Summary We report a study of a potassium-selective channel in the membrane delineating cytoplasmic drops fromChara australis. The relatively large conductance (170 pS in 150 mol/m3 (mm) KCl), high ion selectivity (P Cl/P K=0.015±0.01) and voltagedependent kinetics of this channel indicate that it is a type of maxi-K channel commonly found in animal cells but not previously detected in any plant cell.The current-voltage (I/V) characteristic of these channels was examined in drop-attached and in excised outside-out patches using the patch-clamp technique, over the unusually large voltage range of –250 to 200 mV. TheI/V characteristic is nonlinear and shows saturation at extreme voltages; the current also saturates at high [K+]. In solutions with symmetrical KCl concentrations the saturation behavior of the current is asymmetrical. The permeability of the channel depends on whether it is observed in excised or in drop-attached membrane patches.Here we investigate the main factors affecting the permeation of K+ ions through this maxi-K channel. We present the first direct evidence for the importance of diffusion external to the pore in limiting ion flow through maxi-K channels. The data are consistent with an ion translocation mechanism whose current is limited (i) at high voltages by ion diffusion external to the pore and (ii) at high [K+] by the maximum transport rate of the channel. We fit the data to a diffusion-limited pore model in which the pore exhibits saturation described by Michaelis-Menten kinetics with aK m=50±25 mol/m3 andG max=300±20 pS.  相似文献

Summary Internodal cells ofChara, grown in culture either at pH 5.7, 6.5 or 7.5, were studied to determine their chloride influx capability, the quantitative aspects of charasome morphology and the degree to which these two parameters could be correlated. In cells grown at pH 5.7 the charasomes were relatively small, were widely spaced on the plasma membrane, and contributed only a 0.6% increase to the surface area of the plasma membrane in the acid region of the cell. In contrast, the charasome membrane surface area of cells grown at pH 7.5 had increased × 19, the density of charasomes on the cell surface increased × 42, thus producing a × 3.57 increase in the acid region plasma membrane surface area. Chloride influx in cells grown at pH 7.5 was × 8.7–12.7 greater than in cells grown at pH 5.7. Cells that had been starved of chloride exhibited a × 2.4 average increase in the rate of chloride influx. Our observations establish the existence of a positive correlation between the rate of chloride influx and the increase in membrane surface area due to charasomes, although other factors, such as the effect of pH on transport-related enzymes, and the effect of charasome structure on chemical equilibria, may also be of importance.  相似文献

Summary Progress in patch-clamping the plasmalemma ofChara australis has been slow because of the difficulty in obtaining a clean membrane surface upon which obtain a high resistance seal with the patch pipette. A surgical method is described here for cutting an access window in the cell wall of internodal cells fromChara australis. The method has a high success rate and also has the advantage of obviating the need for enzyme treatments. The patch-clamp technique is applied to theChara plasmalemma. Five types of channel activity are observed of which four of these are selective for Cl over K+.  相似文献

Summary Previous current/voltage (I/V) investigations of theChara K+ state have been extended by increasing the voltage range (up to +200 mV) through blocking the action potential with La3+. A region of negative slope was found in theI/V characteristics at positive PD's, similar to that already observed at PD's more negative than the resting level. These decreases in membrane currents at PD's more negative than –150 mV and at PD's close to 0 or positive are thought to arise from the K+ channel closure. Both the negative slope regions could be reversibly abolished by 0.1mm K+, 20mm Na+, more than 10mm Ca2+ or 5mm tetraethylammonium (TEA). The K+ channels are therefore blocked by TEA, closed by low [K+] o or high [Ca2+] o and are highly selective to K+ over Na+. With the K+ channels closed, the remainingI/V profile was approximately linear over the interval of 400 mV (suggesting a leakage current), but large rectifying currents were observed at PD's more positive than +50 mV. These currents showed a substantial decrease in high [Ca2+] o , sometimes displayed a slight shift to more positive PD's with increasing [K+] o and were unaffected by TEA or changes in [Na+] o . The slope of the linear part of theI/V profile was steeper in low [K+] o than in TEA or high [Na+] o (indicating participation of K+, but not Na+, in the leak current). Diethylstilbestrol (DES) was employed to inhibit the proton pump, but it was found that the leakage current and later the K+ channels were also strongly affected.  相似文献

