Male (anther culture) and female (Hordeum bulbosum) derived, doubled haploid populations were used to map the barley genome and thus determine the different recombination rates occurring during meiosis in the F1 hybrid donor plants. The anther culture-derived (male recombination) population showed an 18% overall increase in recombination rate. This increased recombination rate was observed for every chromosome and most of the chromosome arms. Examination of linkage distances between individual markers revealed eight segments with significantly higher recombination in the anther culture-derived population, and one in the Hordeum bulbosum-derived population. Very strong distortions of single locus segregations were observed in the anther culture-derived population, but map distances were not affected significantly by these distortions. There were 1.047 and 0.912 recombinations per chromosome in the anther culture and Hordeum bulbosum-derived doubled haploid populations, respectively.  相似文献

Summary Microbodies, usually spherical and about 0.2 m in diameter, were found to be associated with prophase nuclei in vegetative cells and meiocytes of the red algaPalmaria palmata. Nucleus-associated microbodies in meiocytes were numerous, but they did not react to the DAB cytochemical test for catalase and peroxidase activity. Microbodies not associated with nuclei in the same cells were intensely DAB-positive. Neither aminotriazole nor potassium cyanide inhibited the DAB reaction.  相似文献

Chromosome arrangement in interphase has been inferred from an analysis of the relative positions of the chromosomes and the chromosome arms in untreated haploid pollen grain metaphases ofUvularia grandiflora. The distances between centromeres forming the smallest possible circle were measured in 43 metaphases. The relative positions of the chromosomes did not differ significantly from randomness. Neither did similar-sized chromosome arms show any tendency to be next to each other. The results thus disagree both with the hypothesis ofComings (1968) that each chromosome occupies a definite position in the interphase nucleus and with the claim ofBennett (1982) that similar-sized chromosome arms lie next to each other.  相似文献

Two emulsions and two water-based-formulations of freeze dried submerged spores of Metarhiziumanisopliae var. acridum (Metch.) Sorokin(isolate IMI 330189) were compared with aerialconidia (GREEN MUSCLETM) for their efficacyagainst Hieroglyphus daganensis (Krauss)and Locusta migratoria (R. & F.). Thefield experiments were conducted in East Nigeron H. daganensis whereas the laboratoryinvestigations were carried out in Germanyusing L. migratoria. In the fields, allformulations were applied on one hectare plotsusing ULV application techniques. Direct andresidual spray effects were assessed. In allcases there were highly significant (p < 0.001) differences between formulations asregards to total mortalities and mediansurvival times (MST). In both direct and sprayresidue effect assessments, aerial conidiaformulated in diesel oil showed over 95%mortality with significantly shorter MST (3 to8 days) under field conditions. These werefollowed by emulsions of submerged spores,which resulted in a mortality ranging from 56to 92% (MST = 8 to 16 days) for the directspray and 90 to 97% (MST = 7 to 12 days) forspray residue effect assessments. Experimentsin the laboratory positively confirmed theseresults. There were no apparent differencesbetween water-based formulations and thecontrol with respect to mortality and MST.These results emphasize the importance ofemploying oil carriers to protect spores fromenvironmental stress and thus enhance efficacy.The study also demonstrated the importance ofsecondary spore pick up from the sprayresidues. Spores in all formulations persistedover five days and caused mortalities rangingfrom 62 to 100% on healthy grasshoppersexposed to the spray residue from treatedvegetation. The results of this researchsuggest that emulsions may be an effectiveoption to improve efficacy of submerged sporesfor ultra low volume application under Sahelianconditions.  相似文献

A mutation of the ctsA locus of Aspergillus nidulans affects both the radial growth and conidiation of the mould when grown in the presence of citrate. The ctsA locus was allocated to linkage group IV but it recombines freely with inoB2 and pyroA4 (which are also in linkage group IV). It is recessive in heterozygous diploids. A possible role for this gene in maintaining membrane integrity is discussed.The authors are with the Departamento de Genética, FMRP-USP, Av. The genusCirsium comprises both gynodioecious and dioecious species. The observation of microsporogenesis in female plants ofC. montanum, C. oleraceum, C. palustre andC. spinosissimum shows that the male sterility is due to a degeneration of the tapetum. This degeneration occurs more or less early according to the species and, in the light of these results, a scheme of evolution in the male sterility mechanism is proposed. Furthermore, the male sterility mechanism inC. montanum is very similar to that previously found in female plants of the dioecious speciesC. arvense. This fact enhances the possibility of evolution of the dioecy ofC. arvense from the gynodioecy found in other species. According to these results, a general scheme of evolution of sexes in the genusCirsium is proposed.  相似文献

