Plant establishment patterns suggest that ectomycorrhizal fungal (EMF) inoculant is not found ubiquitously. The role of animal vectors dispersing viable EMF spores is well documented. Here we investigate the role of wind in basidiospore dispersal for six EMF species, Inocybe lacera, Laccaria laccata, Lactarius rufus, Suillus brevipes, Suillus tomentosus and Thelephora americana. Basidiospores adhered to microscope slides placed on three 60 cm transects radiating from sporocarps. Morphological characteristics of species as well as average basidiospore volume were recorded. Number of basidiospores was quantified at specific distances to produce actual dispersal gradients. We found a negative exponential decay model using characteristics for each species fit the field data well. The 95% modeled downwind dispersal distance of basidiospores was calculated for each species. The 95% modeled downwind dispersal distance increased with increasing cap height and decreasing basidiospore volume for the species sampled, with 95% of basidiospores predicted to fall within 58 cm of the cap. Differences in anatomical characteristics of EMF species influence how far basidiospores are dispersed by wind. We discuss the role of wind dispersal leading to patterns of EMF establishment during primary succession.  相似文献

Muscina angustifrons (Diptera: Muscidae) is a mycophagous species that exploits a variety of fungi, including ectomycorrhizal fungi. Larvae of this species have been shown to feed on sporocarps (including spores), and full-grown larvae leave sporocarps and pupate 0–6?cm below the soil surface. In this study, we examined whether M. angustifrons larvae are capable of transporting ectomycorrhizal fungal spores and enhancing ectomycorrhiza growth on host-plant roots. Full-grown larvae were found to move horizontally 10–20?cm from their feeding sites and burrow underground. These wandering larvae retained ectomycorrhizal fungal spores in their intestines, which were excreted following relocation to underground pupation sites. Excreted spores retained germination and infection capacities to form ectomycorrhiza on host-plant roots. In the infection experiments, ectomycorrhizal fungal spores applied in the vicinity of underground host-plant roots were more effective in forming ectomycorrhiza than those applied to the ground surface, suggesting that belowground transportation of spores by M. angustifrons larvae could enhance ectomycorrhizal formation. These results suggested that M. angustifrons larvae act as a short-distance spore transporter of ectomycorrhizal fungi.  相似文献

Effects of manipulation of litter and humus layers (removal, doubling and control treatments) on the colonization potential of ectomycorrhizal fungi were studied in two secondary stands of Pinus sylvestris (5 and 18 years old) in The Netherlands. Five-mont-hold, sterile-grown Scots pine seedlings, inoculated with Laccaria bicolor, Paxillus involutus or Rhizopogon luteolus and noninoculated seedlings were used as baits. The seedlings were harvested after one growing season. For comparison, sporocarps of ectomycorrhizal fungi were also investigated. Genus composition on the seedlings was independent of initial inoculum, but determined by both treatment and age of the stands. In both stands, removal of litter and humus layers increased, and addition of organic material decreased the number of ectomycorrhizal types on the seedlings. Not all indigenous genera were observed by either outplanting seedlings or sporocarp surveys.  相似文献

To understand the reproduction of the pioneer ectomycorrhizal fungi Laccaria amethystina and Laccaria laccata in a volcanic desert on Mount Fuji, Japan, the in situ genet dynamics of sporocarps were analysed. Sporocarps of the two Laccaria species were sampled at fine and large scales for 3 and 2 consecutive years, respectively, and were genotyped using microsatellite markers. In the fine-scale analysis, we found many small genets, the majority of which appeared and disappeared annually. The high densities and annual renewal of Laccaria genets indicate frequent turnover by sexual reproduction via spores. In the large-scale analysis, we found positive spatial autocorrelations in the shortest distance class. An allele-clustering analysis also showed that several alleles were distributed in only a small, localised region. These results indicate that Laccaria spores contributing to sexual reproduction may be dispersed only short distances from sporocarps that would have themselves been established via rare, long-distance spore dispersal. This combination of rare, long-distance and frequent, short-distance Laccaria spore dispersal is reflected in the establishment pattern of seeds of their host, Salix reinii.  相似文献

