Physalopteroides arnoensis n. sp. (Nematoda: Physalopteroidea) from the intestinal tract of the gecko Lepidodactylus lugubris is described and illustrated. Physalopteroides arnoensis is the 18th species to be assigned to the genus and is distinguished from other oriental species by the distribution pattern of caudal papillae of the male: 22 posterior subventral pedunculated papillae; 8 precloacal papillae in symmetrical pairs; 14 postcloacal papillae, 6 papillae immediately postcloacal in symmetrial pairs and remaining 8 papillae asymmetrically placed, 3 on right, 5 on left. Other helminths found include Pharyngodon lepidodactylus, Hedruris hanleyae, Maxvachonia chabaudi, and larvae of Ascarops sp. Our review of Physalopteroides removes all subspecies designations; Physalopteroides minor Caballero, 1969 n. comb is proposed.  相似文献

SNMP-1 (sensory neuron membrane protein 1) is an olfactory-specific membrane-bound protein which is homologous with the CD36 receptor family. Previous light level immunocytochemical studies suggested that SNMP-1 was localized in the dendrites and distal cell body of sex-pheromone-specific olfactory receptor neurons (ORN); these studies further suggested SNMP-1 was expressed in only one of two to three neurons in male-specific pheromone-sensitive trichoid sensilla. To better understand the expression and localization of SNMP-1, an immunocytochemical study was performed using electron microscopy to visualize the distribution of SNMP-1 among the neurons of several classes of olfactory sensilla of both male and female antennae of the silkmoth Antheraea polyphemus. SNMP-1 antigenicity was primarily restricted to the receptive dendritic membranes of ORNs of all sensilla types examined and was observed in cytosolic granules, but not plasma membranes, of the cell soma. Mean labeling densities ranged from 1 to 16 gold particles per micrometer of dendrite circumference; dendrites of trichoid and intermediate sensilla showed significantly higher labeling densities than those of basiconic sensilla. Larger dendrites of trichoid sensilla showed significantly higher mean labeling densities (13-16/micron) than smaller diameter dendrites (3-7/micron). Immunofluorescence studies using baculovirus expressed SNMP-1 and multiphoton photon laser scanning microscopy (MPLSM) indicated that rSNMP-1, which was post-translationally processed to the in vivo molecular weight, was inserted into the plasma membrane in a topography presenting extracellular epitopes. These studies suggest SNMP-1 is a common feature of the ORNs, is asymmetrically expressed among functionally distinct neurons, and possesses a topography which permits interaction with components of the extracellular sensillum lymph.  相似文献

Inhibitors of porcine trypsin were prepared from aqueous extracts of the parasitic nematodes Ascaris suum (hogs) and Ascaris lumbricoides (human). In this study three experiments were performed. (1) Polyclonal antibodies were prepared against one isoform of trypsin inhibitor from each parasitic nematode. Each antibody reacted with all isoforms from itself as well as all isoforms from the other parasite. (2) Association equilibrium constants were measured by titrating host trypsins (porcine or human) with the isoforms of trypsin inhibitors from A. suum and A. lumbricoides. While three of the combinations formed tight complexes that can be precipitated, the fourth complex, A. suum trypsin inhibitor-human trypsin has a Ka that is a 300 to 1000 times weaker interaction than the three other titration pairs. (3) Live A. suum worms were incubated in isosmotic media that contained either porcine trypsin or human trypsin. A suum worms survived in porcine trypsin and in the controls but were killed and digested after exposure for 5 days in human trypsin. The first experiment suggests that the trypsin inhibitors from A. suum and A. lumbricoides have similar epitopes, while the second experiment suggests that there are differences near the reactive site of the inhibitors. The consequences of these differences are dramatically demonstrated by the third experiment in which live A. suum worms in the presence of human trypsin die and are digested but those in porcine trypsin survive. These experiments suggest that in order to parasitize a host, a nematode requires a complement of protease inhibitors that interact strongly with those host proteases that are in their environment.  相似文献

Upon completion of cytokinesis, the midbody ring is transported asymmetrically into one of the two daughter cells where it becomes a midbody ring derivative that is degraded by autophagy. In this study we showed that the ubiquitin-binding autophagy receptor SQSTM1/p62 and the interacting adaptor protein WDFY3/ALFY form a complex with the ubiquitin E3 ligase TRAF6 and that these proteins, as well as NBR1, are important for efficient clearance of midbody ring derivatives by autophagy. The number of ubiquitinated midbody ring derivatives decreases in TRAF6-depleted cells and we showed that TRAF6 mediates ubiquitination of the midbody ring localized protein KIF23/MKLP1. We conclude that TRAF6-mediated ubiquitination of the midbody ring is important for its subsequent recognition by ubiquitin-binding autophagy receptors and degradation by selective autophagy.  相似文献

