Although theCyclamen subg.Cyclamen spp. are morphologically variable, previous studies suggest a superficial cytological uniformity. New chromosome numbers and an indication of karyotypic instability are reported inC. hederifolium andC. africanum that reveal that the cytology of the subgenus is more complicated than previous accounts suggest. The possible significance of these phenomena is discussed. The diploid status ofC. purpurascens has been confirmed and a distinction between the three diploid karyotypes has been described. The cytological variation may help to explain the well documented morphological variation exhibited in these species.  相似文献

Summary Variation in nuclear DNA amounts found in different species of Cucumis was surveyed. The DNA amounts varied from 1.373 to 2.483 pg in diploids and from 2.846 to 3.886 pg in tetraploids. DNA amount was not correlated with chromosome number and periodicity. Tetraploids were found to have double the quantity of nuclear DNA of diploids. A positive linear relationship was established between the nuclear DNA amounts and volume of chromosomes. The botanical varieties within a particular species do not differ significantly for 2C DNA amounts. A comparison of the distribution of DNA amounts among different chromosomes of haploid complement in different species revealed that the quantitative DNA changes associated with speciation affected all chromosomes. DNA changes were not however, of the same magnitude in all chromosomes of the complement. Speciation in Cucumis thus seems to have occurred through amplification or diminution of DNA proportionate to the size of chromosomes. The relationship between the basic numbers, x=7 and x=12, will have to be considered relative to the high DNA amount noticed in some species with x=12.  相似文献

A South African carnation species Dianthus zeyheri subsp. natalensis was cultured in vitro using techniques similar to those developed for the cut carnation (Dianthus caryophyllus L.). Crepis dinarica andC. froelichiana are two closely related species of theC. praemorsa complex. Even though they exhibit the same chromosome number (2n = 8) and similar idiogram shape, they differ widely in quantity and distribution of heterochromatin bands. The hybrids between these two species comprise three morphological types. Parental genomes were distinguished in hybrids by Giemsa differential staining (C-banding). Although meiosis presents only a few abnormalities (about 2.4%), the percentage of aborted pollen grains is very high (90%).  相似文献

Variability and genetic divergence of 11 field populations of seven species of theCampanula elatines andC. fragilis rock-plants group from the Mediterranean and pre-Alpine areas have been studied by starch-gel electrophoretic techniques.Campanula isophylla, C. elatines, C. elatinoides, C. fragilis subsp.fragilis and subs.cavolinii, C. garganica were collected in Italy, whilstC. fenestrellata subsp.istriaca andC. portenschlagiana came from W. Jugoslavia. Twelve enzymatic loci for each population were genetically analysed: PGI-1 and 2, PGM-2, IDH-1 and -2, SKDH, ME, ADH, GOT-2, MPI-1 and -2, SOD-1. The genetic distances among the above mentioned entities have been calculated by Nei's index and depicted in a dendrogram.  相似文献

Summary Somatic fusions between the cultivated potato Solanum tuberosum and the wild species S. circaeifolium subsp. circaeifolium Bitter were produced in order to incorporate desirable traits into the potato gene pool. Selection of the putative hybrids was based on a difference in callus morphology between the hybrids and their parents, with the hybrids showing typical purple-colored cells in otherwise green calli. In all, 17 individual calli regenerated to plants. Of the nine plants that could be transferred to the greenhouse, eight showed a hybrid and one a parental morphology. Restriction fragment length polymorphism (RFLP) analysis confirmed the hybrid character in the former group. Chloroplast counts in stomatal guard cells and flow cytometric determination of nuclear DNA content showed that four hybrid plants were tetraploid (4x), one was mixoploid (5x–8x), and the others were polyploid (6x; 8x). Three out of four tetraploid hybrids were found to be fully resistant to Phytophthora infestans, and all four hybrids were resistant to Globodera pallida pathotypes Pa2 and Pa3. It was further observed that the type and amount of steroidal glycoalkaloids varied among the tubers of the parents and the hybrids. Using the hybrids as female parents in crosses with S. tuberosum, viable seeds could be obtained. This demonstrates the potential of these hybrids in practical plant breeding.  相似文献

