Virus infection is established when viral proteins can interact with host factors to execute replication and/or cell-to-cell movement. Even after the virus infection has started, host resistance reactions, if trigged, would suppress further virus propagation. We would like to introduce what we understand about host factors as determinants of infection establishment and as key resistance molecules. Genome-wide information of Arabidopsis is providing us much information about such host factors involved in virus infection.  相似文献

ABSTRACT. A microsporidian parasite, Nosema muscidifuracis n. sp., has been found in Muscidifurax raptor , a parasitoid of muscoid flies. Stages of the parasite developed in direct contact with the host cell cytoplasm and were detected in midgut epithelium, Malpighian tubules, ovaries (including oocytes) and fat body of larvae and adults. Spores were also detected within eggs deposited on the host. Light and electron microscopy revealed a developmental cycle with diplokaryotic stages dividing by binary fission and disporous sporulation sequences producing diplokaryotic spores of three morphological classes, differing significantly only in length of the polar filament. Two of the classes were found in larvae, pupae and adults. One of these, with about five turns in the coiled polar filament, is presumed to be responsible for transmission from cell to cell within the host (autoinfection) and the other, with about 10 turns, responsible for transmission from host to host. A third class, with about 15 turns in the polar filament, was found in eggs of M. raptor . It is, presumably, either involved in initiation and spread of the infection at eclosion or is responsible for horizontal transmission to a new host individual when eggs are cannibalized.  相似文献

Gas exchange characteristics of a hemiparasiteMelampyrum arvense L. before and after attachment to the hostCapsella bursa pastoris (L.) Med. were compared. The net photosynthetic rates (PN) on a leaf area basis were extremely low and in comparison to the value obtained for the host were about 15 % and 23 % for the unattached and attached hemiparasite, respectively. Also the concentration of photosynthetic pigments was low (as compared with the host the content of chlorophylls was about 33 % and 49 % and of carotenoids about 38 % and 36 % in the unattached and attached hemiparasite, respectively). On the other hand the rates of respiration were high (about 1.8 and 2.6 times higher in the unattached and attached hemiparasite, respectively, than in the host). In darkness stomatal conductance (gS) of the host and the unattached hemiparasite was rapidly reduced to 10 % of the value obtained in light, gS of the attached hemiparasite was decreased only by about 30%. A total reduction of gS occurred at relative water content (RWC) of 85 %, 75% and 45 % for the unattached hemiparasite, the host, and the attached hemiparasite, respectively. The transpiration (E) rate in the preparasitic stage was very low, being 2.6 and 4.5 times smaller than in the host and the attached hemiparasite, respectively. In the attached hemiparasite WUE was 7.5 and 3 times poorer than in the host and in the preparasitic stage, respectively.  相似文献

Abstract.— Virulence is of central importance in host-parasite interactions, yet little is known about how it changes over extended evolutionary periods. In this study, all four species in the testacea species group of Drosophila were experimentally infected with sympatric and allopatric nematodes in the Howardula aoronymphium species complex, and the effect of parasite infection on three components of host fitness was determined. The Drosophila species show striking differences in their responses to infection, with reductions reaching 80% in adult lifespan, 100% in female fertility, and 90% in male fertility. Female sterility appears to be determined by the host; species that are sterilized by their local nematodes are also sterilized by the other allopatric nematodes in the H. aoronymphium complex. Host species that are not sterilized by their local parasite are not sterilized by other nematodes in the complex. In contrast, reductions in host adult lifespan and male fertility depend on both the host and the parasite. Whereas all nematodes reduced the survival of their local host species equally (about 40–45%), survival of two host species was drastically reduced (about 80%) when infected with an allopatric parasite. Thus, virulence is evolutionarily labile in associations between Drosophila testacea group species and their Howardula parasites. The data suggest that changes in the sterility component of virulence are due primarily to host evolution, whereas changes in the host mortality component are due in large part to parasite evolution.  相似文献

