Summary In the crested newt, the ultrastructural organization of the pars nervosa is analogous to that already known in non-mammal tetrapods. An orderly array of ependymal cells makes up the inner limiting layer while less abundant pituicytes are irregularly distributed within this organ. Light and dark pituicytes can be distinguished on the basis of the relative density of the cytoplasmic matrix and the distribution of the cell organelles.Both the ependymal cells and pituicytes are rich in dense bodies and possess extensive processes which ramify among the nerve fibers, often reaching the pericapillary space which they can line for long distances.The main components of the pars nervosa are nerve fibers and nerve terminals (type A), containing electron dense granules 1200–2000 Å in diameter together with clear vesicles averaging 250–400 Å. These fibers are likely to correspond to the aldehyde fuchsin positive neurosecretory fibers revealed by light microscopy. Differences in the granule size within the fibers and terminals lead to further recognition of two subgroups (A1 and A2).Other fibers and terminals (type B) containing clear vesicles and granular vesicles 600 to 1000 Å in diameter, possibly of aminergic type, are also encountered. These fibers are rare and can be seen only in the portion of the pars nervosa near the pars intermedia of the adenohypophysis.Lastly, fibers and terminals containing only clear vesicles ranging from 250 to 400 Å (type C) are occasionally found.Nerve endings are often formed by type A fibers on the perivascular space and on the perivascular processes of the ependymal cells and pituicytes. Summary The folliculo-stellate network of the avian adenohypophysis consists of stellate cells surrounding colloid-containing follicular cavities into which cilia and microvilli project. Other identifying criteria are agranularity, junctional complexes at the apical pole, presence of cytoplasmic processes ramifying between adjacent secretory cells, and close appositions of plasma membranes linking folliculo-stellate cells and presumptive thyrotropic cells.Transmission electron microscopy reveals that TRH and L-DOPA induce simultaneous ultrastructural changes in the folliculo-stellate network and in the thyrotropic cells. TRH transforms at cell of the cephalic lobe into a highly hypertrophic cell in which enlargement of cisterns of rough endoplasmic reticulum containing secretory granules, development of a large Golgi complex, presence of newly synthesized secretory granules, and granulation of the cytoplasm are the main features. In the meantime, the follicular cavities become dilated by large amounts of homogeneous colloid. The administration of L-DOPA also leads to the development of dilated cisterns in presumptive thyrotropic cells of the cephalic lobe. Intracisternal granules, immature secretory granules, and large Golgi complexes, however, are not observed. Degranulation of the cytoplasm is obvious. The follicular cavities of both cephalic and caudal lobes are enlarged and filled with colloid in which granular elements are noted.The ultrastructural changes observed in thyrotropic cells and in the folliculo-stellate network reflect functional changes induced by the experimental manipulation. Summary Using the immunoperoxidase technique and antisera to the specific beta () subunits of bovine and rat TSH1, selective immunocytochemical staining was localized in a specific cell population in the pars distalis of the dog pituitary gland. These TSH cells were found to be positive to aldehyde fuchsin, alcian blue, periodic acid-Schiff (PAS) and aniline blue. With the performic acidalcian blue (pH 0.2) -PAS-orange G procedure these cells stained blue-purple, demonstrating FSH/LH cells (blue or turquoise), ACTH/MSH cells (redpurple) and PRL cells (orange-red). The TSH cells were further differentiated from other functional cell types of the pars distalis on the basis of their typical cytological features, intraglandular distribution and by immunocytochemical double staining. In the pars distalis of adult male dogs the TSH cells were mostly shown to be smaller in size and less numerous than in bitches in the anestrous phase of the sexual cycle. Moreover, cytological alterations in the immunoreactive thyrotrophs in the pituitary of male and female dogs generally paralleled the spontaneous changes in thyroid function associated with thyroid atrophy and/or pituitary insufficiency, and thyroid hyperplasia or goiter. In conclusion, because of their specificity and high potency, the antisera to the -subunits of bovine and rat TSH represent an effective tool for the selective immunocytochemical localization of TSH in the dog pituitary. Summary An electron microscopic analysis was performed on the pars distalis of the hypophysis of hepatectomized mice. Intact and sham operated mice served as controls. The STH cells presented striking changes that were most intense and widespread in those animals sacrificed at midnight of the second day after hepatectomy. These changes can be summarized as follows: 1) Hypertrophy of the Golgi complex with increased number of immature granules within the Golgi zone. This change appeared also in otherwise unmodified STH cells. 