The aim of this investigation was to achieve the first step toward a comprehensive model of the lymphatic system. A numerical model has been constructed of a lymphatic vessel, consisting of a short series chain of contractile segments (lymphangions) and of intersegmental valves. The changing diameter of a segment governs the difference between the flows through inlet and outlet valves and is itself governed by a balance between transmural pressure and passive and active wall properties. The compliance of segments is maximal at intermediate diameters and decreases when the segments are subject to greatly positive or negative transmural pressure. Fluid flow is the result of time-varying active contraction causing diameter to reduce and is limited by segmental viscous and valvular resistance. The valves effect a smooth transition from low forward-flow resistance to high backflow resistance. Contraction occurs sequentially in successive lymphangions in the forward-flow direction. The behavior of chains of one to five lymphangions was investigated by means of pump function curves, with variation of valve opening parameters, maximum contractility, lymphangion size gradation, number of lymphangions, and phase delay between adjacent lymphangion contractions. The model was reasonably robust numerically, with mean flow-rate generally reducing as adverse pressure was increased. Sequential contraction was found to be much more efficient than synchronized contraction. At the highest adverse pressures, pumping failed by one of two mechanisms, depending on parameter settings: either mean leakback flow exceeded forward pumping or contraction failed to open the lymphangion outlet valve. Maximum pressure and maximum flow-rate were both sensitive to the contractile state; maximum pressure was also determined by the number of lymphangions in series. Maximum flow-rate was highly sensitive to the transmural pressure experienced by the most upstream lymphangions, suggesting that many feeding lymphatics would be needed to supply one downstream lymphangion chain pumping at optimal transmural pressure.  相似文献

Isolated polytene chromosomes were stretched in a 0.125 M NaCl solution with constant speed, by constant force and by cyclically changing force. For each regime, the dependence of chromosome length on the time and force magnitude were recorded. From this it may be concluded that three processes are involved in chromosome stretching: viscoelastic deformation, viscous flow of DNP segments, and cristallization, i.e. intermolecular cross-linking of neighbour segments. At a high rate stretching (V greater than Vo) chromosome may be torn like at small deformation; when rate is V greater than Vo chromosome deformation is mostly viscoelastic; at rates V approximately Vo viscous flow of DNP segments if predominant. We estimate Vo approximately less than 3--6 mum/s. Electron microscopy shows that during chromosome stretching its DNP fibers are oriented along chromosome axis without detectable breaks.  相似文献

The distribution in the spinal cord of the trigeminal primary projections in the frog Rana ridibunda was studied by means of the anterograde transport of horseradish peroxidase (HRP). Upon entering the medulla via the single trigeminal root, a conspicuous descending tract that reaches the cervical spinal cord segments is established. This projection arises in the ophthalmic (V1), maxillary (V2), and mandibular (V3) trigeminal nerve subdivisions. In the spinal cord, only a minor somatotopic arrangement of the trigeminal fibers was observed, with the fibers arising in V3 terminating somewhat more medially than those from V1 and V2. A dense projection to the medial aspect of the spinal cord, above the central canal, primarily involves V3. Each trigeminal branch sends projections at cervical levels to the contralateral dorsal field, and those from V2 are most abundant. Bilateral experiments with HRP application show convergence of primary trigeminal and spinal afferents within the dorsal field of the spinal cord. The pattern of arrangement of the trigeminal primary afferent fibers in the spinal cord of this frog largely resembles that of amniotes. However, the organization seems simpler and the slight somatotopic distribution of V1, V2, and V3 fibers is similar to the condition in other anamniotes. © 1993 Wiley-Liss, Inc.  相似文献

Topical application of sodium nitroprusside on rat mesentery has a marked influence on lymph microvessels function. The drug causes a dilation of majority of lymphangions and decrease of the pacemaker activity of the vessel wall structures and valves. These changes do not lead to lymph stasis, and lymph flow velocity does not reduce. The non-selective inhibitor of NO synthase (N-nitro-L-arginine) intensifies vasomotions of lymph microvessels, modulates phasic contractile activity and increases lymph flow velocity. There is a time dependent dynamic of changes in action of N-nitro-L-arginine. During inhibition of endogenous NO synthesis the responses of lymph microvessels on sodium nitroprusside application are modified: the constriction of majority lymphangions and activation of valve work are observed.  相似文献

