Krill (Euphausiids) play a vital ecosystem role in many of the world’s most productive marine regions, providing an important trophic linkage. We introduce a robust modeling approach to link Cassin’s auklet (Ptychoramphus aleuticus) abundance and distribution to large-scale and local oceanic and atmospheric conditions and relate these patterns to similarly modeled distributions of an important prey resource, krill. We carried out at-sea strip transect bird surveys and hydroacoustic assessments of euphausiids (2004–2013). Data informed separate, spatially-explicit predictive models of Cassin’s auklet abundance (zero-inflated negative binomial regression) and krill biomass (two-part model) based on these surveys. We established the type of prey responsible for acoustic backscatter by conducting net tows of the upper 50 m during surveys. We determined the types of prey fed to Cassin’s auklet chicks by collecting diet samples from provisioning adults. Using time-depth-recorders, we found Cassin’s auklets utilized consistent areas in the upper water column, less than 30 m, where krill could be found (99.5% of dives were less than 30 m). Birds primarily preyed upon two species of euphausiids, Euphausia pacifica and Thysanoessa spinifera, which were available in the upper water column. Cassin’s auklet abundance was best predicted by both large scale and localized oceanic processes (upwelling) while krill biomass was best predicted by local factors (temperature, salinity, and fluorescence) and both large scale and localized oceanic processes (upwelling). Models predicted varying krill and bird distribution by month and year. Our work informs the use of Cassin’s auklet as a valuable indicator or krill abundance and distribution and strengthens our understanding of the link between Cassin’s auklet and its primary prey. We expect future increases in frequency and magnitude of anomalous ocean conditions will result in decreased availability of krill leading to declines in the Farallon Islands population of Cassin’s auklets.  相似文献

Nuclear factor-kappaB (NF-κB) is critical for the expression of multiple genes involved in inflammatory responses and cellular survival. NF-κB is normally sequestered in the cytoplasm through interaction with an inhibitor of NF-κB (IκB), but inflammatory stimulation induces proteasomal degradation of IκB, followed by NF-κB nuclear translocation. The degradation of IκB is mediated by a SCF (Skp1-Cullin1-F-box protein)-type ubiquitin ligase complex that is post-translationaly modified by a ubiquitin-like molecule Nedd8. In this study, we report that BRCA1-associated protein 2 (Brap2) is a novel Nedd8-binding protein that interacts with SCF complex, and is involved in NF-κB translocation following TNF-α stimulation. We also found a putative neddylation site in Brap2 associated with NF-κB activity. Our findings suggest that Brap2 is a novel modulator that associates with SCF complex and controls TNF-α-induced NF-κB nuclear translocation.  相似文献

Although reserpine and chlorpromazine had tranquilizing effect on a number of geriatric and alcoholic patients in a state hospital, several complicating results were noted, some with a bearing on the patients'' health and others that might affect the assignment of personnel.Lipoprotein studies carried out on patients receiving reserpine seemed to indicate that a reduction in the blood levels of the denser lipoprotein molecules took place during therapy.Several elderly patients receiving chlorpromazine died of diseases that were not as sharply manifest by symptoms as they might be expected to be. Hence the need for closer observation than a limited staff could afford seemed a matter for consideration. Another consideration of the same order was the possible need for increased personnel for psychotherapy in light of the more receptive condition of the patients.  相似文献

