The present study used wobbegong sharks (genus Orectolobus) to assess the threshold value proposed by previous research to categorize strict lecithotrophic from incipient histotrophic species. Totals of 236 and 135 ornate wobbegong Orectolobus ornatus and spotted wobbegong Orectolobus maculatus, respectively, were collected from the New South Wales commercial fishery between June 2003 and May 2006. Eight pregnant gulf wobbegong Orectolobus halei were also recorded outside the sampling period for the first time. The three species were reproductively synchronous with a gestation of c. 10-11 months. Embryos started to be macroscopically visible during January and external yolk sacs were fully absorbed by June to July when embryos were c. 200 mm total length (L(T) ). Internal yolk sacs were first observed during April to May when embryos were c. 160 mm L(T) , reached a peak during June and persisted in embryos immediately prior to parturition. The total wet mass from uterine egg to full-term embryos increased by 44-89% and 45-62%, whereas the total organic mass decreased by 32-33% and 26%, for O. ornatus and O. maculatus, respectively, suggesting that these species are strict lecithotrophic yolk-sac viviparous sharks with no maternal nutrient input. A review of the literature identified various issues and suggested that the previously proposed organic mass loss threshold value separating strict lecithotrophic species from incipient histotrophic species might not be appropriate. Instead, it is recommended that a combination of methods (e.g. estimation of organic mass gain or loss between ovarian egg and developed embryo, histology and electron microscopy of the uterus, radio-tracer assay and uterine fluid analysis throughout gestation) is used to discern between strict lecithotrophic and incipient histotrophic species.  相似文献

Embryos of the viviparous dwarf ornate wobbegong shark (Orectolobus ornatus) develop without a placenta, unattached to the uterine wall of their mother. Here, we present the first light microscopy study of the uterus of O. ornatus throughout pregnancy. At the beginning of pregnancy, the uterine luminal epithelium and underlying connective tissue become folded to form uterine ridges. By mid to late pregnancy, the luminal surface is extensively folded and long luminal uterine villi are abundant. Compared to the nonpregnant uterus, uterine vasculature is increased during pregnancy. Additionally, as pregnancy progresses the uterine epithelium is attenuated so that there is minimal uterine tissue separating large maternal blood vessels from the fluid that surrounds developing embryos. We conclude that the uterus of O. ornatus undergoes an extensive morphological transformation during pregnancy. These uterine modifications likely support developing embryos via embryonic respiratory gas exchange, waste removal, water balance, and mineral transfer.  相似文献

Maternal behavior and serum prolactin were measured in pregnant and virgin female rats. Pregnant rats were either ovariectomized or shamovariectomized on Day 17 of pregnancy, while virgin females were ovariectomized at the same age. Two days after surgery nests were rated and the three treatment groups were tested for responsiveness to rat pups. Both pregnant treatment groups built superior nests compared to the virgin group and also responded more frequently to rat pups within a 1 hr test period than the virgin controls. In addition, significantly more ovariectomized pregnant subjects responded to pups than did intact pregnant females. Serum prolactin levels did not differ among the three treatments nor did exposure to pups affect serum prolactin levels. In each treatment group serum prolactin was less than 15 ng/ml, well below the 139.7 ng/ml mean found on Day 23 of pregnancy. These data suggest that high levels of serum prolactin during late pregnancy are not essential for the initiation of maternal behavior in the rat.  相似文献

锦绣龙虾(Panulirus ornatus)是世界名贵的经济虾类, 也是我国捕捞和养殖的大型虾类, 但资源匮乏,育苗还没有取得成功, 因此, 需要开展人工繁殖研究以保护和增殖资源。实验用人工养殖26 月龄的锦绣龙虾进行人工繁殖和胚胎发育研究, 结果表明, 锦绣龙虾在5 个月内繁殖2 次。切除锦绣龙虾单侧眼柄能加速其性腺发育, 在第一和第二次繁殖中, 切除眼柄的雌龙虾比对照组的卵巢成熟时间分别缩短17.7d 和11.3d。锦绣龙虾在交配后1—10d 内产卵。锦绣龙虾切除眼柄与否对怀卵量没有显著影响。第一、二次繁殖平均产卵量分别为40.34×104 粒和32.97×104 粒, 平均孵化率分别为77.29%和77.72%, 孵化出叶状幼体平均数量分别为31.38×104/尾和25.62×104/尾。锦绣龙虾的胚胎发育分为11 个时期: 受精卵、卵裂期、囊胚期、原肠期、膜内无节幼体期、七对附肢期、九对附肢期、十一对附肢期、复眼色素形成期、准备孵化期和孵化期。在水温29.2℃, 盐度30, 受精卵经22—23d 孵化出叶状幼体。    相似文献

