A novel endophytic actinomycete, designated strain NEAU-J3T, was isolated from soybean root (Glycine max (L.) Merr) and characterized using a polyphasic approach. Phylogenetic analysis based on 16S rRNA gene sequences suggested that strain NEAU-J3T fell within the family Micromonosporaceae. The strain was observed to form an extensively branched substrate mycelium, which carried non-motile oval spores with a smooth surface. The cell walls of strain NEAU-J3T were determined to contain meso-diaminopimelic acid and galactose, ribose and glucose were detected as whole-cell sugars. The major menaquinones were determined to be MK-9(H4) and MK-9(H6). The phospholipids detected were phosphatidylcholine and phosphatidylethanolamine. The major cellular fatty acids were determined to be C16:0, C18:1 ω9c, C18:0, C17:0, C17:1 ω7c, anteiso-C17:0, C16:1 ω7c and C15:0. The DNA G + C content was 62.5 mol%. On the basis of the morphological and chemotaxonomic characteristics, phylogenetic analysis and characteristic patterns of 16S rRNA gene signature nucleotides, strain NEAU-J3T is considered to represent a novel species of a new genus within the family Micromonosporaceae, for which the name Wangella harbinensis gen. nov., sp. nov. is proposed. An alkalitolerant, thermotolerant, strictly aerobic and Gram-staining negative bacterial strain, designated YIM 78140T, was isolated from a water sample in Hehua hot spring, Tengchong, Yunnan province, south-west China. The colonies were light brown, convex and circular. Phylogenetic analysis of the 16S rRNA gene sequence of strain YIM 78140T indicated that it was clustered with members of β-Proteobacteria (with the similarity from 96.9 to 93.6 %). Good growth occurred at 40–50 °C, pH 8.0–9.0 and in the presence of 0–3 % (w/v) NaCl. The predominant ubiquinones were Q-8 and Q-9. The major fatty acids were C16:0, C17:0 cyclo, C18:1 ω7c and summed feature 3. The G+C content of genomic DNA was 70.8 mol%. The results of physiological and biochemical characteristics, phylogenetic analysis allowed the phenotypic and genotypic differentiation of strain YIM 78140T from its closest phylogenetic neighbours. Therefore, the strain YIM 78140T represents a novel genus of the family Comamonadaceae, for which the name Zhizhongheella caldifontis gen. nov., sp. nov. is proposed. A Gram-negative, non-spore-forming, catalase- and oxidase-positive, aerobic, curved-rod shaped bacterium with polar or subpolar flagellum, designated strain JYr2T, was isolated from a sediment sample collected from an amphioxus breeding zone in the coastal region of Qingdao, China. The organism grew optimally at 37 °C, pH 8–9 and in the presence of 3 % (w/v) NaCl or 3–4 % sea salts (w/v; Sigma). Salt was required for growth. The strain contained isoprenoid quinone 8 (Q-8, 100 %) as the predominant isoprenoid quinone and C16:0 (24.1 %) and C16:1 ω7c and/or C16:1 ω6c (35.8 %) as major fatty acids. Phosphatidylethanolamine, phosphatidylglycerol, aminophospholipid (PN) and two aminolipid (AL1, AL2) were the major constituents of the phospholipids. A phylogenetic analysis based on 16S rRNA gene sequences indicated that strain JYr2T formed a distinct evolutionary lineage within the family Colwelliaceae. It showed <95.0 % sequence similarities to all validly published species in the family Colwelliaceae, except 95.2 % to Thalassomonas viridans DSM 13754T and 95.0 % to Colwellia polaris JCM 537T. The G+C content of the DNA was 42.7 mol%. On the basis of the polyphasic taxonomic study, strain JYr2T (= CGMCC 1.10794T = JCM 17549T) was considered to represent a novel genus and species in Gammaproteobacteria, for which the name Litorilituus sediminis gen. nov. sp. nov. was proposed.  相似文献

