Over a period of three years, 165 cyclic or anestrous Hanoverian mares received 177 treatments with 2, 3 or 4 mg of a novel luteolytic prostaglandin analog, K 11941. Heat and ovulations, indicating luteolysis, were observed after an average of 3.98 and 7.62 days, respectively, in 88.04% of 142 treated cyclic, postpartum and anestrous mares, and mares after an early loss of the conceptus. All doses tested were effective in inducing luteolysis as confirmed by determination of progesterone blood levels in samples collected daily, in 70 of 80 mares studied (87.5%). Attempts to achieve control of the cycle by various methods revealed that with K 11941 given once or twice, alone or in combination with hCG and/or an GnRH analog (Hoe 766), one can effectively concentrate estrus periods and follicular growth patterns, but can neither synchronize nor concentrate ovulations. Since most of the mares treated were confirmed problem mares, the pregnancy rate of 40.0% from first insemination at drug induced estrus, was regarded as satisfactory when compared to a pregnancy rate of 43.8% obtained with natural breeding in the same population. No drug related clinical signs of side-effects were observed.  相似文献

The effects of oxytocin and two prostaglandin (PG) F(2)alpha analogues, prostalene and alfaprostol, on uterine pressure in the mare were measured using balloon-tipped catheters connected to pressure transducers. The PGF(2)alpha analogues caused increased uterine pressure beginning 7 to 15 min postinjection and persisting for the duration of each 60 min recording session. Forty postpartum mares of light-horse breed were used to evaluate the effects of prostalene on postpartum pregnancy rate. Eighteen mares were injected by aseptic technique subcutaneously with 1 mg prostalene twice daily, beginning on the day of foaling (Day 0) and continuing for 10 consecutive days (Day 10) or until the mare was first bred at foal heat. Twenty-two postpartum mares were injected with 1.0 ml sterile saline by the same technique as the controls. Of treated mares, 76.9% were diagnosed pregnant after breeding versus 44.4% of the control mares (P = 0.07). Of treated mares, 66.7% bred at their second postpartum estrus became pregnant versus 28.6% of control mares (P = 0.03). Prostalene, given at 1 mg twice daily for 10 d postpartum, produced an increased pregnancy rate after both foal heat and second postpartum estrus breedings in the mare.  相似文献

In two field trials, the parturition inducing and MMA preventing effects of the prostaglandin F(2alpha) analog (PGFA), K 11941, were explored. In trial 1, 100 sows were treated with 2 mg and 30 with 3 mg of K 11941 i.m. on day 112 of gestation; 125 sows treated on day 112 with 175 mcg cloprostenol (Planate, ICI) served as positive controls, and 248 sows treated with saline on day 112 of gestation were used as negative controls. Both PGF analogs were equally effective in inducing early parturiton: 84.6% and 83.2% of the sows had farrowed between 20 and 30 hours after treatment. Both treatments reduced stillbirth rate and piglet losses during the first 10 days of life, and lowered the incidence of MMA significantly (6.2 and 5.6% vs 27.3 and 24.2%; p<.001). In a second trial, parturitions were induced in four groups of 60 sows each with 2 mg K 11941, housed in farms with either a chronic high incidence of MMA (A: 45 to 65% or only a seasonal rise in MMA during summer (B: 10 to 50%). Groups were treated either in April/May or August/September; 240 salinetreated, control sows were used. K 11941-induced parturition reduced MMA incidence in farm A during both seasons and prevented the summer rise in farm B (p<.01). Fertility after weaning was impaired in herds with an increased incidence of MMA.  相似文献

A prostaglandin F analogue was studied in anestrous mares: a dose-response study; a study in mares presumed pregnant; and a field evaluation of effective doses in breeding establishments. A dose of 2.0mg given by single subcutaneous injection to mares with initial plasma progesterone levels greater than 1.0ng/ml, caused luteolysis on the basis of decline in plasma progesterone concentrations. Follicle maturation leading to ovulation, accompanied by estrus, was observed, and fertility at mating either by natural service or artificial insemination was satisfactory. A dose of 1.0mg was generally effective for luteolysis, but pregnancy rates were lower than after 2.0mg. A proportion of mares which had less than 1.0mg of plasma progesterone at the time of injection ovulated and became pregnant.  相似文献

