Silver and bighead carp were stocked in a large pen to control the nuisance cyanobacterial blooms in Meiliang Bay of Lake Taihu. Plankton abundance and water quality were investigated about once a week from 9 May to 7 July in 2005. Biomass of both total crustacean zooplankton and cladocerans was significantly suppressed by the predation of pen-cultured fishes. There was a significant negative correlation between the N:P weight ratio and phytoplankton biomass. The size-selective predation by the two carps had no effect on the biomass of green alga Ulothrix sp. It may be attributed to the low fish stocking density (less than 40 g m−3) before June. When Microcystis dominated in the water of fish pen, the pen-cultured carps effectively suppressed the biomass of Microcystis, as indicated by the significant decline of chlorophyll a in the >38 μm fractions of the fish pen. Based on the results of our experiment and previous other studies, we conclude that silver and bighead carp are two efficient biomanipulation tools to control cyanobacterial (Microcystis) blooms in the tropical/subtropical eutrophic waters. Moreover, we should maintain an enough stocking density for an effective control of phytoplankton biomass.  相似文献

A bloom of the cyanobacteria Microcystis aeruginosa was sampled over the summer and fall in order to determine if the spatial and temporal patterns in cell density, chlorophyll a (chl a) concentration, total microcystins concentration, and percent microcystins composition varied with environmental conditions in San Francisco Estuary. It was hypothesized that the seasonal variation in Microcystis cell density and microcystin concentration was ecologically important because it could influence the transfer of toxic microcystins into the aquatic food web. Sampling for Microcystis cell density, chl a concentration, total microcystins concentration and a suite of environmental conditions was conducted biweekly at nine stations throughout the freshwater tidal and brackish water regions of the estuary between July and November 2004. Total microcystins in zooplankton and clam tissue was also sampled in August and October. Microcystis cell density, chl a concentration and total microcystins concentration varied by an order of magnitude and peaked during August and September when and αB were high. Low streamflow and high water temperature were strongly correlated with the seasonal variation of Microcystis cell density, total microcystins concentration (cell)−1 and total microcystins concentration (chl a)−1 in canonical correlation analyses. Nutrient concentrations and ratios were of secondary importance in the analysis and may be of lesser importance to seasonal variation of the bloom in this nutrient rich estuary. The seasonal variation of Microcystis density and biomass was potentially important for the structure and function of the estuarine aquatic food web, because total microcystins concentration was high at the base of the food web in mesozooplankton, amphipod, clam, and worm tissue during the peak of the bloom. Water quality experienced changes throughout the 3-year ecological engineering experiment in the drinking water source in Meiliang Bay of Lake Taihu. Average concentrations of TN, TP, NH4+, BOD5 and transparency in the drinking water source during the period of July–December 2005 were 1.85, 0.13, 0.23, 3.03 mg L−1 and 27.5 cm, respectively, decreasing by 47.9%, 21.2%, 83.3%, 54.4% and 24.2%, compared to concentrations from the same period in 2003. Concentrations of chlorophyll a and COD were 89.9 μg L−1 and 6.45 mg L−1, increasing by 27.9% and 17.7%, compared to the values in 2003. Cyanobacteria (mainly Microcystis) dominated the phytoplankton community in the ecological engineering area during July–December 2005. Densities of cyanobacteria and Microcystis were higher in 2005 than in 2004 and higher inside the engineering area than outside. Density percentages of cyanobacteria and Microcystis to total algae were above 90% and 60% during the bloom period. Average density of flagellate algae was higher during July–December 2005 than in 2004. Changes in water quality in the engineering area resulted mainly from the weakening of waves, decrease in concentrations of suspended solids, and assimilation of mass algae and periphytons. In spite of initial improvement of water quality, cyanobacterial bloom still determined the phytoplankton dynamics and variations. Additionally, nutrient concentration still remained at a high level without control of external loading. Therefore, a more holistic approach and long-term management should be adopted in Lake Taihu.  相似文献

Fed-batch cultures of recombinant Escherichia coli BL21 for producing human-like collagen were performed at different specific growth rates (0.1~0.25 h−1) before induction and at a constant value of 0.05 h−1 after induction by the method of pseudo-exponential feeding. Although the final biomass (around 69 g l−1) was almost the same in all fed-batch cultures, the highest product concentration (13.6 g l−1) was achieved at the specific growth rate of 0.15 h−1 and the lowest (9.6 g l−1) at 0.25 h−1. The mean productivity of human-like collagen was the highest at 0.15 h−1 (0.57 g l−1 h−1) and the lowest at 0.1 h−1 (0.35 g l−1 h−1). In the phase before induction, the cell yield coefficient (YX/S) decreased when the specific growth rate increased, while the formation of acetic acid increased upto 2.5 g l−1 at 0.25 h−1. The mean product yield coefficient (YP/S) also decreased with specific growth rate increasing. The respiration quotient (RQ) increased slightly with specific growth rate increasing before induction, and the mean value of RQ was around 72%. The optimum growth rate for human-like collagen production was 0.15~0.2 h−1.  相似文献

