Bacterial inoculation may influence Brassica juncea growth and heavy metal (Ni, Cr, and Cd) accumulation. Three metal tolerant bacterial isolates (BCr3, BCd33, and BNi11) recovered from mine tailings, identified as Pseudomonas aeruginosa KP717554, Alcaligenes feacalis KP717561, and Bacillus subtilis KP717559 were used. The isolates exhibited multiple plant growth beneficial characteristics including the production of indole-3-acetic acid, hydrogen cyanide, ammonia, insoluble phosphate solubilization together with the potential to protect plants against fungal pathogens. Bacterial inoculation improved seeds germination of B. juncea plant in the presence of 0.1 mM Cr, Cd, and Ni, as compared to the control treatment. Compared with control treatment, soil inoculation with bacterial isolates significantly increased the amount of soluble heavy metals in soil by 51% (Cr), 50% (Cd), and 44% (Ni) respectively. Pot experiment of B. juncea grown in soil spiked with 100 mg kg?1 of NiCl2, 100 mg kg?1 of CdCl2, and 150 mg kg?1 of K2Cr2O7, revealed that inoculation with metal tolerant bacteria not only protected plants against the toxic effects of heavy metals, but also increased growth and metal accumulation of plants significantly. These findings suggest that such metal tolerant, plant growth promoting bacteria are valuable tools which could be used to develop bio-inoculants for enhancing the efficiency of phytoextraction.  相似文献

The microbial abundance, the percentage of viable bacteria, and the diversity of bacterial isolates from different regions of a 83.45-m ice core from the Puruogangri glacier on the Tibetan Plateau (China) have been investigated. Small subunit 16S rRNA sequences and phylogenetic relationships have been studied for 108 bacterial isolates recovered under aerobic growth conditions from different regions of the ice core. The genomic fingerprints based on ERIC (enterobacterial repetitive intergenic consensus)-polymerase chain reaction and physiological heterogeneity of the closely evolutionary related bacterial strains isolated from different ice core depths were analyzed as well. The results showed that the total microbial cell, percentages of live cells, and the bacterial CFU ranged from 104 to 105 cell ml−1 (Mean, 9.47 × 104; SD, 5.7 × 104, n = 20), 25–81%, and 0–760 cfu ml−1, respectively. The majority of the isolates had 16S rRNA sequences similar to previously determined sequences, ranging from 92 to 99% identical to database sequences. Based on their 16S rRNA sequences, 42.6% of the isolates were high-G + C-content (HGC) gram-positive bacteria, 35.2% were low-G + C (LGC) gram-positive bacteria, 16.6% were Proteobacteria, and 5.6% were CFB group. There were clear differences in the depth distribution of the bacterial isolates. The isolates tested exhibited unique phenotypic properties and high genetic heterogeneity, which showed no clear correlation with depths of bacterial isolation. This layered distribution and high heterogeneity of bacterial isolates presumably reflect the diverse bacterial sources and the differences in bacteria inhabiting the glacier’s surface under different past climate conditions.  相似文献

Effects of heavy metals on the isopod Asellus aquaticus (L.) are studied by static toxicity tests. Results demonstrate that the species is sensitive to Cd+2, Cr+6, Cu+2, Fe+3, Hg+2, Ni+2 Pb+2 and Zn+2, but the toxicity of each metal is different. Differences are also found between adults and between adults and juveniles. The comparative analysis of all data on the toxicity has been performed on the concentrations of metal ions and not on metal compound concentrations.Criteria for establishing water quality in order to guarantee protection of the environment are discussed.  相似文献