It has become a common practice in the equine breeding industry to send 2 insemination doses for breeding with transported cooled semen, one to be used for the initial insemination upon arrival, and the other to be held a second insemination the next day. One fertile stallion and 36 fertile mares were used to determine if breeding once with 1 dose of semen cooled for 24 h would improve fertility compared with breeding twice, 1 d apart, with half the dose of semen cooled for 24 h on the first day of breeding and half cooled for 48 h on the second day of breeding. Mares were given two intramuscular injections of 10 mg PGF2 alpha 14 d apart. Following the second injection, mares were teased with a stallion and their ovaries were scanned by transrectal ultrasonography daily. When a dominant follicle (> 35 mm diameter) was detected, 1500 units hCG were injected intravenously, and the mares were inseminated. Semen was collected in advance of anticipated breeding, mixed in nonfat dry milk solids-glucose extender to a concentration of 25 million sperm/mL, and placed in 2 commercial cooling containers for 24 or 48 h of storage prior to breeding. Mares were randomly assigned to 1 of 2 insemination treatment groups: 1) Group T1 (n = 18), in which mares were inseminated on the day of hCG injection with 500 million spermatozoa cooled for 24 h, or 2) Group T2 (n = 18), in which mares were inseminated on the day of hCG injection with 250 million spermatozoa cooled for 24 h, and again on the following day with 250 million spermatozoa cooled for 48 h. Pregnancy status was confirmed by transrectal ultrasonographic examination at 14 and 16 d after ovulation. Pregnancy rates were the same for both insemination treatment groups (12/18; 67%). There was no advantage to holding half of the insemination dose for rebreeding on the following day.  相似文献

To evaluate the technique of ultrasound-guided luteal injection in mares, PGF2alpha was administered under ultrasound guidance to horse mares (n = 7 to 9 per group) on Day 9 postovulation via either a systemic (i.m.; zero, 0.01, 0.1, or 5 mg/dose) route or a local intraluteal (i.l.; zero, 0.01 or 0.1 mg/dose) route. The luteolytic efficacy of each treatment was determined based on post-treatment decreases in progesterone concentration, interval to uterine edema (IE) and interovulatory interval (IOI). Local administration of PGF2alpha directly into the CL consistently induced luteolysis, at doses up to 50-fold lower than the lowest effective systemic dose. Significant decreases in IOI and IE occurred in mares treated with 5 mg PGF2alpha i.m. or 0.1 mg PGF2alpha i.l., but did not occur in mares treated with 0.1 or 0.01 mg PGF2alpha i.m., 0.01 mg PGF i.l., vehicle i.l. or vehicle i.m.. Progesterone concentrations were reduced to less than 10% of pretreatment values by two days post treatment in mares treated with 5 mg PGF2alpha i.m. or 0.1 mg PGF2alpha i.l.. PGF2alpha doses of 0.1 mg i.m. and 0.01 mg i.l. were associated with smaller but significant progesterone decreases (to 66% and 46% of pre-treatment values, respectively) by two days post treatment. Progesterone values after administration of i.l. vehicle did not differ from pre-treatment values by two days post treatment, but were significantly lower (53% of pre-treatment values) by four days post treatment. Intramuscular treatment with vehicle or 0.01 mg of PGF2alpha did not significantly reduce progesterone concentrations below pretreatment values. Overall, the minimum effective luteolytic dose of PGF2alpha given intraluteally was between 0.01 and 0.1 mg. Based on the results of this study, ultrasound-guided i.l. injection appears to be a repeatable method for studying the direct effect of other chemicals on luteal function. However, the current procedure carries some risk, since three i.l. injections were associated with ovarian abscesses.  相似文献

