Proteolytic activity was estimated in germinated finger millet seedlings using the endogenous trypsin/amylase inhibitor as substrate and also with haemoglobin and albumin as substrates. The maximal proteolytic activity was observed on the third day of germination. With the inhibitor as substrate, the proteolytic activity was maximal at pH 2.5. The protease that acted on the inhibitor required sulphydryl groups for maximal activity and was suppressed by diazoacetyl norleucine methyl ester and Pepstatin. The protease that acted on haemoglobin with optimum pH of 5.0, was more stable on storage, did not depend on sulphydryl groups for activity and was unaffected by reagents that react with carboxyl groups.  相似文献

The seeds of twelve common species of legumes were examined for the release of proteinase inhibitor activity during germination. All species released inhibitory activity against bovine trypsin (EC 3.4.21.4), ranging from 1.0 unit per g dry wt. of seed in 24 hr for soybean (Glycine max), to 0.07 unit per g for broad beans (Vicia faba) and sugar pod peas (Pisum sativum). This release corresponds to approximately 1–13 % of the total trypsin inhibitory activity of the seed, with lentils (Lens culinaris) releasing the greatest percentage, and the scarlet runner bean (Phaseolus coccineus) the least. In most species the amount of inhibitor released increases until 24–48 hr of germination, and then remains roughly the same or decreases slightly by 72 hr of germination. Five species of legumes were also examined for the release of inhibitory activity against bovine chymotrypsin (EC 3.4.21.1). In each case chymotryptic inhibitory activity was released in a manner similar to the trypsin inhibitor.  相似文献

Refuse compost and sewage sludge were mixed with a loamy sand at various rates in pots and sown withBrassica chinensis, Daucus carota andLycopersicon esculentum in a glasshouse. A commercial fertilizer was also applied to the same soil for comparison. Dry matter production of the three crops and contents of Cd, Cu, Mn, Pb and Zn in the harvested tissues were determined at the end of the experiment. In general, crop yield in refuse compost treatment was improved over that in sandy soil alone, but was less than that in the sludge and fertilizer treatments. Despite the relatively high heavy metal contents of refuse compost, crops grown on compost-treated soils accumulated lower levels of metal than those grown on sludge-treated soils. This is probably due to the high pH and organic matter content of the composted refuse. Higher levels of heavy metals were found in the roots than in the aerial parts ofB. chinensis andL. esculentum, but the reverse was found inD. carota. In the edible tissue of the three crops,L. esculentum accumulated metals to a lesser extent than the other two.  相似文献

The effect of various abiotic stresses on germination rate, growth and soluble sugar content in Sorghum bicolor (L.) Moench cv. CSH 6 seed embryos and endosperm during early germination was investigated. Under stress conditions germination, water potential and tissue water content decreased markedly. Subsequently, this reduction resulted in marked decreases in fresh weight both in embryos and endosperm. Conversely, a substantial increase in dry weight was observed. Furthermore, a considerable increase in the sugar contents in both embryo and endosperm was detected. The fructose level was always higher than glucose and sucrose in response to various stresses. However, as compared to the control the level of glucose and sucrose was higher in embryos and endosperm after stress treatments. Based upon these results a possible physiological role of sugars in the germination of sorghum seeds is discussed.  相似文献

Summary A procedure for culturing detached panicles of sorghum, Sorghum bicolor (L.) Moench, was developed to achieve flowering, fertilization, and subsequent seed development and maturation in vitro. Sixteen sorghum genotypes (five high and eleven low in tannin) were tested for their ability to develop normally in culture. Panicles collected one to two days before the initiation of anthesis were cultured in flasks containing liquid medium. Contamination and medium darkening were the major obstacles encountered. Up to 55% of the panicles cultured reached physiological maturity in vitro. The frequency of seed set ranged from 30 to 97% depending upon genotype and medium. Seed and glume color were normal. Seed produced in vitro resembled those grown in vivo and germinated well, but were smaller than normal (100 kernel weight reached 50 to 70% of the control). Grain polyphenols were synthesized in the cultured panicles. Seed of high tannin genotypes produced in vitro were lower in total phenols and tannins and higher in flavan-4-ols and the 3-deoxyanthocyanidin pigments than control seed. This technique can be used for harvesting late-maturing stocks and for various sorghum studies.  相似文献

