Many fruits on Golden King plum trees inoculated through the stalks with Xanthomonas campestris pv. pruni developed unusual lesions extending from the exocarp to the endocarp. A few uninoculated, diseased fruits had similar lesions. The pathogen was isolated from both inoculated and uninoculated stalks and from seeds inside fruits. Scanning electron microscopy of inoculated stalks and mature fruits with unusual lesions revealed that vascular channels of the stalk, seed coat, stony endo, carp, and mesocarp were filled with masses of X. campestris pv. pruni. Bacterial colonies also occurred in other tissues of these fruit parts but were apparently absent from the starchy endosperm or surface of the diseased exocarp. This is the first full report of systemic movement of X. campestris pv. pruni to seed and fruit through stalks.  相似文献

As part of efforts to identify the causal agent of the rose rosette disease (RRD) of multiflora rose (Rosa multiflora Thunb.), root tip extracts from both symptomatic and nonsymptomatic roses were used to mechanically inoculate leaves of Nicotiana glutinosa. Pale green spots were observed along the margins of the major leaf veins only on leaves inoculated with extracts prepared from symptomatic rose plants. Light microscopy revealed abnormal development of the palisade and spongy mesophyll cells in the symptomatic tissue, although no virus‐like particles (VLPs) were observed by electron microscopy. However, VLPs were observed in cells from tissue adjacent to the leaf veins and bordered by the pale green spots. Inoculation of N. benthamiana with extracts from symptomatic N. glutinosa initially did not result in visible symptoms on N. benthamiana inoculated leaves. However, approximately 4 wk post inoculation, splitting of leaf tissue across and along major leaf veins in expanding leaves occurred. In later stages of leaf expansion some leaves split in regions not associated with veins. Light microscopy of thick sections revealed separation between palisade cells and groups of small dead cells in the mesophyll tissue of expanding systemically infected leaf blades. Electron microscopy revealed crystalline arrays in the cytoplasm of mesophyll cells. No abnormal cellular changes were observed in plants inoculated with extracts prepared from nonsymptomatic rose plants.  相似文献

Endophytic fungi of healthy twig tissues act as dominant primary colonizers of twig litters and are exposed to the attacks of saprobic secondary colonizers. In the present study, we examined the effect of interspecific interactions on the decay rate of twigs in laboratory experiments using two endophytic ascomycetes (Phomopsis sp. and Xylaria sp.) and two saprobic basidiomycetes (Mycena polygramma and Phanerochaete filamentosa) on twigs of Japanese beech (Fagus crenata) as a model system. Both endophytes were defensive against two saprobes on 2% malt agar, and were not replaced by the saprobes in the twigs during the incubation period. However, inoculation of the saprobes reduced the mass loss of twig components (acid-unhydrolyzable residue (AUR), holocellulose, soluble carbohydrate, and polyphenol). Especially when the twigs previously inoculated with Xylaria sp. or Phomopsis sp. were successively inoculated with M. polygramma the net weight of AUR increased, which was probably due to the synthesized melanin in hyphae of the endophytes. Our findings indicate that interspecific interactions between endophytic and saprobic fungi do affect the total mass of humic substances produced during the litter decay process.  相似文献

Age-related Resistance in Bell Pepper to Cucumber mosaic virus   总被引:2,自引:0,他引:2  
We demonstrated the occurrence of mature plant resistance in Capsicum annuum‘Early Calwonder’ to Cucumber mosaic virus (CMV) under greenhouse conditions. When Early Calwonder plants were sown at 10 day intervals and transplanted to 10‐cm square pots, three distinct plant sizes were identified that were designated small, medium and large. Trials conducted during each season showed that CMV accumulated in inoculated leaves of all plants of each size category. All small plants (with the exception of the winter trial) developed a systemic infection that included accumulation of CMV in uninoculated leaves and severe systemic symptoms. Medium plants had a range of responses that included no systemic infection to detection of CMV in uninoculated leaves with the systemically infected plants being either symptomless or expressing only mild symptoms. None of the large plants contained detectable amounts of CMV in uninoculated leaves or developed symptoms. When plants were challenged by inoculation of leaves positioned at different locations along the stem or different numbers of leaves were inoculated, large plants continued to accumulate CMV in inoculated leaves but no systemic infection was observed. When systemic infection of large plants did occur, e.g. when CMV‐infected pepper was used as a source of inoculum, virus accumulation in uninoculated leaves was relatively low and plants remained symptomless. A time‐course study of CMV accumulation in inoculated leaves revealed no difference between small and large plants. Analyses to examine movement of CMV into the petiole of inoculated leaves and throughout the stem showed a range in the extent of infection. While all large plants contained CMV in inoculated leaves, some had no detectable amounts of virus beyond the leaf blade, whereas others contained virus throughout the length of the stem but with limited accumulation relative to controls.  相似文献

