We examined secular changes in mean age at menarche among 5577 Colombian women born between 1941 and 1989, and correlated those changes with nation-wide rates of homicide and real gross domestic product per capita (GDP) at the year of birth and at the year at age 5, within predefined historical periods. The mean (standard error) rate of change in age at menarche by year of birth was −0.55 (0.02) years/decade. The rate of change was not constant, but varied between historical periods as follows: −1.44, −0.14, −0.60, and −0.36 years/decade for the periods 1941–1947, 1948–1958, 1959–1978, and 1979–1989, respectively. The changes in age at menarche correlated positively with the changes in the nation-wide rates of homicide within such periods; i.e. decelerations in the menarcheal trend coincided with increases in the rates of homicide and vice versa. The correlation was higher with the rates of homicide when women were 5 years of age (r = 0.99, p = 0.01) compared to the rates of homicide at the year of birth (0.55, p = 0.45). There were negative correlations between the changes in age at menarche and the changes in GDP, but they were weaker than those with the rates of homicide. These results could suggest a potential impact on maturation of psychosocial stress in childhood due to exposure to a generalized atmosphere of violence and fear.  相似文献

Dose–response relationship between resistance of wheat seedlings (Triticum aestivum, cultivar Zarya) to Erysiphe graminis f. sp. tritici Marchal. (Syn. Blumeria graminis), a causal organism of wheat powdery mildew and exogenous zeatin has been investigated. Two-week-old seedlings were inoculated with the pathogen. Zeatin or zeatinriboside were added to the nutrient solution immediately after inoculation. The dose–response curve of cytokinin in the most cases was multiphasic, with peaks of increased susceptibility occurring at 0.25–1.5 and 1.5–9 μM cytokinin, separated by a region of increased resistance at 0.5–3 μM cytokinin. The change in mineral nutrition or simultaneous treatment with thidiazuron revealed alterations of the dose–response curve ranging from a curve with maximum of resistance to a curve with maximum of susceptibility. Both multiphase nature of dose–response and its variability were proposed as possible explanations for earlier observed discrepancies in experimental data on modification of disease resistance by cytokinins. A mathematical model for two metabolic processes with substrate inhibition connected in-series was suggested to explain the multiphase dose–response. In this model, the product of the first reaction was used as substrate for the second reaction. Numerical experiments showed the changes in the shape of dose–response curve with changes in parameters dependent of cytokinin metabolism.  相似文献

GTPase activating protein (GAP) is a known regulator of ras-p21 activity and is a likely target of ras-induced mitogenic signaling. The domains of GAP that may be involved in this signaling are unknown. In order to infer which domains of GAP may be involved, we have performed molecular dynamics calculations of GAP complexed to wild-type and oncogenic (Val 12–containing) ras-p21, both bound to GTP. We have computed and superimposed the average structures for both complexes and find that there are four domains of GAP that undergo major changes in conformation: residues 821–851, 917–924, 943–953, and 1003–1020. With the exception of the 943–953 domain, none of these domains is involved in making contacts with ras-p21, and all of them occur on the surface of the protein, making them good candidates for effector domains. In addition, three ras-p21 domains undergo major structural changes in the oncogenic p21-GAP complex: 71–76 from the switch 2 domain; 100–108, which interacts with SOS, jun and jun kinase (JNK); and residues 122–138. The change in conformation of the 71–76 domain appears to be induced by changes in conformation in the switch 1 domain (residues 32–40) and in the adjacent domain involving residues 21–31. In an accompanying paper, we present results from microinjection of peptides corresponding to each of these domains into oocytes induced to undergo maturation by oncogenic ras-p21 and by insulin-activated wild-type cellular p21 to determine whether these domain peptides may be involved in ras signaling through GAP.  相似文献

