Agglutinating activity often varies both between and within the algal species assayed. However, it is difficulty to interpret such variation without further analysis. We report a statistical analysis of agglutinating activities against human, cow, sheep, and pig erythrocytes, using cell extracts from 43 taxa (strains) of freshwater microalgae. Most of the extracts agglutinated erythrocytes from at least one of the sources, but pig erythrocytes appeared to be most suitable for the detection of agglutination reactions. Chlorella cell extracts preferentially agglutinated human erythrocytes, whereas extracts of other taxa were less active against mammalian erythrocytes. Cluster analysis generated four distinct subclusters of taxa, characterized by different specificities for antigens or carbohydrate receptors on the erythrocytes. Principal component analysis further separated the agglutination characteristics of Chlamydomonas from Chlorella on the first two components. Specificity for pig erythrocytes accounted for most of the clustering or grouping of algal taxa in multivariate analysis. However, clustering or grouping patterns of Chlorella species on haemagglutinating activity resembled that based on DNA sequences, revealing a possible genetic connection of agglutinins and their biochemical characteristics in algal cells. Variability of agglutination reactions among the algae investigated is simplified and interpreted most easily using multivariate analysis.  相似文献

Protein extracts from 22 species of marine macroalgae from Florida and North Carolina were compared for their abilities to agglutinate sheep and rabbit erythrocytes. Protein extracts from 21 algal species agglutinated rabbit erythrocytes compared to 19 for sheep erythrocytes. However, agglutination by brown algal extracts was variable. The agglutination produced by protein extracts from Dictyota dichotoma could be blocked by addition of polyvinylpyrrolidone. Protein extracts from North Carolina macroalgae were also tested against five bacterial species. Three of these agglutinated bacterial cells. Ulva curvata and Bryopsis plumosa agglutinated all five species. Protein extracts from five species of Florida algae were tested for their effects on mitogenesis in mouse splenocytes and human lymphocytes. Gracilaria tikvahiae HBOI Strain G-5, Ulva rigida and Gracilaria verrucosa HBOI Strain G-16S stimulated mitogenesis in mouse splenocytes, while Gracilaria tikvahiae HBOI Strain G-16stimulated mitogenesis in human lymphocytes.  相似文献

1. Fifteen marine algal species were analyzed for agglutinins to rabbit, sheep and human A, B and O blood group erythrocytes. 2. Protein extracts from all marine algae agglutinated rabbit erythrocytes, whereas twelve and five extracts agglutinated sheep and human erythrocytes, respectively. 3. The highest agglutination titers were consistently observed with rabbit erythrocytes. 4. Dictyota dichotoma strongly agglutinated human B blood group erythrocytes relative to A and O group erythrocytes. 5. Agglutination titer of rabbit erythrocytes by six algal extracts was not inhibited by mono- or polysaccharides, yet was reduced by glycoproteins.  相似文献

We analysed the haemagglutinating activity of algal extracts from 44 species of freshwater microalgae against native and trypsin/papain-treated cow, pig, sheep, and human A-, B-, and O-type erythrocytes. Algal extracts obtained with aqueous ethanol exhibited higher haemagglutinating activity than those obtained with aqueous acetone. Most of the algal extracts agglutinated at least one of the erythrocyte types analysed. Human erythrocytes were the most sensitive of the cell types analysed. In the other species, the sensitivity of algal haemagglutinating activity for erythrocytes was pig > sheep > cow. Pre-treating erythrocytes with trypsin and papain improved the detection of most algal agglutinins and increased the haemagglutination titre; pre-treatment with papain was most effective for pig erythrocytes. Algal extracts stored at –20 °C for 4 months lost their haemagglutinating activity. Algal extracts also exhibited strong antibiotic activity against food pathogenic bacteria, especially against Bacillus. Our numerical taxonomy data showed that these microalgae might be grouped into several clusters according to their haemagglutinating activity. The detection of haemagglutinating activity may provide an efficient biochemical or physiological character to classify and differentiate microalgae. Our results suggest that freshwater microalgae might provide a potent source of haemagglutinins and antibacterial compounds for biochemical and medical studies and applications.  相似文献