A Gram-negative, aerobic, motile rod strain, designated Ma-20T, was isolated from a pool of marine Spirulina platensis cultivation, Sanya, China, and was subjected to a polyphasic taxonomy study. Strain Ma-20T can grow in the presence of 0.5–11 % (w/v) NaCl, 10–43 °C and pH 6–10, and grew optimally at 30 °C, pH 7.5–9.0 in natural seawater medium. The polar lipids were composed of phosphatidylethanolamine, three unidentified phospholipids and three unidentified polar lipids. The respiratory quinone was ubiquinone 8 (Q-8) and the major fatty acids were C18:1ω6c/C18:1ω7c (summed feature 8, 32.84 %), C16:1ω6c/C16:1ω7c (summed feature 3, 30.76 %), C16:0 (13.54 %), C12:03-OH (4.63 %), and C12:0 (4.09 %). The DNA G+C content of strain Ma-20T was 58 mol %. Phylogenetic analyses based on 16S rRNA gene sequences showed that strain Ma-20T belonging to Gammaproteobacteria, it shared 88.46–91.55 and 89.21–91.26 % 16S rRNA gene sequence similarity to the type strains in genus Hahella and Marinobacter, respectively. In addition to the large 16S rRNA gene sequence difference, Ma-20T can also be distinguished from the reference type strains Hahella ganghwensis FR1050T and Marinobacter hydrocarbonoclasticus sp. 17T by several phenotypic characteristics and chemotaxonomic properties. On the basis of phenotypic, chemotaxonomic and phylogenetic properties, strain Ma-20T is suggested to represent a novel species of a new genus in Gammaproteobacteria, for which the name Nonhongiella spirulinensis gen. nov., sp. nov. is proposed. A novel, Gram-negative, facultative anaerobic, motile and short rod-shaped bacterium, strain KBL006T was isolated from the larval gut of Hermetia illucens, Black soldier fly. The 16S rRNA gene sequence of strain KBL006T showed 96.4 % similarity to that of Wohlfahrtiimonas chitiniclastica S5T. Strain KBL006T grew optimally at 30 °C, at pH 8.0 and in the presence of 1–2 % (w/v) NaCl. Oxidase activity and catalase activity were positive. The major fatty acids were C18:1 ω7c, C14:0, and C16:0. The major respiratory quinone was ubiquinone-8 (Q-8). The major polar lipids were phosphatidylethanolamine, phosphatidylglycerol, and diphosphatidylglycerol, and two phospholipids. The G+C content of the genomic DNA was 45.2 mol%. Based on these polyphasic data, strain KBL006T is considered to represent a novel species in the genus Wohlfahrtiimonas, for which the name Wohlfahrtiimonas larvae sp. nov. is proposed. The taxonomic status of Rhodococcus equi, originally isolated from foal specimens, has been the subject of discussion for a number of years. The chequered history of the taxon has prompted this polyphasic analysis of R. equi strains, close members of the genus Rhodococcus and representatives of other genera classified in the order Corynebacteriales, to establish the taxonomic position of this taxon. Thirty one R. equi strains, including the type strain, were examined for genotypic and numerical taxonomic properties. The resultant data are consistent with their classification in the order Corynebacteriales but the R. equi strains formed a distinct phyletic clade away from representatives of other members of the genus Rhodococcus in the 16S rRNA gene tree. Representatives of this clade shared their highest pairwise 16S rRNA gene sequence similarities with the type strain of Rhodococcus kunmingensis (95.2–98.1 %). However, the R. equi taxon was readily distinguished from R. kunmingensis and from the other members of the order Corynebacteriales using a combination of genotypic, chemotypic and phenotypic properties. On the basis of these data the R. equi strains are considered to represent a new genus. The name proposed for this taxon is Prescottia gen. nov., with Prescottia equi comb. nov. as the type species containing the type strain, C 7T (= ATCC 25729T = ATCC 6939T = CCUG 892T = CIP 54.72T = DSM 20307T = HAMBI 2061T = NBRC 14956T = JCM 1311T = JCM 3209T = LMG 18452T = NBRC 101255T = NCTC 1621T = NRRL B-16538T = VKM Ac-953T).  相似文献

