The incidence of Phoma exigua var. foveata and Polyscytalum pustulans in dry soil and dust from potato stores was assessed at 10 farms in Scotland producing virus-tested stocks of seed potatoes derived from stem cuttings (VTSC). Samples were collected on three occasions during 1976: during the storage period (February-March), soon after the potatoes were planted (May) and just before the new crop was lifted and stored (August-October). Both pathogens were detected at all three sampling times but P. exigua var. foveata was more frequently detected at the last one than P. pustulans. Soil and dust from various sites within the stores, such as floors, ledges, graders and boxes, were contaminated by these pathogens. Propagules of both pathogens remained airborne for at least 12–17 min after the floors were swept. P. exigua var. foveata remained viable for at least 16 months in dry soil from tuber surfaces stored at 4–6°C. When VTSC tubers were dusted before planting with a dry store soil contaminated by P. exigua var. foveata the gangrene potential of the progeny tubers was much greater than that from untreated tubers or from tubers treated with an uncontaminated soil. The role of inoculum surviving in dry soil is discussed in relation to reinfection of VTSC stocks.  相似文献

In 1 out of 2 years' field trials benomyl applied as a dust treatment at time of planting seed potatoes resulted in an increase in potato gangrene in the progeny of both gangrene-free tubers planted in land contaminated with Phoma exigua var. foveata and gangrene-diseased tubers planted in clean land. Gangrene-infected seed tubers treated with benomyl also produced more stems infected with var. foveata than untreated tubers. Two hypotheses are presented to account for this increase in gangrene which does not occur in seed potatoes treated shortly after lifting. In all cultivars tested an organo-mercury dip-treatment increased total numbers of tubers in the seed and chat-size grades without increasing total weight whereas benomyl dust increased the numbers in these grades in Majestic only. The treatment of gangrene-diseased seed with benomyl dust affected neither total weight nor total number of tubers.  相似文献

以辣根过氧化物酶标记马铃薯卷叶病毒抗体,采用双抗体夹心ELISA方法鉴定了马铃薯和洋酸浆的茎、叶、根及马铃薯块茎中的马铃薯卷叶病毒(Potato Leafroll Virus,PLRV),结果表明,对提纯的PLRV可测出的最低浓度为25ng/ml,当包被抗体浓度为40μg/ml、酶标记抗体稀释度为1/120时,可测出马铃薯茎、叶和根汁液中的PLRV,感染PLRV的洋酸浆茎、叶和根汁液的消光值,均比无病对照者高二倍以上,虽然感染PLRV的马铃薯休眠块茎维管束组织汁液的消光值高于无病毒对照,且脐部维管束组织消光值高于顶端,但测定打破休眠的感病块茎顶端维管束组织的阳性结果更为可靠和明显。  相似文献

Common scab, caused by Streptomyces scabiei is an economically important potato disease worldwide. The potato industry in New Brunswick, Canada experience $1.2 million loss every year due to this disease. Superficial, raised, or deep-pitted brownish lesions on infected tubers reduce the quality and marketability of both fresh-market and processing potatoes, and hence, common scab is considered a priority disease for which adequate control measures are lacking. The objective of this research was to compare various potential treatments in suppressing the disease. Two field experiments were conducted at McCain's Research Farm, Florenceville-Bristol, New Brunswick, Canada, in 2008 and 2009 to assess the efficacy of Bacillus subtilis, Enterobacter cloacae, Chloropicrin, Pic-Plus, manganese sulphate and mustard meal in comparison with the chemical controls fludioxonil and mancozeb against common scab of potato. The disease incidence was significantly reduced by 36% due to the addition of mustard meal to the soil; 35.4% due to fludioxonil seed treatment; 30.0% due to soil fumigation with Pic-Plus; or by 27.2% due to soil fumigation with Chloropicrin. Potato tubers with scab severity ≥5% which are considered unmarketable in Canada were significantly reduced by 56.1% due to seed treatments with B. subtilis; by 57.8% due to fludioxonil; or by 63.1% due to the soil addition of mustard meal. The same treatments significantly increased marketable yield by 32.5%, 24.6%, and 24.6%, respectively. Soil fumigation with Chloropicrin or Pic-Plus increased marketable yield by 9.5% or 7.1%, respectively. These findings indicate that, in addition to the fludioxonil seed treatment, the seed treatment with the biopesticide containing B. subtilis and the soil addition of mustard meal treatments are potential alternatives for managing common scab of potatoes.  相似文献

