The adaptation ofCamellia rusticana, an evergreen broad-leaved shrub found in areas of heavy snowfall in Japan, to heavy snowfall environments, and the mechanisms by which it is damaged in winter above the snow, were investigated. The stomatal response and photosynthetic characteristics ofC. rusticana were compared to those ofCamellia japonica found in areas of light snowfall. In field conditions, the mean net photosynthesis ofC. rusticana at photon flux density (PFD) over 200 μmol m−2s−1 (Pn(>200). was 50% larger than that ofC. japonica, but in both light saturated and CO2 saturated conditions, the O2 evolution rate (Pc) ofC. rusticana was not different from that ofC. japonica. Mean leaf conductance at PFD over 200 μmol m−2s−1 (gl(>200)) was about 100% larger than that ofC. japonica in the field. The Pn(>200)) was 50% ratio ofC. rusticana was 37% higher than that ofC. japonica which suggests thatC. rusticana's larger Pn(>200) can be explained by its larger gl(>200). WhenC. rusticana trees wintering underneath the snow were projected above it, the leaves of these plants showed serious drought within five days in non-freezing conditions. Their Pc and the maximum stomatal conductance decreased by half and did not recover. The leaves ofC. rusticana showed larger gl(>200) and a less sensitive stomatal response to the decrease of leaf water potential than that ofC. japonica. The stomata characteristics ofC. rusticana caused larger net photosynthesis than that ofC. japonica during the no snow period, and caused the need for snow cover in winter as protector from winter drought.  相似文献

Camelliarusticana is an evergreen broad-leaved shrub found only in areas of heavy snowfall in Japan. The ecophysiological tolerance of this species to prolonged snow cover was studied in comparison with those of Camellia japonica found in areas of light snowfall. The shoots of C. rusticana and C. japonica were stored under a simulated snow cover for 1 year. During the experimental period, about 20% of the leaves of C. japonica defoliated after 90–140 days of storage and more than 80% of the leaves browned within 360 days. The proliferation of fungi was observed on all shoots of C. japonica after 200 days. In contrast, the leaves of C. rusticana showed no visible changes throughout the experimental period. The decline in the rate of soluble carbohydrate content in C. rusticana was about one-sixth of that in C.japonica. The photosynthetic capacity (O2 exchange rates at saturated light and CO2 at 20°C) of C.japonica dropped to about half its initial value after 140 days, while that of the C. rusticana did not change even after 360 days. Under dark conditions, the stomata of C. rusticana were always closed through the experiment, but those of C. japonica were not completely closed after 90 days. The differences in stomata characteristics and the consumption rate of soluble carbohydrates under snow-covered conditions between the two Camellia species are important factors for determining their habitat segregation by snow depth.  相似文献

Simple sequence repeat (SSR) markers were developed for Japanese white birch, Betula platyphylla var. japonica, using previously designed primer pairs derived from expressed sequence tags (ESTs). Out of 98 unpublished primer pairs, 35 yielded clear PCR amplification products, 11 of which revealed polymorphism in eight individuals sampled across the species’ range. The number of alleles detected and the expected heterozygosity ranged from 1 to 10 and 0.000 to 0.857, respectively, when these 11 loci were examined in 24 individuals from a single B. platyphylla var. japonica population. In cross-species transferability tests most of the 11 loci were also polymorphic in three other Betula species examined, but not B. maximowicziana. We have now developed a total of 25 polymorphic EST-SSRs for the genus Betula (including 14 we previously developed), which are likely to be highly useful in studies of various aspects of population genetics, including hybridization dynamics, in the genus.  相似文献