Neonatal predation in multispecies aquarium exhibits can prevent detection of captive breeding by wobbegong sharks. We used ultrasonography and isolation strategies to prevent neonatal predation and maximize survival/growth of the dwarf ornate wobbegong (Orectolobus ornatus de Vis, 1883). We captured seven free‐living wobbegongs (two males and five females) and subjected each animal to a health assessment which led to the euthanasia of one female with a retained hook. Ultrasonography showed that females were pregnant, one was preovulatory, and one was in a resting phase. Two females (one pregnant) and one male were placed in isolation in each of two tanks. In October 2006, 25 neonates were born overnight with the two litters placed into separate neonate tanks. Over the ~6.5‐month monitoring period, four neonates with reduced body condition died without premonitory signs resulting in a 63.0% annual survival rate. Finite growth rates did not differ between sexes or litters and averaged (±SE) 12.2 (1.5) cm/year and 156.4 (26.4) g/year. At the cessation of monitoring, total length had increased by ~30%, whereas total weight had almost doubled with neonatal body condition in line with free‐living wobbegongs. Our efficacious, six‐step manipulative, the approach should be applicable with all wobbegongs given their reproductive similarities, but we recommend that efforts focus on the dwarf ornate, tasselled and Japanese wobbegongs because all are small in size and have bred in aquaria. Ultimately, this approach should produce self‐sustaining aquarium populations, place less reliance on the wild acquisition and provide animals for other aquaria, population restocking, or scientific research.  相似文献

In prairie voles (Microtus ochrogaster), most virgin females are infanticidal. To determine the onset of maternal responsiveness, female prairie voles were tested for maternal behavior as virgins and at different times throughout pregnancy. Female voles that were infanticidal as virgins by and large remained infanticidal throughout pregnancy. In contrast, about 30% of voles that were maternal as virgins became infanticidal during pregnancy. To test whether events associated with parturition facilitate the onset of maternal behavior, females had their litters delivered by Caesarean section within a day of expected delivery or were allowed to give birth naturally with sham surgery occurring shortly before or after birth. Females that gave birth naturally were fully maternal and did not attack unrelated pups, but females subjected to artificial delivery remained infanticidal. This suggests that events closely related to parturition are crucial for full development of maternal behavior in female prairie voles.  相似文献

We report full-term development of nuclear transfer embryos following nuclear exchange at the 2-cell stage. Nuclei from 2-cell rat embryos were transferred into enucleated 2-cell embryos and developed to term after transfer to recipients (NT2). Pronuclear exchange in zygotes was used for comparison (NT1). Zygotes and 2-cell embryos were harvested from 4-week-old female Sprague-Dawley rats. Nuclear transfer was performed by transferring the pronuclei or karyoplasts into the perivitelline space of recipient embryos followed by electrofusion to reconstruct embryos. Fused couplets were cultured for 4 or 24 h before being transferred into day 1 pseudopregnant recipients (Hooded Wistar) at the 1- or 2-cell stage. In vitro culture was also carried out to check the developmental competence of the embryos. In vitro development to the blastocyst stage was not significantly different between the two groups (NT1, 34.3%; NT2, 45.0%). Two of three recipients from NT1 and two of five recipients from NT2 became pregnant. Six pups (3 from NT1, 3 from NT2) were delivered from the four foster mothers. Three female pups survived; 2 from NT1 and 1 from NT2. At 2 months of age these pups appeared healthy, and were mated with Sprague-Dawley males. One rat derived from NT1 delivered 15 pups (5 males, 10 females) as did the rat from NT2 (7 males, 8 females). Our results show that by using karyoplasts from 2-cell stage embryos as nuclear donors and reconstructing them with enucleated 2-cell embryos, healthy rats can be produced.  相似文献