2) Strong dilatation of the endoplasmic reticulum whose cisternae contained much electron dense material. 3) Granules with partially diminished electron density, some of them in spatial relation with the plasma membrane and others swelling and bursting within the cytoplasm. All transitions between unchanged 350–400 m granules and extremely altered ones, were seen. 4) Release sites, characterized by dense zones in the plasmalemma, close to aggregates of electron lucent microvesicles, and almost empty granule membranes. 5) Increase in the density of the mitochondria which appeared grouped near the Golgi zone. 6) Increase in the number of large lysosomes of the autophagic vacuole type. 7) Irregular nuclear outlines. Summary Three different immunocytoenzymatic techniques were used to identify and characterize the thyrotropic cells in the pituitary of normal guinea pigs at the ultrastructural level (superimposition technique, immunocytochemical technique using P.A.P. and indirect immunohistoenzymatic method before embedding).These cells are characterized by a dark cytoplasm with granules ranging from 1500 to 2000 Å in diameter. The appearance of these granules is very variable: some display a marked electron density and are homogeneous but some have a less marked electron density with a more electron dense peripherally situated region.The TSH molecules are essentially confined to the granules but when the immunocytochemical reactions are carried out before embedding, positive staining is also seen in the cytoplasm and the outer surface of most of the rough endoplasmic reticulum membranes. Summary An electron microscopic analysis was performed on the pars distalis of the hypophysis of mice bearing transplanted hepatomas. Normal mice served as controls. In animals bearing a fast growing as well as in those bearing a slow growing hepatoma, the STH cells presented changes indicating increased release and synthesis of its secretory product. These changes can be summarized as follows: 1) Increased number of granuledepleted STH cells which were very irregular in size and shape. 2) Very irregular and sometimes bizarre nuclei. 3) Plasma membranes irregular and convoluted. 4) Increased quantity of endoplasmic reticulum which appeared sometimes dilated and contained material of varying electron density. 5) Abundant, irregular and frequently ramified mitochondria. 6) Hypertrophic Golgi system with increased number of microvesicles. 7) Increased number of large lysosomes of different type. 8) Increased number of fat droplets.Work carried out with the financial assistance of grants of the Consejo Nacional de Investigaciones Científicas y Técnicas de la Republica Argentina and the Comisión de Investigación Científica de la Universidad Nacional de La Plata. An investigation of N-terminal pro-opiocortin peptides in the rat pituitary   总被引:1,自引:0,他引:1  
Extracts of neurointermediate lobe (NIL) and anterior lobe (AL) of the rat pituitary, and material released from perfused rat pars distalis (PD) and pars intermedia (PI) cells were gel chromatographed and monitored using three antisera, each recognizing different regions of the non-corticotropin (ACTH)-lipotropin (LPH) portion of pro-opiocortin (POC). Two peaks (termed N-POC I) which emerged close to the elution position of rat beta-LPH were detected. The first peak was reduced significantly in the PI. Two smaller N-POC fragments which eluted near beta-endorphin were detected only in extracts and secretions of intermediate lobe tissue. One peak cross-reacted in the gamma 3-melanotropin (MSH) assay (N-POC III) whereas the other peak possessed amino (N)-terminal N-POC immunoreactivity (N-POC II). The results demonstrated differences in the distribution and nature of N-POC peptides released and extracted from the PD and PI of the rat pituitary, and suggest that the enzymic processing of N-POC is different in the two pituitary lobes.  相似文献

Summary The effects of synthetic thyrotropin-releasing hormone on pituitary prolactin and thyrotropic cells were investigated in adult male Rana perezi (formerly Rana ridibunda) frogs. Animals were given daily injections of synthetic thyrotropin-releasing hormone into the dorsal lymph sac. Prolactin and thyrotropic cells were identified by the colloidal-gold method, using anti-human prolactin and anti-human--thyrotropin hormone as primary antisera. The stereological parameters of the rough endoplasmic reticulum, Golgi complex, and secretory granules of prolactin and thyrotropic cells were evaluated by ultrastructural morphometry (point-counting method). Thyrotropin-releasing hormone caused cytological changes in both cell-types which were consistent with increased synthesis and release of both prolactin and thryrotropin. These changes were still significant after 48 h treatment in the case of thyrotropic cells, while in prolactin cells the thyrotropin-releasing hormone increased the number of secretory granules. After 6 days, the cells resembled essentially those used as controls. These results indicate that thyrotropin-releasing hormone stimulates the synthesis and release of prolactin and thyrotropin, and that the response of each cell type to this hypothalamic stimulus follows a different time-course.This work has been supported by grants no. 2184-83 and PB 86-0095 from the Comisión Interministerial para la Ciencia y Tecnología, Spain  相似文献

