An allozymic study of three wild species of Antirrhinum L. A. lopesiamum Rothm., A. mollissimum Roihni. and A. microphyllum Rothm. is described. All are members of subsection Kickiella Rothm., and are narrow-range endemics of the Iberian Peninsula. The variability of the different loci, as well as the number and mobility of the alleles, differ among the three species, a demonstration of the usefulness of allozymes for the systematics of the genus. The finding of alleles unique to each species indicates high divergence among species suggesting ancient diversification, and supports the hypothesis of a geographical model of speciation. All three species show high levels of within-species variability, mainly partitioned within populations, while between populations genetic differentiation is low. Correlations between population size, sample size and genetic variability, and the usefulness of allozymic data for conservation purposes, are discussed.  相似文献

Two new N-glycosidase type-1 ribosome-inactivating proteins (RIPs), denoted petroglaucin 1 and petrograndin, respectively, were isolated from the plantsPetrocoptis glaucifolia (Lag.) Boiss sp.viscosa (Rothm.) Laínz andPetrocoptis grandiflora Rothm. These new RIPs do not share H2N-terminal amino-acid sequence homology with petroglaucin (now denoted as petroglaucin 2), the only other type-1 RIP to be isolated fromP. glaucifolia (Arias et al. (1992) Planta186, 532–540). Petroglaucin 1 shares amino-acid sequence homology with RIPs from Cucurbitaceae while petroglaucin 2 and petrograndin do so with saporins and dianthin 30 (Caryophyllaceae). The new RIPs strongly inhibited protein synthesis at subnanomolar concentrations in rabbit reticulocyte lysates and other eukaryotic cell-free systems, but they were inactive on bacterial ribosomes.  相似文献

Allozyme diversity was studied within and among populations of five related taxa of Antirrhinum L. endemic to the Iberian Peninsula ( A. graniticum Rothm. ssp. graniticum , ssp. brachycalyx Sutton and ssp. ambiguum (Lange) Mateu & Segarra, A. boissieri Rothm. and A. onubensis (Fdez. Casas) Fdez. Casas). All of the studied taxa are obligate outcrossing endemic perennial herbs which form isolated populations. However, the taxa vary in range and population sizes, and are found on different soil types. The level and distribution of allozyme diversity differed widely between taxa: A. graniticum ssp. brachycalyx had the lowest level of allozyme diversity (HT = 0.09), whilst the highest level was detected in A. boissieri (HT = 0.25). Total variation was partitioned into within- and among-population variation. The proportion attributable to variation within populations varied from about 67% up to 84.3% and 89.5% in A. graniticum ssp. brachycalyx and A. graniticum ssp. ambiguum , respectively. Both these subspecies also showed little population divergence (GST = 0.10 and 0.09, respectively) and had high levels of estimated gene flow (Nm = 2.18 and 2.62, respectively). These results are discussed in relation to geographical proximity of populations and habitat continuity. Isolation by distance was not detected in any of the studied taxa. This result suggests that divergence among populations is due to random genetic drift.  © 2003 The Linnean Society of London . Biological Journal of the Linnean Society , 2003, 79 , 299–307.  相似文献

Petrocoptis grandiflora Rothm. is a species endemic to the Iberian Peninsula with restricted distribution. We studied various aspects of its pollination ecology, nectar characteristics and standing crop, pollinators and nectar-robbers, and the effects of nectar robbing and supplementary pollination on fruit set. The nectar is moderately rich in saccharose (saccharose/hexose ratio 0.54), and is largely consumed by long-tongued bees (Anthophoridae). The incidence of nectar robbing by Xylocopa violncea differed between the four plant populations studied, and it had significant effects on fruit set when the population was simultaneously considered. Supplementary pollination did not improve fruit set significantly.  相似文献

Cytotaxonomic investigations of the autumn-flowering squills, Prospero autumnale (L.) Speta ≡ Scilla autumnalis L., Prospero obtusifolium (Poir.) Speta ≡ Scilla obtusifolia Poir., Barnardia numidica (Poir.) Speta ≡ Scilla numidica Poir., and Hyacinthoides lingulata (Poir.) Rothm. ≡ Scilla lingulata Poir. were performed in 20 populations from northern Algeria located between Tipasa and La Vieille Calle. Various chromosome numbers were found, including a new cytotype, 2n = 8, for the flora of Algeria, concerning plants identified as Prospero obtusifolium (Poir.) Speta [including P. fallax (Steinh.) Speta = S. autumnalis L. ssp. fallax (Steinh.) Batt.]. The numbers 2n = 14, 28, and 42 correspond, respectively, to diploid, tetraploid, and hexaploid levels of P. autumnale s.l. [including P. pulchellum (Munby) Speta ≡ Scilla pulchella Munby = S. autumnalis var. pulchella (Munby) Batt.], with x = 7. The cytotypes of Barnardia numidica (Poir.) Speta with 2n = 18 and Hyacinthoides lingulata (Poir.) Rothm. with 2n = 16 chromosomes were confirmed.  相似文献

