Crossing experiments were carried out between the stiped-egg form collected from Sapporo (43°04N) and Machida (35°33N), and the stipeless-egg form ofPanonychus akitanus Ehara from Tomakomai (42°37N) and Imagane (42°34N). Intra-form crosses gave a high proportion of female progeny, but inter-form crosses only male progeny, suggesting that fertilization did not take place. The number of eggs laid during the first 10 days of the oviposition period was greater in intra-form crosses than in inter-form crosses (P<0.001). The latter, except for the cross between the Sapporo and the Imagane populations, had similar egg production to virgin females (P<0.05).Mated females showed a greater daily oviposition rate than virgin females. However, the latter drastically increased their oviposition rate after mating with their sons. The two forms are thus reproductively isolated.  相似文献

The gap dynamics of two Nothofagus pumilio (lenga) stands have been investigated. We evaluated and compared tree diameter distributions, spatial patterns, tree fall and gap characteristics and regeneration responses in gaps in two old-growth forests of Nothofagus pumilio in Southern Chile (Shangri-La: 36°60 S, 71°30 W; Reserva Coyhaique: 45°52 S, 72°00 W). In addition, we describe relationships between gap size and regeneration characteristics. We detected some differences in tree and sapling densities between sites. The causes of gap formation and the gap size distribution, mean gap area, and gap fraction were similar, but gap abundance was different at the two sites. The Reserva Coyhaique site had 15 gaps/ha compared to 10 gaps/ha for Shangri-La. Sizes of clumps of trees were within the range of sizes of canopy gaps at both sites. The density of saplings was higher in gaps than under closed forest at R. Coyhaique, but not at Shangri-La. We found that sapling densities were unrelated to gap size in both sites. The lower sapling density in gaps at Shangri-La might be explained by the presence of Chusquea quila, a competitive pioneer bamboo species. The height increment was related to gap size at Shangri-La, but not at R. Coyhaique. Gap size itself does not account for all the variation in recruitment performance in these Southern beech stands.  相似文献

The degradation of the phenylcoumaran substructure model compound methyl dehydrodiconiferyl alcohol by the white-rot wood decay fungus Phanerochaete chrysosporium was investigated using culture conditions optimized for lignin oxidation. Initial attack was in the cinnamyl alcohol side chain, which was oxidized to a glycerol structure. This was subsequently converted by loss of the two terminal carbon atoms, C and C, to yield a C-aldehyde structure, which was further oxidized to the C-acid compound. The next detected intermediate, a phenylcoumarone, was produced by double bond formation between C and C, and oxidation of the C-alcohol to an aldehyde group. Further oxidation of C to an acid yielded the next intermediate. The final identified degradation product was veratric acid. No products from the 5-substituted aromatic ring, and no phenolic products, were found. Summary The three haploid yeast strains T2tmp1-3, T2tmp1-1, and T6tmp1-51 auxotrophic for 5-dTMP differ in their requirement for thymidylate: 72, 16, and 3 g 5-dTMP/ml will restore optimal growth, respectively. Thymidylate low requirement in strain T2tmp1-1 and T6tmp1-51 is termed tlrA and tlrC, respectively. When the growth medium is made 5x10-4 M for 5-dTMP only strain T6tmp1-51 is severely inhibited in RNA and DNA synthesis. This inhibition is reversible after removal of excessive 5-dTMP. The inhibitory characteristic is in marked contrast to thymineless death due to the lack of 5-dTMP in strain T6tmp1-51 where only DNA synthesis stops while RNA synthesis continues. The inhibitory effect of 5x10-4 M 5-dTMP is not due to the 5-dTMP auxotrophy but to the thymidylate low requiring character (tlrC) in strain T6tmp1-51. The arrest of RNA and DNA synthesis by high concentrations of exogenous 5-dTMP suggests a regulatory role of either the monoor triphosphate on nucleoside or nucleotide biosynthesis in yeast.  相似文献

