Summary In the teleost fish, Gillichthys mirabilis, vasoactive intestinal polypeptide (VIP)-like immunoreactivity is present in some nerve fibres of the principal pancreatic islet and surrounding tissues, the vagus and splanchnic nerves, the coeliac ganglion and the wall of the intestine. The nerves of the pancreatic islet that contain VIP-like immunoreactivity probably correspond to one of the two types of non-cholinergic, non-adrenergic (peptidergic) nerve previously described in this fish. Similarities in the localisation of hormonal peptides in fish and mammals suggest that the regulation of gastroenteropancreatic physiology in fish may resemble that of mammals.This work was supported by grants from the Medical Research Council of Great Britain and the National Institutes of Health, USA (Grant No. AM 17161)  相似文献

Summary The principal pancreatic islets of the teleost Scorpaena scropha are found ultrastructurally to contain four different kinds of parenchymal cells, viz. 1-(= D), 2-, -and agranular cells. The -cells show considerable variations in the shape of the secretory granules. A peculiar feature is that many of these granules are composed of fibrillar subunits, often in parallel arrangement. All -granules are surrounded by membranes and between the membrane and the granule core there is a moderately wide electron lucent space. The electron density of the cytoplasm in the -cells varies somewhat. The 2-cells possess typical secretory granules with an electron dense core and a closely applied membrane. The secretory granules in the 1-cells show also a closely applied membrane but a less dense core. Also in the -cells the electron opacity of the cytoplasm varies. The agranular cells are mainly characterized by low cytoplasmic electron density, narrow cisterns of endoplasmic reticulum and sometimes a laminated Golgi complex. Small immature secretory granules are occasionally seen in the cytoplasm of these cells. Summary Immunocytochemical staining has revealed that the pancreatic islets of various teleost fishes contain a pancreatic polypeptide (PP)-like substance which cross-reacts with antibodies to mammalian and avian PP. The PP cells were frequently found at the periphery of the islets, as in mammals, and were of irregular shape.Dr. J.R. Summary Light- and electron-microscopic autoradiographic studies of pituitaries of the molly Poecilia latipinna, after their incubation with tritiated serotonin, revealed the presence of labelled cells in the proximal pars distalis, together with cell processes or nerve fibres throughout all regions of the gland except the prolactin cell zone. The serotonincon-centrating cells and most of the fibres contained small dense-cored vesicles, but some labelled fibres contained larger granules similar in ultrastructure to those of vasotocinergic fibres.This work was supported by a SERC studentship to DJG and SERC grant GR/6379-06 to Professor J.N. Retinal extracts have been prepared from dark-adapted mudsuckers by treatment of retinal tissue or of isolated outer segments of the visual cells with digitonin solution. The extracts were examined spectrophotometrically and found to absorb light maximally between the wave lengths of 488 and 510 mµ, depending on the proportion of yellow impurities and light-sensitive pigment present. This photosensitive pigment was shown to be homogeneous by partial bleaching of the extracts with monochromatic light of various wave lengths from 390 to 660 mµ. The mudsucker pigment was specifically demonstrated not to be a mixture of rhodopsin and porphyropsin; the adequacy of the method used to analyze such mixtures was shown by performing a control experiment with an artificial mixture of bullfrog rhodopsin and carp porphyropsin. Comparison of the hydroxylamine difference spectrum and of the absorption maximum of the purest retinal extract located the mudsucker photosensitive pigment maximum at 512 ± 1 mµ. Extraction of retinal tissue with a fat solvent after exposure to white light gave a preparation which after the addition of antimony chloride reagent developed the absorption band maximal near 664 mµ, which is characteristic of retinene1. If an hour intervened between exposure of the retinal tissue to light and extraction of the carotenoid, the antimony trichloride test gave a color band maximal at 620 mµ, characteristic of vitamin A1. No evidence of retinene2 or vitamin A2 was obtained. The euryhaline mudsucker has, therefore, a photosensitive retinal pigment with an absorption maximum halfway between the peaks of rhodopsins and of porphyropsins and belonging to the retinene1 system characteristic of rhodopsins. The pigment is therefore named a retinene1 pigment 512 of the mudsucker, Gillichthys mirabilis. It is uncertain whether this type of photosensitive pigment will be found in other euryhaline fishes.  相似文献

Summary Morphological changes in the adrenergic innervation of pancreatic islets after chemical sympathectomy by use of 6-hydroxydopamine and the influence of the sympatho-adrenal system on insulin secretion were investigated in the mouse and rat.Fluorescence histochemistry revealed a clear-cut reduction in the number of adrenergic nerve fibers in the pancreatic islets 2 days after administration of 6-hydroxydopamine; the reduction was more pronounced in the rat than in the mouse. In the rat, a partial regeneration was seen after 6 weeks. In the pancreas of the mouse, after administration of 6-hydroxydopamine, a severe damage of unmyelinated nerve fibers was revealed electron microscopically. However, no ultrastructural or immunohistochemical alterations could be demonstrated in the endocrine cells of the islets.6-Hydroxydopamine induced a depression of basal plasma insulin concentrations in mice and an elevation in rats. Adrenalectomy depressed basal plasma insulin levels in mice.The -adrenoceptor antagonist phentolamine enhanced insulin secretion in normal mice. The secretory response of insulin to phentolamine was diminished by chemical sympathectomy and almost abolished by adrenalectomy or the combination of chemical sympathectomy and adrenalectomy. Thus, the effect of phentolamine is probably mediated by liberated catecholamines.It is concluded that basal insulin secretion is partially regulated by the sympatho-adrenal system and that species differences exist in this respect. In addition, the results suggest that endogenous catecholamines have the ability to promote insulin secretion.  相似文献