The effect of heavy metals on community structure of a heavy metal tolerant sulfidogenic consortium was evaluated by using a combination of denaturing gradient gel electrophoresis (DGGE) of 16S rRNA gene and dissimilatory sulfite reductase (dsrB) gene fragments, 16S rRNA gene cloning analysis and fluorescence in situ hybridization (FISH). For this purpose, four anaerobic semi-continuous stirred tank reactors (referred as R1–R4) were run in parallel for 12 weeks at heavy metal loading rates of 1.5, 3, 4.5 and 7.5 mg l?1 d?1 each of Cu2+, Ni2+, Zn2+, and Cr6+, respectively. The abundance ratio of Desulfovibrio vulgaris detected by FISH to total cell counts was consistent with the obtained results of cloning and DGGE. This indicated that D. vulgaris was dominant in all analyzed samples and played a key role in heavy metal removal in R1, R2, and R3. In contrast, after 4 weeks of operation of R4, a distinct biomass loss was observed and no positive hybridized cells were detected by specific probes for the domain Bacteria, sulfate-reducing bacteria and D. vulgris. High removal efficiencies of heavy metals were achieved in R1, R2 and R3 after 12 weeks, whereas the precipitation of heavy metals in R4 was significantly decreased after 4 weeks and almost not observed after 6 weeks of operation. In addition, the anaerobic bacteria, such as Pertrimonas sulfuriphila, Clostridium sp., Citrobacter amalonaticus, and Klebsiella sp., identified from DGGE bands and clone library were hypothesized as heavy metal resistant bacteria at a loading rate of 1.5 mg l?1 d?1 of Cu2+, Ni2+, Zn2+, and Cr6+.  相似文献

Eleven bacterial strains were isolated from soil samples collected from mine tailings. Bacterial strains were checked for tolerance against heavy metals (Cr, Cd, Ni), using the agar dilution method. All the strains showed multiple tolerances against heavy metals, but the most promising results appeared in strains BCr3, BCd33, and BNi11: they were tolerant to 15 mM of Cr6+, 7.5 mM of Cd2+, and 10 mM of Ni2+, respectively. The effect of heavy metals on bacterial growth was tested together with their ability to grow in different pH, NaCl, and temperature values. Bacterial isolates grew well between pH 7.5 and 8.5. The optimum temperature for maximum growth was between 35 and 37°C, and no significant change in bacterial growth was observed in the presence of 2% NaCl. In addition, the bioaccumulation potential of bacterial strains was investigated. Bacterial strains BCr3, BCd33, and BNi11 showed high bioaccumulation ability of Cr (68.7%), Cd (72.4%), and Ni (69.8%), respectively. All bacterial isolates were identified by 16S rRNA gene sequencing. Analysis of plasmid content revealed that all bacterial isolates contained a single plasmid. Further, polymerase chain reaction together with DNA sequence analysis was used to screen all bacterial strains for the presence metal tolerance genes (czcD, chrA, chrB, czcB, czcC, nccA, and cadA) on both plasmid and chromosomal genomes.  相似文献

Environmental pollution with toxic heavy metals is increasing throughout the world alongside industrial development. Microorganisms and microbial products can be highly efficient bioaccumulators of soluble and particulate forms of metals, especially dilute external solutions. Microbe related technologies (Biotechnology) may provide an alternative or additive conventional method for metal removal or metal recovery. This study dealt with isolation, identification and characterization of heavy metal-resistant (Pb2+, Cd2+, Al3+, Cu2+, Ag2+ and Sn2+) bacteria from sewage wastewater at Taif Province, Saudi Arabia. Nine bacterial isolates were selected by using an enrichment isolation procedure based on high level of heavy metal resistance. All the isolates showed high resistance to heavy metals with Minimum Inhibitor Concentration (MIC) ranging from 800 μg/ml to 1400 μg/ml. All nine resistant isolates showed multiple tolerances to heavy metals. On the basis of morphological, biochemical and 16S rRNA characterization, the most potent isolates (Cd1-1, Ag1-1, Ag1-3 and Sn1-1) were identified as Alcaligenes faecalis. Scanning electron microscope analysis showed that the morphology of Alcaligenes faecalis Ag1-1 was unchanged after growth in medium without and with addition of Ag2+ indicative Ag2+ is not toxic to the isolate under the conditions tested. The ability of Alcaligenes faecalis Ag1-1 to synthesize sliver nanoparticles was examined. The heavy metal-resistant bacteria obtained could be useful for the bioremediation of heavy metal-contaminated environment.  相似文献

