Superovulated beef cows and heifers were nonsurgically collected 6 to 8 days post estrus. Commercial production results for 1976 through 1978 were 4979 pregnancies from 7814 embryos transferred for an overall pregnancy rate of 63%. In 1978, 519 superovulation procedures averaged 9.95 +/- 8.4 (S.D.) ova collected, 8.2 +/- 7.55 ova fertilized, 5.96 +/- 5.37 embryos transferred and 3.63 +/- 5.37 pregnancies per procedure. Embryos were transferred to recipient cows in estrus 12 hr before the donor (-12) the same time (0) or 12 hr after the donor (+12). The +12 group had a significantly lower pregnancy rate (61%, P<.05) than the 0 group (67%) or -12 group (66%). Transfer of early morula stage embryos resulted in a lower pregnancy rate (61%, P<.05) than late morula (67%) early blastocyst (67%) or late blastocyst (71%) stage embryos. A higher pregnancy rate (P<.05) was obtained with embryos of good morphological quality (71%) than with embryos graded fair and poor (55%). The pregnancy rate for embryos transferred nonsurgically was lower (44%) than the pregnancy rate for embryos transferred surgically during the same time period (66%). Pregnancy rates for three operators performing the nonsurgical transfers were 48%, 53%, 28%. No difference in pregnancy rate was found between embryos cultured 24 hr in BMOC-3 at 37C (62%) and embryos transferred the same day as collection (60%). Pregnancy rates for cultured embryos transferred to recipient cows in estrus 12, 24 or 36 hr after the donor were 68%, 62% and 60%, respectively. Embryos recovered on days 6, 7 and 8 were frozen in 1.5M DMSO and stored in liquid nitrogen several days to several weeks. Of 68 embryos frozen, 34 were viable post thaw. Upon transfer to recipient cows, the 34 viable embryos produced 23 confirmed pregnancies.  相似文献

We describe the first complete embryo transfer program, including flushing of embryos from the oviducts via the uterine horns, transfer of embryos into the Fallopian tubes or the uterine horns and recording of the number of piglets born live. The described procedure is minimally invasive and allows the use of pigs simultaneously for embryo collection and production of normal pregnancies. A 30 degrees forward oblique endoscope provided optimal visualization of the reproductive organs and free access to the organs for embryo flushing and transfer. In contrast to surgical and nonsurgical methods, endoscopy allows to pre-examine the genital tract for reproductive abnormalities and successful ovulation. A total of 95 prepuberal gilts or cyclic sows were used in this trial. Embryos or oocytes were collected from hormonally treated pigs via endoscopy(n = 17) on Day 3 and via laparotomy or post mortem after slaughter (control group, n = 38) on Day 3 and 6 after insemination. One (unilateral collection, n = 7) or both oviducts (bilateral collection, n = 10) were flushed endoscopically. We recovered 114 (average 16/pig) and 279 (average 28/pig) oocytes or embryos with fertilization rates of 89% and 72%, respectively. In the control group 834 oocytes or embryos were collected at Day 3 and 6 after insemination (fertilization rate 64%, total 534 embryos, 33 at 2-, 367 at 4-, 2 at 8-cell stage, 24 morulae and 108 blastocysts). Of 836 embryos recovered by endoscopy, surgery or slaughter 528 Day 3 embryos at 2- to 4-cell stage were transferred into (one) oviducts (n = 27 pigs, about 20/pig) resulting in 9 pregnant pigs diagnosed at Day 28 by sonography. Of the 9, 8 carried a total of 49 piglets to term. A total of 195 Day 6 embryos were transferred into uterine horns (n = 12 pigs, about 16/pig), resulting in 5 pregnant pigs carrying a total of 38 offspring to term. The use of endoscopy in assisted reproduction of pigs has the advantages of allowing easy access to the ovary, oviduct and uterus, clear view of the organ manipulation without exposure and exteriorization of viscera during surgery.  相似文献

This review introduces newly developed serum-free media (IVD101 and IVMD101), that are effective for producing high yields of transferable embryos of good quality from in vitro-matured and -fertilized oocytes. Both serum-free media produced better results than serum-containing medium, including increased rates of blastocyst formation, post-thaw embryo viability, and pregnancy after transfer. In addition, reduced risks of calf mortality and large calf syndrome were also observed for the serum-free-derived embryos. Serum-derived embryos contained a large number of lipid droplets and immature mitochondria in their cytoplasm that may account for the lower production of transferable embryos and poor embryo quality. A non-invasive technique using scanning electrochemical microscopy was successful in quantitatively measuring oxygen consumption of single embryos. This technique may prove to be reliable for predicting embryo viability and subsequent developmental ability.  相似文献

