The aim of this work was to evaluate the response to biobleaching of steam exploded kraft pulps and to compare the results with the controls. For this end, a laccase-mediator treatment using commercial laccase (Novozyme 51003) and a natural mediator (acetosyringone) were assayed, followed by alkaline extraction and hydrogen peroxide stages.Our approach resulted in exploded biobleached pulps with lower kappa number and improved optical properties compared to controls, even after subjecting pulps to accelerated ageing. Additionally, use of hydrogen peroxide was reduced. The LMS (laccase-mediator system) had a smaller impact on the properties of the bleached pulps and on hydrogen peroxide consumption than the steam explosion process did.  相似文献

The effects of multiple xylanase treatments were assessed during the peroxide bleaching of three pulps: Douglas-fir (kraft); Western hemlock (oxygen delignified kraft); and trembling Aspen (kraft). The addition of a xylanase treatment stage, either before or after the peroxide bleaching stage(s), resulted in the enhanced brightening of all pulps. A higher brightness was achieved using two enzyme treatments, one before and one after the peroxide stage(s). Both bleach boosting and direct brightening seemed to contribute to the enhancement of the peroxide bleaching. Compared to xylanase prebleaching, xylanase posttreatment of peroxide bleached pulps solubilized less lignin and chromophores and made smaller amounts of these materials alkaline soluble. Nevertheless, the final brightness achieved by xylanase posttreatment was similar or superior to that achieved with xylanase prebleaching of the corresponding unbleached pulps. Abstract: The use of white-rot fungi, especially Trametes versicolor and isolate IZU-154, to delignify and brighten kraft pulps is reviewed. The fungal treatments are effective but slow; the responsible enzymes are being studied with a view to accelerating the process. Manganese peroxidase, or laccase with a co-substrate, can demethylate and partially solubilize the lignin in pulps, mimicking the early steps of the fungal delignification.  相似文献

Four xylanase preparations that are commercially available, namely Cartazyme from Sandoz, Ecopulp from Alko-ICI, Irgazyme from Ciba-Genencor and Pulpzyme HB from Novo Nordisk, were tested in bleaching experiments of kraft pulps from Pinus radiata. The main objective of this study was to optimize a reduction in the consumption of chlorine dioxide in the bleaching sequences C90/D10EoDED, C70/D30EoDED and D100EDED. Enzymatic treatments led to savings of ClO2 between 3.5 and 3.9 kg per air-dried tons (ADT) in the three bleaching sequences, without affecting the target brightness of the pulps. In these assays, some minor although reproducible differences in the performance of the enzymes were observed. In most cases, xylanase treatment partially affected the beatability of the pulps, measured as the number of revolutions in the PFI mill required to reach the same tensile index as the respective controls.  相似文献

Parameters influencing the mutagenic properties of spent bleaching liquors from sulphite pulps have been studied. In addition a comparison has been made between the properties of spent liquors from sulphite and kraft pulp bleaching. In the sulphite process the cooking base had no influence on the mutagenicity of the chlorination stage. In contrast, removing the extractives before chlorination especially for dissolving pulp resulted in an increase in mutagenic activity. The mutagenicity decreased significantly after substituting 40% of the chlorine with chlorine dioxide. Sequential addition of chlorine and chlorine dioxide resulted in higher activity than simultaneous or premixed chlorination as observed for liquors from kraft pulp. Increasing the pH of the extracts or addition of sulphur dioxide decreased the mutagenicity. Expressed as 10(7) revertants per kappa number and ton pulp the mutagenicity varied between 10 and 40 for sulphite pulp while the corresponding figures for kraft pulp were 100-225.  相似文献

The effect of a pretreatment consisting of steam explosion (SE) followed by a laccase mediator system (LMS) stage on Eucalyptus globulus kraft pulping has been evaluated and compared with fungal pretreatments. Pretreatment with SE and LMS was more efficient than pretreatments using Pycnoporus sanguineus and Trametes sp. I-62. Steam explosion not only improved the enzyme penetration into the wood chips and shortened the pulping process by 60%, but also extracted around 50% of the hemicelluloses which could be converted into value-added products. The optimal conditions for the LMS treatment were 3 h, 3 UA/g and 40 °C. Compared to SE, the SE/LMS treatment yielded an increase in delignification of 13.9% without affecting pulp properties, provided a similar screened kraft yield, and reduced consumption of chemical reagents Na2S and NaOH by 11.5% and 6.3%, respectively. Therefore, SE/LMS is a promising pretreatment for converting the pulp mill into a forest bio-refinery.  相似文献