With the astonishing rate that genomic and metagenomic sequence data sets are accumulating, there are many reasons to constrain the data analyses. One approach to such constrained analyses is to focus on select subsets of gene families that are particularly well suited for the tasks at hand. Such gene families have generally been referred to as “marker” genes. We are particularly interested in identifying and using such marker genes for phylogenetic and phylogeny-driven ecological studies of microbes and their communities (e.g., construction of species trees, phylogenetic based assignment of metagenomic sequence reads to taxonomic groups, phylogeny-based assessment of alpha- and beta-diversity of microbial communities from metagenomic data). We therefore refer to these as PhyEco (for phylogenetic and phylogenetic ecology) markers. The dual use of these PhyEco markers means that we needed to develop and apply a set of somewhat novel criteria for identification of the best candidates for such markers. The criteria we focused on included universality across the taxa of interest, ability to be used to produce robust phylogenetic trees that reflect as much as possible the evolution of the species from which the genes come, and low variation in copy number across taxa.We describe here an automated protocol for identifying potential PhyEco markers from a set of complete genome sequences. The protocol combines rapid searching, clustering and phylogenetic tree building algorithms to generate protein families that meet the criteria listed above. We report here the identification of PhyEco markers for different taxonomic levels including 40 for “all bacteria and archaea”, 114 for “all bacteria (greatly expanding on the ∼30 commonly used), and 100 s to 1000 s for some of the individual phyla of bacteria. This new list of PhyEco markers should allow much more detailed automated phylogenetic and phylogenetic ecology analyses of these groups than possible previously.  相似文献

The endo-beta-1, 3-glucanase (beta-1, 3-glucan 3-glucanhydrolase, EC 3.2.1.6) extracted from Phaseolus vulgaris L. cv. Red Kidney had a pH optimum of 5 and a temperature optimum of 50 C. Excision of plant tissue resulted in an increase in beta-1, 3-glucanase activity after a 6-hour lag period. The increase could be prevented by indole-3-acetic acid, gibberellic acid, and cytokinins. Ethylene (half-maximal concentration = 0.1 microliter/liter) promoted the synthesis of beta-1, 3-glucanase, and 10% CO(2) overcame some of the ethylene effect. Cycloheximide prevented the induction of beta-1, 3-glucanase, but actinomycin D and chromomycin A(3) had only a partial effect.The amount of callose in sieve tube cells correlated with levels of beta-1, 3-glucanase, suggesting that this enzyme played a role in the degradation of beta-1, 3-glucans.  相似文献

The chytrid fungus Batrachochytrium dendrobatidis (Bd) has been devastating amphibians globally. Two general scenarios have been proposed for the nature and spread of this pathogen: Bd is an epidemic, spreading as a wave and wiping out individuals, populations, and species in its path; and Bd is endemic, widespread throughout many geographic regions on every continent except Antarctica. To explore these hypotheses, we conducted a transcontinental transect of United States Department of Defense (DoD) installations along U.S. Highway 66 from California to central Illinois, and continuing eastward to the Atlantic Seaboard along U.S. Interstate 64 (in sum from Marine Corps Base Camp Pendleton in California to Naval Air Station Oceana in Virginia). We addressed the following questions: 1) Does Bd occur in amphibian populations on protected DoD environments? 2) Is there a temporal pattern to the presence of Bd? 3) Is there a spatial pattern to the presence of Bd? and 4) In these limited human-traffic areas, is Bd acting as an epidemic (i.e., with evidence of recent introduction and/or die-offs due to chytridiomycosis), or as an endemic (present without clinical signs of disease)? Bd was detected on 13 of the 15 bases sampled. Samples from 30 amphibian species were collected (10% of known United States'' species); half (15) tested Bd positive. There was a strong temporal (seasonal) component; in total, 78.5% of all positive samples came in the first (spring/early-summer) sampling period. There was also a strong spatial component—the eleven temperate DoD installations had higher prevalences of Bd infection (20.8%) than the four arid (<60 mm annual precipitation) bases (8.5%). These data support the conclusion that Bd is now widespread, and promote the idea that Bd can today be considered endemic across much of North America, extending from coast-to-coast, with the exception of remote pockets of naïve populations.  相似文献