Algae of Peter the Great Bay (Sea of Japan) were for the first time bioassayed as a source of lectins. From 28 algal species of three orders, only some extracts from brown (Phaeophyta) and red (Rhodophyta) seaweeds were found to cause agglutination of human erythrocytes. The hemagglutinating activity of extracts from three species of brown algae and the red alga Tichocarpus crinitus was caused by lectins; for a majority of extracts from the investigated algae, this activity was due to the presence of substances of non-lectin nature.  相似文献

Antibiotic activity of lectins from marine algae against marine vibrios   总被引:6,自引:0,他引:6  
Saline and aqueous ethanol extracts of marine algae and the lectins from two red algal species were assayed for their antibiotic activity against marine vibrios. Experimental studies were also carried out on the influence of environmental factors on such activity, using batch cultures. The results indicated that many of the saline extracts of the algal species were active and that the activity was selective against those vibrios assayed. The algal extracts were active against Vibrio pelagius and the fish pathogen V. vulnificus, but inactive against V. neresis. Algal lectins from Eucheuma serra (ESA) and Galaxaura marginata (GMA) strongly inhibited V. vulnificus but were inactive against the other two vibrios. The antibacterial activity of algal extracts was inhibited by pretreatment with various sugars and glycoprotein. Extracts of the two red algae, E. serra and Pterocladia capillacea, in saline and aqueous ethanol, inhibited markedly the growth rate of V. vulnificus at very low concentrations. Culture results indicated that metabolites active against V. vulnificus were invariably produced in P. capillacea over a wide range of temperature, light intensity, and nutritional conditions. Enhanced antibacterial activity occurred when P. capillacea was grown under higher irradiance, severe nutrient stress and moderate temperature (20 °C), reflecting the specific antibiotic characteristics of this alga. The strong antibiotic activity of lectins towards fish pathogenic bacteria reveals one of the important roles played by algal lectins, as well as the potential high economic value of those marine algae assayed for aquaculture and for biomedical purposes.  相似文献

Aqueous extracts from 42 species of Vietnamese marine macroalgae, including 17 Chlorophyta, 22 Rhodophyta, and three Phaeophyta species, were examined for hemagglutination activity using native and enzyme-treated different animal and human erythrocytes. All extracts agglutinated at least one type of erythrocytes tested. Strong activity was detected in extracts from four Chlorophyta (Caulerpa serulata var. boryana, Caulerpa sertularioides f. longipes, Halimeda velasquezii, and Halimeda discoidea) and two Rhodophyta species (Gelidiella acerosa and Titanophora pulchra) with enzyme-treated rabbit and horse erythrocytes. The hemagglutinins of some active species were examined for sugar-binding specificity, pH, temperature stability, and divalent cation independency using ammonium sulfate precipitates prepared from their extracts. In a hemagglutination–inhibition test with various monosaccharides and glycoproteins, none of the hemagglutinins had affinity for monosaccharides. The activity of the hemagglutinins was inhibited by some glycoproteins tested. The inhibition profiles with glycoproteins were different depending on hemagglutinin species, suggesting the presence of lectins specific for complex N-glycans, high mannose N-glycans or O-glycans. On the other hand, the activities of almost all algal hemagglutinins were stable over a wide range of pH and temperature, and independent of the presence of divalent cations, except Gelidiopsis scoparia hemagglutinin, its activity was dependent on the presence of divalent cations. These results suggest that Vietnamese marine macroalgae may be good sources of useful lectins for many biological applications.  相似文献

