Summary.  Quercus × warburgii A. Camus, the Cambridge Oak, is a semi‐evergreen tree of hybrid origin, possibly between Q. rugosa Née and Q. robur L., and is only known in cultivation. Mature trees in gardens are extremely rare. Its history and cultivation are discussed.  相似文献

The Pied and Collared Flycatchers Ficedula hypoleuca and F. albicollis hybridize on the Baltic islands of Öland and Gotland. Field studies of hatching success in clutches from 36 breeding hybrid males indicate that male hybrids reproduce successfully. In comparison, 25 hybrid females had normal clutch sizes but failed to hatch nestlings, suggesting female hybrid sterility. The paternity of seven hybrid males was investigated by DNA fingerprinting using two hypervariable minisatellite probes. Of these hybrid males, six were assessed as fertile. Three of the hybrid male families (43%) contained nestlings with extra-pair paternity and the frequency of extra-pair fertilization among 37 nestlings was 22%. One nestling originated from intraspeciflc female nest parasitism. The sterility pattern in hybrids between these two flycatchers with sterile female and fertile male hybrids is in agreement with Haldane's rule.  相似文献

Adult roach, bream and their presumed F1 hybrid from an Anglian Water reservoir were identified on the basis of morphological and meristic characteristics. The hybrid was clearly intermediate. Four hybrid breeding crosses were induced to spawn by hypophysis. A bream × roach cross (female named first) failed to produce fertile eggs, whereas F1 hybrid × roach, roach × F1 hybrid and F1 hybrid × F1 hybrid all produced fry. Fertility (defined as survival of eggs to hatching) was high for the F1 hybrid × roach back-cross (56%) but low for the others (<2%), in comparison to the pure species controls (roach 69%, bream 76%). Progeny from these crosses were reared until anal fin rays could be counted. These counts indicated intermediacy between the parents and back-crossed individuals, and similarity between F1 hybrids and their F2 progeny.  相似文献

Data on the production of offspring of 4×× 2× crosses in potato are presented. The ploidy composition of seedlings varied from year to year and parent to parent. Products of dihaploid induction crosses were chiefly 2× or 3× whereas crosses between S. tuberosum cultivars and unreduced gamete-producing S. phureja clones were mainly 3× or 4×. Substantial percentages of 3× seedlings were produced by some crosses in some years. It was concluded that the so-called ‘triploid block’ in potatoes is a variable phenomenon and that factors which increase seed production may suppress the proportion of triploids produced.  相似文献

Restriction fragment length polymorphisms in the internal transcribed spacer region of ribosomal DNA and in the chloroplast genome combine to confirm the existence of Potamogeton×bottnicus ( P. pectinatus × P. vaginatus ) in Britain. One of the parents, P. vaginatus , is not currently a member of the British flora, but did occur here in the past (the latest fossil fruits date from 30000 BP) and the hybrid may therefore owe its origin to ancient cross-pollination involving indigenous material or to more recent long-distance dispersal.  相似文献

In this study we report the first documentation of mating between harp ( Phoca groenlandica ) and hooded seals ( Cystophora cristata ). The production of this hybrid was quite unusual, being the result of a cross between parents of different genera which are morphologically dissimilar and have quite different mating behavior and dramatically different body sizes. The molecular techniques (mtDNA and macrosatellite nuclear DNA banding patterns) used in this study will undoubtedly be applied widely to many different taxa in the near future, allowing us to re-examine many suspected cases of hybridization among marine mammals and, in a larger context, the meaning of the species concept.  相似文献

以开花期的椒样薄荷(Mentha × piperita)、薄荷(M. haplocalyx)和苏格兰留兰香(M. × gentilis)叶片部位提取的精油为研究对象, 通过GC-MS分析, 并采用纸片扩散法研究了3种精油单独使用及与抗生素联合使用时对金黄色葡萄球菌、蜡状芽孢杆菌、大肠杆菌、绿脓杆菌和肺炎克雷伯氏菌的抑制情况。结果表明, (1) 椒样薄荷与薄荷精油中含量最高的成分为薄荷醇、薄荷酮和异薄荷酮, 苏格兰留兰香精油的主要成分为香芹酮和柠檬烯。薄荷和苏格兰留兰香精油符合欧洲药典与ISO标准, 椒样薄荷需要继续改良以提高其精油品质与抑菌功能。(2) 精油单独使用时, Pseudomonas aeruginosa ATCC 15442对椒样薄荷精油和薄荷精油敏感; P. aeruginosa ATCC 27853对薄荷精油和苏格兰留兰香精油敏感。精油与抗生素联合使用时抑菌范围和强度均有所改变: 绿脓杆菌的2个菌株对精油与抗生素的组合最为敏感, 其中, 椒样薄荷精油与头孢他啶的组合对P. aeruginosa ATCC 15442显示出最强的增效作用, 薄荷精油与头孢他啶混合之后对P. aeruginosa ATCC 27853出现拮抗作用。Staphylococcus aureus ATCC 25923对所有精油以及精油与抗生素混合物均有抗性。(3) 椒样薄荷、薄荷和苏格兰留兰香精油的不同成分及其含量差异不仅对精油品质有影响, 而且影响精油对测试菌种的抑制作用, 可考虑将其作为薄荷属植物品质育种的参考指标。  相似文献