We have examined whole-cell K+ currents and a Ca2+-dependent K+ channel at the single channel level in rostral pars distalis cells of Gillichthys mirabilis. Whole-cell K+ currents activated by depolarizing pulses have an inactivating component and a sustained component. The magnitude of both of these components is increased when a hyperpolarizing prepulse is delivered prior to depolarization. Both components are partially blocked by application of 5 mM TEA+. The Ca-dependent K+ channel, (K(Ca)), was sensitive to 2 mM TEA+ in outside-out patches (O/O) but not in inside-out patches (I/O). Channel open probability (P(o)) was dependent on membrane potential (Vm), with depolarization leading to an increase in P(o). Calcium on the cytoplasmic face of I/O patches increased channel P(o) in a dose-dependent manner. A portion of the single K(Ca) channels studied displayed inactivation after depolarizing pulses. These channels may be a component of the inactivating whole-cell current.  相似文献

Succinate dehydrogenase activity was measured in rat pancreatic islet homogenates incubated in the presence of [1,4-14C]succinate, the reaction velocity being judged through the generation of 14CO2 in the auxiliary reactions catalysed by pig heart fumarase and chicken liver NADP-malate dehydrogenase. In the presence of 1·0 mM succinate, the reaction velocity averaged 5·53 ± 0·44 pmol min?1 μg?1 islet protein. The Km for succinate was close to 0·4 mM and the enzymic activity was restricted to mitochondria. These kinetic results indicate that, under the present experimental conditions, the activity of succinate dehydrogenase does not vastly exceed that of either NAD-isocitrate dehydrogenase or the 2-ketoglutarate dehydrogenase complex, at least when the latter enzymes are activated by ADP and/or Ca2+. Nevertheless, the activity of succinate dehydrogenase is sufficient to account for the increase in O2 uptake evoked in intact islets by the monomethyl ester of succinic acid. It could become a rate-limiting step of the Krebs cycle in models of B-cell dysfunction.  相似文献

We have previously described a preferential reduction in the secretory response to nutrient secretagogues in pancreatic mouse islets maintained in culture after in vitro exposure to streptozotocin (SZ). This reduction was associated with an impaired substrate metabolism at the mitochondrial level. To further clarify this issue, mouse pancreatic islets were exposed in vitro to 2.2 mM SZ for 30 min. At 4 h after SZ treatment ultrastructural changes were apparent in the endoplasmic reticulum and Golgi areas of the B-cells. However, 2 and 6 days following SZ exposure the B-cells appeared well preserved, except for a marked decrease in the number of insulin-containing secretory granules. A morphometric analysis of the B-cells 6 days after SZ exposure showed a normal B-cell size and a normal volume fraction of B-cell mitochondria. However, there was a decrease in total islet size and a 13% decrease in the volume fraction of B-cells in the islets. These mouse islets exhibited a decreased content of the mitochondrial DNA-encoded cytochrome b mRNA, as evaluated by dot-blot analysis. As a whole, the data obtained indicate that SZ treatment does not induce a decrease in the number of mitochondria or long-lasting ultrastructural damage to this organelle. However, there is a clear decrease in the cytochrome b mRNA, suggesting that SZ can induce damage to the mitochondrial DNA.  相似文献

Insulin is stored in pancreatic islets as a zinc-insulin complex, and stimulating the islets results in the release of insulin and zinc. Simulant pancreatic islet beads have been developed using agarose beads (50-250 micro m diameter) derivatized with iminodiacetic acid that have been loaded with zinc. A qualitative comparison of the simulant beads with pancreatic islets has been made by staining with dithizone and a zinc-binding fluorescent dye, TSQ. The binding capacity of simulant beads was determined to be 34 micro mol Zn(2+)/g of dried beads using anodic stripping voltammetry. Hydrochloric acid was used to release zinc from beads to mimic the secretion of insulin from pancreatic islets and a release profile was established. The simulant beads can be used to optimize the islet isolation process and reduce the use of real islets in method development.  相似文献

