达乌里胡枝子(Lespedezadavurica)是黄土高原地区重要的生态修复草种,但土壤盐碱化极大影响了达乌里胡枝子的生长,制约了其经济和生态价值。接种根际促生菌剂是提高植物对盐碱胁迫抗性的重要途径。【目的】验证醋酸钙不动杆菌(Acinetobacter calcoaceticus)对盐碱胁迫下达乌里胡枝子幼苗和成株的促生效应,初步揭示其促进豆科植物生长的潜在机理,为田间应用提供理论依据。【方法】采用解钾培养基对2株达乌里胡枝子根际分离的醋酸钙不动杆菌的解钾能力进行测定;采用液体发酵法对菌株分泌铁载体、吲哚乙酸(indole-3-acetic acid, IAA)的能力进行定量测定;分别在培养皿和盆栽条件下,验证菌株的促生能力及其机制。【结果】菌株DP25和DP27具有解钾和分泌铁载体、IAA的能力,其中相对铁载体表达量值(siderophore, SU)分别为53.13%和86.67%,IAA含量分别为1.01 mg/L和17.31 mg/L;菌株DP25显著提高了培养皿中达乌里胡枝子幼苗的茎长和根长;菌株DP25和DP27对盐碱胁迫下达乌里胡枝子成株的光合色素合成及其光合作用产生...  相似文献

Aims: To synthesize two heterologous endochitinases in Escherichia coli and demonstrate their potential for applied use in generating antibacterial chitin-derived oligosaccharides (OGS). Methods and Results: Heterologous endochitinase genes, chiA Nima and chiA74, were expressed in E. coli. Endochitinases were secreted by the E. coli export machinery and by ∼20 h maximal chitinolytic activity was observed. The highest chitinolytic activity was observed with ChiA Nima, which produced antibacterial OGS with activities against Enterobacter cloacae, Escherichia coli, Staphylococcus aureus and S. xylosus. Conclusions: It was shown that the export machinery of E. coli is well suited for the secretion of bioactive ChiA74 and ChiA Nima endochitinases, and that the latter can generate antibacterial OGS. Significance and Impact of the Study: Our study suggests that it is feasible to synthesize endochitinases ChiA Nima and ChiA74 codified by E. coli and mass-produce these enzymes in culture supernatants. As signal peptides in native ChiA Nima and ChiA74 were recognized by the protein export molecular apparatus in E. coli, these short peptides could be included as signal sequences for transport in E. coli of other proteins with applied value. This is the first report suggesting that ChiA Nima can be used to produce OGS to control food-borne pathogenic bacteria.  相似文献

固氮鱼腥藻作为光能自养型微生物,具备良好的碳氮固存能力。施加固氮鱼腥藻能够提升土壤肥力并减少化肥施用量。然而,解析固氮鱼腥藻在土壤中的碳氮固存机制以及不同菌株固存效率差异仍有待深入研究。因此,从单细胞水平对土壤固氮鱼腥藻菌株进行筛选和碳氮固存过程的研究至关重要。针对固氮鱼腥藻在单细胞水平上发生的复杂而动态的元素变化过程,本研究综述了土壤固氮鱼腥藻碳氮固存过程,并探讨了利用纳米二次离子质谱法与稳定同位素标记结合(nano-secondary ion mass spectrometry-stable isotopic probing, NanoSIMS-SIP)技术和拉曼光谱成像与稳定同位素标记结合(Raman spectroscopy imaging-stable isotopic probing, Raman-SIP)技术解析单细胞水平上的碳氮元素的时空分布的原理、进展与难点。重点关注了单细胞稳定同位素技术定量可视化固氮鱼腥藻碳氮固存的最新技术发展与应用。同时,对该类可视化技术的未来研究进行了展望。本研究对于理解固氮鱼腥藻在土壤中的碳氮固存机制和固氮效率差异具有重要的科学意义,为农业生产中减少化肥使用、提高土壤肥力提供理论依据。  相似文献