Horses are about five times more sensitive to the luteolytic effect of prostaglandin F2alpha (PGF) than cattle, as indicated by a recommended clinical dose of 5 mg in horses and 25 mg in cattle. Novel evaluations of the PGF plasma disappearance curves were made in mares and in heifers, and the two species were compared. Mares and heifers (n = 5) of similar body weight were injected (Min 0) intravenously with PGF (5 mg per animal). Blood was sampled every 10 sec until Min 3, every 30 sec until Min 5, every 10 min until Min 60, and every 30 min until Min 240. The mean PGF concentration was greater (P < 0.05) in mares than in heifers at Min 1 through Min 60 and at Mins 180 and 240. The mean time to maximum PGF concentration was not different between mares (42.0 ± 8.6 sec) and heifers (35.0 ± 2.9 sec). The apparent plasma clearance, distribution half-life, elimination half-life, and maximum plasma PGF concentration were 3.3 ± 0.5 L h(-1) kg(-1), 94.2 ± 15.9 sec, 25.9 ± 5.0 min, and 249.1 ± 36.8 ng/ml, respectively, in mares and 15.4 ± 2.3 L h(-1) kg(-1), 29.2 ± 3.9 sec, 9.0 ± 0.9 min, and 51.4 ± 22.6 ng/ml, respectively, in heifers. Plasma clearance was about five times less (P < 0.0005), maximum plasma PGF concentration was five times greater (P < 0.002), and the distribution half-life and elimination half-life were about three times longer (P < 0.005) in mares than in heifers. The fivefold greater plasma clearance of PGF in heifers than in mares corresponds to the recommended fivefold greater clinical dose of PGF in cattle and supported the hypothesis that the metabolic clearance of PGF is slower in mares than heifers.  相似文献

Equine pituitary extract (EPE) has been reported to induce heightened follicular development in mares, but the response is inconsistent and lower than results obtained in ruminants undergoing standard superovulatory protocols. Three separate experiments were conducted to improve the ovarian response to EPE by evaluating: (1) effect of increasing the frequency or dose of EPE treatment; (2) use of a potent gonadotropin-releasing hormone agonist (GnRH-a) prior to EPE stimulation; (3) administration of EPE twice daily in successively decreasing doses. In the first experiment, 50 mares were randomly assigned to one of four treatment groups. Mares received (1) 25 mg EPE once daily; (2) 50 mg EPE once daily; (3) 12.5 mg EPE twice daily; or (4) 25 mg EPE twice daily. All mares began EPE treatment 5 days after detection of ovulation and received a single dose of cloprostenol sodium 7 days postovulation. EPE was discontinued once half of a cohort of follicles reached a diameter of >35 mm and hCG was administered. Mares receiving 50 mg of EPE once daily developed a greater number (P = 0.008) of preovulatory follicles than the remaining groups of EPE-treated mares, and more (P = 0.06) ovulations were detected for mares receiving 25 mg EPE twice daily compared to those receiving either 25 mg EPE once daily and 12.5 mg EPE twice daily. Embryo recovery per mare was greater (P = 0.05) in the mares that received 12.5 mg EPE twice daily than those that received 25 mg EPE once daily. In Experiment 2, 20 randomly selected mares received either 25 mg EPE twice daily beginning 5 days after a spontaneous ovulation, or two doses of a GnRH-a agonist upon detection of a follicle >35 mm and 25 mg EPE twice daily beginning 5 days after ovulation. Twenty-four hours after administration of hCG, oocytes were recovered by transvaginal aspiration from all follicles >35 mm. No differences were observed between groups in the numbers of preovulatory follicles generated (P = 0.54) and oocytes recovered (P = 0.40) per mare. In Experiment 3, 18 mares were randomly assigned to one of two treatment groups. Then, 6-11 days after ovulation, mares were administered a dose of PGF2, and concomitantly began twice-daily treatments with EPE given in successively declining doses, or a dose of PGF2alpha, but no EPE treatment. Mares administered EPE developed a higher (P = 0.0004) number of follicles > or = 35 mm, experienced more (P = 0.02) ovulations, and yielded a greater (P = 0.0006) number of embryos than untreated mares. In summary, doubling the dose of EPE generated a greater ovarian response, while increasing the frequency of treatment, but not necessarily the dose, improved embryo collection. Additionally, pretreatment with a GnRH-a prior to ovarian stimulation did not enhance the response to EPE or oocyte recovery rates.  相似文献