Extracts from 44 species of Vietnamese marine algae, including 15 Chlorophyta, 18 Rhodophyta and 11 Phaeophyta species, were examined for hemagglutination activity with a variety of different animal and human erythrocytes that were untreated or treated with enzymes. Almost all extracts showed activity toward at least one type of erythrocytes, although those from three Chlorophyta and two Rhodophyta species showed no hemagglutination with any type of erythrocytes examined. Strong activity was detected in extracts from two Chlorophyta (Anadyomene plicata and Avrainvillea erecta) and four Rhodophyta species (Gracilaria eucheumatoides, Gracilaria salicornia, Kappaphycus alvarezii, and Kappaphycus striatum) with enzyme-treated rabbit and sheep erythrocytes. The hemagglutinins of seven Chlorophyta and eight Rhodophyta species were examined for sugar-binding specificity, pH- and temperature-stability, and divalent cation-independency of hemagglutination using ammonium sulfate-precipitates prepared from their extracts. In a hemagglutination-inhibition test with various monosaccharides and glycoproteins, none of the hemagglutinins had affinity for monosaccharides, except the Codium arabicum and Gracilaria euchematoides hemagglutinins, whose activities were inhibited by both N-acetyl-d-galactosamine and N-acetyl-d-glucosamine. On the other hand, all of the hemagglutinins activities were inhibited by some glycoproteins. The inhibition profiles with glycoproteins were different depending on hemagglutinin species, and suggest the presence of lectins specific for high mannose N-glycans, complex N-glycans, or O-glycans. The activities of these algal hemagglutinins were stable over a wide range of pH and temperature, and independent of the presence of divalent cations. These results indicate that Vietnamese marine algae are a good source of novel and useful lectins.  相似文献

Aqueous protein extracts from 30 Brazilian marine algae were examined for haemagglutinating activity using native and enzyme-treated rabbit, chicken, sheep and human erythrocytes. Most extracts agglutinated at least one of the blood cells used. Sheep and rabbit erythrocytes were more suitable for detection of the agglutinating activity. The minimum protein concentration necessary to produce positive agglutination was usually lower with enzyme-treated erythrocytes than native ones. The five algal protein extracts showing the greatest haemagglutination titre were tested for sugar-binding specificity. Only the activity present in the green alga Cauler pacupressoides was inhibited by simple sugars and not by the glycoproteins tested. The activity of the other four extracts was inhibited by at least one of the glycoproteins utilised. A tetrameric lectin, with hemagglutinating activity toward rabbit erythrocytes and with specificity toward d-mannosamine and d(+)-mannose, was isolated from the ovaries of a teleost, the cobia Rachycentron canadum. The isolation protocol comprised ion exchange chromatography on CM-cellulose and Q-Sepharose, ion exchange chromatography by fast protein liquid chromatography (FPLC) on Mono Q, and finally gel filtration by FPLC on Superose 12. The lectin was adsorbed on all ion exchangers used. It exhibited a molecular mass of 180 kDa in gel filtration on Superose 12 and a single 45-kDa band in sodium dodecyl sulfate-polyacrylamide gel electrophoresis, indicating that it is a tetrameric protein. The hemagglutinating activity of the lectin was stable up to 40°C and between pH 4 and pH 10. All hemagglutinating activity disappeared at 60°C and at pH 1 and pH 13. The hemagglutinating activity was doubled in the presence of 0.1 μM FeCl3. The lectin exerted antibacterial activity against Escherichia coli with 50% inhibition at 250 μg. There was no antifungal activity toward Coprinus comatus, Fusarium oxysporum, Mycosphaerella arachidicola, and Rhizoctonia solani at a dose of 300 μg. The lectin exhibited maximal mitogenic response from mouse splenocytes at a concentration of 14 μM.  相似文献