Summary Crossability between the diploid species S. circaeifolium subsp. circaeifolium (crc) and other diploid species, primarily diploid S. tuberosum subsp. tuberosum (tbr-2x), was studied. Forty-seven hybrids were obtained from crosses between crc as female parent and tbr-2x and some other species from series Tuberosa as male parents. Of these hybrids 17% were diploids; the other 83% were triploids, probably carrying two genomes of crc. Female fertility was sufficient to obtain offspring from backcrosses with the cultivated parent. Pollen stainability of the f1 varied, and micro-pollen as well as unreduced pollen occurred. During meiosis of the diploids and triploids a rather high proportion of univalents was found, and in the triploids on average two or three trivalents per cell were found. All hybrids were resistant to Globodera pallida pathotypes 2 and 3, and 75% of the tested genotypes were highly resistant to Phytophthora infestans. Solanidine, tomatidine, tomatidenol, and demissidine glycosides were found in tubers of the hybrids. Comparisons with somatic hybrids between crc and tbr-2x are made. It is concluded that crc is a valuable Solanum species that can and should be included in potato breeding programs.  相似文献

Investigations on interspecific crossability in 8Cucumis species (2n = 24) and chromosome pairing and pollen fertility of their hybrids from 15 combinations have been utilized for tracing the phylogenetic relationships among these taxa and factors responsible for their differentiation. A collective evaluation of data suggests that there are three broad groups of species, one of the spiny fruited interfertile species, whose hybrids show varying degree of chromosome associations and low to high pollen fertility; the second of species with non-spiny fruits, which are completely incompatible with the former but weakly compatible with the cultivated species,C. melo L. to produce partly developed seeds, and the third group ofC. metuliferus E. Mey. exSchrad. andC. melo and its different botanical varieties. Somatic chromosome numbers have been determined for the followingCerastium taxa:C. eriophorum (2n = 36),C. alpinum (2n = 72),C. transsylvanicum (2n = 108),C. arcticum (2n = 108),C. latifolium (2n = 36),C. carinthiacum (2n = 36),C. banaticum (2n = 36),C. arvense subsp.glandulosum (2n = 36),C. arvense subsp.arvense (2n = 72) andC. fontanum (2n = 144). Karyotypes of three diploid species (C. eriophorum, C. banaticum andC. latifolium), belonging to three different taxonomic groups, were analysed and found to be similar. The relative nuclear DNA contents of all taxa were determined by flow cytometry and, for five species, also by Feulgen cytophotometry. The values obtained by the two methods are similar. A comparison of nuclear DNA contents among diploids shows that values differ significantly between different taxonomic groups, and are correlated with average chromosome size. Within closely related polyploid groups nuclear DNA amounts increase from 2x- to 4x- and 6x taxa as 1 : 1.4 : 2.4 in theC. alpinum complex, whereas DNA amounts are doubled comparing 2x- and 4x-subspecies in theC. arvense complex.  相似文献

Summary A restriction map of the Cucumis melo L. (melon) plastome was constructed by using several mapping approaches: single and double digestions of the chloroplast DNA (chlDNA) with endonucleases (XhoI, SmaI, SacI and PvuII) and hybridization to heterologous chlDNA probes and to isolated melon chlDNA fragments. Four plastome-coded genes were located using heterologous probes. The overall organization and gene position of the melon plastome was found to be similar to that of tobacco and other angiosperm species. Restriction patterns based on digestion of the chlDNA with nine endonucleases were obtained in over 20 wild species and cultivated varieties of Cucumis. These led to mutational analysis of the restiction sites yielding the most parsimonious phylogenetic tree of the Cucumis plastome. Most African species from a compact group (Anguria group) which is distant from the melon, the cucumber and a few other species (C. sagittatus, C. metuliferus and C. humifructus). All of these are also far apart from each other. The distribution of polymorphic restriction sites along the Cucumis plastome is described and conservative regions as well as hot spots are suggested.  相似文献

Summary The 68C puff is a highly transcribed region of theDrosophila melanogaster salivary gland polytene chromosomes. Three different classes of messenger RNA originate in a 5000-bp region in the puff; each class is translated to one of the salivary gland glue proteins sgs-3, sgs-7, or sgs-8. These messenger RNA classes are coordinately controlled, with each RNA appearing in the third larval instar and disappearing at the time of puparium formation. Their disappearance is initiated by the action of the steroid hormone ecdysterone. In the work reported here, we studied evolution of this hormone-regulated gene cluster in themelanogaster species subgroup ofDrosophila. Genome blot hybridization experiments showed that five other species of this subgroup have DNA sequences that hybridize toD. melanogaster 68C sequences, and that these sequences are divided into a highly conserved region, which does not contain the glue genes, and an extraordinarily diverged region, which does. Molecular cloning of this DNA fromD. simulans, D. erecta, D. yakuba, andD. teissieri confirmed the division of the region into a slowly and a rapidly evolving protion, and also showed that the rapidly evolving region of each species codes for third instar larval salivary gland RNAs homologous to theD. melanogaster glue mRNAs. The highly conserved region is at least 13,000 bp long, and is not known to code for any RNAs.  相似文献

