Evidence that face perception is mediated by special cognitive and neural mechanisms comes from fMRI studies of the fusiform face area (FFA) and behavioral studies of the face inversion effect. Here, we used these two methods to ask whether face perception mechanisms are stimulus specific, process specific, or both. Subjects discriminated pairs of upright or inverted faces or house stimuli that differed in either the spatial distance among parts (configuration) or the shape of the parts. The FFA showed a much higher response to faces than to houses, but no preference for the configuration task over the part task. Similarly, the behavioral inversion effect was as large in the part task as the configuration task for faces, but absent in both part and configuration tasks for houses. These findings indicate that face perception mechanisms are not process specific for parts or configuration but are domain specific for face stimuli per se.  相似文献

Many bushcrickets produce specific song spectra for acoustic communication. Song detection and/ or recognition may make use of such specificity. Where in the nervous system are the filters for song frequency situated? A peripheral tuning for song frequency typically does not exist. Auditory receptor cells of bushcrickets connect to local and ascending neurons in the prothoracic ganglion. One of the ascending neurons (1) may function as a frequency filter in a group of four related bushcrickets (genera Ancistrura, Barbitistes). The frequency response of ascending neuron 1 is species-specific roughly corresponding to the frequency of the conspecific male song. The species-specific tuning of the neuron is not brought about by specific excitation, but by specific inhibition. By eliminating this frequency-dependent and species-specific inhibition the former filter neuron is transformed into an unspecific broad-band neuron in all four species. Its tuning then does not differ from omega neuron 1, a local neuron which is rather unspecific for frequency. Also, the supra-threshold responses of ascending neuron 1, which are different in intact animals, are similar to each other and similar to omega neuron 1 following elimination of inhibition. Only ascending neuron 1 of Ancistrura retains some species-specific features at low frequencies. In conclusion, evolution changed inhibition, not excitation of a species-specific neuron.  相似文献

Protein folding is often accompanied by formation of non-native conformations leading to protein aggregation. A number of reports indicate that antibodies can facilitate folding and prevent aggregation of protein antigens. The influence of antibodies on folding is strictly antigen specific. Chaperone-like antibody activity may be due to the stabilization of native antigen conformations or folding transition states, or screening of aggregating hydrophobic surfaces. Taking advantage of chaperone-like activity of antibodies for immunotherapy may prove to be a promising approach to the treatment of Alzheimers and prion-related diseases. The role of phosphatidylinositol-specific phospholipase C (PIase C) in a) the enigmatic phosphatidylinositol (PI) turnover and b) in our understanding of membrane enzyme-PI interactions is the subject matter of this article. PIase C is present in both procaryotes and eukaryotes. This enzyme is considered to be involved in the cells PI breakdown which occurs in response to several external stimuli. Recent information on the physical properties, Ca2+ requirement, cellular localization and modulation of the activity of PIase C of mammalian systems can help to evaluate the PI turnover from a new angle. Existing evidence suggests that Ca2+-dependent PI breakdown is probably mediated through the cytosolic and particulate PIase C while a Ca2+ independent pathway is catalyzed by a lysosomal enzyme. Apparently PI turnover may be operating through more than one mechanism. The association of this phenomenon with a membrane receptor event linked with “Ca2+ gating” may have to be reconsidered. Modulation of the PIase C activity by unsaturated amphiphiles or the presence of this enzyme in different physico-chemical forms could be a potential regulatory feature.Hydrolysis of membrane PI of a number of cells and tissues by the bacterial PIase C has been shown to cause substantial release of acetylcholinesterase, alkaline phosphatase and 5′-nucleotidase in free, soluble form. Other membrane enzymes, e.g., alkaline phosphodiesterase I, L-leucyl-β naphtyl amidase and Ca2+ or Mg2+ ATPase are not affected. These results indicate a specific interaction between PI and certain enzymes in membranes. The chemical nature of this linkage, whether it is covalent or non-covalent, has also been explored and has provided intriguing insight into this phenomenon. New findings also indicate that hydrolysis of PI by PIase C also can cause modifications in membrane-enzyme activities, e.g., adenylate cyclase.  相似文献

The differential analysis of genes between microarrays from several experimental conditions or treatments routinely estimates which genes change significantly between groups. As genes are never regulated individually, observed behavior may be a consequence of changes in other genes. Existing approaches like co-expression analysis aim to resolve such patterns from a wide range of experiments. The knowledge of such a background set of experiments can be used to compute expected gene behavior based on known links. It is particularly interesting to detect previously unseen specific effects in other experiments. Here, a new method to spot genes deviating from expected behavior (PAttern DEviation SCOring--Padesco) is devised. It uses linear regression models learned from a background set to arrive at gene specific prediction accuracy distributions. For a given experiment, it is then decided whether each gene is predicted better or worse than expected. This provides a novel way to estimate the experiment specificity of each gene. We propose a validation procedure to estimate the detection of such specific candidates and show that these can be identified with an average accuracy of about 85%.  相似文献

Staphylokinase is a 135 amino acid protein produced by certain strains of Staphylococcus aureus. It belongs to fibrin-specific plasminogen activator. Staphylokinase converts plasminogen--the inactive proenzyme--to the plasmin, which dissolves the fibrin of a blood clots. When the immune system encounters alloantigen it can respond in any one of a number of different ways. The choice that is made will take into account factors such as where, when and how the contact with the alloantigen takes place, as well as the environmental conditions that prevail at the time the alloantigen is encountered. Alloantigen administration before transplantation either alone or in combination with therapeutic agents that modulate the functional activity of the responding leucocytes can be a powerful way of inducing specific unresponsiveness to alloantigens in vivo. The molecular mechanisms that influence the way the outcome of the immune response to alloantigen develops, either activation or unresponsiveness to the triggering antigen, hold the key to our ability to manipulate the immune system effectively by exposing it to donor antigen for therapeutic purposes. This review will focus on alloantigen-induced immunological unresponsiveness and how insights into the mechanisms of unresponsiveness have driven the development of novel tolerance-induction strategies that show promise for translation into the clinic in the future.  相似文献

Attitudes of the physician and attitudes of the patient may affect the rational use of specific therapies. An example is the use of penicillin for the common cold. A survey showed that 25 per cent of physicians and 16 per cent to 22 per cent of three other groups polled had received penicillin for a cold, although there is no rational basis for such therapy. Evaluation of both the physicians' and patients' attitudes toward specific therapies might lead to a more rational treatment of the patient as a total personality.  相似文献