Abstract The hemagglutinating activity of Clostridium perfringens enterotoxin (CPE) was studied after trypsin treatment. Untreated CPE did not show any hemagglutinating activity to human type A, B, and O, sheep, chicken, horse, guinea-pig, or rabbit erythrocytes. Trypsinized CPE resulted in a more than 100-fold increase in hemagglutinating activity with rabbit erythrocytes only. Other erythrocytes and trypsinized rabbit erythrocytes were not agglutinated at all. The hemagglutinating activity of CPE was also found on treatment with a lysine-specific proteinase. On the other hand, trypsinized CPE did not significantly increase the cytotoxic and enterotoxic activities. The binding reaction between trypsinized and rabbit erythrocytes was not inhibited by any mono-, di-, or polysaccharides, glycoproteins or ganglioside mixtures. These results suggest that the hydrolysis of bonds involving lysine residues is mainly required for hemagglutinating activity, and that the receptor for trypsinized CPE on rabbit erythrocytes is probably the protein moiety.  相似文献

Thirty-eight strains ofCorynebacterium pyogenes isolated from cases of heifer- and dry-cow mastitis and from other infections of sheep, cows, pigs, and man were screened for agglutination of sheep erythrocytes. Bacteria grown either in serum broth or on blood agar in the presence of CO2 hemagglutinated. Performance of titrations at 4°C avoided the hemolytic effects ofC. pyogenes. Erythrocytes of cat, chicken, cow, dog, guinea pig, horse, man (Group A), pig, and rabbit were also agglutinated. Pretreatment of sheep erythrocytes with trypsin, pepsin, A1 proteinase or pronase had no effect on agglutinability. Pretreatment ofC. pyogenes with pronase, but not with trypsin, A1 proteinase, or pepsin, abolished hemagglutinating capacity. The hemagglutinin was inactivated by exposure to 60°C for 10 min. Agglutination of sheep erythrocytes was inhibited by five glycoproteins. None of 12 mono-, di-, or trisaccharides nor heparin, chondroitin sulfate, or dextrin inhibited hemagglutination. These data suggest that the receptor may possibly be an oligohexosyl group of a glycoconjugate of lipid nature. Although a few cells of three mastitic strains ofC. pyogenes possessed fimbriae-like surface structures, no correlation between fimbriation and hemagglutinating activity was apparent.  相似文献

The protease elaborated by Vibrio mimicus is known to possess hemagglutinating ability to chicken erythrocytes, the well-known HA/protease. A non-protease hemagglutinin (HA) with strong agglutinating ability towards rabbit erythrocytes was obtained from 32 hr culture supernatant of a pathogenic environmental strain of V. mimicus. This HA (V. mimicus HA: VMHA) appeared stable at relatively higher temperature and agglutinated the erythrocytes from rabbit, guinea pig and mouse but not the erythrocytes from chicken, bovine, horse and sheep. Simple sugars, metal ions and chelating agents failed to inhibit the activity of VMHA. The activity of VMHA was found to be sensitive to digestion by proteolytic enzymes including HA/protease. These results provide evidence for the existence of novel HA other than HA/protease in V. mimicus.  相似文献

This paper presents seasonal in situ monitoring data on benthic coverage and coralalgae interactions in high-latitude fringing reefs of the Northern Red Sea over a period of 19 months. More than 30% of all hermatypic corals were involved in interaction with benthic reef algae during winter compared to 17% during summer, but significant correlation between the occurrence of coralalgae interactions and monitored environmental factors such as temperature and inorganic nutrient availability was not detected. Between 5 and 10-m water depth, the macroalgae Caulerpa serrulata, Peyssonnelia capensis and filamentous turf algae represented almost 100% of the benthic algae involved in interaction with corals. Turf algae were most frequently (between 77 and 90% of all interactions) involved in interactions with hermatypic corals and caused most tissue damage to them. Maximum coral tissue loss of 0.75% day−1 was observed for Acropora-turf algae interaction during fall, while an equilibrium between both groups of organisms appeared during summer. Slow-growing massive corals were more resistant against negative algal influence than fast-growing branching corals. Branching corals of the genus Acropora partly exhibited a newly observed phenotypic plasticity mechanism, by development of a bulge towards the competing organism, when in interaction with algae. These findings may contribute to understand the dynamics of phase shifts in coral reefs by providing seasonally resolved in situ monitoring data on the abundance and the competitive dynamic of coralalgae interactions.  相似文献

