In vitro fertilization (IVF) systems using isolated male and female gametes have been utilized to dissect fertilization-induced events in angiosperms, such as egg activation, zygote development and early embryogenesis, as the female gametophytes of plants are deeply embedded within ovaries. In this study, a rice IVF system was established to take advantage of the abundant resources stemming from rice research for investigations into the mechanisms of fertilization and early embryogenesis. Fusion of gametes was performed using a modified electrofusion method, and the fusion product, a zygote, formed cell wall and an additional nucleolus. The zygote divided into a two-celled embryo 15–24 h after fusion, and developed into a globular-like embryo consisting of an average of 15–16 cells by 48 h after fusion. Comparison of the developmental processes of zygotes produced by IVF with those of zygotes generated in planta suggested that zygotes produced by IVF develop and grow into early globular stage embryos in a highly similar manner to those in planta. Although the IVF-produced globular embryos did not develop into late globular-stage or differentiated embryos, but into irregularly shaped cell masses, fertile plants were regenerated from the cell masses and the seeds harvested from these plants germinated normally. The rice IVF system reported here will be a powerful tool for studying the molecular mechanisms involved in the early embryogenesis of angiosperms and for making new cultivars.  相似文献

Summary A study was made of the cytogenetic effect of mutagens with a delayed effect (ethylenimine and ethyl methanesulphonate) on Crepis capillaris seeds. The effect was found to depend on the physiological condition of the seeds. In seeds not subjected to prolonged storage, where only chromatid aberrations were occurring spontaneously, mutagens also induced chromatid aberrations only. If, however, because of physiological changes in the seeds (e.g. upon prolonged storage or when seeds were kept at an elevated temperature and humidity) a large number of chromosome-type aberrations appeared, they also appeared when the seeds were acted upon by mutagens with a delayed effect. The action of such mutagens was also found to depend on spontaneous mutation in seeds with different rates of germination. Special experiments showed that the interaction of ethylenimine with the metabolites of seeds in vitro leads to the formation of secondary active mutagens differing from ethylenimine in the nature of their action. The induction of chromosome-type aberrations by treating seeds with alkylating compounds may be due to the action of secondary mutagens.  相似文献

Understanding the dispersal processes that influence genetic structure in marine species requires estimating gene flow in a dynamic, fluid environment that is often poorly characterized at scales relevant to multiple dispersive stages (e.g. spores, gametes, zygotes, larvae, adults). We examine genetic structure in the marine alga Fucus vesiculosus L., which inhabits moderately exposed shores in the northern Atlantic but releases gametes only under sunny, calm conditions. We predicted genetic structure would correlate with coastal topography because weather frequently varies across coastal promontories on the Maine shore when F. vesiculosus is reproductive, which causes one side to experience high levels of water motion (= no gamete release) while one side is calm (= gamete release). Furthermore, we expected that the effect of low dispersal capacities of gametes and zygotes would result in spatial genetic structure over short distances. Using surface drifters, we characterized near-shore circulation patterns around the study sites to investigate whether directionality of gene flow was correlated with directionality of currents. We found significant genetic differentiation among sites sampled at two different peninsulas, but patterns of differentiation were unrelated to coastal topography and there was no within-site spatial structuring. Our genetic and near-shore circulation data, combined with an examination of gamete longevity, support the dependency of gene flow on storm-detached, rafting, reproductive adults. This study highlights the significance of rafting as a mechanism for structuring established populations of macroalgae and associated biota and demonstrates the importance of coupling population genetics' research with relevant hydrodynamic studies.  相似文献

Chlamydomonas reinhardi, a haploid isogamous green alga, presents a classic case of uniparental inheritance of chloroplast genes. Since the molecular basis of this phenomenon is poorly understood, an examination of the cytology of the C. reinhardi plastid DNA was made in gametes, newly formed zygotes, maturing zygotes, and at zygote germination.The single plastid per cell of Chlamydomonas contains a small number of DNA aggregates (‘nucleoids’) which can be seen after staining with DNA-binding fluorochromes. In zygotes formed by pre-stained gametes, the fluorescing nucleoids disappear from the plastid of mating type minus (male) gamete plastids but not from the plastid of mating type plus (female) gamete plastids about 1 h after zygote formation. Subsequently, nucleoids aggregate slowly to a final average of two or three in the single plastid of the mature zygote.Quantitative microspectrofluorimetry indicates that gametes of both mating types have equal amounts of plastid DNA, and that zoospores arising from zygotes have 3.5 × as much as gametes. Assuming degradation of male plastid DNA, there must be a very major synthesis of plastid DNA between zygote formation and zoospore release when zygotes produce the typical 8–16 zoospores. That synthesis appears to occur at germination, where there is a massive increase in plastid DNA and nucleoid number beginning just prior to meiosis. The results support the theory that uniparental inheritance results from degradation of plastid DNA entering the zygote via the male gamete and suggest further studies, using mutants and altered conditions, which might explain how male plastid DNA sometimes survives.  相似文献