Marine bacteria possess a wide variety of bioremediation potential which is beneficial environmentally and economically. In this study, bacterial isolates from marine waters were screened for tolerance and growth in high concentrations of chromate (Cr6+). Two isolates, capable of tolerating Cr6+ concentrations 300 µg mL?1 or higher, and found to completely reduce 20 µg mL?1 Cr6+ were grown in Cr6+ (50 and 100 mg kg?1) spiked garden soil. Notably, both facilitated normal germination and growth of mung (Vigna radiata) seeds, which could hardly germinate in Cr6+ spiked garden soil without either of these bacteria. In fact, large percent of mung seeds failed to sprout in the Cr6+ spiked garden soil and could not grow any further. Apparently, chromate detoxification by marine bacterial isolates and the ability of mung plants to deal with the reduced form appear to work complementarily. This study provides an insight into marine bacterial abilities with respect to chromium and potential applications in promoting growth of leguminous plants-similar to mung in particular-in Cr6+ contaminated soil.  相似文献

In order to evaluate the effect of microwave radiation on immobilization of heavy metals (Cu 2 + , Cr 6 + , Zn 2 + and Pb 2 + ) in sediment sludge, leaching tests were run under different test conditions to compare microwave radiation with conventional blast heating and drying process for their effectiveness in immobilizing heavy metals within sediment sludge. Experimental results indicate that microwave radiation can decrease the concentration of heavy metals in leachate by 63% ~ 70% more than conventional blast heating and drying process in an aclinic shake leaching test. Under the same simulated natural water conditions, the concentration of heavy metals in the leachate using microwave radiation is well below the concentration of heavy metals in leachate using conventional blast heating and drying process. It is therefore concluded that microwave radiation is much more effective than conventional blast heating and drying process in immobilizing heavy metals in sediment sludge.  相似文献

In this study, we fabricated a novel recycled aggregate (RA) bio-carrier with sulfate-reducing bacteria (SRB) for the effective removal of the heavy metals Zn2+, Ni2+, and Cr6+ from contaminated seawater. By using only RA carriers, Zn2+, Ni2+, and Cr6+ could be removed up to 35.6, 11.0, and 61.3% and SRB-immobilized RA biocarriers could effectively lead to the removal of Zn2+, Ni2+, and Cr6+ up to 100.0, 98.0, and 87.8% for 100 ppm concentration of the heavy metals at 37°C, respectively. These results indicate that recycled aggregate bio-carriers with SRB hold great potential for use in the removal of cationic heavy metal species from marine environment.  相似文献

Pollution of soil with heavy metals, herbicides, antibiotics and other chemicals is known to have a negative effect on microbial activities. Therefore, the aim of this study was to isolate cultures of Azotobacter sp. from polluted and unpolluted soils and to study the effect of these pollutants on their growth. A total of 120 Azotobacter sp. were isolated from soils irrigated with wastewater (contaminated soils) and groundwater (uncontaminated soils). These isolates were screened for resistance to heavy metals, herbicide and antibiotics. Also, the soils from which the cultures were isolated were analyzed for the concentrations of Zn2+, Cd2+, Cu2+, Pb2+ and Mn2+ they contained. Contaminated soil showed high levels of heavy metals as compared to uncontaminated soil. The size of the Azotobacter population in contaminated soil was lower than that in uncontaminated soil. Of the Azotobacter isolates, 64 that were recovered from contaminated soil exhibited high resistance to heavy metals (Hg2+, Cd2+, Cu2+, Cr3+, Co2+, Ni2+, Zn2+ and Pb2+) and herbicide 2,4-D compared to 56 isolates from uncontaminated soil. Also, isolates from contaminated soil showed high resistance to chloramphenicol, nitrofurantoin and co-trimoxazole compared to those isolated from uncontaminated soil. The majority of Azotobacter isolates from contaminated soil showed multiple-resistance to different metal ions and antibiotics. All isolates failed to grow at pH less than 6. Salt concentration (5%) was found to be inhibitory to all isolates. The most potent isolates from contaminated soil that showed multiresistance to all substances tested were identified on the basis of morphological and biochemical characteristics, and 16S rRNA as A. chroococcum. These resistant isolates could be employed in contaminated soils and/or bioremediation.  相似文献