The presence of explosives in soils and the interaction with drought stress and nutrient limitation are among the environmental factors that severely affect plant growth on military soils. In this study, we seek to isolate and identify the cultivable bacteria of a 2,4‐dinitrotoluene (DNT) contaminated soil (DS) and an adjacent grassland soil (GS) of a military training area aiming to isolate new plant growth‐promoting (PGP) and 2,4‐DNT‐degrading strains. Metabolic profiling revealed disturbances in Ecocarbon use in the bare DS; isolation of cultivable strains revealed a lower colony‐forming‐unit count and a less diverse community associated with DS in comparison with GS. New 2,4‐DNT‐tolerant strains were identified by selective enrichments, which were further characterized by auxanography for 2,4‐DNT use, resistance to drought stress, cold, nutrient starvation and PGP features. By selecting multiple beneficial PGP and abiotic stress‐resistant strains, efficient 2,4‐DNT‐degrading consortia were composed. After inoculation, consortium UHasselt Sofie 3 with seven members belonging to Burkholderia, Variovorax, Bacillus, Pseudomonas and Ralstonia species was capable to successfully enhance root length of Arabidopsis under 2,4‐DNT stress. After 9 days, doubling of main root length was observed. Our results indicate that beneficial bacteria inhabiting a disturbed environment have the potential to improve plant growth and alleviate 2,4‐DNT stress.  相似文献

Furazolidone (FZD) has been widely used as an antibacterial and antiprotozoal feed additive for poultry, cattle and farmed fish. Since FZD has been shown to have mutagenic, genotoxic and potentially carcinogenic properties when tested in a variety of systems, there is an increasing need to find a way to remove FZD from contaminated environments. In this report, three bacterial strains Acinetobacter calcoaceticus T32, Pseudomonas putida SP1 and Proteus mirabilis V7 capable of degrading FZD effectively were isolated, identified and characterized. The reduced FZD concentration after degradation was determined by HPLC. After bacterial cells were grown in the media containing 5 mg l−1 FZD for 5 days, almost all FZD was degraded by A. calcoaceticus T32, and more than 50% of FZD was degraded by P. putida SP1 and P. mirabilis V7, respectively. Bacterial GST activity of A. calcoaceticus T32, P. putida SP1 and Pmirabilis V7 was determined to be influenced by different FZD concentrations. Cytotoxicity analysis showed that FZD was degraded to the metabolites with far less cytotoxicity compared to FZD. The inoculation of bacterial strains A. calcoaceticus T32, P. putida SP1 and P. mirabilis V7 into FZD-contained media resulted in a higher degradation efficiency than natural degradation, which indicated the potential application of these strains in treatment of FZD-polluted freshwater or seawater environments.  相似文献

During a screening of phosphate solubilizing bacteria (PSB) in agricultural soils, two strains, IH9 and OCI1, were isolated from the rhizosphere of grasses in Spain, and they showed a high ability to solubilize phosphate in vitro. Inoculation experiments in chickpea and barley were conducted with both strains and the results demonstrated their ability to promote plant growth. The 16S rRNA gene sequences of these strains were nearly identical to each other and to those of Acinetobacter calcoaceticus DSM 30006T, as well as the strain CIP 70.29 representing genomospecies 3. Their phenotypic characteristics also coincided with those of strains forming the A. calcoaceticus–baumannii complex. They differed from A. calcoaceticus in the utilization of l-tartrate as a carbon source and from genomospecies 3 in the use of d-asparagine as a carbon source. The 16S–23S intergenic spacer (ITS) sequences of the two isolates showed nearly 98% identities to those of A. calcoaceticus, confirming that they belong to this phylogenetic group. However, the isolates appeared as a separate branch from the A. calcoaceticus sequences, indicating their molecular separation from other A. calcoaceticus strains. The analysis of three housekeeping genes, recA, rpoD and gyrB, confirmed that IH9 and OCI1 form a distinct lineage within A. calcoaceticus. These results were congruent with those from DNA–DNA hybridization, indicating that strains IH9 and OCI1 constitute a new genomovar for which we propose the name A. calcoaceticus genomovar rhizosphaerae.  相似文献