The carbon balance in reproductive organs over the whole reproductive period was estimated for 20 woody species on the basis of the published data on the final dry weight and CO2 exchange of reproductive organs. The relationship between net respiration and dry weight growth was formulated by a linear function with a negative y-intercept. From this linear relationship, linear relations of dry weight growth and net respiration to net translocation were derived. The size dependence of net respiration showed that the net respiration changed from negative to positive as the final dry weight of the reproductive organ exceeded 0.481 g. The cost for translocation per unit dry weight was much lower in the species with small reproductive organs (<0.481 g) because of their photosynthetic contribution to the weight growth. The relation of dry weight growth/net translocation ratio to net translocation was expressed as a hyperbolic function. The dry weight growth/net translocation ratio remained at 0.650, in the higher range of net translocation, irrespective of woody species. Electronic Publication  相似文献

Phenolamides were extracted from reproductive organs and different capsule parts (fruit wall, placenta and seeds) of dormant and non-dormant lines derived from the species Petunia hybrida hort,. Petunia axillaries Lam. B.S.P. and Petunia parodii Steere. The different compounds were separated by high-pressure liquid chromatography and analyzed before pollination, during ripening, at ripeness and after 2 weeks of after-ripening of the seeds. The extracts were examined particularly for the presence of an alkaline compound (X), which is a phenolic derivative of tryptophanamide and appears to be a biochemical marker for the dormancy of ripe dry seeds of Petunia. During capsule growth and seed ripening, compound X was not a marker for dormancy, although it occurred in the reproductive organs of seed-parent plants. After pollination and fertilization, accumulation of compound X started mainly in the fruit wall, but in ripe dry capsules it was present entirely in the seeds. In contrast to compound X, neutral phenolamides were not detected in extracts from the fruit wall: and from the early stages of the capsule growth substantial accumulation took place only in the seeds.
Absence of compound X from ripe dry seeds appears as a necessary but not a sufficient condition for non-dormancy.  相似文献

Sensitivity to activation of germination and growth was studied using unfertilized female gametes and zygotes ofAllomyces macrogynus. When placed in nutrient medium, female gametes formed a branched rhizoid and a single, unbranched hypha that never formed reproductive structures during the 48-hr period of the experiment. Although partial female development was very asynchronous when compared to zygote development, nuclear divisions occurred and the nuclear cycle time was similar to that in zygote-derived hyphae over the first 7 hours of growth. After this time few divisions occurred in the female-derived hyphal nuclei. Comparison of nuclear number versus hyphal length in female- and zygote-derived hyphae showed no statistical difference, demonstrating that the actual number of nuclei was not the reason for incomplete development from female gametes. Female gametes and zygotes did not germinate in cycloheximide nor form a hyphal germ tube in actinomycin D. Motile zygotes plated on nonnutrient agar, or induced in nutrient medium before inoculation into nonnutrient liquid medium, developed a single, unbranched hypha with a small sporangium at the apical end. Ultracytochemistry with diaminobenzidine showed the presence of catalase-containing microbodies often in association with lipid bodies in female gametes. This suggested that although lipid may supply energy for female partial development, since zygotes completed development on nonnutrient medium, endogenous energy reserves are not the cause of incomplete female development.  相似文献

Germ-cell mutations may occur during meiosis, giving rise to independent mutant gametes in a Poisson process, or before meiosis, giving rise to multiple copies of identical mutant gametes at a much higher probability than the Poisson expectation. We report that the occurrence of these early premeiotic clusters of new identical mutant alleles increases the variance-to-mean ratio of mutation rate (R(u) > 1). This leads to an expected variance-to-mean ratio (R(t)) of the molecular clock that is always greater than one and may cover the observed range of R(t) values. Hence, the molecular clock may not be over-dispersed based on this new mutational model that includes clusters. To get a better estimation of R(u) and R(t), one needs measurements of the intrageneration variation of reproductive success (N(i)/N(e(i))), population dynamics (k(i)), and the proportion of new mutations that occur in clusters (r(c)), especially those formed before germ-cell differentiation.  相似文献