50 dissections of the human inferior V. cava have been performed in order to measure its right renal vein - diaphragm, retrohepatic, and suprahepatic segments. We conclude that some individual parameters as skin type, age, height, weight did not influence the magnitude of the studied segments. The average measurements of the different parameters proposed for the inferior V. cava are: 1. The distances between the right renal vein and the diaphragm and between the right renal vein and the right atrium are 113.94 mm and 135.16 mm, respectively; 2. the length of the retrohepatic portion of the inferior V. cava and the suprahepatic one were 78.34 mm and 19.34 mm respectively; 3. the valve of the inferior V. cava is present in 46% of the observations; its length and width averages are 31 mm and 10.22 mm, respectively.  相似文献

In the human T-cell rearranging gamma (TRG gamma) locus, five joining (J) segments have been identified: J1, J2 and three additional segments JP, JP1 and JP2. We report the sequence of the germline JP1 segment and compare it with the other human and mouse J gamma segments. We also demonstrate that rearrangements to the three additional J gamma segments can be identified by hybridization of the KpnI digests to the J gamma 1 probe pH60. Since rearrangements to J1 or J2 can be assigned, using the same pH60 probe, to one of the nine variable (V) gamma genes known to rearrange [(1987) EMBO J. 6, 1945-1950], our results show that a unique probe can detect all the TRG gamma rearrangements and be particularly useful for assessing the preferential usage of V gamma and J gamma segments in the TRG gamma-expressing cells.  相似文献

An existing lumped-parameter model of multiple lymphangions (lymphatic vascular segments) in series is adapted for the incorporation of recent physiological measurements of lymphatic vascular properties. The new data show very marked nonlinearity of the passive pressure–diameter relation during distension, relative to comparable blood vessels, and complex valve behaviour. Since lymph is transported as a result of either the active contraction or the passive squeezing of vascular segments situated between two one-way valves, the performance of these valves is of primary importance. The valves display hysteresis (the opening and closing pressure drop thresholds differ), a bias to staying open (both state changes occur when the trans-valve pressure drop is adverse) and pressure-drop threshold dependence on transmural pressure. These properties, in combination with the strong nonlinearity that valve operation represents, have in turn caused intriguing numerical problems in the model, and we describe numerical stratagems by which we have overcome the problems. The principal problem is also generalised into a relatively simple mathematical example, for which solution detail is provided using two different solvers.  相似文献

The aim of this study was to evaluate changes in heart rate (HR), QT and RR intervals and corrected QT (QTc) values in conscious male and female New Zealand rabbits which intravenously received oxytocin (OXT) at different dosages. Animals were divided into 6 equal groups: group I (n = 6 male, received 0.75 U OXT per animal); group II (n = 6 male, received 1.5 U OXT per animal); group III (n = 6 male, received 3 U OXT per animal); group IV (n = 6 female, received 0.75 U OXT per animal); group V (n = 6 female, received 1.5 U OXT per animal); group VI (n = 6 female, received 3 U OXT per animal). ECG recording were taken from all animals before injection and then at 2, 4, 6, 8, 10, 15 and 20 min of OXT administration. QT and RR intervals obtained at 2 min of OXT administration were significantly prolonged in all groups (p < 0.05) with one exception that is the 1.5 U OXT injected female group where only QT interval did not change. The prolongation of QT and RR intervals persisted for 20 min in 1.5 U OXT injected male group while only QT interval prolongation was obvious for 20 min in 3 U OXT injected female group as for the other groups the prolonged interval were observed for 8-10 min and then returned to baseline values. Generally, a significant prolongation of QTc was noticed in both male and female rabbits at the 2 and 4 min in all groups and bradycardia was noticed at 2 min of OXT administration in all groups. Heart beats returned to normal values in all groups after 8 min of OXT administration. The change of HR, RR, QT and QTc was gender- but not dose-dependent (p < 0.001). The male rabbits were more sensitive to OXT effect then female rabbits. In conclusion, OXT used in therapeutic dosages decreased heart rate and prolonged QT and QTc intervals. Although cardiovascular effect of OXT are of short duration, its use in patient with risk factors for malignant arrhythmias requires more attention.  相似文献