Aims: To isolate and characterize actinomycetes with probiotic activities from healthy goat faeces. Methods and Results: Faecal actinomycetes were isolated by dilution methods and identified by 16S rRNA gene sequence analysis. The hydrolytic enzyme activities were analysed by clear zone formation. The antimicrobial activities and resistance to heavy metals were tested by growth inhibition methods. The isolates belong to a small group of actinobacterial genera, including Streptomyces, Nocardiopsis and Oerskovia. The Oerskovia was the most widely distributed genus among the cultures. The proportion of streptomycete‐like strains producing amylase or protease is significantly higher than those of other actinomycetes (P < 0·05). Compared with streptomycete‐like strains, a higher proportion of (α‐ or β‐) galactase‐producing other actinomycetes was found in goat faeces. More than 50% of streptomycete‐like strains showed activities against test fungi. Streptomycetes could tolerate 0·25 mmol l?1 Cr2O72?, 2 mmol l?1 Ni2+; however, other actinomycetes are liable to 40 mmol l?1 Fe3+ and 0·25 mmol l?1 Cr2O72? and resistant to 5 mmol l?1 Ni2+ and 2 mmol l?1 Cu2+. Conclusions: The different physiological characteristics of the actinomycetes suggested that the cooperation in the actinomycetes might be involved in their association with goat. Significance and Impact of the Study: Probiotic mixtures based on faecal actinomycetes showed potentials in animal production.  相似文献

The effects of heavy metals (Cd, Cr and Cd + Cr) on the motility parameters and oxidative stress of sterlet (Acipenser ruthenus) sperm were investigated in vitro. Sturgeon sperm were exposed for 2 h to heavy metals at environmental related concentrations (0.1 mg L?1 Cr, 0.001 mg L?1 Cd, 0.1 mg L?1 Cr + 0.001 mg L?1 Cd) and higher concentrations (5.0 mg L?1 Cr, 0.05 mg L?1 Cd, 5.0 mg L?1 Cr + 0.05 mg L?1 Cd). Results revealed that environmental concentrations of heavy metals had no significant influence on motility parameters and antioxidant responses indices in sturgeon sperm, except for LPO level and SOD activity. But higher concentrations of these metals induced oxidative tress in sturgeon sperm in vitro, associated with sperm motility parameters inhibition. Our results suggest that using of sperm in vitro assays may provide a novel and efficiently means for evaluating the effects of residual heavy metals in aquatic environment on sturgeon.  相似文献

By combining two functions of alginate gel and activated carbon, an activated carbon-containing alginate bead (AC-AB) adsorbent was developed and successfully used to simultaneously remove heavy metal ions and toxic organics. Quantitative analysis showed that almost all of the adsorption of toxic organics, such as p-toluic acid, is caused by the activated carbon in the AC-AB adsorbent, whereas the alginate component has a major role in the removal of heavy metals. A 50-L solution containing eight heavy metals (Pb2+, Mn2+, Cd2+, Cu2+, Zn2+, Fe2+, Al3+ and Hg2+) and four mineral ions was run continuously through a filter cartridge packed with 160 g of the AC-AB adsorbent. The adsorbent showed a high capacity to remove heavy metals completely from the water, while allowing essential minerals, such as K+, Na+, Mg2+ and Ca2+, to pass through the filter. The adsorbent could be regenerated using eluents, such as HNO3, and reused repeatedly without considerable loss of its metal uptake capacity through 10 subsequent cycles of adsorption and desorption. With its high capacity and high selectivity for toxic heavy metals, the AC-AB adsorbent has enormous potential for application in drinking water treatment technologies.  相似文献