Abstract

Antirrhinum lopesianum Rothm. is a narrow endemic of the Lusitan Duriensean biogeographical sector (central western Spain and north-eastern Portugal). The species is listed as threatened in several Spanish documents, although it does not figure as such in any Portuguese document. This paper provides a detailed study of its distribution, estimates of the sizes of its populations, the threats it faces, and its current conservation status. The total number of individuals thought to exist is only 768, distributed along the valley of the River Duero on the Spanish – Portuguese border (562, 71.2%), and in the Portuguese Sabor River valley (206, 26.8%). The main threat to the species is loss of habitat: about one third of the Iberian populations can be considered threatened; one population containing 37.6% of all these plants (289) is severely threatened. To determine the Area of Occupancy and the Extent of Occurrence, an exhaustive bibliographical survey was carried out, and herbarium specimens deposited in several institutions were revised. It is, therefore, classifiable as Critically Endangered in Portugal and Endangered in Spain.  相似文献

BACKGROUND AND AIMS: Fifty-two populations were sampled in order to establish the taxonomic delimitation and relationships of eight taxa belonging to the A. majus L. and A. siculum Miller groups. METHODS: Data on 13 allozyme loci were recorded after extraction of fresh leaves and electrophoresis on horizontal 10% starch gels. KEY RESULTS: Genetic distances between conspecific populations are lower than for other species of the genus. CONCLUSIONS: These results support the recognition of A. majus, A. tortuosum, A. linkianum, A. cirrigherum, A. litigiosum and A. barrelieri at specific rank. The genetic distances, together with the lack of morphological differences and the sympatric distribution ranges, support the inclusion of A. australe into A. tortuosum, A. dielsianum into A. siculum, and A. latifolium subsp. intermedium as a synonym of A. latifolium. The results support separation of the taxa studied into two groups, coinciding with series Sicula Rothm. and Majora, but disagreeing with the arrangement of species into them. According to our results, Sicula consist of A. siculum and Majora consists of A. latifolium, A. majus, A. tortuosum, A. linkianum, A. cirrigherum, A. litigiosum and A. barrelieri.  相似文献

Random amplified polymorphic DNA (RAPD) markers were used to study the spatial genetic structure in two populations (Bolarque and Entrepe?as) of endangered cliff specialist Antirrhinum microphyllum Rothm. (Scrophulariaceae). Mantel tests found no significant linear correlations between geographic and genetic data. However, redundancy analysis (RDA) models developed using the spatial data as the constraining matrix were highly significant, and spatial data explained 13.6% and 11.1% of total genetic variation in Bolarque and Entrepe?as, respectively. Moran's I correlograms and Mantel correlograms revealed a positive autocorrelation in the first distance class (15 m), which suggests a patchy distribution of genetic diversity. This distribution is consistent with the genetic vicinities that are expected from the territorial behavior of main pollinator Rhodanthidium sticticum (Megachilidae), the predominant short-distance seed dispersal, and the patchy spatial distribution of available safe sites. The gradient pattern obtained in Entrepe?as was consistent with standard isolation-by-distance models. However, a differential sinusoidal pattern was obtained in Bolarque, which would indicate a more frequent gene flow between patches and might be due to lower plant density there. The spatial genetic structure coexists with a strict self-incompatibility system in the species. Simplified RDA models obtained using a stepwise forward selection comprised the easting component in Entrepe?as and the easting and northing components in Bolarque. Similar results were obtained with directional correlograms. These differential patterns can be explained by the distinct spatial arrangement of the populations (linear and bidimensional in Entrepe?as and Bolarque, respectively).  相似文献

A multivariate analysis has been performed in order to study the polymorphisms of the three species ofPotentilla sect.Recta (Th. Wolf)Juzepchuk represented in the Iberian Peninsula:P. asturica Rothm.,P. hirta L. andP. recta L. The variability of 20 characters (quantitative, binary and qualitative) has been evaluated in 76 collections mostly from the Iberian Peninsula and from other European territories, but also from North America, and North Africa. The analyses performed provide support for the recognition of the three species. The most discriminant among the quantitative characters is the width of the long trichomes on the stem, and then the width of the long trichomes on the leaves and the maximum length of the long trichomes. The interval of number of carpels, presence or absence of short eglandular trichomes and the abundance of long eglandular trichomes (>3 mm) are the most discriminant ones among the qualitative ones. The taxonomical position ofP. asturica, which has been previously subordinated as a subspecies within the other two taxa, is discussed. Our results support a good delimitation of this taxon which, on the basis of the morphological characters studied by us, seems to be morphologically more close toP. hirta than toP. recta, but differs significantly from both. A key to the three species is provided.  相似文献