The effect of nitrogen limitation in a medium on the composition of intracellular lipids in the alga Botryococcus braunii Kutz IPPAS H-252 in the course of culture development was investigated. Under the conditions of nitrogen limitation, the alga under investigation accumulated lipids as triacylglycerols, and this process was accompanied by substantial changes in the total fatty acid (FA) composition, which were manifested in a decrease in trienoic acids (from 52.8–57.2 to 19.5–24.7% of total FAs) and an increase in the content of oleic (from 1.1–1.2 to 17.1–24.4%) and saturated (from 23.7–26.0 to 32.9–46.1%) acids. In the control culture, the directionality of FA redistribution was less marked, and these changes were noticed at the later stages of culture development. Under nitrogen limitation, marked changes in the FA composition of polar lipids occurred by the 13th day, and they were characterized by an increase in the content of saturated acids (up to 76.8%) and a dramatic decrease in the content of all polyenoic acids (up to 6.8%). The changes in the FA composition of triacylglycerols were noticed as early as by the 7th day; these changes consisted in an increase in the content of oleic acid, and its high content (28.4–38.4%) was maintained up to the end of culturing. Lake Kilpisjärvi was stocked with a total number of 440 000 migratory and benthic whitefish fry during the period of 1959–1964. Thereafter, the Diphyllobothrium ditremum plerocercoid infection became a problem decreasing the feasibility and commercial exploitation of the whitefish stock. Since dense fish populations may cause stunting, all stockings in the study area were stopped in an attempt to enhance the fisheries. The aim of the present study was to explore the effect of the cessation of the stocking on the growth, condition, catches and D. ditremum infection of the whitefish population. A total of 1594 whitefish were sampled with benthic gill nets. Growth was estimated from length-at-age and mass–length relationship was estimated with a non-linear regression in 1982–1983, 1992–1993 and 1997. Parasite infection was studied in 1992–1993 and 1997. Growth and condition were poor in the beginning of the 1980s. In 1992–1993, the growth and condition of the fish improved as the numerical catch per unit effort decreased from 1982 to 1992. The mean catches of other fish species were low. There were no significant changes in whitefish growth or condition from 1992–1993 to 1997. From the 1970s to 1982–1983, during slow growth and poor condition of the fish, the median gillraker count was decreasing to 19 (min–max 14–26). In 1992–1993, the gillraker count had increased significantly to 23 (min–max 19–26). However, the improved growth and condition did not cause decrease in the D. ditremum burden. On the contrary, based on our own results and literature notes, the parasite abundance seems to increase during the observation period of 1964–1997.  相似文献

The kinetics of xanthan formation in Xanthomonas campestris continuous and fed-batch fermentations was studied along with metabolic changes due to growth rate variation. A maximum growth rate within the range 0.11–0.12 h–1 was obtained from the continuous culture data in defined medium, producing xanthan at rates up to 0.36 g l–1 h–1 corresponding to a maximum 67% glucose conversion at a dilution rate (D) of 0.05 h–1. Comparatively, fed-batch cultivation was more efficient, producing maximum xanthan at 0.75 g l–1 h–1 and 63% glucose conversion at 0.1 h–1. When reaching D=0.062 h–1 in continuous cultures, a change was observed and the values of the specific rate of substrate consumption shifted, initiating an uncoupled growth region expressing a lack of balance of the catabolic and anabolic reactions. The deviation was not accompanied by a change in specific xanthan production indicating that xanthan metabolism was not affected by D. For fed-batch-grown X. campestris cells within the range D=0.03–0.1 h–1, both metabolic parameters changed linearly with the growth rate showing a wide region coupled to growth. Outdoor chemostat cultures of the marine microalgaIsochrysis galbana at constant dilution rate (0.034 h–1 ) have been carried out under different weather conditions. Steady-state biomass concentrations were 1.61±0.03 kg m–3 in May and 0.95±0.04 kg m–3 in July, resulting in biomass output rates of 0.54 kg m–3 d–1 and 0.32 kg m–3 d–1 in May and July, respectively. Two patterns of daily variation with the solar cycle were observed in the fatty acid content. Saturated and mono-unsaturated fatty acids (16:0 and 16:1n7) show significant variation with the solar cycle, associated with short-term changes in environmental factors. Palmitic and palmitoleic acids are generated during daylight and consumed during the dark period. However, polyunsaturated fatty acids do not show a significant response to the solar cycle and their changes are associated with long-term variation in environmental factors. The maximum EPA productivity was obtained in May, 14.1 g m–3 d–1, which is close to that found in the literature for indoor continuous cultures. Nonetheless, the outdoor EPA content (up to 2.61 % d.wt) was lower than the indoor EPA content from a previous study (5% d.wt).  相似文献

