Isolated internodes of Chara corallina and Nitella flexilis have been used to determine the concentration of one passively permeating solute in the presence of non-permeating solutes. The technique was based on the fact that the shape of the peaks of the biphasic responses of cell turgor (as measured in a conventional way using the cell pressure probe) depended on the concentration and composition of the solution and on the permeability and reflection coefficients of the solutes. Peak sizes were proportional to the concentration of the permeating solute applied to the cell. Thus, using the selective properties of the cell membrane as the sensing element and changes of turgor pressure as the physical signal, plant cells have been used as a new type of biosensor based on osmotic principles. Upon applying osmotic solutions, the responses of cell turgor (P) exactly followed the P(t) curves predicted from the theory based on the linear force/flow relations of irreversible thermodynamics. The complete agreement between theory and experiment was demonstrated by comparing measured curves with those obtained by either numerically solving the differential equations for volume (water) and solute flow or by using an explicit solution of the equations. The explicit solution neglected the solvent drag which was shown to be negligible to a very good approximation. Different kinds of local beers (regular and de-alcoholized) were used as test solutions to apply the system for measuring concentrations of ethanol. The results showed a very good agreement between alcohol concentrations measured by the sensor technique and those obtained from conventional techniques (enzymatic determination using alcohol dehydrogenase or from measurement of the density and refraction index of beer). However, with beer as the test solution, the characean internodes did show irreversible changes of the transport properties of the membranes leading to a shift in the responses when cells were treated for longer than 1 h with diluted beer. The accuracy and sensitivity of the osmotic biosensor technique as well as its possible applications are discussed.  相似文献

Abstract A ``double-water-film electrode technique' has been developed for the long-term characterization of the electrical properties across the interface between the nodal (N) and internodal (A or B) cells and the vacuole along the length of an internode of Chara as a function of time and temperature. The electrode unit consisted of a pair of the water-film electrodes described elsewhere (Chilcott 1988; Chilcott and others 1983; Coster and others 1984; Lucas 1985; and Ogata 1983). The distance between two water-film probes was fixed at 1.0 cm. By scanning the electrode unit, the spatial variations in electrical resistance and capacitance along the longitudinal axis of Chara were observed. Analysis was performed by applying an electrical equivalent circuit for the biomembrane (Philippson 1921). Across the internode (−A or −B)/central nodal cells interface, the specific parallel resistance (Rm) and the parallel capacitance (Cm) at 20°C were 30 ± 5 × 10−3Ωm2 and 1.5 ± 0.5 × 10−1Fm−2 (at 30 Hz), respectively. And the series resistance, corresponding to the vacuole of the internode was 8 × 10−3Ωm2. Study of temperature dependencies of Rm and Cm suggested that a dynamic homeostatic regulation was operating at the interface where numerous plasmodesmata were observed with an electron microscope (Pickett-Heaps 1967; Spanswick and Costerton 1967). Assuming that the individual cylinder of plasmodesma was filled only with cytoplasm, the number of plasmodesma per interface was estimated at 2.6 × 105. In Pisum sativum L. a third, more severe, allele at the internodelength locus le is identified and named led. Plants homozygousfor led possess shorter internodes and appear relatively lessresponsive to GA20 than comparable le (dwarf) plants. Gene ledmay act by reducing the 3ß-hydroxylation of GA20 tothe highly active GA1 more effectively than does gene le. Theresults indicate that le is a leaky mutant and therefore thatendogenous GA1 influences internode elongation in dwarf (le)plants. Fall dormancy (FD) in alfalfa (Medicago sativa L.) can be described using 11 FD ratings, is widely used as an important indicator of stress resistance, productive performance and spring growth. However, the contrasting growth strategies in internode length and internode number in alfalfa cultivars with different FD rating are poorly understood. Here, a growth chamber study was conducted to investigate the effect of FD on plant height, aboveground biomass, internode length, and internode number in alfalfa individuals in the early growth stages. In order to simulate the alfalfa growth environment in the early stage, 11 alfalfa cultivars with FD ratings from one to 11 were chosen and seeded at the greenhouse, and then were transplanted into an artificial growth chamber. The experimental design was a randomized complete block in a split-plot arrangement with three replicates. Plant height, above-ground biomass, internode length, and internode number were measured in early growth stage in all individuals. Our findings showed that plant height and the aboveground biomass of alfalfa did not significantly differ among 11 different FD rated cultivars. Also, internode length and internode number positively affected plant height and the aboveground biomass of alfalfa individuals and the average internode length significantly increased with increasing FD rating. However, internode number tended to sharply decline when the FD rating increased. Moreover, there were no correlations, slightly negative correlations, and strongly negative correlations between internode length and internode number in alfalfa individuals among the three scales, including within-FD ratings, within-FD categories and inter-FD ratings, respectively. Therefore, our results highlighted that contrasting growth strategies in stem elongation were adopted by alfalfa with different FD ratings in the early growth stage. Alfalfa cultivars with a high FD rating have longer internodes, whereas more dormant alfalfa cultivars have a larger number of internodes. There were tradeoffs between internode length and internode number in response to FD in alfalfa, which reflected certain scale-dependence.  相似文献

