Abstract A rapid and simple method has been developed for the simultaneous measurement of juvenile hormone (JH) and JH acid synthesized in vitro by larval corpora allata (CA) of the tobacco hornworm, Manduca sexta. An organic solvent partition of incubation medium efficiently separates JH acid from JH, and a radioimmunoassay which recognizes the two moieties equivalently is then employed to quantify each. The change in the biosynthetic product of the CA from JH to JH acid appears to begin slowly at the time of ecdysis to the last (fifth) larval stadium and is not complete until just prior to wandering (day 4). The inclusion of the JH esterase inhibitor S-benzoyl-O-ethyl phosphoramidothiolate in incubations of corpora allata revealed that the activity of JH esterases from the gland parallels gland activity and that significant hydrolysis of newly synthesized JH by these esterases occurs in incubations of glands taken at the beginnings of the fourth and fifth larval stadia. An allatostatin, which is proposed to inhibit the corpus allatum during the time of the change in its product, inhibits both JH I and JH I acid synthesis. 相似文献

We determined the changes in hemocyte titer and in the abundance of hemocyte types of the tobacco hornworm Manduca sexta during the fourth and fifth larval stadium and the beginning of the pupal stadium. As we analyzed the samples of individual insects at daily intervals, we were able to correlate phenotypical features, body weight, as well as total protein content and lysozyme activity in the hemolymph with the observations on hemocytes. In the course of the fifth larval stadium, the hemocyte titer decreased slightly and declined further after pupation. Using calculated values for total hemocyte numbers, females had about five times and males three times more hemocytes in the circulating population at the beginning of the wandering stage (in the middle of the fifth larval stadium) than immediately after the last larval--larval molt (from the fourth to the fifth larval stadium). This sexual difference was mainly due to an increase in the number of plasmatocytes, which was more prominent in females than in males. Granular cells were dominant in early fifth larval stadium while plasmatocytes were the most abundant cells in pupae. Oenocytoids and spherule cells disappeared during the wandering stage. Lysozyme activity in the hemolymph rose to a maximum during the wandering stage, with females having lysozyme values twice as high as those for males. These changes in lysozyme activity, however, did not correlate with the increase of total hemolymph protein titer which occurred already at the beginning of the wandering stage. We postulate that changes in hemocyte titers are under direct hormonal control, which has to be proven in future experiments. 相似文献

ABSTRACT. Feeding behaviour of fifth instar tobacco hornworm caterpillars, Manduca sexta (Johansen) (Lepidoptera; Sphingidae), eating tobacco or artificial diet, is quantitatively described. The insects grow at the same rate on both foods. There is no daily rhythm of feeding behaviour. For most insects, feeding on either food occurs in bouts with the lengths of interfeed gaps and of feeding bouts appearing to be distributed randomly. However, in many insects there is a strong correlation between the length of a feeding period and that of the preceding non-feeding period.
The proportion of time spent feeding on tobacco is much greater than on artificial diet. On tobacco, feeding periods are separated by shorter interfeed gaps than on the artificial diet, while the rate of bout initiation is similar on either food.
On both tobacco and artificial diet, the proportion of time spent feeding increases as the fifth stadium proceeds. This is due to both longer feeding bouts and shorter gaps. The rate of food acquisition within bouts does not change during the stadium. 相似文献

Midgut mitochondria from fifth larval instar Manduca sexta exhibited a transhydrogenase that catalyzes the following reversible reaction: NADPH + NAD(+) <--> NADP(+) + NADH. The NADPH-forming transhydrogenation occurred as a nonenergy- and energy-linked activity. Energy for the latter was derived from the electron transport-dependent utilization of NADH or succinate, or from Mg++-dependent ATP hydrolysis by ATPase. The NADH-forming and all of the NADPH-forming reactions appeared optimal at pH 7.5, were stable to prolonged dialysis, and displayed thermal lability. N,N'-dicyclohexylcarbodiimide (DCCD) inhibited the NADPH --> NAD(+) and energy-linked NADH --> NADP(+) transhydrogenations, but not the nonenergy-linked NADH --> NADP(+) reaction. Oligomycin only inhibited the ATP-dependent energy-linked activity. The NADH-forming, nonenergy-linked NADPH-forming, and the energy-linked NADPH-forming activities were membrane-associated in M. sexta mitochondria. This is the first demonstration of the reversibility of the M. sexta mitochondrial transhydrogenase and, more importantly, the occurrence of nonenergy-linked and energy-linked NADH --> NADP(+) transhydrogenations. The potential relationship of the transhydrogenase to the mitochondrial, NADPH-utilizing ecdysone-20 monooxygenase of M. sexta is considered. 相似文献