The purification and characterization of psychro‐thermoalkalistable protease from psychrotrophic Pseudomonas putida isolate is being reported for the first time. A ~53 kDa protease was purified 21.4‐folds with 57.2% recovery by ultrafiltration and hydrophobic interaction chromatography. Kinetic analyses revealed the Km and Vmax to be 1.169 mg mL?1 and 0.833 mg mL?1 min?1, respectively. The kcat value of 3.05 × 102 s?1 indicated high affinity and catalytic efficiency toward casein. The protease was most active at pH 9.5 and 40°C, with 100% stability in pH and temperature range of 6.0–11.0 and 10–40°C, respectively. Presence of Zn2+ increased the thermostability of protease (at 70°C) by 433%. Ethylene diamine tetra acetic acid (EDTA) and 1,10‐phenanthroline were inhibitory, whereas phenyl methyl sulfonyl fluoride (PMSF), p‐chloro mercuric benzoate (PCMB), and β‐mercaptoethanol were ineffective, revealing the enzyme to be a metalloprotease. Zinc, calcium, iron, nickel, and copper at 1 mM increased the enzyme activity (102–134%). Complete reversion of enzyme inhibition (caused by Ethylene diamine tetra acetic acid [EDTA]) by Zn2+ affirmed this enzyme as zinc‐dependent metalloprotease. At 0.1% concentration, Triton X‐100 and Tween 80 slightly increased, while SDS and H2O2 reduced the protease activity. In the presence of 0.1% commercial detergents, the enzyme was fairly stable (54–81%). In the presence of organic solvent, the protease was remarkably stable exhibiting 72–191% activities. In contrast, savinase exhibited good stability in the presence of hydrophilic solvents, while chymotrypsin showed elevated activities with benzene, toluene, and xylene only. Circular dichroism analysis revealed the protease as a β‐rich protein, having large fraction (~40%) of β‐sheets. Presence of different environmental conditions altered the β‐content, which accordingly affected the protease activity. © 2012 American Institute of Chemical Engineers Biotechnol. Long-term preservation of complex engineered tissues and organs at cryogenic temperatures in the absence of ice has been prevented to date by the difficulty of discovering combinations of cryoprotectants that are both sufficiently non-toxic and sufficiently stable to allow viability to be maintained and ice formation to be avoided during slow cooling to the glass transition temperature and subsequent slow rewarming. A new theory of the origin of non-specific cryoprotectant toxicity was shown to account, in a rabbit renal cortical slice model, for the toxicities of 20 vitrification solutions and to permit the design of new solutions that are dramatically less toxic than previously known solutions for diverse biological systems. Unfertilized mouse ova vitrified with one of the new solutions were successfully fertilized and regained 80% of the absolute control (untreated) rate of development to blastocysts, whereas ova vitrified in VSDP, the best previous solution, developed to blastocysts at a rate only 30% of that of controls. Whole rabbit kidneys perfused at -3 degrees C with another new solution at a concentration of cryoprotectant (8.4M) that was previously 100% lethal at this temperature exhibited no damage after transplantation and immediate contralateral nephrectomy. It appears that cryoprotectant solutions that are composed to be at the minimum concentrations needed for vitrification at moderate cooling rates are toxic in direct proportion to the average strength of water hydrogen bonding by the polar groups on the permeating cryoprotectants in the solution. Vitrification solutions that are based on minimal perturbation of intracellular water appear to be superior and provide new hope that the successful vitrification of natural organs as well as tissue engineered or clonally produced organ and tissue replacements can be achieved.  相似文献

Objective

To compare the effect of novel direct cover vitrification (DCV) and conventional vitrification (CV) for human ovarian tissue.