Sequence comparisons and a revised classification of the Euglenophyceae were based on 92 new SSU rDNA sequences obtained from strains of Euglena, Astasia, Phacus, Trachelomonas, Colacium, Cryptoglena, Lepocinclis, Eutreptia, Eutreptiella and Tetreutreptia. Sequence data also provided molecular signatures for taxa from genus to class level in the SSU rRNA secondary structure, revealed by a novel approach (search for non-homoplasious synapomorphies) and used for taxonomic diagnoses. Photosynthetic euglenoids and secondary heterotrophs formed a clade, designated as Euglenophyceae (emend.) with two orders: Euglenales and Eutreptiales. The mostly marine Eutreptiales (Eutreptia, Eutreptiella; not Distigma) comprised taxa with two or four emergent flagella (the quadriflagellate Tetreutreptia was integrated within Eutreptiella). The Euglenales (freshwater genera with < or = one emergent flagellum) formed nine clades and two individual branches (single strains); however, only two clades were congruent with traditional genera: Trachelomonas (incl. Strombomonas) and Colacium. Euglena was polyphyletic and diverged into four independent clades (intermixed with Astasia, Khawkinea and Lepocinclis) and two individual branches (e.g. E. polymorpha). Phacus was also subdivided into Phacus s. str. and two combined lineages (mixed with Lepocinclis spp. or Cryptoglena). In consequence, Euglena (s. str.), Phacus and other genera were emended and one lineage (mixed Phacus/Lepocinclis-clade) was recognized as the previously neglected genus Monomorphina Mereschkowsky (1877). The sister clade of Phacus s. str. (mixed Euglena/Lepocinclis-clade) was identified as Lepocinclis Perty (emended).  相似文献

Dietary mixing by nymphs of Schistocerca americana was studied in the laboratory using artificial diet cakes. Individuals were given either two different inadequate but complementary diet cakes or two adequate and identical ones. When unique flavors (coumarin or NHT) were added to the diet cakes, insects given the inadequate diet treatment switched between cakes more than insects exposed to the adequate diets. The corpora allata (CA) of various insects express enzymes with fixation resistant NADPHdiaphorase activity. In female grasshoppers, juvenile hormone (JH) released from the CA is necessary to establish reproductive readiness, including sound production. Previous studies demonstrated that female sound production is also promoted by systemic inhibition of nitric oxide (NO) formation. In addition, allatotropin and allatostatin expressing central brain neurons were located in close vicinity of NO generating cells. It was therefore speculated that NO signaling may contribute to the control of juvenile hormone release from the CA.This study demonstrates the presence of NO/cGMP signaling in the CA of female Chorthippus biguttulus. CA parenchymal cells exhibit NADPHdiaphorase activity, express anti NOS immunoreactivity and accumulate citrulline, which is generated as a byproduct of NO generation. Varicose terminals from brain neurons in the dorsal pars intercerebralis and pars lateralis that accumulate cGMP upon stimulation with NO donors serve as intrinsic targets of NO in the CA. Both accumulation of citrulline and cyclic GMP were inhibited by the NOS inhibitor aminoguanidine, suggesting that NO in CA is produced by NOS. These results suggest that NO is a retrograde transmitter that provides feedback to projection neurons controlling JH production. Combined immunostainings and backfill experiments detected CA cells with processes extending into the CC and the protocerebrum that expressed immunoreactivity against the pan-neural marker anti-HRP. Allatostatin and allatotropin immunopositive brain neurons do not express NOS but subpopulations accumulate cGMP upon NO-formation. Direct innervation of CA by these peptidergic neurons was not observed.  相似文献

We report an additional case of long-term persistence of Paranosema locustae in grasshoppers of Argentina. The pathogen was introduced from North America on rangeland at Loncopué, Neuquén province. Microsporidia were not detected in pre-introduction samples whereas infected grasshoppers were found 11 years after introduction. Affected grasshoppers were the melanoplines Dichroplus elongatus, Dichroplus maculipennis, and Scotussa lemniscata, some of them with high spore loads. The case highlights the ability of P. locustae to recycle in local grasshopper communities by parasitizing susceptible species other than the natural hosts.  相似文献

Summary Cellular morphology, contact, and arrangement in the late blastula and in various stages of gastrulation ofXenopus were examined by SEM of specimens dissected after fixation or fractured in amyl acetate. The prospective ectoderm of the blastocoel roof consists of several layers of interdigitating cells connected by numerous small protrusions which may function in the decrease in number of cell layers observed during ectodermal epiboly. During gastrulation, prospective mesoderm is regionally differentiated by cellular morphology and arrangement into preinvolution mesoderm, the mesodermal involution zone, and involuted mesoderm. The involuted anterodorsal (head), lateral, and ventral mesoderm consists of a stream of loosely-packed, irregularly shaped cells having large extensions of the cell body attached locally to other cells by small protrusions. Involuted posterodorsal mesoderm (chordamesoderm) consists of elongated cells arranged in palisade fashion and connected by similar protrusions. Involuted mesodermal cells in all regions are attached to the overlying prospective ectodermal cells by numerous small protrusions along the entire interface between the two cell layers. Suprablastoporal endodermal cells involute as an epithelial sheet, changing in shape in the process, to form the roof of the archenteron. Bottle cell morphology, arrangement, and position with respect to the mesodermal cell stream is described. Evidence presented here and elsewhere suggests that involution of mesoderm and of the archenteron roof inXenopus is dependent primarily upon the relative movement of the mesodermal cell stream and of the overlying ectoderm.  相似文献

