Nine strains of Aeromonas hydrophila isolated from diseased fish or soft-shelled tortoise were tested for the presence of three virulence genes including the genes encoding aerolysin,hemolysin,and extracellular serine protease (i.e.,aerA,hlyA,and ahpA,respectively).These genes were investigated using polymerase chain reaction (PCR)with specific primers for each gene.And the pathogenicities to Carrassius auratus ibebio of these strains were also assayed.PCR results demonstrated that the distribution patterns of aerA,hlyA,and ahpA were different in these strains.6/9 of A.hydrophila strains were aer A positive,8/9 of strains hly A positive,7/9 of strains ahp A positive,respectively.However,the assay for pathogenesis showed that two strains (A.hydrophila XS91-4-1 and C2)were strong virulent,two strains (A.hydrophila ST78-3-3 and 58-20-9)avirulent and the rest middle virulent was to the fish.In conclusion,there are significant correlation between the distribution pattern of the three virulence genes and the pathogenicity to Carrassius auratus ibebio.All strong virulent A.hydrophila strains were aerA+hlyA+ahpA+genotype,and all aerA+hlyA+ahpA+strains were virulent.Strains with the genotype of aerA-hlyA-ahpA+have middle pathogenicity.In the present study,we found for the first time that all A.hydrophila isolated from the ahpA positive were virulent to Carrassius auratus ibebio.Additionally,there was a positive correlation between the virulence of A.hydrophila and the presence of aerA and ahpA.  相似文献

以携带质粒pAM12 0 (Tcr Tn916 )的大肠杆菌CG12 0株为供体菌 ,采用滤膜接合法与受体菌嗜水气单胞菌J_1株 (cfzr)进行接合转移 ,在含Tc和cfz选择平板上进行筛选。共获接合转移菌落 380 0个 ,其接合频率为 3× 10 - 5(按供体细胞计算 )。任取 38个接合子 ,提取基因组DNA ,以嗜水气单胞菌特异性 16SrDNA引物进行PCR扩增 ,所有接合子均阳性。为证明Tn916确实插入基因组 ,以四环素基因 (tet)引物进行PCR扩增 ,结果所有抗性接合子均扩增出一条特异条带。与亲本J_1株相比 ,所有接合子的主要毒力因子如蛋白酶、溶血素、DNA酶和淀粉酶等均不表达 ,对小鼠失去致病力 ,其LD50 大于 10 9CFU。接合子连传 10次后 ,四环素抗性消失 ,但毒力未恢复 ,说明通过转座子Tn916的插入可获得稳定的无毒嗜水气单胞菌突变株。Tn916引起嗜水气单胞菌毒力性状改变的机制有待研究 ,推测可能与该菌染色体上存在Tn916的热点或毒力岛有关。  相似文献

A non-enterotoxigenic strain of Aeromonas hydrophila isolated from diarrheic stools of an 8-month-old child was found to cause vacuolation in Caco-2 cells. The vacuoles became prominent 60 min after addition of the bacterial culture to the cell monolayers and, after 120 min, a complete disruption of the monolayers was observed. Cell vacuolation was not detected when Caco-2 monolayers were tested with sterile filtrates of overnight cultures of the A. hydrophila vacuolating strain AH14846. This strain produced a diffuse adherence pattern in Caco-2 cell monolayers, but did not produce detectable cytotonic enterotoxin in the suckling mouse test and only produced small quantities of aerolysin. By demonstrating the ability to induce vacuolation in mammalian cells of enterocytic lineage, the current study raises the possibility that such activity might contribute to gastrointestinal symptoms in infections involving Aeromonas strains which do not express well-established enterotoxins.  相似文献