Summary Uptake of water and magnesium chloride solution was investigated through the outer surface of twigs of Picea abies (L.) Karst. Water uptake was determined by using pressure/volume (P/V) curves of the twigs as a basis for calculation to avoid problems of superficial extraneous water. When water was sprayed on bark and needles of 3- to 7-year-old twigs at a xylem water potential of -1.00 MPa, they absorbed as much as 80 mm3 water in 200 min/g twig dry weight as the twig water potential recovered to -0.15 MPa. With fluorescent dyes, pathways for absorption of water and solutes through the twig bark were found, particularly through the radially orientated ray tissue. In addition to uptake by mass flow, magnesium could also diffuse along a concentration gradient from the twig surface into the xylem. In the field, the magnitude of these uptake processes would depend on the concentration of elements deposited by atmospheric precipitation, the concentration gradient between the plant surface and the xylem sap, the xylem water potential and the intensity and duration of each precipitation event.  相似文献

Shot hole disease is one of the most important diseases of stone fruit trees in Iran. The disease is wide spread among orchards of Prunus spp. During spring and summer of 2007, 80 monoconidial isolates of the pathogen were recovered from infected leaves, fruits and twigs of different Prunus spp. in West Azerbaijan, Tehran, Ghazvin and Razavi Khorasan provinces of Iran and were studied taxonomically. Based on morphological and physiological characteristics and growth optimal temperature, all isolates were identified as Wilsonomyces carpophilus. Seedlings of stone fruits (apricot, almond, peach, nectarine, plum, sweet cherry and sour cherry) were used for pathogenicity tests. All seedlings were susceptible to the fungal isolates and showed disease symptoms on twigs, leaves, buds and petioles. Genetic diversity of 28 selected fungal isolates was investigated based on DNA fingerprinting by random amplified polymorphic DNA-polymerase chain reaction (RAPD-PCR), using four random primers. Based on cluster analysis of the PCR results from the four primers, 10 fingerprinting groups (clonal lineages) and 27 haplotypes were identified. Clonal lineages “C”, “D” and “E”, each with six haplotypes formed the biggest clonal lineages, but other clonal lineages (“B”, “F”, “G”, “H”, “I” and “J”) included only one isolate. No correlation was detected among clonal lineages with the location of selected isolates and their host species. A correlation was found between the substrate (fruit, twig or leaf) and clonal lineages, particularly in “C” clonal lineage. The results showed that the fungus population had high genetic diversity which is distributed among the different areas of Iran.  相似文献

During 2009–2011, a dieback disease of mango (Mangifera indica) has recently emerged on mango trees in Panzhihua City, Sichuan province of China. The disease is characterized by large irregular brown‐coloured speckles on the petioles and twigs, vascular necrosis and dry leaves and complete twig mortality. Fusarium species were isolated repeatedly from the infected petioles and twigs. The species was identified as Fusarium decemcellulare Brick based on morphology and sequence analysis of Translation Elongation Factor‐1alpha (TEF‐1α) gene. Koch's postulates were fulfilled by pathogenicity tests on potted mango seedlings. To our knowledge, this is the first record of dieback on mango caused by Fdecemcellulare in China.  相似文献

During a recent survey of the inoculum source of twig dieback of tea caused by Macrophoma theicola, symptoms of canker were observed on branches of a number of tea cultivars. Most cankered areas contained minute black pycnidia of M. theicola, which ooze white conidial masses in strands when incubated under moist conditions. Macrophoma theicola was also consistently isolated from cankered branches. When healthy branches of tea were inoculated with conidia of M. theicola obtained from diseased branches, cankers similar to those occurring in nature developed in the inoculated areas, and M. theicola was re‐isolated from all experimentally induced cankers. Isolates derived from conidia of M. theicola also caused dieback on healthy tea twigs, indicating that fruiting bodies on the cankered branches are the main inoculum source of twig dieback of tea. There was a positive correlation between the severity of twig dieback and the prevalence of branch canker among the tea cultivars surveyed. Six of the cultivars surveyed were highly resistant to M. theicola.  相似文献