The suspension feeding of Bithynia tentaculata was tested in laboratory experiments. The animals were fed in 1-1 aerated glass beakers, and filtration rates were calculated from changes in cell concentrations during the 6-h experiment. Temperature influenced the filtering rate, with minimum values of 5ml · ind–1 · h–1 at 5° C and maxima of 17.2 ml · ind–1 · h–1 at 18° C. Three food species of different size, motility and cell surface characteristics (Chlamydomonas reinhardii, Chlorella vulgaris and Chlorogonium elongatum) did not affect filtration rates. Suspension feeding increased with increasing food concentrations up to 12 nl · ml–1, above which feeding rate was kept constant by lowering the filtering rates. Even the smallest animals tested (<4 mm body length) were found to be feeding on suspended food at a rate of 2.7 ml · ind–1 · h–1, and increasing rates up to 8.4 ml were found in the 6–7 mm size class. All size classes of Bithynia showed a circannual fluctuation of their filtration rates. The ecological consequences of Bithynia's ability to switch between two feeding modes, grazing and suspension feeding, are discussed.  相似文献

Aerated lagoons are commonly used for domestic and industrial wastewater treatment due to their low cost and minimal need of operational requirements. However, little information is known regarding microbial communities that inhabit these ecosystems. In this study, a 16S-DGGE approach was used to estimate bacterial diversity and to monitor community changes in two aerated lagoons from a wastewater treatment plant receiving urban and industrial effluents. Pronounced shifts between bacterial communities collected in winter–spring and summer–autumn months were detected. Temperature, dissolved oxygen (DO) and pH were the variables that most influenced the bacterial communities. Phylogenetic affiliation of predominant members was assessed by the determination of the 16S rDNA sequence of correspondent bands. Affiliations to CytophagaFlexibacterBacteroides (CFB) group, Firmicutes, and β- and ε-proteobacteria were found.  相似文献

Zooplankton community transformation in response to soft water acidification was studied in two experimental series using the microcosm method. The dynamics of total abundance, changes in the proportions of the main zooplankton groups, stability of the dominant complex, and species diversity were evaluated. Zooplankton population proved to decrease at all studied low pH (5–6, 4–5, and 3–4) within the first two weeks relative to the control. At water pH 5–6, either Rotifera-Copepoda or Cladocera-Copepoda zooplankton complex was established depending on the initial community composition; while at pH 4–5, Copepoda predominated in the established zooplankton community. The community edificator species was replaced at pH 4–5. The lowest diversity index was also observed at this pH (1.0–1.5 bit/ind.). In addition, the proportion and absolute population of copepod nauplii and cladoceran Scapholeberis mucronata increased at low pH. Lactic acid was added to batch very high gravity (VHG) fermentations and to continuous VHG fermentations equilibrated to steady state with Saccharomyces cerevisiae. A 53% reduction in colony-forming units (CFU) ml–1 of S. cerevisiae was observed in continuous fermentation at an undissociated lactic acid concentration of 3.44% w/v; and greater than 99.9% reduction was evident at 5.35% w/v lactic acid. The differences in yeast cell number in these fermentations were not due to pH, since batch fermentations over a pH range of 2.5–5.0 did not lead to changes in growth rate. Similar fermentations performed in batch showed that growth inhibition with added lactic acid was nearly identical. This indicates that the apparent high resistance of S. cerevisiae to lactic acid in continuous VHG fermentations is not a function of culture mode. Although the total amount of ethanol decreased from 48.7 g l–1 to 14.5 g l–1 when 4.74% w/v undissociated lactic acid was added, the specific ethanol productivity increased ca. 3.2-fold (from 7.42×10–7 g to 24.0×10–7 g ethanol CFU–1 h–1), which indicated that lactic acid stress improved the ethanol production of each surviving cell. In multistage continuous fermentations, lactic acid was not responsible for the 83% (CFU ml–1) reduction in viable S. cerevisiae yeasts when Lactobacillus paracasei was introduced to the system at a controlled pH of 6.0. The competition for trace nutrients in those fermentations and not lactic acid produced by L. paracasei likely caused the yeast inhibition.  相似文献

Summary The possible regulation of Cl influx inChara by the cytoplasmic Cl concentration and cytoplasmic pH was investigated using both intact and intracellularly perfused cells. In perfused cells Cl influx was sensitive to changes in the internal Cl concentration but only when the concentration was less than 1mm.In intact cells the metabolic inhibitors, CCCP, DCMU, and oligomycin which inhibit Cl influx also reduced the cytoplasmic pH. A correlation between ATP concentration and cytoplasmic pH was shown to apply when the ATP concentration was lowered using these inhibitors. The possible relationships between ATP status, cytoplasmic pH, and Cl influx are discussed.  相似文献