The optic fibers in the retinas of diverse species may be selectively stained and viewed en bloc in the embryonic and adult states. Treat the eye as follows: 1) 50% pyridine for at least 16 hr, 2) distilled water 3--4 hr, 3) 20% H2O2 until the eye is a light brown, 4) 95% ethanol overnight, 5) 1.5% AgNO3 for 2--6 days at 37 C, 6) in water, remove the vitreous, then direct 0.25% pyrogallic acid in 1.25% formalin against the retina for 2--5 secs until the optic fibers are reduced to a coffee-copper color (1--4 minutes), 7) dissect the retina and mount flat on a glass slide, 8) cover with glycerin, apply a coverslip, and fix in place with nail polish. Variants for particular species are given. The technique offers an advantage over Golgi and methylene blue methods which tend to stain only a small percentage of fibers and frequently do not work at the earliest stages of development.  相似文献

We examined the inhibitory activity of type V collagen on cell attachment and cell growth and the role of stress fibers and beta 1 integrin in cultured human endothelial cells. Human endothelial cells cultured on type V collagen attached temporarily to the substrate and formed stress fibers. However, the cells failed to proliferate and gradually detached from the substrate. After 24 h, the cells on type V collagen lacked discernible stress fibers (F-actin filaments) and exhibited dots in small aggregates of F-actin. In addition, the cells expressed little or no proteins as focal adhesions, including vinculin and beta 1 integrin. In contrast, the cells on fibronectin and type I collagen formed complete F-actin filaments, exhibited sufficient vinculin and beta 1 integrin, and grew logarithmically from 2 days. On the other hand, human smooth muscle cells formed complete F-actin filaments, revealed typical focal adhesions, and started to proliferate rapidly after 24 h on type V collagen as well as on fibronectin and type I collagen. Thus, the disassembly of F-actin filaments was observed as a specific phenomenon in human endothelial cells cultured on type V collagen. Moreover, the F-actin filaments disappeared from endothelial cells treated with cytochalasin D after 24 h and the cells detached from fibronectin and type I collagen with time, a result consistent with the observations on type V collagen. Accordingly, the disassembly of F-actin filaments in focal adhesions may result in the detachment of endothelial cells from type V collagen.  相似文献

Lymphatic vessels of the small intestine mesentery have been investigated in 36 human fetuses 3-6-month-old. Lymphatic postcapillaries are the predominant link in the lymphomicrocirculatory bed studied. They differ from the capillaries only by presence of valves. Intersegmentary valves and velves at the places where the lymphatic capillaries get into the postcapillaries are described; they are situated along the whole length of the postcapillary. Both types of the valves can be uni- and besinusoid, and according to the number of endotheliocytes, covering the sinuses, uni- and multicellular. Cells of fibroblastic line and developing collagenous fibers are inevitable structural components and situate in their base. Ultrastructural peculiarities of the lymphatic postcapillary wall are described. Structural mechanisms responsible for dilatation and narrowing of the vascular lumen (formation of the pulse wave) are discussed. The data obtained support the conception successively elaborated by V. V. Kuprianov on the lymphatic postcapillary as an inevitable link of the lymphomicrocirculatory bed.  相似文献

Ventricular contraction occurred simultaneously over the shole chamber. The passage of blood into the posterior aorta could be restricted by the protuberances on its wall. 5. Pericardial cavity pressure rose by about 5 cm H2O at shell valve adduction and 0-25--0-6 cm H2O at ventricular diastole. 6. The structure of the nerve fibres in the intact and neurally isolated intestine of the cat was studied after 6-OHDA treatment by fluorescence and electron microscopy. A gradual disappearance of green fluorescence in the monoaminergic fibers was observed. Degeneration of nerve fibres containing 30--60 nm agranular and 30--60 nm densecore (granular) vesicles could be seen in all layers of the small intestine. Sporadically, degeneration of the presynaptic elements occurred also in the synapses of myenteric ganglia. A large number of degenerating fibres could be observed in close relation to blood vessels. These were thought to be monoaminergic because 6-OHDA selectively affected their terminals. In the chronically isolated small intestine degenerated nerve processes could not be observed following 6-OHDA treatment. It is concluded that the intrinsic nerve elements do not contain monoamines. Accordingly, the previously observed yellow fluorescence of the nerve elements of the isolated small intestine might be due to some other kind of neurotransmitter, possibly tryptamine.  相似文献