Cut twigs of Monterey pine(Pinus radiata D.Don) were banded with a 14C labelled morphactin formulation and the lower half of the twig placed in a Scholander type pressure bomb. Water was pumped through the twig from a reservoir inside the bomb and exudate collected from the protruding end of the twig. Using this method it was possible to determine that the slowest step in the translocation of morphactin was the passage across the outer layer of bark. Subsequent passage through the twig was not affected by girdling the bark suggesting that morphactin was translocated upwards via the xylem. The 14C compound in the exudate from live twigs differed from morphactin and was tentatively identified as the carboxylic acid derivative of the morphactin ester. The cut twig system facilitated studies on the uptake, metabolism and translocation of morphactin and should prove useful for studying the uptake and fate of other chemicals considered for bark application to trees.  相似文献

In recent years, leaf necrosis and twig dieback in the olive crop have been detected in Sicily (Italy). In this article, we identify the predominant fungal species associated with symptomatic leaves and twigs, using morphological features and DNA sequencing of the internal transcribed spacer (ITS) region, as Alternaria alternata, Arthrinium phaeospermum, Phoma cladoniicola and Ulocladium consortiale. The pathogenicity of these four species was tested on olive plants cv. Biancolilla. All species were pathogenic on leaves, but only U. consortiale produced cortical lesions on twigs, thus suggesting its main role in the Olea europaea twig dieback. To our knowledge, this is the first report of A. phaeospermum, P. cladoniicola and U. consortiale as olive pathogens.  相似文献

Cucumber mosaic virus (CMV) is known to systemically infect Arabidopsis thaliana ecotype Columbia plants. In order to identify the host factors involved in the multiplication of CMV, we isolated an A. thaliana mutant in which the accumulation of the coat protein (CP) of CMV in upper uninoculated leaves was delayed. Genetic analyses suggested that the phenotype of delayed accumulation of CMV CP in the mutant plants was caused by a single, nuclear and recessive mutation designated cum1-1, which was located on chromosome IV. The cum1-1 mutation did not affect the multiplication of tobacco mosaic virus, turnip crinkle virus or turnip yellow mosaic virus, which belong to different taxonomic groups from CMV. Accumulation of CMV CP in the inoculated leaves of cum1-1 plants was also delayed either when CMV virion or CMV virion RNA was inoculated. On the other hand, when cum1-1 and the wild-type Col-0 protoplasts were inoculated with CMV virion RNA by electroporation, the accumulations of CMV-related RNAs and the coat protein were similar. These results suggest that the cum1-1 mutation did not affect the uncoating of CMV virion and subsequent replication in an initially infected cell but affected the spreading of CMV within an infected leaf, possibly the cell-to-cell movement of CMV in a virus-specific manner.  相似文献

ABSTRACT Snow affects the nutritional ecology of northern ungulates during winter through burial of important winter forages. We used nonlinear regression analyses to model snow-burial dynamics of blueberry (Vaccinium spp.) browse biomass, a key winter food item of Sitka black-tailed deer (Odocoileus hemionus sitchensis) in southeastern Alaska, USA. During November 2003—March 2004 we collected data from 546 individually marked twigs located on 100 plants of differing sizes and architectures across a range of snow depths. In general, browse biomass became buried and unavailable to deer at snow depths substantially lower than prewinter twig heights. Plant architecture and plant height were related to the probability of a twig being buried. Probability of twig burial was higher on plants with lateral than on those with erect architectures. Twig height also affected the probability of burial by snow but the relationship was complex. For twigs located on erect plants, probability of burial was greatest for twigs near the bottom and top of the plant due to ground-up burial and bending of flexible apex stems, respectively. We used estimated nonlinear equations to model blueberry browse availability in a simulated upland old-growth habitat patch subject to a range of snow depths. We then compared subsequent estimates of deer winter nutritional carrying capacity for this habitat patch to findings derived using an alternative, simple linear (ground-up) model of winter-browse burial by snow. Comparisons indicated that ground-up models of browse burial overestimated browse availability and nutritional carrying capacity for most snow depths. Our findings demonstrate the importance of applying detailed snow-burial models when characterizing nutritional landscape of northern ungulates during winter.  相似文献

Leaves of the scab-susceptible apple (Malus domestica) cultivar Golden Delicious were harvested from May to August 2008 and 2009. Some leaves were healthy and some infected with fungus Venturia inaequalis. The phenolic compounds were analysed in healthy leaves, infected leaves and in the scab spot tissue. In comparison to healthy leaves, the infected leaves showed higher contents of hydroxycinnamic acid, flavanols and phloridzin, while lower contents on procyanidins, quercetins and phloretin. The total amount of phenolic compounds in the infected tissue was 10 to 20 % higher than in the healthy leaves. Accumulation of phenolic compounds is a post-infection response, and probably their further transformation is a prerequisite for plant resistance.  相似文献