A polyphasic taxonomic study using morphological, biochemical, chemotaxonomic and molecular genetic methods was performed on six strains of unknown Gram-positive, nonspore-forming, facultative anaerobic coccus-shaped bacteria isolated from a swine-manure storage pit. On the basis of the 16S rRNA, RNA polymerase α-subunit (rpoA) and 60 kDa chaperonin (cpn60) gene sequence analyses, it was shown that all the isolates were enterococci but formed two separate lines of descent. Pairwise 16S rRNA gene sequence comparisons demonstrated that the two novel organisms were most closely related to each other (97.9 %) and to Enterococcus aquimarinus (97.8 %). Both organisms contained major amounts of C16:0, C16:1 ω7c, C16:1 ω7c, and C18:1 ω7c/12t/9t as the major cellular fatty acids. Based on biochemical, chemotaxonomic and phylogenetic evidence, the names Enterococcus lemanii sp. nov. (type strain PC32T = CCUG 61260T = NRRL B-59661T) and Enterococcus eurekensis sp. nov. (type strain PC4BT = CCUG 61259T = NRRL B-59662T) are proposed for these hitherto undescribed species.  相似文献

An aerobic, Gram-negative, coccoid to short rod-shaped and non-flagellated marine bacterial strain S354T was isolated from seawater of Micronesia. The strain was capable to degrade agar-forming slight depression into agar plate. Growth occurred at a temperature range of 12–44 °C, a pH range of 5–9, and a salinity range of 1–7 % (w/v) NaCl. Phylogenetic analyses based on 16S rRNA gene sequences suggested that S354T belongs to the family Flammeovirgaceae. The novel strain was most closely related to Limibacter armeniacum YM 11-185T with similarity of 92.5 %. The DNA G+C content was 43.8 mol%. The major fatty acids (>10 %) were iso-C15:0 and C16:1 ω5c. The predominant isoprenoid quinone was determined to be MK-7. Polar lipid profile of S354T consisted of phosphatidylethanolamine, unknown polar lipid, and unknown glycolipids. Based on the phenotypic, phylogenetic, biochemical, and physiological tests conducted in this study, S354T is proposed to represent a type strain of a novel genus and species. The 16S rRNA gene sequence of S354T is registered in GenBank under the accession number JQ639084. The type of strain Algivirga pacifica gen. nov., sp. nov. is S354T (=KCCM 90107T=JCM 18326T).  相似文献

Two novel aerobic p-n-nonylphenol-degrading bacterial strains were isolated from seawater obtained from the coastal region of Ogasawara Islands, Japan. The 16S rRNA gene sequence analysis indicated that the strains are affiliated with the order Alteromonadales within the class Gammaproteobacteria. One isolate, strain KU41G2, is most closely related to Maricurvus nonylphenolicus (99.2 % similarity), and is tentatively identified as M. nonylphenolicus. The other isolate, strain KU41GT, is also most closely related to M. nonylphenolicus; however, the 16S rRNA gene sequence similarity was only 94.7 %. Cells of strain KU41GT are Gram-negative rods with a single polar flagellum. The predominant respiratory lipoquinone was ubiquinone-8, and the major cellular fatty acids were C17:1 ω8c (24.2 %); C15:0 iso 2-OH; and/or C16:1 ω7c (16.3 %), C15:0 (10.3 %), C11:0 3-OH (9.5 %), C9:0 3-OH (6.7 %), C10:0 3-OH (6.4 %), and C18:1 ω7c (5.5 %). The DNA G+C content was 53.3 mol%. On the basis of physiological, chemotaxonomic, and phylogenetic data, strain KU41GT is suggested to represent a novel species of a new genus, for which we propose the name Pseudomaricurvus alkylphenolicus gen. nov., sp. nov. A Gram-negative, strictly aerobic, motile, short rod-shaped bacterium with peritrichous flagella, designated JYr12T, was isolated from sediment of an amphioxus breeding zone in the coastal region of Qingdao, China. Strain JYr12T was found to grow optimally at 28 °C, at pH 7.0 and in the presence of 2–3 % (w/v) NaCl. The organism was determined to contain C16:0 (28.3 %) and C16:1 ω7c and/or C16:1 ω6c (34.8 %) as the major fatty acids and ubiquinone-8 (90 %) as the predominant isoprenoid ubiquinone. The polar lipids were found to consist of phosphatidylglycerol, phosphatidylethanolamine, diphosphatidylglycerol and an unknown glycolipid. The G+C content of the genomic DNA was determined to be 42.3 mol %. Comparative 16S rRNA gene sequence analysis revealed that strain JYr12T was placed in the class