In view of recent findings which suggest that renal prostaglandins mediate the effect of hypoxia on erythropoietin production, we have studied whether hypoxia is a stimulus for in vitro prostaglandin synthesis. Studies were carried out in rat renal mesangial cell cultures which produce erythropoietin in an oxygen-dependent manner. Production rates of PGE2 and in specified samples also of 6-keto-PGF, as a measure of PGI2, and PGF were determined by radioimmunoassay after incubation at either 20% O2 (normoxic) or 2% O2 (hypoxic) in gas permeable dishes for 24 hrs. Considerable variation in PGE2 production was noted among independent cell lines. PGE2 production appeared to be inversely correlated to the cellular density of the cultures. In addition, PGE2 production was enhanced in hypoxic cell cultures. The mean increase was 50 to 60%. PGF and 6-keto-PGF increased by about the same rate. These results indicate that hypoxia is a stimulus for in vitro prostaglandin production.  相似文献

Preliminary characterization indicated the presence of separate prostaglandin (PG)E1 and (PG)F binding sites in membrane fractions prepared from bovine corpora lutea. These differ in the rate and temperature dependence of the specific binding. Equilibrium binding data indicate the apparent dissociation constants as 1.32 × 10−9M and 2.1 × 10−8M for PGE1 and PGF, respectively. Competition of several natural prostaglandins for the PGE1 and PGF bovine luteal specific binding sites indicates specificity for the 9-keto or 9α-hydroxyl moiety, respectively. Differences in relative ability to inhibit 3H-PG binding were found due to sensitivity to the absence or presence of the 5,6-cis-double bond as well.Bovine luteal function was affected following treatment of heifers with 25 mg PGF as measured by reduced estrous cycle length, decreased corpus luteum size and significantly decreased plasma progesterone levels. In contrast, treatment with 25 mg PGE1 resulted in cycle lengths comparable to those of non-treated herdmates with no apparent modification in corpus luteum size. However, plasma progesterone levels were increased significantly following PGE1 treatment compared to pretreatment values. In so far as data obtained in vitro on PGF relative binding affinity to the bovine CL can be compared to data obtained independently in vitro on PGF induced luteolysis in the bovine, PGF relative binding to the CL and luteolysis appeared to be associated. By similar reasoning, there was no apparent relationship between PGE1 relative binding affinity in the luteal fractions and luteolysis in estrous cyclic cattle.  相似文献

In vitro synthesis of progesterone and estradiol-17β from endogenous precursors was studied in the placenta from women in early stage of gestation (< 7 weeks). Radioimmunoassay techniques were used to measure progesterone and estradiol-17β.It was shown that placental tissue from as early as six weeks of gestation can synthesize both progesterone and estradiol-17β in vitro. Prostaglandins F and E2 in concentration of 100 μg/ml of the incubation media did not have any significant effect on the in vitro synthesis of progesterone and estradiol-17β in the placental tissue.It seems unlikely that the abortifacient effect of natural prostaglandins PGE2 and PGF is due to their direct action on the synthesis of progesterone and estradiol-17β in the placenta.  相似文献

The ability of various prostaglandins (PGs) to affect the in vitro anamnestic immune response of keyhole limpet hemocyanin (KLH)-primed rabbit popliteal lymph node cells was investigated. Of the four PGs studied (PGA1, PGE2 and PGF), PGE1 was found to have a stimulatory effect, whereas PGA1, PGE2 and PGF were ineffective in stimulating or inhibiting the in vitro anamnestic response. Under the conditions studied, a 3.5-fold increase in antibody production was obtained in PGE1-treated, KLH-stimulated cultures. Maximum enhancement was obtained when 0.2 μg of PGE1 were added at the time of culture initiation and were allowed to remain in contact with the lymph node cells for 24 hours.  相似文献

Ten to 60 minutes following a single i.v. injection of PGE2 (500 μg/rat) into male rats of 30 to 35 days of age FSH concentration in the serum was raised significantly. The rise in FSH was maintained from 10 to 60 minutes after treatment, then at 90 minutes FSH had declined and was not significantly different from that of the control before treatment. Prostaglandin E1, E2 or F (670μg/rat) significantly increased the serum prolactin level 10 to 60 minutes after a single i.v. injection in spayed rats primed with estrogen and progesterone. And, rats primed with estrogen and progesterone. And, increases in prolactin in the serum were observed with as little as 2μg of PGE1 or E2, and 20μg of PGF. Twenty μg of PGE2, and 200μg of PGE1 or F gave the maximum stimulation. These results indicate that release of pituitary hormones is affected by prostaglandins.Prostaglandins (PGs) are widely distributed in mammalian tissues, and they have been reported to have an almost equally wide variety of endocrine and metabolic effects. It was recently postulated that PGs may be involved in the process of ovulation because ovulation was blocked by inhibitors of PG synthesis (1–5).  相似文献