Summary Host type choice and sex allocation were examined using the solitary parasitic waspDinarmus basalis (Pteromalidae, Hymenoptera) parasitizing larvae or pupae of the bean weevilCallosobruchus chinensis (Bruchidae, Coleoptera) within azuki beans (Vigna angularis). The wasps were offered two types of host; one was hard for the mother to lay eggs in, but was more beneficial for the offspring; the other was easy for the mother to lay eggs in, but was less beneficial for the offspring. The two types of host were one large host (17-day old host) in one bean and 6 small hosts (12-, or 13-day old hosts) in one bean. The same number of each host was presented at the same time to female wasps. The wasps accepted more 17-day old hosts than 12-day old hosts, and more 13-day old hosts than 17-day old hosts in each pair-wise choice experiment. The proportions of accepted host types were different from the proportions predicted by optimization models of random prey encounter with known or unknown prey densities. The wasps showed partial preference of host types. Incomplete information about prey densities, and about the costs and benefits of the two types of host may have generated the partial preference. Two predictions of host sizemodels, that (1) there should be a negative relationship between host size and offspring sex ratio (proportion of male offsprings), and (2) the sex ratio in each size host changes with the relative frequency of each size host utilized, were qualitatively supported.  相似文献

Plasmid DNA was used to study gamma-radiation-induced recombination and mutagenesis in Escherichia coli host cells. Plasmid pBRP1, a derivative of pBR322 containing the lac operon of E. coli, was irradiated with 60Co gamma rays prior to transformation into E. coli strains of different recA and lac genotypes. Plasmid-chromosome recombination was assayed in lacY1 host cells, whereas plasmid mutagenesis was assayed in delta lac host cells lacking chromosomal sequences homologous to the plasmid. Both recombinant and mutant plasmids were identified by the phenotypic changes in lactose utilization, and confirmed by restriction analysis of isolated plasmids. Plasmid-chromosome recombination was induced to high levels (about 20% of survivors at 700 Gy) and was dependent on the host recA gene. Plasmid mutagenesis occurred at lower levels (about 1.5% of survivors at 600 Gy) and was relatively independent of the recA gene. Plasmid survival was unaffected by the presence or absence of host recA mutations or the potential for plasmid-chromosome recombination.  相似文献

The content of abscisic acid (ABA) in abaxial leaf epidermis of the host (Capsella bursa pastoris) and the unattached hemiparasiteMelampyrum arvense showed diurnal changes. ABA content increased during the light period and declined rapidly upon the darkening of leaves. In an attached hemiparasite the content of ABA in the epidermis was maintained at an almost constant level irrespective of the diurnal cycle. As compared with the maximum level in the host, at the end of the light phase the content of ABA in abaxial epidermis constituted about 70 % and 164 % in the unattached and attached hemiparasite, respectively. No significant changes in ABA content were recorded in adaxial epidermis. In all the samples abaxial/adaxial epidermis ABA content ratio was about 3.6:1 in light phase. In darkness this ratio decreased to about 1.1:1 in the host and the unattached hemiparasite and did not show significant change after attachment. ABA content ratio in mesophyll was 1:0.7:1.5 for the host, the unattached, and attached hemiparasite, respectively. In comparison with the host the concentration of ABA in xylem sap of the hemiparasite constituted about 31 % and 152 % for the unattached and attachedM. arvense, respectively.  相似文献

For the majority of eriophyoid species, host ranges have been established purely on the basis of collection records, usually without quantitative data. The aim of this study was to: (1) quantitatively examine published literature to explore whether relevant analyses of field-collected quantitative data were used to assess host specificity of herbivores; (2) propose a protocol for data analysis that could be applied to plant-feeding mites; (3) analyse host specificity of the grass-feeding Aceria species as a case study. Field data were collected in Central and Northern Europe over a period of 11 years, and included 73 grass species. For the eight Aceria species found, infestation parameters and host specificity indexes were assessed. Accumulation curves were calculated to study how the sampling effort influenced estimates of host specificity indexes. A literature analysis showed that among the studies that declared an aim of estimating the host range only 56% of them applied any quantitative analysis or informed on estimation reliability. The analysis of field-collected data and its interpretation showed the most complete and reliable conclusions about the host specificity of Aceria species when all indices were considered and, if available, other information about the mite’s ecology and biology. It was shown that estimates of host specificity could be strongly affected by sampling effort, and that several hundreds of samples should be collected for measuring the host specificity of grass-infesting mites, at least. Recommendations regarding host specificity estimation on the basis of field data are given.  相似文献