Gammaproteobacteria and shared ~91.20, 91.18, 91.01, 90.95, 90.80 and 90.40 % sequence similarities with representatives of the genus Thalassolituus, Nitrincola, Oceanospirillum, Marinobacterium, Marinomonas and Microbulbifer, respectively. On the basis of phenotypic, chemotaxonomic and genotypic data, strain JYr12T is considered to represent a novel species in a new genus within the Gammaproteobacteria, for which the name Litoribacillus peritrichatus gen. nov., sp. nov. is proposed. A strain designated as S85T was isolated from a seaweed collected from coastal area of Chuuk State in Micronesia. The strain was gram-negative, rod-shaped, and non-motile and formed yellow colonies on the SWY agar (0.2 % yeast extract and 1.5 % agar in seawater) and Marine agar 2216. The strain grew at pH 5–9 (optimum, pH 8), at 15–40 °C (optimum, 25–28 °C), and with 1–9 % (w/v) NaCl (optimum, 3 %). The phylogenetic analysis based on 16S rRNA gene sequence showed that strain S85T was related to Lutibacter litoralis CL-TF09T and Maritimimonas rapanae A31T with 91.4 % and with 90.5 % similarity, respectively. The dominant fatty acids were iso-C15:0, iso-C15:0 3-OH and iso-C17:0 3-OH, C16:0 3-OH and summed feature 3 (C16:1 ω7c and/or iso-C15:0 2-OH). The major isoprenoid quinone was MK-6. The DNA G+C content of the type strain was 34.6 mol %. The major polar lipids were phosphatidylethanolamine, an unknown glycolipid and two unknown polar lipids. Based on this polyphasic taxonomic data, strain S85T stands for a novel species of a new genus, and we propose the name Ochrovirga pacifica gen. nov., sp. nov. The type strain of O. pacifica is S85T (=KCCM 90106 =JCM 18327T).  相似文献

A Gram-negative, aerobic, non-flagellated, non-gliding and rod-shaped bacterial strain, designated M-M16T, was isolated from seashore sand around a seaweed farm on the South Sea, South Korea, and its taxonomic position was investigated by using a polyphasic study. Strain M-M16T grew optimally at 30 °C, at pH 7.0–8.0 and in the presence of 2 % (w/v) NaCl. Strain M-M16T exhibited the highest 16S rRNA gene sequence similarity values to the type strains of Gaetbulibacter lutimaris (96.5 %) and Flaviramulus basaltis (95.8 %). Neighbour-joining and maximum-parsimony phylogenetic trees based on 16S rRNA gene sequences revealed that strain M-M16T clustered with the type strains of Gaetbulibacter species and F. basaltis. Strain M-M16T contained MK-6 as the predominant menaquinone and iso-C15:1 G, iso-C15:0 and iso-C17:0 3-OH as the major fatty acids. The major polar lipids detected in strain M-M16T were phosphatidylethanolamine and one unidentified lipid. The DNA G+C content of strain M-M16T was 37.4 mol%. The phylogenetic and chemotaxonomic data and other phenotypic properties revealed that strain M-M16T represents a novel genus and species within the family Flavobacteriaceae, for which the name Sabulilitoribacter multivorans gen. nov., sp. nov. is proposed. A novel marine, Gram-negative, rod-shaped bacterium, designated strain N9T, was isolated from a water sample of the sea shore at Visakhapatnam, Andhra Pradesh (India). Strain N9T was found to be positive for oxidase and catalase activities. The fatty acids were found to be dominated by C16:0, C18:1 ω7c and summed in feature 3 (C16:1 ω7c and/or C16:1 ω6c). Strain N9T was determined to contain Q-10 as the major respiratory quinone and phosphatidylethanolamine, phosphatidylglycerol, two aminophospholipids, two phospholipids and four unidentified lipids as polar lipids. The DNA G+C content of the strain N9T was found to be 63 mol%. 