微生物产生的胞外多糖(exopolysaccharides, EPS)可促进大粒径土壤团聚体形成,高产EPS的菌株在土壤改良、促进作物生长方面具有较好的应用前景。【目的】从土壤样品中筛选高产胞外多糖的细菌,研究其在土壤改良、环境适应性、广谱抗病等方面的功能,为制备土壤改良型功能菌剂提供候选菌株。【方法】采用蒽酮硫酸法测定菌株胞外多糖的产量,通过形态学观察、生理生化试验及16S rRNA基因序列测定确定其分类地位,结合土壤培养试验研究菌株对土壤团聚体形成的影响。【结果】获得3株胞外多糖产量大于500 mg/L的细菌,经鉴定A-5为地衣芽孢杆菌(Bacillus licheniformis),XJ-3为萎缩芽孢杆菌(Bacillus atrophaeus),KW3-10为耐盐芽孢杆菌(Bacillus halotolerans)。菌株A-5、XJ-3、KW3-10处理后,土壤大团聚体(>0.25 mm)含量较对照分别提高了4.07、2.14和3.16倍。3株菌株对疮痂链霉菌(Streptomyces scabies)、尖孢镰刀菌(Fusarium oxysporum)、茄链格孢菌(Alternaria solani)和立枯丝核菌(Rhizoctonia solani)等多种植物病原菌具有明显的抑制效果,可耐受pH为5-9和NaCl含量1%‒9%的盐碱环境,促进植物生长,其中KW3-10的代谢产物中IAA含量为25.58 mg/L。【结论】菌株A-5、XJ-3、KW3-10可显著促进土壤团粒结构形成,具有较好的广谱抗病性和促生长特性,可作为高效复合功能菌剂的候选菌株。  相似文献

Broad-host-range plasmids, belonging to IncP (RP4 and pUPI102) and IncC (R57.b), were studied for intrageneric and intergeneric gene transfer in three different soil microcosms. RP4 was transferred intragenerically in clay loam, sandy loam, and sandy microcosms at frequencies of 0.71×10–2, 0.83×10–2, and 0.41×10–2 respectively, optimally at 37°C and at 100% vol/wt moisture content. Under similar conditions, R57.b was also transferred at frequencies of 0.38×10–2, 0.58×10–2, and 0.80×10–5 respectively at 30°C. Both RP4 and R57.b were transferred at low frequency at 20°C. Kinetics of plasmid transfer revealed that 48 h was the optimum time for intrageneric conjugal gene transfer. Gene transfer frequency was tenfold higher in all nutrient-amended soil microcosms than in the absence of nutrient amendment. RP4 was transferred to an indigenous soil bacteriumBeijerinckia indica in a nonsterile soil microcosm and to other indigenous soil bacteria, viz.Xanthomonas campestris, Azotobacter chroococcum, Acinetobacter calcoaceticus, Achromobacter agili, andRhizobium meliloti in sterile soil microcosms. pUPI102 was transferred fromA. calcoaceticus BD413 toEscherichia coli K12 J53 at a frequency of 0.75×10–6 and 1.1×10–6 in clay loam and sandy loam microcosms respectively. However, no gene transfer was observed in any soil microcosm when strains ofA. calcoaceticus BD413 (pUPI102) andE. coli K12 J53.2 (RP4) were used for conjugal mating. 【目的】固氮菌和氨化细菌是氮循环产生生物有效氮的关键起始环节,直接影响了外来入侵植物的生长速度和扩散进程。然而,关于典型入侵植物薇甘菊根际可培养固氮菌和氨化细菌的研究尚未见报道,这在很大程度上制约了我们对薇甘菊根际高效的氮素转化机制的深刻理解。【方法】采用传统平板涂布培养法对野外采集的薇甘菊根际土壤中的可培养固氮菌和氨化细菌进行了分离鉴定,并进行了接种验证实验。【结果】结果表明,入侵植物薇甘菊根际土壤中的固氮菌和氨化细菌的菌群密度显著高于两个本地伴生植物(火炭母和鸡屎藤),其固氮效率及有机氮矿化效率也优于2个本地种;系统发育分析表明:薇甘菊根际的固氮菌菌株归类于5个属,分别为伯克霍尔德氏菌属(Burkholderia)、肠杆菌属(Enterobacter)、植物杆菌属(Phytobacter)、新肠杆菌属(Kosakonia)和根瘤菌属(Rhizobium);氨化细菌归类于7个属,分别为沙雷氏菌属(Serratia)、不动杆菌属(Acinetobacter)、假单胞菌属(Pseudomonas)、博德特氏菌属(Bordetella)、寡养单胞菌属(Stenotrophomonas)、苍...  相似文献