Three minor sulfur-containing arsenic metabolites: monomethylmonothioarsonic acid (MMMTA(V)), dimethylmonothioarsinic acid (DMMTA(V)), and dimethyldithioarsinic acid (DMDTA(V)) were recently found in human and animal urine after exposure to inorganic arsenic. However, it remains unclear how the thioarsenicals are formed in the body and then excreted into the urine. It is hypothesized that the generation of thioarsenicals occurs during enterohepatic circulation. To address this hypothesis, male Sprague Dawley (SD) rats and Eisai hyperbilirubinuric (EHB) rats (with deficiency of multidrug resistance-associated protein 2) were orally administered a single dose of inorganic arsenite (iAs(III)) at 3.0 mg kg(-1) of body weight. Five hours after dosing, less than 1.0% of the dose was recovered in the bile of EHB rats, while more than 27% of the dose was recovered in the bile of SD rats, with the majority being monomethylarsinodiglutathione [MMA(SG)(2)] with a small amount of arsenic triglutathione [iAs(SG)(3)]. During the early time periods (3 h and 6 h) the arsenic levels in the liver, red blood cells (RBCs) and plasma of EHB rats were higher than those of SD rats, and approximately 76% and 87% of the dose was recovered in the RBCs of SD and EHB rats, respectively, at day 5 after dosing. However, there were no significant differences in arsenic concentration in urine between the two types of animal. Regarding the arsenic species in the urine of both types of rat, significant levels of thiolated arsenicals MMMTA(V) and DMMTA(V) were detected in SD rat urine, however in EHB rat urine only low levels of DMMTA(V) were detected. The present result of the metabolic balance and speciation study suggests that the formation of MMMTA(V) and DMMTA(V) in rats is dependent on enterohepatic circulation. In addition, in vitro experiments indicated that arsenicals excreted from bile may be transformed by gastrointestinal microbiota into MMMTA(V) and DMMTA(V), which are then absorbed into the bloodstream and finally excreted into the urine.  相似文献

Background

There is a paucity of information on structural organization of muscular bundles in the interatrial septum (IAS). The aim was to investigate histologic and ultrastructural organization of muscular bundles in human IAS, including fossa ovalis (FO) and flap valve.

Methods

Macroscopic and light microscopy evaluations of IAS were performed from postmortem studies of 40 patients. Twenty three IAS specimens underwent serial transverse sectioning, and 17 - longitudinal sectioning. The transverse sections from 10 patients were immunolabeled for HCN4, Caveolin3 and Connexin43. IAS specimens from 6 other patients underwent electron microscopy.

Results

In all IAS specimens sections the FO, its rims and the flap valve had muscle fibers consisting of working cardiac myocytes. Besides the typical cardiomyocytes there were unusual cells: tortuous and horseshoe-shaped intertangled myocytes, small and large rounded myocytes with pale cytoplasm. The cells were aggregated in a definite structure in 38 (95%) cases, which was surrounded by fibro-fatty tissue. The height of the structure on transverse sections positively correlated with age (P = 0.03) and AF history (P = 0.045). Immunohistochemistry showed positive staining of the cells for HCN4 and Caveolin3. Electron microscopy identified cells with characteristics similar to electrical conduction cells.

Conclusions

Specialized conduction cells in human IAS have been identified, specifically in the FO and its flap valve. The cells are aggregated in a structure, which is surrounded by fibrous and fatty tissue. Further investigations are warranted to explore electrophysiological characteristics of this structure.  相似文献