The current study was designed to evaluate the antioxidant, anti‐inflammatory and antimicrobial activities of Alchemilla mollis (Buser ) Rothm . (Rosaceae) aerial parts extracts. Chemical composition was analyzed by spectrophotometric and chromatographic (HPLC) techniques. The antioxidant properties assessed included DPPH· and ABTS·+ radical scavenging, β‐carotene‐linoleic acid co‐oxidation assay. Antimicrobial activity was evaluated with disc diffusion and micro dilution method. In order to evaluate toxicity of the extracts, with the sulforhodamine B colorimetric assay L929 cell line (mouse fibroblast) was used. The anti‐inflammatory activities of the potent antioxidant extracts (methanol, 70% methanol, and water extracts) were determined by measuring the inhibitory effects on NO production and pro‐inflammatory cytokine TNF‐α levels in lipopolysaccharide stimulated RAW 264.7 cells. 70% methanol and water extracts which were found to be rich in phenolic compounds (184.79 and 172.60 mg GAE/g extract) showed higher antioxidant activity. Luteolin‐7‐O‐glucoside was the main compound in the extracts. Ethyl acetate and 70% methanol extracts showed higher antibacterial activity against Staphylococcus aureus and Salmonella enteritidis with MIC value of 125 μg/ml. 70% methanol extract potentially inhibited the NO and TNF‐α production (18.43 μm and 1556.22 pg/ml, respectively, 6 h).  相似文献

Background and aims

Microbe-assisted phytoremediation is particularly effective for organic pollutants. The leguminous shrub Cytisus striatus (Hill) Rothm. has been proposed as a candidate species for the rhizoremediation of hexachlorocyclohexane (HCH)-contaminated sites. The aim of this study was to improve the performance of this species using microbial inoculants.

Methods

C. striatus was grown in substrates contaminated with 0, 10 and 35 mg HCH kg?1 for 8 weeks. Plants were either not inoculated (NI), or inoculated with the endophyte Rhodococcus erythropolis ET54b and the HCH-degrader Sphingomonas sp. D4 (isolated from a HCH-contaminated soil) on their own or in combination (ET, D4 and ETD4).

Results

Inoculation with both bacterial strains (ETD4) resulted in decreased HCH phytotoxicity and improved plant growth. HCH-exposed plants inoculated with ETD4 presented a 120–160 % increase in root, and 140–160 % increase in shoot biomass, and led to a decrease in the activities of enzymes involved in anti-oxidative defence. APOD activity was reduced by up to 37 % in shoot tissues and 25 % in root tissues, and corresponding activities of SOD were reduced by up to 35 % and 30 %. HCH dissipation was enhanced in the presence of C. striatus but no significant effect of microbial inoculants was observed.

Conclusions

Inoculating C. striatus with this combination of bacterial strains is a promising approach for the remediation of HCH-contaminated sites.  相似文献

The organochlorine 1,2,3,4,5,6 hexachlorocyclohexane (HCH) is a broad-spectrum insecticide that was used on a large-scale worldwide. The soil–plant–microbe system and its influence on HCH biodegradation are evaluated. A greenhouse experiment was designed to evaluate HCH dissipation and several microbial parameters among rhizosphere and bulk soil of two contrasting plants, Cytisus striatus (Hill) Rothm and Holcus lanatus L. Plants were grown for 180 days in three treatments: uncontaminated soil (control), uncontaminated soil inoculated with soil (3% w/w) from a HCH-contaminated site (INOC), and uncontaminated soil inoculated with soil (3% w/w) from the HCH-contaminated site and artificially contaminated to obtain 100 mg HCH kg−1 dry soil (100HCH-INOC). At harvest, plant biomass, soil water-extractable organic C, pH and Cl concentration, rhizosphere microbial densities (total heterotrophs, ammonifiers, amylolytics) and C substrate utilization patterns, and degradation of α-, β-, δ- and γ-HCH isomers were determined in bulk and rhizosphere soils. Soil solution Cl concentration was determined every 30 days throughout the entire growth period. Results demonstrate that both Cytisus striatus and Holcus lanatus can grow in soils with up to 100 mg HCH kg−1. An enhanced degradation of α-HCH, but not β- or δ-HCH, was observed in the rhizosphere. Significant changes in the microbial densities were observed between bulk and rhizosphere soils of Cytisus, and an increase in C source utilization indicated changes in community level physiological profiles (CLPP) in the rhizosphere of this species when grown in contaminated soils. HCH dissipation was also greater in soils planted with this species. In accordance, increases in soil extractable C, Cl concentration and acidity were greater at the rhizosphere of Cytisus. Concentration of Cl in soil solutions also indicates greater HCH dechlorination in soils planted with Cytisus than Holcus. Results suggest that phytostimulation of bacteria present or added to soil is a promising approach to cleaning HCH-contaminated sites, and especially for biodegradation of α-HCH.  相似文献