Study design

Ovarian biopsy specimens obtained from 12 patients were randomly allocated into five groups: Fresh, DCV1, DCV2, DCV3 and CV. Three concentrations of cryoprotectants were used in DCV group. The equilibration solution of DCV1, DCV2, DCV3 was 5% EG + 5% DMSO + DPBS, 7.5% EG + 7.5%DMSO + DPBS, 10% EG + 10% DMSO + DPBS, respectively. And the vitrification solution of DCV1, DCV2, DCV3 was 10% EG + 10% DMSO + DPBS, 15%EG+15% DMSO + DPBS, 20% EG + 20% DMSO + DPBS, respectively. The equilibration solution and the vitrification solution of CV group was same as DCV3 group. The effects of cryopreserved procedure on human ovarian tissue were studied by histology, TUNEL assay, transmission electron microscopy (TEM) and heterotopic allograft.

Results

The percentages of morphologically normal and viable follicles of DCV2 were significantly higher than DCV1, DCV3 and CV groups (P < 0.05). TUNEL assay demonstrated that the incidence of apoptotic cell in vitrification ovarian tissue was significantly higher than fresh tissue (P < 0.05), but there were no difference in various groups with cryopreservation. TEM showed that less damage was detected in DCV2 group. After grafting, the follicle density of DCV2 was greater than DCV1, DCV3 and CV groups (P < 0.05).

Conclusions

The novel cover vitrification with optimal concentration of cryoprotectants is superior to conventional vitrification. It is suitable for human ovarian tissue fragments with high efficiency and facility.  相似文献

Vitrification is a process in which a liquid begins to behave as a solid during cooling without any substantial change in molecular arrangement or thermodynamic state variables. The physical phenomenon of vitrification is relevant to both cryopreservation by freezing, in which cells survive in glass between ice crystals, and cryopreservation by vitrification in which a whole sample is vitrified. The change from liquid to solid behavior is called the glass transition. It is coincident with liquid viscosity reaching 1013 Poise during cooling, which corresponds to a shear stress relaxation time of several minutes. The glass transition can be understood on a molecular level as a loss of rotational and translational degrees of freedom over a particular measurement timescale, leaving only bond vibration within a fixed molecular structure. Reduced freedom of molecular movement results in decreased heat capacity and thermal expansivity in glass relative to the liquid state. In cryoprotectant solutions, the change from liquid to solid properties happens over a ∼10 °C temperature interval centered on a glass transition temperature, typically near −120 °C (±10 °C) for solutions used for vitrification. Loss of freedom to quickly rearrange molecular position causes liquids to depart from thermodynamic equilibrium as they turn into a glass during vitrification. Residual molecular mobility below the glass transition temperature allows glass to very slowly contract, release heat, and decrease entropy during relaxation toward equilibrium. Although diffusion is practically non-existent below the glass transition temperature, small local movements of molecules related to relaxation have consequences for cryobiology. In particular, ice nucleation in supercooled vitrification solutions occurs at remarkable speed until at least 15 °C below the glass transition temperature.  相似文献

The Coxiella burnetii icd gene encoding an immunogenic dimeric NADP(+)-dependent isocitrate dehydrogenase (IDH) was cloned by screening a C. burnetii genomic library with a human positive serum and sequenced. The predicted gene product consists of 427 amino acids (M(r) = 46,600) and showed high identity to the IDHs of Escherichia coli (74%), Salmonella enterica (73%) and IDH-I of Vibrio sp. (71%). The cloned gene complemented an icd-defective E. coli mutant producing a recombinant IDH that had the same biochemical properties as the enzyme from purified C. burnetii. Unlike the homologs from other bacteria, the cloned enzyme was expressed to the highest level in low pH conditions. This distinct property of the cloned IDH suggests that C. burnetii icd gene may have a role in the adaptation of the organism to the harsh acidic environment of the eucaryotic phagolysosomes.  相似文献

Faúopia × bohemica is a good example of a hybrid occurring when two introduced species come in contact in their adventive range. Although only recognised in the early 1980's, herbarium material and the current distribution pattern suggest that it has long been an unrecognised part of the Japanese Knotweed s.l. population. The identification, distribution, cytological background, history and some of the ecological determinants of the spread in Europe are provided along with a substantial citation of field observations and herbarium specimens used to produce the distribution map. This is the first attempt to map its European distribution, and must be regarded as provisional since comprehensive recording of its distribution is limited to a handful of countries. The unexpected discovery for the first time of this taxon in the Mediterranean is also reported. It is to be hoped that this paper will provide the impetus for a more comprehensive study of this invidious and under-recorded taxon.  相似文献
