The heavy metal hyperaccumulator Thlaspi caerulescens occurs both on heavy metal polluted soils (metallicolous ecotype MET) and on soils with normal heavy metal content (non-metallicolous ecotype: NMET). In order to assess the extent and structure of variation in growth, shoot accumulation of Cd, Zn and mineral element (Ca, Mg, K, Fe), a MET ecotype from Belgium and a NMET ecotype from Luxembourg were studied. Seven maternal families from two populations of each ecotype were grown on both Cd and Zn contaminated soil. Although both ecotypes presented a similar heavy metal tolerance in the experimental conditions tested, they differed in several points. The MET populations had markedly higher biomass and higher root:shoot ratio compared to NMET populations. The Zn, and at lesser extent, the Cd hyperaccumulation capacity tended to be higher in the NMET populations. The same trend was observed for the foliar concentrations of Mg, Ca and Fe with NMET populations having higher concentrations compared to MET ones. Cd and Zn concentrations were negatively correlated with the biomass of both ecotype. However, the negative correlation between the Zn and biomass was much lower in MET ecotype suggesting a tighter control of internal Zn concentration in this ecotype. Finally, although the Cd phytoextraction capacity was similar in both ecotype, a higher Zn phytoextraction capacity was detected in NMET ecotype when these plants grow on moderate Cd and Zn concentrations.  相似文献

Thlaspi caerulescens J. & C. Presl is a distinctive metallophyte of central and western Europe that almost invariably hyperaccumulates Zn to> 1.0% of shoot dry biomass in its natural habitats, and can hyperaccumulate Ni to> 0.1% when growing on serpentine soils. Populations from the Ganges region of southern France also have a remarkable ability to accumulate Cd in their shoots to concentrations well in excess of 0.01% without apparent toxicity symptoms. Because hyperaccumulation of Cd appears to be highly variable in this species, the relationship between Cd tolerance and metal accumulation was investigated for seven contrasting populations of T. caerulescens grown under controlled conditions in solution culture. The populations varied considerably in average plant biomass (3.1‐fold), shoot : root ratio (2.2‐fold), Cd hyperaccumulation (3.5‐fold), shoot : root Cd‐concentration ratio (3.1‐fold), and shoot Cd : Zn ratio (2.6‐fold), but the degree of hyperaccumulation of Cd and Zn were strongly correlated. Two populations from the Ganges region were distinct in exhibiting high degrees of both Cd tolerance and hyperaccumulation (one requiring 3 µM Cd for optimal growth), whereas across the other five populations there was an inverse relationship between Cd tolerance and hyperaccumulation, as has been noted previously for Zn. Metal hyperaccumulation was negatively correlated with shoot : root ratio, which could account quantitatively for the differences between populations in shoot Zn (but not Cd) concentrations. On exposure to 30 µM Cd, the two Ganges populations showed marked reductions in shoot Zn and Fe concentrations, although Cd accumulation was not inhibited by elevated Zn; in the other five populations, 30 µM Cd had little or no effect on Zn or Fe accumulation but markedly reduced shoot Ca concentration. These results support a proposal that Cd is taken up predominantly via a high‐affinity uptake system for Fe in the Ganges populations, but via a lower‐affinity pathway for Ca in other populations. Total shoot Cd accumulated per plant was much more closely related to population Cd tolerance than Cd hyperaccumulation, indicating that metal tolerance may be the more important selection criterion in developing lines with greatest phytoremediation potential.  相似文献

通过花药培养筛选出三个水稻耐镉突变体并都获得再生植株。再生植株及由根尖诱导产生的愈伤组织都保持稳定的耐镉性。突变植株的花药经再培养鉴定也具有耐镉性,从而表明所获突变体的耐镉特性是可以遗传的。在对愈伤组织耐镉机理进行的分析中发现,突变体愈伤组织中的胱氨酸和半胱氨酸的含量高于对照。将一定量的胱氨酸加到含镉的培养基中用于检查野生型愈伤组织的增殖情况,观察到胱氨酸可以降低镉对细胞的危害,表明突变体耐镉机理可能与细胞中积累有较多的胱氨酸和半胱氨酸有关。  相似文献