Blue-green algal blooms formed by Microcystis and Oscillatoria often occur in shallow eutrophic lakes, such as Lake Taihu (China) and Lake Kasumigaura (Japan). Growth characteristics and competitions between Microcystis aeruginosa and Oscillatoria mougeotii were investigated using lake simulator systems (microcosms) at various temperatures. Oscillatoria was the superior competitor, which suppressed Microcystis, when temperature was <20°C, whereas the opposite phenomenon occurred at 30°C. Oscillatoria had a long exponential phase (20 day) and a low growth rate of 0.22 day−1 and 0.20 day−1 at 15°C and 20°C, respectively, whereas Microcystis had a shorter exponential phase (2–3 days) at 30°C and a higher growth rate (0.86 day−1). Interactions between the algae were stronger and more complex in the lake simulator system than flask systems. Algal growth in the lake simulator system was susceptible to light attenuation and pH change, and algae biomasses were lower than those in flasks. The outcome of competition between Microcystis and Oscillatoria at different temperatures agrees with field observations of algal communities in Lake Taihu, indicating that temperature is a significant factor affecting competition between Microcystis and Oscillatoria in shallow, eutrophic lakes.  相似文献

Experiments on growth characters and ecological functions of the macroalgae Gracilaria lemaneiformis, collected from south China, were conducted in polyculture areas of kelp and filter-feeding bivalve in Sanggou Bay in Weihai City, Shandong, in north China from May 2002 to May 2003. The results of 116 days cultivation showed that the average wet weight of alga increased 89 times from 0.1 to 8.9 kg rope−1, with an average specific growth rate (based on wet weight) of 3.95% per day. The most favorable water layer for its growth was 1.0–1.8 m below the surface in July and August, with an average specific growth rate of 8.2% per day in 30-day experiments. Photosynthetic activity changed seasonally, with an average of 7.3 mg O2 g dw−1 h−1. The maximum rate (14.4 mg O2 g dw−1 h−1) was recorded in July, or 19.3 mg CO2 g dw−1 h−1, while the minimum (0.40 mg CO2 g dw−1 h−1) was in April. This study indicated that the culture of G. lemaneiformis is an effective way to improve water quality where scallops are cultivated intensively.  相似文献

In fall 1997, the toxic cyanobacterium Microcystis aeruginosa was documented in Lake Sammamish (western Washington, U.S.A.) for the first time. Cyanobacterial activity and environmental conditions that may promote toxic cyanobacteria were investigated during summer and fall 1999. Development of toxic Microcystis was hypothesized to be due to runoff of nutrients from the watershed (external loading hypothesis) or from vertical migration of dormant cyanobacteria from the nutrient-rich sediments into the water column (cyanobacterial migration hypothesis). Microcystins were detected using an enzyme-linked immunosorbent assay during late August and early September 1999 despite low cyanobacterial abundance. Microcystin concentrations ranged between 0.19–3.8 g l–1 throughout the lake and at all depths with the exception of the boat launch where concentrations reached 43 g l–1. Comparison of the conditions associated with the toxic episodes in 1997 and 1999 indicate that Microcystis is associated with a stable water column, increased surface total phosphorus concentrations (> 10 g l–1), surface temperatures greater than 22°C, high total nitrogen to phosphorus ratios (> 30), and increased water column transparency (up to 5.5 m). Migration of the cyanobacteria, Microcystis and Anabaena, occurred in both the deep and shallow portions of the lake. Microcystis dominated (89–99%) the migrating cyanobacteria with greater migration from the shallow station. External loading of nutrients due to the large rainfall preceding the 1997 toxic episode may have provided the nutrients needed to fuel that bloom. However, toxic Microcystis occurred in 1999 despite the lack of rain and subsequent external runoff. The migration of Microcystis from the nutrient-rich sediments may have been the inoculum for the toxic population detected in 1999.  相似文献