Although transfer of bovine embryos is much less likely to result in transmission of pathogens than transport of postnatal cattle, the epidemiologic risk associated with bovine embryo transfer merits examination. Much research has validated the efficacy of internationally approved processing protocols to render bovine in vivo-derived embryos free of specified pathogens. The purpose of this review is to summarize current sanitary recommendations for bovine embryo transfer, while emphasizing recent research to develop and validate novel approaches to biosecurity. Continued research will enable the development and validation of novel embryo treatments and culture reagents to minimize requirements for testing of embryo or oocyte donors, and testing of embryo recipients.  相似文献

The objective of this study was to use an enzyme-linked immunosorbent assay to measure progesterone (P(4)) in donor cows as an indicator of reproductive normality and response to superovulatory treatment. Forty-eight Holstein cows were superovulated, and blood and milk samples were collected four times: 1) at initial follicle stimulating hormone treatment, 2) just prior to administration of prostaglandin F(2)-alpha, 3) at estrus and 4) at the time of embryo recovery, 6 to 7 d after estrus. Cows were inseminated twice during estrus. The P(4) profiles (ng/ml) obtained at the four collections, were, respectively, as follows: blood plasma, 2.4, 2.7, 0.2 and 11.2; defatted milk analyzed by radioimmunoassay, 3.7, 3.8, 0.5 and 14.6; preserved whole milk analyzed by ELISA, 6.3, 7.8, 0.7 and 19.1; frozen whole milk analyzed by ELISA, 9.8, 10.0, 0.8 and 21.1. All four assays resulted in the expected pattern of P(4) values for most cows at the four stages sampled. Approximately 10% of the cows had P(4) concentrations outside of the expected range of values. A majority of these cows were aberrant, based upon behavior and palpation of the reproductive organs. The rapidly available P(4) results from the ELISA were useful in making appropriate decisions regarding management of donor cows.  相似文献

Our objective was to evaluate factors that affected the success of embryo transfer programs in large dairy herds. Non-lactating donor cows produced a larger number of ova/embryos (P<0.01) and viable embryos (P<0.01) than lactating cows. The interaction between season and donor class was correlated with the proportion of ova/embryos classified as fertilized (P=0.03), because lactating donors had fewer fertilized ova in the summer. There was no correlation between 305-day mature equivalent milk yield and response to superstimulation. Although the interval between superstimulation protocols was correlated with the number of ova/embryos (P=0.03), there was no correlation with the number of viable embryos. Pregnancy per embryo transfer (P/ET) in heifer recipients was correlated with embryo quality grade (P<0.01), season (P=0.04), and whether embryos were fresh or frozen/thawed (P<0.01). Lactating recipient cows tended to have a lower rate of P/ET during the summer (P=0.12 to P=0.08). Synchronization protocols tended to be (P=0.06; Herd 1) or were (P=0.02; Herd 2) correlated with P/ET. Lactating cows receiving vitrified IVF embryos had a lower (P=0.01) P/ET than those receiving fresh IVF embryos, especially in the summer (P=0.09). Milk yield was not correlated with P/ET. The use of heat abatement systems is critical to improve embryo production and P/ET. Synchronization protocols that optimized synchrony of ovulation may increase fertility of recipient cows and eliminate the need for estrous detection.  相似文献

Embryos were non-surgically recovered from superovulated donors. Two trials of ipsilateral single embryo transfer were performed with the Japanese AI instrument.For the first trial, neither the AI instrument nor the cervical expander were ensheathed. The overall pregnancy rate was 33.3 % (2226). Pregnancy rates obtained from three groups with different media were almost identical.In the second trial, both the AI instrument and the cervical expander were covered with a paper sheath to minimize uterine infection. The overall pregnancy rate after the second trial was 59.1 % (1322).  相似文献

Crossbred gilts and sows (n=116) were used for the collection of 1-cell zygotes for DNA microinjection and transfer. Retrospectively, estrus synchronization and superovulation schemes were evaluated to assess practicality for zygote collection. Four synchronization and superovulation procedures were used: 1) sows were observed for natural estrous behavior; 1000 IU human chorionic gonadotrophin (hCG) was administered at the onset of estrus (NAT); 2) cyclic gilts were synchronized with 17.6 mg altrenogest (ALT)/day for 15 to 19 days followed by superovulation with 1500 IU pregnant mares serum gonadotropin (PMSG) and 500 IU hCG (LALT); 3) gilts between 11 and 16 days of the estrous cycle received 17.6 mg ALT for 5 to 9 days and PMSG and hCG were used to induce superovulation (SALT); and 4) precocious ovulation was induced in prepubertal gilts with PMSG and hCG (PRE). A total of 505 DNA microinjected embryos transferred into 17 recipients produced 7 litters and 50 piglets, of which 8 were transgenic. The NAT sows had less (P < 0.05) ovarian activity than gilts synchronized and superovulated by all the other procedures. Synchronization treatments with PMSG did not differ (P > 0.05) in the number of corpora hemorrhagica or unovulated follicles, but SALT and PRE treaments had higher ovulation rates than LALT (24.7 +/- 2.9, 24.3 +/- 1.8 vs 11.6 +/- 2.7 ovulations; X +/- SEM). The SALT and PRE treatments yielded 12.3 +/- 2.6 and 17.7 +/- 1.7 zygotes. Successful transgenesis was accomplished with SALT and PRE procedures for estrus synchronization and superovulation.  相似文献