报道我国内陆水体单殖吸虫的宿主特异性的研究。研究表明单殖吸虫有很强的宿主特异性,但不同种类的特异的有很大的判别。从目前已有的记录来看,60%以上的单殖吸虫中只有1种宿主,约75%的单殖吸虫的宿主仅为1属,超过97%的单殖吸虫的宿主在1科之内。没有1种单殖吸虫可生于超过3个料的宿主中,但大钩指环虫可在23属33种宿主中寄生。在科和发放持异性水平,以锚首虫科和指环虫科单殖吸虫的特异性较强,三代虫科次之  相似文献

Omic approaches to the analysis of plant-virus interactions are becoming increasingly popular. These types of data, in combination with models of interaction networks, will aid in revealing not only host components that are important for the virus life cycle, but also general patterns about the way in which different viruses manipulate host regulation of gene expression for their own benefit and possible mechanisms by which viruses evade host defenses. Here, we review studies identifying host genes regulated by viruses and discuss how these genes integrate in host regulatory and interaction networks, with a particular focus on the physical properties of these networks.  相似文献

The Oscillation Hypothesis posits that plant‐eating insect diversity is generated by cycles of diet breadth expansion and contraction. Although at any given time most plant‐eating insect species are host specialists, host‐use evolution and speciation tend to entail a phase of generalism. The main evidence for this comes from comparative phylogenetic studies, but with mixed support. Here, I review and add to this evidence. I show that some of the original work that inspired the Oscillation Hypothesis is flawed in a way that leads to spurious inferences about trends in the evolution of diet diversity. And I present a new analysis which fails to support its predictions about patterns of species diversity. On the other hand, some of the published work that claims to reject the Oscillation Hypothesis may actually provide some of the strongest support for it, and I present new analyses which support its prediction that host‐use generalism facilitates host‐use evolution. In summary, the Oscillation Hypothesis successfully predicts some phylogenetic patterns but not others. Generalism appears to facilitate host‐use evolution, but it does not appear to be inevitably chased by host‐use specialization and speciation.  相似文献

Research on effectors secreted by pathogens during host attack has dominated the field of molecular plant-microbe interactions over recent years. Functional analysis of type III secreted effectors injected by pathogenic bacteria into host cells has significantly advanced the field and demonstrated that many function to suppress host defense. Fungal and oomycete effectors are delivered outside the host plasma membrane, and although research has lagged behind on bacterial effectors, we are gradually learning more and more about the functions of these effectors. While some function outside the host cell to disarm defense, others exploit host cellular uptake mechanisms to suppress defense or liberate nutrients intracellularly. Comparative genomics suggests that the organization of effector genes drives effector evolution in many pathogen genomes.  相似文献

Plasmodium falciparum, the most virulent agent of human malaria, undergoes both asexual cycling and sexual differentiation inside erythrocytes. As the intraerythrocytic parasite develops it increases in size and alters the permeability of the host cell plasma membrane. An intriguing question is: how is the integrity of the host erythrocyte maintained during the intraerythrocytic cycle? We have used water window cryo X-ray tomography to determine cell morphology and hemoglobin content at different stages of asexual and sexual differentiation. The cryo stabilization preserves native structure permitting accurate analyses of parasite and host cell volumes. Absorption of soft X-rays by protein adheres to Beer–Lambert’s law permitting quantitation of the concentration of hemoglobin in the host cell compartment. During asexual development the volume of the parasite reaches about 50% of the uninfected erythrocyte volume but the infected erythrocyte volume remains relatively constant. The total hemoglobin content gradually decreases during the 48 h cycle but its concentration remains constant until early trophozoite stage, decreases by 25%, then remains constant again until just prior to rupture. During early sexual development the gametocyte has a similar morphology to a trophozoite but then undergoes a dramatic shape change. Our cryo X-ray tomography analysis reveals that about 70% of the host cell hemoglobin is taken up and digested during gametocyte development and the parasite eventually occupies about 50% of the uninfected erythrocyte volume. The total volume of the infected erythrocyte remains constant, apart from some reversible shrinkage at stage IV, while the concentration of hemoglobin decreases to about 70% of that in an uninfected erythrocyte.  相似文献