In the summer of 2004, a harmful bloom caused by Chattonella ovata (Raphidophyceae) occurred over almost the entire area of the Seto Inland Sea and caused fishery damages. This incident was the first record of a bloom and damage to the fisheries caused by this species in Japanese waters. In order to elucidate the mechanism of the bloom outbreak, we examined the nutrition and the growth kinetics in nitrogen (N)- and phosphorus (P)-limited semi-continuous cultures of this species. Inorganic N compounds, such as nitrate, nitrite, and ammonium, were found to be good nitrogen sources for the growth of C. ovata, while organic nitrogen (urea and uric acid) was not utilized. This species was capable of using ATP, ADP and inorganic phosphorus compounds, but did not utilize phosphate monoesters as a sole P source. Under both N-limited and P-limited steady state conditions, the growth rate as a function of cell nitrogen and phosphorus quota, respectively, followed the Droop equation. Kinetic parameters μm (maximum growth rate) and kq (minimum cell quota) obtained for N- and P-limited cultures were 0.79 day−1 and 5.5 pmol N cell−1 and 0.86 day−1 and 0.48 pmol P cell−1, respectively. The minimum cell quotas were 23–30% lower than those of C. antiqua. The nutrient availability and kinetic parameters of C. ovata are compared with other harmful algae and the ecological implications of these characteristics discussed.  相似文献

“Red tide” events are frequent and periodical in Bahía de Mazatlán, Sinaloa, México. Yet, the ones observed from 4 February to 4 June 2000, showed some distinctive features: First, the dinoflagellates Prorocentrum balticum (85%), P. mexicanum (5%), and Ceratium furca (5%), dominated the composition of the blooms; Second, the average cell abundance by date was 1.3×106 cells l−1, with a range of 3.5×103 to 24,500 × 103 cells l−1, well above previous records; Third, the temperature registered at 10–20 m deep was unusually cold (19 °C), below the normal average of 21.5 °C observed over the last 10 years. Salinity was high (35.9 psu) and showed very little influence on the water density gradient. A mean thermal stratification index (TSI), of 3.4, with a maximum of 7 °C, was observed throughout the period, in spite of a weak upwelling activity and intermittent strong NW winds which were unable to break up water column stratification. Temperature fluctuations at the surface and at the bottom layers showed a 30-day periodicity, suggesting some association with the lunar cycle. To explain the characteristics of the “red tides” registered in Bahía de Mazatlán during the winter–spring period of year 2000, it is proposed that the temperature and density stratification, stabilized further by internal waves that compensated for the weak upwelling activity and provided the necessary nutrients to the surface layer, favored the persistence and intensity of the harmful algal bloom events then observed.  相似文献

The elemental composition of cells of Anabaena flos-aquae was investigated by energy-dispersive electron probe X-ray microanalysis (10?kV) of mixed phytoplankton samples obtained from different depths within a stratified eutrophic freshwater lake. Preliminary studies had indicated that at 10?kV, X-ray information was derived mainly from the peripheral region (up to 1–2?μm) of cells. Routinely detectable elements (present in at least 50% of cells) included Mg (overall mean 112?mmol?kg?1 dry mass), Si (1850), P(313), S (113), Cl (92), K (300) and Ca (56). Peaks of Na and Al were also frequently present. Individual elements showed a wide range of intracellular concentrations at each depth within the lake. Si had a clear bimodal distribution at depths of 1–8?m, indicating both low-Si (mean concentrations 54–290–3800?mmol?Si?kg?1) and high-Si (2770–3800?mmol?Si?kg?1) cell subpopulations. Low-Si cells had significantly higher concentrations of other detectable elements compared with high-Si cells, but could not be distinguished from the latter in terms of morphology or stage of cell cycle (comparison of dividing and non-dividing cells). High-Si cells were intermixed with low-Si cells in individual Anabaena colonies and decreased proportionally with depth, ranging from approximately 75% in the surface sample to 10% at 8?m. With the exception of Si, mean elemental concentrations of Anabaena populations were closely similar throughout the epilimnion. These differed from the metalimnion sample, which had significantly higher concentrations of Mg, P, S and K, and a lower concentration of Si. Mean ratios of certain elements (Mg, P, S and K) and ion groups (monovalent/divalent cations) were highly constant throughout the sampled water column. At each depth, most of the cell elements were significantly and positively inter-correlated (Pearson analysis), with Mg, P, S and K typically constituting the major group (factor analysis). Correlations with Si were invariably negative, suggesting a separate (possibly cell wall) location of this element in the cell.  相似文献