Summary An electrogenic K+–Na+ symport with a high affinity for K+ has been found inChara (Smith & Walker, 1989). Under voltage-clamp conditions, the symport shows up as a change in membrane current upon adding either K+ or Na+ to the bathing medium in the presence of the other. Estimation of kinetic parameters for this transport has been difficult when using intact cells, since K+–Na+ current changes show a rapid falling off with time at K+ concentrations above 50 m. Cytoplasm-enriched cell fragments are used to overcome this difficulty since they do not show the rapid falling off of current change seen with intact cells. Current-voltage curves for the membrane in the absence or presence of either K+ or Na+ are obtained, yielding difference current-voltage curves which isolate the symport currents from other transport processes. The kinetic parameters describing this transport are found to be voltage dependent, withK m for K+ ranging from 30 down to 2 m as membrane potential varies from –140 to –400 mV, andK m for Na+ ranging between 470 and 700 m over a membrane potential range of –140 to –310 mV.Two different models for this transport system have been investigated. One of these involves the simultaneous transport of both the driver and substrate ions across the membrane, while the other allows for the possibility of the two ions being transported consecutively in two distinct reaction steps. The experimental results are shown to be consistent with either of these cotransport models, but they do suggest that binding of K+ occurs before that of Na+, and that movement of charge across the membrane (the voltage-dependent step) occurs when the transport protein has neither K+ nor Na+ bound to it.  相似文献

Effects of salinity and cutting on the development of Phragmites australis   总被引:1,自引:0,他引:1  
The effects of increased salinity and cutting the above ground biomass on the growth of Phragmites australis were evaluated by investigating four experimental reed stands grown in outdoor tanks. Two stands were treated with 30 salinity and the other two stands with freshwater; one stand of each treatment was cut to 20 cm during the second growing season. Growth conditions were observed until all the plants were dead at the end of the second year. The number of shoots emerged from the freshwater-treated stand was about 70% higher than that of the saltwater-treated stand. The number of shoots emerged from cut plant stands were markedly lower than uncut stands. The average shoot height was negatively affected by salinity and shoots that emerged after cutting further decreased in height. The average number of leaves on a shoot was not significantly affected by salinity, but reduced by cutting in both treatments. Leaf length, width and the distance between leaves were decreased by both salinity and cutting. In the freshwater-treated uncut stand more than 50% of the shoots formed panicles, but this proportion was reduced to 6% by salinity, to 15% by cutting, and to 0% by the combination of salinity and cutting. This study showed again that salinity reduces the growth of aboveground components. The growth, however, was most severely retarded by cutting combined with salinity, which has many implications for better management of P. australis stands.  相似文献

Summary Cytoplasmic drops, covered by a membrane derived from the tonoplast, were obtained from the internodal cells ofChara australis. Patch-clamp measurements were made on this membrane using the droplet-attached configuration with the membrane patch voltage clamped at values from –250 to 50 mV. Single-channel records, filtered at 5 kHz, were analyzed to elucidate the kinetics of the ion gating reaction of the K+-selective channel. The current-voltage characteristics for single channels exhibit saturation and are shown to be consistent with Läuger's theory of diffusion-limited ion flow through pores (P. Läuger,Biochim. Biophys. Acta 455:493–509, 1976). The time-averaged behavior of the K+ conductance has a maximum at –100 to –150 mV which is produced by the combination of two distinct mechanisms: (1) The channel spending more time in long-lived closed states at positive voltages and (2) a large decrease in the mean open lifetime at more negative voltages. The channel activity shows bursting behavior with opening and closing rates that are voltage-dependent. The mean open time is the kinetic parameter most sensitive to membrane potential, showing a maximum between –100 to –150 mV. The distribution of open times is dominated by one exponential component (time constant 0.3 to 10 msec). In some cases an additional rapidly decaying exponential component was detectable (time constant=0.1 msec). The closed distributions contained were observed to obtain up to four exponential components with time constants over the range 0.1 to 200 msec. However, the voltage dependence of the closed-time distributions suggests an eight-state model for this channel.  相似文献