Abstract. Fifth stadium larvae of the tobacco hornworm, Manduca sexta (L.), ate larger meals than usual when they had been deprived of food for periods of time longer than the usual intermeal interval (c. 45 min). Meal size increased with time since the last meal until 180 min, when it was about 3 times normal. There was no evidence of a role for volumetric feedback from the gut in controlling meal size. Injections of a paraffin oil/wax mixture, or of petroleum jelly (Vaseline) into the foregut, midgut or rectum failed to decrease meal size. Cutting the recurrent nerve failed to alter meal size compared to sham-operated controls (although both groups took smaller meals than unoperated controls). By contrast, injections of an extract of soluble nutrients from the diet into the midgut inhibited feeding in some insects and reduced subsequent meal size in others. Appropriate controls showed that these effects were not due to the volumetric or osmotic effects of the injections. These results imply that nutrient feedback plays an important role in controlling meal size in Manduca caterpillars, while volumetric feedback is probably unimportant. 相似文献

The levels of three alternatively spliced mRNAs from the Manduca sexta allatotropin (Manse-AT) gene were determined following physiological manipulations during the larval, pupal and adult stages; starvation of larvae, induction of pupal diapause and adult mating experience. The juvenile hormone biosynthetic activity of the corpora allata (CA) was also determined in starved larvae and in mated and unmated females. Starvation of early fifth instar larvae specifically increased the amount of one Manse-AT mRNA that is predicted to encode Manse-AT and two related peptides, Manse-ATL-I and -II. The normal rapid decrease in the activity of the CA in last instar larvae was not observed in starved insects which maintained a relatively high rate of JH biosynthesis for at least 3 days. Diapause induction resulted in a small increase in one Manse-AT mRNA, but levels were much lower compared to those observed in larvae or adults. During the first 4 days of adult life, Manse-AT mRNA levels were not changed as a result of mating. However, in mated females, the rate of JH biosynthesis gradually increased, in sharp contrast to the relatively low level of CA activity seen in virgin females. These observations suggest the elevated activity of the CA in mated females is not simply due to the increased level of Manse-AT mRNA. 相似文献

Nodule formation is the predominant insect cellular defense reaction to bacterial challenges, responsible for clearing the largest proportion of infecting bacteria from hemolymph circulation. Hemocyte spreading behavior is a critical step in the nodulation process. It has been suggested that eicosanoids mediate several steps in the process. However, the influence of eicosanoids on hemocyte spreading has not been investigated in detail. To test the hypothesis that eicosanoids mediate hemocyte spreading behavior, I treated larvae of the tobacco hornworm, Manduca sexta, with eicosanoid biosynthesis inhibitors and later assessed plasmatocyte elongation on glass slides. Plasmatocytes from larvae treated with dexamethasone did not elongate to the extent of plasmatocytes from untreated control larvae. The dexamethasone effect on plasmatocyte elongation was expressed in a dose-dependent manner and was reversed by injecting dexamethasone-treated larvae with the eicosanoid-precursor fatty acid, arachidonic acid. Palmitic acid, which is not substrate for eicosanoid biosynthesis, did not reverse the influence of dexamethasone on plasmatocyte elongation. Finally, plasmatocytes from larvae treated with a range of eicosanoid biosynthesis inhibitors did not elongate to the extent of plasmatocytes from control larvae. Plasmatocyte width did not appear to be influenced in this study. These findings strongly support the idea that insect plasmatocyte elongation is influenced by eicosanoids. 相似文献

