Background  

The underlying cellular and molecular mechanisms that coordinate the physiological processes in digestion are complex, cryptic, and involve the integration of multiple cellular and organ systems. In all intestines, peristaltic action of the gut moves food through the various stages of digestion from the anterior end towards the posterior, with the rate of flow dependent on signals, both intrinsic and extrinsic to the gut itself.  相似文献

The gastric evacuation time (GET) and the passage of food through the intestines was investigated in Sarotherodon mossambicus (Peters) fry of two size groups, viz. 0·2–1·0g and 1·0–3·0g, in response to five diets. In the experiments voluntary feeding was adopted and markers were not used. The passage of food through the stomach was described by the exponential model.
The GET increased with body weight for any particular diet, with one exception. The GET ranged from 290 min to 1104 min in fry of 1·0–3·0 g on Diet 2. There was evidence to indicate that the GET was negatively correlated to the specific gravity of the diet.
The passage of food through the intestines showed a consistent trend from diet to diet; e.g. the algal diet being moved into the intestine in 'finite' quanta as opposed to the others. The importance of these observations and the implications of the dependence of the GET on the specific gravity of the diet in the use of markers in digestibility studies are discussed.  相似文献

Background  

Gene transfer into the amniotic fluid using recombinant adenovirus vectors was shown previously to result in high efficiency transfer of transgenes into the lungs and intestines. Adenovirus mediated in utero gene therapy, however, resulted in expression of the transgene for less than 30 days. Recombinant adenovirus associated viruses (rAAV) have the advantage of maintaining the viral genome in daughter cells thus providing for long-term expression of transgenes.  相似文献

Background

Helicobacter pylori is unevenly distributed in hypochlorhydric environments. The study aim was to elucidate the risk factors for a negative Giemsa staining finding in seropositive subjects by measuring the secretory ability of the stomach.

Methods

Subjects aged over 18 years were included consecutively after endoscopic biopsy at gastric lesions with color or structural changes. Blood was sampled for the serum pepsinogen (PG) assay and H. pylori serology test. After excluding the subjects with past H. pylori eradication, the risk factors for a negative Giemsa staining finding in seropositive subjects were analyzed.

Results

Among 872 included subjects, a discrepancy between the serum anti‐H. pylori IgG and Giemsa staining findings was found in 158 (18.1%) subjects, including 145 Giemsa‐negative, seropositive subjects. Gastric adenocarcinoma/adenoma (OR = 11.090, 95% CI = 3.490‐35.236) and low serum PG II level (OR = 0.931, 95% CI = 0.899‐0.963) were the independent risk factors for a negative Giemsa staining finding in seropositive subjects. The cutoff value of serum PG II level was 7.45 ng/mL (area under curve [AUC] = 0.904, 95% CI = 0.881‐0.927). Follow‐up studies of Giemsa staining at different sites of the stomach revealed that 75% of the Giemsa‐negative seropositive subjects with adenocarcinoma are positive, whereas none of those with low serum PG II level of <7.45 ng/mL revealed positive findings.

Conclusions

The risk of a negative Giemsa staining finding in seropositive subjects is increased in gastric adenocarcinoma/adenoma specimens and in subjects with a diminished gastric secretory ability with low serum PG II level of <7.45 ng/mL. A false‐negative Giemsa staining finding is common in subjects with adenocarcinoma, and therefore, additional biopsies at different sites should be performed in these subjects.  相似文献

Clinical isolates of the gastric pathogen Helicobacter pylori display a high level of genetic macro- and microheterogeneity, featuring a panmictic, rather than clonal structure. The ability of H. pylori to survive the stomach acid is due, in part, to the arginase-urease enzyme system. Objectives

This study has intended to investigate longevity of subcutaneous fat‐derived mesenchymal stem cells (SF‐MSCs) under extensive culturing. It has also focused on optimization of culture media for them over prolonged periods in vitro.

Materials and methods

We evaluated SF‐MSCs with reference to phenotypic characterization, proliferative ability, karyotype stability and differentiation potency with early (P3) and late passage (P20) conditions, using four different media, DMEM‐LG, ALPHA‐MEM, DMEM‐F12 and DMEM‐KO.

Results

This study unravels retention of SF‐MSC characteristics in facets of phenotypic expression profile (CD 90, CD 105, CD 73, CD 34, CD 29, CD 54, CD 49d, CD 117, HLA‐DR, CD 166, CD 31, CD 44), proliferative characteristics, karyotyping and differentiation potency prolonged culturing to P25 in all media. Population doubling time (PDT) in Alpha MEM, DMEM LG, DMEM F 12, DMEM KO were identified to be (1.81, 1.84, 1.9, 2.08 days) at early passage and (2.93, 2.94, 3.12, 3.06 days) at late passage. As a corollary, Alpha MEM and DMEM LG serve as appropriate basal media for SF‐MSC when proliferative potency is considered.