The biodiversity of saprotrophic and ectomycorrhizal basidiomycetous macrofungi growing on seven islands in central Japan were compared to examine colonizing success within the context of island biogeography theory. Two hypotheses were tested: that the number of the fungal species depends on island area and that the slope of the species-area curve for saprotrophic and ectomycorrhizal macrofungi differ in response to differences in their nutritional requirements. Data for the number of species that were identified based on sporocarps closely fit the conventional species-area curve. The slopes of the species-area curve for saprotrophic fungi (0.316) and ectomycorrhizal fungi (0.469) were similar to those reported for insects and birds, and plants on other archipelagos, respectively. In addition species-area curve data showed that ectomycorrhizal fungi colonized only islands > 630 m(2). While the species composition of saprotrophic fungi found on any pair of islands was positively correlated to the ratio of the areas of the island pair being compared (smaller/larger), no such relationship was observed for ectomycorrhizal fungi. Conversely similar ectomycorrhizal fungi, mostly those belonging to the genera Amanita, Inocybe, Boletellus and Russula, were found on pairs of islands with similar vegetation in the same geographic region. These results suggested that the colonizing success by ectomycorrhizal fungi is limited by host plant diversity, which is lower on smaller islands, instead of restricted immigration resulting from limited spore dispersal ability.  相似文献

食用外共生菌根菌是一类可食用并与植物共生的大型真菌,其在纯培养条件下菌丝生长缓慢,不会扭结发育成原基,不能完成生活史。因此关于食用菌根菌纯培养条件下生活史的研究报道极少。本研究的兰茂牛肝菌Lanmaoa asiatica已能在纯培养条件下诱导出原基,这使生活史的研究得以完成。利用扫描电子显微镜、透射电子显微镜及荧光显微镜对兰茂牛肝菌子实体担子、担孢子、纯培养菌丝和原基的核相进行了观察。结果表明,子实体菌丝细胞为双核,担子经过减数分裂形成4个单核担孢子;担孢子萌发时长轴端长出芽状突起伸长为菌丝,原孢子中的细胞核不分裂,直接进入菌丝,形成单核初生菌丝;初生菌丝5d后变为双核菌丝;纯培养菌丝及原基细胞均为双核,其菌丝表面光滑,未观察到锁状联合。  相似文献

In most ectomycorrhizal (EM) community studies involving molecular identification methods there is a poor correspondence between fungi that appear dominant as sporocarps and those that appear dominant on EM roots and the species richness belowground is higher than that above ground. As a consequence, many fungi from root tip samples remain unidentified. In most studies, genetic data from multiple samples of an EM morphotype collected from various sampling locations are compared to genetic data from one to a few sporocarps of each species for identification purposes. The mismatch between above- and belowground species richness may be influenced by these different sampling efforts. To address this, intra-specific variation in the ITS region first investigated in Kårén et al. (1997) is revisited here, but at a spatial scale in which variation is expected to be low. Sporocarps were collected across a 7 km region of the Oregon Dunes National Recreation Area in western North America. ITS–RFLP data are presented for 3-18 sporocarps from each of 44 EM species in 18 genera. A total of 311 sporocarps were analyzed. Fifty-three ITS–RFLP types were observed. Of the 44 species, 38 (86% of total) yielded a single, species specific, RFLP type. No 2 species had the same RFLP type. Polymorphic ITS–RFLP types were observed in six species (14%). The following three species had two ITS-RFLP types with one type dominating: Inocybe lacera, Laccaria proxima, and Rhizopogon subcaerulescens. The following three species had three RFLP types with no type dominating: Laccaria laccata, Lactarius deliciosus, and Tricholoma flavovirens. A phylogenetic analysis of ITS sequences in Tricholoma revealed that two of the RFLP types in T. flavovirenswere apparently the result of intra-specific variation, while the third RFLP type was likely a cryptic species. All the other RFLP types observed in Tricholoma represented unique phylogenetic species. These results suggest that ITS–RFLP data from single samples (sporocarp or EM) are robust for characterizing most of the species at this scale. However, restriction endonucleases detect a limited amount of existing nucleotide variation and thus have limited value to detect cryptic species. Therefore, additional analyses of sequence data should be added to the RFLP matching technique to identify unknown RFLP types. These data also suggest that polymorphic RFLP types within species do not adequately explain the mismatch between above- and belowground views of EM species richness.  相似文献