Both qualitative and quantitative differences in the minor nucleotide constituents of rRNA from normally grown and from etiolated wheat plants (Triticum aestivum L.) were established. Using different degradation methods and separation techniques the 18S+26S RNA of 8-day-old wheat seedlings grown in the light was found to contain 5-methylcytidine, 3-methylcytidine, 5-methyluridine, 3-methyluridine, 5-carboxymethyluridine, 1-methyladenine, N-methyladenine, 5-hydroxymethylcytidine, O2-methyluridine, O2-methylcytidine, pseudouridine, O2-methylpseudouridine, N2,N2-dimethylguanine, 1-methylguanine, ribothymidine and some unknown minor constituents. On the other hand, there were only a few minor nucleotides in the rRNA of etiolated wheat seedlings. Cycloheximide, a cytoplasmic protein synthesis inhibitor, simulated etiolation in that it reduced the number of minor nucleotides in rRNA, whereas chloramphenicol, a chloroplast protein synthesis inhibitor, had no significant effect on the minor nucleotide content of rRNA. Base-labile 6-chloro-2,3,5-tri-O-acetylpurine riboside (1c) and 2-amino-6-chloro-2,3,5-tri-O-acetylpurine riboside (1d) were fully deacetylated through Candida antarctica B lipase hydrolysis, affording respectively 6-chloropurine riboside (2c) and 2-amino-6-chloro-purine riboside (2d). Quantitative results were found at pH 7 and 60 °C in 24 h for 1c and 72 h for 1d. This mild and simple enzymatic technique represents a convenient procedure for the removal of acetyl groups from such base labile halogenated nucleosides.  相似文献

When tested in a poly(U)-dependent polyphenylalanine synthesizing system and in a postnuclear supernatant, both derived from Ehrlich ascites tumor cells, 2(3),5-ADP did not affect chain elongation of polypeptide synthesis. In a cell-free system which was dependent on initiation and programmed by natural mRNA, however, the amino acid incorporating activity was suppressed to about 10% of the control in the presence of 1 mM 2(3),5-ADP. The inhibitor was shown not to interfere with the attachment of poly(U) to the small ribosomal subunit and with the formation of mRNA-80S ribosome complexes in a complete protein synthesizing system. The subsequent attachment of a 40S ribosomal subunit to the mRNA-80S ribosome complex and the formation of polysomes, however, was depressed by the inhibitor. The experimental results suggest that 2(3),5-ADP inhibits initiation-dependent protein synthesis between monosome formation and the formation of the first peptide bond(s).  相似文献

Using a protease (at 100 g l–1) from Bacillus licheniformis, enzymatic acryloylation of sucrose (1 M) with vinyl acrylate (4 M) was carried out in anhydrous pyridine and yielded sucrose acrylate esters with more than 90% of sucrose converted in 24 h. After 5 days of reaction, the ester products consisted of 70% sucrose monoacrylate and 30% sucrose diacrylate. The monoester product was a sucrose 1-acrylate and the diester products consisted of sucrose 6,1-diacrylate and sucrose 6,1-diacrylate in the ratio of 3:2.  相似文献

Species of the genus Actinostola are known for high variability of features. Anatomy, histology and cnidae of type specimens of five species from South America and Antarctica originally described as members of Actinostola and one species of Stomphia were compared to specimens of Actinostola chilensis collected during this study. None of these traditionally used features clearly distinguish the examined Actinostola species. I therefore propose new distinctive taxonomic features, including in vivo and in situ data. I provide an emended diagnosis of the genus Actinostola and a revised list of its species. I accept the synonymy of A. excelsa, A. pergamentacea and A. intermedia with A. crassicornis, and reject the synonymy of A. chilensis with A. crassicornis and A. intermedia. I re-describe A. chilensis in detail, including in situ information. Specimens of A. chilensis inhabit exposed positions of rocky substrate from 22 m depth down in south Chilean fjords between Puerto Montt (41°3535S, 72°53W) and Puyuhuapi (44°3136S; 72°326W); the most conspicuous features are its relatively large size, bright-orange colour, smooth, tough column and numerous and clearly entacmaeic tentacles.Electronic Supplementary Material  Supplementary material is available in the online version of this article at The publisher regrets that during copy-editing of this article the names of the authors of species were styled incorrectly. The phytophagous bug Riptortus clavatus (Thunberg) (Heteroptera: Alydidae) produces two or three generations per year in Central Japan and overwinters in the adult stage. In bugs from the Kyoto population (35°00 N, 135°45 E), we studied (1) the effects of day-length on the nymphal and preoviposition periods under constant photoperiod at 20.5 °C, and (2) photoperiodic induction of adult diapause at 20.5 °C and under a combination of constant photoperiod and natural daily rhythm of temperature in the forest-steppe zone of Russia (50°38 N, 35°58 E). Then, we examined (3) the timing of diapause induction under quasi-natural conditions in the same region, far outside the species' natural geographical range. At 20.5 °C, the nymphal period in both males and females was significantly longer under regimes with shorter photophases than under those with longer photophases. The preoviposition period in females was significantly longer under the near-critical long-day regime L14:D10 than under typical long-day regimes (L15:D9, L16:D8, and L17:D7). The critical day-length for diapause induction was shorter under conditions of natural daily rhythm of temperature than those reported at constant 20, 25, and 30 °C. Under quasi-natural conditions in the forest-steppe zone, R. clavatus entered diapause in September, much later than the local populations of true bugs studied to date. This experiment showed that R. clavatus was maladapted to new environmental conditions: diapause was induced too late with the result that all or most nymphs hatched in late August or early September will die.  相似文献