Conclusions

In research, it is imperative that SF‐MSC uphold their expansion potency in the aforesaid attributes in all media over extensive culturing, thereby transforming their colossal in vitro potency, with the aim of curing a wide horizon of diseases.  相似文献

Background  

The Blood Brain Barrier (BBB) maintains the homeostasis of central nervous system by preventing the free passage of macromolecules from the systemic circulation into the brain. This normal physiological function of the BBB presents a challenge for delivery of therapeutic compounds into the brain. Recent studies have shown that the application of focused ultrasound together with ultrasound contrast agent (microbubbles) temporarily increases the permeability of the BBB. This effect is associated with breakdown of tight junctions, the structures that regulate the paracellular permeability of the endothelial cell layer. The influence of this ultrasound effect on the activation of intracellular signaling proteins is currently not well understood. Potential therapeutic use of mesenchymal stem cells (MSCs) is likely to require large-scale in vitro expansion of the cells before transplantation. MSCs from adipose tissue can be cultured extensively until senescence. However, little is known on the differentiation potential of adipose stem cells (ASCs) upon extended culture and on associated epigenetic alterations. Background

The present study was carried out to determine whether the p53 pathway played a role in the spontaneous immortalization of the SC-2 chicken embryo fibroblast (CEF) cell line that has been in continuous culture for over three years.

Results

The SC-2 cell line emerged from an extended crisis period with a considerably slower growth rate than primary CEF cells. The phenotype of the SC-2 cells changed dramatically at about passage 80, appearing smaller than at earlier passages (e.g., passage 43) and possessing a small, compact morphology. This morphological change coincided with an increase in growth rate. Passage 43 SC-2 cells expressed undetectable levels of p53 mRNA, but by passage 95, the levels were elevated compared to primary passage 6 CEF cells and similar to levels in senescent CEF cells. However, the high level of p53 mRNA detected in passage 95 SC-2 cells did not correlate to functional protein activity. The expression levels of the p53-regulated p21WAF1 gene were significantly decreased in all SC-2 passages that were analyzed. Examination of the Rb pathway revealed that E2F-1 and p15INK4b expression fluctuated with increasing passages, with levels higher in passage 95 SC-2 cells compared to primary passage 6 CEF cells.

Conclusion

The present study suggests that altered expression of genes involved in the p53 and Rb pathways, specifically, p53 and p21WAF1, may have contributed to the immortalization of the SC-2 CEF cell line.  相似文献

Ghrelin is a 28-amino acid octanolyated peptide, synthesised primarily in the stomach. It stimulates growth hormone release, food intake and exhibits many other diverse effects. Our group have previously determined that ghrelin inhibited human contractility in vitro. The aim of this study therefore, was to investigate the expression of ghrelin, its receptor, the growth hormone secretagogue receptor type 1 (GHS-R1), ghrelin O-acyltransferase (GOAT) which catalyses ghrelin octanoylation, prohormone convertase 1/3 (PC1/3) responsible for pro-ghrelin processing, in human myometrium, during pregnancy prior to labour, during labour and in the non-pregnant state. Young rabbits consume milk, feed and soft faeces during the pre-weaning period, but the biological rhythm of intake were scarcely described. This work aimed to analyse the behaviour of ingestion and caecotrophy by analysing the stomach and rectal contents of young rabbits. Chronological samplings were performed according to a 4 × 3 factorial experimental design (17, 22, 28 and 35 days at 3, 8 and 23 h after suckling, respectively). Both digestive contents were examined and scored according to a discrete 4-step scale. Milk intake peaked around 22 days (26.6 g/rabbit) and decreased by 50% at 28 days. Stomach score for milk content showed a negative relationship with age. Dry feed intake reached 7.2 g/rabbit at 22 days, but was relatively variable among litters (CV = 44%). After 22 days, nutrient intake from the solid feed exceeded that of the milk. A higher pellet intake was observed during the night period (18:00–09:00 h) whatever the age. Dry feed in the stomach content was affected by age (17 versus 22, 28 and 35 days). From 22 days, milk and feed components were particularly distributed in the stomach: the coagulated milk was close to the wall of the fundus, whereas the feed was homogeneously spread. Soft faeces were not found until 28 days of age, and were located in the fundic region (25–30% of stomach profile). Stomach weight increased almost threefold from 17 to 35 days of age, but showed a slower development than the rest of the body from 22 to 35 days (relative stomach weight from 1.65 to 1.38%). High pH values (5.5–3.0) were found in the fundus at 3 and 8 hours after suckling. The pH in the antrum decreased from 4.19 to 1.90 at 17 and 22 days, respectively. The excretion of hard and soft faeces was indirectly recorded by checking the content of distal colon and rectum. From 22 days onwards, the amount of hard faeces in distal colon and rectum reached a peak at 8 h after suckling (2.3 ± 0.2). On the contrary, soft faeces were only observed at 3 and 23 h after suckling from 28 days of age.  相似文献