Breeding of mares at the foal heat is desirable, but the low pregnancy rate and the not infrequent occurrence of subsequent anestrus are a deterrent. A PGF2a analogue, Synchrocept? (prostalene), as a single 2 mg subcutaneous injection was used to induce ovulation, either during the corpus luteum phase subsequent to a foal heat (test group 1) or at a fixed interval of 20 days from parturition (test group 2) in mares that also had a foal heat. Mares with no signs of a foal heat were treated 20 days postpartum (test group 3). A total of 80, mostly Thoroughbred, mares were treated in more than 15 breeding centers in Northern Germany between mid-January and mid-May 1975. At the same locations and over the same time periods, 60 control mares (control group 1) were bred at the foal heat and 40 additional control mares (control group 2) were bred later than 25 days postpartum.In groups 1 and 2, treatment induced estrus in 95% of the mares and in group 3 estrus was induced in 68% of the mares. Of these animals, 92% were bred and 81% conceived on the average 28 days after parturition. The number of services per conception in mares treated was 25% lower (2.5) than in controls (3.2). Synchrocept acted as a luteolysin, but seems also to have induced heat and estrus in animals with plasma progesterone levels below 1 ng/ml, as has been reported previously. Neither mares nor foals showed detectable clinical signs of drug intolerance.  相似文献

The objective was to evaluate the effects of giving prostaglandin F (PGF) to donor mares 48 h prior to embryo collection. Non-lactating donor mares (n = 20 estrous cycles in 10 mares), ranging from 2.5 to 10 y of age and 400 to 500 kg of body weight were used from September 2004 to February 2005 in the southern hemisphere (Brazil). Donor mares were randomly assigned in a cross-over design study. During a Treated cycle, 7.5 mg PGF was given 48 h prior to embryo collection, whereas in the Control cycle, 7.5 mg PGF was given at embryo collection. In Treated Cycles, serum progesterone concentrations decreased between the day of PGF treatment and the day of embryo collection (13.9 ± 5.4 and 0.5 ± 0.3 ng/mL, respectively; P < 0.05). In Treated versus Control cycles, the interovulatory interval was shorter (14.9 ± 0.9 vs 17.5 ± 1.1 d, P < 0.05). However, there was no significant difference between these groups for the interval from PGF to ovulation (average, 9.8 d), embryo recovery rate (average, 75%), embryo quality, uterine protein concentration, and pregnancy rate in recipient mares (average, 87% at 15 d after ovulation, with no pregnancy loss detected by 60 d). In conclusion, giving donor mares PGF 48 h prior to embryo collection reduced the average interovulatory interval by approximately 2.5 d, thereby potentially increasing the numbers of embryos that could be collected during a breeding season, with no deleterious effects on embryo recovery rate, embryo quality, or pregnancy rate in recipient mares.  相似文献

In the present study, the kinetics of the prostaglandin F2alpha (PGF2alpha)-metabolite 15-keto-13,14-dihydro-PGF2alpha after a single intramuscular application of various doses of the natural PGF2alpha dinoprost at Day 7 of the cycle in the mare were investigated. Effects of low doses on estrous cycle length and life span of corpus luteum were examined, because release of PGF2alpha is still under discussion to have detrimental influence on success rates of transcervical transfer of equine embryos. Eight Shetland pony mares were each randomly assigned to each of four treatments: (a) 0.8 mg/100 kg (group T1), (b) 0.4 mg/100 kg (group T2), (c) 0.2 mg/100 kg BM dinoprost i.m. (group T3), and (d) 1 ml physiological saline i.m. (group CO). Treatments were administered as single doses on Day 7 of the estrous cycle. Administration of dinoprost caused dose-dependent rises of plasma concentrations of PGF2alpha-metabolite, although values of individual mares showed great variation within groups. Prostaglandin treatments resulted in a distinct decrease of plasma progesterone concentrations to values between 1.6 and 7.9 ng/ml within 24 h. Treatment groups had significantly lower progesterone area under the curve (AUC: T1 942.8+/-175.9, T2 1050+/-181.2 and T3 1117+/-179.8 ng/ml/h) when compared with controls (CO 1601.9+/-227.6; t-test, P<0.05 ). There was a small, but significant negative correlation between AUC of progesterone and of PGF2alpha-metabolite ( R=-0.4; P=0.05 ). Administration of PGF2alpha caused secretion of oxytocin in three (T1, T2) and two (T3) mares out of eight ranging from 19.3 to 63.1 pg/ml. The AUC of oxytocin was positively correlated with AUC of PGF2alpha-metabolite ( R=0.4, P<0.05) and negatively correlated with AUC of progesterone ( R=-0.4, P<0.05). Administration of dinoprost yielded significantly shorter intervals from treatment to estrus and ovulation (values in parentheses), respectively, when compared with controls: T1 3.9+/-0.7 days ( 12.1+/-0.7 days), T2 4.5+/-0.6 ( 12.3+/-0.6 ), T3 4.9+/-0.5 ( 12.3+/-0.6 ), and CO 8.9+/-0.6 days ( 16.5+/-0.8 days) (t-test, P<0.01 ) (Fig. 2). Different doses of PGF2alpha caused similar effects. Data suggest that progesterone concentrations at applications influence efficacy of treatments more than doses administered, as demonstrated by their high correlation with estrous cycle patterns. It is important to note that differences we achieved are gradual and that all mares responded to treatment by luteolysis and premature estrus, regardless of doses applied.  相似文献