The success rate of in vitro fertilization (IVF) in dogs is low and only early embryos have been obtained. In the present study, we investigated the use of intrauterine transfer of early-stage canine embryos to obtain pups. Twenty-three female dogs, in which the date of ovulation (based on plasma progesterone concentrations) differed by +/-1 day, were used (10 donors and 13 recipients). The uterine tube was extirpated under general anesthesia 1-4 days after mating (5-7 days after ovulation), and descending perfusion was done to collect embryos. Embryos were examined and transferred into the uterine horn of a recipient, ipsilateral to the ovary with the most corpora lutea. Pregnancy was established in one of eight bitches that received early embryos (zygote to 4-cell embryos); she received two zygotes and one 2-cell embryo and delivered two puppies. Although intrauterine transfer of early embryos (zygote to 4-cell embryos) was difficult, pregnancy was achieved, suggesting that uterine tube transfer is appropriate for these early-stage embryos.  相似文献

This study reports on evidence for reproductive isolation among Orectolobus ornatus and Orectolobus halei, two previously cryptic and recently redescribed species of wobbegong shark (Orectolobiformes: Orectolobidae) from the east coast of Australia. The evidence is based on disparity in size at sexual maturity, diagnostic nuclear and mitochondrial DNA variants, and marked phylogenetic divergence. Plots of total length (LT) and maturity for the two species were non‐overlapping and illustrative of statistically significant size dimorphism. Genetic analyses and phylogenetic reconstruction did not provide indication of hybridization between O. ornatus and O. halei. In fact, sequence divergence between them was higher than in comparisons with another congeneric and largely co‐distributed wobbegong species (Orectolobus maculatus). The assumption of a molecular clock revealed that the two species have evolved in isolation for c. 3·9 million years. These results challenge a paradigm often mentioned in the biodiversity literature that most cryptic species are the product of recent speciation events and will contribute to the development of effective management strategies for wobbegong sharks.  相似文献

The aim of this study was to investigate whether roscovitine (the cyclin-dependent kinase 2 inhibitor) effectively induces synchronization of the donor cell cycle at G0/G1 and to examine the effect of donor cell cycle synchronization protocols on canine somatic cell nucleus transfer. Canine fibroblasts were obtained from skin biopsy cultures taken from a 7-yr-old retriever. The donor cell cycle was synchronized either by culturing cells to reach confluency or by treating cells with 15 μg/mL roscovitine for 24 h. Cell cycle stages and apoptosis were analyzed by flow cytometry. After synchronization of the donor cell cycle, cells were placed with enucleated in vivo-matured dog oocytes, fused by electric stimulation, activated, and transferred into 18 naturally estrus-synchronized surrogates. There was no significant difference in cell cycle synchronization and apoptosis rates between the confluent and roscovitine groups. After transfer of reconstructed embryos, pregnancy was detected in three of nine surrogates that received cloned embryos reconstructed with roscovitine-treated cells, whereas only one of nine surrogates was pregnant after transfer of cloned embryos reconstructed with confluent cells. One pregnant female from the confluent cell group delivered one live and one dead pup, but the live one died within 5 days after birth. Three pregnant females from the roscovitine-treated cell group delivered eight live pups and one dead pup, and one of eight live pups died within 6 days after birth. In conclusion, the current results demonstrated that reconstructing embryos with roscovitine-treated cells resulted in increased efficiency of canine somatic cell nucleus transfer.  相似文献

This study examined anatomical and histological characteristics of genital organs of 38 black agouti females in the wild in different reproductive stages, collected by rural hunters in the North-eastern Peruvian Amazon. Females in the follicular phase of the estrous cycle had greater antral follicle sizes than other females, the largest antral follicle measuring 2.34mm. Antral follicles in pregnant females and females in luteal phase of the estrous cycle had an average maximum diameter smaller than 1mm. In black agouti females in follicular phase, some antral follicles are selected to continue to growth, reaching a pre-ovulatory diameter of 2mm. Mean ovulation rate was 2.5 follicles and litter size was 2.1 embryos or fetuses per pregnant female, resulting in a rate of ovum mortality of 20.8%. Many follicles from which ovulation did not occur of 1-mm maximum diameter luteinize forming accessory CL. The constituent active luteal tissues of the ovary are functional and accessory CL. Although all females had accessory CL, transformation of follicles into accessory CL occurred especially in pregnant females, resulting in a contribution from 9% to 23% of the total luteal volume as pregnancy advances. The persistence of functional CL throughout pregnancy might reflect the importance for the maintenance of gestation and may be essential for the continuous hormonal production. The duplex uterus of the agouti female is composed by two completely independent uterine horns with correspondent separate cervices opening into the vagina. In pregnant females, most remarkable observed uterine adaptations were induced by the progressive enlargement caused by the normal pregnancy evolution. The wild black agouti showed different vaginal epithelium features in accordance with the reproductive state of the female.  相似文献