The change in dried rhizome samples that were left to decompose was investigated to elucidate the effects of rhizome age on the decomposition rate of Phragmites australis. Rhizomes were classified into five age categories and placed 30 cm below the soil surface of a reed stand. After 369 days of decay, new (i.e., aged less than one year) rhizomes had lost 84% of their original dry mass, compared with a loss of 41–62% for that of older rhizomes. The exponential decay rates of older rhizomes were nearly identical to that of aboveground biomass. The nitrogen (N) concentration increased to two times its original values, but the phosphorus (P) concentration remained constant after an initial loss by leaching. The carbon to nitrogen (C:N) and carbon to phosphorus ratios (C:P) leveled out at 22:1 to 38:1 and 828:1 to 1431:1, respectively, regardless of rhizome age. The results are important to understand the nutrient cycles of reed-dominant marsh ecosystems.  相似文献

Seed-production ability was studied in Utricularia australis R. Br. f. australis Komiya and Shibata through field observation, pollination experiment, pollen culture, and isozyme analysis for populations in various regions of Hokkaido, northern Japan. Utricularia australis f. australis has previously been postulated to be sterile in Japan, however, in the present study, fertile populations were found in eastern Hokkaido. Bisexual sterility and male sterility were suspected to be present in populations in the western part of Hokkaido. There was also a strain that seemed to produce no seed because of intra-clonal inbreeding depression or self-incompatibility. Isozyme analysis of three enzyme systems indicated a uniformity of multienzymatic phenotype (MEP) in each population or region. The MEP grouping of populations corresponded to the type of sterility and fertility revealed by pollination and pollen culture experiments. These experiments suggested that each regional strain had its own sterility or fertility type.  相似文献

The influence of zinc (Zn) on physiological and biochemical parameters was studied to elucidate the mechanism of Zn resistance in Phragmites australis. Zn concentrations in roots, stems and leaves increased with exogenous Zn concentration, while Zn content in roots was much higher than in shoots. X-ray microanalysis was used to reveal compartments in which Zn accumulated in root cortex. Zinc concentrations followed a gradient with the sequence: intercellular space>cell wall >vacuole >cytoplasm, indicating that most Zn was immobilized in the apoplast or sequestered into the vacuolar lumen. Sequential extraction of various Zn chelates revealed that the ratio of Zn extracted with different extraction media was markedly different. Ethanol, HAc (acetic acid) and NaCl-extractable Zn were dominant in both roots and leaves of P. australis. Zn-binding protein fractions were found in the roots and leaves after gel filtration chromatography, among which a polypeptide with an apparent molecular mass of 14kDa bound Zn most effectively. Two newly synthesized polypeptides of 58 and 45kDa appeared under Zn pollution, whereas a prominent fraction of 72kDa disappeared. The involvement of Zn distribution in plant tissues, subcellular compartments and chelates and Zn-inducing proteins in the acclimation mechanism of P. australis to Zn pollution is discussed.  相似文献

Efforts to manage or prevent Phragmites australis invasion in salt and brackish marshes are complicated by the lack of a general causal role for specific human activities. The pattern of invasion within a marsh differs among sites, and each may have different causal histories. A review of the literature finds three establishment/invasion patterns: (1) from stands established on ditch- or creek-bank levees toward interior portions of high marshes, (2) from stands along upland borders toward high marsh interiors, and (3) centroid spread from high marsh stands established in ostensibly random locations. Each invasion pattern seems to have different anthropogenic precursors, therefore preventing generalizations about the role of any one human activity in all sites. However, historical and experimental evidence suggests that regardless of invasion pattern, establishment is much more likely at sites where rhizomes are buried in well-drained, low salinity marsh areas. Any human activity that buries large rhizomes, increases drainage, or lowers salinity increases chances of establishing invasive clones. To integrate these patterns and improve our understanding of the rapid spread of Phragmites, recent evidence has been synthesized into a dichotomous flow chart which poses questions about current site conditions and the potential for proposed activities to change site conditions that may facilitate invasion. This simple framework could help managers assess susceptibility and take preventative measures in coastal marshes before invasion occurs or before removal becomes very expensive.  相似文献