We examined the floral morph of tetraploid Ophiorrhiza japonica Blume var. amamiana Hatus. and diploid O. japonica var. japonica to elucidate the association of distyly and ploidy levels. Chloroplast DNA phylogeny was reconstructed to determine the number of tetraploidization events and floral morph shifts in O. japonica. All individuals of O. japonica var. amamiana proved to be long-homostylous, whereas O. japonica var. japonica was distylous with typical long- and short-styled flowers. Distyly is related to the ploidy level. The bagging treatment of flowers indicated that O. japonica var. amamiana is self-compatible and potentially automatically self-pollinating. In cpDNA sequencing analysis, no haplotype was shared between the two varieties. The cpDNA haplotype network displayed the monophyly of O. japonica var. amamiana, suggesting a single origin of this variety. Hence, both tetraploidization and the breakdown of distyly to homostyly in O. japonica var. amamiana likely occurred just once. Because O. japonica var. amamiana having the morphological and cytological entity is recognized as a single lineage and clearly separated from O. japonica var. japonica, this variety can be considered to be a distinct species. We therefore propose to raise O. japonica var. amamiana to the rank of species. Plant adaptation to an environment subject to heavy snowfalls was investigated in four species of evergreen shrubs growing in a Fagus crenata forest in an area of Honshu on the Sea of Japan. These shrubs stored carbohydrates in some organs before the snowy season and were covered with snow for 4–5 months. Aucuba japonica var. borealis, Camellia rusticana, and Ilex crenata var. paludosa maintained a reserve of carbohydrates during the snowy season. In Daphniphyllum macropodum var. humile, the reserve of carbohydrates decreased during winter. The respiration rates in the first three species decreased from autumn to winter, whereas the decrease in D. macropodum was slight. It was found that the first three species could use reserve carbohydrates for the growth of new shoots after the thaw, whereas in the last species the growth of new shoots depends on high photosynthetic activity in late spring. Our findings suggest some types of matter economy in evergreen shrubs for wintering in an environment of heavy snow.  相似文献

Aucuba japonica varieties are common evergreen understory shrubs in Japan.Aucuba japonica var.borealis is distributed on the Sea of Japan side of Honshu and Hokkaido where heavy snow cover lasts for more than 3 months in winter.Aucuba japonica var.japonica is distributed in areas with shallow or no snow on the Pacific Ocean side of Honshu and Shikoku. The ecophysiological characteristics of var.borealis were compared with those of var.japonica to examine the effects of heavy and long-term snow cover on the life cycle of var.borealis. Shoots of both varieties were shaded in crushed ice for 110 days, but their photosynthetic activities, chlorophyll contents and the chlorophylla/b ratio was not affected. The leaves of var.borealis were no less frost tolerant than those of var.japonica. In spite of the difference in environmental factors, both varieties had similar characteristics in seasonal changes of photosynthesis, respiration and chlorophylla/b ratio. These results suggest that var.japonica could survive in areas with heavy snow where it does not normally occur. Leaf net production (LNP) was estimated based on the microclimatic data and seasonal photosynthetic and respiration rates. The difference in the annual LNP between the two varieties was equivalent to the difference in the LNP during the snow season. One of the major effects of snow cover is to interrupt and reduce the production period of var.borealis.  相似文献

Intrapopulational spatial genetic structure was examined in two populations ofChionographis japonica var.japonica, a self-incompatible perennial, by spatial autocorrelation analysis of enzyme polymorphism. Although most spatial autocorrelation indices (Moran'sI) in the shortes distance class were significantly positive, most in the other distance classes did not significantly deviate from the values expected from random distributions of genotypes in both populations. This contrasts with a spatial genetic pattern previously reported for a population of the predominantly selfing congener,C. japonica var.kurohimensis, indicating that pollen-mediated gene flow highly impedes genetic substructuring within populations of outcrossingC. japonica var.japonica. Genetic similarity in very proximate distance found in outcrossingC. japonica var.japonica is probably due to restricted dispersal of seeds.  相似文献