观赏灌木小枝和叶性状在林下庇荫环境中的权衡关系   总被引:2,自引:0,他引:2  
小枝与叶片间的关系能够很好地反映植物对环境的适应策略。目前,从枝叶间权衡关系的角度研究树木对庇荫环境适应性的报道尚少,其中主要包括"生长-生长"权衡和"生长-生存"权衡两种假说。因此,为了探讨灌木在庇荫环境中,是否会在小枝和叶性状间采取权衡策略,来提高光照资源的利用能力或增强生存和防御能力,在城市绿地的林隙和林冠下两种光环境中,选择了没有人工修枝、整形等经营措施的金银木(Lonicera maackii)、小花溲疏(Deutzia parviflora)和连翘(Forsythia suspensa)等11种、79株观赏灌木作为研究对象,采集其当年生小枝,观测并计算了每棵灌木所处环境的有效光合辐射(PAR)相对累积光量和红光/远红光(R/FR),以及灌木的小枝干重(TDW)、单位小枝叶干重(LDW)、出叶强度(LN/TDW)、叶面积支持效率(LA/TDW)、比枝长(TL/TDW)和单叶面积(ILA) 6个性状。对数据进行标准化和数据转换后,运用Pearson相关检验分析光环境指标和灌木枝叶性状间的相关性;采用标准化主轴估计法进行异速生长方程的参数估计;使用多元回归分析不同光强下各性状的相互关系。结果表明:(1) PAR相对累积光量和R/FR与TDW和LDW呈极显著(P0.01或P0.001)正相关,与TL/TDW、LA/TDW、LN/TDW呈极显著(P0.001)负相关,与ILA相关性不显著(P 0.05)。整体上,R/FR与各灌木性状的相关性大于PAR相对累积光量;(2) TDW和LDW存在极显著(P0.001)的异速生长正相关关系。随着光强的减小,灌木当年生小枝和叶片的生物量都趋于减小,但是表现出相对偏向于叶片生物量的投资偏好;(3) LA/TDW和TL/TDW存在极显著(P0.001)的等速生长正相关关系。但随着光强的减小,比枝长随叶面积支持效率增加而增加的速率却减小,说明灌木在庇荫条件下,更倾向于采取忍耐型的光资源利用策略;(4) ILA和LN/TDW呈极显著(P0.001)异速生长负相关关系。随着光强的减小,灌木趋向于表现出单位小枝上着生大量小叶的现象。所以总体上,庇荫环境下的观赏灌木存在投资偏好和权衡,倾向于通过枝叶生长提高光截获能力来适应弱光环境,与"生长-生长"的权衡假说相符,但整体上,观赏灌木的耐阴性较差,故不建议种植在庇荫环境中。  相似文献

1. Gregarious behaviours in phytophagous insects are common, but their adaptive value is rarely well understood. In the present study, we document a novel case of cooperative behaviour, the aggregative oviposition of a leaf beetle, Pyrrhalta viburni Paykull, and the realised fitness benefit of overcoming a plant defensive response (wound tissue production). 2. In laboratory choice‐tests, females exhibited aggregative oviposition, characterised by (i) a preference for twigs already infested by conspecifics, and (ii) positioning of new egg masses adjacent to existing ones. Field observations supported laboratory results, showing that P. viburni egg masses are most commonly found aggregated in large clusters. 3. In a field experiment using three host plants (Viburnum dentatum L., Viburnum opulus L. and Viburnum×bodnantense Aberc. ex Stearn), mean egg survivorship and twig mortality increased, while twig wound response decreased, with an increasing level of infestation. Egg survivorship was consistently higher on twigs that died than on twigs that remained alive. 4. Overall, these results suggest that, by aggregating their egg masses, P. viburni females overwhelm the twig wound response, often killing the twig in the process. Aggregative oviposition and low defences of V. dentatum to P. viburni could have facilitated the establishment and spread of this beetle in its introduced range (northeastern North America), in areas where V. dentatum is abundant.  相似文献