Summary An electrophoretic comparison of 29 nuclear-coded enzymes was carried out for 21 Cucumis species, and a phylogeny based on pairwise measurements of the respective genetic distances was computed. This phylogeny was compared to the one based on chlDNA cariation (Perl-Treves and Galun 1985). The two phylogenies were found to share the main dendrogram features; they also agree well with most taxonomic data available on Cucumis. Accordingly, most of the African Cucumis species form a close group (Anguria group — Group A), which is distant from the melon (C. melo), and from a few other distinct species, all of which are far apart from each other. The cucumber (C. sativus) is the most distant species within the genus. Some specific taxonomic implications as well as some general evolutionary problems related to such a parallel investigation of the nuclear genome and the plastome are evaluated.This publication is dedicated to Dr. T. Seed storage proteins were analyzed for variation in polypeptide patterns and proportion of four protein fractions in Cucumis melo and for interrelationships of different taxa of the genus Cucumis. SDS-PAGE of total protein extracts of 11 lines of C. melo representing different geographic regions showed considerable variation in their polypeptides in the range of molecular weights 50–55, 35–39, 20–26 and 16–19 kDa. As compared to one subunit pair reported earlier, two-dimensional gel electrophoresis revealed subunit pairs of molecular weights 53, 52, 50, 42, 39 and 23 kDa, each consisting of a large and a small subunit like those of legumin-like 11S proteins of leguminous seeds. The salt-soluble globulins represented the major protein fraction (44.8–48.1%), followed by glutelins and albumins with prolamins being the lowest in the seed. Based on the protein profiles of seed protein extracts of 28 Cucumis taxa, similarity matrix indices and UPGMA dendrogram showed that eight taxa of C. melo were clustered in two subclusters of four taxa each. Five taxa of C. sativus along with C. zeyheri belonged to another cluster, sister to the third cluster of remaining taxa. In this third cluster, C. anguria var. longaculeatus showed 100% similarity with C. myriocarpus and C. myriocarpus ssp. leptodermis as compared to only 64% similarity with C. anguria; C. myriocarpus ssp. myriocarpus occurred along with C. prophetarum in the same group rather than with C. myriocarpus. The accession PI-299570 representing an undescribed taxon of Cucumis exhibited 100% similarity with C. pustulatus, and C. sagittatus was the most distant from all Cucumis taxa.  相似文献

Genomic relationships of 13 tetraploid species within the AsiaticElymus parviglumis group containing the SY genomes were assessed by analysing chromosome pairing at metaphase I of the parental species and their interspecific hybrids. Two major genomic subgroups among the tetraploids were identified from the cluster analysis of the averaged c-values, namely, theE. caucasicus subgroup (two species) and theE. parviglumis subgroup (11 species). Summary The structure of the ribosomal DNA has been analyzed in three species of theCynareae tribe using Southern blot hybridization.Silybum marianum possesses two types of ribosomal genes 12.3 and 13.4 kb long;Cirsium vulgare has at least four types of rDNA repeats 10.6, 10.5, 11.7 and 11.9 kb long;Carlina acaulis only one type of ribosomal genes 9.7 kb long. The rRNA genes of the three species studied showed an identical restriction mapping in the 18 S and 25 S regions. However species differentiation in length and/or nucleotide sequences are present in the external spacer and very probably in the internal transcribed spacer.By cytophotometric studies and byin vitro rRNA/DNA filter hybridization, the DNA amount/4 C nucleus and the rDNA percentage were calculated in nine species of theCynareae tribe:Cynara cardunculus subsp.scolymus (artichoke),Cynara cardunculus subsp.cardunculus (wild artichoke),Onopordon acanthium, O. illyricum, Galactites tomentosa, Carduus nutans, Silybum marianum, Cirsium vulgare andCarlina acaulis. The DNA amount/4 C nucleus in eight species are similar, ranging from 4.24 pg inGalactites tomentosa to 5.96 pg inCirsium vulgare, whileCarlina acaulis has a DNA amount/ 4C nucleus of 11.8 pg. The rDNA percentages range from 0.192% inOnopordon acanthium to 1.022% inSilybum marianum, whileCarlina acaulis has 0.038% of rDNA. This latter species is clearly distinct within theCynareae tribe.  相似文献