The Salicaceae family comprises a large number of high‐biomass species with remarkable genetic variability and adaptation to ecological niches. Salix caprea survives in heavy metal contaminated areas, translocates and accumulates Zn/Cd in leaves. To reveal potential selective effects of long‐term heavy metal contaminations on the genetic structure and Zn/Cd accumulation capacity, 170 S. caprea isolates of four metal‐contaminated and three non‐contaminated middle European sites were analysed with microsatellite markers using Wright's F statistics. The differentiation of populations North of the Alps are more pronounced compared to the Southern ones. By grouping the isolates based on their contamination status, a weak but significant differentiation was calculated between Northern metallicolous and non‐metallicolous populations. To quantify if the contamination and genetic status of the populations correlate with Zn/Cd tolerance and the accumulation capacity, the S. caprea isolates were exposed to elevated Cd/Zn concentrations in perlite‐based cultures. Consistent with the genetic data nested anova analyses for the physiological traits find a significant difference in the Cd accumulation capacity between the Northern and Southern populations. Our data suggest that natural populations are a profitable source to uncover genetic mechanisms of heavy metal accumulation and biomass production, traits that are essential for improving phytoextraction strategies.  相似文献

为探讨铅锌矿废弃地优势植物在重金属污染土壤植物修复中的应用潜力,利用野外采样分析法,从粤东梅县丙村铅锌尾矿区采集其三种优势植物类芦、黄荆、盐肤木的根、茎、叶和土壤样品,测定和分析Pb、Zn、Cu、Cd四种重金属含量.结果表明:该矿区土壤污染严重,Pb、Zn、Cd含量远超土壤环境质量的三级标准,Cu超出二级标准;根际土壤和非根际土壤重金属含量均为Pb>Zn>Cu>Cd,但根际土壤的重金属含量显著低于非根际土壤;这三种植物对Pb、Zn、Cu的转移系数大于1.0,对Cu的富集系数最高,Pb最小,但对四种重金属的富集系数均小于1.0,均未达到超富集植物临界含量标准.三种植物为该矿区的优势植物,说明它们对土壤的重金属污染有很强的耐性,虽然并非典型的超富集植物,但对污染土壤仍有较好的修复效果.  相似文献

Restoration of metalliferous mine soils requires using plant species tolerant to high metal concentrations and adapted to nutrient‐poor soil. Legumes can increase plant productivity through N2‐fixation, but they are often scarce in metalliferous sites. We examined survival, growth, and tolerance of four populations of a legume, Anthyllis vulneraria, from two metalliferous (MET) Zn‐Pb mine sites, Avinières (AV) ([Zn‐EDTA] = 26,000 mg/kg) and Eylie (EY) ([Zn‐EDTA] = 4,632 mg/kg), and two non‐metalliferous (NMET) sites located in the south of France with the aim to select the most appropriate populations for restoration of mined soils. In a common garden experiment, plants from each population were reciprocally grown in soil from the provenance of each population. The two NMET populations exhibited high mortality and low growth rates in soil from the mined sites. The AV MET exhibited a high growth rate in metalliferous soils, but showed high mortality in non‐metalliferous soils. The growth of the EY MET was very low in the AV‐contaminated soil, but was the highest of all populations in moderately and non‐metalliferous soils. Plants from the AV MET population showed a high growth and survival in metalliferous soil and would be appropriate in the restoration of metal‐contaminated sites (>30,000 mg Zn kg?1). The EY MET population would be adapted to the restoration of moderate metal‐contaminated soils (<30,000 mg Zn kg?1). Taking into account the broad distribution of A. vulneraria, these two populations could be suitable for the restoration of derelict mine sites in mediterranean and temperate regions of Europe and North America.  相似文献

Callus of Trema orientalis derived from both contaminated and uncontaminated sources were tested in vitro for their relative tolerance to chromium and nickel. The calluses derived from contaminated source were metal-tolerant and showed better growth than those obtained from uncontaminated plants. The specificity of metal tolerance shown by the parent material was maintained in the calluses. Compared to the uncontaminated explants, the calli derived from contaminated sources exhibited higher catalase and peroxidase activities but a reduced acid phosphatase activity. Biochemical studies, provided evidence that the contaminated sources were physiologically distinct from the uncontaminated ones. Thus, this study indicated that seeds of Trema orientalis collected from contaminated sites were tolerant to chromium and nickel, and may have the advantage of being used in sustainable revegetation programmes on chromiferous minewastes.  相似文献

