应用解剖学方法,对铁筷子(Helleborus thibetanusFranch.)(毛茛科)营养器官的结构进行了研究。结果表明,铁筷子根的初生结构观察到三原型、四原型和六原型。营养器官中的维管束在横切面上木质部中的导管分子不呈“V”字形排列;根状茎的次生结构由外向内为表皮、皮层和维管柱,髓射线发达。茎的初生结构中多个维管束排列成环状,维管束鞘分化不明显,节部为单隙三迹,叶迹分别来自于3条维管束或同一条维管束。叶为两面叶,表皮细胞不规则;气孔器只分布于下表皮,为毛茛科典型的无规则型气孔。从铁筷子营养器官的解剖学特点来看,与毛茛科其它植物基本相同,但在营养器官中维管束木质部不呈“V”字形、维管束鞘分化不明显、节部具单叶隙等特征上与其它毛茛科植物不同。  相似文献

The vascular system of the grapevine (Vitis vinifera L.) flower is a channel for transporting water and nutrients to the ovary. It plays an important role in the development of the ovary and fertilization through pollination. However, the vascular bundles in the flower are so tiny that they are difficult to sample and observe by traditional slicing techniques. In this study, ‘Summer Black’ grape flowers were selected as the test materials, and the tissue samples were treated by the optical clearing technique. After simple compaction, the structure and development of the vasculature were observed by common microscopy, fluorescence microscopy and laser confocal microscopy. The results showed that the transparency effects of 3% NaOH and a saturated trichloroacetaldehyde composite agreed well with the observations of the vascular structure and the developmental process of the flower in different periods. Moreover, the samples after optical clearing could be reconstructed in 3D, which helped us know more about its development and function. According to these observations, the vasculature of the ‘Summer Black’ flower can be divided into ovule vascular bundles, peripheral vascular bundles and central vascular bundles. The peripheral vascular bundles were composed of the first-order vascular bundles and the inferior vascular bundles which branched from the superior vascular bundles. These bundles branched in different directions with no discernible pattern. The two different branching methods were as follows. First, the inferior vascular bundle was directly connected to a superior vascular bundle. Secondly, some of the superior vascular bundles bent in different ways, forming the inferior vascular bundle connecting the superior vascular bundles by a metamorphosed vessel with a triangular shape. In a comparison of the developmental changes in various periods, the growth of vascular bundles at each period was directly proportional to the growth of the flower. Laser confocal scanning was used to explore the three-dimensional morphology of the peripheral vascular bundle and showed that the peripheral vascular bundle of grapes was not completely parallel to the flower’s epidermal cells. As a result, the optical clearing technique was convenient and authentic compared with the traditional slicing operation for tiny flower organs. With these advantages according to the observations, this study provides a feasible technique and useful information for the study of vascular bundle development in grape flower organs.  相似文献

Lectin-histochemical studies were performed on formalin-fixed, paraffin-embedded tissues from ten mammalian species to demonstrate the pattern of carbohydrate residues in vascular endothelium. Ten different biotinylated lectins were used as probes and avidin-biotin-peroxidase complex (ABC) was used as visualant. Ricinus communis agglutinin-I (RCA-I) and wheat germ agglutinin (WGA) stained vascular endothelium in all species. Peanut agglutinin (PNA) stained vascular endothelium in all species only after preincubation with neuraminidase. Bandeirea simplicifolia agglutinin-I (BS-I) stained vascular endothelium in all species but human, while Ulex europeus agglutinin-I (UEA-I) stained only human endothelium. Individual differences in staining of human vascular endothelium were noted with BS-I and succinylated-WGA (SWGA). Similarly, individual differences in staining of animal vascular endothelium were noted with soybean agglutinin (SBA) after preincubation with neuraminidase. Finally, Concanavalia ensiformis agglutinin (Con A), Dolichos biflorus agglutinin (DBA) and Lens culinaris agglutinin (LCA) did not stain vascular endothelium in any of the species studied.  相似文献

