用正交试验法探讨了以麦秸为原料进行纤维素酶降解的工艺条件。正交试验的结果表明,影响麦秸纤维素降解的因素的主次顺序为A(酶添加量)>B(底物浓度)>E(时间)>C(温度)>D(pH值),纤维素酶解麦秸纤维素的最佳组合为A3B1E3C3D2,即纤维素酶的添加量为0.2%,底物浓度为5%,反应时间为2h,反应温度50℃,pH5.0时为最佳条件。在比常规酶解法时间缩短12-30倍的条件下,能使纤维素降解葡萄糖的转化率达22.3%。  相似文献

本研究尝试将氨基磺酸应用于甘蔗渣预处理,探究其作为酸预处理试剂对甘蔗渣成分和酶解的影响。氨基磺酸预处理最优条件为浓度3%,温度121℃,预处理1 h。在该条件下,甘蔗渣的固体回收率为64.45%,半纤维素和木质素去除率分别为70.81%和25.10%,纤维素损失率仅7.56%。与硫酸、盐酸预处理相比,氨基磺酸的半纤维素和木质素去除率不如硫酸、盐酸预处理,但固体回收率更高,纤维素损失率低,能保留更多纤维素有效成分。进一步酶解显示,氨基磺酸预处理的纤维素转化率高于硫酸、盐酸预处理。氨基磺酸作为一种新的酸预处理试剂,在木质纤维素降解上有良好应用前景。  相似文献

以单酸正交试验结果为基础,对二酸混合法和三酸混合法降解植物纤维素的最佳工艺条件进行了探讨。二酸混合法的结果表明,酸的添加顺序严重影响纤维素向葡萄糖的转化。强酸与弱酸同时混用均使葡萄糖的转化率降低。但盐酸与乙酸的混用效果则因所选最佳条件的不同而不同,虽然硫酸与盐酸均为无机强酸,与乙酸混用时添加的顺序亦一样即先弱酸后强酸,但其结果差别甚大,前者使葡萄糖的转化率降低,后者则使葡萄糖的转化率由40.9%锰增到62.7%。三酸混合法的结果表明,在本研究所选用的四种酸中,无论是哪三种酸同时混用,均使葡萄糖的转化率降低;当按一定顺序添加时,其结果亦不如二酸混合法,结论:在用多种酸降解植物纤维素时,一定要按一定的顺序添加,最好用两种酸即可。  相似文献

以小麦秸秆为原料,通过正交实验对纤维素酶降解秸秆纤维的影响因素进行了研究.结果表明,影响小麦秸秆降解的因素依次为:酶量>酶解时间>料液比>反应温度,其最适条件是:加酶量为40 u/g,酶解时间为10h,反应温度为40℃,料液比为1∶3,总糖含量达到43.24%.以米氏方程为基础,建立起最适酶解条件下总纤维素降解的动力学模型.  相似文献

以大黄酸含量为参考指标,通过单因素实验分别考察硫酸浓度(A)、液料比(B)、提取加热时间(C)、提取加热温度(D)对大黄中大黄酸提取量的影响,并根据单因素试验结果设计正交试验。结果表明,影响大黄中大黄酸提取工艺的主次因素为:加热温度D>加热时间C>硫酸浓度A>液料比B,大黄中大黄酸的最佳提取工艺为:15%硫酸、5∶1液料比、55℃加热回流2.5 h。此法操作性强,优选出最佳提取工艺条件,为开发大黄中泻下有效部位提供了理论依据。  相似文献

以挥发油提取率为指标,通过单因素和L9(34)正交试验设计确定了水蒸气蒸馏法提取大蒜挥发油的最佳提取工艺及条件。结果表明,各因素作用主次顺序为蒸馏时间(D)>料液比(C)>发酵温度(A)>浸泡时间(B),各因子的最优组合为A2B2C2D3,即以1∶3.5的料液比(m∶V),在35℃时浸泡3h后蒸馏2h为大蒜挥发油的最佳提取工艺,此时挥发油的提取率达到0.32%,证明此方法是大蒜挥发油提取的可靠、易行的科学方法。  相似文献

本文介绍了出分解纤维素的嗜热厌氧菌Clostridium celluloflavus sp.nov.直接发酵纤维素产乙醇的初步研究、发酵于60℃下进仃,其主要产物为乙醇、乙酸、氢气和二氧化碳。文中介绍了间歇发酵的若干特征与影响发酵的因素,1%纤维素发酵至120小时,大约有70%纤维素被分解;乙醇的转化率约为0.36g/g降解纤维素;发酵液中乙醇浓度达到56至61mM。发酵中乙醇与乙酸浓度的比值因发酵时间与其它发酵条件的不同而不同。  相似文献

以薯蓣皂苷元的提取率为指标,应用L9(3^4)正交试验设计优化了从脱多糖、脱脂胡芦巴豆粉中提取甾体皂苷的工艺条件,并且筛选了ZTC澄清剂对胡芦巴甾体皂苷水提取液的澄清条件。实验结果表明,影响水提取的主次因素为:提取次数>提取温度>固液比>提取时间;最佳水提取条件为:固液比为1:10,在70℃下提取3次,每次120min得出胡芦巴种子水提取液的澄清条件如下:ZTCl 1澄清剂的加入次序是先加B组分再加入A组分;澄清剂B和A的最佳用量分别为1.0g/L和O.5g/L;加入组分A或B后在80℃下作用30min即可达到澄清目的。  相似文献