Bacillus polymyxa grown in a recycling fermentor shows the same behavior previously observed with Escherichia coli: 3 successive growth phases. In the last 2 phases the growth rate is linear and the apparent maintenance energy demand rate and the molar growth yield are both independent of the specific growth rate, , and of the cells mass. The final phase of very slow growth is an indefinitely prolonged state of strong, stringent control, the regulatory system based on guanosine 3-diphosphate 5-diphosphate, and guanosine 3-diphosphate 5-triphosphate. The maximum cost of this stringent response is calculated to be 9% of the energy available to these energy-limited cells. There is a further energy cost contained in substantial amounts of DNA, RNA, and protein released from the cells during the latter 2 growth phases. Benzyladenine (BAP) uptake and metabolism were characterized during the key stages of shoot organogenesis in leaf explants of Petunia MD1. Using leaf explant transfer experiments, it was shown that exposure to 2.2 M BAP for 6, 8 or 10 days induced shoot formation on 27, 80 and 100% of the explants respectively, with a concomitant increase in the number of shoots per explant. BAP uptake and metabolism were characterized in leaf explants after 1, 3, 6 or 10 days exposure to [3H]BAP or 10 days exposure plus an additional 2 days on basal medium (10+2). BAP and 9--D-ribofuranosyl-BAP ([9R]BAP) were detected at days 1 and 3 only. Therefore, the BAP free base was not detectable during the shoot induction period between days 6 and 10, as defined by leaf transfer experiments. The BAP ribotide pool was largest on day 1 and decreased to day 10+2. It is possible that the BAP ribotide pool provided either the active cytokinin itself or acted as a short-term storage form for the active cytokinin in petunia shoot organogenesis. The effects of the removal of a heated water discharge from Lake Wabamun, Alberta (longitudes 114°26 and 114°44W; latitudes 53°30 and 53°35N) upon epiphytic algae has been documented. Due to the timing of the cessation of flow the first year without elevated temperatures was one of transition. Standing crops returned to normal lake levels the following year. Species composition changes occurred, and species such as Epithemia turgida, Amphipleura pellucida, Nitzschia fonticola, Rhopalodia gibba, Rivularia haematites, and Scytonema alatum, all of which were detrimentally affected by the elevated temperatures, returned to the previously affected area, while species such as Oedogonium sp., Spirogyra sp. and Cladophora glomerata, dominant at times in the heated area, diminished in importance. The ability of Cocconeis placentula to grow at low light intensities (15–110 µEinsteins m–2 s–1) was again demonstrated.  相似文献

The movements of nine breeding adult emperor penguins Aptenodytes forsteri from two colonies, Auster (67° 23S 64° 04E) and Taylor Glacier (67° 28S 60° 54E), were determined by satellite telemetry on their pre-moult foraging trips. While preparing for their annual moult the penguins travelled for 22–38 days and reached distances of up to 618 km from the colony. Six of the nine tracked penguins were followed to three different moult locations all to the west of their breeding colonies and near other known emperor penguins colonies, such as Kloa Point (66°38S, 59°23E) and Fold Island (67°17S, 59°23E). Sea-ice conditions changed throughout the tracking period; as the birds travelled north the sea-ice contracted south.  相似文献