The goal of the research is to determine the effect of pretreated poultry manure and irrigation on the yield and quality of potatoes (Solanum tuberosum) grown in fields located on the southern steppe of the Republic of Bashkortostan. Field experiments were repeated every three years. During vegetation, potato leaves and tubers were studied at the full blossom, leaves’ decay, and potato harvest. The results showed that applying pretreated poultry manure at the rate of 120 t/ha favored higher tuber weight both on the rainfed and irrigated plots (from 0.23 to 0.82 kg/plant and from 0.24 to 1.02 kg/plant, respectively). On the rainfed and irrigated plots where poultry mature was not applied (control fields) the tubers’ weight ranged from 0.08 to 0.31 kg/plant and from 0.16 to 0.50 kg/plant, respectively. Upon application of 40 t/ha of manure, under irrigation, the highest marketable value of tubers was 78%. On the rainfed plots the same value was 72% when applying poultry manure at a dose of 120 t/ha. Soil moisture monitoring showed that the potatoes did not get adequate water during the growing season. When the soil moisture on irrigated plots was 70% less than its minimum water capacity, potato plantings were watered.  相似文献

In 1982 – 88, potatoes were grown in 2-, 4- and 6-course rotations with spring barley on a field infested with Globodera rostochiensis. Severity of stem canker and black scurf increased with increasing frequency of previous potato crops, and seed tuber treatment with tolclofos-methyl became less effective in controlling diseases. This suggested that previous crops had increased the amounts of soil-borne inoculum of Rhizoctonia solani. Oxamyl soil treatment increased stem canker in one year and decreased black scurf in four years. Seed tuber treatment with imazalil or prochloraz decreased stem base infection by Polyscytalum pustulans and skin spot and silver scurf on tubers. Black dot was prevalent on tubers in all years and was not affected by seed tuber treatment or previous cropping. Oxamyl increased black dot and common scab in five years and decreased % tuber dry matter in six years. Cysts of G. rostochiensis were found attached to Désirée but not to Maris Piper tubers in August. At harvest tubers of both cultivars were affected by superficial pitting and its severity was related to soil populations of G. rostochiensis at planting. This damage was controlled by oxamyl. It is suggested that the pitting developed from holes made in the tuber skin at larval invasion. In 1989, Désirée seed tubers and healthy mini tubers were planted in all plots and severity of stem canker and black scurf increased with increasing proximity of previous potato crops and with the number of previous crops. Black dot on stems and tubers was not affected by previous cropping but was much less severe in a plot that had not grown potatoes during the seven years of the experiment. The severity of common scab generally decreased as the number of preceding potato crops increased.  相似文献

Western Australia has a unique seed potato production scheme which has remained virtually unchanged for more than 60 years, consisting of summer plantings of predominantly one cultivar in wind-exposed coastal swamplands. No rotation is used and the scheme relies on natural winter flooding and 'grazing' by sheep to eliminate unharvested tubers. Stocks are recycled every year with only limited inputs of pathogen-tested seed tubers in recent times. Virus spread in the crop is controlled by selecting large tubers for planting, roguing, aphicide application and growing season inspections. Potato viruses X and S were commonly detected in old seed stocks produced by this scheme attaining 100% infection in some. Both viruses were less frequently found in newly introduced seed stocks. By contrast, potato virus Y was never detected and potato leaf roll virus rarely found.  相似文献

Surveys were made for the presence of potato virus Y (PVY) in the planted seed and harvested tubers in ware potato crops of cv. Record grown at three sites in England in 1994 (survey 1) and seven sites in 1995 (survey 2). PVY was not found in samples of planted seed, but high levels of infection were found in many, but not all, harvested crops. However, plants of volunteer potatoes (VP) (i.e. plants arising from tubers or true seed derived from previous crops and surviving in the soil) were frequently found to be infected. Infection in tubers harvested from crops in the first survey ranged from 2–52%. In 1995, VP were collected from two of the three English sites where potato crops had been grown the previous season and also from a site in Scotland where PVY infection in an experimental crop of cv. Record had been monitored in 1994. The percentages of infected VP ranged from 2–54%. PVYN was the predominant strain found in sampled VP, with only two plants (out of 300 infected) containing PVYO. In the second survey, VP were assessed within the 1995 ware crops and were found at four sites, at which they comprised between 4–8% of emerged potato plants. Between 31–93% of VP were infected. Again, PVYN was the predominant strain with one plant containing PVYO and another PVYC (out of 189 infected). A sample of harvested tubers from each site was also tested for PVY. At those sites which had many infected VP, the harvested crop contained a large percentage of infected tubers, ranging from 60–97%. Two sites which had not previously been used for cropping potatoes had no VP and a very low incidence of PVY infection in the harvested tubers (1% and 2%). However, although no VP were found at one site, 31% of harvested tubers were infected, suggesting that alternative inoculum sources may be important.  相似文献