Summary Viable mutations for ear characters in two cultures of foxtail millet (Setaria italica Beauv.) were induced by individual and combined treatments of gamma rays, EMS (ethyl methanesulphonate), and dES (diethylsulphate). Four doses of gamma rays (10Kr to 40Kr), and four durations (6hrs to 24hrs) each of EMS (0.1 %) and dES (0.1 %) were used. In the combined treatments, only two durations (6hrs, 12hrs) of the chemical mutagens were used following each of the four doses of gamma rays.The mutation frequencies were recorded as mutants per 100 M2 plants and these were found to be higher in MU-1 than in MU-2 in all the treatments except dES. The frequencies also increased with increase in dose or duration of treatments and were found to be much higher in the combined treatments than in the individual treatments. In several combined treatments, a synergistic effect was observed in culture MU-1, the degree of synergism ranging from 1.01 to 1.62. Thus there was a differential response by the two cultures to the mutagenic treatments. The mutation spectrum was also found to be wider in culture MU-1, where 11 different kinds of mutants were recorded compared with only eight kinds of mutants recorded in MU-2.  相似文献

The in vitro fertilization system of maize is the well characterized model system for the fertilization process and early zygotic embryogenesis of higher plants. Application of molecular methods to the in vitro fertilization system led to the isolation of new genes and uncovered specific expression patterns of cell cycle regulators. Recent studies showed that expression of transgenes is possible in gametes and zygotes, thus transgenic approaches might offer an opportunity to unravel the roles of genes during fertilization and early development. The competence of gametes and zygotes to express transgenes will also enable the expression of GFP based reporter genes for the visualization of subcellular components in these cells in vivo. This review focuses on the data concerning the expression of transgenes in gametes and zygotes and describes some examples of recent developments in transgenic technology illustrating the emerging possibilities in experimental design by combining this technology with in vitro fertilization. The effects of the spider toxin alpha-latrotoxin (alpha-LTX) on gametes, zygotes, and early embryos of the sea urchin Paracentrotus lividus have been investigated by in vivo experiments and by histochemical studies of acetylcholinesterase (AChE) activity. Treatment of unfertilized eggs with nanomolar amounts (1 to 0.3 nmol/l) of alpha-LTX neither triggered cortical granule exocytosis, nor prevented the elevation of the fertilization layer by sperms. Instead, fertilized eggs exposed to alpha-LTX showed noticeable alterations in cell surface topography, including the appearance of prominent membrane-limited blebs. Moreover, the zygotes treated with 1 nmol/l alpha-LTX failed to cleave. The histochemical staining of treated zygotes revealed a very strong AChE activity in the cortical region, including blebs. An enzyme reaction was also found in the perivitellin space. Our results suggest the hypothesis that some alpha-LTX receptors may appear after fertilization, supporting the awareness that fertilized eggs display excitable cell features.  相似文献

对内蒙古中东部草原分布的克氏针茅进行了种群内和种群间的形态差异分析。结果表明:(1)种群内不同的形态性状存在不同程度的差异;(2)种群间生殖枝中的一部分形态性状以及营养枝高度和营养枝干重均存在较大差异;(3)每穗小花数、每穗籽粒数、每穗小穗数、生殖枝高、穗干重、生殖枝干重、每穗种子重、种子重/生殖枝重比等性状的变化趋势相同,均表现为随生境条件变差而增加,即在生殖上投入增加以增大适合度;(4)主成分分析表明穗长、种子重/生殖枝重比、每穗小花数、每穗籽粒数、生殖枝高、基盘长、第二芒柱长、营养枝高、千粒重、芒针长是不同种群差异的主要指标。这些形态性状的差异可以看作克氏针茅对不同生境的适应性表现。  相似文献

Methods have been developed which enable us to obtain in vitro fusion of pairs of sperm and egg cells, and sperm and central cells of angiosperms. Cultured products of such cell fusions develop progressively into zygotes, embryos and fertile plants, and endosperm, respectively. In vitro fusion of isolated gametes allows precisely timed examination of the earliest developmental processes following fertilization. When cultured, in vitro produced zygotes and primary endosperm cells organize themselves independently, and without any requirement for supporting tissues. This technology thus constitutes a unique model system for studies of early stages of zygotic embryogenesis and endosperm development. Following the adaptation of molecular techniques for use with only a few cells, it has proved possible to investigate developmental processes in these systems. This review describes the successful combination of molecular techniques with in vitro fertilization methods, and highlights results obtained with small numbers of reproductive cells isolated by microdissection.  相似文献

The inheritance of chloroplast genes in Chlamydomonas is regulated by methylation of chloroplast DNA during gametogenesis. The wild-type pattern of maternal inheritance results from the methylation of chloroplast DNA in female (mt+) but not in male (mt-) gametes, leading to preferential degradation of chloroplast DNA of male origin in zygotes. This paper describes the distribution of 5-methyl cytosine residues in restriction fragments of chloroplast DNA sampled during gametogenesis by two methods: ethidium bromide staining of agarose gels, and binding of antibody directed against 5-methyl cytosine onto restriction fragments blotted to nitro-cellulose paper. Methylated cytosines are located in most if not all Eco RI and Msp I fragments, but the extent of methylation is not proportional to fragment size. The mat-1 mutation carried by males converts maternal inheritance. Chloroplast DNA of male gametes carrying the mat-1 mutation becomes methylated during gametogenesis. This methylation protects against restriction enzyme-promoted degradation in zygotes, as shown by physical data demonstrating the transmission to progeny of chloroplast genes carried on chloroplast DNA of the mat-1 male parent. Thus the mat-1 gene, which is linked to the mating-type locus, determines whether or not methylation of chloroplast DNA will occur in males during gametogenesis.  相似文献