Enzymatic hydrolysis of glucosinolates, a class of compounds found in Brassica species, results in a number of products with potential to inhibit seed germination. To investigate the impact of both volatile and water-soluble allelochemicals, germination bioassays using Lactuca sativa seeds were conducted with root and combined leaf and stem tissues of Brassica napus. Tissues in which glucosinolates were hydrolyzed to remove volatile glucosinolate degradation products were compared with intact tissues and water controls. Only tissues containing glucosinolates produced volatiles that inhibited germination. Volatiles were trapped and identified using GC-MS. Volatiles produced in greater quanitity from intact tissues than from tissues without glucosinolates were almost exclusively glucosinolate hydrolysis products. Water-soluble components also inhibited germination. Chemical analysis of extracts confirmed the presence of glucosinolate hydrolysis products, but indicated the involvement of additional allelochemicals, especially in leaf and stem tissues. The oligochaete worm Nais elinguis was counted during a year and a half in a full-scale, completely mixed, municipal activated sludge plant consisting of four aeration tanks connected in parallel. Simultaneously the operating variables, i.e. effluent quality, energy costs in kWh for oxygen supply in the aeration tanks, and sludge-disposal were measured. The number of worms varied both seasonally and among the aeration tanks. A major worm bloom resulted in a low sludge volume-index, lower energy consumption for oxygen supply expressed in kWh and, depending on the temperature, less sludge-disposal. The worms had no influence on the effluent quality.  相似文献

Conditions have been identified that allow reproducible amplification of RAPD markers in sorghum. High resolution of RAPD markers was accomplished by radiolabeling PCR-amplified DNAs followed by separation on denaturing 5% polyacrylamide gels. Reaction parameters including MgCl2 concentration and temperature significantly influenced yield and the type of amplification products synthesized. Unexplained amplified DNAs increased when more than 35 cycles of PCR amplification were used. Under standard conditions, approximately 80% of the primers tested amplified DNA, and most revealed 1–5 polymorphisms between BTx 623 and IS 3620C. Primers were used to amplify RAPDs in 32 genotypes of sorghum. In addition, 8 primers detected RAPDs in a population previously used to create an RFLP map for sorghum. These RAPDs were mapped successfully using a population of 50 F2 plants.  相似文献

A short period (15–30 min) at 30° C promotes germination of seeds of Lactuca sativa L. cv. Repolhuda in darkness. Far-red light reverses this stimulation, and the escape curves for phytochrome and high-temperature action are quite similar, indicating that the two factors act at a common point in the chain of events leading to germination. Raffinose family oligosaccharides (RFOs) are almost ubiquitous in seeds and have been hypothesized to constitute an important energy source during germination. To test this hypothesis we applied a specific alpha-galactosidase inhibitor (1-deoxygalactonojirimycin, DGJ) to germinating pea seeds, resulting in a complete blocking of RFO breakdown. The germination rates of DGJ-treated seeds dropped drastically to about 25% of controls two days after imbibition. Similarly, the activities of the key enzymes in the galactose salvage pathway galactokinase, UDP-galactose pyrophosphorylase and UDP-galactose 4'-epimerase, were also significantly lower in seeds treated with the inhibitor. The inhibitory effect on germination could be relieved by galactose but only partially by sucrose, indicating that galactose, in addition to providing easily available energy for growth, may also be an important component of the sugar signaling pathway during germination. Taken together our study, for the first time, provides clear evidence that RFOs play an important role for early germination.  相似文献