Abstract The body of Hirudo medicinalis consists of 32 segments. The quinquannulate midbody segments 3–18 bear 14 sensilla on the central (neural) annulus. Elsewhere segments are represented by fewer than five annuli but sensilla are retained on the neural annulus. From neural sensilla protrude S hairs (cilia from uniciliate cells) which are thought to detect water currents. Two categories of multiciliate cell, of unknown function, are also present within neural sensilla; (i) grouped cilia extending beyond the cuticle (G hairs), and (ii) cilia which radiate out beneath the cuticle. Studies of the entire external surface of leech body wall with a scanning electron microscope revealed the presence of large numbers of sensilla on every annulus including the neural annuli. These sensilla lack S hairs. Our results show: (i) annular sensilla have a significantly smaller surface area than neural sensilla (p < 0.001 Mann-Whitney test, 2 tailed), (ii) The position and number of the small sensilla varies from annulus to annulus, segment to segment and individual to individual, (iii) Significantly higher numbers of small sensilla were found where non-neural annuli remain single and have not undergone a further division during development as happens in the quinquannulate segments. The data suggest that small sensilla continue to be added during the adult life of the leech.  相似文献

Summary Sensilla on legs and ovipositor of the moth Ostrinia nubilalis were investigated by light and scanning electron microscopy. The ovipositor is composed of two papillae densely packed with medium length mechanoreceptor sensilla (MRb: 80–160 m, n=420–460). Long mechanoreceptor sensilla (MRa: 250–300 m, n=20–24) and contact chemoreceptors (CRa: 30–40 m, n=20–28) are evenly distributed at the periphery of these papillae. Legs support contact chemoreceptors (CRa), scattered among the scales. The pretarsus structure of each leg includes a single contact chemoreceptor (125 m) inserted dorsally. The fifth tarsomere bears a ventral area without scales on which contact chemoreceptors are disposed in two parallel rows (CRb: 40–60 m). A sexual dimorphism was found in the number and density of these sensilla (females: mean = 5.3, SD=1.0; males: mean = 3.3, SD=0.7), and in the size of the sensory field. The possible involvement of these sensory structures in oviposition site selection is discussed.  相似文献

We here analyze the population structure in the pig roundworm, Ascaris suum, among domestic pigs in Denmark using a whole-genome DNA fingerprinting technique, "amplified fragment length polymorphism" (AFLP) analysis. With these data, we can extract absolute gene frequency variance components and G-statistics for 135 independent nucleotide polymorphisms. The average proportion of total variance partitioned between Jutland and Zealand is less than 3% of the total variance, implying no restriction in gene flow between worms from different regions in Denmark. The average gene frequency difference between two farms widely separated in Jutland represents 5% of the total genetic variance of these two farms combined. Conversely, worms from different hosts within these two farms are more subdivided, with an average of 12% of the total variance in gene frequencies within farms being distributed between hosts. This result implies substantial single generation inbreeding due to founder effects in the establishment of adult worms in single hosts. Absolute variance components extracted from the gene diversities also showed significant differences, with the among-host variance being greater that the between-farm and between-region values. This little geographical variation is discussed in relation to the hierarchic structure of the Danish swine production system. Comparison of our results with other studies on parasitic roundworms, suggests that patterns of host dispersal effectively control patterns of worm gene flow. Furthermore, the potential spread of anthelminth resistance among A. suum may thus be rapid, due to the flow of infected hosts within the domestic swine stocks in Denmark.  相似文献

The nematode Drilomermis leioderma n. gen., n. sp. (Merrnithidae) is described from larvae of Cybister fimbriolatus (Say) (Dytiscidae: Coleoptera) in Louisiana. Diagnostic characters of the genus Drilomermis are: medium-sized nematodes with the cuticle appearing smooth (lacking cross fibers) under the light microscope, six cephalic papillae, without mouth papillae, six hypodermal cords at midbody, 2 extremely long spicules (longer than 10 times body width at anus) which are separate and parallel (not twisted), an S-shaped vagina, medium-sized amphids located near head papillae, and postparasitic juvenile with a tail appendage. D. leioderma possesses a ventrally displaced mouth, very long vagina, and male genital papillae arranged in 3 double rows in the vicinity of the cloacal opening. Even when containing multiple parasites, about 40% of the hosts sulwived emergence of the memithids and lived several more days. In nature, some of these hosts may be able to continue their development, which is unusual since most mermithid-parasitized hosts die soon after the nematode emerges.  相似文献