Summary Improving the availability of oxygen by adding polydimethylsiloxanes (PDMS) oxygen carriers to Trametes versicolor cultures increased pulp brightening. The presence of the oxygen carriers in cultures of T. versicolor with hardwood kraft pulp increased the growth rate of the fungus, but not the ultimate biomass yield. The PDMS also stimulated brightening of hardwood kraft pulp by it T. versicolor immobilized in polyurethane foam. A threefold increase in the oxygen uptake rate in T. versicolor cultures with PDMS was observed. Lipophilic wood extractives commonly referred to as pitch, cause significant problems for the pulp and paper industries. The reduction of these extractives is an important aspect that concerns industries around the world. In the present work the change in the amount and chemical composition of lipophilic extractives from Eucalyptus spp. stored for 20, 40, 60, 100, 140 and 180 days after harvesting was investigated. The results showed a decrease in extractives content with storage time, with the most significant decrease occurring 60 days after harvesting. In addition, fatty acids and sterols were the main classes of compounds responsible for the significant decrease in extractive content. Data were analyzed by principal component analysis. PC1 explains approximately 99% of the total variance, and beta-sitosterol was the major compound responsible for the differentiation. These studies demonstrate that in terms of economical aspects, quality of the pulp and paper and minimization of pitch formation, the best period of wood storage is 60 days.  相似文献

The possibility of using xylanase preparations for hydrolyzing hemicelluloses in a non-bleached kraft pulp in order to facilitate its bleaching was studied. The effects of enzymatic preparations of fungal and bacterial origins were examined, and the optimal conditions for xylanase activity were determined. UV spectroscopy demonstrated that the treatment of kraft pulp with enzymatic preparations containing xylanase facilitated the subsequent removal of lignin and increased the brightness by 5%. The effect of enzymatic treatment was retained in the case of peroxide bleaching. The enzymatic preparations studied are promising for the development of chlorine-free pulp bleaching technologies.  相似文献

The chemical composition of the epicuticular wax from the fruits of Eucalyptus globulus was studied by GC-MS before and after alkaline hydrolysis. The wax had two main components, ursolic acid and tritriacontan-16,18-dione, together with several other triterpenic acids. After alkaline hydrolysis, a large increase in the amounts of triterpenic acids and fatty acids (particularly in hexadecanoic acid) was observed, suggesting that these components were present predominantly in esterified forms in the fruit wax. Six compounds were isolated from the fruits by preparative chromatography, and were identified as 8-desmethyleucalyptin, sesamin, tritriacontan-16,18-dione, ursolic acid, 3beta-hydroxyurs-11-en-13beta(28)-olide (ursolic acid lactone) and 3beta,11alpha-dihydroxyurs-12-en-28-oic acid, the latter of which was identified for the first time.  相似文献

Summary Flooding for up to 40 days induced morphological changes and reduced growth of 6-week-old Eucalyptus camaldulensis and Eucalyptus globulus seedlings. However, the specific responses to flooding varied markedly between these species and with duration of flooding. Both species produced abundant adventitious roots that originated near the tap root and original lateral roots, but only E. camaldulensis produced adventitious roots on submerged portions of the stem. Flooding induced leaf epinasty and reduced total dry weight increment of seedling of both species but growth of E. globulus was reduced more. In both species dry weight increment of shoots was reduced more than dry weight increment of roots, reflecting compensatory growth of adventitious roots. Spontaneous bleeding of sugar-rich sap from cambial-deep incisions in the bark of trunks was demonstrated for Eucalyptus globulus and other eucalypts across a range of localities and seasonal conditions in south-west Australia. High levels of sucrose and raffinose (up to 31% w/v total sugars) were present in the exudates, and upward and downward gradients in exudate sugar concentrations were recorded between samples obtained at different heights up trunks of E. globulus. The data indicated a phloem origin for the exudates, with source:sink pressure gradients driving translocation. Concentration ratios of sugars to amino acids were consistently lower in exudate from upper (distal) than basal regions of trunks, suggesting preferential partitioning of nitrogen upwards towards the trunk apex. A comparison of phloem and xylem sap composition from one plantation over a season showed nitrate in xylem but not phloem and substantial amounts of sodium, and high concentrations of chloride and sulphate relative to phosphate in xylem and phloem. Phloem sap sampled across a range of 29 contrasting plantations of E. globulus at peak stress (autumn) showed great inter-site variability in concentrations of amino acids, sulphur, sodium and certain trace elements and in C:N and Na:K ratios of sap. Carbon isotope ratios (δ13C) were strongly correlated with sugar concentrations of the sap samples from these and other plantations. Use of sap compositional attributes of phloem and δ13C values of translocated carbon is suggested for assessing the current nutritional condition and water status of E. globulus plantings. It is generally agreed that unsaturated fatty acids (UFA) are an important class of target molecule for reaction with ozone when polluted air is inhaled. Most discussions have implicated the UFA in cell membranes, but lung lining fluids also contain fatty acids that are from 20 to 40% unsaturated. Since UFA in lung lining fluids exist in a highly aquated environment, ozonation would be expected to produce aldehydes and hydrogen peroxide, rather than the Criegee ozonide. In agreement with this expectation, we find that ozonations of emulsions of fatty acids containing from one to four double bonds give one mole of H2O2 for each mole of ozone reacted. Ozonation of oleic acid emulsions and dioleoyl phosphatidyl choline gives similar results. with two moles of aldehydes and one mole of H2O2 formed per mole of ozone reacted. The net reaction that occurs when ozone reacts with pulmonary lipids is suggested to be given by equation 1. [formula: see text]. From 5 to 10% yields of Criegee ozonides also appear to be formed. In addition, a direct reaction of unknown mechanism occurs between ozone and UFA in homogeneous organic solution, in homogeneous solutions in water, in aqueous emulsions, and in lipid bilayers to give organic radicals that can be spin trapped. These radicals are suggested to be responsible for initiating lipid peroxidation of polyunsaturated fatty acids. Thus, aldehydes, hydrogen peroxide, and directly produced organic radicals are suggested to be mediators of ozone-induced pathology.  相似文献

