The humoral immune response of Balb/c mice to live infective larvae or adult worm extract of Dipetalonema viteae is composed of two antibody populations either with or without specificity for phosphorylcholine. Absorption of immune serum on phosphorylcholine-Sepharose and separation of the antibody population demonstrated that anti-larvae serum contains a larger ratio of phosphorylcholine versus non-phosphorylcholine antibodies as compared to anti-adult serum. Immunofluorescence on crossections of female worms revealed that antigen expressing phosphorylcholine determinants were mainly found on certain internal structures, like egg, uterine, and intestinal membranes, but not on the cuticle. Immunoblotting using an adult worm extract demonstrated that protein bands reacted with either one or both populations of antibodies. The patterns were heterogeneous and moreover differed between the anti-larvae serum and the anti-adult serum.  相似文献

The jird, Meriones unguiculatus, infected with 80 normal infective larvae of Dipetalonema viteae, revealed a recovery rate of 27.9% 12 weeks after infection. A pretreatment by three injections of 50 normal larvae each and challenge by 80 larvae resulted in a recovery rate of 10.7%. The recovered worms were longer than those from the challenge control animals. When three times 50 irradiated larvae (35 krad) were inoculated, the recovery rate of the challenge decreased to 2.6%, representing a protection of 90.7%. The surviving adult worms were stunted and derived exclusively from the 80 normal larvae given for challenge, since absolutely no adult worms were recovered in eight animals inoculated three times with 50 irradiated larvae only. Sera of all pretreated jirds contained IgG and IgM antibodies which bound in immunoblotting experiments bound predominantly to three proteins of larvae with molecular masses of 68,140, and 165 kDa, respectively. Enzymatic surface iodination revealed that the three antigens were exposed on the larval surface. The coincidence of a partial resistance to a challenge infection and of an antibody response against surface proteins of infective larvae suggests an importance of these antigens for the rejection of D. viteae mediated by an acquired immunological resistance of M. unguiculatus.  相似文献

The effect of the gas phase on the in vitro growth and development of Dipetalonema viteae (Nematoda: Filarioidea) third-stage larvae obtained from the tick vector and 3 day infections of jirds was examined. Measurements of the oxygen (pO2) and carbon dioxide (pCO2) tensions and the pH in the medium were made for each gas phase. In cultures gassed with 5% carbon dioxide in nitrogen the pO2 was between 32 and 50 mm Hg, the pCO2 ranged from 25 to 40 mm Hg and the pH was between 7.2 and 7.4. This gas phase resulted in the best growth and development of third-stage larvae to the fourth-stage. Survival and development of larvae were decreased in cultures with oxygen tensions less than 20 mm Hg and greater than 50 mm Hg.  相似文献

The egg shell of Dipetalonema viteae separated from the oolemma and became highly convoluted at an early stage of development. No second oolemma or trilaminate membrane was seen. Channels containing electron dense material (thought to be nutrient material from the uterine wall) were formed between adjacent embryos. Many developing embryos died. Microvilli were formed by the uterine wall and developing embryos were closely apposed to these (again presumably to obtain nutrient). Embryos emerged from the egg in the uterus and were born as unsheathed microfilariae.  相似文献

Infective third-stage larvae of Strongyloides stercoralis were frozen over liquid nitrogen and remained infective to dogs when thawed. Successful cryopreservation depended on a 30-60-min incubation in a cryoprotectant (10% DMSO and 10% dextran) before freezing and thawing the frozen larvae into RPMI. First-stage larvae could also be frozen by this method. Thawed first-stage larvae remained viable and continued their development to third-stage larvae, which were shown to be infective to dogs.  相似文献