The influence of concurrent pregnancy until the third lactation on reproduction was studied in four strains of mice (SHN, SLN, C3H/He and GRS/A) as a possible step to evaluate the efficiency of this system on offspring production. In all strains, little difference was observed between sequential and concurrent pregnancy and lactation groups in any parameter as the index of reproductivity--delivery interval, litter size, average weight and growth rate of pups, mother weight, rearing rate, still-birth rate and rate of still-born pups. Also, no difference was seen between groups of any strain, in the number of females which had three litters. The estimated number of pups weaned per female during 50 days after first being placed with males was higher in the concurrent pregnancy groups than in the sequential pregnancy groups of all strains. Meanwhile, the incidence of spontaneous mammary tumors, the major characteristic of these strains, was consistently lower in the concurrent pregnancy group in all strains. These results have demonstrated that the concurrent pregnancy and lactation system is an efficient means of mouse production.  相似文献

The role of accumulated intrauterine fluid in rats at oestrus was investigated by experimentally altering the fluid volume in 106 rats. Complete evacuation of uterine fluid or low natural fluid volume did not significantly reduce the number of embryos on Day 10 of pregnancy overall. Transuterine (but not transcervical) sperm transport was reduced by uterine fluid removal, but did not vary significantly with naturally occurring variations in fluid content. Irrespective of fluid content, transuterine sperm transport was, on average, greater in right than left horns. Distensible balloons inserted into the uterus indicated that increasing and decreasing volume increased and decreased respectively myometrial activity at oestrus and dioestrus. Left horns contracted less frequently than did right horns at low volumes. While uterine fluid volume clearly affected uterine contractility and transuterine sperm transport, we were unable to demonstrate a major role of fluid for fertility.  相似文献

Surgical embryo transfer in the silver fox was investigated as part of a larger project concerning the conservation of endangered canine species using modern artificial reproduction techniques with the farmed fox as a model. The animals were chosen on the basis of synchrony in natural oestrus. The timing of ovulation and artificial insemination was determined by measuring electrical resistance in the vagina. Twenty-nine embryos were flushed from eight humanely killed donor females and transferred surgically into the uteri of eight recipients. One recipient female gave birth to two male pups 47 days after the transfer of four expanded blastocysts and one embryo at the 16-cell stage derived from a donor female flushed 10 days after artificial insemination.  相似文献

Baseline and stress-responsive glucocorticoid (GC) levels were assessed during early pregnancy, late pregnancy, and lactation in female variable flying foxes (Pteropus hypomelanus) and in males over the same time period. Animals were maintained in a breeding colony in captivity. High levels of both cortisol and corticosterone were detected, with total plasma GC levels being among the highest documented in vertebrates (up to 3000 ng/ml in individual animals, with cortisol being the primary GC, accounting for approximately 78% of total GCs), and significantly greater in males than in females. Plasma levels of cortisol and corticosterone showed nearly identical profiles within each sex, with the exception of females in late pregnancy, in which corticosterone, but not cortisol, increased significantly. Baseline levels of plasma cortisol were highest in September (when pups were between 1 and 2 months of age) in both sexes, which may be related to the approaching onset of the mating period. There was a continuum in the magnitude of the response to stress (handling and sampling) over time in females, with the greatest stress response in early pregnancy, a dampened response during late pregnancy, and no significant stress response during lactation. Surprisingly, males failed to exhibit elevated GCs after this stress, but did have significant stress-induced hyperglycemia and suppression of plasma testosterone levels. This may be due to their high (perhaps maximal) baseline levels, which suggests that being in a breeding group was chronically stressful for males.  相似文献