【背景】细菌生物膜是造成病原菌耐药性增强和持续感染的主要因素,但目前尚无针对抗菌膜的特效药物。特境植物根际微生物可产生大量具有提高宿主免疫功能的活性成分,极具抗生物膜药源开发潜力。【目的】了解滇西北高寒特境白马雪山分布的云南黄芪与灰毛康定黄芪植物根际微生物的物种多样性,并对可培养菌株进行抑菌与抗生物膜活性筛选。【方法】采用宏基因组技术结合传统微生物培养方法,对采自我国云南迪庆藏族自治州德钦县白马雪山的云南黄芪与灰毛康定黄芪的根际微生物进行物种多样性研究,并通过“孔板法”测定其可培养菌株发酵液乙酸乙酯粗浸膏的抗菌、抗生物膜活性。【结果】宏基因组测序结果显示,云南黄芪根际土壤样本中的微生物来自6门7纲8目8科9属10种,其中栖热菌属为优势菌群;灰毛康定黄芪根际土壤样本中的微生物来自6门8纲10目11科14属15种,其中慢生根瘤菌属为优势菌群。通过纯培养共获得145株可培养菌株,包括112株细菌和33株真菌。其中,云南黄芪根际细菌59株,共计16属35种,优势属为假单胞菌属和链霉菌属;根际真菌19株,共计4属5种,优势属为曲霉属;灰毛康定黄芪根际细菌53株,归属于16属29种,优势属为芽孢杆菌属与寡养单胞菌属;根际真菌14株,归属于3属4种,优势属为曲霉属。从不同种水平上选择51株细菌和7株真菌为代表菌株进行抗生素药源评估,发现5株细菌及1株真菌发酵液的乙酸乙酯粗浸膏具有中等至较强的抗革兰阳性菌活性,而且其中4株具有抗MRSA生物膜活性,最终确定了链霉属放线菌Streptomyces fulvissimus KTA1和曲霉属真菌Aspergillus fumigatus YNF5为潜力活性菌株。【结论】首次报道了滇西北地区高寒特境黄芪属植物根际微生物具有较好的物种多样性,而且具有一定的抗生素药用资源开发潜力。本研究对滇西北高寒特境特色植物来源的微生物资源开发利用与保护具有重要的借鉴意义。  相似文献

Aspergillus flavus colonizes agricultural commodities worldwide and contaminates them with carcinogenic aflatoxins. The high genetic diversity of A. flavus populations is largely due to sexual reproduction characterized by the formation of ascospore-bearing ascocarps embedded within sclerotia. A. flavus is heterothallic and laboratory crosses between strains of the opposite mating type produce progeny showing genetic recombination. Sclerotia formed in crops are dispersed onto the soil surface at harvest and are predominantly produced by single strains of one mating type. Less commonly, sclerotia may be fertilized during co-infection of crops with sexually compatible strains. In this study, laboratory and field experiments were performed to examine sexual reproduction in single-strain and fertilized sclerotia following exposure of sclerotia to natural fungal populations in soil. Female and male roles and mitochondrial inheritance in A. flavus were also examined through reciprocal crosses between sclerotia and conidia. Single-strain sclerotia produced ascospores on soil and progeny showed biparental inheritance that included novel alleles originating from fertilization by native soil strains. Sclerotia fertilized in the laboratory and applied to soil before ascocarp formation also produced ascospores with evidence of recombination in progeny, but only known parental alleles were detected. In reciprocal crosses, sclerotia and conidia from both strains functioned as female and male, respectively, indicating A. flavus is hermaphroditic, although the degree of fertility depended upon the parental sources of sclerotia and conidia. All progeny showed maternal inheritance of mitochondria from the sclerotia. Compared to A. flavus populations in crops, soil populations would provide a higher likelihood of exposure of sclerotia to sexually compatible strains and a more diverse source of genetic material for outcrossing.  相似文献

