171 mutations conferring resistance to the indole analogue 5-fluoroindole (5 FI) were isolated in the filamentous basidiomycete fungus Coprinus cinereus. 5 FI is thought to be toxic because it is converted intracellularly to 5-fluorotryptophan (5 FT) which feedback inhibits the first enzyme of the tryptophan biosynthetic pathway, anthranilate synthase. Mutations were assigned to five loci, iar-1-iar-5 on the basis of functional analyses and mapping experiments. iar-5 mutations mapped in the anthranilate synthase structural gene and gave rise to an enzyme feedback resistant to tryptophan and its analogue. Mutants at other loci had regulatory changes. iar-1 and iar-3 mutants had elevated levels of two pathway enzymes measured (anthranilate synthase and tryptophan synthase) and were cross resistant to analogues of other aromatic amino acids suggesting that the entire aromatic pathway was derepressed. iar-3 mutants were unable to degrade metabolically derived typtophan to anthranilic acid unlike iar-1 mutants which excreted high levels of anthranilic acid. iar-2 mutants appeared to have a constitutive degradative pathway. iar-4 mutants had a blocked degradative pathway and unusual levels of tryptophan pathway enzymes.Abbreviations 5 FI 5-fluoroindole - 5 FT 5-fluorotryptophan - pFP para-fluorophenylalanine - mFT meta-fluoro-tyrosine  相似文献

Psychological distance to reward, or the segmentation effect, refers to the preference for a terminal link of a concurrent-chains schedule consisting of a simple reinforcement schedule (e.g. fixed interval [FI] 30s) relative to its chained-schedule counterpart (e.g. chained FI 15s FI 15s). This experiment was conducted to examine whether the segmentation effect is due to the number of terminal-link stimulus and response segments per se. Three pigeons pecked under a concurrent-chains schedule in which identical variable-interval (VI) schedules operated in the initial links. In each session, half the terminal-link entries followed one initial-link key and the other half followed the other initial-link key. The initial-link keys correlated with the different terminal links were manipulated across conditions. In the first three conditions, each terminal link contained a chained fixed-time (FT) FT schedule, and in the final three conditions, each terminal link contained a chained FI FI schedule. In each condition, in one terminal link (alternating), the order of two key colors correlated with the different schedule segments alternated across terminal-link entries, whereas in the other terminal link (constant), the order of two other key colors was identical for each entry. With the chained FT FT schedule terminal links, there was indifference between the alternating and constant terminal links within and across pigeons, as indexed by initial-link choice proportions. In addition, terminal-link response rates were relatively low. With the chained FI FI schedule terminal links, for each pigeon, there was relatively more preference for the alternating terminal link and terminal-link response rates increased relative to conditions with the chained FT FT schedule terminal links. These data suggest that the segmentation effect is not due simply to the number of terminal-link stimulus or response segments per se, but rather to a required period of responding during a stimulus segment that never is paired with reinforcement.  相似文献

5-Fluorotryptophan (5FT), indolmycin (IM), 4-fluorotryptophan and 7-azatryptophan were found on screening to be tryptophan antagonists among various chemically synthesized and naturally occurring tryptophan analogues for the isolation of l-tryptophan (l-Trp) producing mutants of Bacillus subtilis K.

From among 5FT resistant mutants, potent l-Trp producers were obtained using an improved isolation medium. Growth of the isolated 5FT-resistant l-Trp producer, AJ 11709, was inhibited by IM. From among 5FT and IM resistant mutants, the best strain, AJ 11979, which produced 9.0 g/liter of l-Trp from 13% glucose on 120hr cultivation, was selected.  相似文献