During a goat transgenic program that took place in Israel from July 1995 to February 1996, Saanen (n = 343) and Nubian x Damascus (n = 378) crossbred goats of mixed ages were used as donors (n = 433) and recipients (n = 288). The effects of season, age, number of surgical procedures, previous hormonal treatments and ovulation rate on the number of microinjectable embryos collected were studied. Likewise, the effects of these parameters on the pregnancy rate as well as the number of embryos transplanted, endogenous progesterone concentrations and exogenous progesterone supplementation were studied in recipient does. Following superovulation with ovine follicle stimulating hormone, 85% of the does responded with 13.6 +/- 5.7 (mean +/- S D) ovulations/doe. Age, month and number of previous hormonal treatments significantly affected the ovulation rate. The average recovery rate was 70%, and it was affected only by the ovulation rate. Pronuclei were visualized in about 30% of the flushed embryos (including unfertilized ova), and those were microinjected with human serum albumin gene construct. About 68% of the injected embryos underwent at least one division during an overnight incubation, and those embryos were transferred, giving about 2.0 transferred embryos per ovulated donor. Of the recipients, 86% responded following synchronization with 3.1 +/- 1.6 (mean +/- S D) ovulations per doe. Breed and month had a significant effect on the ovulation rate. Two or three microinjected embryos were transferred to each recipient, resulting in more than a 40% pregnancy rate during September to November. Lower pregnancy rates were obtained before and after that period. By monitoring plasma progesterone concentrations in the recipients it was found that progesterone concentration was correlated with the ovulation rate. However, the pregnancy rate was not affected by progesterone concentration. During January and February, 30 to 50% of the recipients failed to develop functional corpora lutea (CL) following embryo transfer, which explained the lower pregnancy rate in those months. Nowadays, the development of new technological solutions in the medical field, in particular biosensors, is a priority and a ground for great scientific and financial investment. From glucose sensors to highly sensible and more precise molecular tools, this biotechnological field has gone through an exponential growth, but still the applications are very limited to the future potential foreseen in the medical area. In the last decade, the advances in the genomic field have permitted the identification of specific biomarkers related to certain diseases, becoming one of the main approaches used in clinical diagnosis. Biomarkers have different clinical values, in the sense that they may provide preventive, predictive, prognostic and therapeutic response related information, not being exclusively used for diagnostic purposes, but also be applied in health management and disease treatment. Therefore, biomarkers allied with biosensors have the potential to revolutionize the way healthcare is managed. The vast choice of bioreceptors such as nucleic acids, antibodies, antigens, enzymes and even whole cells, consents the diagnosis of diseases ranging from viral and bacterial infections to cancer and metabolism disorders. The major appeal of these sensing platforms is that it provides a fast, cost-effective, reliable, highly sensitive and easy way to obtain an earlier clinical diagnosis, which can significantly affect the survival rate or patient's prognosis. A mathematical model was used to evaluate the dynamics and risks in bovine embryo transfer. Variables included embryo collection, fertilization, and transfer rates, plus overall pregnancy rates. Decision analysis was applied to three sets of 500 simulated flushes to test three strategies: 1) nonsuperovulation, 2) superovulation using follicle stimulating hormone (FSH) and 3) superovulation using pregnant mare serum gonadotropin (PMSG). Model validation involved comparing model performance against standards derived from 39 published studies. The model's repeatability was +/- 0.10 against standards in 93% of cases, and never exceeded +/- 0.13 in all 1500 simulations. Model outcomes were accurate to +/- 3% using average results. No pregnancies occurred in 22% of nonsuperovulated donors, 8% of FSH superovulated donors, and 6% of PMSG superovulated donors. PMSG treatment averaged more pregnancies per flush than FSH treatment (4.4 vs 3.9) but showed greater variation in response (64 vs 51%). Decision analysis suggests that a PMSG-induced flush would net $105 more than an FSH-induced flush, and that either superovulation strategy would yield approximately 10 times the net income of a nonsuperovulated flush. Pricing by response (PMSG) or by transfer (FSH) is optimal for the provider of embryo transfer services.  相似文献