The oxazine dye Nile blue A and its fluorescent oxazone form, Nile red, were used to develop a simple and highly sensitive staining method to detect poly(3-hydroxybutyric acid) and other polyhydroxyalkanoic acids (PHAs) directly in growing bacterial colonies. In contrast to previously described methods, these dyes were directly included in the medium at concentrations of only 0.5 μg/ml, and growth of the cells occurred in the presence of the dyes. This allowed an estimation of the presence of PHAs in viable colonies at any time during the growth experiment and a powerful discrimination between PHA-negative and PHA-positive strains. The presence of Nile red or Nile blue A did not affect growth of the bacteria. This viable-colony staining method was in particular applicable to gram-negative bacteria such as Azotobacter vinelandii, Escherichia coli, Pseudomonas putida, and Ralstonia eutropha. It was less suitable for discriminating between PHA-negative and PHA-positive strains of gram-positive bacteria such as Bacillus megaterium or Rhodococcus ruber, but it could also be used to discriminate between wax-ester- and triacylglycerol-negative and -positive strains of Acinetobacter calcoaceticus or Rhodococcus opacus. The potential of this new method and its application to further investigations of PHA synthases and PHA biosynthesis pathways are discussed. The Flooding Pampa (FP) is the most important area for cattle breeding in Argentina. In this region, persistence and yield of typical forage legumes are strongly limited by soil salinity and alkalinity, which affect around 30% of the total area. Instead, naturalized Lotus tenuis is the main forage legume in this region. Rhizobial strains currently used for inoculating L. tenuis in the FP are exotic or native from non-saline soils of this region, their taxonomic identity being unknown. Assuming that rhizobia native from the most restrictive environments are well adapted to adverse conditions, the use of such isolates could improve the productivity of L. tenuis in the FP. Hence, the goal of this study was to evaluate the symbiotic efficiency of selected L. tenuis rhizobia native from the FP, as compared with strains currently used for field inoculation of this legume. Under non-stressing conditions, the symbiotic performance of native strains of FP exceeded those ones currently used for L. tenuis. Moreover, the symbiotic performance of the native strain ML103 was considerably high under salt stress, compared with strains currently used as inoculants. Analysis of 16S rRNA gene sequencing revealed that unclassified rhizobia currently used for field inoculation of L. tenuis and native strains grouped with the genus Mesorhizobium. As a whole, results obtained demonstrate that soils of the FP are a source of efficient and diverse rhizobia that could be used as a sustainable agronomic tool to formulate inoculants that improve forage yield of L. tenuis in this region.  相似文献