Representatives of Brassicaceae species—the hyperaccumulator Noccaea caerulescens F.K. Mey and the metal excluder Thlaspi arvense L.—were compared in terms of their ability to accumulate nickel (Ni) and zinc (Zn) and their tolerance to these metals. Four ecotypes of N. caerulescens were used: the ecotypes La Calamine (LC, Belgium) and Saint Felix de Palliéres (SF, France) grow naturally on calamine soils rich in Zn, Cd, and Pb; the ecotype Monte Prinzera (MP, Italy) originates from serpentine soils rich in Ni, Co, and Cr; and the ecotype Lellingen (LE, Luxembourg) inhabits non-metalliferous soils. The plants of N. caerulescens were grown for 8 weeks in a half-strength Hoagland solution supplemented with 25, 100, 200, 300, and 400 μM Ni(NO3)2 (ecotypes LC, SF, MP, LE) or 100, 200, 400, 800, and 1000 μM Zn(NO3)2 (ecotypes LC, SF, LE); the plants of T. arvense were grown in the presence of 10, 20, 25, and 30 μM Ni(NO3)2 or 40, 50, 60, 70, 80 μM Zn(NO3)2. The toxic effect of Ni and Zn was assessed from changes in dry matter of roots and shoots of treated plants compared to untreated. The content of metals in roots and shoots was determined by means of atomic absorption spectrophotometry. The Ni-accumulating capacity of N. caerulescens ecotypes increased in the order: LC < SF < LE < MP, and the Zn-accumulating capacity increased in the row: LC < SF < LE. In the hyperaccumulating plant N. caerulescens, the increments of biomass started to decrease at a lower metal content in roots than in shoots, whereas the opposite pattern was observed in the metal excluder T. arvense. Since T. arvense plants accumulated Ni and Zn in roots, whereas N. caerulescens accumulated these metals in shoots, one may assume that the greater sensitivity of root growth compared with shoots in N. caerulescens was determined by more effective mechanisms of metal detoxification in shoots. Conversely, the higher sensitivity of shoot growth compared to root growth in T. arvense was determined by more effective mechanisms of metal detoxification in roots. Being more tolerant to Ni and Zn than T. arvense plants, the N. caerulescens ecotypes differed substantially in terms of metal-accumulating capacity and their tolerance to heavy metals. The ecotype originating from non-metalliferous soils (LE) accumulated larger amounts of Zn, but was less tolerant compared with ecotypes growing naturally on calamine soils (SF and LC), whereas the ecotype occurring on serpentine soils (MP) exhibited a markedly greater tolerance to Ni, compared with other ecotypes examined, as well as the largest accumulation of this metal. The results indicate the existence of different mechanisms responsible for plant tolerance to Ni and Zn; the study of these mechanisms is a promising direction for future research.  相似文献

The capacity to accumulate cadmium (Cd) and zinc (Zn) was compared in Thlaspi goesingense and four populations of Thlaspi caerulescens . Two populations of T. caerulescens were grown in hydroponics at five concentrations of Cd. In addition, plants were grown in pots containing compost in which three different concentrations of Cd and two concentrations of Zn were added. A field trial was conducted to compare Zn and Cd uptake by three populations of T. caerulescens on nine selected plots of the Woburn Market Garden Experiment (UK) which had been contaminated to different degrees with heavy metals owing to past applications of sewage sludge. Results show that the four populations of T. caerulescens had the same ability to hyperaccumulate Zn but were significantly different in terms of Cd accumulation. Two populations of T. caerulescens from Southern France accumulated much more Cd than the populations from Prayon (Belgium) and Whitesike (UK). Generally, uptake of Cd was not decreased by increased concentrations of Zn in the substrate. These results indicate that the mechanisms of Cd and Zn hyperaccumulation are not identical in this species. This is the first report of hyperaccumulation of Cd by T. goesingense , but the growth of this species was markedly reduced by the large concentrations of Zn in the substrate. Future work should focus on the differences between Cd and Zn uptake in hyperaccumulator plants at the species and population level.  相似文献