嗜水气单胞菌感染现状及耐药分析   总被引:7,自引:0,他引:7  
目的调查湖州市中心医院嗜水气单胞菌感染现状和耐药情况。方法采用常规方法分离,用VITEK-32全自动微生物分析仪进行菌种鉴定为嗜水气单胞菌或豚鼠气单胞菌,依据葡萄糖产气反应鉴定为嗜水气单胞菌。并根据配套药敏卡进行药敏试验。结果共分离到34株嗜水气单胞菌,主要来自痰液、胆汁、腹腔引流液或腹水。嗜水气单胞菌对哌拉西林、替卡西林、阿莫西彬克拉维酸、妥布霉素、头孢呋辛、头孢噻肟、头孢他啶、环丙沙星、复方新诺明耐药率为52.9%~73.5%。结论目前嗜水气单胞菌也呈现多重耐药现象,临床上应予以重视。  相似文献

Abstract

Microbial transglycosylation is useful as a green alternative in the preparation of purine nucleosides and analogues, especially for those that display pharmacological activities. In a search for new transglycosylation biocatalysts, two Aeromonas hydrophila strains were selected. The substrate specificity of both micro-organisms was studied and, as a result, several nucleoside analogues have been prepared. Among them, ribavirin, a broad spectrum antiviral, and the well-known anti HIV didanosine, were prepared, in 77 and 62% yield using A. hydrophila CECT 4226 and A. hydrophila CECT 4221, respectively. In order to scale-up the processes, the reaction conditions, product purification and biocatalyst preparation were analyzed and optimized.  相似文献

分别利用葡萄糖或葡萄糖酸钠与十一碳酸、月桂酸与十一碳酸为混合碳源进行嗜水气单孢菌 (Aeromonashydrophila)菌株 4AK4的摇瓶培养 ,实现了含有 3 羟基戊酸 (3HV)单体的聚羟基脂肪酸酯的微生物合成。当使用葡萄糖或葡萄糖酸钠与十一碳酸为混合碳源时 ,野生型A .hydrophila 4AK4及含有 3 羟基丁酸辅酶A合成基因phaA和phaB的重组A .hydrophila 4AK4 (pTG01)能够合成-3-羟基丁酸(3HB)与-3HV的共聚物 ,且葡萄糖或葡萄糖酸钠与十一碳酸比例为 1∶1时最利于细胞生长和PHA的积累。当使用月桂酸和十一碳酸为混合碳源时 ,A .hydrophila4AK4能够合成-3HB、3HV与 β-羟基己酸 (3HHx)的共聚物 ,且随着混合碳源中十一碳酸的含量增加 ,A .hydrophila4AK4合成的PHA中-3HV的比例增加 ,而-3HB和-3HHx的比例降低.  相似文献

Aims: To evaluate the frequency of the aerolysin (aerA), cytotoxic enterotoxin (alt) and serine protease (ahp) genes in Aeromonas hydrophila isolates from different sources, and to determine the relationship between the presence of these genes and virulence of A. hydrophila in zebrafish. Methods and Results: Aeromonas hydrophila isolates from clinical cases (n = 40), from healthy fish (n = 22) and from water environment (n = 21) were analysed with respect to the prevalence of aerA, alt and ahp genes by PCR assay. These virulence factors occur among clinical isolates as well as among isolates from healthy fish and water environment. The majority (97·6%) of the strains examined carried one or more virulence genes. The isolates were divided into seven genetic profiles on the basis of PCR result: aerA+alt+ahp+ (62·7%), aerA+alt+ahp? (13·3%), aerA+alt?ahp+ (10·8%), aerA?alt+ahp+ (4·8%), aerA?alt?ahp+ (3·6%), aerA+alt?ahp? (2·4%) and aerA?alt?ahp? (2·4%). A higher frequency of genetic group aerA+alt+ahp+ was determined in the isolates from diseased animals compared to those from healthy fish or water environments. Virulence properties of 26 representative strains belonging to the seven genetic profiles were further characterized. Results demonstrated that as the present of virulence genes increased, the proteolytic, haemolytic and cytotoxic activities of extracellular products also increased. And the 50% lethal doses (LD50s) of aerA+alt+ahp+ isolates (<105) in zebrafish were lower when compared with the strains expressing one or combinations of two virulence genes (>106). Conclusions: Virulence properties of A. hydrophila correlated well with the presence of virulence genes tested. aerA+alt+ahp+ was more frequent virulence genotype in A. hydrophila isolates from clinical diseases than from healthy fish and water environment, and the aerA+alt+ahp+ isolates were more virulent to zebrafish compared to the other six genetic profiles. Significant and Impact of the Study: The detection for aerA, alt and ahp can be used for virulence typing of A. hydrophila isolates.  相似文献