Human erythrocytes were exposed to extracts of Codium fragile ssp. tomentosoides, Ascophyllum nodosum, Dolichos biflorus seeds and a solution of purified polyphenols from Ascophyllum nodosum. In each case the erythrocytes appeared to agglutinate. Agglutinates were examined by scanning electron microscopy. Photomicrographs of the erythrocytes exposed to lectin-containing extracts of C. fragile and D. biflorus showed the typical appearance of agglutinated cells. Photomicrographs of the erythrocytes exposed to the crude extract and purified polyphenols from A. nodosum were similar to each other, but quite different in appearance to erythrocytes agglutinated by the lectins from C. fragile and D. biflorus. This evidence adds further support to the contention that lectin presence recorded in brown algal extracts is largely due to ‘pseudoagglutination’ produced by polyphenols contained in these extracts.  相似文献

Simple separation of anticoagulant sulfated galactan from marine red algae   总被引:1,自引:0,他引:1  
In this study, hot water extracts of 22 red algal species were evaluated for their potential anticoagulant activities. The extracts from eight species (Grateloupia elliptica, Sinkoraena lancifolia, Halymenia dilatata, Grateloupia lanceolata, Lomentaria catenata, Martensia denticulata, Schizymenia dubyi, Chondrus crispus) showed potent activated partial thromboplastin time (APTT). Of these eight algae, the crude polysaccharide fraction (CpoF) from the hot water extracts of L. catenata and S. dubyi showed the highest APTT activity. Lomentaria catenata and S. dubyi were selected and an enzymatic digestion process which could effectively separate a crude polysaccharide fraction with higher yields from raw algae materials was applied. The 10 enzymes tested included five carbohydrases and five proteases. The Ultraflo and Celluclast digests of L. catenata and the AMG digest of S. dubyi exhibited the most potent anticoagulant activity. Furthermore, in both species, the active compounds were mainly concentrated in the >30 kDa fraction through ultrafiltration and showed strong APTT (>1000) and thrombin time (TT) >1000 activity. The active compounds were shown to be sulfated galactans with a greater than 80% galactose content and an 0.22 ∼ 0.31 sulfate to total sugar ratio.  相似文献

We examined the sub-ice algal community in the Chukchi Sea during June 1998 using a remotely operated vehicle (ROV). Ice algae were observed on the under-ice surface at all ten stations (from 70°29′N to 72°26′N; 162°00′W to 153°56′W) and varied in abundance and distribution from small aggregations limited to depressions in the ice to nets, curtains and strands of Melosira. There was no relationship between percent cover of sub-ice algae and physical factors at the kilometer scale, but at the scale of individual ice floes the percent cover of sub-ice algae was positively correlated with distance from the floe edge and negatively correlated with snow depth. A significant positive relationship between the concentration of sediment pigments and percent cover of sub-ice could indicate a coupling between ice algal and benthic systems. Pieces of ice algae that appeared to be Melosira were observed on the seafloor to a depth of over 100 m and cells or spores of obligate ice algal taxa were collected from sediments from 44-m to 1,000-m deep. The large biomass of sub-ice algae observed at many stations in the Chukchi Sea and the presence of ice algae on the seafloor indicates that the distribution and abundance of sub-ice algae needs to be understood if we are to evaluate the role of ice algae in the Arctic marine ecosystem.  相似文献