Summary The pancreatic islets ofCarassius carassius have been studied by electron microscopy. 1. Besides A-, B- and D-cells, two new cell types, the fourth and the fifth, have been identified. The fourth cell type is numerous; it occurs interposed among the other types of islet cells or in small clusters. The secretory granules (90–280 mg in diameter) are round or oval and usually with much lower electron density than α- and δ-granules. The secretory granules of the fifth type of cell (approximately 140–240 mμ in diameter) contain finely granular material and an electron dense core that is round or often tetra- or hexagonal. 2. The islet cells with clear cytoplasmic matrix generally contain large numbers of fine, agranular and cored vesicles 400–680 ? in diameter. They appear, in bead-like chains, or randomly scattered throughout the cytoplasm, or often clustered in aggregates close to the secretory granules and show evidence of incorporation into the secretory granules. The two types of vesicles may be formed by constriction or pinching-off of the tubular smooth endoplasmic reticulum.  相似文献

Summary By means of correlative light and electron microscopy, five pancreatic islet cell categories are described in the teleost fish, Xiphophorus helleri, each of which has specific light microscopic appearance and fine structure. Different histochemical techniques have been used, including immunofluorescence with antiporcine insulin and glucagon sera. In addition to B- and A1-cells, two categories of A2-cells have been observed, both reacting with antiporcine glucagon serum: A2-cells with round granules gave a positive reaction for tryptophan; A2-cells with crystalline granules gave a negative reaction with the same staining technique on the same section. The clear cells, the last category, were not specifically stained by any of the staining methods carried out in this investigation. Summary The fine structure of the collecting tubules of the trout and killifish kidney was studied. These tubules are surrounded by layers of smooth muscle cells which are commonly innervated. The nerve terminals contain synaptic vesicles and, occasionally, a few dense-cored granules as well. Capillaries occur in the connective tissue space between these smooth muscle cells and the collecting tubule. Epithelial cells of the collecting tubules contain abundant mitochondria and a well developed membrane system displaying parallel arrays, and were considered to be actively involved in the transport of materials. In the trout, the collecting tubules contain peculiar cells in addition to regular tubule cells. The fine structure of these peculiar cells is highly reminiscent of that of gill chloride cells. The significance of these findings may be summarized as follows: If the smooth muscles around the collecting tubule contract under neural influence, intratubular pressure may be increased and, thus affect glomerular filtration rate. The contraction of these muscles may also cause the collapse of peritubular capillaries, affecting the transport activity of tubule cells.  相似文献

Leptin suppresses basal insulin secretion from rat pancreatic islets   总被引:2,自引:0,他引:2  
The effects of leptin on insulin secretion from pancreatic islets of Sprague–Dawley rats were examined in vitro. In a basal glucose medium (5.5 mM), insulin secretion from isolated islets was significantly decreased after addition of a recombinant leptin (80 nM) (3.20±0.14 nmol/10 islets/h) compared with that before the addition (4.41±0.30 nmol/10 islets/h). Although significant leptin suppression of insulin secretion was not observed under a glucose-stimulated (11.1 mM) condition, these results suggest that a negative feedback system may exist between leptin and insulin, which increases the production of leptin from adipose tissues.  相似文献

Mast cells are important mediators of normal angiogenesis, and participate in normal would healing, i.e. processes involved in pancreatic islet engraftment. The aim of the study was to evaluate if mast cells are present in islet grafts. For this purpose, male normoglycaemic Wistar-Furth rats were either untreated or syngeneically implanted with 250 islets under the renal capsule. The animals were killed 1 month later, and the kidneys and endogenous pancreas were removed, fixed and embedded in paraffin. The distribution of mast cells was studied in Alcian Blue stained sections. Mast cells were rarely encountered in endogenous islets, but were frequent in the renal capsule adjacent to islet grafts. Mast cells interspersed between graft endocrine cells were as rare as in the endogenous pancreas. We conclude that mast cells may contribute to the engraftment after islet transplantation.  相似文献

Summary The structure of the bulbus arteriosus of a wide range of teleost fish is described with particular reference to the vascularization and innervation. The adventitia of the organ consists of blood vessels and large nerve bundles in a collagen matrix. The nerve bundles contain monoamines, and fluorescence studies show small terminal bundles penetrating the muscular media; this is confirmed by electron microscopy. The media consists of an extensive elastic tissue matrix with a spiral arrangement of smooth muscle cells joined end to end by desmosomes and presumed electrotonic junctions. The muscle cells are innervated only at the adventitia/media boundary and the significance of this innervation is discussed. It is proposed that there is a correlation between the degree of vascularization and innervation and the activity of a particular species offish.  相似文献

In rat pancreatic islet membranes exposed to [alpha-32P]NAD, cholera toxin stimulated the labelling of three peptides with Mr close to 22 000, 42 000 and 48 000, respectively. In the islets, the toxin-stimulated ADP-ribosylation of the heavy form of the Ns alpha-subunit predominated over that of the light form, in mirror image of the situation found in the exocrine pancreas. When intact islets were preincubated with cholera toxin, the adenylate cyclase activity of a subcellular particulate fraction was increased. The responsiveness of adenylate cyclase to GTP was also augmented, but that to NaF was decreased. In intact islets, the production of cyclic AMP and the glucose-stimulated release of insulin were also enhanced after pretreatment with cholera toxin. These findings reveal the presence in pancreatic islets of the guanyl nucleotide regulatory protein of adenylate cyclase, with an unusual predominance of the heavy form of the Ns alpha-subunit.  相似文献
