Eighty soil samples were collected from various sites of Bahrain and screened for presence of keratinophilic fungi using hair baiting techniques for isolation. Thirty-six isolates were recovered and identified. The cultures were identified using macro- and micromorphological features. Their identification was also confirmed by the BLAST search of sequences of ITS1-5.8S-ITS2 rDNA region against the NCBI/Gene bank data and compared with deposited sequences for confirmation. Eight species of five genera were isolated viz. Aphanoascus fulvuscence (8.75%), Aphanoascus punsolae (20.00%), Chrysosporium indicum (2.50%), Chrysosporium tropicum (2.50%), Chrysosporium zonatum (3.75%), Spiromastix warcupii (1.25%), Microsporum gypseum (3.75%), and Trichophyton mentagrophytes (2.50%). In conclusion, our study indicates that keratinophilic fungi do occur in the various soils of Bahrain. Moreover, the narrow diversity and low density of keratinophilic fungi in the investigated soils is expected and is emblematic to other hot arid environments.  相似文献

以焦化废水处理系统的微生物群落为对象,对3种不同培养基(YPG、LB、WW)分离细菌的能力及分离物的种群多样性组成进行了比较研究。同一悬浮污泥样品在YPG、LB和WW培养基上的活菌计数结果分别为1.6×10.6 CFU/mL、7.0×10.5 CFU/mL和98×10.5CFU/mL。从每种培养基10-4稀释度平板上共分离137株分离物。将所有分离物扩增近全长的16S rDNA并用限制性内切酶HinfI对PCR产物进行ARDRA(Amplified rDNA restriction analysis)多态性分析,共得到14种不同的操作分类单元(Operational Taxonomic Unit, OUT)。其中YPG培养基上的分离物显示了8种不同的OTUs,而WW培养基和LB培养基分离物只分别显示了6种和4种OTUs。YPGOTU1和WWOTU6所包含的菌株分别占到总分离物的30%和22.3%,为优势分离物。ERICPCR基因组指纹图分析表明,前者的34株分离物共有20种不同的指纹图类型,而后者的25株分离物只有3种。因此,就分离焦化废水处理系统中的细菌及对分离物进行种群多样性的研究而言,YPG培养基比其他两种培养基更合适。  相似文献

A nested PCR protocol was used to analyze five endemic salamander species from Central Texas for the presence of the emerging pathogen, chytrid fungus (Batrachochytrium dendrobatidis). Chytrid fungus was detected from samples of each of the five species sampled: with low abundance, in the Texas salamander (Eurycea neotenes) (1 positive out of 16 individuals tested; 1/16), the Blanco River Springs salamander (E. pterophila) (1/20), the threatened San Marcos salamander (E. nana) (1/17), and the endangered Barton Springs salamander (E. sosorum) (1/7); much higher abundance was obtained for the Jollyville Plateau salamander (E. tonkawae) (6/14), which has recently been petitioned for addition to the USA endangered species list. With one exception, sequences of PCR products were identical to the 5.8S rRNA gene, and nearly so for the flanking internal transcribed spacer (ITS) regions of B. dendrobatidis which confirmed the detection of chytrid fungus, and thus demonstrated the presence of this pathogen in populations of endangered species in Central Texas. These confirmations were obtained from nonconsumptive tail clippings which confirms the applicability of historically collected samples from other studies in the examination of the fungus across time.  相似文献

Host fitness is impacted by trillions of bacteria in the gastrointestinal tract that facilitate development and are inextricably tied to life history. During development, microbial colonization primes the gut metabolism and physiology, thereby setting the stage for adult nutrition and health. However, the ecological rules governing microbial succession are poorly understood. In this study, we examined the relationship between host lineage, captive diet, and life stage and gut microbiota characteristics in three primate species (infraorder, Lemuriformes). Fecal samples were collected from captive lemur mothers and their infants, from birth to weaning. Microbial DNA was extracted and the v4 region of 16S rDNA was sequenced on the Illumina platform using protocols from the Earth Microbiome Project. Here, we show that colonization proceeds along different successional trajectories in developing infants from species with differing dietary regimes and ecological profiles: frugivorous (fruit-eating) Varecia variegata, generalist Lemur catta, and folivorous (leaf-eating) Propithecus coquereli. Our analyses reveal community membership and succession patterns consistent with previous studies of human infants, suggesting that lemurs may serve as a useful model of microbial ecology in the primate gut. Each lemur species exhibits distinct species-specific bacterial diversity signatures correlating to life stages and life history traits, implying that gut microbial community assembly primes developing infants at species-specific rates for their respective adult feeding strategies.  相似文献