During the last decade, the metal hyperaccumulating plants have attracted considerable attention because of their potential use in decontamination of heavy metal contaminated soils. However, in most species, little is known regarding the function, the ecological and the evolutionary significances of hyperaccumulation. In our study, we investigated the parameters influencing the Cd concentration in plants as well as the biological implications of Cd hyperaccumulation in nine natural populations of Thlaspi caerulescens. First, we showed that Cd concentration in the plant was positively correlated with plant Zn, Fe, and Cu concentrations. This suggested that the physiological and/or molecular mechanisms for uptake, transport and/or accumulation of these four heavy metals interact with each other. Second, we specified a measure of Cd hyperaccumulation capacity by populations and showed that T. caerulescens plants originating from populations with high Cd hyperaccumulation capacity had better growth, by developing more and bigger leaves, taller stems, and produced more fruits and heavier seeds. These results suggest a tolerance/disposal role of Cd hyperaccumulation in this species.  相似文献

Mine tailings can have a specific assemblage of plant species due to their unique physicochemical properties, and this process can be important in developing ecological theory and restoration practice. Physicochemical properties and natural colonization of plants on five lead/zinc (Pb/Zn) mine tailings in southern China were investigated. The tailings studied included Fankou and Lechang in Guangdong Province, and Huangshaping, Shuikoushan, and Taolin in Hunan Province. Physicochemical properties of the tailings varied greatly both among and within tailings ponds, but in general, all contained high concentrations of heavy metals (Pb, Zn, Cu, and Cd) and low concentrations of N, P, and organic matter. Toxic levels of heavy metals and deficiency of major nutrients appeared to be the major constraints for colonization of plants on these Pb/Zn tailings and were reflected in the metal concentration of the plant tissues. The natural colonization of plants on these tailings was limited, with only some small patches distributed mainly on the edge of tailing ponds and even fewer patches on the center of the ponds. In total 54 plant species belonging to 51 genera and 24 families were recorded on the five tailings ponds, of which the 13 species belonging to Gramineae were major components of the tailings’ flora. Species establishing on the tailings at the initial colonization phase greatly depended on their seed‐dispersal capacity. Further establishment and growth were then dependent on at least one of the three ecological strategies: (1) microsite (avoidance) strategy: plant establishment on tailings depended on dispersing onto microsites of relatively favorable edaphic conditions; (2) tolerance strategy: plant establishment was a result of evolving metal‐tolerant ecotypes or constitutional metal tolerance; and (3) rhizome strategy: plant establishment on tailings depended on clonal growth by rhizomatous extension.  相似文献

It was demonstrated recently that isolated tonoplast vesicles derived from plants of a Zn-tolerant ecotype of Silene vulgaris accumulate more Zn than vesicles derived from a Zn-sensitive ecotype. We have now characterized the tonoplast-transport system that causes this uptake difference and demonstrated its genetic correlation to Zn tolerance using plant crosses. We conclude that the tonoplast Zn uptake system of the tolerant ecotype differs greatly in its characteristics from that of the sensitive one, with the most prominent differences being its insensitivity to protonophores and ortho-vanadate and its stimulation by Mg-GTP. These differences in characteristics are most likely due to the fact that Zn can be taken up by two or more parallel pathways, which are not present in the same proportions in both ecotypes. In both ecotypes, Zn is actively transported across the tonoplast (temperature coefficient > 1.6), most likely as a free ion, since citrate does not accumulate in vesicles. Most importantly, the uptake difference found using the ecotypes was also found between homozygous Zn-tolerant and Zn-sensitive F3 plants, proving the genetic correlation between increased tonoplast Zn transport and naturally selected Zn tolerance in S. vulgaris.  相似文献