Allozyme variation was examined in three diploid taxaChionographis japonica var.japonica, var.kurokamiana, andC. koidzumiana and three tetraploid taxaC. japonica var.kurohimensis, ssp.hisauchiana, and ssp.minoensis. Results show thatC. japonica var.kurokamiana is genetically closer toC. koidzumiana than to var.japonica. In the tetraploid taxa, fixed heterozygosities were found at several loci, and this supports the hypothesis that these taxa are allotetraploids. Furthermore, the tetraploid taxa have many unique alleles not found in the diploid taxa. This suggests that sufficient time has passed since the origin of tetraploids for new mutations to have been fixed.  相似文献

RAPD and isozyme analyses based on numerous markers have been used for the first time to investigate patterns of phenetic and genetic differentiation among and within nine wild populations of the genus Chaenomeles represented by the species C. japonica, C. speciosa, C. cathayensis and C. thibetica. Highly significant correlations were found between the two different marker systems for both phenetic distances and gene diversity estimates. In agreement with previous studies on cultivated Chaenomeles material, C. japonica was clearly differentiated from C. speciosa and C. cathayensis. The recently recognised species C. thibetica appeared to be rather closely related to C. cathayensis. Populations of C. japonica and C. speciosa were considerably more diverse than populations of C. cathayensis and C. thibetica. Correspondingly, most of the total variability could be attributed to the within-population differentiation in the case of C. japonica and C. speciosa, and to the between-population differentiation in the case of C. cathayensis. Differences in mating systems among the species can be suggested as a possible explanation of the results. A discordant pattern was found between RAPDs and isozymes in the analyses of population structure within C. japonica. This may be explained by a higher proportion of non-neutral markers for isozymes than for RAPDs. Microsatellites were isolated and characterized for the Japanese oak species, Quercus mongolica var. crispula, distributed in temperate deciduous forests of Japan. Eleven of the 48 primer pairs designed successfully amplified unambiguous and polymorphic single loci among 67 Q. mongolica var. crispula individuals within a plot in southwestern Japan. The observed and expected heterozygosities of the 11 microsatellite markers ranged from 0.522 to 0.896 and from 0.536 to 0.882, respectively. These polymorphic microsatellite markers are useful for estimating pollen‐mediated gene flow in Q. mongolica var. crispula.  相似文献

Garden experiments focused on vegetative regeneration were carried out with four invasive taxa of the genus Reynoutria (R. japonica var. japonica, R. japonica var. compacta, R. sachalinensis and a hybrid between R. sachalinensis and R. japonica var. japonica, R. ×bohemica). Regeneration ability of stems and rhizomes, timing of shoot emergence and biomass production were studied under the following treatments: laid horizontally on the soil surface; placed upright; buried in the soil; floating in water. Two different soils (sand and garden loam) representing contrasting nutrient levels were applied. Differences were found in the capability and speed of regeneration, as well as in the quality of shoots produced. Regeneration from stems was less efficient than that from rhizomes in all taxa except R. sachalinensis. R. ×bohemica exhibits higher regeneration potential (61%) than all other taxa and can be considered as the most successful taxon of the Czech representatives of the genus Reynoutria in terms of regeneration and establishment of new shoots. High regeneration capacity was also exhibited by R. japonica var. compacta (52%). Other taxa showed generally lower regeneration rates (R. japonica var. japonica 39% and R. sachalinensis 21%), but under some treatments the percentage of regenerated segments was high, too. R. japonica var. japonica rhizomes regenerated successfully in all three soil treatments but not in the water. An opposite pattern was found for its stems: they regenerated well if exposed to water treatment but in the soil, they did not regenerate at all. Particular taxa responded to the soil type in a contrasting way. R. sachalinensis and R. ×bohemica regenerated better in loam while the opposite was true in R. japonica var. japonica. R. japonica var. compacta produced the tallest and R. ×bohemica the heaviest and most robust shoots. It is concluded that rhizomes are more crucial than stems for the spread of knotweeds through fragmentation and clonal growth, suggesting the importance of soil disturbance.  相似文献