Dirofilaria immitis infection is an important parastic disease in many mammals, especially canids, but has not been reported in bird hosts. Filarial worms were isolated from the lumen of the right atrium of the heart and the connective tissue of the lung of a captive female Humboldt penguin, Spheniscus humboldti, that died at a zoo in Japan. One of these worms was observed morphologically and identified as D. immitis by features such as 4 pairs of cephalic papillae, 1 pair of cervical papillae, esophagus divided into 2 regions, 4 pairs of pre-anal papillae, 5 pairs of post-anal papillae, and unequal spicules. In addition, the partial DNA sequence (234 bp) of mitochondrial CO / gene of the filarial worm was identical to that of D. immitis. This is the first report of D. immitis infection in a bird.  相似文献

Purebred Hampshire pigs, farrowed and maintained under conditions precluding extraneous helminth infection, were exposed to a single dose of 10,000 Ascaris suum infective eggs. The pigs were killed at intervals of 28, 41, 55, 86, 115, 145, 175, and 206 days after infection. At necropsy, no gross lesions were found in the lungs or livers of infected pigs. The worms were recovered from the small intestine, identified, counted, and fixed. The heads were excised, critical point dried, mounted en face, and examined by scanning electron microscopy. Worms 28 to 115 days old had unworn denticles that were triangular when viewed laterally but blunt when viewed tangentially. Wearing of the denticles was observed first with 145-day-old worms; wearing increased with age both in numbers of denticles affected and in degree of wear so that by 206 days after inoculation, almost all denticles in the center of the lip were worn. Worn denticles appear truncated when viewed from any angle. The denticles outside the central area were not affected by wear. The size of the denticles varies not only between specimens of the same age, but also on each specimen. However, average denticle size is directly related to the size and, accordingly, to the age of the worm. External to each denticle is a corresponding depression that we have called the denticular groove. One 28-day-old specimen had some extra denticles aligned irregularly along the lip; this irregularity gave the appearance of a double row. The denticles of the two subventral lips are similar to those of the dorsal and are equally affected by wear. There was no detectable difference in denticles of male and female worms. Since wear can now be specifically correlated with age, we conclude that the denticles are functional and become worn through use. Consequently, adult A. suum may be an even more injurious pathogen than heretofore supposed.  相似文献

Live Ascaris suum females were incubated in medium containing chymotrypsin liganded to fluorescein-5-isothiocyanate, and eggs in the parasite's genital tract took up the probe and fluoresced. Eggs passed by these worms into the medium containing fluorescent probe retained their fluorescence in formaldehyde-saline and by 65 days had developed into second stage infective larvae. Eggs passed naturally by untreated worms were incubated in media containing fluorescent probes and all of the eggs exposed to chymotrypsin liganded to fluorescein-5-isothiocyanate were extensively labeled. Control eggs were labeled sporadically and less intensely, indicating specificity in the uptake of environmental proteins. Chymotrypsin from the parasite's environment can bind to A. suum eggs, and this occurs both inside the worm's genital tract and outside of the parasite. Immunoperoxidase studies showed that IgG developed against chymotrypsin or against A. suum chymotrypsin/elastase isoinhibitors A or C, binds to antigens in cross sections of second stage larvae and their egg shell coats. This suggests that host chymotrypsin is retained during development and may be complexed to A. suum isoinhibitors A and C.  相似文献

Cone snails, predatory marine gastropods, have developed a specialized prey capture method in which a long, distensible proboscis is used to identify prey and inject venom via a hydraulically propelled hollow radular tooth. Using brightfield, epifluorescence, confocal, and transmission and scanning electron microscopy, we describe the morphology of ciliated sensory structures concentrated on the tip of the proboscis. The number and morphology of these sensory papillae are linked to the type of preferred prey: cone snails feeding on worms and mollusks have short, cone or finger‐shaped papillae, whereas fish‐hunting cone snails have long tubular papillae in addition to short conical papillae. Sensory papillae are well positioned to provide information necessary to locate, identify, and dispatch prey. Proboscis tips and their sensory papillae regenerated within 10 d following experimental ablation, and snails with regenerated proboscis tips were able to locate and envenomate prey. The remarkable intrageneric variation found in the morphology of these sensory structures is probably linked to the specialized prey that cone snail species have evolved to hunt.  相似文献