In long-term experiments comparing the results of single and repeated treatment of dwarf peas with gibberellic acid, both once-treated and weekly-treated plants formed more internodes than control plants. The initial high rate of internode formation in the once-treated peas dropped rapidly reaching that of control plants three weeks after treatment. Weekly-treated plants maintained a higher rate of internode formation. The ratio of internode lengths of once-treated plants to control plants was high (over 3) for the internodes which were elongating during or shortly after the single treatment but dropped rapidly and approached unity in later-developed internodes. The ratio of internode lengths of weekly-treated plants to control plants continued to be high throughout the experiment.  相似文献

以水稻培矮64S作对照,对隐性长穗颈温敏核不育水稻(Oryza sativa L.)长选3S幼穗各发育时期穗颈节间长度及细胞数量和长度进行比较分析。结果表明,长选3S在幼穗分化的二核期至始花期穗颈节间伸长速度最快,其节间伸长的长度是培矮64S的2.1倍;在花粉母细胞减数分裂期至二核期及始花当天至始花后第3天两个时段,穗颈节间伸长速度慢。穗颈节间的细胞个数和平均长度两者呈现相似的变化规律,但从花粉母细胞减数分裂期至始花期,长选3S穗颈节间薄壁细胞增加3759个,细胞平均长度为72.9μm,培矮64S增加3134个,细胞平均长度为38μm,长选3S穗颈节间的伸长主要是由幼穗分化的花粉母细胞减数分裂期至始花期细胞分裂和细胞伸长共同作用的结果,其中后者的作用更显著。  相似文献

Emerging evidence suggests that cytoplasmic streaming can regulate the plasma-membrane H+ transport and photosynthetic electron flow. Microfluorometric and surface pH measurements on Chara corallina internodes revealed the transmission of photoinduced signals by the cytoplasmic flow for a distance of few millimeters from the site of stimulus application. When a 30-s pulse of bright light was locally applied, the downstream cell regions responded with either release or enhancement of non-photochemical quenching of chlorophyll fluorescence, depending on the background irradiance of the analyzed cell area. Under dim background irradiance (<20 μmol m?2 s?1), the arrival of the distant signal from the brightly illuminated 400-μm-wide zone elevated the maximal fluorescence F m in the analyzed downstream area, whereas at higher background irradiances it induced strong quenching of F m . At intermediate irradiances the increase and decrease in F m appeared as two successive waves. The transition between the F m responses of opposite polarities occurred at a narrow threshold range of irradiances. This indicates that inevitable slight variations in irradiance at the bottom chloroplast layer combined with the cyclosis-transmitted signals may contribute to the formation of a photosynthetic activity pattern. The rapid cyclosis-mediated release of non-photochemical quenching, unlike the delayed response of opposite polarity, was associated with opening of H+ (OH?)-conducting plasma membrane channels, as evidenced by the concurrent alkaline pH shift on the cell surface. It is proposed that the initial increase in F m after application of a distant photostimulus is determined, among other factors, by the wave of alkaline cytoplasmic pH.  相似文献

The effects of altered endogenous indole-3-acetic (IAA) levels on elongation in garden pea (Pisum sativum L.) plants were investigated. The auxin transport inhibitors 2,3,5-triiodobenzoic acid (TIBA) and 9-hydroxyfluorene-9-carboxylic acid (HFCA) were applied to elongating internodes of wild-type and mutant lkb plants. The lkb mutant was included because elongating lkb internodes contained 2- to 3-fold less free IAA than those of the wild type. In the wild type, TIBA reduced both the IAA level and internode elongation below the site of application. Both TIBA and HFCA strongly promoted the elongation of lkb internodes and also raised IAA levels above the application site. The synthetic auxin 2,4-dichlorophenoxyacetic acid (2,4-D) also markedly increased internode elongation in lkb plants and virtually restored petioles and tendrils to their wild-type length. In contrast, treatment of wild-type plants with TIBA, HFCA, or 2,4-D caused little or no increase in elongation above the application site. The ethylene synthesis inhibitor aminoethoxyvinylglycine also increased stem elongation in lkb plants, and combined application of HFCA and aminoethoxy-vinylglycine restored lkb internodes to the wild-type length. It is concluded that the level of IAA in wild-type internodes is necessary for normal elongation, and that the reduced stature of lkb plants is at least partially attributable to a reduction in free IAA level in this mutant.  相似文献