Pulsed light and nitrate exhibit an interactive effect on the germination ofPaulownia tomentosa Steud. seeds that require long periods of light irradiation. Two pulses of red light (R), separated by an adequately long dark interval, substitute for continuous prolonged irradiation. A far-red (FR) pulse given at the beginning of the dark interval inhibits germination, while it has no effect if given at the end. The requirement for certain ratios of the far-red-absorbing form of phytochrome/total phytochrome (Pfr/Ptot) differs when a FR+R-pulse is given as the first or second of two pulses (FR+R or R) separated by a dark interval. An equal decrease of the Pfr/Ptot ratio leads to a more pronounced decrease in germination when the pulse of the same FR+R ratio is given as the second pulse at the end of the dark interval. The length of dark interval between light pulses needed for maximal germination, differed in (i) seeds with a natural requirement for long periods of light irradiation from that in (ii) seeds with their long light requirement imposed by two weeks of imbibition in darkness or by (iii) imbibition in 40% heavy water. However, a single R pulse was sufficient to induce a high percentage of germination if the seeds were supplied with KNO3 (10 mM) from the onset of imbibition up to the onset of light. This effect decreased with a delayed time of application, and was prevented if FR preceded the KNO3 application. Callus was successfully initiated on root, mesocotyl and leaf base segments of 3- to 4-day-old seedlings of ragi (Eleusine coracana Gaertn.). 2,4-D along with casein hydrolysate for Murashige and Skoog's basal medium was found to be most effective for callus initiation and maintenance. Mesocotyl and leaf base tissue derived calli gave shoot buds in medium in which the 2,4-D concentration was lowered.  相似文献

Water stress resulted in a specific response leading to a large and significant increase (80-fold) in free proline content of ragi (Eleusine coracana leaves and seedlings. L-Proline protected ornithine aminotransferase, an enzyme in the pathway for proline biosynthesis, isolated from normal and stressed ragi leaves against heat inactivation and denaturation by urea and guanidinium chloride. The protection of the stressed enzyme by L-proline was much more complete than that of the enzyme isolated from normal leaves. While L-ornithine, one of the substrates, protected the stressed enzyme against inactivation, it enhanced the rate of inactivation of the normal enzyme. α-Ketoglutarate protected both the normal and stressed enzyme against inactivation and denaturation. These results support the suggestion that ornithine aminotransferase has undergone a structural alteration during water stress. In view of the causal relationship between elevated temperature and water stress of plants under natural conditions, the protection afforded by proline against inactivation and denaturation of the enzyme from stressed leaves assumes significance. These results provide an explanation for a possible functional importance of proline accumulation during water stress.  相似文献

Eight cytokinins detected in germinated chick-pea (Cicer arietinum L. var. Castellana) seeds were first present in the embryonic axes but appeared in the cotyledons after 12h of germination. The cytokinins detected in the cotyledons originate in the embryonic axes, but no passage of these substances from the cotyledons to the axes was detected, except when the seeds were treated with red light.It is concluded that the role played by the embryonic axis in mobilizating the main reserves of the cotyledons is mainly effected through these cytokinins. Anin situ study of the P-uptake ability ofAcinetobacter calcoaceticus was carried out using the alginate immobilization technique. ImmobilizedA. calcoaceticus cells displayed a high P-uptake ability (>97% P-accumulating cells) when immersed in the aerobic zone of an activated sludge system for 30–240 min. The overall P-accumulation pattern of the anaerobic zone depicted a typical P-release mechanism. However, limited P-accumulation was also observed at this stage. Growth and anaerobiosis were not prerequisites for P-uptake. The immobilized cell retention time in the anaerobic zone did not affect remarkably the inherent P-uptake ability of immobilizedA. calcoaceticus when exposed to the aerobic stage. P-uptake and release were reversible and depended on the environmental conditions to which immobilized cells were exposed. Immobilization ofA. calcoaceticus using alginate can be regarded as a reliable method of studying pure cultures in the activated sludge process.  相似文献

Experiments were conducted in a Striga-sick field to study the effect of soil temperature, moisture and nitrogen on Striga parasitism on sorghum. Striga seeds contained in nylon bags and buried at 2 cm in the soil, were exposed to different temperature and moisture treatments. Clear polythene, hay mulch and bare soil treatments were used to vary soil temperature. These treatments gave mean maximum temperatures of 60°, 48° and 37°C, respectively at 2-cm soil depth. Irrigation levels of 0, 30 and 60 mm were applied to change soil moisture. Striga seed germination, viability and emergence were studied. After 34 days of preconditioning, the exhumed Striga seeds from polythene-covered plots (solarized plots) did not germinate or retain viability when these seeds were exposed to sorghum root exudate. However, seeds similarly buried under hay mulch or bare soil, with mean maximum soil temperatures of 48° and 37°C, respectively, had similar germination and viability percentages. Of these 75% germinated and 85% of them were viable, regardless of the temperature treatment. Although seeds stored at high temperature and humidity (solarization) were killed, more Striga plants emerged under the polythene treatment compared to hay mulch and bare soil treatments. The observed Striga plants in the polythene mulch treatment were, therefore, assumed to have come from deeper layers where solarization was not effective. Irrigation treatments did not have significant effects on Striga seed germination and viability, but a slightly higher number of plants emerged at 60-mm irrigation level than at 30-mm and 0-mm. Striga emergence, on the other hand, was directly related to the rate of N application. Nitrogen rates of 0, 25, 50 and 100 kg ha–1 resulted in the emergence of 11, 34, 38 and 40 Striga plants per plot, respectively. Despite the high infestation at high N levels, sorghum plants did not show a loss of vigor. Nitrogen application, therefore, does not reduce Striga incidence, but seems to neutralize the harmful effects of Striga without reducing the extent of parasitism.  相似文献