嗜水气单胞菌菌蜕的制备及其对银鲫的口服免疫   总被引:1,自引:0,他引:1  
菌蜕系统是一个自身具有佐剂性质的新型疫苗体系,不含细胞质内容物但具有细菌的完整表面抗原结构,可诱导机体的体液、细胞免疫应答及增强黏膜免疫反应.本研究通过将带有裂解基因E的质粒pElysis转化至嗜水气单胞菌J-1株中,对Ah J-1(pElysis)进行温度诱导,温度从28℃升至42℃,每隔15min检测菌液的OD600值,测定其溶菌动力学,并做无菌检验,用扫描电镜观察裂解后的细菌形态,研究其作为口服疫苗对银鲫的效果.结果显示,通过温度诱导,嗜水气单胞菌J-1(pElysis)OD值在诱导30min后开始持续下降,75min时开始趋于平稳,到120min溶菌效率达99.99%,诱导16h后进行无菌检验,证实其无活菌.扫描电镜观察绝大部分菌体经诱导后形成菌蜕,细胞两端有溶菌通道.动物试验表明,用菌蜕口服免疫的银鲫,在第5周产生较高的凝集抗体,达到27,并能维持2周;而甲醛灭活苗组为26,维持时间仅一周;生理盐水对照组效价仅2.攻击试验表明,菌蜕疫苗组和甲醛灭活疫苗组对嗜水气单胞菌强毒株J.1的攻击均有保护作用,其相对保护率分别为16/20(78.95%)和12/20(57.9%),显示菌蜕疫苗比普通灭活疫苗能更有效地激活机体的免疫保护.  相似文献

嗜水气单胞菌生物被膜对其耐药性的影响   总被引:9,自引:0,他引:9  
建立了嗜水气单胞菌(Aeromonas hydrophila, Ah)的生物被膜(Bacterial biofilm, BF) 体外形成模型, 并对11种抗菌药物对BF细菌和浮游(Freecell, FC)细菌的清除作用进行了研究。将Ah J1株在放有硅胶膜的TSB中培养7d,用银染法鉴定,发现可形成良好的BF。FC细菌对青霉素具有耐药性, 最低杀菌浓度(MBC)为256μg/mL;对蒽诺沙星和氟哌酸最敏感,MBC分别为003μg/mL和0.25μg/mL。氟苯尼考对BF细菌的清除能力最强,作用于BF细菌和FC细菌的MBC之比为2∶1;卡那霉素、青霉素、新霉素的MBC比值在32∶1以上。扫描电镜观察蒽诺沙星作用于FC及BF细菌前后的形态变化,并测定其杀菌曲线。发现4×MBC时可完全清除FC细菌,但不能完全清除BF细菌;在32×MBC时,4h内可完全清除FC细菌,而24h内完全清除BF细菌。结果表明形成BF的Ah对抗菌药物可形成强耐受性,其潜在影响应引起足够重视。  相似文献

Abstract The relationship between virulence, O-serogroup, and some cell-surface features (self-pelleting [SP] and precipitation after boiling [PAB], profile of lipopolysaccharides [LPSs]) and outer membrane proteins [OMPs] was investigated in strains of the pathogenic species Aeromonas hydrophila and A. jandaei isolated from eels. Virulent strains of A. hydrophila reacted mostly with O:19 antiserum, and those of A. jandaei reacted with O:4, O:11, O:15 and O:29 antisera (Guinée and Jansen system). Regarding the PAB and LPS profiles two groups could be distinguished; (i) five PAB+ strains of serotype O:19 that possessed a homogeneous O polysaccharide side chain and (ii) thirteen PAB strains antigenically diverse that either exhibited a heterogenous side chain or were side chain deficient. A major 50 kDa protein was only found in the PAB+ strains, whereas major OMPs detected in PAB strains ranged from 33 to 45 kDa irrespective of the species. Epizootic eel isolates of A. hydrophila belong to serotype O:19 and share cell-surface features with the Aeromonas highly virulent for other hosts. In contrast, epizootic A. jandaei isolates were antigenically diverse. These findings reinforce the importance of an O-serotype as an epidemiological marker in motile Aeromonas strains pathogenic for eels.  相似文献