Ovum pick-up (OPU) by transvaginal ultrasound guided aspiration (TUGA) is a procedure applied in equine-assisted reproduction programs such as oocyte transfer and in vitro embryo production. Despite a large number of studies reporting that it is a repeatable and safe technique, little information is available about the effect of repeated punctures on fertility of mares. Moreover, even if flushing follicles improves the oocyte recovery rate, to our knowledge the efficiency of flushing estrous and diestrous follicles has not been evaluated. The aims of the present study were (1) evaluate if repeated TUGAs negatively effects fertility and (2) investigate the influence of flushing the follicular cavity (as compared to aspiration only-unflushed) on the recovery rate from follicles of different sizes and in different stages of the estrous cycle. Seventy-six TUGAs were carried out on 20 mares during the breeding season; 153 follicles were aspirated and 31 oocytes were recovered (20.3% per follicle; 40.8% per TUGA attempt). Of the 76 aspirations, 52 were carried out during estrus and 24 in diestrus. Flushing the follicular cavity significantly increased (P < 0.01) the oocyte recovery rate from estrous follicles (13/28, 46.4% flushed versus 3/24, 12.5% aspirated only) but not (P > 0.05) from diestrous follicles of different diameters (3/30, 10% flushed versus 2/36, 5.6% aspirated only for follicles <2 cm in diameter; 6/20, 30% flushed versus 4/15, 26.7% aspirated only for follicles > or =2 cm in diameter). Mares underwent ultrasonic examinations after every aspiration and no alteration was found with the exception of two mares in which the corpus luteum (CL) did not form following aspiration of estrous follicle. Of the 20 mares involved in this study, 10 were artificially inseminated with fresh semen from a single fertile stallion at the first spontaneous heat following the previous aspiration. Of the 10 inseminated mares, 7 were found to be pregnant 16, 30 and 50 days after artificial insemination (AI), indicating that repeated TUGAs did not adversely affect fertility.  相似文献

Two experiments were conducted to compare estrous response to three doses (8, 16 and 24 mg) of prostaglandin F(2alpha) (PGF(2alpha)) administered by intramuscular injection to ewes between day 6 and 12 of the estrous cycle (Experiment I) and to ewes on unknown days of the estrous cycle during four different stages of the breeding season (Experiment II). In experiment I, a total of 41 ewes were treated with PGF(2alpha). The injection of 24 mg PGF(2alpha) resulted in a higher proportion of ewes exhibiting estrus (13 14 , 92.9%) within 5 days after treatment as compared to the other two doses (2 12 and 10 15 , for 8 and 16 mg PGF(2alpha), respectively). However, there was no significant difference for the proportion between 16 mg and 24 mg PGF(2alpha). In experiment II, PGF(2alpha) was given to ewes on the 3rd of February (early breeding season), the 28th of February (mid-early breeding season), the 10th of April (mid breeding season) and the 27th of May (late breeding season). These was a significant difference for the proportion of ewes exhibiting estrus between the early breeding season and the other three seasons (P < 0.05) but not for ewes ovulating. Throughout the breeding season, 16 mg PGF(2alpha) appeared to be slightly better than the other two doses (8 and 24 mg) although there was no overall difference in the estrous responses to treatment among the three doses. However, a significant difference in the proportion of ewes ovulating was found among the three doses of PGF(2alpha) (P < 0.05). Especially, 16 mg PGF(2alpha) was significantly superior to 8 mg (P < 0.01) and 24 mg (P < 0.05). It was considered that there was a complicated relationship between the doses of PGF(2alpha) and the stages of the breeding season for induction of estrus and ovulation in the ewe.  相似文献