Arbuscular mycorrhizas, associations between plant roots and soil fungi, are ubiquitous among land plants. Arbuscular mycorrhizas can be beneficial for plants by overcoming limitations in nutrient supply. Hyphae, which are long and thin fungal filaments extending from the root surface into the soil, increase the volume of soil accessible for plant nutrient uptake. However, no models so far specifically consider individual hyphae. We developed a mathematical model for nutrient uptake by individual fungal hyphae in order to assess suitable temporal and spatial scales for a new experimental design where fungal uptake parameters are measured on the single hyphal scale. The model was developed based on the conservation of nutrients in an artificial cylindrical soil pore (capillary tube) with adsorbing wall, and analysed based on parameter estimation and non-dimensionalisation. An approximate analytical solution was derived using matched asymptotic expansion. Results show that nutrient influx into a hypha from a small capillary tube is characterized by three phases: Firstly, uptake rapidly decreases as the hypha takes up nutrients, secondly, the depletion zone reaches the capillary wall and thus uptake is sustained by desorption of nutrients from the capillary wall, and finally, uptake goes to zero after nutrients held on the capillary wall have been completely depleted. Simulating different parameter regimes resulted in recommending the use of capillaries filled with hydrogel instead of water in order to design an experiment operating over measurable time scales.  相似文献

Freshwater fishes that have been isolated by artificial dams have become models for studying the effects of recent barriers on genetic variation and population differentiation. In this study, we examined the genetic structure of 11 populations of white-spotted charr (Salvelinus leucomaenis) by using polymorphic microsatellite loci. Reduced genetic diversity, expressed as the number of alleles and the expected heterozygosity, was observed in all above-dam relative to below-dam populations. Highly significant genetic differentiation (F ST) was found for all pairwise comparisons among populations, with F ST-values ranging from 0.023 to 0.639. Both multiple regression analysis and a randomization test revealed that genetic differentiation above and below dams was negatively related to the habitat size of above-dam populations, and was positively related to the time period of isolation. This study is one of the few attempts to predict the population genetic structure of such variable spatial–temporal scales. We conclude that differences in genetic structure above and below dams are related to recent historical population size, whereby sites with a lower effective number of adults are more prone to temporal stochasticity in gene frequencies.  相似文献

Osteoclasts are bone-resorbing cells that are critical for the normal formation and maintenance of teeth and skeleton. Osteoclast deficiency can contribute to heterotopic ossification (HO), a pathology that is particularly detrimental to the mechanical functions of joints, valves and blood vessels. On the other hand, osteoclast over-activity is a major cause of osteoporosis. A reliable method for controlled generation of osteoclasts would be useful as a potential autologous cell therapy for HO, as well as high-throughput drug screening for anti-osteoporotic drugs. In this report, we describe the development of a cell engineering approach to control monocytic precursor cell differentiation to osteoclasts. Oligomerization of receptor activator of nuclear factor κB (RANK) is known to be essential for osteoclast differentiation from monocyte/macrophage precursors. We engineered a murine monocytic cell line, RAW264.7 to express a fusion protein comprising the intracellular RANK signaling domain and FK506-derived dimerization domains that bind to a small molecule chemical inducer of dimerization (CID). Virally infected cells expressing this fusion protein were treated with CID and dose-dependent induction of tartrate-resistant acid phosphatase activity, as well as multinucleated osteoclast formation were observed. Furthermore, NF-κB signaling was upregulated in a CID-dependent fashion, demonstrating effective RANK intracellular signaling. Functionally CID-induced osteoclasts had robust mineral resorptive activity in both two-dimensional and three-dimensional in vitro resorption assays. In addition, the CID-induced osteoclasts have the same life span as native RANKL-induced osteoclasts. Most importantly and crucially, the engineered cells differentiated into osteoclasts that were resistant to the potent osteoclast inhibitor, osteoprotegerin. Taken together, these studies are the first to describe a method for inducible control of monocytic precursor differentiation to osteoclasts that may be useful for future development of an engineered autologous cell therapy as well as high-throughput drug testing systems to treat diseases of osteoclast over-activity that are independent of osteoprotegerin.  相似文献

This study aims to examine the spatio-temporal dynamics of Rocky Mountain spotted fever (RMSF) prevalence in four contiguous states of Midwestern United States, and to determine the impact of environmental and socio–economic factors associated with this disease. Bayesian hierarchical models were used to quantify space and time only trends and spatio–temporal interaction effect in the case reports submitted to the state health departments in the region. Various socio–economic, environmental and climatic covariates screened a priori in a bivariate procedure were added to a main–effects Bayesian model in progressive steps to evaluate important drivers of RMSF space-time patterns in the region. Our results show a steady increase in RMSF incidence over the study period to newer geographic areas, and the posterior probabilities of county-specific trends indicate clustering of high risk counties in the central and southern parts of the study region. At the spatial scale of a county, the prevalence levels of RMSF is influenced by poverty status, average relative humidity, and average land surface temperature (>35°C) in the region, and the relevance of these factors in the context of climate–change impacts on tick–borne diseases are discussed.  相似文献

