Summary AeratedVicia faba root meristems were irradiated with 1.9 MeV monoenergetic neutrons. This source of neutrons optimally provides one class of particles (recoil protons) with ranges able to traverse cell nuclei at moderate to high-LET. The volumes of theVicia faba nuclei were log-normally distributed with a mean of 1100 µm3. The yield of chromatid-type aberrations was linear against absorbed dose and near-constant over 5 collection periods (2–12 h), after irradiation. Energy deposition events (recoil protons) determined by microdosimetry were related to cytological changes with the finding that 19% of incident recoil protons initiate visible changes inVicia faba chromosomes. It is probable that a substantial fraction of recoil proton track length and deposited energy is in insensitive (non-DNA containing) portions of the nuclear volume.  相似文献

Inverted repeated DNA sequences have been isolated from sheared Vicia faba DNA by hydroxylapatite column chromatography, treated with nuclease S1, tritiated by the nick translation method and hybridized in situ on squashes of Vicia faba root tips. Silver grains appear grouped in a rather limited portion of interphase nuclei and form a sort of band across them. The central regions of metaphase chromosomes are preferentially labeled, labeling being excluded from telomeres, centromeres and secondary constrictions. These results are briefly discussed in relation to those obtained in other species and the functional significance of inverted repeats.  相似文献

The speciesVicia bithynica andVicia narbonensis, from the same subgeneric section ofVicia faba, show variations in nuclear DNA content Nuclear DNAs, extracted from root tips of the twoVicia species, were characterized by thermal denaturation, analytical ultracentrifugation and reassociation kinetics. The thermal denaturations of DNA, the number of DNA components reassociating with second order kinetics, the proportion of repeated DNA sequences, the frequency of the repeated DNA classes are reported and compared to previous data onVicia faba DNA. Feulgen absorptions at different thresholds of optical density+ of interphase nuclei in cytological preparations of the root meristems ofV. bithynica andV. narbonensis are determined and compared withV. faba analogous determinations. The results, confirming that plant genome is highly flexible, are discussed in relation to other data on the interspecific variations of the nuclear DNA content.  相似文献

Chromosomes of the broad bean Vicia faba were isolated and air-dried on slides after incorporation of BUdR into DNA (BUdR substitution) for two rounds of replication. Then the preparations were embedded in a buffer solution containing trypsin as well as fluorescence dye (acridine orange or Hoechst 33258). We observed chromosomes with a fluorescence microscope at various times after embedding. After about 15 min one sister chromatid of some of the metaphase chromosomes showed enhanced darkening and disintegration within 1–4 min (melting effect) during observation. We suppose that fragmentation of BUdR-substituted DNA by the acridine orange-visible light system in acridine orange staining and by irradiation with wavelengths around the transition from UV to visible light in Hoechst 33258 staining is responsible for this phenomenon. The disintegration of one sister chromatid in BUdR-substituted chromosomes can also be produced by UV irradiation during trypsin treatment when fluorescence dyes are not present.  相似文献

After digestion with trypsin the metaphase chromatids of Vicia faba reveal two length structures (half-chromatids). This confirms the results of other authors. According to our observations the half-chromatids are connected at their ends in such a way that a U-shaped configuration is formed. This finding can be explained in different ways. The ends of chromatid fragments obtained after X-irradiation are either U-shaped (“closed”) like the “natural” chromatid ends or “open”, i.e. the two half-chromatids do not seem to be connected at their ends. As a by-product of our experiments the structure of X-ray-induced achromatic lesions (gaps) was studied in trypsin-treated chromosomes.  相似文献

The hypothesis of indirect mitotic nondisjunction was tested in plant and mammalian cells. This hypothesis states that micronuclei derived from lagging chromosomes or chromatids are able to perform DNA synthesis and undergo mitotic condensation synchronously with main nuclei. Hence, as chromosomes, they can be moved to spindle poles together with the chromosomes of the main nuclei during mitosis. In that way chromosomes lost as micro-nuclei can be reincorporated in the main nuclei. In order to test this, both Vicia faba meristematic cells and cells of a Chinese hamster line (Cl-1) were treated with low doses of colchicine. Mitotic anomalies, micronuclei and cells with a polyploid or aneuploid karyotype were scored at different fixation times. A detailed analysis was performed on single chromosome misdistributions, as well as on micronuclei and cells with aneuploid karyotypes derived from single chromosome misdistributions. Indirect mitotic nondisjunction was shown to play a primary role in the origin of aneuploid karyotypes in Vicia faba, but not in Cl-1 cells.  相似文献