The hyperpermeability of the microvasculature supplying solid tumors is largely attributable to a heterodimeric Mr 34,000-43,000 tumor-secreted protein, vascular permeability factor. Upon reduction, the vascular permeability factor secreted by line 10 tumor cells is resolved by SDS-PAGE into 3 discrete bands of Mr 24,000, 19,500, and 15,000. We demonstrate here that line 10 vascular permeability factor is an N-linked glycoprotein. Nonglycosylated vascular permeability factor migrates on reduced SDS-PAGE as two bands of Mr 20,000 and 15,000. Pulse-chase studies demonstrated that all three chains of native vascular permeability factor were secreted rapidly following synthesis and at equal rates, with a cellular half-retention time of approximately 37 min. When glycosylation was prevented by tunicamycin, individual bands of nonglycosylated vascular permeability factor were also secreted at equivalent rates, but much more slowly (approximately 60 min) than native glycoprotein. Both glycosylated and nonglycosylated forms of vascular permeability factor were equally potent at increasing dermal vessel permeability.  相似文献

In grassland communities vascular plants and bryophytes form two distinct layers. In order to understand the factors responsible for plant community structure, more information about interactions between these plant groups is needed. Often negative correlations between vascular plant and bryophyte covers have been reported, suggesting competition. Here we tested experimentally whether different grassland vascular plant species (Trifolium pratense, Festuca pratensis, Prunella vulgaris) had different influences on the cover of two bryophyte species (Rhytidiadelphus squarrosus, Brachythecium rutabulum). In a two-year garden pot experiment one bryophyte species and one vascular plant species were planted per pot. Bryophytes were planted at a constant density, vascular plants in four densities. The cover of both bryophyte species increased with increasing vascular plant cover, showing the facilitative effect of vascular plants through creating better microclimate, e.g., optimising temperature. Bryophyte responses to vascular plant species were species-specific. Festuca had significantly positive effects on both bryophyte species in the second year, and Trifolium on Brachythecium in both years, whereas Prunella had no significant effect on bryophytes. The facilitative effect of vascular plants was stronger at the second experimental year. In summary, the biotic effects between bryophytes and grassland vascular plants are species-specific and positive interactions are prevailing at low vascular plant densities.  相似文献

Yes-associated protein (YAP), a major effector of the Hippo signalling pathway, is widely implicated in vascular pathophysiology processes. Here, we identify a new role of YAP in the regulation of vascular senescence. The inhibition or deficiency and overexpression of YAP were performed in human umbilical vein endothelial cells (HUVECs) and isolated vascular tissues. Cellular and vascular senescence was assessed by analysis of the senescence-associated β-galactosidase (SA-β-gal) and expression of senescence markers P16, P21, P53, TERT and TRF1. We found that YAP was highly expressed in old vascular tissues, inhibition and knockdown of YAP decreased senescence, while overexpression of YAP increased the senescence in both HUVECs and vascular tissues. In addition, autophagic flux blockage and mTOR pathway activation were observed during YAP-induced HUVECs and vascular senescence, which could be relieved by the inhibition and knockdown of YAP. Moreover, YAP-promoted cellular and vascular senescence could be relieved by mTOR inhibition. Collectively, our findings indicate that YAP may serve as a potential therapeutic target for ageing-associated cardiovascular disease.  相似文献

针对一些纺织型人造血管管壁的临床选择性破损特征,本文报告两种机织型人造血管管壁的均匀特性研究。通过对其整体、纱线、纤维三个层次的纺织结构性能以及相应的生物力学性能的研究,揭示此类人造血管不同的管壁纺织结构对其生物力学性能整体均匀性的影响,并由此从纺织结构的角度预测该类人造血管在临床应用中可能导致提前降解或破损的原因。研究结果提示在新型人造血管的设计中,要避免此类纺织结构的弱节与不均匀性。  相似文献

