A sensitive and easy-to-perform instrumentational method for the detection of Escherichia coli O157 in raw minced beef is described. The detection is based on a light scattering immunoassay and a charge-coupled device (CCD) direct readout spectrometer measuring the scattered light spectral signals at an optimized angle of 20° to the axis of transmitted light. Using latex particles coated with antibodies for E. coli O157, the method sensitivity has significantly improved comparing with the visual immunoassay assessment method when detecting the presence of this bacterium in spiked beef samples. The method is capable of detecting E. coli O157 at the level of 103 cfu mL-1 after 6 h of incubation of the spiked samples. This study has demonstrated a faster technique (within 8 h) for the detection of E. coli O157 in raw beef and a possible new application for the CCD based light scattering instrument.  相似文献

本文从红细胞前向光散射的观点出发对红细胞的尺寸、红细胞的变形性研究以及红细胞容积、血红蛋白浓度等参数的测定作了系统的思考。提出在红细胞与周围悬浮介质折射车相差不大时,反常衍射比夫朗和费衍射更适合用于红细胞前向光散射的研究。利用两组不同角间隔的前向散射光来同时测量红细胞容积和血红蛋白浓度。同时其它一些标识红细胞的参数如平均红细胞容量(MCV)、平均血红蛋白量(MCH)等均可直接或间接由这两项参数导出。最后还将红细胞的光散射与Mie理论作了对比.  相似文献

用常规方法从患典型白底板病黄沙鳖的心脏、肝脏等处进行细菌的接种分离, 通过人工感染确定分离菌株的致病性, API 20NE、16S rRNA基因序列分析进行病原菌鉴定和确定其系统发育地位, K-B纸片扩散法进行药敏试验, PCR检测病原菌的6种毒力基因。试验结果, 共分离到13株病原菌, 其中嗜水气单胞菌9株, 温和气单胞菌4株。在9株嗜水气单胞菌中, 有5株与Aeromonas hydrophila ATCC 7966菌株亲源关系最近, 4株与Aeromonas hydrophila北京株QDC01的亲源关系最近; 而4株温和气单胞菌与Aeromonas sobria ATCC 43979的亲源关系最近。药敏试验结果, 仅头孢哌酮对13株病原菌都高度敏感, 来源于不同养殖区域的病原菌药敏结果相差较大。6种毒力基因的阳性率, Aer、Act和ahp均为100%, hly和Alt为92.31%, ahal为76.92%; 毒力基因型共有4种, 嗜水气单胞菌主要为hly+Aer+Alt+Act+ahal+ahp+基因型, 而温和气单胞菌主要为 hly+Aer+Alt-Act+ahal+ahp+基因型, 同时携带hly基因的菌株其致病力更强。    相似文献

为设计稻曲病菌(Ustilaginoidea virens)专化性PCR引物,测定了1991-2001年采集的多个水稻品种、不同水稻产区的菌株的ITS和5.8S rDNA区序列。U. virens的ITS1、ITS2和5.8S rDNA区域的长度为 624-625bp, 序列高度保守。在与麦角菌科其它种比较的基础上,设计了U. virens专化性嵌合引物。采用PCR方法可以灵敏地检测目标真菌,并且与传统的组织观察结果很好地吻合。这一结果为深入研究稻曲病的侵染规律和建立田间早期诊断技术提供了可能。  相似文献

动态光散射是研究生物大分子溶液物化行为和结晶性能的重要工具。以溶菌酶为模型蛋白 ,对其溶液的DLS研究表明 ,一定范围的蛋白质浓度不会影响蛋白质聚合性质 ,而起沉淀剂作用的盐对多色散性的影响较大。考虑这些研究结果 ,应用DLS研究对空间微重力下晶体生长样品的质量做了考察 ,这对于提高空间样品准备水平 ,进而提高空间实验成功率是一重要途径。  相似文献

The article presents the neural methods of calibration of gas sensors for use in an artificial electronic nose for gas measurements. Different neural network solutions will be presented and compared. They include the classical multilayer perceptron, neuro-fuzzy networks and support vector machines. The other aspect illustrated in the article is the introductory preprocessing of the measured sensor signals in order to attain the highest possible efficiency of the gas measuring system. The theoretical considerations will be supported by the numerical experiments concerning the application of the electronic nose. The first practical aspect is concerned with the application of the developed system for classification problems and will be illustrated in the examples of the recognition of the biocomponents in the gasoline and the recognition of smells of cosmetic cream at the aging process. The second one belongs to the estimation problem and is concerned with the determination of the concentration of the particular gas components in the mixture of gases.

PRACTICAL APPLICATIONS


The results presented in the article may find practical application for calibration of the electronic nose in gas measurements. The electronic nose is widely used for smell recognition. It may find practical application in the petroleum, cosmetics or food industry for the assessment of the quality of their products. Military application is also of great interest.  相似文献

本文应用新建立的快速微波-Envision两步法免疫组织化学技术,分别对福尔马林固定和石蜡包埋切片的良恶性肿瘤进行免疫组织化学标记,观察该法的敏感性和特异性。所有23种抗原均获得强阳性、低背景。整个程序在18分钟内完成。结果表明:该法是一种特异性强、敏感性高、背景染色轻、快速和简便的新方法,可用于常规病理诊断。  相似文献

1. It is generally agreed that the blackening of osmium tetroxide by unsaturated lipid is too unpredictable to demonstrate lipid in tissues.
2. At neutral pH osmium tetroxide combines with the double bonds in the lipoproteins of cellular membranes (mitochondria, etc.) and the deep colour reaction of ethyl gallate with this osmium provides good staining of lipid for the light microscope.
3. Osmium taken up by tissue proteins at neutral pH is only a small fraction of that taken up by the lipid. (After acid fixatives osmium tetroxide is a general protein stain.)
4. The uptake of Sudan black B by partition from dilute solution is a specific test for lipid, but in normally fixed tissue most of the structural lipid is 'bound' and is not accessible to the dye.
5. Cautious treatment of fixed tissue with dilute sodium hypochlorite will unmask this lipid for viewing by the light microscope.
6. Direct fixation with neutral osmium tetroxide is an effective method for visualizing lipid for the electron microscope (as in the ethyl gallate method for the light microscope). But the poor penetration of osmium limits its use in this way.
7. After formol/glutaraldehyde fixation much of the lipid in the tissues is 'bound' and does not take up osmium. It can be unmasked by a saturated aqueous solution of thymol.
8. The unmasked lipid can then be rendered more osmiophil by partition in a solution of the highly unsaturated terpene farnesol, thus increasing the uptake of osmium in a renewed application.
9. Some of the novel observations on tissue lipids made by these methods are reviewed.  相似文献

Two simple methods were followed to determine detection thresholds for the taste of substances in aqueous solution. The methods applied were: a modification of the ascending method of limits and a method based on the use of scales. Detection thresholds were calculated for the four basic tastes (sweet, salty, acid, and bitterness), umami and metallic. Reference substances for each taste were sucrose, sodium chloride, citric acid, caffeine, monosodium glutamate and iron (II) sulfate heptahydrate and the results of the two methods were compared. We found that the threshold values calculated by method ASTM-679 was within the range of concentrations identified with the scales method.  相似文献