Summary We have investigated the connectivity of four classes of mechanosensory afferents to giant interneurons in the earthwormLumbricus. Three of these classes of afferents change their specification for connection to medial giant (MGF) and lateral giant (LGF) fibers along the length of the animal. Near the caudal end, stimulation of touch, pressure and small tactile fibers generates excitatory post-synaptic potentials, epsp's, in the two LGF's but not in the MGF. Near the rostral end these afferents produce much smaller epsp's in the LGFs but produce large epsp's in the MGF. In the middle region of the animal an overlap region exists where both giant fibers receive approximately equal inputs from these afferents. The amplitude of these inputs are reduced compared to the maxima seen at either end. The fourth class of sensory afferents investigated, the stretch neurons, have no synaptic effect on the giant fibers anywhere in the nerve cord.These results explain at least part of the basis, in neuronal connectivity, for the differences in response to tactile stimulation of the head and tail segments previously characterized in terms of behavior and giant fiber impulse activity. 为了探究三峡工程建成运行后洞庭湖区洲滩生境中小型兽类的群落状况,对洞庭湖区洲滩生境的小型兽类进行调查。调查采用铗日法,于2010至2018年,对洞庭湖区洲滩小型兽类群落的多样性进行了调查,并计算优势度、群落多样性指数、均匀性指数。对多样性指数采用t值法进行比较,各年份、生境的捕获率及鼠种组成差异则采用卡方检验。2010至2018年,共布放34 116铗日,捕获可以分辨种类的样本2 129只,主要有啮齿目(Rodentia)和鼩形目(Soricomorpha)2类,共7种,分别是啮齿目的东方田鼠(Microtus fortis)、黑线姬鼠(Apodemus agrarius)、褐家鼠(Rattus norvegicus)、巢鼠(Micromys minutes)、社鼠(Niviventer confucianus)、黄毛鼠(R. losea)和鼩形目的臭鼩(Suncus murinus)。总捕获率为6.36%,东方田鼠、黑线姬鼠捕获率较高,分别为1.83%、3.94%。从小型兽类组成看,黑线姬鼠和东方田鼠在群落中组成的比例较高,分别为63.18%和29.26%。在调查的5类生境中,植被组成以苔草(Carex sp.)、南荻(Miscanthus sp.)和美洲黑杨(Populus deltoides)为主的生境小型兽类群落多样性指数(1.0772)和均匀度指数(0.6662)均最大,其次是以苔草为主的生境(多样性指数和均匀度指数分别为0.9275、0.5723)、以南荻为主的生境(多样性指数和均匀度指数分别为0.8856、0.550 2)和以苔草加南荻为主的生境(多样性指数和均匀度指数分别为0.775 6、0.481 9),以美洲黑杨为主的生境多样性指数(0.323 6)和均匀度指数(0.180 6)最低,但其优势度最高,为0.881 2。结果表明,随着三峡工程的运行,洞庭湖区植被演替发生了变化,从而引起小型兽类群落动态的变化。在洞庭湖洲滩上,生境越复杂,小型兽类物种多样性越高。单一生境下,人类活动干扰程度越大,小型兽类物种多样性越低。目前,黑线姬鼠已经成为洞庭湖洲滩生境的第一优势鼠种。  相似文献

As part of an investigation of the immune system in the developing zebrafish, Danio rerio, we cloned and characterized a genomic V(H) segment and a number of cDNAs encoding IgM heavy chains. The genomic V(H) has the characteristic features of V(H) in other vertebrates, including a leader segment interrupted by a short intron, and regulatory and recombination signal sequences. The V(H) sequences of 75 cDNA clones fell into four distinct groups or families. All of these families cluster most closely with other teleost V(H); one of the families, V(H)3, is also closely related to other vertebrate V(H). Analysis by Southern blotting suggests that there are a number of germline genes corresponding to each family. Most variability within the V(H)1 family is confined to codons 30-32, at the end of the first framework region and in the beginning of the first complementarity-determining region. The nucleotide sequence of the genomic V(H) in this region contains motifs associated with a relatively high frequency of somatic mutation. There is much variability in segments of the clones corresponding to the third complementarity-determining region. The cDNA sequences encoding the fourth framework region are consistent with the contribution of four distinct J(H) segments.  相似文献

Maximum velocity of the actomyosin ATPase reaction (V(max) ATPase) and ATP consumption rate during maximum isometric activation (ATP(iso)) were determined in human vastus lateralis (VL) muscle fibers expressing different myosin heavy chain (MHC) isoforms. We hypothesized that the reserve capacity for ATP consumption [1 -- (ratio of ATP(iso) to V(max) ATPase)] varies across VL muscle fibers expressing different MHC isoforms. Biopsies were obtained from 12 subjects (10 men and 2 women; age 21--66 yr). A quantitative histochemical procedure was used to measure V(max) ATPase. In permeabilized fibers, ATP(iso) was measured using an NADH-linked fluorometric procedure. The reserve capacity for ATP consumption was lower for fibers coexpressing MHC(2X) and MHC(2A) compared with fibers singularly expressing MHC(2A) and MHC(slow) (39 vs. 52 and 56%, respectively). Tension cost (ratio of ATP(iso) to generated force) also varied with fiber type, being highest in fibers coexpressing MHC(2X) and MHC(2A). We conclude that fiber-type differences in the reserve capacity for ATP consumption and tension cost reflect functional differences such as susceptibility to fatigue.  相似文献