Bacteria were isolated from deep terrestrial subsurface sediments underlying the coastal plain of South Carolina. A total of 163 isolates from deep sediments, surface soil, and return drill muds were examined for plasmid DNA content and resistance to the antibiotics penicillin, ampicillin, carbenicillin, streptomycin, kanamycin, and tetracycline. MICs of Cu2+, Cr3+, and Hg2+ for each isolate were also determined. The overall frequency of plasmid occurrence in the subsurface bacteria was 33%. Resistance was most frequent to penicillin (70% of all isolates), ampicillin (49%), and carbenicillin (32%) and was concluded to be related to the concentrations of the individual antibiotics in the disks used for assaying resistance and to the production of low levels of β-lactamase. The frequencies of resistance to penicillin and ampicillin were significantly greater for isolates bearing plasmids than for plasmidless isolates; however, resistance was not transferable to penicillin-sensitive Escherichia coli. Hybridization of subsurface bacterial plasmids and chromosomal DNA with a whole-TOL-plasmid (pWWO) probe revealed some homology of subsurface bacterial plasmid and chromosomal DNAs, indicating a potential for those bacteria to harbor catabolic genes on plasmids or chromosomes. The incidences of antibiotic resistance and MICs of metals for subsurface bacteria were significantly different from those for drill mud bacteria, ruling out the possibility that bacteria from sediments were derived from drill muds.  相似文献

Pistia stratiotes is used for the epuration of domestic sewage in the Biyem Assi phytopurification station. During the process, Fe2+, Mn2+, Zn2+ and Pb2+ are absorbed in substantial amounts by the plant. These metals modify the H+/K+ exchange system at the root level. H+ efflux is inhibited by Fe2+ and by Zn2+ and enhanced by Mn2+ and Pb2+. K+ influx is inhibited by Fe2+, by Zn2+ and by Pb2+ and enhanced by Mn2+. It is shown that the purification capacity ofPistia stratiotes can vary with the composition of the heavy metals in the surrounding medium.  相似文献

Riparian sediment is an important active interface zone for nitrogen-biogeochemical cycles. However, the distribution characteristics of anammox bacteria are not adequately described in the riparian sediment of urban rivers. Therefore, the distribution characteristics of anammox bacteria of Guyun river riparian sediment, which is the representative urban river, were investigated in this study. Research results showed that anammox bacterial abundance showed a significant spatial variation at the study area. Abundance values ranged from 2.00?×?105 to 1.92?×?107 copies·g?1?dry sediment. And the average value was 6.64×106 copies·g?1 dry sediment in six collected samples. Abundance of anammox bacteria was significantly related to TN, NH4+-N, and pH. Slight spatial variations of richness and diversity in Guyun river riparian sediment were obtained and physicochemical parameters of sediment samples did not significantly influence the biodiversity index of anammox bacteria. Phylogenetic analysis indicated that all amplified sequences fell within the known anammox bacterial group, i.e., Brocadia, Anammoxoglobus/Jettenia, and Kuenenia, with proportions of 33.91%, 24.35%, and 41.74%, respectively. The relative abundance of anammox bacteria showed distinctive spatial heterogeneity in six different sample sites. This finding indicated a distinct spatial variation of anammox bacterial in Guyun river riparian sediment.  相似文献

To provide a basis for using indigenous bacteria for bioremediation of heavy metal contaminated soil, the heavy metal resistance and plant growth-promoting activity of 136 isolates from V-Ti magnetite mine tailing soil were systematically analyzed. Among the 13 identified bacterial genera, the most abundant genus was Bacillus (79 isolates) out of which 32 represented B. subtilis and 14 B. pumilus, followed by Rhizobium sp. (29 isolates) and Ochrobactrum intermedium (13 isolates). Altogether 93 isolates tolerated the highest concentration (1000 mg kg−1) of at least one of the six tested heavy metals. Five strains were tolerant against all the tested heavy metals, 71 strains tolerated 1,000 mg kg−1 cadmium whereas only one strain tolerated 1,000 mg kg−1 cobalt. Altogether 67% of the bacteria produced indoleacetic acid (IAA), a plant growth-promoting phytohormone. The concentration of IAA produced by 53 isolates was higher than 20 µg ml−1. In total 21% of the bacteria produced siderophore (5.50–167.67 µg ml−1) with two Bacillus sp. producing more than 100 µg ml−1. Eighteen isolates produced both IAA and siderophore. The results suggested that the indigenous bacteria in the soil have beneficial characteristics for remediating the contaminated mine tailing soil.  相似文献