In two experiments we examined the influence of response and time factors on the speed of acquisition of temporal control on FI schedules. In Experiment 1, prior exposure to FT accelerated the development of temporal control on FI schedules of the same temporal value. It was also found that the slower acquisition on FI with prior RT was similar to that of rats with prior standard training. In Experiment 2, prior exposure to FT accelerated the development of temporal control on a FI schedule with a threefold increase in temporal value. Additionally, it was found that with prior FI 30s training, acquisition of temporal control on FI 90s was even faster than with prior FT 30s. Measures of head-entries into the feeder along the experiments indicated that temporal control was already developed during the periodic but not during the non-periodic histories and that this control transferred to lever press during FI testing phase.  相似文献

The mechanism of killing of A9 fibroblasts by 5-fluorotryptophan has been studied. L-tryptophan competitively relieves the growth inhibition caused by 5FT. After incubation with 5FT, 3H-5FT was incorporated into protein, replacing tryptophan residues. During the initial hours of incubation with 5FT, a specific inhibition was observed of the incorporation of L-tryptophan into protein; later this inhibition was followed by a general inhibition of protein synthesis and cell division. However, nuclear division continued after cell division had ceased. While 5FT was observed to be incorporated into protein after a 1 hour period in MEM + 0.40 mM 5FT in A9, no 3H-5FT was incorporated into protein in a mutant isolated by its resistance to killingy by 5FT. These results support the hypothesis that cell death occurs due to malfunctioning proteins which contain 5FT residues.  相似文献

Experiment I used non-naive pigeons having previously performed on both keypecking and treadlepressing Fixed Interval schedules. In condition IT, treadlepressing was reinforced on successive Fixed Interval 60 seconds, Fixed Time 60 seconds and Fixed Interval 60 seconds schedules. Subsequently (condition IK), the same subjects pecked a key on an identical schedule sequence (FI60, FT60, FI60). In Experiment II, separate groups of naïve subjects were assigned either to treadlepressing (condition IIT) or keypecking (condition IIK) and to the same schedule sequence (FI60, FT60, FI60). Treadle pressing and keypecking decreased greatly in Fixed Time schedules. Curvature indices, pauses and running rates were less sensitive than response rates to the switching from one schedule to the other. Experiments I and II yielded similar results, experimental history accounting only for minor differences. The results were discussed in relation to interspecies differences in the temporal regulation of behavior and operant versus respondent control of the response and schedule-induced behaviour.  相似文献

The present study was undertaken in the natural temperate Himalayan forests of Himachal Pradesh, India, to assess the tree species composition and diversity. For this purpose, six major forest types (FT) viz., FT1- Upper Himalayan Pinus roxburghii forest, FT2- Quercus leucotrichophora forest, FT3- Low-level P. wallichiana forest, FT4- Moist C. deodara forest, FT5- Western Mixed Coniferous Forest, FT6- Pinus gerardiana forest were selected. Detailed sampling was carried out in these forest types, and the sample plots in each forest type were laid out using the stratified random approach. Tree stem density varied from 191.11 N ha?1 (FT6) to 441.11 N ha?1 (FT2), whereas the tree total basal cover varied from 20.01 m2 ha?1 (FT6) to 47.59 m2 ha?1 (FT5). The diversity indices reflected that a total of 21 tree species (16 genera, 11 families) were identified, with tree species richness ranging from 3 (FT6) to 9 (FT1 & FT5). The forest type FT5 recorded the maximum Shannon index of diversity (2.36), Simpson Index of diversity (0.75), Margalef's Index of richness (1.37), pielou equitability (0.74), menheink index of species richness (0.49), whereas the highest Shannon Index of diversity (0.73) and species heterogeneity (0.85) in FT6 forest type. Furthermore, dominance-diversity (d-d) curves drawn that all the six forest types showed geometric curves reveals that one or two tree species are dominant in a particular forest type. Simultaneously, the research area's species diversity, tree stem density, and tree total basal cover were equivalent to those seen in other sections of Western Himalayas.  相似文献