The present study determined whether a 5-d progesterone-based CO-Synch protocol with a single dose of prostaglandin F (PGF) at progesterone withdrawal on Day 5, would yield a timed AI pregnancy rate similar to two doses of PGF given 6 h apart on Day 5. Angus cross beef heifers (N = 562) at six locations were used. All heifers received 100 µg of gonadorelin hydrochloride (GnRH) and a controlled internal drug release (CIDR) insert on Day 0. Within farm, heifers were randomly allocated to receive one dose of 25 mg dinoprost (PGF) at CIDR removal on Day 5 (1 PGF; N = 264), or two doses of 25 mg PGF, with the first dose given on Day 5 at CIDR removal, and the second dose 6 h later (2 PGF; N = 298). Most heifers (N = 415) received a heat detector patch at CIDR removal. After CIDR removal, heifers were observed twice daily through Day 7 for estrus and heat detector aid status was recorded. On Day 8, heifers were given 100 µg of GnRH, heat detector aid status was recorded, and heifers were inseminated approximately 72 h after CIDR removal. Accounting for significant variables such as location (P < 0.01), heifers in estrus at or prior to AI (P < 0.001), and a treatment by location interaction (P < 0.01), two doses of PGF on Day 5 tended to have higher pregnancy rates to timed AI compared to those that received one dose of PGF (P = 0.06). In conclusion, heifers given two doses of PGF at CIDR removal on Day 5, in a 5-d CIDR-CO-Synch protocol, tended to have a higher pregnancy rate than those that received only one dose of PGF.  相似文献

A prostaglandin F analogue caused luteolysis in normal cycling non-lactating mares, and lactating mares (treated after the foal estrus). Effective doses ranged from 1.0 to 4.0mg given as a single subcutaneous injection 8–10 days after ovulation. A dose of 0.5mg was ineffective, hence the dose-response relationship was steep, indicative of a quantal type of response. Mares usually returned to estrus within 2–4 days and ovulated by 7 days after treatment. Mares bred naturally or by artificial insemination at the induced estrus and ovulation were fertile. The compound was without side-effects, and hence should be of value in manipulating the estrous cycle of the mare.  相似文献

The present study describes the effect of a single dose of dexamethasone administered to mares at time of breeding. In an initial experiment, the authors investigated safety of treatment. In a second experiment the effect of treatment on the uterine environment, fetal development and pregnancy outcome was examined. In the final part of the study, mares susceptible to persistent mating induced endometritis were identified, by means of a risk factor score system and the effect of treatment evaluated. Results indicated that dexamethasone administered at breeding time did not negatively impact on mares medical and reproductive traits. A reduced inflammatory response was observed post-mating in treated versus control mares and mares with multiple risk factors for susceptibility to persistent mating induced endometritis showed improved pregnancy rates following treatment. The authors concluded that a single dose of dexamethasone administered at the time of breeding is safe and can be used to modulate the uterine inflammatory response to breeding in susceptible mares.  相似文献

It has recently been reported that large insemination volumes might affect fertility of mares. The results from these studies are confounded by other factors, however, such as inadequate number of spermatozoa in the inseminate. We conducted a study to test whether volume alone affects fertility when sufficient numbers of spermatozoa are present. Semen from one stallion was collected, extended at 50 x 10(6) spermatozoa/ml, and stored in a commercial semen cooling device for 18 to 30 h before insemination. Ten pony mares were assigned during estrus in random pairs to be bred every other day with either 30 or 120 ml of extended cooled semen. Pregnancy was diagnosed by ultrasonography per rectum on Days 11, 12 and 13 after ovulation. On Day 13, the mares were given a luteolytic dose (5 mg) of PGF(2alpha). On the subsequent cycle, the mares were given the alternate treatment. The pregnancy rates in the 30- and 120-ml insemination volume groups were 7 9 and 10 10 , respectively; this difference was not significant (P=0.2). Embryonic growth from Day 11 to Day 13 was not different (P>0.05) between groups. We conclude that insemination volumes as large as 120 ml have no adverse effect on fertility.  相似文献