Cauda epididymal spermatozoa of the golden hamster were labelled with the thiol-alkylating reagent, monobromobimane (MB). Female hamsters underwent uterine insemination with labelled spermatozoa at laparotomy under metofane anesthesia. All 12 females examined between 5 and 54 h postinsemination yielded a total of 83/100 (83%) eggs in the process of fertilization or embryos. Under ultraviolet (UV) exposure all exhibited a fluorescent tail which, in the 4- and 8-cell embryos, could be seen to be fraying or disintegrating. As cleavage progressed, labelled tail components came to be restricted among the blastomeres such that at the 4- and 8-cell stage the tail could be seen in only one to three blastomeres. To study complete development and pregnancy another 12 females received uterine insemination. After recovery from anesthesia (approximately equal to 4 h) these females were mated with a vasectomized male bearing a dominant genetic marker (black eyes) to allow unequivocal determination of paternity in the fetuses and young produced. Seven became pregnant with one female losing her pregnancy about Day 7 of gestation. Two females sacrificed on Day 13 produced 17 normal fetuses and one resorption. Four females delivered 16 young, all normal at birth and in subsequent growth and fertility. In addition, insemination of female rabbits with MB-labelled spermatozoa yielded normal embryos (50/52 96%) from 3 does on Day 2 and (38/64 60%) from 4 does on Days 4 or 5. We examined the nesting and nursing behavior of females of the caviomorph rodent Octodon degus. We recorded the behavior of two, three-female groups of lactating degus kept in captivity and compared it with that of singly housed lactating females. Grouped females spontaneously nested communally. Five of six lactating females had non-offspring pups hanging from their teats, which suggests that some non-offspring nursing took place. Non-mothers increased their maternal behavior after the delivery of their own pups. Although time allocated to parental care by each communally nesting dam did not differ from that of solitary lactating females, young of communal litters spent less time alone and enjoyed more time cared for by a lactating female than young of single dams. A zebra shark, Stegostoma fasciatum, held in captivity at the Burj Al Arab aquarium, produced embryos and pups in the absence of a male. A total of 15 pups were produced from eggs laid within the aquarium over a period of four consecutive years commencing 2007. Parthenogenesis was confirmed through DNA analysis for three pups sampled during the first two consecutive egg cycles and is presumed to be the method of reproduction responsible thereafter.  相似文献

Monoclonal antibodies to histocompatibility (H-Y) antigen, of IgM subclass, were used to immunologically determine the sex of mouse embryos prior to transfer to pseudopregnant recipients. Two experiments were performed, one using cytolysis of H-Y positive embryos and the other using binding of a Fluorescein Isothiocyanate-(FITC) labeled second antibody. Eight- to 16-cell embryos used in the cytolytic assay were cultured in Whitten's Medium without bovine serum albumin (WM), to which monoclonal antibody and normal guinea pig serum were added. Embryos were classified as affected or unaffected, based on morphology of the embryo and its blastomeres. A total of 550 embryos were cultured; 294 (53.5%) were scored as unaffected and 263 of these were transferred to recipients. Forty-three (81.1%) of 53 pups born were female. Morulae and early blastocysts were used in the FITC-labeled second antibody assay. Embryos were cultured in WM containing monoclonal antibody, washed and placed in drops of WM containing FITC-labeled anti-IgM. Following another wash embryos were individually evaluated at 200X for fluorescence. Fifty-five percent (169 of 305) of the embryos displayed cell-specific fluorescence. A total of twenty-three pups, 18 males (78.3%) and five females (21.7%), were born following transfer of 156 fluorescing embryos. Four male (17.4%) and nineteen female (82.6%) pups resulted from embryos classified as non-fluorescing.  相似文献

The purposes of this experiment were 1) to test the hypothesis that placing rabbit embryos into the mare's uterus would hasten oviduct transport and 2) to determine if placing fluid into the uterus of bred mares on Day 4 and/or Day 5 would subsequently disrupt the mare's pregnancy. The hypothesis that placing rabbit embryos into the mare's uterus would hasten oviduct transport was not supported, since the uterine recovery rate of equine embryos on Day 5 was not significantly higher (P>0.05) for mares receiving rabbit embryos on Day 4 than for mares receiving no uterine infusion on Day 4 (1 10 vs 0 10 , respectively). However, placing fluid into the mare's uterus on Day 4 was apparently responsible for hastened oviduct transport, since mares with media infused into the uterus on Day 4 had a significantly higher (P<0.05) recovery rate of equine embryos on Day 5 than did mares receiving either rabbit embryos or no uterine infusion on Day 4 post ovulation (5 10 vs 1 10 or 0 10 , respectively). The Day-14 pregnancy rate was significantly higher (P<0.05) for mares receiving no uterine infusion on Day 4 or Day 5 than for mares receiving uterine infusion on Day 5 or uterine infusion on both Days 4 and 5 (9 10 vs 4 10 , 2 10 and 0 10 , respectively).  相似文献