Inoculation with soil samples proved that the fungus causing dry rot is frequently present in field soils in Cheshire and in soil adhering to imported seed tubers. The fungus was viable in soils having a wide range of p H values and in fields which had not grown potatoes for 5-6 years. Other sources of infection include lofts, used sacks, seed boxes, diseased tubers, and knives used for cutting seed potatoes.
Bruised tubers stored in heavily contaminated boxes developed much dry rot; far less disease occurred in unbruised tubers. In boxes containing own-saved seed, healthy tubers in contact with diseased ones remained sound. Bruised tubers in contact with, or contaminated by, diseased tubers contracted dry rot. Cutting seed with a contaminated knife increased the disease seven-fold.
Inoculation of tubers attached to the parent plant showed that little or no infection occurred before lifting. In field trials severe dry rot developed in several varieties 6-8 weeks after lifting.

Attempts were made to extend the storage life of seed potatoes by treating them with trace elements as antimicrobial and antisprouting agents. In tests to measure the growth of Fusarium oxysporum, and Erwinia carotovora pv. carotovora it was found to be decreased more by iron, copper and zinc than by boron, manganese and molybdenum. Dipping seed tubers in aqueous solutions of trace element salts decreased both sprouting and microbial spoilage during storage at ambient conditions. Field testing at the end of storage proved that treating seed potatoes with ferrous sulphate did not decrease the yield of tubers or increase their iron content. It is suggested that trace elements may be used to extend the storage life of seed potato tubers, in tropical conditions.  相似文献

Four treatments applied on potato seed tubers i.e. talc only, chitosan only, Streptomyces melanosporofaciens strain EF-76 in talc and S. melanosporofaciens strain EF-76 in chitosan were compared for their efficiency to reduce common scab incidence. As previously reported the joint application of the geldanamycin-producing S. melanosporofaciens strain EF-76 and chitosan was effective to control common scab of potato. Nevertheless, field application of S. melanosporofaciens EF-76 did not allow the selection of geldanamycin-resistant actinomycetes (GRA) in the bulk soil during the potato growing season. The number of GRA on harvested potato tubers was, however, significantly higher in treatments that contained chitosan than in other treatments suggesting that chitosan might promote the establishment of the antagonistic actinomycete on progeny tubers. Biolog EcoPlates were used to determine the metabolic profiles of the bacterial soil communities. A permutation MANOVA analysis detected significant differences within the metabolic profiles of the bacterial communities at the potato flowering period but not at the beginning of the season or a week before harvest. A combination of S. melanosporofaciens EF-76 and chitosan thus represents a promising tool against common scab, with low short-term impact on soil bacterial communities.  相似文献

In laboratory and field experiments from 1952 to 1955, 2:3:5:6-tetrachloro-I-nitrobenzene (tecnazene) delayed the development of sprouts on treated potato tubers, and 2:3:4:6-TCNB retarded sprout development in two laboratory experiments. At the usual commercial rates of application the 2:3:4:5 isomer had no effect on sprout development and very little effect at higher rates. The crop from 2:3:5:6-TCNB-treated seed tubers usually contained a higher proportion of small tubers than that from untreated or 2:3:4:5-TCNB-treated seed. Marketable yield from 2:3:5:6-TCNB-treated seed was sometimes below that from untreated seed, but when this happened the yield from 2:3:4:5-TCNB-treated seed was higher.
Both isomers produced good control of dry rot, Fusarium caeruleum (Lib.) Sacc., on five varieties of potatoes, but did not produce a commercial control on Doon Star. To achieve control, the isomers had to be present at the time of infection. There was some evidence that the development of disease symptoms was delayed in treated, infected tubers. Since 2:3:4:5-tetrachloro-1-nitrobenzene has no sprout depressant action it merits further trial as a fungicidal treatment for seed potatoes.  相似文献