Abstract: We analyzed moose (Alces alces)-vehicle collisions (MVCs) in western Maine, USA, from 1992 to 2005 (n = 8,156) using Geographic Information Systems to identify patterns of temporal and spatial distribution and develop predictive models based on road and landscape characteristics. We used chi-square and correlation analyses to assess temporal characteristics of MVCs, K-function and kernel analyses to identify spatial clusters of MVCs, and logistic regression to relate covariates for traffic, land-cover, land-form, and relative moose abundance to probability of MVC. We evaluated candidate models using Akaike's Information Criterion, area under the receiver operating characteristic curve (AUC), and the percentage of correctly classified observations. Most (81.6%) MVCs occurred from May to October, with peak monthly frequencies in June (18.6%). Moose-vehicle collisions were clustered spatially on roads at local (0–4 km) and regional scales (22–41 km and 45–54 km), but not at intermediate scales. Traffic-related covariates predicting MVCs included traffic volume and speed limit. For each additional 500 vehicles/day, odds of a location being an MVC increased by 57%. For each 8-km/hr increase in speed limit, odds of an MVC increased by 35%. Landscape composition covariates best predicted MVCs within a 2.5-km radius of the collision site. Mean percent cover within 2.5 km of MVCs was comprised of 36% more cutover forest, 10% more coniferous forest, 5% less deciduous-mixed forest, and 10% less nonwoody wetland than for random points. For every 5% increase in percent cutover and coniferous forest within 2.5 km of the road, predicted odds of MVC increased by 36% and 19%, respectively. Landscape configuration covariates best predicted MVCs within the 5.0-km radius. Moose-vehicle collisions were associated with areas of less interspersion of cover types; for each 5% increase in an index of interspersion-juxtaposition, predicted odds of MVC decreased by 11%. Our final model attained high predictive power (AUC = 0.835) and validation accuracy (75.0%). The model also proved robust to physiographic variation, exhibiting high predictive power (AUC = 0.828) and validation accuracy (68.8%). Managers seeking to prioritize resources for reducing MVCs or predicting future areas of high MVC probability should assess land-cover composition and configuration surrounding MVC hotspots at geographic extents out to 2.5–5 km and use this information to plan expensive roadside management practices such as fencing. The importance of traffic and landscape covariates in our modeling suggests that effective management to reduce MVCs will require a complex combination of driving speed reductions and modifications to forest management along roads.  相似文献

Growing evidence indicates that the melanocortin 1 receptor (MC1R) and its ligand α–melanocyte-stimulating hormone (α-MSH) have other functions in the skin in addition to pigment production. Activation of the MC1R/α-MSH signaling pathway has been implicated in the regulation of both inflammation and extracellular matrix homeostasis. However, little is known about the role of MC1R/α-MSH signaling in the regulation of inflammatory and fibroproliferative responses to cutaneous injury. Although MC1R and α-MSH localization has been described in uninjured skin, their spatial and temporal expression during cutaneous wound repair has not been investigated. In this study, the authors report the localization of MC1R and α-MSH in murine cutaneous wounds, human acute burns, and hypertrophic scars. During murine wound repair, MC1R and α-MSH were detected in inflammatory cells and suprabasal keratinocytes at the leading edge of the migrating epithelial tongue. MC1R and α-MSH protein levels were upregulated in human burn wounds and hypertrophic scars compared to uninjured human skin, where receptor and ligand were absent. In burn wounds and hypertrophic scars, MC1R and α-MSH localized to epidermal keratinocytes and dermal fibroblasts. This spatiotemporal localization of MC1R and α-MSH in cutaneous wounds warrants future investigation into the role of MC1R/α-MSH signaling in the inflammatory and fibroproliferative responses to cutaneous injury.
