimmings was conducted during the summer of 1996 and winter of 1997 in NW Patagonia using: (i) static piles for fish offal and (ii) turning piles for biosolids. Fish offal was mixed with sawdust + wood shavings (FOC) at 3:1 ratio by weight and biosolids with wood shavings (BCw) and yard trimmings (BCt) at 1:1 ratio by volume. Samples were taken at six dates during the composting period and analyzed to determine the factors that predict compost maturity. Composting of biosolids was affected by the type of bulking agent during winter. Thermophilic temperatures > or = 55 degrees C were sustained long enough to satisfy the USEPA requirements for processes to further reduce pathogens (PFRP) in FOC and BCt, and for processes to significantly reduce pathogens (PSRP) in summer BCw, while in winter BCw temperatures were lower than those recommended for effective pathogen reduction. However, coliform fecal content in all BC treatments was less than 10 most probable number (MPN) g(-1) dry sample at the end of the process. The ratio of water soluble carbon (WSC) to total nitrogen (TN) appeared to be a more adequate index to predict compost maturity than the ratio of total organic carbon to nitrogen.  相似文献

Investigations were carried out to find out the relationship between temperature and microbial activity in dairy cattle manure composting using oxygen uptake rate, specific growth rate and enzymatic activities during autothermal and isothermal composting experiments. In autothermal composting, oxygen uptake rate and specific growth rate were found to be most intensive in order of 43 degrees C, 60 degrees C and 54 degrees C. Isothermal composting at 54 degrees C resulted highest levels of enzymatic activity and promoted the volatile solids reduction. Based on the maximum enzymatic activity, specific growth rate appeared to be more closely linked with microbial activity in compost than with oxygen uptake rate. The enhancement of specific growth rate, enzymatic activity and volatile solids reduction were induced at 54 degrees C in cattle manure composting.  相似文献

In this study, microcosms were used to investigate the influence of temperature (4 and 28 degrees C) and water content (45% and 90% WHC) on microbial communities and activities in carbon-rich fen soil. Bacterial, archaeal and denitrifier community composition was assessed during incubation of microcosms for 12 weeks using terminal restriction fragment length polymorphism (T-RFLP) profiling of 16S rRNA and nitrous oxide reductase (nosZ) genes. In addition, microbial and denitrifier abundance, potential denitrification activity and production of greenhouse gases were measured. No detectable changes were observed in prokaryote or denitrifier abundance. In general, cumulatively after 12 weeks more carbon was respired at the higher temperature (3.7 mg CO(2) g(-1) soil), irrespective of the water content, whereas nitrous oxide production was greater under wet conditions (98-336 microg N(2)O g(-1) soil). After an initial lag phase, methane emissions (963 microg CH(4) g(-1) soil) were observed only under warm and wet conditions. T-RFLP analyses of bacterial 16S rRNA and nosZ genes revealed small or undetectable community changes in response to temperature and water content, suggesting that bacterial and denitrifying microbial communities are stable and do not respond significantly to seasonal changes in soil conditions. In contrast, archaeal microbial community structure was more dynamic and was strongly influenced by temperature.  相似文献

This study aimed to determine the aeration rate and its kinetics in aerobic composting of agricultural wastes. For this aim compost materials were prepared by mixing grass trimmings, tomato, pepper, and eggplant wastes. Four vertical forced aeration type reactors and one vertical natural convection type reactor were manufactured to apply four different aeration rates. CO2 rate and temperature changes were recorded in three different places in the reactors. Moisture content, pH and organic material rate were recorded each day. While process-monitoring parameters (CO2, temperature, pH, moisture content) were used for interpretation of the process, organic material degradation was used for interpretation of the process success. The seven different kinetic models were applied for modeling decomposition rate to the experimental values. According to the results, four of these models were found applicable to this study. These models were analyzed with some statistical methods as root mean square error (RMSE), chi-square (chi2), and modeling efficiency (EF). According to the statistical results of these models, the best model was found as: [Formula: see text] where kT is the rate of decomposition (g VS/g VS day); T the process temperature (degrees C); Mc the daily moisture content (%wb); C the daily CO2 rate in composting reactor (%) and a, b, c, d are constants. According to the results, the highest organic matter degradation and temperature value were obtained at the aeration rate of 0.4 l air min(-1)kg(om)(-1). The feasibility of high-rate composting of barley dregs and sewage sludge was examined using a pilot scale bioreactor. A central composite design (CCD) was used to optimize the mix ratio of barley dregs/sewage sludge and moisture content. The performance of the bioreactor was monitored as a function of carbon decomposition rate (CDR) and total volatile solids (TVS) loss rate. The optimum range of mix ratio and moisture content was found to be 35-40% and 55-60%, respectively. High CO2 evolution rate (CER) and TVS loss rate were observed after 3 days of the composting and the compost was matured/stable after 7 days. Cardinal temperature model with inflection (CTMI) was used to analyze the compost stability with respect to CER as a parameter of composting efficiency. After examining the phytotoxicity, the compost can be promoted for land application.  相似文献

以尖果沙枣1年生实生苗为材料,研究了自然干旱时不同土壤相对含水量对幼苗叶片细胞质膜相对透性、叶片相对含水量、根系活力、光合色素含量和光合参数等指标的影响.结果表明:土壤相对含水量从70%(CK)降到40%时,幼苗根系活力和净光合速率均逐渐上升并达到最大值,分别为1178μg.g-1.h-1和21.9μmol.m-2.s-1;光合色素含量稳步上升;蒸腾速率和水分利用效率均保持稳定;叶片细胞质膜相对透性保持较低水平.土壤相对含水量从40%降到20%时,幼苗叶片相对含水量仍在50%以上,叶片细胞质膜相对透性仍保持较低水平;根系活力和光合色素含量仍较高;但其他光合参数开始缓慢下降.土壤相对含水量从10%降到5%时,幼苗叶片细胞质膜相对透性急剧上升;叶片相对含水量、根系活力、总叶绿素含量、光合参数均极显著下降;而土壤相对含水量为10%时幼苗表现出最高的水分利用效率.尖果沙枣土壤相对含水量最好控制在40%~50%,其1年生实生苗的永久萎蔫系数为4.3%(土壤相对含水量).  相似文献