As part of its aerobic metabolism, Streptococcus pneumoniae generates high levels of H(2)O(2) by pyruvate oxidase (SpxB), which can be further reduced to yield the damaging hydroxyl radicals via the Fenton reaction. A universal conserved adaptation response observed among bacteria is the adjustment of the membrane fatty acids to various growth conditions. The aim of the present study was to reveal the effect of endogenous reactive oxygen species (ROS) formation on membrane composition of S. pneumoniae. Blocking carbon aerobic metabolism, by growing the bacteria at anaerobic conditions or by the truncation of the spxB gene, resulted in a significant enhancement in fatty acid unsaturation, mainly cis-vaccenic acid. Moreover, reducing the level of OH(.) by growing the bacteria at acidic pH, or in the presence of an OH(.) scavenger (salicylate), resulted in increased fatty acid unsaturation, similar to that obtained under anaerobic conditions. RT-PCR results demonstrated that this change does not originate from a change in mRNA expression level of the fatty acid synthase II genes. We suggest that endogenous ROS play an important regulatory role in membrane adaptation, allowing the survival of this anaerobic organism at aerobic environments of the host.  相似文献

Abstract. In experiments where mung beans ( Vigna radiata L.) and peas ( Pisum sativum L.) have been pre-exposed to ethylene and afterwards treated with ozone, it has been shown that such ethylenepretreated plants may become more resistant to ozone. Further experiments with hydrogen peroxide (H2O2) and the herbicide paraquat suggest that this increased resistance against ozone depends on the stimulation of ascorbate peroxidase activity which provides cells with increased resistance against the formation of H2O2 which is also formed when plants are fumigated with ozone. These results explain why increased production of ethylene can be observed in plants exposed with ozone or other oxidative stress and clearly demonstrate that in plants, as well as animals, peroxidases protect cells against harmful concentrations of hydroperoxides.  相似文献

The plant stress hypothesis suggests that some herbivores favour stressed plants, whereas the plant vigour hypothesis proposes that other herbivores prefer vigorous plants. The effects of a prior stress, that of frost damage, were examined on the subsequent growth of Eucalyptus globulus globulus and on the response of insect herbivores. Frost damage affected tree growth by reducing new leaf area and increasing specific leaf area (SLA). However, herbivore abundance was not affected by prior frost damage. Two feeding trials using Anoplognathus chloropyrus and Hyalarcta huebneri and a morphometric study of Ctenarytaina eucalypti were conducted to assess the performance of herbivores on trees that had suffered more or less frost damage. Consumption by A. chloropyrus and H. huebneri was unaffected by foliage origin (damaged versus healthy). Hyalarcta huebneri grew faster when fed leaves from previously damaged trees, and C. eucalypti from previously damaged trees were larger than those from healthy trees. Enhanced insect performance on frost damaged plants may have resulted from the high specific leaf area (most likely thinner) leaves. The herbivore abundance data did not support the hypothesis that previously frost damaged plants are preferred by insects. However, increased growth of H. huebneri and larger body size of C. eucalypti on damaged trees indicates that previously stressed trees may produce leaves of higher nutritional value.  相似文献

The extracellular productions of beta-xylanase, beta-xylosidase, beta-glucosidase, beta-mannanase, arabinosidase, alpha-glucuronidase, alpha-galactosidase and Fpase from Bacillus pumilus CBMAI 0008 were investigated with three different xylan sources as substrate. The enzymatic profiles on birchwood, Eucalyptus grandis and oat were studied at alkaline and acidic pH conditions. B. pumilus CBMAI 0008 grown on the three carbon sources produced mainly beta-xylanase. At pH 10, the levels of xylanase were 328, 160 and 136 U/ml, for birch, oat and E. grandis, respectively. beta-Mannanase production was induced on E. grandis (5 U/ml) and arabinofuranosidase on oat (5 U/ml). Although small quantities of alpha-glucuronidase had been produced at pH 10, activity at pH 4.8 was 1.5 U/ml, higher than observed for Aspergillus sp. in literature reports. Preliminary assays carried out on E. grandis kraft pulp from an industrial paper mill (RIPASA S.A. Celulose e Papel, Limeira, SP, Brazil) showed a reduction of 0.3% of chlorine use in the pulp treated with the enzymes, resulting in increased brightness, compared to conventional bleaching. The enzymes were more efficient if applied before the initial bleaching sequence, in a non-pre-oxygenated pulp.  相似文献