Summary Patterns of herbivory over a two year period on Quercus emoryi (Fagaceae) were correlated with seasonal and yearly changes in tannin and protein content. Quantitative defense theory predicts that tannin and protein content in apparent plants should be negatively and positively correlated, respectively, with degree of herbivory. Most herbivory occurred early in the growing season, but the pattern varied between the two years. Tannin and protein content sometimes varied negatively and sometimes positively with degree of herbivory; they did not consistently covary with herbivory. Protein content was positively correlated with herbivory in 1981–1982 but not in 1982–1983. Condensed tannin content was negatively correlated with herbivory in 1981–1982 but not in 1982–1983. Hydrolysable tannin content was positively correlated with herbivory activity. Multiple regression analyses indicated these phytochemical variables explained either no significant variation in herbivory (1982–1983) or did so in a fashion opposite (1981–1982) to the predictions of the theory of quantitative defense.Feeding by oak insects was not solely a function of seasonal changes in quantitative defenses and nutrients. Obviously, population dynamics of the insects are sensitive to factors other than phytochemistry of the trees and I discuss other factors that can influence patterns of herbivory.  相似文献

Audouinella saviana (Meneghini) Woelkerling was cultured at a constant temperature (24 °C) and different irradiances (from 1 µmol to 30 µmol photons m–2 s–1) of blue (430–470 nm) and green (500–560 nm) light in order to study its adaptive response. Modifications in colour, morphology and ultrastructure of the thalli, together with changes in pigment composition and in the spectral properties of chlorophyll a and R-phycoerythrin, were observed both by means of light and electron microscopy (TEM, SEM) and spectrophotometric and spectrofluorimetric analyses. In this paper we report the adaptive response of the seaweed to blue and green radiation by focussing on the cell wall and on the photosynthetic apparatus, particularly on phycobilisomes in situ and on R-PE after extraction. PBSs were fully structured only under blue light at low irradiance whilst they were absent under green light, whatever the irradiance, in spite of the high R-PE content. This fact, together with the spectral changes of R-PE, suggests adaptation at a molecular level, presumably referable to changes in aggregation state.  相似文献

The R-R intervals were measured in 25 female volunteers aged 21–34 years after exposure to lower body negative pressure (LBNP). A number of advantages of information entropy (h) as an index of cardiovascular performance were revealed. With h used as a controlled variable, it was possible to ensure individual targeted changes in the functional state (FS) during LBNP. It was found that an individual exposure to LBNP caused regular changes in h and FS. Cluster roots are adaptations for nutrient acquisition, found throughout the world in many different plant families and habitats. They arise from changes in root initiation, meristem maintenance and physiology. In Lupinus albus cluster roots form under low internal plant phosphate and low internal plant iron levels. In this study, we compare morphology, structure and physiology of cluster roots formed under –P and –Fe conditions. –Fe cluster roots had a lower density of shorter rootlets than –P roots, and were yellow in colour, probably because of increased phenolics due to down-regulation of peroxidase. Rootlet length and width was reduced in –Fe conditions. The change in exudation of citrate, over time, of –P and –Fe cluster roots shared identical temporal dynamics, with an exudative burst occurring in day 3. However, the –Fe cluster roots displayed much higher rates of exudation than the –P cluster roots. Results are discussed within the context of structural and functional control.  相似文献