The aim of this study is to assess the mycorrhizal status of Eucalyptus plantations in south China and to determine the need for inoculation. In four provinces in south China, 155 plantations were sampled for sporocarps of ectomycorrhizal (ECM) fungi, spores of arbuscular mycorrhizal (AM) fungi, and mycorrhizas over 2 years. This study revealed a low above-ground diversity of ECM fungi consisting of 15 taxa fruiting beneath Eucalyptus plantations. The most common ECM genera were Scleroderma and Pisolithus, but they were infrequent. A total of 21 AM fungi, mostly Glomus species, were recognized from spores collected from eucalypt plantations. Four Glomus species were frequently present in soils, but spore density and relative abundance of AM fungi were generally low. Eucalypt roots from all plantation sites were poorly colonized by either ECM fungi or AM fungi. A bioassay with E. urophylla as a bait host, using soils collected from 11 eucalypt plantations, confirmed low levels of inoculum of both ECM and AM fungi in field soil. This is the first integrated study on the mycorrhizal status of eucalypt plantations in China. Findings from this research can be used to encourage adoption of mycorrhizal technology by eucalypt nurseries in the region. The potential of using spores of compatible ECM fungi or collections for forest nurseries is discussed.  相似文献

Across three tropical Australian sclerophyll forest types, site-specific environmental variables could explain the distribution of both quantity (abundance and biomass) and richness (genus and species) of hypogeous fungi sporocarps. Quantity was significantly higher in the Allocasuarina forest sites that had high soil nitrogen but low phosphorous. Three genera of hypogeous fungi were found exclusively in Allocasuarina forest sites including Gummiglobus, Labyrinthomyces and Octaviania, as were some species of Castoreum, Chondrogaster, Endogone, Hysterangium and Russula. However, the forest types did not all group according to site-scale variables and subsequently the taxonomic assemblages were not significantly different between the three forest types. At site scale, significant negative relationships were found between phosphorous concentration and the quantity of hypogeous fungi sporocarps. Using a multivariate information theoretic approach, there were other more plausible models to explain the patterns of sporocarp richness. Both the mean number of fungal genera and species increased with the number of Allocasuarina stems, at the same time decreasing with the number of Eucalyptus stems. The optimal conditions for promoting hypogeous fungi sporocarp quantity and sporocarp richness appear to be related to the presence and abundance of Allocasuarina (Casuarinaceae) host trees. Allocasuarina tree species may have a higher host receptivity for ectomycorrhizal hypogeous fungi species that provide an important food resource for Australian mycophagous animals.  相似文献

Laccaria and Hydnangium typically undergo a postmeiotic mitotic nuclear division in basidiospores, basidia, or less commonly, in sterigmata resulting in eight nuclei. Basidiospores formed on tetrasterigmate basidia are binucleate, and those on bisterigmate basidia are tetranucleate in both genera. While Hydnangium and Laccaria likely comprise a monophyletic group—a hypothesis based on a number of shared characters—the presence of multinucleate basidiospores in several putative outgroups precludes using this shared character state as a justification for recognizing these two genera in a separate family, Hydnangiaceae, as had been proposed. Based on a review of the literature, the presence and location of postmeiotic mitotic divisions in basidia and basidiospores of Agaricales do not appear to be appropriate for assessing phylogenetic relationships among genera or families because of a high level of homoplasy.  相似文献

Millions of tons of fungal spores are dispersed in the atmosphere every year. These living cells, along with plant spores and pollen grains, may act as nuclei for condensation of water in clouds. Basidiospores released by mushrooms form a significant proportion of these aerosols, particularly above tropical forests. Mushroom spores are discharged from gills by the rapid displacement of a droplet of fluid on the cell surface. This droplet is formed by the condensation of water on the spore surface stimulated by the secretion of mannitol and other hygroscopic sugars. This fluid is carried with the spore during discharge, but evaporates once the spore is airborne. Using environmental electron microscopy, we have demonstrated that droplets reform on spores in humid air. The kinetics of this process suggest that basidiospores are especially effective as nuclei for the formation of large water drops in clouds. Through this mechanism, mushroom spores may promote rainfall in ecosystems that support large populations of ectomycorrhizal and saprotrophic basidiomycetes. Our research heightens interest in the global significance of the fungi and raises additional concerns about the sustainability of forests that depend on heavy precipitation.  相似文献