Abortion or delivery were induced by extra-amniotic instillation of Rivanol during the second trimester in twelve patients and during the third trimester in two patients with fetal death and one patient with fetal acrania. Serial sampling of amniotic fluid was performed through a transabdominal catheter and the levels of free arachidonic acid (AA), prostaglandin F2α (PGF2α), prostaglandin E2 (PGE2), 6-keto-prostaglandin F1α (6-keto-PGF1α) and thromboxane B2 (TXB2) were determined. The levels of AA, PGF2α, PGE2, 6-keto-PGF1α and TXB2 in amniotic fluid increased significantly during induction with the exception of AA in fetal death which was high and remained constant during induction. Furthermore, PGF2α, 6-keto-PGF1α and TXB2 were all significantly correlated to AA.These observations suggested that free AA is released during Rivanol-induction of abortion and labour giving an increased synthesis of PGF2α, PGE2 prostacyclin and thromboxane A2 in the fetal membranes and the decidua but not in the fetus. This increase might be relevant for the initiation and progress of abortion and labour in these patients.  相似文献

Prostaglandins E2 and D2 were both converted to prostaglandin F (9α, 11α) by an enzyme present in sheep blood. Neither the 9β, 11α epimer nor the 9α, 11β epimer was produced from PGE2 or PGD2 respectively. Guinea pig ear epidermal cells (keratinocytes) were established in primary cultures using trypsin, and treated in their proliferative phase of growth with prostaglandins E1, D1, F, E2, D2, or F. This phase is induced by the addition of retinoic acid during cell plating. Intracellular content of cAMP and cGMP was measured by radioimmunoassay at various times after treatment.Maximum stimulation of cAMP levels was observed with PGD2, smaller increases with PGE2 and relatively transient rises with PGF which were of low significance, but confirm earlier data. Similar results were observed with PGD1, PGE1, and PGF with smaller increases. The effects of D and E PGs were biphasic. Significant increases in cGMP were immediately observed with PGD2 and PGE2. With PGF, maximum cGMP levels were noted after some delay.All PGs tested showed some effect in elevating cyclic nucleotides in keratinocytes. The most striking result was the increase in cAMP on PGD2 treatment.  相似文献

The relative potencies of the prostaglandins A1, A2, E1, E2, F and their 15-keto-, 15-keto-13,14-dihydro-, and 13,14-dihydro-metabolites were investigated on isolated lamb ductus arteriosus preparations contracted by exposure to elevated PO2. All the prostaglandins (except PGF and its 15-keto-metabolites) relaxed the tissue. However, only PGE1, E2, and their 13,14-dihydro-metabolites, were effective at concentrations below 10−8 M. Therefore, events that alter metabolism of circulating PGs in the perinatal period may have significant effects on the relative patency or closure of the ductus arteriosus.  相似文献

Dose-response curves to prostaglandins (PG) E1, E2, F, F, A1 and A2 were carried out in the rat stomach fundus, rat colon, chick rectum and rabbit aortic strip superfused with Krebs solution containing adrenergic, cholinergic, serotonergic and histaminergic blockers, and arranged in a cascade fashion. In the stomach and rectum, the order of potency was PGE's > PGF's > PGA's. In the aorta and colon, PG2's were more active than their corresponding PG1's. By determining the colon/stomach and aorta/colon activity ratios, it was possible to define differential patterns of activity for PGE1, PGE2, PGF, PGF and PGA's. The present results indicate the importance of determining complete dose-response curves in the cascade for the characterization of PG-like activity.  相似文献

Radioimmunoassays of platelet prostaglandins E1 and F in platelet rich plasma or platelet suspension, demonstrate that both PGE1 and PGF are present at higher concentrations than prostaglandins E2 and F. Gas chromatography — mass spectrometry determinations of prostaglandins E1 and E2 in resting washed platelets confirm this difference. Lastly, there is a greater incorporation of [1-14C] acetate into prostaglandins E1 and F compared to that into prostaglandins E2 and F.  相似文献