The effects of prostaglandin E2 (PGE2) and indomethacin on the vascular reactivity to norepinephrine were tested in three different isolated rat vascular beds (mesenteric artery, hind limb and splenic artery) perfused with the Krebs bicarbonate solution. In these vascular beds PGE2 (0.1–64 ng/ml) or indomethacin (0.1–96 μg/ml) in the perfusate did not change the basal pressure. In the mesenteric vascular bed and the hind limb, PGE2 dose-dependently potentiated the vascular response to norepinephrine, whereas PGE2 dose-dependently inhibited the vascular response to noreinephrine in the splenic artery. In these three vascular beds indomethacin in the perfusate dose-dependently attenuated the vascular response to norepinephrine. In the mesenteric artery and the hind limb PGE2 restored the effect of indomethacin, but in the splenic artery PGE2 did not restore the inhibitory effect of indomethacin. These results indicate that the modulating effect of exogenously administrated PGE2 on the vascular action to norepinephrine varies in different vascular beds. It is also suggested that the contribution of endogenous PGE2 synthesized in the vascular wall to the vascular reactivity to norepinephrine is, as well as the effect of exogenous PGE2, different in different vascular beds.  相似文献

The biphasic effects of locally applied static magnetic fields (SMF) on the cutaneous microcirculation within a rabbit ear chamber (REC) were evaluated under conscious conditions. The microcirculatory vasomotion within a REC was measured continuously and analyzed multilaterally by microphotoelectric plethysmography, a real-time image analyzer and an image shearing monitor. SMF intensities at the REC were controlled at 1 mT and the duration of exposure was 10 min. Seventy-eight experimental trials were carried out on 22 healthy adult rabbits weighing 2.6-3.5 kg. Five experimental groups were chosen at random: 1) no pharmacological treatment or SMF exposure, 2) increased vascular tone induced by noradrenaline infusion without SMF exposure, 3) increased vascular tone induced by noradrenaline infusion with SMF exposure, 4) decreased vascular tone induced by acetylcholine infusion without SMF exposure, 5) decreased vascular tone induced by acetylcholine infusion with SMF exposure. The results demonstrated that SMF significantly enhanced vasodilatation, with increased vasomotion under noradrenaline-induced high vascular tone as well as vasoconstriction with reduced vasomotion under acetylcholine-induced low vascular tone. This suggests, therefore, that SMF can modulate vascular tone due to biphasic modification of vasomotion in the cutaneous tissue.  相似文献

This study was designed to examine the effects of a high-fat, high-sucrose (HFHS) diet on vascular and metabolic actions of insulin. Male rats were randomized to receive an HFHS or regular chow diet for 4 wk. In a first series of experiments, the rats had pulsed Doppler flow probes and intravascular catheters implanted to measure blood pressure, heart rate, and regional blood flows. Insulin sensitivity and vascular responses to insulin were assessed during a euglycemic hyperinsulinemic clamp performed in conscious rats. In a second series of experiments, new groups of rats were used to examine skeletal muscle glucose transport activity and to determine in vitro vascular reactivity, endothelial nitric oxide synthase (eNOS) protein expression in muscle and vascular tissues and endothelin content, nitrotyrosine formation, and NAD(P)H oxidase protein expression in vascular tissues. The HFHS-fed rats displayed insulin resistance, hyperinsulinemia, hypertriglyceridemia, hyperlipidemia, elevated blood pressure, and impaired insulin-mediated renal and skeletal muscle vasodilator responses. A reduction in endothelium-dependent vasorelaxation, accompanied by a decreased eNOS protein expression in muscles and blood vessel endothelium, and increased vascular endothelin-1 protein content were also noted in HFHS-fed rats compared with control rats. Furthermore, the HFHS diet induced a reduced insulin-stimulated glucose transport activity in muscles and increased levels of NAD(P)H oxidase protein and nitrotyrosine formation in vascular tissues. These findings support the importance of eNOS protein in linking metabolic and vascular disease and indicate the ability of a Westernized diet to induce endothelial dysfunction and to alter metabolic and vascular homeostasis.  相似文献

The effects of endothelin, ET-1, on pulmonary and systemic hemodynamics were studied in the open chest dog and changes in systemic arterial pressure in dogs under conscious and anesthetized states were compared. Rapid intravenous (IV) bolus injections of ET-1, 100-1,000 nanograms/kg, significantly decreased systemic arterial pressure, and significantly decreased systemic vascular resistance whereas left atrial pressure and pulmonary vascular resistance were not altered. Reductions in systemic arterial pressure in response to bolus injection of ET-1, 100 and 300 nanograms/kg IV, during conscious state and during anesthesia were similar, respectively. The present data suggest that ET-1 dilates the systemic vascular bed independent of the animal's state of consciousness. The present data also suggest that when compared to the systemic vascular bed, the pulmonary vascular bed is less responsive to bolus administration of ET-1.  相似文献