To construct a molecular-marker-assisted selection (MAS) system, research was done on identifying molecular markers linking to longer frond length, a crucial selection index in the breeding of the commercially important seaweed Saccharina japonica. An F2-segregant population of 92 individuals was obtained by crossing two prominent S. japonica strains. Genomic DNA from ten individuals with the longest frond and ten individuals with the shortest frond in the F2-segregant population were mixed to create two DNA pools for screening polymorphic markers. In bulked-segregant analysis (BSA), out of 100 random amplified polymorphic DNA (RAPD) primers only two produced three polymorphic RAPD markers between the two DNA pools. In conversion of the three RAPD markers into sequence-characterized amplified region (SCAR) markers, only one was successfully converted into a SCAR marker FL-569 linking to the trait of longer frond. Test of the marker FL-569 showed that 80% of the individuals with longest fronds in a wild population and 87.5% of individuals with the longest fronds in an inbred line “Zhongke No. 2” could be detected by FL-569. Additionally, genetic linkage analysis showed that the SCAR marker could be integrated into the reported genetic map and QTL mapping showed that FL-569 linking to qL1-1. The obtained marker FL-569 will be beneficial to MAS in S. japonica breeding.  相似文献

We have examined morphological and chromosomal variation inFallopia sect.Reynoutria in Korea to clarify their taxonomic identities and to determine whether their morphological variability is associated with ploidy levels. Principal components analysis (PCA) of individuals from 21 populations, using major distinguishing characters, revealed the presence of four major entiries of sect.Reynoutria in Korea; these includeF. sachalinensis, F. japonica var.japonica, F. forbesii, and the Nonsan population consisting of presumed hybrids. Based on morphology, it is hypothesized that the Nonsan population was probably derived from multiple hybridization events involving the three named taxa. The results also indicate thatF. forbesii is distinct fromF. japonica var.japonica. Polyploidy is more prevalent in sect.Reynoutria than has been previously recognized.Fallopia sachalinensis in Korea occurs as dodecaploids with 2n=132; our count is the first dodecaploid count for the species, and represents the highest chromosome number known in the genus.Fallopia japonica var.japonica occurs as tetraploids (2n=44), hexaploids (2n=66), and octoploids (2n=88), whileF. forbesii occurs as hexaploids (2n=66) and octoploids (2n=88); our counts appear to be the first reported chromosome numbers forF. forbesii. Morphological analysis indicates that there is no apparent correlation between the ploidy levels in these taxa and the morphological characters that we have considered in this study except that the tetraploids ofF. japonica var.japonica tend to have somewhat thicker leaves.  相似文献

Japanese knotweed s.l. comprises Fallopia japonica, F. sachalinensis, F. × bohemica and any F2s or backcrosses. The parental taxa were introduced from the East to the West as garden ornamentals in the nineteenth century, and soon spread beyond the confines of the garden to become widespread and persistent weeds. Since only female F. japonica var. japonica was introduced, its impressive spread has occurred solely by vegetative means. However, the initial lack of genetic variability has been complemented by an extensive series of hybridisations in the adventive range. We examine the history, spread, reproductive biology and ecological impact of these species in the West. The role and importance of polyploidy and hybridisation in their invasion of the West is discussed, as are the implications of these factors for the potential further evolution of the group.  相似文献

Chattonella marina var. antiqua and C. marina var. marina (Raphidophyceae) are red tide‐forming, harmful phytoplankton species. We investigated the genetic diversity and genetic relationship among the populations using microsatellite markers to identify putative sources of C. marina var. antiqua and C. marina var. marina in Japanese coastal populations. A positive correlation between genetic divergence and geographical distance (isolation by distance) was recognized for C. marina var. antiqua. The C. marina var. antiqua populations were established throughout a geological time scale, and genetic divergence had progressed in each population with gene flow depending on geographic distances. In contrast, isolation by distance was not observed for C. marina var. marina populations, and the genetic divergence among populations was extremely high. The Tokyo Bay population of C. marina var. marina, which was first recognized in 2008, had many private alleles but was related to the Kagoshima Bay population. The Tokyo Bay population may have been established by several invasions from the Kagoshima Bay population and other regions.  相似文献