The central complement inhibitor factor I (FI) degrades activated complement factors C4b and C3b in the presence of cofactors such as C4b-binding protein, factor H, complement receptor 1, and membrane cofactor protein. FI is a serine protease composed of two chains. The light chain comprises the serine protease domain, whereas the heavy chain contains several domains; that is, the FI and membrane attack complex domain (FIMAC), CD5, low density lipoprotein receptor 1 (LDLr1) and LDLr2 domains. To understand better how FI acts as a complement inhibitor, we used homology-based models of FI domains to predict potential binding sites. Specific amino acids were then mutated to yield 16 well expressed mutants, which were then purified from media of eukaryotic cells for functional analyses. The Michaelis constant (Km) of all FI mutants toward a small substrate was not altered, whereas some mutants showed increased maximum initial velocity (Vmax). All the mutations in the FIMAC domain affected the ability of FI to degrade C4b and C3b irrespective of the cofactor used, whereas only some mutations in the CD5 and LDLr1/2 domains had a similar effect. These same mutants also showed impaired binding to C3met. In conclusion, the FIMAC domain appears to harbor the main binding sites important for the ability of FI to degrade C4b and C3b.  相似文献

Essential tryptophan residues were specifically modified in soybean lipoxygenase-1 by N-bromosuccinimide (NBS). Both linoleate oxidation and pigment bleaching (β-carotene or chlorophyll a) activities were significantly reduced with the modified enzyme under aerobic conditions. However, the effect on the reduction of linoleate oxidation was more marked. Pigment bleaching under anaerobic conditions was almost completely blocked with the modified enzyme. On the basis of spectral studies it was elucidated that soybean lipoxygenase-1 contains two essential tryptophan residues in its active site.  相似文献

The growth of Bacillus subtilis TR–44, a prototrophic transductant from one of inosine producers, was completely inhibited by 200 µg/ml of 5-fiuorotryptophan, a tryptophan analogue, and the inhibition was reversed by the addition of L-tryptophan.

Several mutants resistant to 5FT* produced L-tryptophan in the growing cultures. The best producer, strain FT–39, which was selected on a medium containing 1500 µg/ml of 5FT, produced 2 g/liter of L-tryptophan, when cultured in a medium containing 8% of glucose but without any tryptophan precursors. In this mutant, anthranilate synthetase, a key enzyme of the tryptophan biosynthesis, had increased over 280-fold, presumably owing to a genetic derepression. From FT–39, mutants resistant to 7000 µg/ml of 5FT were derived. Among them, strain FF–25 produced 4 g/liter of L-tryptophan, twice as much as did the parental strain. Since this strain produced large amount of L-phenylalanine as well as L-tryptophan, the genetic alteration seemed to be involved in some metabolic regulation of common part of the aromatic amino acid biosynthetic pathway.

Further, some auxotrophs derived from these 5FT resistant mutants produced more L-tryptophan than did the parental strains.

Relationships between the accumulation of L-tryptophan and the regulation mechanisms of the L-tryptophan biosynthesis were discussed.  相似文献

Aims It has been well recognized that understory vegetation plays an important role in driving forest ecosystem processes and functioning. In subtropical plantation forests, understory removal and fertilization have been widely applied; however, our understanding on how understory removal affects soil respiration and how the process is regulated by fertilization is limited. Here, we conducted an understory removal experiment combined with fertilization to evaluate the effects of the two forest management practices and their interactions on soil respiration in subtropical forest in southern China.Methods The study was conducted in a split-plot design with fertilization as the whole-plot factor, understory removal as the subplot factor and block as the random factor in subtropical Eucalyptus plantations. In total, there were four treatments: control with unfertilized and intact understory (CK), understory removal but without fertilization (UR), with fertilization but without understory removal (FT) and with fertilization + understory removal (FT + UR). Eucalyptus above- and belowground biomass increment, fine root biomass, soil temperature, soil moisture and soil respiration were measured in the present study. Understory respiration (R U) was quantified in different ways: R u = R CK ? R UR or R u = R FT ? R (FT + UR); fertilization increased soil respiration (R FI) was also quantified in different ways: R FI = R FT ? R CK or R FI = R (FT + UR) ? R UR .Important findings Over a 2-year experiment, our data indicate that understory removal significantly decreased soil respiration, while fertilization increased soil respiration. Understory removal decreased soil respiration by 28.8% under fertilization, but only 15.2% without fertilization. Fertilization significantly increased soil respiration by 23.6% with the presence of understory vegetation, and only increased by 3.7% when understory was removed, indicating that fertilization increased soil respiration mainly by increasing the contribution of the understory. Our study advances our understanding of the interactive effects of understory management and fertilization on soil respiration in subtropical plantations.  相似文献