The present study has been performed to investigate how PGs would participate the hatching process. Effects of indomethacin, an antagonist to PGs biosynthesis, on the hatching of mouse blastocysts were examined in vitro. Furthermore, it was studied that prostaglandin E2 (PGE2), prostaglandin F (PGF) or 6-keto-prostaglandin F (6-keto-PGF) were added to the culture media with indomethacin. (1) The hatching was inhibited by indomethacin yet the inhibition was reversible. (2) In the groups with indomethacin and PGE2, no improvement was seen in the inhibition of hatching and the inhibition was irreversible. (3) In the groups with indomethacin and PGF, inhibition of hatching was improved in comparison with the group with indomethacin. (4) In the groups with indomethacin and 6-keto-PGF, no improvement was seen. The above results indicated that PGF possibly had an accelerating effect on hatching and a high concentration of PGE2 would exert cytotoxic effect on blastocysts.  相似文献

We have investigated the direct effects of prostaglandins E1, E2, F and D2 on renin release from rabbit renal cortical slices. Prostaglandin E1 (PGE1) was the most potent stimulant of renin release, while PGE2 was 20–30 fold less active. PGF was found not to be an inhibitor of renin release as reported by others, but rather a weak agonist. PGD2 up to a concentration of 10 μg/ml had no activity in this system. That the stimulation of renin release by PGE1 is a direct effect is supported by the finding that PGE1-induced release is not blocked by L-propranolol or by Δ5,8,11,14-eicosatetraynoic acid (ETYA), a prostaglandin synthesis is inhibitor. The fatty acid precursor of PGE1, Δ8,11,14-eicosatrienoic acid, also stimulated renin release, an effect which was blocked by ETYA. In addition to the above findings, ethanol, a compound frequently used to dissolve prostaglandins, was shown to inhibit renin release.  相似文献

To elucidate the role of prostaglandins in adrenal steroidogenesis, we studied aldosterone and corticosterone responses to
of prostaglandin E2 (PGE2), prostaglandin F (PGF), prostacyclin (PGI2), and arachidonic acid (AA) in collagenase dispersed rat adrenal capsular and decapsular cells. Whereas adrenocorticotrophic hormone (ACTH) and angiotensin II (AII) stimulated aldosterone production in capsular cells and ACTH stimulated corticosterone production in decapsular cells in a dose dependent fashion, aldosterone and corticosterone production were not stimulated significantly by PGE2, PGF, PGI2, and AA. Although preincubation of dispersed adrenal cells with indomethacin ( ) markedly inhibited PGE2 synthesis, ACTH- and AII-stimulated aldosterone production and ACTH-stimulated corticosterone production were not attenuated despite prostaglandin blockade. These results indicate that prostaglandins are unlikely to play an important role in adrenal steroidogenesis.  相似文献

The dose-dependent effects of 9 prostanoids (PGA1, PGA2, PGE1, PGE2, PGF, PGF, PGD2, PGI2, 6 keto- PGF) on metabolism of cultured bovine articular chrondrocytes were investigated. Most prostanoids dose-dependently inhibited 35SO4= and 3H-glycine incorporation. At 25 μg/ml, the inhibitory sequence was A2D2>E2 = E1 = A1>6 keto-F1α>F1>F2, but sensivity (lowest dose eliciting inhibition) followed the sequence E2 > 6 keto-F1α = F1 > A2 = D2>E1>A1. At 25 μg/ml PGA2 also inhibited incorporation of 3H-cytidine and #H-thymidine, but had no significant effect on 3H-glucose or 14C-xylose incorporation. The inhibitory effect of PGA2 was apparent after 30 minutes exposure for 35SO4= and after 60 minutesd for 3H-cytidine, and was still present up to 72 hours following incubation in fresh non-PG-containing medium. PGI2 had no significant effect of 35SO4= incorporation but at concentrations below 10 μg/ml enhanced uptake of 3H-glycine.The PG-induced inhibitory effect was apparently not due to cell damage as indicated by measurement of 3H-glucose metabolism and lactate production.  相似文献