【背景】由互隔交链孢(Alternaria alternata)引起的黑斑病是库尔勒香梨采后贮期的主要病害之一,严重影响果实品质,造成巨大的经济损失。【目的】筛选对梨黑斑病菌具有高拮抗活性的菌株并探究其可能的抑菌机制,为香梨采后保鲜提供理论依据。【方法】从库尔勒香梨园健康植株根际采集土壤样品,采用稀释涂布法分离细菌并进行纯化,通过平板对峙法和琼脂打孔法筛选对梨黑斑病菌具有显著拮抗作用的菌株,结合形态学观察和16S rRNA基因序列分析进行初步鉴定;通过平板对峙法对拮抗菌抑菌谱进行测定;采用特异性平板定性检测拮抗菌产酶活性,采用3,5-二硝基水杨酸比色法法及福林-酚法定量酶活;通过孢子萌发抑制试验和扫描电镜观察,测定拮抗菌对梨黑斑病菌的拮抗作用。【结果】筛选得到2株对梨黑斑病菌有较强抑制活性的菌株JE53和JE56,经分析鉴定为多粘类芽孢杆菌(Paenibacillus polymyxa)及枯草芽孢杆菌(Bacillus subtilis),其发酵上清液对梨黑斑病菌的抑菌圈直径分别为22.49 mm和22.40 mm。抑菌谱测定结果表明,菌株JE53和JE56对马铃薯黑痣病菌等9种植物病原菌均有不同程度的抑制作用。胞外酶测定结果显示这2株菌可产生4种胞外酶;其发酵上清液能够显著抑制病原菌孢子萌发,抑制率分别为70.30%和88.87%;扫描电镜发现这2株菌可导致病原菌菌丝表面粗糙、扭曲变形。【结论】JE53和JE56对梨黑斑病菌有较好的抑制效果,具有进一步开发和应用的潜力。  相似文献

The natural transformation of Acinetobacter calcoaceticus BD413 (trp E27) was characterized with respect to features that might be important for a possible gene transfer by extracellular DNA in natural environments. Transformation of competent cells with chromosomal DNA (marker trp +) occurred in aqueous solutions of single divalent cations. Uptake of DNA into the DNase I-resistant state but not the binding of DNA to cells was strongly stimulated by divalent cations. An increase of transformation of nearly 3 orders of magnitude was obtained as a response to the presence of 0.25 mM Ca2+. With CaCl2 solutions the transformation frequencies approached the highest values obtained under standard broth conditions, followed by MnCl2 and MgCl2. It is concluded that transformation requires divalent cations. DNA competition experiments showed that A. calcoaceticus does not discriminate between homologous and heterologous DNA. Furthermore, circular plasmid DNA competed with chromosomal DNA fragments and vice versa. The equally efficient transformation with plasmid pKT210 isolated from A. calcoaceticus or Escherichia coli indicated absence of DNA restriction in transformation. High efficiency plasmid transformation was obtained in samples of non-sterile natural groundwater and in non-sterile extracts of fresh and air-dried soil. Heat-treatment (10 min, 80°C) of the non-sterile liquid samples increased transformation only in the dried soil extract, probably by inactivation of DNases. The results presented suggest that competent cells of A. calcoaceticus can take up free high molecular weight DNA including plasmids of any source in natural environments such as soil, sediment or groundwater.  相似文献

Inga edulis Mart. is a tropical legume tree used for shade in coffee and cacao plantations and as a hedgerow in alley-cropping practices. Little information can be found concerning N2 fixation in this species. This study was conducted to characterize the rhizobia of I. edulis and determine if it is capable of fixing substantial amounts of N2. Four strains of fast-growing, Gram-negative rhizobia-type bacteria were isolated from I. edulis nodules. The strains were identified by sequencing of partial 16S–23S rDNA internal spacer region. Nitrogenase activity was determined using acetylene reduction assay (ARA). Dinitrogen fixation was measured under controlled conditions by the 15N isotope dilution technique using two non-N2-fixing reference species, Vochysia guatemalensis Donn. Sm, and Gmelina arborea Roxb. ex. Sm. Seedlings were grown in three growth media: native soil and naturally N-depleted sand amended to a low and high N level. The four strains of symbiotic bacteria were closely related to Bradyrhizobium japonicum and to Bradyrhizobium liaoningense. Nodules demonstrated nitrogenase activity as measured by ARA. Vochysia guatemalensis was a better non-N2-fixing reference than G. arborea. When V. guatemalensis was used as the non-N2-fixing reference, the estimate of the percentage of N fixed from atmosphere out of total N in I. edulis seedlings was ca. 40 in the two sand media treatments and 10 in the native soil.  相似文献

Acinetohacter calcoaceticus LMD 79.41 is a unique bacterium containing a soluble quinoprotein D-glucose dehydrogenase (sGDH) in addition to the membrane-bound quinoprotein D-glucose dehydrogenase (mGDH) which is distributed extensively in Gram-negative bacteria. sGDH has been shown to be a distinct enzyme from mGDH, though both enzymes contain a tightly bound pyrroloquinoline quinone (PQQ) as their prosthetic group. In this study, sGDH was detectable in all strains tested of A. calcoaceticus but not in other Gram-negative bacteria tested, indicating that sGDH can be useful as a taxonomic marker for A. calcoaceticus.

