Sucrose accumulation and enzyme activities in callus culture of sugarcane   总被引:1,自引:0,他引:1  
The activities of sucrose phosphate synthase (SPS), sucrose synthase (SUSY), neutral invertase (NI) and soluble acid invertase (SAI) were measured in callus cultures of four Mexican sugarcane cultivars (Saccharum spp.) with a different capacity to accumulate sucrose in stem parenchyma cells. The results indicated that sucrose accumulation in callus was positively correlated to the activity of SPS and SUSY and negatively to the activity of SAI and NI while SPS explained most of the variation found for sucrose accumulation and NI least.The research was funded by the department of Biotechnology and Bioengineering CINVESTAV Mexico City, and F. Screening for drought tolerance in Sorghum using cell culture   总被引:4,自引:0,他引:4  
Summary Callus growth from 10 cultivars ofSorghum bicolor (L.) Moench was measured with increasing levels of polyethylene glycol (PEG) as an osmoticum in the medium to determine whether differences among these cultivars at the cellular level in response to osmotic stress existed. These cellular ratings were compared to field ratings from the 10 tolerant-to-susceptible cultivars when grown under drought conditions to determine whether cellular ratings corresponded to differences in drought tolerance at the plant level. Callus cultures were grown on Murashige and Skoog inorganic salt formulation plus vitamins, 2,4-dichlorophenoxyacetic acid (2,4-D), kinetin and sucrose, supplemented with 0 to 25% (wt/vol) PEG corresponding to −0.2 to −1.62 MPa osmotic potential. Results suggest that PEG-induced osmotic stress on callus cultures can be used to screen sorghum cultivars for potential early field (preflowering) drought tolerance. This implies that at least a component of the early field drought tolerance in sorghum may have a cellular basis. The effect of exogenously supplied reduced nitrogen and sucrose on high-frequency somatic embryogenesis in petiole-derived tissue cultures of a diploid and a tetraploid regenerable clone of Medicago sativa ssp. falcata was investigated. There was an absolute requirement for ammonium during embryo induction and differentiation, with 5mM being the optimum for induction and 10–20 mM the optimum for differentiation of somatic embryos. Exogenous amino acids were not essential for differentiation and often even inhibitory, except 1 or 2 g/l casein hydrolysate or 4.4 mM glutamine with 3.1 mM proline which, under certain conditions, resulted in increases of 20–30% in the number of embryos obtained. High and low sucrose concentrations inhibited somatic embryogenesis and there was no reason to deviate from the 3% (0.088 M) sucrose level commonly used in plant tissue culture media. Selected clones from three M. sativa cultivars showed a response similar to the highly regenerable ssp. falcata clone F1.1.  相似文献