Seasonal and diurnal changes in concentrations of airborne basidiomycete spores (basidiospores, rusts, smuts) were studied, using Burkard volumetric spore traps, in two areas of Mexico City with different degrees of urbanization and related to changes in climatic variables through 1991. Basidiomycete spores formed a large component of the total airborne fungal spore load in the atmosphere of Mexico City. They were the second most abundant spore type after Deuteromycotina (Hyphomycetes), forming 32% of the total fungal spores trapped in an urban-residential area and 28% in an urban-commercial area. The most abundant basidiomycete spores were basidiospores although smut-type spores were trapped on more days than basidiospores and rusts on fewer. Basidiospores occurred in concentrations up to 2,000 spores m-3 in the urban-residential area. Basidiospores showed a marked seasonal distribution, especially in the southern area, with their greatest abundance during the wet season. The correlation coefficients associated with regressions between basidiospore concentration and some environmental factors were increased when a lag period of 2 to 4 days was used between environmental measurements and the day of spore collection. Basidiospore concentrations exceeded the 75 percentile concentration (>400 spores m-3) most often when rainfall was up to 6 mm and relative humidity was >70%. Basidiospores showed a diurnal periodicity with greatest concentrations in the early morning. The most common basidiospore type was Coprinus which formed 67% of basidiospores trapped in the southern area and 63% in the central area. Smut spores were trapped on 87% of days through the year while rust spores occurred in only 35%. Both rusts and smuts were present in only small concentrations.  相似文献

The aim of the present work was to study the possibility of using the fatty acid content in the basidiospores as a taxonomic tool. Basidiospores of Armillaria borealis, Amanita muscaria, Agaricus sylvicola, Hypholoma capnoides, Cortinarius nemorensis and Russula delica were used. The content of fatty acids as well as other substances may vary to a certain degree depending on the part (pileus, stipe, lamella) or stage of development of the actual basidiocarp analysed. Moreover, substances from fungivorous invertebrates, parasitic fungi or bacteria may be found in the chemical analyses of the basidiocarps. Chemotaxonomic conclusions may, therefore, be burdened with serious uncertainties. On the other hand, the ripe basidiospores are terminated structures and belong to the most homogenous structures encountered from a basidiocarp. Their shape, size, colour and ornamentation are considerably homogenous within an actual species. Therefore, the basidiospores are often used as a reliable differentiating characteristic separating species as well as taxa of higher categories. From a practical point of view, ripe spores are easy to obtain in relatively large quantities with simple techniques, and they are not so prone to decay as the carpophore tissue. In the present study, gas chromatography (GC) and gas chromatography-mass spectrometry (GC-MS), after methanolysis of the fungus spores, were used to map essential fatty acids in basidiomycetes. Gas chromatography and gas chromatography-mass spectrometry revealed the presence of fatty acids of C12:0-C24:0 size in the basidiospores of these higher basidiomycetes. The major fatty acid in H. capnoides is C18:2, and the major fatty acid in the other species is C18:1. The basidiospores proved to be a good source of fatty acids for chemotaxonomic investigations of agarics.  相似文献

Resupinate thelephoroid fungi (hereafter called tomentelloid fungi) have a world-wide distribution and comprise approximately 70 basidiomycete species with inconspicuous, resupinate sporocarps. It is only recently that their ability to form ectomycorrhizas (EM) has been realized, so their distribution, abundance and significance as mycobionts in forest ecosystems is still largely unexplored. In order to provide baseline data for future ecological studies of tomentelloid fungi, we explored their presence and abundance in nine Swedish boreal forests in which the EM communities had been analysed. Phylogenetic analyses were used to compare the internal transcribed spacer of nuclear ribosomal DNA (ITS rDNA) sequence data obtained from mycobionts on single ectomycorrhizal tips with that obtained from sporocarps of identified tomentelloid fungi. Five species of Tomentella and one species of Pseudotomentella were identified as ectomycorrhizal fungi. The symbiotic nature of Tomentella bryophila, T. stuposa, T. badia and T. atramentaria is demonstrated for the first time. T. stuposa and Pseudotomentella tristis were the most commonly encountered tomentelloid fungi, with the other species, including T. sublilacina, only being recorded from single stands. Overall, tomentelloid fungi were found in five of the studies, colonizing between 1 and 8% of the mycorrhizal root tips. Two of the five sites supported several tomentelloid species. Tomentelloid fungi appear to be relatively common ectomycorrhizal symbionts with a wide distribution in Swedish coniferous forests. The results are in accordance with accumulating data that fungal species which lack conspicuous sporocarps may be of considerable importance in EM communities.  相似文献