16S rRNA gene sequence analysis indicated that Rhodobacter sphaeroides, Rhodobacter johrii, Pseudorhodobacter ferrugineus, Rhodobacter azotoformans, Rhodobacter ovatus and Pseudorhodobacter aquimaris were the nearest phylogenetic neighbours, with pair-wise sequence similarities of 95.43, 95.36, 94.24, 95.31, 95.60 and 94.74 %, respectively. Phylogenetic analysis showed that strain N9T formed a distinct branch within the family Rhodobacteraceae and clustered with the clade comprising species of the genus Pseudorhodobacter, together with species of the genera Roseicitreum, Roseinatronobacter, Roseibaca and Rhodobaca. Species of the genus Pseudorhodobacter are phylogenetically close with a 16S rRNA gene sequence dissimilarity of 5.9–7.3 % (92.7–94.1 % similarity). Based on the above-mentioned phenotypic characteristics and on phylogenetic inference, strain N9T is proposed as a representative of a new genus and a novel species of the family Rhodobacteraceae as Albirhodobacter marinus gen. nov., sp. nov. A novel Gram-negative, motile, rod-shaped, facultative anaerobic bacterial strain, KMK6T, was isolated from soil contaminated with textile dyes from an industrial estate located at Ichalkaranji, Maharashtra, India, and its taxonomical position was established by using a polyphasic approach. The major cellular fatty acids included C17:1ω8c, summed feature 3 (C16:1ω7c and/or iso-C15:0 2-OH), C17:0, C16:0, and C18:1ω7c. The DNA G+C content of strain KMK6T was 48.8 mol %. 16S rRNA gene sequence analysis confirmed its placement in the genus Alishewanella, and exhibited sequence similarity levels of below 97 % to the type strains of validly published Alishewanella species. On the basis of genotypic and phenotypic evidence, strains KMK6T is considered to be a novel species of the genus Alishewanella, for which we propose that strain KMK6T (=NCIM 5295T =BCRC 17848T) is assigned to a novel species, Alishewanella solinquinati sp. nov.  相似文献

Optimal growth of strain YIM 78007 occurs at 40–50 °C and pH 8.0–10.0. The predominant ubiquinone was identified as Q-10 and the major fatty acids were identified as C18:1 ω7c and C16:0. The polar lipids were identified as diphosphatidylglycerol, phosphatidylethanolamine, phosphatidylcholine, two unidentified aminolipids and one unknown phospholipid. The G + C content of the genomic DNA was determined to be 63 mol %. The levels of DNA–DNA hybridization relatedness between strain YIM 78007T and its closet neighbours (Roseomonas lacus JCM 13283T and Roseomonas terrae JCM 14592T) were well below the threshold required for the proposal of a novel species. The results of physiological and biochemical characteristics, the phylogenetic analysis, as well as low DNA–DNA hybridization values, allowed the phenotypic and genotypic differentiation of strain YIM 78007T from its closest phylogenetic neighbours. Therefore, strain YIM 78007T is considered to represent a novel species of the genus Roseomonas, for which the name Roseomonas alkaliterrae sp. nov. is proposed. A taxonomic study was carried out on strain 22II-S10r2T, which was isolated from the deep sea sediment of the Atlantic Ocean using oil-degrading enrichment. The bacterium was Gram-negative, oxidase positive and catalase negative, spherical in shape, and motile by polar flagella. Growth was observed at salinities of 0.5–7 % and at temperatures of 10–41 °C. The isolate was capable of aesculin hydrolysis, but unable to reduce nitrate to nitrite or degrade Tween 80 or gelatine. Phylogenetic analysis based on 16S rRNA gene sequences indicated that strain 22II-S10r2T belonged to the family Ectothiorhodospiraceae, with highest sequence similarity to Thioalkalivibrio sulfidiphilus HL-EbGR7T (90.9 % similarity). The principal fatty acids were Sum In Feature 8 (C18:1 ω7c/ω6c (29.9 %), C18:1 ω9c (13.5 %), C16:1 ω5c (12.3 %), C12:03OH (6.8 %), C18:1 ω5c (5.7 %) and C16:0 (5.3 %). The G+C content of the chromosomal DNA was 60.7 mol%. The respiratory quinone was determined to be Q-7 (25 %) and Q-8 (75 %). Phosphatidylethanolamine, phosphatidylglycerol, aminophospholipid, glycolipid, three phospholipids and lipid were present. The strain was aerobic, non-phototrophic and non-chemolithoautotrophic. The combined genotypic and phenotypic data show that strain 22II-S10r2T represents a novel species within a novel genus, for which the name Maricoccus atlantica gen. nov. sp. nov. is proposed, with the type strain 22II-S10r2T (=CGMCC NO.1.12317T = LMG 27155T = MCCC 1A09384T).  相似文献