1. A body wall preparation is described which permits intracellular recording from the somatic muscle cells of the small filarial nematode, Dipetalonema viteae. Using this preparation, resting membrane potentials were measured and spontaneous muscle depolarizations described. 2. Stimulatory effects noted upon the addition of acetylcholine, or the cholinergic agonists suggest the hypothesis that acetylcholine is the excitatory neurotransmitter. However, in contrast with vertebrate tissues, the cholinergic antagonists, d-tubocurarine, hexamethonium and pentolinium do not inhibit somatic muscle activity of the worm. 3. GABA inhibited somatic muscle depolarizations, suggesting the possibility that it may serve as an inhibitory neurotransmitter. 4. The anthelmintic drug, levamisole, produced a depolarizing block. Effects of other pharmacological agents are described, discussed and compared with effects on vertebrate muscles.  相似文献

Third-stage larvae of Dipetalonema viteae obtained from the tick vector developed to the fourth stage in several cell-free culture systems. Survival and development of larvae in a number of commercially available cell culture media, supplemented with serum and other defined and undefined components, were compared. All cultures were gassed with 5% carbon dioxide in nitrogen. Best survival, growth and development were obtained in stationary cultures containing 1:1 (v/v) mixtures of NCTC 135, either RPMI 1640 or Iscove's Modified Dulbecco's Medium, and a supplement of 20% non-heat-inactivated fetal bovine serum. The importance of the medium composition and physical environment of the culture system, for the survival, growth and development of D. viteae was demonstrated.  相似文献

Hamsters were given primary infections of 100, 200, and 300 D. viteae larvae and groups killed at various intervals after infection. In addition, hamsters were sequentially infected with 100, 200, and 300 larvae and groups killed at 100 or 75 days after the secondary and tertiary infection, respectively. Blood microfilariae were detected on Day 60 following a primary infection, reached a maximum on Day 75, declined to low levels by Day 105, and were negative on Day 120. No microfilariae reappeared in the blood of hamsters given secondary or tertiary infections.Between 20–30% of the infecting larval dose had reached the adult stage by Days 75 or 100 postinfection in hamsters given primary, secondary, or tertiary infections. There was no evidence of arrested larval development in hamsters receiving a second or third challenge infection. Almost half of the tertiary infection hamsters developed subcutaneous nodules and their numbers varied greatly among individual animals. The nodules variously contained living worms, pus, and fragmented worms, or pus only. Hamsters given primary infections of 100, 200, or 300 larvae and killed 375 days after infection had no subcutaneous nodules; however, hamsters given the 200 and 300 larval infections were seen to have dead worms in the subcutaneous tissues. No stunting of adult worms was noted and all female worms had uteri packed with microfilariae.  相似文献

Following chronic retinol (vitamin A) deprivation leading to exhaustion of liver vitamin A reserves below 50 I.U. per liver hamsters were fed diets either deficient in ("Rd":250 I.U.A./kg in experiment I, 1000 I.U.A/kg in experiment II) or enriched with retinol ("Rw":10000 I.U.A/kg in experiment I and II). After 4 weeks some of the animals (36 in experiment I, 30 in II) were infected with 150 3rd-stage larvae of D. viteae, while clean animals were kept as controls. The retinol status, the immune response (indirect fluorescent antibody test: IFAT) and parasitological parameters were examined up to 8 (experiment I) and 12 weeks (experiment II) post infection (p.i.). Rd hamsters had levelling off of weight gain or weight loss, severely deficient retinol levels in serum and liver, and high mortality. Weight gain was less in infected than in uninfected hamsters, and the capacity of infected Rw animals to restore liver retinol was significantly lower than that of uninfected Rw animals. IFAT titres were similar in Rd and in Rw animals, but microfilaraemia was significantly enhanced at 8 and 10.5 weeks p.i. in Rd hamsters. While the number of worms recovered from Rd and Rw hamsters was similar, there was a significant increase in the ratio of female to male worms in Rd hamsters. Rd hamsters in experiment I produced 3.3 times the worm mass per 100 g body-weight than Rw hamsters. Also, the average mass per female worm was significantly higher in Rd than Rw in hamsters, and this parameter was negatively correlated with the liver retinol concentration in experiment I(r = -0.89). Retinol deficiency has a marked effect on growth and fertility of D. viteae in hamsters.  相似文献