The reversible, membrane-associated transhydrogenase that catalyzes hydride-ion transfer between NADP(H) and NAD(H) was evaluated and compared to the corresponding NADH oxidase and succinate dehydrogenase activities in midgut and fat body mitochondria from fifth larval instar Manduca sexta. The developmentally significant NADPH-forming transhydrogenation occurs as a nonenergy- or energy-linked activity with energy for the latter derived from either electron transport-dependent NADH or succinate utilization, or ATP hydrolysis by Mg++-dependent ATPase. In general, the plant flavonoids examined (chyrsin, juglone, morine, quercetin, and myricetin) affected all reactions in a dose-dependent fashion. Differences in the responses to the flavonoids were apparent, with the most notable being inhibition of midgut, but stimulation of fat body transhydrogenase by morin, and myricetin as also noted for NADH oxidase and succinate dehydrogenase. Although quercetin inhibited or stimulated transhydrogenase activity depending on the origin of mitochondria, it was without effect on either midgut or fat body NADH oxidase or succinate dehydrogenase. Observed sonication-dependent increases in flavonoid inhibition may well reflect an alteration in membrane configuration, resulting in increased exposure of the enzyme systems to the flavonoids. The effects of flavonoids on the transhydrogenation, NADH oxidase, and succinate dehydrogenase reactions suggest that compounds of this nature may prove valuable in the control of insect populations by affecting these mitochondrial enzyme components. 相似文献

The microstructure of the feeding activity of tobacco hornworm caterpillars (Manduca sexta Johansson) on tomato leaf was examined by means of an automated cafeteria. In this device each activity of the caterpillar generates a characteristic slow electrical change which can be recorded. The apparatus is therefore both accurate and sensitive. Examination of the activity records indicated that larger animals ate more than smaller ones by increasing both bite frequency and the lengths of meals. Meal frequency did not increase. Correlations amongst a variety of measures indicated that there was regulation of feeding both between and within meals. 相似文献

Abstract .Temperature and the protein content of food affect rates of consumption and growth in herbivorous insects in different ways: reduced temperature typically reduces both consumption and growth rates, whereas reduced dietary protein typically increases consumption rate but either reduces or has no effect on growth rate. The interactions between temperature and dietary protein concentration in affecting consumption, growth and efficiency in fifth-instar caterpillars of Manduca sexta were studied, using both short-term (4 h) and long-term (duration of fifth stadium) experiments. The short-term experiments examined constant temperatures between 14 and 42°C, whereas the long-term experiments examined constant temperatures between 18 and 34°C; both experiments considered two levels of dietary protein. In both experiments, caterpillars had significantly higher consumption and frass production rates on low-protein compared with high-protein diets at each test temperature between 18 and 34°C, thereby compensating for the lower diet quality. In contrast, at more extreme temperatures (14 and 42°C) in the short-term studies, consumption and frass production rates were lower on low-protein compared with high-protein diets. As a result, there were substantial interactions between temperature and dietary protein for consumption and frass production rates in the short-term experiments, but not in the long-term experiments. These results suggest that interactions between temperature and dietary protein may emerge because of the failure of compensatory feeding responses at low and high temperatures. It is hypothesized that the failure of compensatory responses is more likely to occur under diurnally fluctuating temperatures than under a constant temperature with the same mean, and it is proposed that interactions between temperature and dietary protein for consumption are relevant to M. sexta and other caterpillars that experience wide diurnal fluctuations in temperature in the field. 相似文献