mRNA differential display has been used to identify cyclohexanone oxidation genes in a mixed microbial community derived from a wastewater bioreactor. Thirteen DNA fragments randomly amplified from the total RNA of an enrichment subculture exposed to cyclohexanone corresponded to genes predicted to be involved in the degradation of cyclohexanone. Nine of these DNA fragments are part of genes encoding three distinct Baeyer-Villiger cyclohexanone monooxygenases from three different bacterial species present in the enrichment culture. In Arthrobacter sp. strain BP2 and Rhodococcus sp. strain Phi2, the monooxygenase is part of a gene cluster that includes all the genes required for the degradation of cyclohexanone, while in Rhodococcus sp. strain Phi1 the genes surrounding the monooxygenase are not predicted to be involved in this degradation pathway but rather seem to belong to a biosynthetic pathway. Furthermore, in the case of Arthrobacter strain BP2, three other genes flanking the monooxygenase were identified by differential display, demonstrating that the repeated sampling of bacterial operons shown earlier for a pure culture (D. M. Walters, R. Russ, H. Knackmuss, and P. E. Rouvière, Gene 273:305-315, 2001) is also possible for microbial communities. The activity of the three cyclohexanone monooxygenases was confirmed and characterized following their expression in Escherichia coli.  相似文献

THE MINIMAL LETHAL CONCENTRATION OF OZONE IN WATER WAS DETERMINED FOR THREE BACTERIAL SPECIES: Escherichia coli, Bacillus cereus, and Bacillus megaterium. A contact period of 5 min was selected. The lethal threshold concentration for the cells of B. cereus was 0.12 mg/liter while that for E. coli and B. megaterium was 0.19 mg/liter. Low concentrations of ozone were ineffective when organic matter was present to interfere with the action of ozone on the bacterial cells. Also determined during the study was the sensitivity of spores of B. cereus and B. megaterium to ozone in water. The threshold concentration required to kill the spores of both species was 2.29 mg/liter. The cells and spores of these organisms exhibited the "all-or-none" die-away phenomenon normally associated with ozone treatment.  相似文献

The harlequin frog, Atelopus mucubajiensis, is a critically endangered species that was believed to be extinct until 2004, when a single adult female was found in the Venezuelan Andes. Its disappearance after 1994 has been attributed to an increase in the prevalence of the fungal pathogen, Batrachochytrium dendrobatidis, in populations of these frogs. Using histology and Real-Time PCR assays, we tested this adult female positive for B. dendrobatidis. We report for the first time, to our knowledge, infection in a live specimen of A. mucubajiensis. The presence of this pathogen in remaining individuals poses a problem for the long-term persistence of this critically endangered species, as new epidemic episodes of chytridiomycosis could jeopardize the survival of remnant populations.  相似文献

Outer surface protein A (OspA) is the most promising candidate for a component vaccine against Lyme disease caused by Borrelia burgdorferi sensu lato. Active cross-protection using a whole-cell vaccine prepared from strains belonging various OspA serotypes observed in Japan and worldwide was examined. No cross-protection was obtained by heterologous OspA-serotype vaccines. Since OspA is a highly variable protein expressed by Borrelia, this suggests that immunologically different OspA serotypes need to be combined for the development of an effective vaccine in Japan.  相似文献

Batrachochytrium dendrobatidis (Bd) is a fungus that can potentially lead to chytridiomycosis, an amphibian disease implicated in die-offs and population declines in many regions of the world. Winter field surveys in the last decade have documented die-offs in populations of the lowland leopard frog Rana yavapaiensis with chytridiomycosis. To test whether the fungus persists in host populations between episodes of observed host mortality, we quantified field-based Bd infection rates during nonwinter months. We used PCR to sample for the presence of Bd in live individuals from nine seemingly healthy populations of the lowland leopard frog as well as four of the American bullfrog R. catesbeiana (a putative vector for Bd) from Arizona. We found Bd in 10 of 13 sampled populations. The overall prevalence of Bd was 43% in lowland leopard frogs and 18% in American bullfrogs. Our results suggest that Bd is widespread in Arizona during nonwinter months and may become virulent only in winter in conjunction with other cofactors, or is now benign in these species. The absence of Bd from two populations associated with thermal springs (water >30°C), despite its presence in nearby ambient waters, suggests that these microhabitats represent refugia from Bd and chytridiomycosis.  相似文献