A transposon mutant from Aeromonas hydrophila AH-3 was obtained which was highly resistant to opsonophagocytosis. The mutation was identified in the ftsE gene and we characterised the operon ftsY, E and X from this bacterium. These genes, as in enteric bacteria, are neighbours to rpoH. The A. hydrophilia ftsE and X genes were fully able to complement Escherichia coli ftsE mutants, and also complement the opsonophagocytosis-resistant phenotype of the A. hydrophila mutant strain. This phenotype seems to be related to the filamentous phenotype at 37 degrees C exhibited by the A. hydrophila ftsE mutant.  相似文献

嗜水气单胞菌Aeromonas hydrophila具有广泛的致病性,是水生动物最常见的致病菌之一。由于抗生素不合理使用、质粒以及耐药基因的水平转移等因素,来自零售市场、超市和餐厅的即食海鲜产品中,均可分离出大量的嗜水气单胞菌耐药菌株及其耐药基因。因此,探明关键控制点、寻求有效缓解抗生素耐药性的防控策略至关重要。文中介绍了我国嗜水气单胞菌的耐药现状,嗜水气单胞菌的主要侵染及耐药机制,以及目前削减和防控耐药性的主要手段和策略,并对水产品耐药性研究方向和重点作出展望。  相似文献

聚羟基脂肪酸酯(Polyhydroxyalkanoate,PHA)是一系列生物合成的高分子材料,其单体可由多种3-羟基脂肪酸(3-hydroxyalkanoate,3HA)构成^[1]。PHA物理和机械性能的变化很大,从高脆性到弹性体,这跟它们的单体成分有很大关系^[2]。短链和中长链单体共聚的PHA比短链单体或中长链单体聚合得到的PHA有着更好的性能^[3]。在1994年,豚鼠气单胞菌(Aeromonas caviae)FA440被发现能以偶数碳原子数脂肪酸或植物油作为碳源在体内积累PHBHHx^[4]其PHA生物合成基因被成功克隆^[5]。根据亚基数目和底物特异性,PHA合成的关键酶,即PHA合酶或PhaC,被分成了3种类型。A.caviae的PHA合酶属于第1类PHA合酶^[6]。PHA合酶的一些类型含有一些保守的基因序列,该特征可被用于克隆,特别是第Ⅱ类PHA合酶^[2,8]。嗜水气单胞菌(Aeromonas hydrophila)WQ和A.hydrophila 4AK4是能够合成PHBHHx的另外两种菌株,其中A.hydrophila 4AK4已被用作大规模生产PHBHHx。就目前来说,不管生长条件怎么改变,其合成的PHBHHx中3羟基己酸单体(3-hydroxyhexanoate,3HHx)的含量始终在12%~17%之间变化^[9]。而A.hydrophila WQ合成的PHBHHx中则含有6%~14% 3HHx。本论文研究了A.hydrophila WQ的PHA生物合成及其分子基础。  相似文献