The regulatory mechanism for l-tryptophan (l-Trp) synthesis was compared between the wild type strain and l-Trp producing mutants of B. subtilis K. In the wild type strain, indolmycin (IM) repressed the synthesis of anthranilate synthetase (AS) more strongly than 5-fluorotryptophan ? (5FT), which repressed AS to the same extent as l-Trp did. 5FT inhibited the activity of AS as strongly as l-Trp did, while IM had no inhibitory effect. In the 5FT resistant strains, the syntheses of AS and tryptophan synthetase (TS-B) were markedly increased by genetic derepression, while AS remained still sensitive to the feedback inhibition by l-Trp. The facts that IM repressed the syntheses of AS and TS-B in the strain which was 5FTr and IMS, and did not repress those in the IM-resistant mutant suggested that IM acts as a co-repressor in a different way from 5FT.  相似文献

High multivitamin gestational diets (HV, 10-fold AIN-93G levels) increase body weight (BW) and food intake (FI) in rat offspring weaned to a recommended multivitamin (RV), but not to a HV diet. We hypothesized that high vitamin A (HA) alone, similar to HV, in post-weaning diets would prevent these effects of the HV maternal diet consistent with gene expression in FI and reward pathways. Male offspring from dams fed HV diets were weaned to a high vitamin A (HA, 10-fold AIN-93G levels), HV or RV diet for 29 weeks. BW, FI, expression of genes involved in regulation of FI and reward and global and gene-specific DNA methylation of pro-opiomelanocortin (POMC) in the hypothalamus were measured. Both HV and HA diets slowed post-weaning weight gain and modified gene expression in offspring compared to offspring fed an RV post-weaning diet. Hypothalamic POMC expression in HA offspring was not different from either HV or RV, and dopamine receptor 1 was 30% (P<.05) higher in HA vs. HV, but not different from RV group. Hippocampal expression of serotonin receptor 1A (40%, P<.01), dopamine receptor 2 (40%, P<.05) and dopamine receptor 5 (70%, P<.0001) was greater in HA vs. RV fed pups and is 40% (P<.01), 50% (P<.05) and 40% (P<.0001) in HA vs. HV pups, respectively. POMC DNA methylation was lower in HA vs. RV offspring (P<.05). We conclude that high vitamin A in post-weaning diets reduces post-weaning weight gain and FI and modifies gene expression in FI and reward pathways.  相似文献

The synthesis of eye pigments has been studied in the seven eye color mutants of the Australian sheep blowfly, Lucilia cuprina. Six appear to be affected primarily in the synthesis of xanthommatin. In wild type, the onset of xanthommatin biosynthesis occurs midway through metamorphosis. Developmental patterns of accumulation of the xanthommatin precursors tryptophan, kynurenine, and 3-hydroxykynurenine have also been established for wild type. By determining the levels of these precursors in late pupae of the mutants, it has been shown that the mutant yellowish accumulates excess tryptophan and the mutant yellow accumulates excess kynurenine. The implications of these results—that yellowish lacks tryptophan oxygenase, thus failing to convert tryptophan to kynurenine, and that yellow lacks kynurenine hydroxylase (blocked in the conversion of kynurenine to 3-hydroxykynurenine)—have been confirmed. This has involved in vitro assays of tryphophan oxygenase and precursor feeding experiments. The precursor accumulation patterns are less clear for the other mutants.  相似文献