Microsatellite markers were established by an improved combined simple sequence repeat (CSSR) approach for the ectomycorrhizal basidiomycete Laccaria amethystina. Six markers delivered codominant polymorphic results, with up to six different alleles. They were tested with DNA originating from sporocarps and ectomycorrhizal root tips from silver fir (Abies alba), collected in the northern Black Forest. Sporocarps and ectoymcorrhizae exhibited similar allelic profiles, without any distorting influence of the host tree species. Results were compared to allelic profiles delivered by three published Asian markers. Allelic frequencies of the new markers showed a higher resolution of individuals within the population community, but partially a lower grade of heterozygosity. Possible reasons are discussed, and further questions addressed.  相似文献

The mating systems of most ectomycorrhizal fungi have not been elucidated because of two reasons. One is the difficulty of obtaining homokaryotic isolates for mating tests caused by the low germination rate of basidiospores, and another is the difficulty of checking dikaryotization caused by the absence or inconsistent production of clamp connections on heterokaryotic mycelia under laboratory conditions. Basidiospore germination of a few ectomycorrhizal fungi has been induced by living roots of their host plants. Based on this information, we examined methods to obtain homokaryotic isolates of Rhizopogon rubescens using its host plant, Pinus thunbergii. The basidiospores of R. rubescens appeared to germinate well on an agar plate, on which axenic pine seedlings were grown in advance to induce germination, even when the seedlings were removed from the plate at the time of spore inoculation. To enhance the production rate of clamp connections on the heterokaryotic mycelia of R. rubescens, the culture medium composition was modified. The pH of the medium was critical for the production of clamp connections, and the optimal pH was higher for the production of clamp connections than for mycelial growth. These findings made it possible to conduct mating tests, and we found that the mating system of R. rubescens is bipolar with a multiallelic mating type factor.  相似文献

Glacier forefronts provide a unique system for studying primary succession of plants and fungi. We constructed a chronosequence of ectomycorrhizal fungus occurrence on the forefront of Lyman Glacier in the North Cascades mountain range in Washington, USA. The plant communities established on non-vegetated substrate as patchily distributed plant individuals and developed towards complex vegetation with a variety of ectomycorrhizal hosts, including Salix commutata, S. phylicifolia, Abies lasiocarpa, Larix lyallii, Pinus contorta, Tsuga mertensiana and additional infrequent taxa. A most probable number assay of non-vegetated substrates over the chronosequence indicated that ectomycorrhizal propagules were few or absent in the non-vegetated areas adjacent to the glacier terminus but increased with time since deglaciation. Ectomycorrhizal fungus sporocarps occurred as soon as the first host plants of substantial size were present. However, none were observed with the most recently established hosts--small A. lasiocarpa seedlings. Only four species (Cortinarius decipiens, C. tenebricus, Inocybe lacera, and Laccaria cf. montana) occurred on substrate deglaciated for less than 40 years. Three of these species (C. tenebricus, I. lacera, and L. cf. montana) occurred along the chronosequence to the terminal moraine on substrate deglaciated for 70-100 years. An additional five species (one unidentified species each of Cortinarius and Lactarius, Cortinarius mutabilis, Lactarius uvidus var. montanus, and Suillus cavipes) occurred only on the oldest substrate. Our results support the current "early- and late-stage" model of ectomycorrhizal fungus succession in that additional species enter the community over time. However, we hypothesize that diversification of the mycorrhizal fungus community in this primary successional habitat resulted from an increasing diversity of host plants along with changing habitat attributes.  相似文献