The binding process of PQQ with both enzymes was examined by using the apoenzymes purified from a PQQ-deficient mutant strain of A. calcoaceticus. sGDH was able to bind two moles of PQQ in one mole of the homodimer with a fairly high affinity. The binding reaction was much faster at alkaline pH than at acidic pH, and required the presence of some divalent cations such as Cd2+, Ca2+, Sr2+, or Mn2+. On the other hand, mGDH bound one mol of PQQ in the monomeric enzyme with a relatively slow reacting process, which was optimum at acidic pH and in the presence of different types of divalent cations such as Mg2+, Ca2+, Zn2+, or Sr2+. Thus, it is suggested that sGDH and mGDH have distinct structures around their PQQ binding site. Furthermore, binding of PQQ affects the conformation of both enzymes, which can be shown from the diminishing intrinsic fluorescence of the enzymes and the increase in resistance against proteolysis upon PQQ binding. Data also suggest that the conformational changes caused by PQQ-binding are more dramatic in sGDH than in mGDH. Based on the results obtained, the differences in PQQ-binding mode between the enzymes and the physiological meanings of sGDH are discussed.  相似文献

The genus Asaia (family Acetobacteraceae) was first introduced with a single species—Asaia bogorensis and later six more species were described namely A. siamensis, A. krungthepensis, A. lannaensis, A. platycodi, A. prunellae, and A. astilbes. Acetobacteraceae family has been divided into ten genera but, only three of them include nitrogen fixing species: Gluconacetobacter, Acetobacter, and Swaminathania. This article originated from our study primarily aimed to isolate new endosymbiotic nitrogen fixer among Acetobacteraceae during which we have isolated, for the first time in India, four different strains of Asaia sp. from three different sources: Michalia champaca flower, Anopheles mosquito, and ant Tetraponera rufonigra. All the endosymbiotic strains isolated possess the ability to fix nitrogen. Evidence for both nitrogenase activity and the presence of nifH gene in isolated Asaia sp. is presented. Asaia bogorensis (MTCC 4041T) and A. siamensis (MTCC 4042T), two of the validated type strains available from the repository, were tested positive for the presence of functional nitrogenase. The nifH gene sequences from these type strains were also confirmed and compared with other nitrogen fixing members of the family Acetobacteraceae. Our result corroborate with the previous reports that Asaia sp. are indeed widely distributed in nature but this is the first time demonstration of their functional nitrogenase activity. This study shows Asaia sp. as fourth genera of nitrogen fixing bacteria in the family Acetobacteraceae.  相似文献

 The use of genetically engineered crop plants has raised concerns about the transfer of their engineered DNA to indigenous microbes in soil. We have evaluated possible horizontal gene transfer from transgenic plants by natural transformation to the soil bacterium Acinetobacter calcoaceticus BD413. The transformation frequencies with DNA from two sources of transgenic plant DNA and different forms of plasmid DNA with an inserted kanamycin resistance gene, nptII, were measured. Clear effects of homology were seen on transformation frequencies, and no transformants were ever detected after using transgenic plant DNA. This implied a transformation frequency of less than 10-13 (transformants per recipient) under optimised conditions, which is expected to drop even further to a minimum of 10-16 due to soil conditions and a lowered concentration of DNA available to cells. Previous studies have shown that chromosomal DNA released to soil is only available to A. calcoaceticus for limited period of time and that A. calcoaceticus does not maintain detectable competence in soil. Taken together, these results suggest that A. calcoaceticus does not take up non-homologous plant DNA at appreciable frequencies under natural conditions.