Summary The analyses of haploid male DNA pooled (MDP) samples can increase the cost efficiency in population genetic studies for a variety of applications in fundamental and applied screening of social Hymenoptera populations. Depending on the research problem addressed it can reduce the number of genotyped samples per study by orders of magnitude. In this study we show the feasibility of MDPs for assessing population allele frequencies, mother queen genotypes and mutation rates for microsatellite loci in the honeybee (Apis mellifera). The main purpose of MDP genotyping is not to replace individual based analyses but to discard non-informative samples and monomorphic loci from further analyses.  相似文献

Biosurfactants have been suggested as a method to control harmful algal blooms (HABs), but warrant further and more in-depth investigation. Here we have investigated the algicidal effect of a biosurfactant produced by the bacterium Pseudomonas aeruginosa on five diverse marine and freshwater HAB species that have not been tested previously. These include Alexandrium minutum (Dinophycaee), Karenia brevis (Dinophyceae), Pseudonitzschia sp. (Bacillariophyceae), in marine ecosystems, and Gonyostomum semen (Raphidophyceae) and Microcystis aeruginosa (Cyanophyecae) in freshwater. We examined not only lethal but also sub-lethal effects of the biosurfactant. In addition, the effect of the biosurfactant on Daphnia was tested. Our conclusions were that very low biosurfactant concentrations (5 μg mL−1) decreased both the photosynthesis efficiency and the cell viability and that higher concentrations (50 μg mL−1) had lethal effects in four of the five HAB species tested. The low concentrations employed in this study and the diversity of HAB genera tested suggest that biosurfactants may be used to either control initial algal blooms without causing negative side effect to the ecosystem, or to provoke lethal effects when necessary.  相似文献

A specific condensed lignin substructure, dibenzodioxocin, was immunolocalized in differentiating cell walls of Norway spruce (Picea abies (L.) H. Karsten) and silver birch (Betula pendula Roth) xylem. A fluorescent probe, Alexa 488 was used as a marker on the dibenzodioxocin-specific secondary antibody. For the detection of this lignin substructure, 25-m cross-sections of xylem were viewed with a confocal laser-scanning microscope with fluorescein isothiocyanate fluorescence filters. In mature cells, fluorescence was detected in the S3 layer of the secondary wall in both tree species, but it was more intense in Norway spruce than in silver birch. In silver birch most of the signal was detected in vessel walls and less in fiber cell walls. In very young tracheids of Norway spruce and vessels and fibers of silver birch, where secondary cell wall layers were not yet formed, the presence of the dibenzodioxocin structure could not be shown.Abbreviation CLSM confocal laser-scanning fluorescence microscopy  相似文献

Alarm pheromones are used in many social insects to elicit an alarm response of nestmates towards disturbances. This chemical alarm channel is especially used by species nesting in closed environments. Polistes gallicus paper wasps typically found their nests on open substrates where visual and vibrational stimuli could be more important than the chemical one to alarm the colony. We investigated, through field bioassays, if, also in this species, workers venom is still the source of alarm pheromones. Our results show that venom volatiles are able to increase the colony defence reaction stimulating more individuals to attack and sting the object of disturbance. Therefore our results suggest the hypothesis that chemical alarm is independent of nesting habit (concealed vs exposed) within the Polistes genus.  相似文献

SNAREs are membrane-associated proteins that play a central role in vesicle targeting and intra-cellular membrane fusion reactions in eukaryotic cells. Here we describe the identification of AtBS14a and AtBS14b, putative SNAREs from Arabidopsis thaliana that share 60% amino acid sequence identity. Both AtBS14a and BS14b are dosage suppressors of the temperature-sensitive growth defect in sft1-1 cells and over-expression of either AtBS14a or AtBS14b can support the growth of sft1Δ cells but not bet1Δ cells. These data together with structure–function and biochemical studies presented herein suggest that AtBS14a and AtBS14b share properties that are consistent with them being members of the Bet1/Sft1 SNARE protein family.  相似文献

This paper studies the seasonal presence and removal of the pathogenous micro-organisms Escherichia coli, total coliforms (TC), Clostridium perfringens (Cp), faecal streptococci (FS), Giardia cysts, Cryptosporidium oocysts and helminth eggs, in a constructed wetland treatment system. The removal efficiency of this system with respect to the indicator micro-organisms achieved maximum values in spring and autumn at 99.9% for E. coli and TC, respectively, in winter at 97.0% for FS, in summer at 100% for Clostridium and throughout the year, also at 100%, in the case of Giardia cysts, Cryptosporidium oocysts and helminth eggs. In general, very low protozoan and helminth egg counts were found, and the system demonstrated efficient reduction of the wastewater indicator pathogens.  相似文献