Three cultivars (cvs.) of Gladiolus hybridus Hort., namely ‘Her Majesty’, ‘Aldebaran’ and ‘Bright Eye’ were successfully micropropagated. The cultures were established using intact cormels or segments of cormels and inflorescence axes on Murashige and Skoog (1962; MS) medium. The response depended on media supplements; both callus formation or direct induction of shoot buds was observed. Shoot differentiation from callus could be obtained on MS medium containing 1.0 μM BA (6–benzyladenine) and 10.0 μM NAA (α-naphthalene acetic acid) in all three cultivars. The same could be achieved by giving a heat shock (HS; 50 °C, 1h) to callus cultures (in case of ‘Her Majesty’ and ‘Aldebaran’ only) maintained on the basal medium. In these two cultivars, high sucrose concentration (0.232, 0.290 or 0.348 M) also favoured growth and proliferation of shoot cultures on a plant growth regulator-free medium at 20 °C in comparison to the cultures kept at 25 °C. On the other hand, shoot cultures maintained on the basal medium at 25 °C containing normal (0.058 M, i.e., 2.0%, w/v) sucrose concentration responded similar to those maintained at 20 °C on a high sucrose medium; reduced response was observed on normal sucrose containing medium at 20 °C. Heat shock enhanced shoot proliferation in the cultures maintained on basal medium, but induced prolific rooting in shoot cultures, within 5 days of HS, on high sucrose (optimum 0.232 M) medium. While the number of roots increased at higher sucrose concentrations in the medium in case of cvs. ‘Her Majesty’ and ‘Aldebaran’, the same was found to be independent of sucrose concentration in cv. ‘Bright Eye’. Generally the rooted plants produced on high sucrose (0.232 M) medium in comparison to medium with normal sucrose concentration showed better survival (ca. 90% as against 40%) in the soil. Summary Somatic embryogenesis from nucellus-derived callus cultures of five cultivars, including three (Caipira, Seleta Vermelha, and Valencia) of sweet oranges (C. sinesis L. Osbeck), Rangpur lime (C. limonia L. Osbeck), and Cleopatra mandarin (C. reticulata Blanco) (lines I and II), were studied. Callus lines maintained on MT medium supplemented with 50 g l−1 sucrose were transferred to MT medium supplemented with different carbohydrate sources: galactose, glucose, lactose, maltose, or sucrose at 18, 37, 75, 110, or 150 mM, or glycerol at 6, 12, 24, 36, or 50 mM. Globular embryos were observed after approximately 4 wk, in several treatments. Cultures of Valencia and Caipira sweet oranges and Cleopatra mandarin (line I) showed high numbers of embryos on medium containing galactose, lactose, and maltose. Histological studies showed somatic embryos in all developmental stages with a normal histodiffeentiation pattern. The other two cultivars (Rangpur lime and Cleopatra mandarin, line II) formed very few embryos, which did not develop further following the globular stage. Some of the abnormalities observed were lack or dedifferentiation of protoderm and absence of apical meristems and procambial strands. Embryos that followed the normal sequence of development were easily converted into plants. Non-embryogenic cultures continued as proliferating callus cultures, eventually forming a few embryos which did not convert into plants. Statistical analyses of the callus response to carbohydrate treatments was done using an overdispersion Poisson model.  相似文献

The effect of media manipulatioss, temperature pretreatment, carbohydrate source, and seasonal variation on tetraploid potato anther cultures was investigated. The anther culture responses of three commercial Nordic potato varieties from Scandinavia and two from Germany were compared on different media manipulations. With most of the varieties, solid MS media gave better yields than other published media manipulations. Pretreatments at +6°C and at +30°C were studied on Pito and Danva varieties. The +6°C pretreatment and no pretreatment had the same effect on the anther culture response of cv. Pito, while with cv. Danva pretreatment at +6°C promoted embryogenesis. The +30°C pretreatment had no positive effect on anther culture response on either cultivar. The effect of maltose, melibiose and mannitol individually and in combination with sucrose were compared to normal sucrose medium in cv. Pito anther cultures. Anthers incubated on normal sucrose medium gave the highest embryoid and plant yields; the second highest plant yields were obtained on pure maltose medium. Strong seasonal variation was observed throughout the year in cv. Pito anther cultures. The percentage of anthers producing embryoids ranged from 15–20% during September and October to just 1–3% from February through May. The annual average embryoid production rate was 6.18%.  相似文献

Summary Anther culture of the Easter Lily (Lilium longiflorum; 2n=2x=24) was attempted in order to evaluate its potential in generating haploids for the production of hybrid cultivars. The effects of genotype, temperature (low temperature treatment of buds and high temperature treatment of cultures), sucrose concentration and growth regulators were tested. The most important factors for callus induction were the genotype and the presence of 2,4-dichlorophenoxyacetic acid. Pre-treatments at low or high temperature had no apparent effect, while high sucrose concentration was inhibitory. Callus was derived from 28 of the 108 genotypes tested and plants were regenerated. Phenotypic variations were observed among these regenerants. Somatic chromosome numbers were determined in 42 plants derived from 10 donor genotypes. Thirteen plants were diploid and 29 were mixoploid with chromosome numbers ranging from 11 to 26. Four of the mixoploid plants had a high proportion of cells with haploid chromosome numbers, particularly at early stages of development. Meiosis was examined in plants with flower buds. Most plants had 12 bivalents at Metaphase I, but also aneuploids were observed. Other irregularities included bridges and laggards at Anaphase I. The occurrence of high frequencies of haploid cells (up to 80%) in root tips suggests that some plants may be of gametic origin. Defined media that promote the initiation and undifferentiated growth of callus derived from stem explants of four cultivars of cassava, Manihot esculenta Crantz, are described. Growth rates and yields of cassava callus after 4 weeks of culture are shown to be comparable to those of callus of Nicotiana tabacum L. cv. Wisconsin No. 38. Nitrogen sources of ammonium nitrate or of ammonium chloride plus succinate supported growth of all four cultivars. Sucrose was superior to glucose as a carbon source. The cassava cultivars differed in their response to increasing concentrations of sucrose between 0.5% (w/v) and 3%, two of them increasing in dry matter with increasing sucrose concentrations of up to 3%. When cultured in the light on defined media that contained higher ratios of cytokinin to auxin, callus of the latter two cultivars turned green. Roots but not shoots differentiated from the callus of all cultivars. The influence of hormone concentrations, sucrose level, and nitrogen source on greening and root formation is summarized.  相似文献