Field and glasshouse experiments on the control of potato mop-top virus   总被引:2,自引:0,他引:2  
Field observations during 3 yr on a stock of potato cv. Red Craigs Royal partially infected with potato mop-top virus (PMTV) confirmed that the virus was passed by an infected mother plant to only a proportion of its progeny tubers, and showed that in this cultivar symptomless plants gave rise only to symptomless progeny. The elimination of PMTV from stocks can therefore be greatly accelerated by removing symptom-bearing plants. Infected potato tubers were not freed from PMTV by treating them at 37 °C for up to 8 wk. Treating ‘seed’ tubers bearing powdery scabs that contain PMTV-carrying resting spores of Spongospora subterranea with formaldehyde or organo-mercurial fungicide greatly decreased PMTV establishment when the tubers were planted in previously uninfective soil, but fumigation with 2-aminobutane was ineffective. Decreasing the pH of infective soil to 5-0 by applying sulphur greatly decreased the infection of potato cv. Arran Pilot with PMTV and S. subterranea in field experiments, but this treatment did not eliminate either; when the pH of treated soil was raised the transmission of PMTV resumed. Treating infective soil with a range of fungicides greatly decreased the infection of Nicotiana debneyi bait seedlings in glasshouse experiments but only calomel at 75 kg/ha controlled spread of PMTV and 5. subterranea to potato in field experiments. In other field experiments, applying zinc frit, zinc sulphate or zinc oxide to infective soil greatly decreased the spread of both to potato. The amount of zinc required increased with increase in clay content of the soil. However, treatment with zinc compounds did not eliminate PMTV-carrying vectors from soil, and when treated soil was diluted with autoclaved soil many of the bait seedlings planted in the mixture became infected. The zinc frit was phytotoxic because of its boron content but zinc sulphate and zinc oxide caused little or no decrease in tuber yield. The zinc content of potato tubers was increased but not doubled in zinc-treated plots, and during the first year after treatment the zinc content of topsoil decreased greatly. The zinc content of ryegrass grown after potatoes was greater than of potato tubers but did not reach a level considered dangerous to livestock. Treatment of soil with sulphur, zinc oxide or calomel may be useful for small plots used in the early stages of propagation of virus-tested potato clones where there is risk of infection with PMTV.  相似文献

Applying 5 ml of a 38% aqueous solution of formaldehyde to 1500 ml of sandy loam prevented potato cyst-nematode, Heterodera rostochiensis, from increasing on the roots of Arran Banner potatoes planted in the soil and increased the weight of tubers produced. A comparable dosage applied to plots of sandy loam and sandy clay in winter did not prevent the nematode from multiplying on Pentland Crown potatoes but increased tuber yields. Smaller amounts of formalin had less effect on tuber yields and drenching dilute formalin solutions onto the soil was no more effective than incorporating undiluted formalin into the soil by rotavation 15 cm deep.  相似文献

The incidence of potato virus Y (PVY) infection was assessed in samples of potato tubers, cv. Record, taken from Scottish seed stocks and English ware crops grown from some of these seed stocks. PVY was readily detected by ELISA of tuber sprouts. PVY-infected tubers were found in 10 seed stocks of 84 tested. The mean level of virus infection was 0.23%, 0.76% and 0.56% in Super Elite, Elite and AA stocks respectively. In 46 commercial ware crops grown from some of these seed stocks, a substantial proportion of the harvested tubers in all but one of the crops were infected with PVY, the mean percentage of infected tubers was 58.5%. Ware crops grown from seven seed stocks in which PVY had been detected (mean 6.2% infection in seed) contained a mean of 70% infected tubers, compared with 56% infection in crops grown from 39 stocks in which PVY was not detected in the seed tubers. The predominant PVY strain detected in the ware crops was the veinal necrosis strain (PVYvn).  相似文献

The main source of inoculum of Helminthosporium solani was the seed tuber; the fungus was not detected in soils in which infected potatoes had been grown the previous year. Lesions spread rapidly on seed tubers after planting, so that within a few weeks the entire surface was covered. As lesions aged they lost the capacity to sporulate, so most inoculum was produced in the first few weeks after planting. Infection of progeny tubers was first detected at the heel (stolon attachment) end soon after their initiation. Once established, lesions spread slowly on the surface of progeny tubers when in the soil; more rapid spread took place during storage. Potatoes which seemed unaffected at lifting became diseased during storage.  相似文献