Monocentric chromosomes of Vicia faba (2n=12) and polycentric chromosomes of Luzula purpurea (2n=6) and L. multiflora (2n=36) were studied by the C-banding technique. C-positive regions exhibited a restricted distribution in V. faba nuclei and were located near the centromeres of the chromosomes. Each chromosome had both a characteristic number and distribution of C-positive regions permitting homologue identification. L. purpurea and L. multiflora C-bands were much more numerous than those of V. faba and were widely distributed throughout both nuclei and chromosomes. Three distinct constitutive heterochromatin distribution patterns were present in L. purpurea metaphase chromosomes permitting homologous chromosome identification. One of three C-band distribution patterns was also evident in L. multiflora chromosomes.  相似文献

Summary DNA sequences reassociating within a Cot value of 1.8×10–1 and those producing a light satellite in a CsCl density gradient were isolated fromVicia faba DNA and hybridizedin situ on squashes of roots of the same species. Silver grains were seen to be scattered over both the interphase nuclei and the metaphase chromosomes after hybridization with fast renaturing DNA sequences, indicating these are fairly regularly interspersed in theV. faba genome. Clustered labeling occurred after hybridization with satellite DNA sequences, indicating these are clustered in the genome. The localization of satellite DNA in chromosomes appeared to correspond closely to the position of the bright bands detectable after staining with quinacrine mustard. After hybridization with both DNA probes, labeling intensity over the nuclei of meristematic cells was higher than that over the nuclei of differentiating and/or differentiated cells. These results are discussed in relation to the structure of the cell nucleus, the mechanism of quinacrine banding and to previous data suggesting underrepresentation of nuclear repeated DNA sequences in differentiatingV. faba root cells.  相似文献

Summary A histochemical study of the mitotic chromosomes and interphase nuclei of rat regenerating liver and of root tips from Vicia faba and Trillium grandiflorum has been performed using the periodic acid-Schiff reaction. Carbohydrates and plasmalogen-like molecules were demonstrated on these cellular structures. The presence of such components was confirmed by biochemical studies upon nuclei isolated from rat liver.  相似文献

Abstract

A comparison has been made between the Quinacrine fluorescence bands and the bands obtained with a denaturating-reannealing-Giemsa technique in Vicia faba. The results show that some of the bands, particularly on the M and, proximally, on the S chromosomes are visible with both techniques. A complex pattern of bands on the S chromosomes is revealed with the Giemsa technique. Both the similarities and the differences between the banding patterns obtained with the two methods in Vicia faba may indicate various degrees of DNA repetitiousness and other physico-chemical properties in the chromosome segments involved.  相似文献

The Giemsa banding patterns of the standard karyotype of Vicia faba and of four new karyotypes with easily interdistinguishable chromosomes due to interchanges and inversions are described and compared with the data of other authors on preferential Giemsa staining in Vicia faba. All karyotypes contain 14 easily reproducible marker bands which characterize chromosome segments known to be heterochromatic. It is shown that the preferential Giemsa staining of chromosome regions is a valuable tool for the localization of translocation and inversion points in the chromosomes of the reconstructed Vicia karyotypes. A close correlation exists between banding patterns, segment extension by incorporation into chromosomal DNA of azacytidine and mutagen-specific clustering of induced chromatid aberrations in the new karyotypes.  相似文献

The cytological analysis of gaps induced by 4-aminouracil in Vicia faba shows that they are characterized by the presence of 1–2 very fine parallel chains, probably a chromatid skeleton mentioned by other authors. These achromatic lesions of the chromosomes are probably due to the loss or despiralization of both DNA and chromosomal proteins.  相似文献

The effect of exogenous DNA of syngeneic origin on the course of reparation of meristematic cells ofVicia faba primary roots followed after treatment with monofunctional alkylating agent, ethyl methanesulphonate (EMS), was tested. Time course of alternations in mitotic activity of investigated cellular population and the dynamics of formation of postmetaphase chromosomal aberrations was evaluated. A reparation of damaged cells was significantly supported by syngeneic DNA; its application induced an increased incidence of cellular division already in the early intervals of the repair which was accompanied by concomitant decrease of microscopically detectable rupture in the chromosomes. The study performed on the localization of induced damages occurring in metaphase chromosomes pointed out an increased sensitivity of small chromosomes ofVicia faba to EMS. Similarly, a reparative action of syngeneic DNA was exhibited by significant decrease of aberrations frequency, predominantly in the same chromosomal group. Per cent representation of individual types, not affected by the action of syngeneic DNA, was established by detailed classification of induced aberrations. In both cases, isochromatide breaks were found of greatest predominance.  相似文献

The mitotic activity of merstematic cells ofVicia faba, the frequency of chromosomal abnormalities and their interchromosomal distribution are evaluated in dependence on the concentration of maleic hydrazide (MH) and on the recovery period. The influence of exogenous DNA of different genetic origin on the course of repair of primary root cells damaged by MH was also studied. Isologous DNA which exhibited a strong repair effect in authors' previous experiments was quite ineffective in the case of maleic hydrazide. Heterologous DNA, on the other hand, had to some extent a parallel effect with MH in breaking down the structural integrity of chromosomes and increased the frequency of aberrations induced by maleic hydrazide in meristematic cells ofVicia faba.  相似文献