Cadmium (Cd) can enter soil through the use of fertilisers, calcareous, pesticides and industrial and/or domestic effluents. Cd can leach into groundwater and be taken up by plants, potentially leading to reductions in plant growth and yield. In soil, plant roots interact with heavy metal (HM)‐tolerant microorganisms that may promote plant growth. Soil microorganisms may also be able to solubilise or mobilise soil metals, thereby acting as bioremediators. A better understanding of the interaction among plants, metals, microorganisms and soil will lead to improved plant tolerance. Two multi‐tolerant bacteria from the Burkholderia genus were isolated from Cd‐contaminated and Cd‐uncontaminated soil of a coffee plantation. In addition to its high tolerance to Cd, the strain SCMS54 produces indole‐acetic acid (IAA), solubilises inorganic phosphate and produces siderophores, demonstrating its potential to contribute to beneficial plant–microorganism interactions. When interacting with tomato plants exposed to Cd, the bacterium led to decreases in plant peroxide and chlorosis levels, promoted relative plant growth and decreased the root absorption of Cd, resulting in increased plant tolerance to this highly toxic HM. The results indicated that the inoculation of tomato plants with Burkholderia sp. SCMS54 promotes better growth in plants cultivated in the presence of Cd. This phenomenon appears to be attributed to a mechanism that decreases Cd concentrations in the roots via a beneficial interaction between the bacteria and the plant roots.  相似文献

Two hydroponic experiments were performed to study the physiological responses to heavy metals (HMs) of two populations of Bituminaria bituminosa (L.) C.H. Stirton (Fabaceae): one (“C2”) from a site contaminated by HMs and one from a non-contaminated site (“LA”). In the first, we studied the effects of elevated concentrations of Zn (12 and 61 μM). Population C2 was more tolerant in terms of root and shoot growth at 61 μM Zn, relative to control plants (1 μM Zn). The similar tissue Zn levels of the two populations suggest that C2 is more tolerant of high tissue Zn. Of the parameters measured that could be related to Zn phytotoxicity (micro and macronutrients, root hydraulic activity, water-extractable Zn and organic acids), none could explain totally the superior tolerance of C2. In the second assay, the effects of Cd (4.4 μM), Cu (7.8 μM) and Zn (76 μM) on plant accumulation of the furanocoumarins (FCs) psoralen and angelicin, which function as feeding deterrents and photo-activated toxins, were assessed. For population C2, all three HMs increased the root FC concentrations, while Cd also raised shoot levels. For LA, Cu raised the root concentrations of both FCs. There was a relationship between plant stress, manifested as proline accumulation and disruption of plant water relations, and increased FC accumulation. Higher tissue levels of FCs likely provide greater protection against bacterial or fungal infection and herbivores.  相似文献

Developmental instability in morphological characters can occur during individual development due to various environmental stresses. Fluctuating asymmetry (FA) is often used as a measurement of developmental instability, but within-environment variation (CVe) is also considered an indicator of developmental instability. Cabbage aphid ( Brevicoryne brassicae ) populations were reared on zinc- (Zn) or cadmium- (Cd) contaminated cabbage and radish plants. Developmental instability indicators were measured and their relations with fitness were explored. Results revealed that cabbage aphids exposed to Cd and Zn displayed considerable developmental instability, particularly fluctuating asymmetry. Differences in developmental instability between the two metals were also detected, as well as differences between the two developmental instability measurements. For almost all measured traits, FA was greater on Cd- and Zn-contaminated compared to non-contaminated host plants. In contrast, CVe of some traits was greater on non-contaminated host plants, yet for other traits CVe was greater on contaminated host plants. There were also non-significant inverse relationships between FA and fitness of cabbage aphid populations. Due to weak correlations between FA and different patterns of two developmental instability measurements, this study does not support the hypothesis that developmental instability is a useful bioindicator of environmental quality.  相似文献

Growth, organic acid and phytochelatin accumulation, as well as the activity of several antioxidative enzymes, i.e. superoxide dismutase (SOD), ascorbate peroxidase (APX) guaiacol peroxidase (POX) and catalase (CAT) were investigated under Zn and Cd stress in hydroponically growing plants of Thlaspi caerulescens population from Plombières, Belgium. Tissue Zn and Cd concentration increased (the highest concentration of both was in roots) as the concentration of these metals increased in the nutrient solution. Increasing Zn concentration enhanced plant growth, while with Cd it declined compared to the control. Both metals stimulated malate accumulation in shoots, Zn also caused citrate to increase. Zn did not induce phytochelatin (PC) accumulation. In plants exposed to Cd, PC concentration increased with increasing Cd concentration, but decreased with time of exposure. Under Zn stress SOD activity increased, but APX activity was higher at 500 and 1000 μM Zn and CAT activity only at 500 μM Zn in comparison with the control. CAT activity decreased in Cd- and Zn-stressed plants. The results suggest that relative to other populations, a T. caerulescens population from Plombières, when grown in hydroponics, was characterized by low Zn and Cd uptake and their translocation to shoots and tolerance to both metals. The accumulation of malate and citrate, but not PC accumulation was responsible for Zn tolerance. Cd tolerance seems to be due to neither PC production nor accumulation of organic acids.  相似文献