Pig faeces in which Ascaris suum eggs had been embryonating for 57 days were placed in buckets of soil containing either 30 or no earthworms (Aporrectodea longa). When present, earthworms consumed the faeces and transported the eggs down into the soil, without inflicting any visible damage on the eggs. In later experiments 10 earthworms from the above experiment were fed to each of ten pigs, and another 40 earthworms were dissected. None of the 10 pigs became infected with A. suum through consumption of earthworms, and none of the dissected earthworms were found to contain A. suum larvae. This experiment indicates that A. longa did not act as a paratenic host for A. suum but shows that earthworms are very efficient in transporting A. suum eggs from faeces deposited on the soil surface into the soil.  相似文献

The tegumental ultrastructure of juvenile and adult Himasthla alincia (Digenea: Echinostomatidae) was observed by scanning electron microscopy. One-, 5- (juveniles) and 20-day-old worms (adults) were harvested from chicks experimentally fed metacercariae from a bivalve, Mactra veneriformis. The juvenile worms were elongated and curved ventrally. The head crown bore 31 collar spines, arranged in a single row. The lip of the oral sucker had 12 paired, and 3 single type I sensory papillae, and the ventral sucker had about 25 type II sensory papillae. The anterolateral surface between the two suckers was densely packed with tegumental spines with 4-7 pointed tips. The adult worms were more elongated and filamentous, and had severe transverse folds over the whole body surface. On the head crown and two suckers, type I and II sensory papillae were more densely distributed than in the juvenile worms. Retractile brush-like spines, with 8-10 digits, were seen on the anterolateral surface, whereas claw-shaped spines, with 2-5 digits, were sparsely distributed posteriorly to the ventral sucker. The cirrus characteristically protruded out, and was armed with small spines distally. The surface ultrastructure of H. alincia was shown to be unique among echinostomes, especially in the digitation of its tegumental spines, the distribution of sensory papillae and by severe folds of the tegument.  相似文献

Fifty-four crossbred, 4-week-old pigs divided into nine equal groups were used to test whether multiple inoculations with high numbers of A. suum eggs with or without anthelmintic would result in patent infections. All pigs exposed to multiple prechallenge inoculations of 500, 1000, 2000, 5000, 10,000 and 20,000 and challenged orally 2 weeks later with 10,000 eggs harboured adult worms. When prechallenge infections were removed by pyrantel tartrate treatment the animals were more susceptible to challenge than controls not previously exposed to infections. The same drug used from 2 days before until 10 days after the last prechallenge infection eliminated that effect. Pigs subjected to the same multiple egg dosing regimen but given feed containing fenbendazole immediately before, during and for 10 days after multiple dosing developed significantly more adult intestinal worms after challenge than any other group. These worms were, however, significantly shorter than those that developed in any group of pigs. Adult worms from all these groups produced eggs that after embryonation were infective to mice.  相似文献

Haller's organ in A. tridentatus consists of a capsule and an anterior group of sensilla. The capsule is the hollow in the cuticle on the dorsal surface of the first tarsus, where 4 pored hairs of olfactory sensilla are situated under the cover of the roof, formed by an anostomosis of the upper brunches of pleomorphs (capsule's bottom non-sensory cuticular outgrowths). The canal of the accessory ampullaceous sensillum opens in a capsule near the bottom. The anterior group of sensilla consists of two parts: proximal part, containing pored grooved and thin hairs, is homologous to the anterior grouf of ixodid ticks, and distal one which has no homologues in ixodids. A model is presented for the arrangement of alpha-helices in globular proteins. In the model, helices are placed on certain ribs of "quasi-spherical" polyhedra. The polyhedra are chosen so as to allow the close packing of helices around a hydrophobic core and to stress the collective interactions of the individual helices. The model predicts a small set of stable architectures for alpha-helices in globular proteins and describes the geometries of the helix packings. Some of the predicted helix arrangements have already been observed in known protein structures; others are new. An analysis of the three-dimensional structures of all proteins for which co-ordinates are available shows that the model closely approximates the arrangements and packing of helices actually observed. The average deviations of the real helix axes from those in the model polyhedra is +/- 20 degrees in orientation and +/- 2 A in position (1 A = 0.1 nm). We also show that for proteins that are not homologous, but whose helix arrangements are described by the same polyhedron, the root-mean-square difference in the position of the C alpha atoms in the helices is 1.6 to 3.0 A.  相似文献