The effects of short-term exposure to chilling temperature (10 °C) on sucrose synthesis in leaves of the cold-tolerant sugarcane cultivars Saccharum sinense R. cv. Yomitanzan and Saccharum sp. cv. NiF4, and the cold-sensitive cultivar S. officinarum L. cv. Badila were studied. Plants were grown at day/night temperatures of 30/25 °C, and then shifted to a constant day/night temperature of 10 °C. After 52-h exposure to the chilling temperature, sucrose content in the leaves of NiF4 and Yomitanzan showed a 2.5- to 3.5-fold increase relative to that of the control plants that had been left on day/night temperatures of 30/25 °C. No such increase was observed in Badila leaves. Similarly, starch content in the leaves of NiF4 and Yomitanzan was maintained high, but starch was depleted in Badila leaves after the 52-h exposure. During the chilling temperature, sucrose phosphate synthase (SPS; E.C.2.4.1.14) activity was relatively stable in the leaves of NiF4 and Yomitanzan, whereas in Badila leaves SPS activity significantly decreased. There was no significant change in cytosolic fructose-1,6-bisphosphatase activity for the three cultivars at the chilling temperature. This supports the hypothesis that: (1) on exposure to chilling temperature, sucrose content in sugarcane leaves is determined by the photosynthetic rate in the leaves, and is not related to SPS activity; (2) SPS activity in sugarcane leaves at chilling temperature is to be determined by sugar concentration in the leaves.  相似文献

Shoot clump cultures of Narcissus cultivars St. Keverne and Hawera were used to investigate the effects of culture medium carbon supply, type of carbohydrate and osmolarity on in vitro bulblet development. Increasing the medium osmolarity using mannitol or sorbitol, which did not act as substrates for growth, failed to stimulate bulblet formation with either cultivar. An exception to this was a relatively small increase in total bulblet dry weight per culture, in the cultivar Hawera only, caused by adding 30 g l –1 sorbitol in combination with 30 g l–1 sucrose. Simultaneously increasing the medium osmolarity and carbon supply using the metabolisable carbohydrate sources, sucrose, glucose, fructose or an equimolar mixture of glucose and fructose stimulated bulblet production, total dry matter accumulation and partitioning into bulblets. At comparable levels of carbon supply up to 19.0 g l–1, bulblet development of both cultivars was similar with monosaccharide and sucrose media. This indicates that substrate supply is more important for bulblet development than osmolarity of the culture medium. The cultivar Hawera also showed similar responses to monosaccharide and sucrose media supplying 37.9 g C l–1, despite the high osmolarity of monosaccharide media (c. 650 m Osm kg–1, equivalent to –1.6 MPa, compared to 380 m Osm kg–1 for sucrose medium). However in St. Keverne total dry matter accumulation and dry weight per bulblet were further stimulated only by increasing the sucrose supply from 19.0 to 37.9 g C l–1, not by increasing the monosaccharide supply. Implications of the findings for Narcissus micropropagation are discussed.  相似文献