Trichiurus australis, a new trichiurid fish, is described on the basis of four specimens collected off Burnett River mouth, Pialba and Thursday Island, Queensland, Australia. The new species strongly resembles Trichiurus brevis Wang and You, 1992 (distributed in the South China Sea), T. russelli Dutt and Thankam, 1967 (the northwestern Bay of Bengal and the northern Gulf of Thailand), and T. nickolensis Burhanuddin and Iwatsuki, 2003 (northwestern Australia) in having the highest point of the supraoccipital crest situated directly above the posterior margin of the eye and being relatively small in size (less than ca. 700mm in total length: TL). It differs from those three species in having spinescent gill rakers almost with 2 equally long cusps [vs. (1, rarely 2 but 1 of them clearly shorter than another)], posterior caudal peduncle vertebrae bearing neural spines (vs. neural spines absent), longer caudal peduncle length (mean 16% TL vs. 6% in T. brevis, 8% in T. nickolensis, and 8% in T. russelli), and shorter precaudal length (83% vs. 93%, 91%, and 91%), preanal length (26% vs. 33%, 31%, and 35%), and head length (9% vs. 12%, 11%, and 13%). We have tentatively classified this new species under the Trichiurus russelli complex because the foregoing diagnostic characters are identical to this species complex except for the presence of neural spines in the posterior caudal vertebrae.  相似文献

The effects of copper on the growth, tolerance indices, mineral composition (N, P, K, Fe, Zn and Mn) and metal uptake of reed (Phragmites australis [Cav. Trin. ex Steudel]) and maize (Zea mays L.) were investigated in hydroponic experiments at copper concentrations ranging from 0.5 to 157 M Cu. A reduction in root length was shown to be a good indicator of copper toxicity, concentrations of 15.7 and 78.7 M Cu inhibiting root growth in maize and reed, respectively. The reed was significantly more tolerant of copper than maize and at 7.85 M Cu (external concentration), reed can be described as a Cu tolerant plant, and maize as a Cu non-tolerant species. As a result of Cu toxicity, the concentrations of macronutrients N, P and K decreased in both shoot and root of maize, while the concentrations were hardly affected in reed tissues. Fe concentration increased in shoots and roots of maize and in roots of reed with increasing Cu treatments, leading to highly significant (p<0.01) linear relationships between tissue Fe and Cu concentrations. The bioconcentration factor (BCF) of Cu was higher in roots than in shoots of both plant species, ranging from 612 to 1592 in reed for the Cu treatments tested. In the roots of maize, BCF of Cu increased from 349 to 1931 when increasing Cu in nutrient solution from 7.85 M to 78.5 M. Therefore, reed could be useful in wastewater treatments for the removal of Cu. However, the use of reed in phytoextraction of Cu from contaminated soils is limited by the low accumulation rate in shoots and although reed can be more efficient than maize for Cu phytoextraction, harvesting the full biomass, including roots, may be required.  相似文献

Summary Brown kiwis are an endangered species of nocturnal, flightless birds which are native to New Zealand. The resting focus of two specimens has previously been studied by retinoscopy in a zoo while the birds were restrained by their keeper (Sivak and Howland 1987). Those birds appeared to be hyperopic (farsighted) by 2–7 D. In this study, examination with infrared photorefraction of the focusing of two unrestrained, feeding birds showed that they could focus objects at infinity and objects in their immediate environment and that they had modest powers of accommodation. Measurements on two 6 month old kiwi chicks showed their corneal radius of curvature to be between 2.90 and 3.00 mm (117 D and 101 D in power).  相似文献