三重PCR检测鱼类致病性嗜水气单胞菌   总被引:8,自引:0,他引:8  
[目的]建立一种能够快速准确地检测致病性嗜水气单胞菌的PCR.方法.[方法]根据嗜水气单胞菌的16S rRNA、气溶素基因(aer)和丝氨酸蛋白酶基因(ahp)的保守序列设计了3对引物,然后进行了PCR反应条件的优化、特异性和敏感性的检测并与普通的细菌分离鉴定进行了临床样本和人工攻毒样本检出率的比较.[结果]该方法特异性好,只对致病性嗜水气单胞菌呈阳性扩增;敏感性高,最低可检测100fg的细菌DNA模版.对临床疑似黄鳝(Monopterus albus)样本的检出率为81.8%,高于细菌分离的40.9%;对人工攻毒鲫鱼(Carassius auratus)样本的检出率为87.5%,高于细菌分离的67.5%.[结论]本方法的成功建立,实现在同一反应管中同时对16SrRNA、aer和ahp的检测,避免了只针对aer或ahp单个毒力基因的PCR检测方法可能存在的漏检和误检,为致病性嗜水气单胞菌的诊断、大规模检疫、流行病学调查等提供了一种快速、准确而有效的检测方法.  相似文献

 蛋白酶是嗜水气单胞菌 (Aeromonashydrophila)的重要致病因子 .为研究其结构与功能之间的关系 ,用DEPC、EDC、PMSF、N AI等 9种化学修饰剂处理嗜水气单胞菌J 1株胞外蛋白酶ECPase54,然后检测残余酶活力 ,借以研究酶分子中氨基酸侧链基团与酶活性中心的关系 .结果表明 ,羧基、丝氨酸、ε 氨基、胍基等残基与酶活性无关 ;半胱氨酸残基与酶活性也无直接关系 ;而色氨酸、组氨酸、酪氨酸残基侧链以及二硫键的化学修饰引起酶活性的大幅度的下降 ,说明色氨酸、组氨酸、酪氨酸残基以及二硫键是酶活力所必需的基团  相似文献

AIMS: The aim of this work was to study the biodegradation of benzyldimethylalkylammonium chloride (BAC) by Aeromonas hydrophila sp. K, an organism isolated from polluted soil and capable of utilizing BAC as sole source of carbon and energy. METHODS AND RESULTS: High performance liquid chromatography and gas chromatography-mass spectrometry (GC-MS) analysis was used to study BAC degradation pathway. It was shown that during BAC biodegradation, formation of benzyldimethylamine, benzylmethylamine, benzylamine, benzaldehyde and benzoic acid occurred. Formation of benzyldimethylamine as the initial metabolite suggested that the cleavage of Calkyl-N bond occurred as the first step of BAC catabolism. Liberation of benzylmethylamine and benzylamine likely resulted from subsequent demethylation reactions, followed by deamination with formation of benzaldehyde. Benzaldehyde was rapidly converted into benzoic acid, which was further degraded. CONCLUSIONS: Aer. hydrophila sp. K is able to degrade BAC. A degradation pathway for BAC and related compounds is proposed. SIGNIFICANCE AND IMPACT OF STUDY: These findings are significant for understanding biodegradation pathways of benzyl-containing quaternary ammonium compounds.  相似文献

Allelic variations in the chromosomal genome of 120 isolates of motile Aeromonas (A. hydrophila, A. sobria and A. caviae) and eight reference strains with one Plesiomonas shigelloides were assessed by analysis of electrophoretically demonstrable polymorphism in 16 genes encoding metabolic enzymes. The strains were collected from humans (n = 59) and from the aquatic environment (n = 61) in canton Tessin, Switzerland. Clustering of the electrophoretic types (ET) from a matrix of pairwise genetic distances, based on the 16 enzyme loci, confirmed the genetic distinctness of the three species. Furthermore, A. hydrophila and A. sobria were divided in three and two groups respectively. For each species clinical strains were well differentiated from those collected in the environment.  相似文献

嗜水气单胞菌(Aeromonas hydrophila,Ah)是淡水鱼暴发性败血症的主要病原,该菌能够引致淡水鱼等的败血症和人的腹泻等^[1]。嗜水气单胞菌有多种致病因子,如毒素、蛋白酶、S层蛋白等^[2],还发现它具有侵袭作用,有报道嗜水气单胞菌粪分离株能侵袭HEp-2细胞^[3],但对于鱼源菌株的侵袭特性知之甚少。仅有一些报道认为嗜水气单胞菌能引致细胞病变^[4,5]。  相似文献