采用正交试验方法 ,研究了鸭毛梗水解制备复合氨基酸的工艺条件 ,结果表明 :温度 /压力是影响氨基酸转化率最主要的因素。鸭毛梗水解制备的复合氨基酸转化率盐酸法最高 (82 .36 % ) ,氢氧化钠法最低 (5 8.4 6 % ) ,硫酸法与盐酸法接近(79.4 4 % )。硫酸法产率较高 ,操作方便 ,环境污染和设备腐蚀均小 ,适合大量生产 ,确定为水解制备复合氨基酸介质 ,其最佳工艺条件为 :水解时间 8.0h ,硫酸浓度 3.0mol·L-1,水解温度 12 5℃。氨基酸分析表明 ,水解液中均含有 18种以上氨基酸。  相似文献

It has now been over twenty years since a novel herpesviral genome was identified in Kaposi's sarcoma biopsies. Since then, the cumulative research effort by molecular biologists, virologists, clinicians, and epidemiologists alike has led to the extensive characterization of this tumor virus, Kaposi's sarcoma-associated herpesvirus(KSHV; also known as human herpesvirus 8(HHV-8)), and its associated diseases. Here we review the current knowledge of KSHV biology and pathogenesis, with a particular emphasis on new and exciting advances in the field of epigenetics. We also discuss the development and practicality of various cell culture and animal model systems to study KSHV replication and pathogenesis.  相似文献

Comprises species occurring mostly in subtidal habitats in tropical, subtropical and warm-temperate areas of the world. An analysis of the type species, V. spiralis (Sonder) Lamouroux ex J. Agardh, a species from Australia, establishes basic characters for distinguishing species in the genus. These characters are (1) branching patterns of thalli, (2) flat blades that may be spiralled on their axis, (3) width of the blade, (4) primary or secondary derivation of sterile and fertile branchlets and (5) position of sterile and fertile branchlets on the thalli. Application of the latter two characters provides an important basic method for separation of species into three major groups. Osmundaria , a genus known only in southern Australia, was studied in relation to Vidalia , and its separation from the Vidalia assemblage is not accepted. Species of Vidalia therefore are transferred to the older genus name, Osmundaria. Two new species, Osmundaria papenfussii and Osmundaria oliveae are described from Natal. Confusion in the usage of the epithet, Vidalia fimbriala Brown ex Turner has been clarified, and Vidalia gregaria Falkenberg, described as an epiphyte on Osmundaria pro/ifera Lamouroux, is revealed to be young branches of the host, Osmundaria prolifera.  相似文献

Fifteen chromosome counts of six Artemisia taxa and one species of each of the genera Brachanthemum, Hippolytia, Kaschgaria, Lepidolopsis and Turaniphytum are reported from Kazakhstan. Three of them are new reports, two are not consistent with previous counts and the remainder are confirmations of very scarce (one to four) earlier records. All the populations studied have the same basic chromosome number, x = 9, with ploidy levels ranging from 2x to 6x. Some correlations between ploidy level, morphological characters and distribution are noted.  相似文献

采用原位分子杂交方法检测HCV RNA及HBV X基因;采用免疫组织化学方法研究HCV核心抗原,非结构区C33c抗原及HBxAg在肝细胞肝癌中的定位及分布.结果表明(1)HCV RNA、HBV X基因在肝细胞肝癌组织检出率分别为40%(55/136)和82%(112/136).HCV RNA定位于癌细胞的胞浆内,阳性细胞呈散在、灶状及弥漫分布三种形式;HBV X基因在肝癌细胞中的分布呈胞浆型、核型及核浆型,阳性细胞也呈上述三种分布形式;(2)HCV C33c抗原、核心抗原在肝细胞肝癌中的阳性率为81%(133/164)及86%(141/164).C33c抗原定位于癌细胞及肝细胞的胞浆内;核心抗原既定位于癌细胞核中,又可定位于胞浆中.C33c抗原阳性细胞以灶状分布为主;而核心抗原阳性细  相似文献

For a plant selection model with frequency-independent viabilities, fertilities and selfing rates, it is shown that apart from global fixation, for certain parameter combinations a protected polymorphism and facultative fixation (either allele may become fixed according to initial frequencies) may both occur. Facultative fixation requires different selling rates for the dominant and recessive type. Protection of the polymorphism requires resource allocation for male and female function. In this connection the problem of purely genetically caused population extinction is discussed.
For general frequency dependence and regular segregation, the chances for establishment of a completely recessive gene are compared to those of a completely dominant gene. It is proven that the process of establishment of the recessive gene, despite a fitness advantage, may be considerably endangered by drift effects if random mating prevails. The recessive gene may reach the same effectivity in establishment as a dominant gene, only if the recessive homozygote mates exclusively with its own type during the period of establishment.  相似文献