Spatial patterns of ectomycorrhizal fungi, ectomycorrhizal plants, and non-ectomycorrhizal plants were investigated in a natural subtropical forest using second-order analysis. The results of spatial pattern analysis showed that the degree of clumping of woody plants and ectomycorrhizal sporocarps were correlated. There was a significantly positive correlation of relative aggregation indices between ectomycorrhizal fungi and both non-ectomycorrhizal trees and ectomycorrhizal saplings. Correlations between percentage of ectomycorrhizal trees and sporocarp occurrence of ectomycorrhizal fungi and between diversities of woody plants and ectomycorrhizal fungi were distance-dependent or scale-related. A significantly high percentage of ectomycorrhizal trees was found only at relatively short distance from ectomycorrhizal fungal sporocarps, and significantly positive correlation of the diversity between woody plants and ectomycorrhizal fungi was found only at relative long distance, which implied that ectomycorrhizal sporocarps prefer ectomycorrhizal-tree-dominant micro-sites at near distances and at relatively large scales, diverse ectomycorrhizal sporocarps could be found in woodlands with high diversity of woody plants. Important factors affecting the spatial distribution, occurrence, and diversity of ectomycorrhizal fungi include spatial pattern of ectomycorrhizal plants and non-ectomycorrhizal plants, percentage of ectomycorrhizal plants, and plant diversity in a natural forest.  相似文献

One hundred and thirty-eight scat (faecal) samples from 17 mammal species native to forests of northeastern Queensland were examined for the presence of spores of both ectomycorrhizal and arbuscular mycorrhizal fungi. Spores of mycorrhizal fungi were found in 57 percent of scat samples representing 12 animal species (Aepyprymnus rufescens, Antechinus godmani, Bettongia tropica, Hypsiprymnodon moschatus, Isoodon macrourus, Melomys ceruinipes, Perameles nasuta, Rattus fuscipes, R. tunneyi, Thylogale stigmatica, Trichourur uulperula, Uromys caudimaculatus). Spores were absent in scats of Antechinus stuartii, Dasyurus hallucatus, Dendrolagus lumholtzi, Petaurus australis and Mesembriomys gouldii. Spores of ectomycorrhizal fungi occurred in 38 percent of scats, and all but one of these samples were from Eucalyptus-dominated sclerophyll forests. Based on the frequency and abundance of spores in scats, five mammals were considered active consumers of hypogeous mycorrhizal sporocarps in sclerophyll forests (A. rufescens, B. tropica, I. macrourus, P. nasuta, and U. caudimaculatus). Individual scats of these animals generally contained a range of distinctive spore types. Spores of arbuscular mycorrhizal fungi were found in low abundance in almost 40 percent of scat samples collected, from both sclerophyll forest and rainforest habitats. We suggest that the majoriry of these spores were acquired incidentally through ingestion of soil during foraging activities on the forest floor. Glasshouse inoculation experiments in which seedlings of Eucalyptus grandis and Sorghum bicolor were inoculated with scat material from several species of mammal demonstrated that the spores of ectomycorrhizal and arbuscular mycorrhizal fungi retained some viability and colonized the roots of host-plant seedlings. Insufficient information is known of the ecology of mycorrhizal fungi in Australia's tropical forests to speculate as to the implications of these findings for forest conservation and rehabilitation.  相似文献

Sporocarps of ectomycorrhizal fungi are strong carbon sinks for the source in host trees, but the details of carbon transfer from the host to the sporocarp are unknown. In this study, single seedlings of Japanese red pine (Pinus densiflora) colonised by Laccaria amethystina were grown on floral foam plates fitted in rhizoboxes, resulting in fruiting on the substrate. The seedlings were photosynthetically labelled with 14CO2; 14C-labelled photosynthate transfer from leaves to sporocarps was then chased using a time-course autoradiography technique. 14C was transferred to healthy, fresh sporocarps in a purple colour ranging from primordial to elongate sporocarps, but hardly to senesced ones that had faded to white or grey, or browned. This suggested that C is transferred only to physiologically active sporocarps. Two seedlings associated with a growing sporocarp were labelled again 7 and 16 days after the first labelling, respectively. 14C accumulation in the sporocarps rose in a stepwise manner after the second labelling, indicating that sporocarps mainly used recently rather than previously photosynthesised C.  相似文献