The elongation response of the gibberellin (GA) deficient genotypes na, ls, and lh of peas (Pisum sativum L.) to a range of GA-precursors was examined. Plants possessing gene na did not respond to precursors in the GA biosynthetic pathway prior to GA12-aldehyde. In contrast, plants possessing lh and ls responded as well as wild-type plants (dwarfed with AMO-1618) to these compounds. The results suggest that GA biosynthesis is blocked prior to ent-kaurene in the lh and ls mutants and between ent-7α-hydroxykaurenoic acid and GA12-aldehyde in the na mutant. Feeds of ent-[3H]kaurenoic acid and [2H]GA12-aldehyde to a range of genotypes supported the above conclusions. The na line WL1766 was shown by gas chromatography-mass spectrometry (GC-MS) to metabolize [2H]GA12-aldehyde to a number of[2H]C19-GAs including GA1. However, there was no indication in na genotypes for the metabolism of ent-[3H]kaurenoic acid to these GAs. In contrast, the expanding shoot tissue of all Na genotypes examined metabolised ent-[3H]kaurenoic acid to radioactive compounds that co-chromatographed with GA1, GA8, GA20, and GA29. However, insufficient material was present for unequivocal identification of the metabolites. The radioactive profiles from HPLC of extracts of the node treated with ent-[3H]kaurenoic acid were similar for both Na and na plants and contained ent-16α,17-dihydroxykaurenoic acid and ent-6α,7α,16β,17-tetrahydroxykaurenoic acid (both characterized by GC-MS), suggesting that the metabolites arose from side branches of the main GA-biosynthetic pathway. The three goals of cardiac assistance are: (1) To maintain systemic blood flow; (2) To reduce cardiac work and tension development; and (3) To increase oxygen availability to the heart. Toward these ends, various devices and techniques have been developed, including several different types of vascular shunts in combination with or without extracorporeal oxygenation of blood, implantable auxiliary ventricle and augmentation of diastolic pressure by direct counter pulsation of blood through femoral cannulae or intra-aortic balloon.The sequenced counter pulsator is an external cardiac assist device being developed for the therapy of low output syndromes. Investigation in the laboratory has shown that it is capable of increasing cardiac output and diastolic systemic pressure with concomitant reduction in left ventricular end-diastolic pressure. Therefore, it appears to be clinically useful in patients with low cardiac output syndromes.  相似文献

We present a Bayesian method for deriving species-sensitivity distributions (SSDs). We employed four Bayesian statistical models to consider differences in tolerance to toxic substances among different taxonomic groups. We first used a Malkov chain Monte Carlo simulation based on these models to estimate the SSD parameters. We then computed deviance information criterion values of the models and compared them in order to select the model with the best predictive ability. We applied this approach to seven substances (zinc, lead, hexavalent chromium, cadmium, nickel, short-chain chloride paraffin, and chloroform) as case examples, and then compared the derived SSDs from the selected models and a model that assumed no tolerance differences among taxonomic groups. We discuss the advantages and limitations of our approach on the basis of our results.  相似文献

Fournier's gangrene is a necrotizing infection of the scrotum or perineum that requires aggressive surgical debridement. Radical debridement of perineal necrotizing fasciitis can leave extensive tissue defects that are difficult to close and often require multiple surgical interventions. Vacuum-assisted closure (VAC) devices have been shown to assist in a more rapid closure of these wounds, but placement of such devices in the perineum can pose significant challenges. Fick's diffusion equations have been solved for the Haxo and Blinks electrochemical determination of photosynthetic O(2) evolution. The model shows the method to be quantitative in the case of a "long" chamber measuring constant rates of O(2) evolution. Transients are amplitude distorted by an amount depending on the space distribution of the O(2) sources. The elongation of the internodes and petioles of Vicia fabahas been studied in the plumules of normal seedlings, in seedlingsfrom which part or all of the cotyledonary reserve has beenremoved, and in axillary shoots developed on seedlings of variousages. The curves obtained by plotting the lengths of the internodesagainst their plastochrone ages reveal constant differencesin behaviour between plumules and axillary branches. As thegrowth of any shoot progresses a greater contribution to thetotal elongation rate cornea to be made by the internodes fartherremoved from the apex. The length of each internode depends throughout its developmenton its position in the stem as well as on the length of theinternode below. The extent to which the elongation of an internodelags behind that of the one below increases upwards, a relationshipsimilar to that previously shown to exist in respect of thevascularization of the internodes. In plumules the length of petiole corresponding to any givenlength of internode undergoes a decrease followed by an increaseas the plumule is traversed from base to apex, a relationshipexactly oppsite to that previously demonstrated between thevascularization of a leaf-trace bundle and that of the internodein which it runs. The effects of age and of removal of cotyledonary reserves aredescribed, and it is concluded that it is unlikely that thecharacteristic difference in internode length within the limitsof a single shoot are wholly due to changes in the availabiltyof reserve substances.  相似文献