The annual algal bloom (February–June) in Lake Kinneret consists almost entirely of the dinoflagellatePeridinium cinctum f.westii (Dinophyceae). To clarify the role of phosphatases in the alga, experiments were carried out using cells from culture or from the lake. In culture, as the external ambient orthophosphate (Pi) concentration decreased, alkaline phosphatase activity increased (and to some extent acid phosphatase activity, as well). Hot water extractable P decreased, although molybdate reactive phosphorus (MRP) appeared to be utilized in preference to the non-MRP component of this pool. Alkaline phosphatase inPeridinium collected from the lake as well as cells grown in culture under a high (3–6 mg l–1) ambient Pi concentration in both continuous light and a 12:12 light-dark cycle, showed a diurnal fluctuation in activity. These results, together with previous observations suggest that the phosphatases inPeridinium are controlled by changes in intracellular phosphorus levels (other than the hot water extractable pool) and/or by other metabolic processes not directly involved in P nutrition.  相似文献

(R)-Phenylacetylcarbinol (PAC), a pharmaceutical precursor, was produced from benzaldehyde and pyruvate by pyruvate decarboxylase (PDC) of Candida utilis in an aqueous/organic two-phase emulsion reactor. When the partially purified enzyme in this previously established in vitro process was replaced with C. utilis cells and the temperature was increased from 4 to 21 °C, a screen of several 1-alcohols (C4–C9) confirmed the suitability of 1-octanol as the organic phase. Benzyl alcohol, the major by-product in the commercial in vivo conversion of benzaldehyde and sugar to PAC by Saccharomyces cerevisiae, was not formed. With a phase volume ratio of 1:1 and 5.6 g C. utilis l−1 (PDC activity 2.5 U ml−1), PAC levels of 103 g l−1 in the octanol phase and 12.8 g l−1 in the aqueous phase were produced in 15 h at 21 °C. In comparison to our previously published process with partially purified PDC in an aqueous/octanol emulsion at 4 °C, PAC was produced at a 4-times increased specific rate (1.54 versus 0.39 mg U−1 h−1) with simplified catalyst production and reduced cooling cost. Compared to traditional in vivo whole cell PAC production, the yield on benzaldehyde was 26% higher, the product concentration increased 3.9-fold (or 6.9-fold based on the organic phase), the productivity improved 3.1-fold (3.9 g l−1 h−1) and the catalyst was 6.9-fold more efficient (PAC/dry cell mass 10.3 g g−1).*Dedicated with gratitude to Prof. Dr. Franz Lingens – “Theo”.  相似文献

Toxic Microcystis blooms frequently occur in eutrophic water bodies and exist in the form of colonial and unicellular cells. In order to understand the mechanism of Microcystis dominance in freshwater bodies, the physiological and biochemical responses of unicellular (4 strains) and colonial (4 strains) Microcystis strains to phosphorus (P) were comparatively studied. The two phenotype strains exhibit physiological differences mainly in terms of their response to low P concentrations. The growth of four unicellular and one small colonial Microcystis strain was significantly inhibited at a P concentration of 0.2 mg l−1; however, that of the large colonial Microcystis strains was not inhibited. The results of phosphate uptake experiments conducted using P-starved cells indicated that the colonial strains had a higher affinity for low levels of P. The unicellular strains consumed more P than the colonial strains. Alkaline phosphatase activity in the unicellular strains was significantly induced by low P concentrations. Under P-limited conditions, the oxygen evolution rate, F v/F m, and ETR max were lower in unicellular strains than in colonial strains. These findings may shed light on the mechanism by which colonial Microcystis strains have an advantage with regard to dominance and persistence in fluctuating P conditions. Handling editor: L. Species identification of the common filamentous green alga Spirogyra is mainly based on the conjugation process and zygospores. However, this genus is mostly found in its vegetative stage, which complicates studies on the ecological demands for individual species. We therefore used a different approach by assessing the relationship between vegetative Spirogyra filament type groups (morphotypes) and environmental conditions (mainly ions, nutrients, light supply and water temperature). Sampling was done at 133 sites in Central Europe and in total 333 different filament types were classified. Spirogyra was found at pH values between 6.2 and 9.1, while total alkalinity ranged from 0.6 to 7.9 mequiv l−1. The genus is colonizing habitats with a specific conductivity between 75 and 1500 μS cm−1. Total phosphorus amounts varied between 1 and 2240 μg l−1 with a median value of 34 μg l−1, indicating meso- to eutrophic conditions as optimal growth range. Filament type grouping by means of cluster analysis was based on cell cross walls (plane or replicate), average cell widths and average chloroplast numbers and resulted in 10 groups with plane cross walls and three with replicate cross walls. Canonical correspondence analysis revealed nutrients to be the key factor for morphotype occurrence: filaments with increased cell widths preferred elevated nutrient conditions. Other environmental variables (ions, buffer capacity, light supply and water temperature) had no significant effects on morphotype occurrence.  相似文献