Cysteine synthase [O-acetyl-L-serine(thiol)lyase] catalyzes the final step for L-cysteine biosynthesis in plants. The tolerance of transgenic tobacco plants over-expressing cysteine synthase cDNA in cytosol (3F), chloroplasts (4F) and in both organelles (F1) was investigated towards heavy metals such as Cd, Se, Ni, Pb and Cu. The transgenic plants were significantly more tolerant than wild-type plants in agar medium containing Cd, Se and Ni. The F1 transgenic plants had a higher resistance than other transgenic lines towards these metals and could enhance accumulation of Cd in shoot. These results suggest that the transgenic plants over-expressing cysteine synthase both in cytosol and chloroplasts can be applicable to phyto-remediation of Cd from contaminated soils.  相似文献

Phytochelatins (PCs) are heavy metal binding peptides that play an important role in sequestration and detoxification of heavy metals in plants. In this study, our goal was to develop transgenic plants with increased tolerance for and accumulation of heavy metals from soil by expressing an Arabidopsis thaliana AtPCS1 gene, encoding phytochelatin synthase (PCS), in Indian mustard (Brassica juncea L.). A 35S promoter fused to a FLAG–tagged AtPCS1 cDNA was expressed in Indian mustard, and transgenic lines, designated pc lines, were evaluated for tolerance to and accumulation of Cd and Zn. Transgenic plants with moderate AtPCS1 expression levels showed significantly higher tolerance to Cd and Zn stress, but accumulated significantly less Cd and Zn than wild type plants in both shoot and root tissues. However, transgenic plants with highest expression of the transgene did not exhibit enhanced Cd and Zn tolerance. Shoots of Cd-treated pc plants had significantly higher levels of phytochelatins and thiols than wild-type plants. Significantly lower concentrations of gluthatione in Cd-treated shoot and root tissues of transgenic plants were observed. Moderate expression levels of phytochelatin synthase improved the ability of Indian mustard to tolerate certain levels of heavy metals, but at the same time did not increase the accumulation potential for Cd and Zn.  相似文献

 植物长期生长在重金属污染的生境中,逐渐进化成不同的生态型。通过调查中国东南部古老Pb/zn矿和非矿山生境中的植物种群,发现生长在古老Pb/Zn矿的东南景天(Sedum alfredii Hance)是一种新的Zn超积累植物。在自然和控制条件下,古老Pb/Zn矿生态型比非矿山生态型植株的茎粗、叶片大、植株高。在矿山土壤Zn有效含量为105.5~325.4mg·kg-1时,矿山生态型东南景天植株地上部Zn含量为4134~5000mg·kg-1;当营养液中Zn浓度为1223.6μmol时,其Zn含量高达2%。在相同Zn浓度下,矿山生态型地上部Zn含量比非矿山生态型高30倍左右。两种生态型体内Zn分布也不同,古老铅锌矿山生态型的不同器官中Zn含量以茎>叶片>根系,而非矿山生态型则以根系>茎>叶片。古老铅锌矿山生态型地上部积累的Zn占植株总积累量的90%以上,其中叶片和茎分别占41.66%±5.46%和54.75%±5.87%;非矿山生态型各器官中积累的Zn远远低于古老铅锌矿山生态型,各器官中积累的Zn以茎>根系>叶片。本研究表明,这两种生态型东南景天的发现,为今后探讨植物耐高Zn胁迫和超积累Zn的微进化过程提供了非常有价值的材料,也为Zn污染土壤的植物修复提供了一种很有潜力的候选材料。  相似文献