Cytological observations were made on developing seed coat of broad bean (Vicia faba L.) by use of light and electron microscopy. Attention was focused on vascular distribution. The seeds were attached by the funiculus to tile large vascular bundles of pericarp of broad bean. The vascular bundle passed through hilum and two layers of palisade, entered the pa- renchyma of seed coat, then diverged in to two routes. One was a complete vascular bundle composed of both'phloem and xylem elements, it stretched down through seed raphe, then upward and terminated near the radical. The other was a two-recurrent-vascular-bundle with only phloem constitutents, they extended forward detoured the micropyle and extended downward, but did not join with the upward complete vascular bundle. The recurrent vascular bundles branched out many small short branches. The obvious difference between phloem of recurrent vascular bundle and of complete vascular bundle was that the companion ceils of the former did not normally modify to transfer ceils, but connected to the adjoining parenchyma cells through abundant plasmodesmata. It is deduced from the structural analysis that the symplastic route may play an important role, particularly in the region of recurrent vascular bundle, in the course of importing assimilates unloading in seed coat and transporting to the embryo.  相似文献

In this study, we attempted to assess the incorporable potential of vascular endothelial cells derived from adult organ blood vessels into tumor blood vessels. Two kinds of adult organ-derived vascular endothelial cells, human aorta endothelial cells (HAEC) and umbilical vein endothelial cells (HUVEC), were administered into murine tumors inoculated to SCID mice. Many human blood vessel networks were visualized in the murine tumors. These cells in solid tumor not only survived and proliferated, but also incorporated into tumor endothelium. These results suggest that adult organ-derived vascular endothelial cells possess the potential to form the neovascular network in various tissues such as vascular endothelial progenitor-like cells in vivo. We propose that these cells can be regarded as a congenic (autologous) vector for vascular regeneration cell therapy and tumor vascular targeting gene therapy.  相似文献

目的:探讨利用CTA原始数据集构建在体女性盆腔静脉血管网数字化三维模型的方法及意义。方法:基于双源CTA技术,获取1例宫颈癌患者的Dicom 3.0原始二维断层图像数据集。利用Mimics 10.01软件分别对骨盆、盆腔动脉血管网以及盆腔静脉血管网进行三维重建并配准融合。结果:构建的盆腔静脉血管网数字化三维模型可以清楚地显示下腔静脉、髂总静脉、髂外静脉、髂内静脉及其初级属支,以及双侧卵巢静脉等。与重建的骨盆、盆腔动脉血管网配准融合后,各支静脉血管的解剖走形及引流区域变得更加清晰明确。结论:基于CTA的计算机三维重建技术是一种研究女性盆腔静脉血管网的好方法,具有较大的运用价值。  相似文献

近年来的研究发现,氧化型低密度脂蛋白(oxi-dizedlowdensitylipoprotein,OX-LDL)是导致动脉粥样硬化发生的重要因素[1].OX-LDL具有双重效应,既有强烈的促细胞生长效应,又可诱导细胞发生凋亡.这主要根据过氧化物量的变化而定,少量的OX-LDL可促进增殖,而长时间大量的OX-LDL作用于平滑肌细胞则可导致其凋亡[2].OX-LDL诱导的平滑肌细胞凋亡有助于氧化脂质的生成,导致动脉粥样硬化形成.在动脉粥样硬化晚期,由于斑块中的平滑肌细胞凋亡,细胞外基质分泌减少,使斑块极不稳定而易于破裂,诱发急性临床事件如心肌梗塞、猝死等的发生[3].OX-…  相似文献