Water stress effects on photosynthesis in different mulberry cultivars   总被引:10,自引:0,他引:10  
The effect of water stress on photosynthesis was determined in five mulberry cultivars (Morus alba L. cv. K-2, MR-2, BC2-59, S-13 and TR-10). Drought was imposed by withholding water and the plants were maintained at different water potentials ranging from 0.5 -MPa to 2.0 -MPa. Photosynthetic rates, activities of ribulose-1,5-bisphosphate carboxylase and sucrose phosphate synthase, photosystem II activity and chlorophyll content were used as key parameters to assess photosynthetic performance. There was a marked variation in the photosynthetic rates and ribulose-1,5-bisphosphate carboxylase activity among the five mulberry cultivars subjected to water stress. Photosystem II (PSII) and sucrose phosphate synthase activities were also severely reduced as measured by drought conditions. Of the five mulberry cultivars, S-13 and BC2-59 showed higher photosynthetic rates, ribulose-1,5-bisphosphate carboxylase activity, high sucrose phosphate synthase activity and photochemical efficiency of PSII compared to the other varieties.  相似文献

Global mean temperatures are expected to rise by 2–4.5°C by 2100, accompanied by an increase in frequency and amplitude of extreme temperature events. Greater climatic extremes and an expanded range of cultivation will expose rice to increasing stress in the future. Understanding gene expression in disparate thermal regimes is important for the engineering of cultivars with tolerance to nonoptimal temperatures. Our study investigated the proteomic responses of rice cell suspension cultures to sudden temperature changes. Cell cultures grown at 28°C were subjected to 3‐day exposure to 12 or 20°C for low‐temperature stress, and 36 or 44°C for high‐temperature stress. Quantitative label‐free shotgun proteomic analysis was performed on biological triplicates of each treatment. Over 1900 proteins were expressed in one or more temperature treatments, and, of these, more than 850 were found to be responsive to either of the temperature extremes. These temperature‐responsive proteins included more than 300 proteins which were uniquely expressed at either 12 or 44°C. Our study also identified 40 novel stress–response proteins and observed that switching between the classical and the alternative pathways of sucrose metabolism occurs in response to extremes of temperature.  相似文献

The level, time of application and duration of the high temperature treatment necessary for embryo production from Brussels sprouts anther culture were examined. The effects of 29, 32, 35, and 38°C given for 24 h immediately following removal of the anthers from the bud, were tested on different cultivars, on different plants within the cultivars and on different occasions for each plant. Most embryos were produced following 32 and 35°C, very few following 30°C and none following 38°C. Although there was a tendency for some cultivars to respond better to one or other of the two more favourable temperatures, this varied considerably between individual plants. Plant to plant variation was also seen in the overall level of the response, although responsiveness tended to decline with successive samplings of the same plant. Experiments with cultivars Hal and Gower suggested that high temperature was required for at least 12 h after anther removal, but beyond that time the optimum period varied from plant to plant. If the excised anthers were held at 25°C for 16 h or more with Hal or 24 h or more with Gower before being exposed to the high temperature treatment, embrogenesis tended to be reduced. It is suggested that apparent non-responsiveness in anther culture may result to a large extent from the specific conditions that are used during the anther culture process.  相似文献

Summary Initial trials with tomato-root cultures disclosed the desirability of employing a gently agitated liquid medium containing iron in the chelated form. For the normal cultivars “Ace” and “Tropic”, subcultures were best achieved by utilizing sectors that possessed one or more newly emerged laterals. Continuous cultures of a nonlateral-forming tomato mutant, “Diageotropica”, and of citron were accomplished by subculturing tips of the elongating primary roots. The tomato roots were cultured in White's medium with the Fe2(SO4)3 replaced by 0.03 mM NaFeEDTA. Sustained growth of citron-root tips necessitated the use of a medium containing Murashige and Skoog salts, 7.5% sucrose, 100 mg per I each of citric acid and thiamine HCl, and 5000 mg per li-inositol. The success with citron-root cultures was extendable to all cultivars ofC. medica L., but not to otherCitrus species relatives. Both citron and “Diageotropica” root cultures manifested undiminished elongation through repeated subcultures; but neither produced laterals in response to any cultural treatments. Climate change predictions foresee a combination of rising CO2, temperature and altered precipitation. Effects of single climatic variables are well defined, but the importance of combined variables and genotypic effects is less known, although pivotal for assessing climate change impacts, for example, with crop growth models. This study provides developmental and physiological data from combined climatic factors for two distinct wheat cultivars (Paragon and Gladius), as a basis to improve predictions for climate change scenarios. The two cultivars were grown in controlled climate chambers in a fully factorial setup of atmospheric CO2 concentration, growth temperature and watering regime. The cultivars differed considerably in their developmental rate, response pattern and the parameters responsible for most of their variation. The growth of Paragon was linked to climatic effects on photosynthesis and mainly affected by temperature. Paragon was overall more negatively affected by all treatment combinations compared to Gladius. Gladius was mostly affected by watering regime. The cultivars' acclimation strategies to climate factors varied significantly. Thus, considering a single factor is an oversimplification very likely impacting the accuracy of crop growth models. Intraspecific crop variation could help understanding genotype by environment variation. Cultivars with high phenotypic plasticity may have greater resilience against climatic variability.  相似文献