The correlation between triacylglycerols containing conjugated diene fatty acyl moieties and pheromone aldehydes in the sex pheromone glands of females of Manduca sexta was investigated. Females decapitated 15 h after adult emergence neither called nor produced pheromone during the natural period of pheromone production on the subsequent two nights. However, these females could be stimulated to produce sex pheromone for prolonged periods by repeated injection of synthetic pheromone biosynthesis activating neuropeptide (PBAN). Gas chromatographic analysis of methanolysis products of lipids extracted from the pheromone glands of decapitated and intact females showed no differences in the amounts of fatty acyl precursors of pheromone. High performance liquid chromatographic analysis of the triacylglycerols containing conjugated diene analogues of the pheromone components (diene TG), obtained 24 and 48 h after decapitation, showed that the total amounts of these components were not affected by decapitation. The amounts of all diene TG peaks declined significantly when decapitated females were stimulated to produce pheromone during a 7 h period by repeated injection of PBAN at 3 h intervals but recovered when pheromone production subsided. A new class of inhibitors of juvenile hormone epoxide hydrolase (JHEH) of Manduca sexta and further in vitro characterization of the enzyme are reported. The compounds are based on urea and amide pharmacophores that were previously demonstrated as effective inhibitors of mammalian soluble and microsomal epoxide hydrolases. The best inhibitors against JHEH activity so far within this class are N-[(Z)-9-octadecenyl]-N′-propylurea and N-hexadecyl-N′-propylurea, which inhibited hydrolysis of a surrogate substrate (t-DPPO) with an IC50 around 90 nM. The importance of substitution number and type was investigated and results indicated that N, N′-disubstitution with asymmetric alkyl groups was favored. Potencies of pharmacophores decreased as follows: amide>urea>carbamate>carbodiimide>thiourea and thiocarbamate for N, N′-disubstituted compounds with symmetric substituents, and urea>amide>carbamate for compounds with asymmetric N, N′-substituents. JHEH hydrolyzes t-DPPO with a Km of 65.6 μM and a Vmax of 59 nmol min−1 mg−1 and has a substantially lower Km of 3.6 μM and higher Vmax of 322 nmol min−1 mg−1 for JH III. Although none of these compounds were potent inhibitors of hydrolysis of JH III by JHEH, they are the first leads toward inhibitors of JHEH that are not potentially subject to metabolism through epoxide degradation. 相似文献

ABSTRACT. The contributions of olfactory and gustatory organs in food plant discrimination were examined in larvae of Manduca sexta (Johan.) (Lepidoptera, Sphingidae). Larvae, from which various chemosensory organs had been removed surgically, were tested for feeding preferences for a host, tomato ( Lycopersicon esculentum ); a weakly acceptable non-host, rape ( Brassica napus ); and an unacceptable non-host canna ( Canna generalis ), using a two-choice disc bioassay.
Removal of all known chemosensory organs resulted in failure to show discriminatory behaviour in a strictly chemosensory bioassay, indicating that all external chemosensory organs have been accounted for. The involvement of non-chemosensory organs results in residual discrimination for leaves by individuals with total chemosensory ablations.
Larvae possessing either olfactory or gustatory organs still exhibit normal preferences for tomato over rape. Gustatory (but not olfactory) organs are required for larvae to show normal preferences for tomato over canna; in fact, olfactory organs do not appear to participate in this decision.
To examine which if any of the plant species is being selected in two-choice tests, larvae were given a choice between each leaf species and a 'neutral' substance (wet filter paper). Both olfactory and gustatory organs are required for normal preferences for tomato, but either alone will suffice for rape. Only gustation is needed to select canna, and participation of either the epipharyngeal sensilla or a single medial sensillum styloconicum is sufficient to elicit complete rejection behaviour.
We conclude that, in larvae of M. sexta , the complement of chemosensory organs needed for food plant discrimination varies with the plant species sampled. An in vitro system for the uptake of 125l-vitellogenin (VG) or vitellin into isolated follicles of the tobacco hornworm, Manduca sexta, is described. After incubation with 125l-VG, follicles were disrupted and the internal yolk contents separated from the follicle membranes. The results showed that 125l-VG was associated principally with the membranes (92%) after incubation at 4°C. However, at 27°C, 125l-VG was mainly in the yolk (92%). Furthermore, trypsin treatment removed approximately 70% of VG bound to the follicles at 4°C. Labeled VG was shown to bind to sonicated follicle membranes with high specificity and affinity (KD ? 1.3 × 10?8 M). This binding was sensitive to pH and calcium concentration. The total binding sites were estimated at 4 × 1014 sites/g of membrane protein. Competition studies showed that binding of 125l-VG to follicle membranes was blocked by excess unlabeled vitellin and deglycosylated vitellogenin but not by lipophorin (the major hemolymph lipoprotein), microvitellogenin, a female-specific protein (Mr ~ 31,000) found in both hemolymph and eggs, and the smaller vitellogenin subunit, apovitellogenin-II (Mr ~ 45,000). These results suggest that selective uptake of M. sexta VG from the hemolymph involves binding to specific receptors located on the follicle membranes. 相似文献