In a recent study, when X-irradiated chromosomes of Vicia faba were treated with trypsin, we observed two types of ends of chromatid fragments, namely “open” and “closed” ends. To put this qualitative finding on a quantitative basis, the fraction of “open” ends among the total number of ends lassified was determined. It amounted to 58.1% (18/31). When the X-irradiation was replaced by treatment with an effective chromosome-breaking agent, namely FUdR (5-fluorodeoxyuridine), again both “open” and “closed” chromatid fragment ends were observed and a similar fraction of “open” ends was found, namely 43.2% (16/37).  相似文献

In this paper we compare the influence of heterologous and isologous DNA on the radiation damage repair of primary root meristematic cells ofVicia faba. Roots, irradiated by exposure of 150 r were cultivated at different time intervals either in tap water, or in a solution of heterologous or isologous DNA. In comparing mitotic activity of meristematic cells it was found that both types of DNA studied enhance the recovery of irradiated cells. The frequency of postmetaphase chromosomal aberrations of irradiated cells was influenced also by post-irradiation action of exogenous DNA. While heterologous DNA exhibited synergical effect with radiation in the sense that it increased the post-irradiation incidence of aberrations in all time intervals studied, isologous DNA had a strong repair effect—the application caused a significant decrease of the percentage of post-metaphase aberrations. Both kinds of DNA caused changes in the relation of chromosome to chromatid aberrations; a higher percentage of chromatid aberrations was registered. The study of the distribution of aberrations between large and small chromosomes ofVicia faba showed that the post-irradiation application of heterologous DNA increases damage of small chromosomes while isologous DNA caused an increased repair ability in this chromosomal group.  相似文献

The induced meiotic abnormalities as a result of sprayingVicia faba plants with aqueous saturated solutions of podophyllin and 8-hydroxyquinoline were studied. The 2 drugs induced the same types of abnormalities including lagging chromosomes, stickiness, and bridges. Summary The DNA content of nuclei from meristematic root tip cells of five coniferous and one deciduous tree species and, for comparison, ofVicia faba was cytophotometrically determined. The DNA values of diploid nuclei fromGinkgo biloba are approximately a quarter lower than those fromVicia faba. The nuclear DNA values of the other tree species are merely a third to a ninth part of those ofVicia faba. In three tree species, as well as diploid, we have found nuclei of different polyploid level.The reliability of different cytochemical methods, which are used for determination of the nuclear DNA content, is critically analyzed. The DNA values of the investigated tree species are discussed in connection with the evolution of the DNA content in higher plants.Dedicated to Professor Dr. F. Differences as well as similarities in the action of ionizing radiation and deoxyribonucleic acids from various sources on mitosis in root cells ofVicia faba were established. The time course of occurrence of aberrations were examined. Whereas in irradiated broad beant the maximum percentage of aberrations was observed immediately after irradiation, the aps plication of non-isologous DNA was followed by maximum aberrations after 8–16 hours. As all the time-intervals studied, an incraasad number of aberrations was found during metaphase-as compared with anaphases, both after irradiation and after application of DNA. A comparison of isologous, homologous and heterologous DNA as inductors of chromosomal aberrations supported our previous findings and showed that the efficiency of DNA depends on the genetic difference between donor and acceptor. During a study of distribution of aberrations between large and small chromosomes of meristematic cells ofVicia faba, at various time-intervals it was obsarved that after irradiation the distribution of aberrations between individual chromosomes is proportional to their total length, whereas the effect of heterologous DNA is mostly in the damage to small chromosomes. It was also found that aftar irradiation mostly chromatid aberrations are formed at shorter time-intervals and only later chromosomal aberrations will appear. On the other hand, heterologous DNA brings about in all time-intervals a predominance of chromatid aberrations.  相似文献

Summary The karyotypes and the meiotic behaviour of two spontaneous reciprocal translocations and one pericentric inversion are described. One of these translocations was characterized by chiasma-formation in the interstitial-segments, the other was not. The types of orientation and distribution of the chromosomes from the chain-configurations in meta-anaphase I are specified, compared mutually and with those of translocation rings and chains in maize and the similarities and differences are pointed out. In the range of the heterozygous pericentric inversion there was no pairing in meiotic prophase and therefore no chiasma formation and sterility. It will be tried to cross especially one of the translocations with the inversion-type in order to rebuild the karyotype ofVicia faba. In this case it would be possible to distinguish between all five pairs of small chromosomes according to their morphological structure, a possibility very important in localizing chromosome aberrations on the chromosomes ofVicia faba.