Rhizoctonia solani, the causal agent of stem canker and black scurf on potato, survives as sclerotia on tubers, in soil and in plant residues. The objective of the present study was to evaluate the importance of inoculum source on disease development. Disease‐free minitubers and seed tubers contaminated with low levels of R. solani were planted in fumigated or artificially inoculated growth mixture in greenhouse experiments. Black scurf incidence and severity were significantly higher when the inoculum was present in both seed tubers and soil, compared with either of them separately. The severity of disease symptoms on the subterranean parts of the plant also were significantly higher in plots where both seed tubers and soil were contaminated, compared with plots where the inoculum source was either the seed tubers or the soil. Thus, both major sources of inoculum, seed tubers and soil, are important in disease development. However, when both sources are present, black scurf incidence and severity are increased, leading to economical damage to tuber yield and quality. Additional results from field trials support these findings. Disease incidence and severity on daughter tubers were correlated with levels of contamination in seed tubers and soil. When seed tubers and soil were heavily infested, the levels of black scurf incidence and severity on daughter tubers were very high; when seed tuber and soil infestation were very low, black scurf incidence and severity on progeny were also lower. Disease levels were reduced by in‐furrow fungicide treatment, but were less effective when the initial levels of the fungus on the seed tubers and in the soil were high.  相似文献

Benomyl and thiabendazole, applied as dusts to seed potatoes before chitting (sprouting), reduced the incidence of silver scurf disease on the progeny at lifting and during subsequent storage. Treatment of seed tubers 4 months prior to planting almost completely suppressed sporulation during storage, even under conditions very favourable for the growth of the fungus. The fungicides appeared not to act systemically because developing tubers grown from treated seed were just as susceptible to infection after inoculation as those from untreated. Control was due to effective inhibition of sporulation on the surface of diseased seed tubers after planting. There was no reduction in the amount of disease in subsequent generations of potato crops without further fungicidal treatment. Seed virtually free from silver scurf, produced from seed treated with fungicide in the previous year, yielded progeny with as much disease as progeny from untreated seed stocks.  相似文献

Transgenic potato plants expressing the gene of hepatitis B surface antigen (HBsAg) under the control of the double promoter of 35S RNA of cauliflower mosaic virus (CaMV 35SS) and the promoter of patatin gene of potato tubers have been obtained. Biochemical analysis of the plants was performed. The amount of HBsAg in leaves, microtubers, and tubers of transgenic potatoes growing in vitro and in vivo was 0.005-0.035% of the total soluble protein. HBsAg content reached 1 microg/g in potato tubers and was maximal in plants expressing the HBsAg gene under the control of CaMV 35SS promoter. In transgenic plants expressing HBsAg gene under the control of tuber-specific patatin promoter, HBsAg was found only in microtubers and tubers and was absent in leaves. Western blot analysis of HBsAg eluted from immunoaffinity protein A-Sepharose matrix has been performed. The molecular weight of HBsAg peptide was approximately 24 kD, which is in agreement with the size of the major protein of the envelope of hepatitis B virus. Using gel filtration, it was determined that the product of HBsAg gene expression in potato plants is converted into high-molecular-weight multimeric particles. Therefore, as well as in recombinant HBsAg-yeast cells, assembling of HBsAg monomers into immunogenic aggregates takes place in HBsAg-transgenic potato, which can be used as a source of recombinant vaccine against hepatitis B virus.  相似文献

采用多管发酵法检测二级水灌溉对马铃薯种植土壤中大肠菌群数量,结果表明:充分灌溉后的土壤中大肠菌群的数量均高于分根交替灌的土壤;地下滴灌的土壤中大肠菌群数量高于沟灌土壤的大肠菌群数量;二级水经加氯消毒后,土壤中大肠菌群数量明显下降。同样采用多管发酵法检测马铃薯块茎,结果发现:采用加氯消毒后的二级水灌溉的小区E和F中生长的马铃薯,其块茎组织中大肠菌群数量低于其他小区,采用地下滴灌灌溉的小区I和J所种植出的马铃薯块茎中大肠菌群的数量高于采用沟灌灌溉的小区K和L。结果表明:马铃薯块茎中大肠菌群的数量与其生境中大肠菌群的数量有密切关系。  相似文献