Mannitol is a sugar polyol claimed to have health-promoting properties. A mannitol-producing strain of Lactococcus lactis was obtained by disruption of two genes of the phosphoenolpyruvate (PEP)-mannitol phosphotransferase system (PTSMtl). Genes mtlA and mtlF were independently deleted by double-crossover recombination in strain L. lactis FI9630 (a food-grade lactate dehydrogenase-deficient strain derived from MG1363), yielding two mutant (ΔldhΔmtlA and ΔldhΔmtlF) strains. The new strains, FI10091 and FI10089, respectively, do not possess any selection marker and are suitable for use in the food industry. The metabolism of glucose in nongrowing cell suspensions of the mutant strains was characterized by in vivo 13C-nuclear magnetic resonance. The intermediate metabolite, mannitol-1-phosphate, accumulated intracellularly to high levels (up to 76 mM). Mannitol was a major end product, one-third of glucose being converted to this hexitol. The double mutants, in contrast to the parent strain, were unable to utilize mannitol even after glucose depletion, showing that mannitol was taken up exclusively by PEP-PTSMtl. Disruption of this system completely blocked mannitol transport in L. lactis, as intended. In addition to mannitol, approximately equimolar amounts of ethanol, 2,3-butanediol, and lactate were produced. A mixed-acid fermentation (formate, ethanol, and acetate) was also observed during growth under controlled conditions of pH and temperature, but mannitol production was low. The reasons for the alteration in the pattern of end products under nongrowing and growing conditions are discussed, and strategies to improve mannitol production during growth are proposed.  相似文献

A photoaffinity probe for corticotropin (ACTH) receptors was prepared by selective modification of the single tryptophan residue in ACTH. A new photoreactive agent, 2,4-dinitro-5-azidophenylsulfenyl chloride, was synthesized and used for introducing the photoreactive group into ACTH. 2,4-Dinitro-5-azidophenylsulfenyl-Trp9-ACTH (DNAPS-ACTH) was also prepared by thiolysis of 2,4-dinitrophenyl-sulfenyl-Trp9-ACTH to form 2-thiol-Trp9-ACTH and reaction of this with 2,4-dinitro-5-azidofluorobenzene. DNAPS-ACTH was characterized by ultraviolet spectra, peptide mapping, and amino acid analysis. Covalent attachment of DNAPS-ACTH to a pituitary protein fraction FI by photolysis was demonstrated by ultraviolet absorption changes as well as by the use of tritiated DNAPS-ACTH.  相似文献

Heat stress in feedlot cattle is known to reduce their performance. The challenge comes in determining reliable predictors of current and near-future changes in thermal status and performance. A 42-d study, using crossbred (Bos taurus) steers was conducted during summer months (July through August) to identify best environmental determinants of rumen temperature (Trumen) and feed intake (FI) in feedlot cattle with access to shade. A further goal was to define the relationship between Trumen and FI. Shade coverage was approximately 50%, and all animals were provided standard feedlot diets and water ad libitum. Intraruminal telemetric boluses recorded Trumen several times each hour. Ear tags, telemetrically connected to a feed monitoring system, provided FI data using RFID technology. Data loggers recorded ambient conditions in sun and shade, along with black globe temperature. Regression analyses identified daylight black globe and air temperatures in shade, with one hour delays, as the best predictors of Trumen. Prediction of FI was much less reliable. Unexpectedly, Trumen was not superior to ambient variables in predicting FI. Maximum daily temperature humidity index, calculated using BG in sun with a 5-d lag, was the best significant predictor of FI. These results indicate for feedlot cattle that although air temperature alone in the shade may be the best predictor of Trumen in the heat, black globe temperature in the sun may be a better determinant of feed intake over time. Additional studies are needed to verify the delayed FI response which seems unusually long.  相似文献