对叉孢苏铁茎的解剖结构进行了仔细的研究,结果表明:叉孢苏铁茎具有发达的皮层,皮层及髓中具有黏液道,有外韧型和周韧型的维管束分布在皮层中,但主要以外韧型为主,在同一切面上能看到横、纵、斜3种不同走向的维管束,所有皮层维官束韧皮部组成成份比较单一。维管组织区由多个生长环组成,每个生长环都具有木质部、维管形成层和韧皮部组成,各生长环韧皮部所占比例均大于木质部,在具4生长环的下部茎的维管组织中,环与环之间间隔2~10层不等的薄壁细胞。从纵向变化上看,从上至下,维管组织区的厚度及生长环数逐渐递增,在同一组织中,含晶及含单宁物质的异细胞呈从无到有,从少到多的分布趋势。  相似文献

作为热带林中一个重要的特征性组分, 附生维管植物对于维持热带森林的物种多样性及其生态系统功能均具有重要作用。该文首次系统地报道了热带天然针叶林中的附生维管植物多样性和分布特征。以海南岛霸王岭国家级自然保护区保存完好的热带天然针叶林(我国唯一较大面积分布的南亚松(Pinus latteri)天然林)中的附生维管植物为研究对象, 通过样带调查(共设置12个10 m × 50 m的样带, 记录每个样带内胸径(DBH) ≥ 5 cm树木上附生维管植物的物种名称、株数及附生高度), 分析附生维管植物的物种多样性和空间分布特征。结果表明: 1)热带针叶林0.6 hm 2面积内共有附生维管植物769株, 分属于7科17属27种, 附生兰科植物和萝摩科植物为优势类群; 2)附生维管植物在水平方向上呈现出聚集分布; 3)附生维管植物在垂直方向上, 在中等高度层次(10-20 m)分布最多, 在下层(0-5 m)也有较多的分布; 4)少数附生维管植物对南亚松表现出一定的选择性, 如华南马尾杉(Phlegmariurus fordii)、玫瑰毛兰(Eria rosea)、眼树莲(Dischidia chinensis)和铁草鞋(Hoya pottsii)等; 5)附生维管植物的物种丰富度及多度与宿主胸径均存在显著的正相关关系。  相似文献

Eriophorum spp. are abundant perennial graminoids in the Arctic tundra and boreal peatlands. Because ecological studies indicated that some plants are unusually productive on infertile and cold sites, the anatomy of the overwintering corms of Eriophorum vaginatum (L.) and Eriophorum scheuchzeri (Hoppe) were examined to determine their involvement in nutrient uptake and storage. Components of the long-distance transport pathways were identified within the plants by using histochemical techniques and transport of apoplastic and symplastic dyes. E. scheuchzeri produced a rhizome that consisted mainly of storage parenchyma cells within which collateral vascular bundles were centrally located and arranged in a circle. By contrast, E. vaginatum developed a ring of horizontally arranged xylem and phloem, in addition to axial amphivasal vascular bundles leading to the leaves, all of which were bordered by transfer cells. As shown by the transport of fluorescein in the phloem and Safranine O in the xylem, each axial bundle and adventitious root contacted the horizontal ring of vascular tissues so that solutes from one vascular bundle were translocated into the vascular ring and circulated to another vascular bundle and/or to the roots. In addition, special groups of sclereids that functioned in both phloem and xylem transport were found at the base of the leaf traces and within junctions of senescing roots. These sclereids were named 'vascular sclerenchyma' and it was hypothesized that they provide a moving end for the vascular system because the corm dies progressively from the distal end as it grows upward from the apical meristem. It was concluded that this unusual vascular system of E. vaginatum is efficient in recycling nutrients internally, which may account for its competitive advantage in infertile and cold sites.  相似文献

细胞衰老是指细胞在各种应激条件下出现周期阻滞,不可逆地丧失增殖能力,其形态、基因表达和功能都发生特定变化的过程。研究表明,血管内皮细胞衰老可以通过削弱血管功能,促进衰老相关血管疾病的发生发展。然而,有关内皮细胞衰老的发生机制以及内皮细胞衰老影响血管功能及衰老相关血管疾病的潜在机制尚待挖掘。本文从血管内皮细胞衰老相关的信号通路,以及血管内皮细胞衰老与血管功能和血管相关疾病(动脉粥样硬化、高血压和糖尿病血管并发症)的最新研究进展进行综述,为进一步认识血管疾病的发病机制,延缓血管衰老提供新的思路。  相似文献