Triglycerides (TG) stores build up in the insect fat body as lipid droplets at times of excess of food. The mobilization of fat body triglyceride (TG) is stimulated by adipokinetic hormones (AKH). The action of AKH involves a rapid activation of cAMP-dependent protein kinase (PKA). Recent in vitro studies have shown that PKA phosphorylates and activates the TG-lipase substrate, the lipid droplets. Conversely, purified TG-lipase from Manduca sexta fat body is phosphorylated by PKA in vitro but is not activated. This study was directed to learn whether or not AKH promotes a change in the state of phosphorylation of the lipase in vivo, and what are the relative contributions of cytosol and lipid droplets to the overall increase of lipolysis triggered by AKH. TG-lipase activity of fat body cytosols isolated from control and AKH-treated insects was determined against the native substrate, in vivo [3H]-TG radiolabeled lipid droplets, obtained from control and AKH-treated insects. The lipase activity of the system composed of AKH-cytosol and AKH-lipid droplets (11.1 +/- 2.1 nmol TG/min-mg) was 3.1-fold higher than that determined with control cytosol and lipid droplets (3.6 +/- 0.5 nmol TG/min-mg). Evaluation of the role of AKH-induced changes in the lipid droplets on lipolysis showed that changes in the lipid droplets are responsible for 70% of the lipolytic response to AKH. The remaining 30% appears to be due to AKH-dependent changes in the cytosol. However, the phosphorylation level of the TG-lipase was unchanged by AKH, indicating that phosphorylation of the TG-lipase plays no role in the activation of lipolysis induced by AKH. 相似文献

Two major pathways of programmed cell death (PCD)--the apoptotic and the autophagic cell death--were investigated in the decomposition process of the larval fat body during the 5th larval stage of Manduca sexta. Several basic aspects of apoptotic and autophagic cell death were analyzed by morphological and biochemical methods in order to disclose whether these mechanisms do have shared common regulatory steps. Morphological examination revealed the definite autophagic wave started on day 4 followed by DNA fragmentation as demonstrated by agarose gel electrophoresis and TUNEL assay. By the end of the wandering period the cells were filled with autophagic vacuoles and protein granules of heterophagic origin and the vast majority of the nuclei were TUNEL-positive. No evidence was found of other aspects of apoptosis, e.g. activation of executioner caspases. Close correlation was disclosed between the onset of autophagy and the nuclear accumulation of the ubiquitin-proteasome system. 相似文献

Both allatotropic and allatostatic activities were found in crude extracts of brain from adult and larval Eri silkworm, Samia cynthia ricini, but it seems that allatotropic activity dominates in each stage. There was a high level of allatotropic activity in the crude extract of brain from newly emerged female adults, but allatostatic activity appeared in the bioassay when excessive amounts of crude extracts of brain were added. Crude extracts of brain from premoulting fourth‐instar larvae and from newly ecdysed fifth‐instar larvae exhibited allatotropic activities, whereas extracts of brain from the second and third day of the fifth‐instar larvae inhibited juvenile hormone (JH) release slightly. Allatotropic activity from the brains of adults and larvae stimulated both adult and larval corpora allata (CA) to synthesize JH. Manduca sexta allatotropin (AT) (Mas‐AT) and M. sexta allatostatin (AST) (Mas‐AST) also stimulated and inhibited both adult and larval S. cynthia ricini CA to synthesize JH, respectively. Higher concentrations of Mas‐AT (10?4 or 10?3 M) showed an inhibitory effect on adult CA. CA from newly emerged female adults were the most sensitive to inhibition by Mas‐AST, whereas CA from female pharate adults at about 6 h before adult emergence were the most sensitive to stimulation by Mas‐AT and S. cynthia ricini brain allatotropic activity. An extract of brain and Mas‐AT induced some of the non‐active female pharate adult CA at 12 h before emergence to synthesize a small amount of JH. 相似文献