Two new photoreactive arylsulfenyl chlorides, 2-nitro-4-azidophenylsulfenyl chloride (2,4-NAPS-Cl) and 2-nitro-5-azidophenylsulfenyl chloride (2,5-NAPS-Cl), have been synthesized and used for the selective modification of corticotropin (ACTH). Both reagents reacted rapidly with N-acetyltryptophanamide and ACTH under acidic conditions. The NAPS derivatives of ACTH were purified by partition chromatography and characterized by absorption spectra, amino acid analysis, and peptide mapping. The spectral changes caused by photolysis as well as the kinetics of photolysis are described. Tritiated 2,5-NAPS-ACTH was attached covalently to a pituitary protein fraction FI by photolysis. The photolabeling of FI was blocked in the presence of excess ACTH.  相似文献

The interrelationships of 5-HT receptors and the increased fluid secretion by isolated salivary glands of Calliphora have been studied using pharmacological techniques. Removal of the 5-OH group (tryptamine) or displacement to position 6 (6-HT) results in a decrease in potency but no change in intrinsic activity of the hormone whereas altering the ethyl amine side chain (5-OH tryptophan) results in a decrease in both potency and intrinsic activity. Removal of the 5-OH group and alteration of the side chain (gramine and tryptophan) results in a total loss of activity. Gramine and tryptophan behave as competitive antagonists of 5-HT.Prostaglandin E1 (PGE1) was found to be a non-competitive inhibitor of 5-OH tryptophan and theophylline whereas the response to 5-HT and cyclic AMP was only slightly diminished indicating a ‘receptor reserve’ for 5-HT.Saturating concentrations of gramine and tryptophan potentiate theophylline revealing a ‘threshold’ for the mediation of the response. It is concluded that 5-HT derivatives are capable of producing graded effects on adenyl cyclase both above and below the range of enzyme activity which evokes graded changes in fluid secretion.  相似文献

Abstract— The activity of tryptophan hydroxylase was measured in whole homogenates of midbrain and forebrain areas of the rat brain. A significant elevation of tryptophan hydroxylase in midbrain and forebrain was found within 1 h after injection of corticosterone hemisuccinate Na salt (10mg/kg) into normal rats. A further elevation of tryptophan hydroxylase at 4 h after injection occurred only in the midbrain region. A rapid alteration of tryptophan hydroxylase was also observed following intracistemal injection of a protein synthesis inhibitor, cydoheximide. A significant depression of 50% of normal levels occurred both in midbrain and forebrain regions within 1 h. However. 4 h after injection only the midbrain tryptophan hydroxylase level was depressed, and this depression was 16% of normal levels. This temporal and spatial pattern following cydoheximide injection was not the result of changes in the ability of cydoheximide to inhibit in vivo protein synthesis since [3H]valine incorporation into protein was shown to be equally depressed at both 1 and 5 h in both the midbrain and forebrain. Puromycin blocked [3H]valine incorporation into proteins in the midbrain and forebrain. but only caused a depression of 16% of tryptophan hydroxylase in the midbrain at 4 h. The aminonucleoside derivative of puromycin has no effect on protein synthesis or on tryptophan hydroxylase. Cydoheximide had no effect on tryptophan hydroxylase in vitro. The data suggest that cydoheximide and corticosterone produce an early (1 h) effect on tryptophan hydroxylase unrelated to de novo protein synthesis in regions known to contain perikaryon (midbrain) and axon terminals (forebrain) of 5-HT-containing neurons. The later (4h) effects of these two compounds and puromycin on tryptophan hydroxylase in the perikaryon (midbrain) region of 5-HT-containing neurons probably result from alteration in de novo protein synthesis. The half time of tryptophan hydroxylase in midbrain region is calculated to be 12 h.  相似文献