Several tannin-tolerant bacteria were isolated from enrichment cultures of rumen microflora of bush duiker, giraffe, Grant's gazelle, sheep, and goat, and established in medium containing crude tannin extracts or tannic acid. The isolates were characterized by classical and molecular methods. The isolates were also tested for the presence of tannin acylhydrolase. Characterization by restriction fragment length polymorphism of the 16S rRNA–PCR product was performed withAlu 1, Dde 1, Msp 1, and Taq 1. Amplified fragment length polymorphism analysis was performed only on the isolates that were curved rods. The nucleotide sequence of PCR products derived from the 16S rRNA genes of the isolates was determined. The classical characterization suggested that, with one exception all the curved rods isolates wereSelenomonas and the coccus was a Streptococcus. Only Selenomonas -like isolates had tannin acylhydrolase activities. One isolate lost the ability to completely hydrolyze tannins after prolonged storage at −70°C. The restriction fragment length polymorphism profiles suggested that the Selenomonas -like isolates exhibited heterogeneity in the ribosomal RNA locus. The coccus had the same profiles as Streptococcus caprinus, while the straight rods appeared to be similar to each other. Amplified fragment length polymorphism analysis suggested that the Selenomonas -like isolates clustered into two major groups. The 16S rRNA sequences of the coccus clustered with that ofStreptococcus species and the Selenomonas -like isolates exhibited a high level of similarity withSelenomonas ruminantium , while the straight rods clustered with Klebsiella species accessions in the databases. A partial 16S sequence strongly indicated that one of the isolates was Butyrivibrio fibrisolvens.  相似文献

Diversity of Bacterial Isolates from Commercial and Homemade Composts   总被引:1,自引:0,他引:1  
The diversity of heterotrophic bacterial isolates of three commercial and two homemade composts was studied. The commercial composts were produced from poultry litter (PC), sewage sludge (SC), municipal solid waste (MC), and homemade composts (thermal compost [DC] and vermicompost [VC]) from food wastes. The taxonomic and physiological diversity of the heterotrophic culturable bacteria was assessed using phenotypic and genotypic characterization and the analysis of the partial 16S rRNA gene sequence. Composts DC and SC presented the higher genotypic diversity, as could be inferred from the number of distinct genotypic patterns observed, 28 and 21, respectively. Gram-positive bacteria, mainly Firmicutes, were predominant in all the composts. Some organisms related with taxa rarely reported in composts, as Rhodanobacter spathiphylli, Moraxella osloensis, Lysobacter, Corynebacterium, Pigmentiphaga kullae, and new taxa were also isolated. The highest relative proportion of isolates able to degrade starch was found in compost SC (>70%), to degrade gelatine in compost DC (>70%), to degrade Tween 80 in compost PC (>90%), and to degrade poly-epsilon-caprolactones in compost DC (>80%). Compost MC presented the lowest relative proportions of isolates able to degrade starch (<25%), gelatine (<20%), and poly-epsilon-caprolactone (<40%). When compared with the others, the homemade composts presented higher relative proportions of Gram-positive isolates able to inhibit the target organisms Staphylococcus aureus, Enterococcus faecalis, Escherichia coli, or Pseudomonas aeruginosa. In compost MC, none of the Gram-positive isolates was able to inhibit those targets.  相似文献

Two-dimensional gel electrophoresis analysis of and total cell protein determination for three bacterial isolates from marine waters at the onset and after 24 h of energy and nutrient deprivation demonstrated that the three isolates exhibited different pathways of starvation survival. 采用纯培养技术对一个降解喹啉的反硝化反应器筛选到的26株优势菌株进行反硝化能力以及好氧降解喹啉能力研究,其中的反硝化菌还测定了反硝化条件下的喹啉降解能力。结果发现Bacillus、Staphylococcus、Pseudomonas、Brucella、Delftia等5个属的10株菌具有反硝化能力,Rhodococcus属的9株细菌能够好氧降解喹啉,揭示了反硝化喹啉降解反应器中主要细菌类型的代谢特性,发现在缺氧反硝化反应器中存在多样的代谢类型的细菌。  相似文献