The biochemical basis for resistance to toxicity is complicated by the great variety of reactions at the molecular and cellular levels even in closely related organisms and tissues. Several strategies for resistance to intoxication have been identified. Metal ion interactions in biology can be divided into three classes representing fast, intermediate and slow exchange with biological ligands. Examples of those elements in fast exchange include the alkali metals Na+ and K+, the alkali earth metals Ca2+ and Mg2+, and, of course, H+. Those which can sometimes be in intermediary exchange are Fe2+ and Mn2+. Examples of those in slow exchange are generally in the active sites of metalloenzymes, e.g., Fe3+, Zn2+, Ni2+, Cu2+. In the presented paper, the cycling of one essential element (nickel) and one non-essential element (mercury) are reviewed with special emphasis on their mobilities in the event of in situ sediment contamination.  相似文献

This paper reports the synthesis of azomethine-modified gold nanoparticles with azomethine (azomethine-AuNPs) in aqueous media, which were characterized by FT-IR spectroscopy, ultraviolet–visible spectroscopy (UV-Vis), dynamic light scattering (DLS), thermogravimetric analysis (TGA), and transmission electron microscopy (TEM). The azomethine-AuNPs were employed as colorimetric for Cr3+ and Co2+ ions at pH 6.2–7.5 and 8.1–9.1, at room temperature in aqueous solution. In the presence of Cr3+ and Co2+, the azomethine-AuNPs induce aggregation of the nanoparticles. Upon aggregation, the surface plasmon absorption band red-shifts so that the nanoparticle solution appears a blue color. The sensitivity of azomethine-AuNPs towards other metal ions, Mg2+, Mn2+, Cr6+, Na+, Ni2+, Ag+, Al3+, Ca2+, Cd2+, Cu2+, Fe2+, Fe3+, Hg2+, Cd2+, K+, Co3+, Ni2+, Pb2+, and Zn2+ are negligible. This highly selective sensor allows a direct quantitative assay of Co2+ and Cr3+ with colorimetric detection limits of 83.22 and 108 nM, respectively.

Aims: Forty‐nine bacteria isolated from four newly‐produced waste samples of a steel industry, which had a high content of CaO, MgO, Cr and P2O5, were characterized molecularly and phenotypically by susceptibility testing against heavy metals. Methods and Results: Phylogenetic analysis using 16S rRNA gene sequences revealed that the isolates belonged to nine genera, Pseudomonas, Micrococcus, Acinetobacter, Bacillus, Dietzia, Kocuria, Diaphorobacter, Staphylococcus and Brevibacillus. Besides, some isolates could be affiliated to species: M. luteus, Ac. junii, Ac. schindleri, B. cereus, K. marina, D. nitroreducens and Staph. warneri. The bacteria that were characterized are taxonomically diverse, and Pseudomonas and Micrococcus predominated. Fingerprinting BOX‐PCR revealed high genomic heterogeneity among the isolates. Among the heavy metal compounds Zn, Ni, Pb and Cu were least toxic to the bacterial isolates, whereas Ag inhibited all isolates at 0·001 mmol l?1. Conclusions: Heterotrophic bacteria, affiliated with several phylogentic groups, were able to colonize different wastes of a steel industry. Significance and Impact of the Study: This study extends our knowledge of the early colonizers bacteria populating siderurgic environments. Some of these bacteria could have potential for recycling siderurgic waste for steel production.  相似文献