Poor uterine perfusion has been proposed as a cause of infertility in mares. The objective of this study was to investigate the effect of isosorbide dinitrate (ISDN), a nitric oxide donor, on uterine and ovarian blood flow resistance during diestrus and early pregnancy in mares. Six Trotter mares, aged 7 to 14 years, were examined daily during the first 11 days of three diestrous periods, and five of those mares were also examined during the first 11 days of two pregnancies. Six mares randomly received a placebo, a low dose (30 mg, ISDN30), or a high dose of ISDN (60 mg, ISDN60) through three nonconsecutive cycles. The treatments were administered orally, every 12 hours from Day 1 to 11 of the cycle (Day 0 = ovulation). Five of the 6 mares received a placebo or 60 mg of ISDN orally every 12 hours from Day 1 to 11 of pregnancy. The mares were short cycled on Day 12 of each trial. Transrectal color Doppler was used to determine blood flow resistance semiquantitatively and expressed as pulsatility index. Mean pulsatility index of both uterine arteries combined and of the dominant (ipsilateral to the CL) ovarian artery was lower (treatment effects: P ≤ 0.01; time effects: P ≤ 0.002) in mares receiving 30 mg or 60 mg of ISDN compared with placebo-treated mares. Blood flow resistance in the dominant ovarian artery was lower in ISDN-treated pregnant mares than in placebo-treated pregnant and cycling mares (treatment effect: P = 0.04; time effect: P = 0.003). Isosorbide dinitrate increases uterine and ovarian perfusion in cycling mares and ovarian perfusion in early pregnant mares. Further studies are needed to investigate these effects in relation to fertility of the mare.  相似文献

Uterine clearance of radiocolloid was measured by scintigraphy in 5 reproductively normal mares and in 4 mares exhibiting a delay in uterine clearance (DUC) after administration of PGF, cloprostenol or fenprostalene. Scintigraphy studies were performed on the second or third day of estrus during 3 consecutive estrous cycles. Drugs, PGF (5 mg IM), cloprostenol (CLO; 2mg IM) or fenprostalene (FEN; 250 μg subcutaneously) were given in random order, with only 1 drug given each estrus. Treatment response curves were generated, and the effect of each drug on uterine clearance of radiocolloid was compared to the clearance of radiocolloid when no drug was given and between treatments. In reproductively normal mares, CLO and PGF caused a rapid clearance of radiocolloid within 60 min (P < 0.01), with <25% of the initial dose of radiocolloid (% IDR) remaining by 120 min. Mean percentage of IDR at 120 min when no drug was given was 39% ± 4. Response of reproductively normal mares to FEN varied, with 3 mares clearing > 85% and 2 mares clearing <35%. In mares exhibiting DUC, all 3 drugs (CLO, PGF and FEN) caused rapid clearance of radiocolloid from the uterus by 60 min (P < 0.0001). Mares cleared significantly more colloid after treatment with CLO at 60 and 120 min than after PGF (P < 0.001). In conclusion, CLO appears to be the best drug of the 3 tested for stimulating clearance of intrauterine fluid since variation in response was observed following treatment with PGF and FEN.  相似文献

Prostaglandins F were quantitated by radioimmunoassay in uterine venous plasma of anesthetized mares on day 7 of estrus, days 2, 6, 10, 14 or 18 of diestrus and days 10, 14 or 18 of pregnancy. The PGF concentration was greater (P less than .01) at day 14 of diestrus than at all other days studied. The concentrations at days 10 and 18 of diestrus and at days 10, 14 and 18 of pregnancy were greater (P less than .05) than at day 7 of estrus and days 2 and 6 of diestrus. PGF concentrations at days 10 and 14 were greater (P less than .01) for diestrous than for pregnant mares.  相似文献