The occurrence of adenosine 5-triphosphate-3-diphosphate-synthesizing activity was detected in five strains of actinomycetes; Streptomyces morookaensis, Streptomyces aspergilloides, Streptomyces hachijoensis, Actinomyces violascens and Streptoverticillium septatum, out of 825 strains of actinomycetes, bacteria, fungi and imperfecti. Purine nucleotide pyrophosphotransferase were extracellularly excreted associating with the cell growth, and were purified partially or to apparent homogeniety from the culture filtrate. Summary WhenActinoplanes strain ATCC 33076, the producer of A-16686 A1, A2 and A3 complex, is fermented in a suitable medium three additional factors, designated A1, A2 and A3 are produced. These were isolated and characterized, and were shown to differ from the parent components of the original complex by lacking one mannose unit. Bioconversion of A factors into A factors was achieved by incubation with the mycelium ofActinoplanes ATCC 33076. Factor A2 has better antibacterial activity than A2 against some bacteria.  相似文献

Gloshedobin, a thrombin-like enzyme from the venom of Gloydius shedaoensis, was expressed in Escherichia coli using expression vector pET-32a(+). The gene was expressed under T7 promotor with a fusion partner of Thx.Tag and a 6xHis.Tag at its 5 terminal. After induction by IPTG for 6 h, the recombinant enzyme was expressed in the cytoplasm. Expression at 25°C gave twice the amount of recombinant gloshedobin in cytoplasm than at 37°C.  相似文献

Lake Taimyr in Siberia is northernmost among the world's large lakes: 73°40–75°20N, 99–106°E. The lake area is up to 4650 km2 in summer, with a maximum depth of 26 m and a mean depth of only 2.8 m. The ice-free period lasts about three months. The water level sinks 5.5–6 m during winter, so that 85% of the bottom surface is frozen into ice for some time and subjected to negative temperatures, probably down to –20 °C. In artificially melted sediment samples, 75–92% of animals survived. The average summer biomass of zoobenthos is about 1 g m–2 wet weight, a half of this being formed by Oligochaeta. Altogether 76 samples with 3742 oligochaete specimens collected by V. N. Grëze in 1943–1944 were studied. At least 14 taxa of Tubificidae, Lumbriculidae, and Enchytraeidae were found in the lake, and some more enchytraeids in an adjacent river. Many immature animals could not be identified to species. Naididae were completely lacking probably due to the absence of macrovegetation. The shallow freezing zone is inhabited mostly by Alexandrovia ringulata. The profundal fauna is dominated by Lamprodrilus isoporus, Stylodrilus sp., and Isochaetides sp.  相似文献

The demography of the acaulescent palm Attalea humilis was studied from 1996 to 1999 in three forest fragments (1.6, 6.4, and 9.9ha) in southeastern Brazil (22°30–22°33 S, 42°15–42°19 W). A human-induced fire damaged the populations in August–September 1997. Palm density was higher in the medium-sized fragment, where it was concentrated in a single disturbed sector. Population ontogenetic stage structure was dependent on fragment and year, but in general later stages were more abundant than initial ones. Population finite growth rates projected from linear, stage-structured demographic models indicated that the populations were increasing before fire. Population growth rate in the smallest fragment was larger due to a single recruitment event, being most sensitive to changes in survival and growth of seedlings, virgins and the immature, and relatively insensitive to changes in shrinkage and fecundity. Observed growth rates for the entire study period showed that populations remained in equilibrium after fire, but the population in the medium-sized fragment showed a declining tendency. Population structure was resilient to fire, which caused a transitory concentration of individuals in the medium-sized stage. The self-sustainability of the population in the largest, most preserved fragment suggests that habitat-protection measures may suffice to ensure the persistence of populations in small fragments. However, population fluctuations and exclusion from hyper-disturbed sectors in the smallest fragment suggest that reduced fragment area, habitat degradation and human-induced fires can interact to threaten the persistence of even abundant, preadapted to fire, species like A. humilis.  相似文献