The receptors mediating prostanoid-induced contraction of guinea-pig isolated trachea have been characterised in terms of a recently proposed general classification of prostanoid receptors. Results obtained on the trachea were compared with those obtained on guinea-pig fundus, which contains a sub-type of PGE2-sensitive (EP-) receptor termed the EP1-receptor, and guinea-pig lung strip, which contains a thromboxane-sensitive or TP-receptor. The following agonists were studied, PGE2, PGF2α and the thromboxane-like agonists U-46619 and Wy17186. The antagonists studied were SC-19220 which selectivity blocks EP1-receptors, and AH19437 which selectively blocks TP-receptors. On guinea-pig fundus the rank order of agonist potency was PGE2 > PGF2α > Wy17186 U-46619, and responses to all agonists were antagonised by SC-19220 but not by AH19437. On guinea-pig lung strip the rank order of potency was U-46619 > Wy17186 PGF2α > PGE2 and responses to all agonists tested were blocked by AH19437 but not by SC-19220. On the trachea, the rank order was PGE2 = U-46619 > Wy17186 = PGF2α. SC-19220 antagonised responses to PGE2 and PGF2α, but not those to U-46619 or Wy17186. Conversely, AH19437 antagonised responses to U-46619 and Wy17186 but not those to PGE2 or PGF2α. It is concluded that prostanoid-induced contractions of guinea-pig trachea can be mediated by both EP1- and TP-receptors.  相似文献

Fluorescent esters of the prostablandins D2, E2, F2α, and 6-keto-F1α and of thromboxane B2 have been prepared using the reagent 4-bromomethyl-7-methoxycoumarin. All of these derivatives can be separated in a single run either by thin-layer or high-performance liquid chromatography (TLC or HPLC). As little as 20 ng of PGE2 can be detected after derivatization and HPLC analysis. Identification of thromboxane B2 produced by human platelets and of 6-keto-PG F1α produced by bovine aortic microsomes has been achieved with this method.  相似文献

The release of prostaglandin E2 and F, thromboxane B2 and 6-keto-prostaglandin F was measured in isolated human placental cotyledons perfused under high- and low-oxygen conditions. Also the effect of reoxygenation on prostaglandin production was studied. During the high-oxygen period, prostaglandin E2 accounted for 44 % and 6-keto-prostaglandin F for 28 % of all prostaglandin release, and the rank order of prostaglandin release was E2 > 6-keto-prostaglandin F > thromboxane B2 > prostaglandin F. Hypoxia had no significant effect on quantitative prostaglandin release, but the ration of prostaglandin E2 to prostaglandin F was significantly increased. After the hypoxic period during reoxygenation the release of 6-keto-prostaglandin F was significantly decreased, as was the ratio of 6-keto-prostaglandin F to thromboxane B2. Also the ratio of the vasodilating prostaglandins (E2, 6-keto-prostaglandin F) to the vasocontricting prostaglandins (thromboxane B2, prostaglandin F) was decreased during reoxygenation period. With the constant flow rate, the perfusion pressure increased during hypoxia in six and was unchanged in three preparation. The results indicate that changes in the tissue oxygenation in the placenta affect prostaglandin release in the fetal placental circulation. This may also have circulatory consequences.  相似文献

Pretreatment of membranes for 1 hr at 4° with up to 0.1% Triton X-100 (TX-100) and sodium desoxycholate (SDC), resulted in a greater loss of [3H] prostaglandin (PG)F2α binding compared to E1 binding. Lubrol WX (LWX) tended to cause a greater loss of [3H]PGF2α than E1 binding. However, the differential loss was not as marked as with TX-100 or SDC. Triton X-305 was relatively ineffective, but loss of [3H]PGE1 binding was greater than for PGF2α. Increasing concentrations of dimethylsulfoxide (DMSO) progressively inhibited PGF2α binding without affecting PGE1 binding. The detergent, but not DMSO, induced losses of membrane PG binding were due to solubilization of the receptors. Greater amounts of membrane protein and phospholipids were solubilized at detergent (TX-100 and SDC) concentrations that solubilized 100% of PGE1 receptors compared to 100% solubilization of F2α receptors. Neither the duration of preincubation nor the amount of membrane protein chosen were responsible for differential PGE1 and F2α receptor losses. These differential membrane PG receptor losses raise the possibility of differences in PGE1 and F2α receptors association with the membrane structure.  相似文献

A subcellular fraction was isolated from uteri of non-pregnant and pregnant cows. ATP-dependent calcium binding was shown to take place in this fraction. This calcium binding was inhibited in a dose related fashion when increasing amounts of prostaglandin (PG) E2 or F were added to the in vitro experimental medium. The physiologically inactive PGF had no inhibitory effect. Oxytocin caused inhibition of calcium binding in preparations from both pregnant and non-pregnant cows. The response to PGE2 and PGF was somewhat greater in preparations from pregnant uteri than from non-pregnant uteri. The response to oxytocin was very much greater in pregnant uteri. Because of the high PG sensitivity of calcium binding in preparations from the non-pregnant uterus, it is concluded that the PGs may be the more suitable agent in the control of reproduction.  相似文献