The phylogenetic relationships within the genus Cucumis (a total of 25 accessions belonging to 17 species) were studied using the nuclear ribosomal DNA internal transcribed spacer (ITS) region. The analysis included commercially important species such as melon (C. melo L.) and cucumber (C. sativus). Two additional cucurbit species, watermelon and zucchini, were also included as outgroups. The data obtained reflected the clustering of Cucumis species in four main groups, comprising accessions from cucumber, melon, C. metuliferus and the wild African species. Some of the species clustered in different positions from those reported in classifications previously described by other authors. The data obtained clearly identify a division between the 2n=2x = 14 species (C. sativus) and the 2n = 2x = 24 ones (C. melo and wild species). Within the wild species we identified a subgroup that included C. sagittatus and C. globosus. Oreosyce africana, also classified as Cucumis membranifolius, was shown to be nested within Cucumis. Three accessions previously classified as independent species were shown to be genotypes of Cucumis melo. A set of melon and cucumber SSRs were also used to analyse the Cucumis species and the results were compared with the ITS data. The differential amplification of the SSRs among the accessions made it possible to distinguish three main groups: melon, cucumber and the wild species, though with less detail than applying ITS. Some SSRs were shown to be specific for melon, but other SSRs were useful for producing PCR fragments in all species of the genus.We are grateful to NCRPIS, IPK in Gatersleben, Semillas Fitó S.A., Michel Pitrat and Fernando Nuez for providing seeds. Populations ofC. cartwrightianus, C. hadriaticus andC. thomasii from the Balkans have 2n = 16 as has the Middle EasternC. pallasii subsp.haussknechtii. C. dispathaceus andC. pallasii subsp.pallasii have 2n = 14 whileC. pallasii subsp.turcicus has 2n = 12.C. niveus has 2n = 28 andC. sativus is an autotriploid with 2n = 24. Phytogeographical, morphological, and molecular evidence for the widespread but sporadic occurrence of sterile hybrids betweenLeucaena leucocephala subsp.glabrata andL. esculenta subsp.esculenta in South-Central Mexico is presented. Most morphological and DNA characters studied in the putative hybrids showed states intermediate between the proposed parental taxa. The occurrence of non-additive nuclear ribosomal DNA phenotypes is discussed and the need to use a suite of nuclear taxon-specific markers to determine hybridity is emphasized. The origin of the hybrid is discussed in relation to the disruption of the distributions of both parental taxa through use by man as minor food plants, providing another example of the important influence of human interference on the evolution ofLeucaena. The successful use of dried leaf material as a source of DNA is highlighted as an efficient way to identify sterile hybrids at the molecular level.  相似文献

Variation at six polymorphic enzymatic loci was studied in 50 populations of the selfing annualPolygonum aviculare from Belgium. The results show that theP. aviculare complex has an allopolyploid origin and comprises two main genetic entities, viz.P. aviculare subsp.depressum (2n = 4x = 40), restricted to urban sites subject to trampling stress, andP. aviculare subsp.aviculare (2n = 4x = 40, 2n = 6x = 60), with a wider ecological amplitude. The latter had a higher genetic diversity and a higher frequency of fixed heterozygosity than the former. WithinP. aviculare subsp.aviculare, genetic diversity and ecological amplitude correlated positively with ploidy level. Additionally, stronger genetic differentiation among and lower variation within populations was found in urban areas than in cultivated fields.  相似文献

Based on an analysis of results from experimental hybridization, the plants assigned byMunz toOenothera subg.Oenothera and subg.Raimannia, now divided into approximately 76 species, are referred to a single section,Oenothera. This section is in turn divided into five subsections:Euoenothera, Munzia, Raimannia, Emersonia, and an undescribed group of three species related toOenothera pubescens. Euoenothera is maintained in the traditional sense, and includes about 14 species of North America, widely naturalized elsewhere.Munzia consists of 45 species, comprising three series, and native to South America.Raimannia is restricted to a group of approximately 11 North American species.Emersonia comprises four rather heterogenous species of northern Mexico and southern New Mexico, of whichOenothera macrosceles, O. maysillesii, andO. organensis have been described. Within these four subsections, interspecific hybrids can be made in general, although plastid differentiation often leads to incompatibilities. With varying degrees of difficulty, hybrids were produced in all intersectional combinations involvingEuoenothera, Emersonia, Munzia, andRaimannia, the most difficult being those betweenEmersonia andRaimannia. Based on their habit and distribution,Emersonia species, and especiallyOenothera maysillesii, appear to resemble most closely the common ancestor of the section,Euoenothera andMunzia to have been derived from it or its common ancestor, andRaimannia perhaps to be more closely related to the phylogenetic branch that leads toEuoenothera.  相似文献