A novel bacterial strain designated GJW-30T was isolated from soil of the lava forest, Gotjawal, located in Aewol, Jeju, Korea. Strain GJW-30T was found to be strictly aerobic, Gram-negative and to form pleomorphic, non-motile rods and white colonies on R2A agar. The major fatty acids were identified as C18:1ω7c, C16:0 and C17:0, the predominant isoprenoid quinone as Q-10, the polar lipids as diphosphatidylglycerol, phosphatidylglycerol, phosphatidylethanolamine, phosphatidylcholine, an unidentified aminolipid and an unidentified lipid. The cell-wall sugar pattern of strain GJW-30T was found to be composed of glucose, ribose and rhamnose and meso-DAP as the diagnostic diamino acid in the cell-wall peptidoglycan. The DNA G+C content of strain GJW-30T is 62.2 mol%. Phylogenetic analysis, based on 16S rRNA gene sequence similarities, showed that strain GJW-30T forms a deep branch within the order Rhizobiales, sharing the highest level of sequence homology with Bradyrhizobium oligotrophicum LMG 10732T (93.6 %). On the basis of the phenotypic, chemotaxonomic and phylogenetic characteristics, strain GJW-30T is considered to represent a novel genus and species, for which the name Variibacter gotjawalensis gen. nov., sp. nov. (the type strain is GJW-30T = KCTC 32391T = CECT 8514T = LMG 28093T) is proposed.  相似文献

Strain Gsoil 348T was isolated from a ginseng field soil sample by selecting micro-colonies from one-fifth strength modified R2A agar medium after a long incubation period. 16S rRNA gene sequence analysis indicated that the strain is related to members of the phylum Armatimonadetes (formerly called candidate phylum OP10). Strain Gsoil 348T is mesophilic, strictly aerobic, non-motile and rod-shaped. It only grows in low nutrient media. The major respiratory quinones are menaquinones MK-11 and MK-10, and the main fatty acids are iso-C15:0, iso-C17:0, C16:0 and C16:1 ω11c. The G+C content is 61.4 mol%. The 16S rRNA gene sequences in public databases belonging to the phylum Armatimonadetes were clustered here into 6 groups. Five of these groups constituted a coherent cluster distinct from the sequences of other phyla in phylogenetic trees that were constructed using multiple-outgroup sequences from 49 different phyla. On the basis of polyphasic taxonomic analyses, it is proposed that strain Gsoil 348T (= KACC 14959T = JCM 17079T) should be placed in Fimbriimonas ginsengisoli gen. nov., sp. nov., as the cultured representative of the Fimbriimonadia class. nov., corresponding with Group 4 of the phylum Armatimonadetes.  相似文献

A new aerobic, Gram-negative, chemo-organotrophic, euryhaline bacterium, designated strain JL2009T, was isolated from surface water of the Caribbean Sea. The strain formed flaxen colonies on marine ager 2216 (MA; Difco) medium. Cells were dimorphic, with stalks or a polar flagellum, and reproduction occurred by means of binary fission. Growth occurred at 15–45 °C (optimum at 35 °C), 0–15 % (w/v) NaCl (optimum at 1–10 %) and pH 5–9 (optimum at pH 7–8). Phylogenetic analysis based on 16S rRNA gene sequences showed that the strain formed a distinct evolutionary lineage within the family Hyphomonadaceae. Strain JL2009T was most closely related to Maricaulis parjimensis MCS 25T (92.2 % DNA sequence similarity), Woodsholea maritime CM2243T (90.9 %), and Oceanicaulis alexandrii C116-18T (90.9 %). The major respiratory quinone was Q-10. The dominant cellular fatty acids were summed feature 8 (C18:1 ω7c), C18:0 and 11-methyl C18:1 ω7c. The polar lipid pattern indicated the presence of phospholipid, phosphatidyl glycerol and glycolipids. The G + C content of the genomic DNA was 70.5 mol%. On the basis of the chemotaxonomic and phenotypic characteristics and the phylogenetic evidence, strain JL2009T represents a novel genus and species in the family Hyphomonadaceae, for which the name Euryhalocaulis caribicus gen. nov., sp. nov. is proposed.