Summary Nine soybean [Glycine max (L.) Merr.] cultivars representing midwestern, mid-south, and southern US growing regions were evaluated at each of three locations (Athens, GA; Lexington, KY; and Wooster, OH) using uniform embryogenic induction and proliferation protocols in order to evaluate the portability of soybean somatic embryogenic protocols to different locations. The experimental design minimized variation between locations by having all cultivars present at all locations on all days. A quantitative weighted score for primary embryo induction was developed on average embryo number per explant and was used to describe non-embryogenic, poorly embryogenic, moderately embryogenic, and highly embryogenic responses. Ranking of cultivars remained similar across all locations, indicating a uniform transportability of the protocol, at least as far as embryo induction is concerned. Continued proliferation of embryogenic cultures was also measured using a repetitive growth measure but few meaningful conclusions could be made due to the high level of variability including inconsistent growth of cultures between each subculture. Overall, several cultivars were identified as being uniformly embryogenic or non-embryogenic at the primary induction phase at all locations, and we predict that those embryogenic cultivars could be used by any laboratory for high-efficiency induction of embryogenesis. The best of these cultivars, ‘Jack’, was uniformly responsive across all locations and should be selected as the genotype most likely to yield positive results when attempting to culture and genetically engineer soybeans via embryogenic protocols.  相似文献

Seventy-six cultivars of alfalfa (Medicago sativa L., M. falcata L. and M. varia Martyn) were tested in vitro for their capacity to produce callus and somatic embryos. A three-step media protocol was used to survey the response of the cotyledons and hypocotyl of each genotype while the epicotyl region was conserved in order to recover highly responding genotypes. The best regeneration response was observed in creepingrooted cultivars which contained a strong genetic contribution of two landrace germplasm sources, defined as M. falcata and Ladak, in their ancestry. The callus and embryogenesis responses showed a high degree of variation both between cultivars and among the plants of many of the 76 cultivars tested. A higher number of plants produced somatic embryos in the high regenerating cultivars compared to the low regenerating cultivars regardless of the media protocol or explant.  相似文献

One cultivar of winter oil seed rape (Brassica napus ssp. oleifera)and three cultivars of spring rape were used in a study of theeffects of sucrose on microspore survival and embryo inductionin cultured anthers. A preliminary study on the winter cultivar(Fiona) revealed that the osmotic pressure of the supernatantof anther homogenates was equivalent to a solution of 17% sucrose.A study of microspore survival and embryo induction in thiscultivar on media containing either 8 %, 12%, 16% or 20% sucroserevealed the highest survival (after 16 d) and the greatestnumber of anthers with induced embryos (after 42 d) occurredon the highest sucrose concentration. A subsequent study on three spring cultivars (Willi, Duplo andTower) examined microspore survival at 8 d and embryo induction(42 d) on media containing either 8 % or 16 % sucrose and againrevealed much higher survival and induction at the higher concentration.The variation in response between the cultivars was also reducedby culture at the higher sucrose concentration. The beneficialeffect of the 16% level occurred regardless of the growth environmentof the donor plants and of the stage of pollen development atthe start of culture. Rhizobium sp. 127E15 fixed nitrogen asymbiotically, when grown in induction media. Highest level of acetylene reduction activity was reached in LNB5 medium and in media containing a combination of two sugars, sucrose and arabinose. In the induced cultures, large pleomorphic forms of bacteroids were produced. Considerable acetylene reduction activity was recorded in the rhizosphere of the lima bean plants that were inoculated with rhizobia and grown in pot cultures. Trace amounts of activity could also be detected in the rhizobia adhering to the rhizoplane.  相似文献