The ornithine aminotransferase [EC 2.6.1.13] content of Morris hepatoma 44 is about 15 times higher than that in normal liver. The turnover rates of ornithine amino-transferase in hepatoma 44 and host liver were determined using L-[14C]leucine. Studies on the incorporation of radioactive leucine into ornithine aminotransferase in rats bearing hepatoma 44 showed that the rate of synthesis of this enzyme in the hepatoma was about 5-fold higher than that in host liver. The half-life of ornithine aminotransferase in host liver was 0.98 day, which was the same as that in normal liver, whereas that in hepatoma 44 was 3.5 days. The rate constant of degradation of ornithine aminotransferase in hepatoma 44 was significantly less than that in host liver. These results show that the high ornithine aminotransferase content of hepatoma 44 is due to both increase in its rate of synthesis and decrease in its rate of degradation.  相似文献

Plants are resistant to most potentially pathogenic microbes. This forces plant pathogens to develop sophisticated strategies to overcome basic plant resistance, either by masking intrusion or by suppression of host defences. This is particularly true for fungal pathogens, which establish long lasting interactions with living host tissue, without causing visible damage to invaded cells. The interactions of cereal crops and Arabidopsis with powdery mildew fungi are model systems for understanding host resistance. Currently, these systems are also promoting the understanding of fungal infection by identifying fungal pathogenicity and virulence factors and host target sites. This minireview focuses on recent findings about host susceptibility and the way powdery mildew fungi might induce it.  相似文献

The quantification of host-parasite associations from field data is a fundamental step towards understanding host-parasite and host-parasite-pathogen dynamics. For parasites that are not rigid host specialists, exemplified in this paper by ticks, the interpretation of host-parasite association data is difficult. Interpretations of tick collection records have largely assumed that off-host collection records offer a valid basis from which to make claims about the host specificity or generality of tick species. A simple simulation analysis of rudimentary tick-host interactions in a hypothetical 50 x 50-cell habitat demonstrates that perceptions of tick-host relationships can be strongly biased by spatial patterns. Regardless of their true level of host specificity or generality, it seems that: (i) more abundant ticks will be perceived as generalists, while rarer species will be considered specialists; and (ii) tick species that have patchy, strongly aggregated distributions will be more likely to be perceived as host specialists than species that have more dispersed or uniform distributions. Since all available evidence suggests that abundances and spatial patterns vary between tick species, there is no way of assessing the true validity of claims about host specificity without first undertaking detailed research on the relative abundances and spatial and temporal patterns of both tick and host distributions.  相似文献

Abstract: Parasitic angiosperms are a taxonomically diverse group of plants that invade host tissues and remove resources via a specialized structure known as the haustorium. Through the haustorium, carbon, water and mineral nutrients are withdrawn, often at the expense of host growth and vigour. In addition to the removal of resources from host plants, many parasitic angiosperms are also able to impact host growth via effects on host photosynthesis. In this paper we review what is known about how parasitic angiosperms affect host photosynthesis and the impact this has on host productivity. Holoparasites, that lack chlorophyll, act as extra sinks for host photosynthates and generally either enhance or have a neutral effect on host photosynthetic productivity. In contrast, hemiparasites, that are capable of some autotrophic carbon fixation, usually have a negative impact on host photosynthesis. Irrespective of the outcome of infection, the mechanisms involved in altering host photosynthesis are diverse and may act at either the leaf or whole-plant level. In some cases, parasites impact directly on host photosynthetic metabolism, while in others the effects are more indirect, for example through changing host architecture.  相似文献

The release of several R factor and chromosomal beta-lactamases by osmotic shock treatment was studied. It was found that those beta-lactamases with a molecular weight of about 20,000 were released, but those with a molecular weight of about 30,000 to 44,000 were not released during osmotic shock. This differential release did not depend on whether the structural genes were on the chromosome or on the genome of an R factor. The release or retention of the beta-lactamases appeared to be a characteristic of the enzyme rather than the host cell since the same results were obtained when the R factors were harbored by a variety of host bacteria. Studies with bacteria which produced more than one beta-lactamase showed that each enzyme reacted independently to the presence of other beta-lactamases produced by the host bacterium.  相似文献