The multimammate rat, Mastomys natalensis was used as a model system to evaluate the chronic effects of infection by Dipetalonema viteae on hepatic mixed function oxidase activity. Total hepatic cytochrome P450 content and related total tissue mixed function oxidase activity were decreased to about 50% of control levels at patent phase of infection. The decrease in total tissue mixed function oxidase activity was due to a large decrease in cytochrome P450 concentration in the endoplasmic reticulum. Although the decrease in total liver monooxygenase activity in two substrates aniline and aminopyrine roughly paralleled the loss in cytochrome P450 content, several other microsomal enzyme markers not related to cytochrome P450 monooxygenation were elevated in proportion to total liver microsomal protein content. These results suggest that in M. natalensis during experimental filariasis, there is proliferation of hepatic cells with normal content of endoplasmic reticulum. Furthermore, there appears to be selective toxicity for hepatic cytochrome P450 and related monooxygenase activities. This may compromise the animal's ability to metabolize and dispose of other drugs to which the animals may be exposed in the course of infection.  相似文献

The antigenic properties of adult male and female of Dipetalonema viteae were studied by immunocytochemistry. Using antisera of the rodents Meriones unguiculatus and Mastomys natalensis infected with D. viteae, the binding of antibodies to sections of filariae embedded in Epon was assayed by the peroxidase-antiperoxidase (PAP) technique and by electron microscopy. The optimal staining intensity was obtained with an antiserum dilution of 1:5000. Control sera were obtained from sex and age matched uninfected animals. Female D. viteae showed maximal antigen-antibody reactions within the uterus: in the inner uterus wall, in the "nutrient channels" between the maturing eggs and the differentiating microfilariae, on the eggshells, in the cuticula of microfilariae and in the spermatheca on the cell membrane of the mature spermatozoa. Male filariae showed binding of antibodies in the vesicula seminalis: in the nucleus and the nuclear membrane of primary spermatocytes and on maturing spermatids. Less pronounced antigen-antibody reactions in the cuticula, muscle and intestine were observed in both sexes. The PAP-technique offers significant improvements in comparison with other techniques, e.g., immunofluorescence, used to detect antigens on filariae: the PAP-technique has an increased sensitivity with a concomitant reduction in nonspecific background and can be used for both light and electron microscopy; moreover, PAP-treated tissues can be stored indefinitely at room temperature.  相似文献

Mature male Dipetalonema viteae released a substance(s) which caused enhanced microfilaraemia in infected hamsters. In hamsters implanted with female D. viteae, the microfilaraemia of a subsequent infection was suppressed. Microfilariae of Litomosoides carinii and Dipetalonema viteae absorbed about ten times as much diethylcarbamazine (DEC) as did their adults, but bound much less. The higher uptake of DEC by the microfilariae might be related to the fact that DEC is microfilaricidal. Binding of DEC with the parasites supports the view that the microfilaricidal action begins with the fixation of the drug to the microfilariae.  相似文献

Radioisotopically labeled glucose and pyruvate were employed to elucidate biochemical mechanisms utilized by the filariid Dipetalonema viteae during cultivation. Adults isolated from amicrofilaremic hamsters were incubated at 37 C in a mixture of NCTC135:IMDM (NI), with either D-[14C-(U)]glucose or [1-14C]pyruvate, under a gas phase of 5% CO2/N2 for 3 days. Labeled organic acids were separated and quantified by ion exchange chromatography. High performance liquid chromatography (HPLC) was used for separation and quantification of the 23 free amino acids in the NI medium. Ion exchange chromatography revealed that lactate was the major glycolytic end product, accounting for 90-97% of the original carbon utilized. Small amounts of radioactivity were recovered in succinate and variably in acetate fractions. HPLC analysis demonstrated that some amino acids increased, some decreased, and some remained at the initial concentration. Alanine exhibited the greatest change, consistently increasing from 2 to 4 times the original concentration. Analyses of purified amino acid peaks revealed radioactivity only in the alanine peak, accounting for 2-4% of the original carbon utilized.  相似文献