Summary Five categories of granulated cells were distinguished by their ultrastructural features, and quantitative analyses were made of the pars distalis cells in normal and castrated lizards. The gonadotropin-producing cell was identified on the basis of its uniform distribution in the gland as well as from cytological changes resulting from castration. The secretory granules of the gonadotropic cell vary in size (100–500 m) and density, and lipid bodies are commonly present. Following castration, the endoplasmic reticulum proliferates, forming many small, rough-surfaced, dilated cisternae which do not coalesce greatly as in other vertebrate species. Degranulation is accompanied by hypertrophy and hyperplasia of the mitochondria and by the appearance in the cytoplasm of conspicuous clusters of microfilaments. The designated gonadotropic cell was the only class of secretory cell showing consistent changes following three weeks of castration.In addition to the uniformly distributed gonadotrope cell, two secretory cells occur mainly in the rostral half of the gland, and two in the caudal half. Summary Differentiated nongranulated (folliculo-stellate) cells were observed in the centre and periphery of the cords of cells in the pars distalis of the adult tammar wallaby (Macropus eugenii). The nongranulated cells formed follicles containing a small lumen; the cell apices were joined by junctional complexes including zonulae adhaerentes and maculae adhaerentes (desmosomes). Follicles of granulated cells were also occasionally found. Follicles were rarely observed in the adult, but were numerous and generally larger in pouch-young wallabies. Moreover, whereas the involvement of granulated cells in the follicles of the adult was infrequent, they were common components of follicular structures in the pouch-young. The marginal cells at the periphery of the pars distalis in contact with the hypophysial cleft had many of the cytological characteristics of the nongranulated cells in the central pars distalis. In both adult and pouch-young wallabies, nongranulated cells at the periphery of the cell cords were juxtaposed such that they formed sinusoid-like spaces or lacunae, presumably as part of the extravascular channel system. The observations are discussed in light of the proposed phagocytotic, metabolite and/or hormone transport and stem cell roles attributed to these cells.This work was supported by a travel grant and grant-in-aid of research from the N.S.E.R.C. Abstract: Administration of histidyl-proline diketopiperazine (cyclo (HisPro)) to rats attenuates ethanol-induced sleep. To understand the role played by cyclo (His-Pro) in the pathophysiology of prolonged alcohol consumption, we have measured the distribution of this peptide in brains of control and alcohol-treated rats. The data show that prolonged alcohol consumption increases the concentration of cyclo (His-Pro) in hypothalamic as well as extrahypothalamic brain. These changes may reflect a physiologic adaptation of the brain during alcohol consumption.  相似文献

The unfertilized egg of the newt, Cynops pyrrhogaster, has a second meiotic spindle at the animal pole and numerous cortical cytasters. After physiologically polyspermic fertilization, all sperm nuclei incorporated into the egg develop sperm asters, and the cortical cytasters change into bundles of cortical microtubules. The size of the sperm asters in the animal hemisphere is ∼5.6-fold larger than that in the vegetal hemisphere. Only one sperm nucleus moves toward the center of the animal hemisphere to form a zygote nucleus with the egg nucleus. This movement is inhibited by nocodazole, but not by cytochalasin B. The centrosome in the zygote nucleus divides into two parts to form a bipolar spindle for the first cleavage synchronously with the nuclear cycle, but centrosomes of accessory sperm nuclei in the vegetal hemisphere remained to form monopolar interphase asters and subsequently degenerate around the first cleavage stage. The size of sperm asters in monospermically fertilized Xenopus eggs was ∼37-fold larger than those in Cynops eggs. Since sperm asters that formed in polyspermically fertilized Xenopus eggs exclude each other, the formation of a zygote nucleus is inhibited. Cynops sperm nuclei form larger asters in Xenopus eggs, whereas Xenopus sperm nuclei form smaller asters in Cynops eggs compared with those in homologous eggs. Since there was no significant difference in the concentration of monomeric tubulin between those eggs, the size of sperm asters is probably regulated by a component(s) in egg cytoplasm. Smaller asters in physiologically polyspermic newt eggs might be useful for selecting only one sperm nucleus to move toward the egg nucleus. Mol. Reprod. The effect of treatment with L-3,5,3'-triiodothyronine (T3) on the levels of pyroglutamyl peptidase I and pyroglutamyl peptidase II in rat brain regions, pituitary, and serum was studied. Pyroglutamyl peptidase I cleaves pyroglutamyl peptides such as thyrotropin releasing hormone (TRH), luteinizing hormone releasing hormone, neurotensin, and bombesin, whereas pyroglutamyl peptidase II appears to be specific for TRH. Acute administration of T3 did not affect pyroglutamyl peptidase I in any of the regions studied, whereas pyroglutamyl peptidase II was significantly elevated in frontal cortex and pituitary. Treatment with T3 for 10 or 14 days significantly elevated pyroglutamyl peptidase I in pituitary, hypothalamus, olfactory bulb, hippocampus, and thalamus. Chronic T3 treatment elevated pyroglutamyl peptidase II in frontal cortex and in serum. These studies demonstrate regulation of neuropeptide degrading enzymes by thyroid hormones in vivo. This regulation may play a role in the negative feedback control of thyroid status by T3.  相似文献