高等植物的卵细胞深藏在子房内的胚珠体细胞组织中,形成了对高等植物受精过程研究的技术障碍。以前采用超微结构观察研究受精过程已取得了一定的结果,但用固定切片技术研究受精机理需将卵细胞杀死,并且不能进行定点追踪观察。将高等植物的精、卵细胞分离出来在体外诱导其融合的离体受精技术可在很大程度上克服这些技术障碍,对雌、雄配子的识别和融合,合子开始胚胎发生等一系列的受精和胚胎发生机理进行研究。分离的雌、雄配子及合子使应用分子生物学方法研究这些细胞的结构和功能成为可能。将合子的二倍性和胚胎发生特性与外源DNA转入技术结合起来可使转基因植物研究的后期工作简单化。另外,异种植物离体精、卵细胞融合和杂种合子的培养也是进行远缘杂交的一条有潜力的途径。  相似文献

Colpomenia sinuosa (Mertens ex Roth) Derbès and Solier (Scytosiphonaceae, Phaeophyceae) is a common species on the rocky intertidal shores of the Azores, where reproductive gametophytes occur throughout the year. Life‐history studies of this species were carried out in culture, and both sexual and asexual reproduction were observed. Anisogamous gametes fused to form zygotes. The zygotes gave rise to a filamentous prostrate sporophyte generation bearing unilocular sporangia, under both short‐day and long‐day conditions at 15 and 22°C, and to both unilocular and plurilocular sporangia, under the lower temperature condition. Unispores developed into gametophytes, and plurispores gave rise to filamentous sporophytes. Asexual reproduction was carried out by unfused female gametes and asexual plurispores produced from the same gametophyte. Unfused gametes developed into filamentous prostrate sporophytes producing unilocular sporangia in both culture conditions, and unispores released from the sporangia gave rise to gametophytes. Asexual plurispores from field gametophytes, under both culture conditions, developed directly into new gametophytes. The species exhibited three types of life history: a heteromorphic, diplohaplontic; a heteromorphic, monophasic (both with alternation between the erect and filamentous prostrate thalli); and a monomorphic, monophasic.  相似文献

Our understanding of the development of intrinsic reproductive isolation is still largely based on theoretical models and thorough empirical studies on a small number of species. Theory suggests that reproductive isolation develops through accumulation of epistatic genic incompatibilities, also known as Bateson–Dobzhansky–Muller (BDM) incompatibilities. We can detect these from marker transmission ratio distortion (TRD) in hybrid progenies of crosses between species or populations, where TRD is expected to result from selection against heterospecific allele combinations in hybrids. TRD may also manifest itself because of intragenomic conflicts or competition between gametes or zygotes. We studied early stage speciation in Arabidopsis lyrata by investigating patterns of TRD across the genome in F2 progenies of three reciprocal crosses between four natural populations. We found that the degree of TRD increases with genetic distance between crossed populations, but also that reciprocal progenies may differ substantially in their degree of TRD. Chromosomes AL6 and especially AL1 appear to be involved in many single- and two-locus distortions, but the location and source of TRD vary between crosses and between reciprocal progenies. We also found that the majority of single- and two-locus TRD appears to have a gametic, as opposed to zygotic, origin. Thus, while theory on BDM incompatibilities is typically illustrated with derived nuclear alleles proving incompatible in hybrid zygotes, our results suggest a prominent role for distortions emerging before zygote formation.  相似文献

Sexual differentiation in the heterothallic alga Chlamydomonas reinhardtii is controlled by two mating-type loci, mt+ and mt-, which behave as a pair of alleles but contain different DNA sequences. A mutation in the mt minus-linked imp11 gene has been shown previously to convert a minus gamete into a pseudo-plus gamete that expresses all the plus gametic traits except the few encoded by the mt+ locus. Here we describe the iso1 mutation which is unlinked to the mt- locus but is expressed only in minus gametes (sex-limited expression). A population of minus gametes carrying the iso1 mutation behaves as a mixture of minus and pseudo-plus gametes: the gametes isoagglutinate but they do not fuse to form zygotes. Further analysis reveals that individual gametes express either plus or minus traits: a given cell displays one type of agglutinin (flagellar glycoprotein used for sexual adhesion) and one type of mating structure. The iso1 mutation identifies a gene unlinked to the mating-type locus that is involved in sex determination and the repression of plus-specific genes.  相似文献