Stachyurus macrocarpus and S. macrocarpus var. prunifolius are critically endangered shrub species in the Bonin (Ogasawara) Islands, Japan. These species are extremely rare, and the numbers of individuals in wild populations are 68 in S. macrocarpus and 13 in S. macrocarpus var. prunifolius. For the investigation of genetic diversity, genetic structure and relatedness among remnant individuals of these endangered species, we developed eight microsatellite markers from S. macrocarpus var. prunifolius and characterized these markers for S. macrocarpus var. prunifolius and S. macrocarpus using all naturally occurring individuals of these species. The expected heterozygosities of these markers ranged from 0.14 to 0.67 in S. macrocarpus var. prunifolius, and from 0.02 to 0.84 in S. macrocarpus. The markers described here will be useful for investigating the genetic diversity, genetic structure and relatedness among remnant individuals, and planning the restoration of these critically endangered species.  相似文献

A new species,Stromatinia cryptomeriae, is described based on a specimen collected in Iwate Prefecture, Japan. It was found on fallen dead branches and twigs of Japanese cedar,Cryptomeria japonica. The morphology of isolates on potato-dextrose agar (PDA), which were obtained from single ascospores ofS. cryptomeriae, was identical withGloeosporidina cryptomeriae, the causal fungus of Japanese cedar twig blight. In an inoculation test using single ascospore isolates, many minute black spots (sclerotioid bodies; sclerotules) and acervuli ofG. cryptomeriae were formed on the necrotic lesions, which developed into typical symptoms of Japanese cedar twig blight. These results show thatStromatinia cryptomeriae is the teleomorph ofG. cryptomeriae. On PDA, the fungus grew over a range of about 1 to 25°C, with the optimum growth at about 15–20°C.  相似文献

We identified diagnostic chloroplast DNA and mitochondrial DNA markers that can (a) discriminate between Larix gmelinii var. japonica and L. kaempferi, and (b) determine the maternal and paternal species of hybrids between them by exploiting the difference in inheritance mode between the two genomes. We also investigated the hybridization rates at a site with two types of interspecific seed orchard—a new type with rows of a single maternal clone of L. gmelinii var japonica among rows of L. kaemferi and a “traditional” type with multiple, intimately mixed clones—in 2 years using chloroplast diagnostic DNA markers. The average hybridization rates in the single maternal clone (SMC) interspecific seed orchards [84.2% (±9.4%) in 2004 and 94.1% (±3.9%) in 2005) were higher than that in the traditional interspecific seed orchards [15.9% (±13.4%) in 2004 and 30.0% (±25.5%) in 2005] because of the self-incompatibility of the L. gmelinii var. japonica clone. We detected significant differences in hybridization rates between the orchard types in both investigated years (P < 0.001, analysis of variance, ANOVA). This finding suggests that SMC interspecific seed orchards can reliably provide seeds with high proportions of hybrids. In the traditional interspecific seed orchard, there were significant differences in the proportions of hybrids among L. gmelinii var. japonica seeds between the two years (P < 0.005, ANOVA), which may have been partly due to differences in the relative amounts of pollen cones produced by L. gmelinii var. japonica and L. kaempferi.  相似文献

Eighteen microsatellite markers were developed from Rosa odorata var. gigantea (Rosaceae), including 11 new microsatellite markers and 7 modified microsatellite loci having been developed from other Rosa species. About 27 wild individuals from 3 populations were used to screen polymorphism of these 18 microsatellite makers. The average allele number of these markers was 3.9 per locus, ranging from 2 to 9. The expected and observed heterozygosities varied from 0.2711 to 0.8043 and from 0.0370 to 0.5556, respectively. Cross-species amplification in other eight Rosa species showed their potential use for congeneric species. These microsatellite primers will be used for population genetics studies, constructing genetic linkage maps or locating quantitative trait locus (QTL) of R. odorata var. gigantea and related species.  相似文献