An aerobiological study was performed to evaluate the potential exposure of animals and workers to dust constituents generated during routine animal house work. Different rooms of air conditioned (A, control) and passively ventilated (B, non-air conditioned) animal facilities were sampled, in order to evaluate total airborne culturable fungi and bacteria, fungal spore concentrations and particle levels. Airborne room particles were analyzed gravimetrically and for endotoxin content. All parameters, except for culturable fungi, were higher in facility B and statistically significant, with respect to those from the control facility A. Median values for airborne particle concentration, endotoxin and fungal spores in facility B were: 115 µg m–3, 25 EU m–3, and 2173 spores m–3, respectively. Median values for facility A were: 66 µg m–3, 9 EU m–3, and 248 fungal spores m–3. Broncheoalveolar lavage from rats kept in the rat room of B, presented median concentrations of total cells and lactate dehydrogenase, higher than those found in the control facility (4.4 × 105 vs. 1.1 × 105 and 2.7 UmL-1 vs. 0.39 UmL–1, respectively). Values of total and biological particles of both facilities, as well as the time spent in different rooms, showed that worker exposure was higher during cage washing. It was especially high in the passively ventilated facility (airborne particles 686 µg m–3 3.5 h–1 vs. 976 µg m–3 3.5 h–1, endotoxin 70 EU m–3 3.5 h–1 vs. 108 EU m–3 3.5 h–1). The type of basidiospores and ascospores found, as well as the significant correlation between particle levels and endotoxin contents suggests that wood chip bedding disturbance during cage washing is an important source for airborne biological particles. The changes in broncheoalveolar lavage components found in rats from these facilities and previously reported changes in pro-inflammatory cellular responses found in workers, indicate that these relatively low levels of exposure are enough to induce a biological response. Studies considering the composition of mixed organic dusts, would be needed to reevaluate current occupational standards.  相似文献

Summary Experiments were performed in intact proximal tubules of the doubly perfused kidney and in fused proximal tubule cells ofRaha esculenta to evaluate the dependence of intracellular pH (pHi) on cell membrane potential applying pH-sensitive and conventional microelectrodes. In proximal tubules an increase of the K concentration in the peritubular perfusate from 3 to 15 mmol/liter decreased the peritubular cell membrane potential from –55±2 to –38±1 mV paralleled by an increase of pH i , from 7.54±0.02 to 7.66±0.02. The stilbene derivative DIDS hyperpolarized the cell membrane potential from –57 ± 2 to –71 ±4 mV and led to a significant increase of the K-induced cell membrane depolarization, but prevented the K-induced intracellular alkalinization. Fused proximal tubule cells were impaled by three microelectrodes simultaneously and cell voltage was clamped stepwise while pH i changes were monitored. Cell membrane hyperpolarization acidified the cell cytoplasm in a linear relationship. This voltage-induced intracellular acidification was reduced to about one-third when HCO3 ions were omitted from the extracellular medium. We conclude that in proximal tubule cells pH i depends on cell voltage due to the rheogenicity of the HCO 3 transport system.  相似文献

The conventional microelectrode technique was applied to study changes in conductance and activation characteristics of potassium and chloride channels in the plasmalemma of characean alga Nitella flexilis(L.) Agardz. during long-term heat treatment. Measurements were conducted at 18–20°C after preliminary exposure of cells to 33°C for 1–25 days. The conductance of outward- and inward-rectifying potassium channels, as well as the currents of excitable chloride channels, decreased after 2–3 days of heat treatment. By the 15th–17th days, the conductance of potassium channels was reduced by a factor of 3–5, whereas the peak values of the chloride current, associated with the action potential, was reduced by a factor of 8–10. These heat-induced changes were long lasting: the restoration of the initial parameters of transport systems after transferring cells to chilling or room temperature occurred within several days. Moreover, the recovery at chilling temperatures (8–10°C) proceeded nearly two times longer than at room temperature. Prolonged hyperthermia accelerated activation and deactivation of outward-rectifying potassium channels and caused the shift of their activation curve towards positive potentials by 35–40 mV. Analysis of current–voltage relations showed that the inward current in inward- and outward-rectifying potassium channels was reduced to a greater extent than the outward current. At the same time, both inward and outward currents of chloride channels were reduced to an equal extent. It is assumed that the changes observed are involved in thermal adaptation and account for the decrease in the intracellular concentrations of potassium and other cations and anions, which represents a nonspecific response of plant cells to stress.  相似文献

The population growth pattern and related changes in both the nitrogen and phosphorus contents in the cell of the dinoflagellate Peridinium penardii (Lemm.) Lemm., which formed a freshwater red tide in a reservoir, were studied in situ. An exponential increase with time in population density was found. A specific growth rate of 0.25 d–1 was observed. The cellular content of phosphorus per cell decreased from 6.0 × 10–5 µg to 9.2 × 10–6 µg along with an increase in population density from 8.0 × 102 cells ml–1 to 2.5 × 104 cells ml–1. A prominent change in the cellular nitrogen did not occur. Decreasing cell content and continuous uptake of phosphorus were advantageous for P. penardii to form a freshwater red tide under P-limited conditions.  相似文献