Synopsis A revitalized view of feeding by herbivorous marine fishes is sought through two questions. First, What characteristics of major taxa of algae identify them as predictably high or low quality foods? Second, are marine algae valuable foods for fishes which do not mechanically disrupt cell walls and do not harbor specialized enzymes or microbes capable of lysing cell walls? Energy, ash and nutrient content of 16 species of marine algae were employed to assess food quality of fleshy red, green, brown and calcareous red algae. On the basis of ash, calories, total protein and total lipid content, fleshy algae should be superior to calcareous algae as foods for fishes; in addition, green algae should be superior to brown algae and brown algae superior to red algae. When the probable digestibility of storage and extracellular carbohydrates is considered, green and red algae are predicted superior to brown algae as food. Two species of damselfishes (Pomacentridae) from the Gulf of California,Eupomacentrus rectifraenum andMicrospathodon dorsalis, eat red and green algae and ignore brown and calcareous algae. They feed, therefore, in a fashion consistent with predictions based only on algal chemistry. These fishes absorb at least 20–24% of the biomass, 57–67% of the protein, 46–56% of the lipid and 37–44% of the carbohydrate contained in algae eaten in the wild. Since these damselfishes do not masticate their food, it appears that herbivorous fishes can digest major fractions of algal nutrients without mechanical destruction of algal cells.  相似文献

摘要:【目的】研究三峡水库神农溪库湾春季水华期间浮游藻类演替及其成因分析。【方法】2014年3–5月在神农溪库湾布置了6个断面(SN01–SN06),在神农溪汇入长江干流河口附近水域设置1个断面CJBD,对浮游藻类、相关环境因子及水动力因子进行了同步监测,据此分析了水体层化结构及水动力特性。【结果】神农溪在监测时段内共检测到浮游藻类6门38种(属);库湾浮游藻类生物量时间上差异显著(ANOVA,P<0.05)。春季浮游藻类群落结构具有明显的演替规律,3月份暴发大面积的硅藻水华(藻密度>100×105 cells/L),小环藻(Cyclotella spp.)为优势藻种;4月在SN02–SN06暴发以小球藻(Chlorella spp.)为主要优势种、衣藻(Chlamydomonas spp.)为次优势种的绿藻水华(藻密度>100×105 cells/L),5月份受水位大幅消落影响,浮游藻类生物量降低且无明显优势藻种。【结论】在具备充足的营养盐的水体中,水体层化结构与水动力特性对浮游藻类演替影响重大。三峡水库水位处于快速消落阶段时,流速成为抑制神农溪库湾藻类生长的主要因素。  相似文献

Goose erythrocytes were agglutinated by five strains of rabies virus grown in monolayer cell cultures at pH 6.4 and at 0 to 4 C. Hemagglutination was not affected by the cell type in which the virus was grown. Prerequisites for occurrence of hemagglutination are absence of hemagglutination inhibitors (such as those contained in bovine serum) and a relatively high virus concentration (> 10(6) plaque-forming units of virus per ml). "Soluble" hemagglutinin was not present in crude preparations of extracellular virus. Treatment of purified preparations of extracellular virus with Tween 80 and ether did not result in release of a "soluble" hemagglutinin. The hemagglutinating property of extracellular virus seemed to be conditioned by the integrity of its coat. Preparations of infectious intracellular virus exhibited about 15 times lower hemagglutinating activity than extracellular virus. This decreased hemagglutinating activity did not seem to be caused by binding of hemagglutination inhibitors to the virus particles. Rabies virus can be quantitatively adsorbed onto and eluted from erythrocytes. Erythrocytes pretreated with rabies virus retained their ability to be agglutinated by the same virus strain. The reaction with rabies virus of erythrocytes treated with the receptor-destroying enzyme or KIO(4) was the same as that of nontreated erythrocytes. The hemagglutinating component of rabies virus, therefore, does not exhibit neuraminidase activity. Treatment of extracellular virus by various agents indicated that the hemagglutinating component consists of protein or lipoprotein. Sulfhydryl groups present in the viral hemagglutinin are essential for hemagglutination.  相似文献