Phragmites australis (the common reed) was collected at six sites in southern Québec and Ontario, Canada, in order to study the accumulation of iron plaque on the roots. The deposition of iron oxides on roots ofP. australis did not correlate directly with soil measurements; however, the amounts of iron-bound-to-carbonates fraction of the soil/sediment, responsible for the iron plaque accumulation, correlated with the % of water, % of organic matter, % of clay and pH of the substrate. Plants located very near flowing water accumulated more iron plaque on the roots than plants in other habitats through the summer; it is hypothesized that carbonates associated with iron come from the flowing water. In wetlands or sites near flowing water, most root iron was found on the surface, as iron plaque, while there was more iron inside the root in dry environments. Radial oxygen loss from the roots is probably the most important source of oxygen for the oxidation of iron.  相似文献

Performance of young Phragmites australis plants was examined after 7 weeks on an artificial nutrient-enriched inorganic substrate and on the same substrate to which an organic sediment from a eutrophic lake was added, at three different water depth treatments. Growth decreased, and proportional allocation of biomass to roots increased, with the addition of sediment. These differences were significant in shallow and deep water, but not at a medium depth. Concentrations of phosphorus and nitrogen in plant biomass decreased, and concentration of iron increased, with addition of sediment.The effects of sediment addition may have resulted from a decreased availability of nutrients in the substrate or from an impaired root functioning. Nutrient exhaustion in the substrate, due to a fast plant growth, can explain the relatively strong effects in shallow water. Deep water, on the other hand, probably restricted oxygen transport to the roots, resulting in an impaired root functioning in the low-redox sediment environment. The results show that, especially in relatively deep water, growth of undisturbed plants of P. australis may be inhibited by eutrophication of sediments, probably because of an impaired root functioning in sediments containing reduced toxic compounds (e.g. ferrous iron).  相似文献

Variations in the height, shoot density, biomasses of above- and below-ground parts and rhizome distributions ofPhragmites australis were investigated along a line-transect in a reed community at Yufutsu Mire, Hokkaido. Relationships of performance of the reed plants to soil conditions and species compositions were also examined. Three types of rhizome development were recognized in reed plants; (1) the central part of the reed community, characterized by well developed rhizomes and dense aerial shoots, (2) the intermediate part, characterized by development of rhizomes along both the peat and surface layers and very dense aerial shoots, and (3) the marginal part, characterized by development of rhizomes only along the peat layer and sparse aerial shoots. Observation showed that rhizomes in the surface layer actively produced aerial shoots, whereas rhizomes in the peat layer contributed to the spreading of their distribution range. With the growth of rhizomes, organic debris originating from dead rhizomes and roots accumulated in the mineral soil to promote organic soil formation. In dense parts of the reed stand, species composition was poor because of the shading and litter accumulation by reed plants. The effects of microtopography and water level on the establishment of reed seedlings were also considered.  相似文献

During the summer of 2002, symptoms of rust disease were observed for the first time on Phragmites australis in Saudi Arabia. Light brown lesions of regular shape indicating uredinia of Puccinia isiacae appeared on the leaves. The morphology and characteristics of the fungus were described in detail with both light and scanning electron microscopy. The possibility of using this fungus as a biological control agent was also discussed.  相似文献

Summary Reducing the pH of the bathing solution from 8.2 to pH 6 can induce an inversion of the extracellular current pattern that develops at the surface ofChara corallina internodal cells. A similar result can be obtained on some cells by changing the medium to a pH value of 10. In noninvertingChara cells the currents were strongly reduced when the pH value of the medium was changed between 3 and 11. Simultaneous measurements of theChara transmembrane potential in the acid and alkaline regions revealed that a light-induced electrical potential gradient of approximately 24 mV was present in the axial (or longitudinal) direction. Correlated to the external current pattern inversion was an inversion of this internal longitudinal voltage gradient. Reillumination ofNitella cells, after a period of darkness, often resulted in a complete inversion of the extracellular current pattern. These results are discussed in terms of spatial and temporal control of membrane transport processes, and in particular the control of current loops that pass through these cells.  相似文献