Increasing sucrose from 20 to 50 g l−1 in Uncaria tomentosa cell suspension cultures enhanced ursolic acid and oleanolic acid production from 129 ± 61 to 553 ± 193 μg g−1 cell dry wt. The maximal concentration of both triterpenes (1680 ± 39 μg g−1 cell dry wt) was 8 days after elicitation by jasmonic acid, while yeast extract or citrus pectin treatments produced 1189 ± 20 or 1120 ± 26 μg g−1 cell dry wt, respectively. Rates of inorganic nitrogen uptake by three Northeast US and three Asian species of Porphyra were compared in short-term incubations to evaluate potential for longer term and larger scale examination of bioremediation of nutrient-loaded effluents from finfish aquaculture facilities. The effects of nitrogen (N) species and concentration, temperature, acclimation history, and irradiance were investigated. Uptake rates increased ca. nine-fold from 20 to 150 μM N. Nitrate and ammonium uptake occurred at similar rates. Irradiance had a strong effect, with uptake at 40 μmol photons m−2 s−1only 55% of uptake at 150 μmol photons m−2 s−1. N-replete tissue took up inorganic nitrogen at rates that averaged only 60% of nutrient-deprived tissue. Although there were species (P. amplissima > (P. purpurea = P. umbilicalis)) and temperature effects (10 °C>5 °C>15 °C), interactions among factors indicated that individual species be considered separately. Overall, P. amplissima was the best Northeast US candidate. It took up ammonium at faster rates than other local species at 10 and 15 °C, two temperatures that fall within the expected range of industrial conditions for finfish operations.  相似文献

Colonial aggregation of Microcystis plays a key role in bloom formation. Limited studies have been reported about effects of environmental factors on the aggregation of Microcystis. Calcium is an important chemical element in water system. In this study, we investigated the effects of a low- (0.015 g l−1) and a high-concentration of calcium (0.100 g l−1) on the aggregation and buoyancy of a colonial strain M. aeruginosa XW01. Results show that compared to the low concentration of calcium, the high-calcium condition results in bigger colonial size, higher level of buoyancy and increased production of extracellular polysaccharides (EPS) of M. aeruginosa XW01. Increased production of EPS induced by the high-calcium concentration should contribute to the colonial aggregation and buoyancy of M. aeruginosa XW01. These results suggest that an increase in calcium concentration may be beneficial for Microcystis blooms occurring in a soft water lake.  相似文献

In order to calibrate carrying capacity models, investigations were conducted into the effects of food concentration and food quality on the feeding rates of small (25–50 mm), medium (60–85 mm) and large (90–115 mm) Greenshell mussels (Perna canaliculus). Experimental diets varying from 3.3 to 6.0 μg l−1 chlorophyll a concentration and 12–25% organic content were fed to mussels housed in individual flow through chambers. Not surprisingly, this study found that the main factor affecting feeding rates is mussel size. Small mussels were observed to maintain a constant filtration rate of approximately 20 mg h−1 irrespective of food concentration or quality, whereas mussels of greater than 60 mm length had more variable filtration rates between 30 and 80 mg h−1. The filtration rates of these large mussels were also observed to increase positively with organic content, and showed no sign of levelling out, even at the highest organic content tested (25%). Highest rejection rates (50–70 mg h−1) were observed when the organic content of the available seston was low, suggesting that P. canaliculus are able to selectively reject organic material, thereby organically enriching their diet. It appears that the organic content of the seston is the primary determinant of the net efficiency with which food is selected from the available seston by the mussel. The present study shows that P. canaliculus of all sizes are capable of adapting their feeding behaviour to compensate for changes in the food supply, which may occur over relatively short time periods, in the culture environment.  相似文献

Cyanobacteria blooms (especially Microcystis spp.) are thought to alter dominance of large-sized daphnids into small-sized metazoan zooplankton. However, several field investigations show different phenomena. Laboratory experiments were conducted based on the hypothesis that different Microcystis spp. concentrations would influence competitive outcomes using two algal combinations of different concentrations and four species of cladocerans. In the algal combination of 50 mg l−1 colonial Microcystis spp. + 1 mg l−1 Scenedesmus obliquus (fresh weight), Daphnia carinata was absent during the experiment in competition with other cladocerans. Decreasing colonial Microcystis spp. concentration (10 mg l−1) resulted in a shift from dominance by small-sized cladocerans to dominance by D. carinata. No significant effects of different concentrations of colonial Microcystis spp. on competitive outcomes were shown among three small-sized cladocerans. These results support the idea that cyanobacteria concentration affects the dominance status of large-bodied daphnid.  相似文献