The germination of seeds of celery (Apium graveolens L.) becomes progressively thermoinhibited on incubation in the dark at high temperatures, the inhibitory temperature being dependent on the cultivar used. In two high-dormancy cultivars of celery, the production of germination inhibitors in seeds incubated in the dark at 26°C gradually increased over a 7-day period. Inhibitor production was measured by incubating seeds of the low-dormancy cultivar Florida 683 in homogenates of the thermoinhibited seeds of the high-dormancy cultivars and recording germination either in the light or with the gibberellins A4 and A7 (GA4/7) in the dark. Most Florida 683 seeds which failed to germinate in the homogenates after 15 days were induced to germinate by addition of N6-benzyladenine (BA). The presence of BA in addition to GA4/7 throughout incubation in the dark completely overcame the inhibitory effects of homogenates. This indicates that thermoinhibition of celery seeds is associated with the accumulation of a germination inhibitor which interacts with cytokinins. Colonization of sorghum by Macrophomina phaseolina in field plots was determined at nitrogen fertilization rates of 0, 56, 112, and 168 kg ha-1 in 1988 and 0, 84, 168, and 256 kg ha-1 in 1989. Above ground plant tissue and roots were sampled monthly to determine total nitrogen and percent colonization of root segments by natural inoculum. Root infection was not affected by nitrogen treatment, but was affected by growth stage and environment. High root infection occurred before reproductive development (growth stage 3) in 1988 and was associated with hot, dry weather early in the growing season. In 1989, when the weather was cool and wet, root infection began after reproductive development (growth stage 4). The effect of nitrogen treatments on lesion length was determined in sorghum stalks artificially inoculated with M. phaseolina. Lesion lengths were significantly affected by both nitrogen treatments and growth stage. Lesions were significantly longer with all nitrogen treatments at growth stage 9 than with the no-nitrogen treatment, and lesions tended to increase with increased levels of nitrogen fertilization. Significant increases in lesion length occurred between growth stages 5, 7, and 9 in 1988 and between 7 and 9 in 1989. This study demonstrates that nitrogen fertilization affects colonization of sorghum stalks but not root infection by M. phaseolina.  相似文献

Summary Two species of bacteria capable of growth onN-phosphonomethylglycine (glyphosate) were isolated from a bench scale sequencing batch reactor degrading a waste stream containing glyphosate. The enrichment and isolation medium contained defined salts and glyphosate as the sole carbon and energy source. Glyphosate was stoichiometrically degraded to aminomethylphosphonic acid (AMPA). A carboxylesterase (EC 3.1.1.1) was purified from germinated finger millet by ammonium sulphate fractionation, diethylaminoethyl-cellulose chromatography and Sephadex G-200 filtration. The homogeneity of the enzyme was established by Polyacrylamide gel electrophoresis, isoelectric focussing and sodium dodecyl sulphate-polyacrylamide gel electrophoresis. The enzyme has a single polypeptide chain with a molecular weight of 70,000. The amino acid analysis of the purified enzyme revealed that it contained a greater number of neutral and acidic, compared to, basic amino acid residues. The isoelectric pH of the enzyme was found to be 5·1. Studies with different organophosphate and carbamate inhibitors showed that this enzyme was more sensitive to organophosphate inhibitors than carbamates. The rate constantsk i andl 50 for different inhibitors were calculated. The product inhibition studies with this enzyme showed linear competitive inhibition with acetate and linear noncompetitive inhibition with 1-naphthol  相似文献