Summary Using immunofluorescent techniques thyrotropin releasing hormone (TRH) is demonstrated in skin of Rana pipiens and R. catesbeiana. Hyperresponsiveness to norepinephrine contributes to post-traumatic stress disorder (PTSD). Prazosin, a brain-active blocker of α1-adrenoceptors, originally used for the treatment of hypertension, has been reported to alleviate trauma nightmares, sleep disturbance and improve global clinical status in war veterans with PTSD. Thyrotropin-releasing hormone (TRH, pGlu-His-Pro-NH2) may play a role in the pathophysiology and treatment of neuropsychiatric disorders such as major depression, and PTSD (an anxiety disorder). To investigate whether TRH or TRH-like peptides (pGlu-X-Pro-NH2, where “X” can be any amino acid residue) participate in the therapeutic effects of prazosin, male rats were injected with prazosin and these peptides then measured in brain and endocrine tissues. Prazosin stimulated TRH and TRH-like peptide release in those tissues with high α1-adrenoceptor levels suggesting that these peptides may play a role in the therapeutic effects of prazosin.  相似文献

The effects of a new thyrotropin releasing hormone (TRH) analogue, YM-14673 (N alpha-[[(S)-4-oxo-2-azetidinyl]carbonyl]-L-histidyl-L-prolinamide dihydrate), on the release of acetylcholine (ACh) in free-moving rats were examined in vivo by intracerebral dialysis. In the frontal cortex, YM-14673 (0.1-0.3 mg/kg) caused a significant dose-dependent increase in the extracellular levels of ACh, suggesting that YM-14673 stimulated the ACh release. These actions of YM-14673 were about 50 times more potent than those of TRH. On the other hand, extracellular levels of ACh in caudate nucleus were not changed following injection of YM-14673 even at 3 mg/kg. TRH and methamphetamine also increased the release of ACh in frontal cortex. Haloperidol prevented the increase in the methamphetamine-induced release of ACh, whereas the increased release of ACh produced by YM-14673 was partially antagonized by haloperidol. These results suggest that the dopaminergic system affects the facilitatory effects on the ACh release in the frontal cortex and that the stimulatory effect of YM-14673 on the frontal cholinergic neurons is partially mediated by dopaminergic neurons.  相似文献

Summary An electron microscopic study aimed at differentiation between castration gonadotrophs and posthepatectomy STH cells was performed on the pars distalis of the pituitary of castrated male mice, after partial hepatectomy. The ultrastructural features observed permit the distinction of both cell types. In the present experiments some remarkable ultrastructural changes, other than those described in a previous report, have been found in STH cells of hepatectomized mice with or without previous castration. Most of them contained masses of heterogeneous electron density, suggesting fusion of granules. These masses and some secretion granules were observed close to the plasma membrane, apparently in the process of discharging material into the pericapillary space. Granular extrusion was more frequent than normally. An increased number of lysosomes, probably related to the digestion of overproduced secretion material, was evident. The appearance of concentric lamellar formations might be related to an increase in cell movements. Summary Intact female rats received transplants of two hypophysial partes distales under the kidney capsule. The plasma levels of prolactin were determined, and the ultrastructure of the grafted gland was studied 15, 45 and 90 days after the operation. Although prolactin levels in the three experimental groups were significantly higher than those in control rats, a decrease in prolactin level was detected in the 45-day samples. Summary The structure of the nongranulated cells in the sea lamprey adenohypophysis and similar cells of the rostral pars distalis in a number of actinopterygian fishes was examined with the aim of determining the role(s) of these cells in pituitary function.A number of possible roles are proposed for the nongranulated cells. In salmonids and Amia calva the cells may be involved in the active movement of material into or from the follicle lumina. The structure of the nongranulated cells in in vitro cultured and in in vivo transplanted trout rostral pars distalis also suggests a phagocytotic role for these cells. In teleosts with a non-follicular rostral pars distalis the nongranulated cells appear to play roles in the release of granules from the prolactin cells and in the subsequent dispersal of the hormone (and/or carrier substance) into the peripheral circulation.We wish to thank Dr. B.I.