国内首次从捕自中国三个地区的猕猴分离到三株逆转录病毒,经ELISA、免疫荧光检查与SRV-1呈阳性反应。免疫蛋白印迹试验加以证实,血清抗体与SRV-1呈阳性反应,其中有一株与SRV-1和STLV血清抗体有交叉反应。电子显微镜下见有大量病毒颗粒,具有典型逆转录病毒结构。  相似文献

本研究采用响应面设计优化超声辅助提取车前总黄酮的最佳条件,然后用此条件提取大车前和平车前总黄酮,并探究大孔树脂纯化对三种车前草总黄酮抗氧化活性的影响。结果表明,在超声温度60℃、乙醇浓度70%的条件下,车前总黄酮最佳提取工艺参数为液料比20:1 m L/g、超声时间80 min、超声功率210 W,车前、大车前和平车前的总黄酮得率分别为5.04%、2.86%和1.22%。无论是纯化前还是纯化后,大车前总黄酮的还原力和对羟基自由基的清除作用最强,平车前最弱;车前总黄酮对DPPH自由基的清除作用最大,平车前最弱。纯化前后的还原力和对DPPH自由基、羟基自由基的清除作用都接近Vc的水平。  相似文献

A comparison of vanadium-rich activity of three species fungi of Basidiomycetes, Ganoderma lucidum, Coprinus comatus, and Grifola frondosa, was studied. By fermentation and atomic absorption spectroscopy analysis, the biomass of G. lucidum and G. frondosa declined rapidly when the concentration of vanadium exceeded 0.3% but the biomass of C. comatus did not decline rapidly until the concentration of vanadium exceeded 0.4% and the content of vanadium accumulated in the mycelia was 3529.3 μg/g. After the mice were administered (intragastrically) with vanadium-rich C. comatus, the blood glucose of alloxan-induced hyperglycemic mice was decreased (p < 0.05) and the body weight of the alloxan-induced hyperglycemic mice was increased gradually. Thus, we selected C. comatus to absorb vanadium and chose 0.4% as the optimal concentration of vanadium for the pharmacological works.  相似文献

Percutaneous osseointegrated implant technology provides a potential alternative to current socket prosthetics for individuals with limb loss. However, similar to other percutaneous devices, there remain concerns of periprosthetic infection. To understand this process of infection, bacterial isolates were collected and characterized from a sheep model of osseointegration. CSA-13, a novel cationic steroid antimicrobial, was used at the skin/implant interface in an attempt to reduce the rate of infection. Results indicated that in this application, normal flora and environmental organisms continued to colonize the skin/implant interface as well as cause infection in the presence of CSA-13. Two factors are believed to have contributed to this outcome: the delivery of CSA-13 and the lack of a skin seal at the skin/implant interface, which would create a biological barrier to infection.  相似文献

For the survival of individual isolates of gram-negative bacteria Pseudomonas putida, Achromobacter xylosoxidans, and the gram-positive bacterium Bacillus megaterium, in an environment polluted with crude oil products, the production of catalases exhibiting both catalase and dianisidine-peroxidase activity is important. Electrophoretic resolution of cell-free extracts of aerobically grown strains in Luria–Bertani medium during exponential phase revealed distinctive expression of catalatic and peroxidatic activities detected with 3,3′-diaminobenzidine tetrahydrochloride. A considerable diversity in microbial catalase and peroxidase responses to 20 or 40 mM H2O2 stress, resulted from hydroperoxidase’s variant of original isolates, indicating an environmental selective pressure. However, catalase was important for the adaptation of cultures to high concentration of 60 mM H2O2. Appreciable differences in the sensitivity to toxic effect of H2O2 (20 or 40 mM) treatment between individual isolates and their adapted variants during growth were observed until the middle of exponential phase, but they were insignificant at the entry to stationary phase. Isolates also exhibited a considerable diversity in catalases responses to phenolic contaminants 1 and 2 mM o- or p-phenylenediamine. Catalase activity of bacterium P. putida was visibly stimulated only by p-phenylenediamine and not by its positional isomer o-PDA. This study contributes to a better understanding of the role catalases play in bacterial responses to a polluted environment.  相似文献

In three forests that differed in annual rainfall and seasonality, the probability of a liana with a stem ≥2.0 cm stem diameter reaching the canopy was >50 percent. Lianas reached the canopy at significantly smaller size‐classes (1.5 cm) in the wet aseasonal forest, suggesting that this estimate changes with forest type. Nevertheless, as a general rule, we suggest that 2.0 cm is the minium stem diameter to examine the abundance and diversity of canopy lianas or canopy competition between lianas and trees.  相似文献