The sensitivity of tissue nutrient concentrations to changes in plant age and the supply of P and N was compared between leaves and associated twigs in two forest species. In a young regrowth stand, tissues were sampled on three occasions from the mid-crown position of karri (Eucalyptus diversicolor F. Muell.) and Bossiaea laidlawiana Tovey and Morris, a major understorey legume. Leaves and twigs were also sampled from young plants of B. laidlawiana growing in a mature eucalypt stand to which P treatments had been applied.Nitrogen application increased N concentrations in twigs of karri and B. laidlawiana, but not in leaves. Phosphorus application increased P concentrations in both leaves and twigs of karri but the average increases were proportionally greater in twigs (65%) than in leaves (36%). Over the sampling period, P concentrations in leaves declined, while those in twigs were relatively stable. In B. laidlawiana, P supply also had a larger effect on P concentrations in twigs than in leaves. Addition of 200 kg P ha–1 increased average P concentrations in twigs by 109% in the regrowth stand and by 215% in the mature stand while the corresponding increases in leaves were only 11% and 27%. Concentrations of other nutrients in both species were also affected by N and P application, the most notable being a decline in the concentrations of the minor nutrients, Zn and Cu, with increased P supply.The increased N concentrations in twigs of karri, and the increased P concentrations in tissues of both species, were associated with responses of karri to added N and P, and of B. laidlawiana to added P. This indicates that tissue concentrations of N and P were generally below critical concentrations where N and P were not applied. The results show that for these species twigs may be a better tissue than leaves for diagnosing deficiencies or predicting N and P requirements. The ratio of P concentrations in twigs to P concentrations in leaves also increased with added P. It is suggested that this ratio may be a useful index if it reduces the variability caused by sampling position within the crown or genetic differences between plants.  相似文献

Enzyme-linked immunosorbent assay (ELISA) was used to test plum trees for prune dwarf (PDV), Prunus necrotic ringspot (NRSV) and apple mosaic (ApMV) viruses, cherry trees for PDV and NRSV, and apple trees for ApMV. Optimum conditions were determined for sampling in large-scale surveys for these viruses. All three viruses were detected throughout the growing season in individual samples of young leaves, or twigs with newly formed buds. However, when single infected leaves were combined with different numbers of healthy leaves, tests were most successful for all three viruses early in the growing season. PDV was detected in 1/40 (infected/total leaves) cherry leaves in April and May and 1/40 plum leaves until July, whereas NRSV was detected in 1/20 cherry leaves until July and 1/20 plum leaves until May. ApMV was detected in 1/20 apple or plum leaves until June but was detected less readily in mature leaves after June than either NRSV or PDV. There was no evidence of uneven distribution of virus infection in the trees. The viruses were detected in leaf samples kept for 8 wk at 3°C but freezing was less reliable for storage especially with ApMV. ApMV was detected in tests on plants held for several weeks at 25°C, and PDV and NRSV in plants held at 30°C.  相似文献

The photosynthetic behavior of leaves and twigs was compared in Hymenoclea salsola T. and G., a subshrub of the Mohave and Sonoran deserts, in which both leaves and green twigs make substantial contributions to whole-plant carbon gain. Light saturated photosynthesis in twigs was 0.62 times that of leaves (36.9 μmol m-2 s-1) when plants were well watered. Similar ratios were consistently observed in contrasting the photosynthetic responses of the two organ types to light, temperature, and intercellular CO2, regardless of whether rates were compared under saturating or highly limiting conditions of light or intercellular CO2. These scalar differences in photosynthetic rate between leaves and green twigs under a wide range of conditions were correlated with contrasting anatomical features such as chlorenchyma volume per projected area. Under normal ambient CO2 concentrations (350 μl 1-1), twigs on well watered plants operated at lower intercellular CO2 concentrations than the leaves. Possible causes of this difference are discussed with respect to performance under well-watered conditions, organ lifespans, and contrasting anatomical constraints. Twigs require larger investments than do leaves of both carbon and nitrogen per projected area of the respective organs, yet they realize lower photosynthetic rates per intercepted light. Twigs, however, fulfill additional roles besides photosynthesis such as structural support and vascular transport which does not allow them to be as anatomically specialized as leaves for photosynthesis. Twigs also have a longer expected lifespan than leaves with a larger fraction of them surviving the summer drought period. This was correlated with a greater tolerance of twig than leaf photosynthesis to low plant water potentials.  相似文献

Summary Root exudates of inoculated and uninoculated berseem (Trifolium alexandrinum) and lucerne (Medicago sativa) were examined chromatographically for their amino acid and reducing-sugar contents after 6 and 9 weeks growth in a sand culture experiment.Aspartic acid was the only amino acid detected in the root exudates of both the inoculated and uninoculated legumes, up to 9 weeks growth.Root exudates of inoculated berseem contained more amino acids than those of lucerne. Glutamic acid and fructose are characterised by their consistent presence in the root exudates of inoculated plants and their absence in those of uninoculated ones.The data presented in this paper is taken from the thesis submitted by the senior author to the P.G.
