The common pistachio psylla, Agonoscena pistaciae Burckhardt & Lauterer (Hemiptera: Psyllidae), is known as the key pest of pistachio orchards in Iran. This pest passes the winter as adults. In this study, energy allocation changes in relation to ambient temperature were investigated in field-collected adults by measuring total body sugar, trehalose, glucose, sorbitol, myoinositol, glycogen, lipid, and protein contents. Glycogen content decreased with decrease in ambient temperature. The decrease in glycogen content was proportional to the increase in total body sugar, trehalose, myoinositol, and sorbitol contents. In January, with mean ambient temperature of 5.4°C, glycogen content was at the lowest level, whereas total body sugar, trehalose, glucose, and sorbitol were at the highest level. Total body sugar, trehalose, myoinositol, and sorbitol contents increased as temperature decreased from 22.7°C in October to 5.4°C in January. In conclusion, low molecular weight carbohydrates and polyols may play a role in winter survival and adaptation to cold of the common pistachio psylla by providing the required cryoprotection. Also, overwintering adults of the common pistachio psylla may store energy in the form of lipid for later utilization during the overwintering.  相似文献

Doubly labeled phytic acid of high specific activity was prepared by incubating whole wheat seeds with [32P]phosphoric acid and [3H]myoinositol for 48 h and purifying by anion-exchange chromatography on AG 1- X 8 resin. Both degradation and synthesis of phytic acid were inhibited by KF to a similar extent, yet the catabolic and anabolic pathways involved distinctly different enzyme systems, as no [3H, 32P]myoinositol tetra- or pentaphosphate could be detected.  相似文献

Regioselective mannosylations of a myoinositol acceptor diol are readily achieved by Lewis acid mediated iodinolysis of n-pentenyl ortho-esters. The procedure affords a psuedotrisaccharide to which the phosphoglyceryl and other lipid residues are added leading to the key biosynthetic intermediate of Mycobacterium species.  相似文献

The extract of the floral parts of Viguiera quinqueremis afforded, in addition to known compounds, six new sesquiterpene lactones as well as a new myoinositol derivative. All compounds were detected in glandular trichomes which were collected micromechanically from the anther appendages and were analyzed by HPLC. Structure identification was performed by 1H NMR measurements including LC NMR and LC MS experiments.  相似文献

We recently described a 125 kd membrane glycoprotein in Saccharomyces cerevisiae which is anchored in the lipid bilayer by an inositol-containing phospholipid. We now find that when S. cerevisiae cells are metabolically labeled with [3H]myoinositol, many glycoproteins become labeled more strongly than the 125 kd protein. Myoinositol is attached to these glycoproteins as part of a phospholipid moiety which resembles glycophospholipid anchors of other organisms. Labeling of proteins with [3H]myoinositol for short times and in secretion mutants blocked at various stages of the secretory pathway shows that these phospholipid moieties can be added to proteins in the endoplasmic reticulum and that these proteins are transported to the Golgi by the regular secretory pathway. sec53, a mutant which cannot produce GDP-mannose at 37 degrees C, does not incorporate myoinositol or palmitic acid into membrane glycoproteins at this temperature, suggesting that GDP-mannose is required for the biosynthesis of these phospholipid moieties. All other secretion and glycosylation mutants tested add phospholipid moieties to proteins normally.  相似文献

Dispersed mouse pancreas acinar cells were prepared in which phosphatidylinositol had been labeled with myo[2-3H]inositol. During incubation with 0.3 microM cholecystokinin octapeptide (CCK-8) for 15 min, there was a loss of [3H]phosphatidylinositol radioactivity (23%) and a 3-fold gain in trichloroacetic acid-soluble radioactivity. Replacement of NaCl by up to 58 mM LiCl did not significantly affect the amount of CCK-8-stimulated [3H]phosphatidylinositol breakdown or the gain in acid-soluble radioactivity. However, in normal medium, the product of phosphatidylinositol breakdown was almost all inositol, whereas in Li+-containing medium, the product was almost all inositol 1-phosphate. Similar results were obtained with acetylcholine which, in the presence of Li+, gave a dose-responsive increase in inositol 1-phosphate over the concentration range of 0.1 to 10 microM. No increased accumulation of [3H]inositol diphosphate or [3H]inositol triphosphate was detected in stimulated cells. Time courses in the presence of Li+ indicated that the formation of inositol 1-phosphate preceded the formation of inositol. Addition of up to 50 mM myoinositol to the incubation medium showed no diluting effect on the amount of [3H]inositol 1-phosphate found. The accumulation of inositol 1-phosphate is presumably due to the known ability of Li+ to inhibit myoinositol 1-phosphatase. The results provide clear evidence that stimulated phosphatidylinositol breakdown involves a phospholipase C type of phosphodiesterase activity. 1.25 mM Li+ gave half-maximal inositol 1-phosphate accumulation. This is close to the range of plasma Li+ levels which is used therapeutically in psychiatric disorders. In unstimulated cells, [3H]inositol 1-phosphate accumulation in the presence of Li+ corresponded to a breakdown rate for [3H]phosphatidylinositol of 2 to 3%/h.  相似文献

We report here the in vivo conversion of [3H]myoinositol to [3H]chiroinositol. After labeling intraperitoneally with [3H]myoinositol for 3 days to reach radioisotope equilibrium in urine, [3H]chiroinositol was isolated from tissues and purified after 6 N HCl hydrolysis by two sequential paper chromatographies and high performance liquid chromatography (HPLC). Percent conversion of [3H]myoinositol to [3H]chiroinositol was highest in urine (36%), liver (8.8%), muscle (8.8%), and blood (7.6%) with intestine, brain, kidney, spleen, and heart decreasing in percentage from 2.8 to 0.7%. Labeling of other inositol isomers including scyllo-, neo-, and epi-, and mucoinositol was minimal, approximately 0.06% of [3H]myoinositol. Glucose was unlabeled, but glucuronate, the product of myoinositol oxidation, was labeled up to 1.5% of the [3H] myoinositol. Acid hydrolysates of combined inositol-containing phospholipids contain significant labeled chiroinositol. [3H]Phosphatidylinositols and [3H]glycosylphosphatidylinositols were extracted from liver, muscle, and blood, isolated by thin layer chromatography, and inositols purified by HPLC after acid hydrolysis. Percent conversion of [3H]myoinositol to [3H] chiroinositol was highest in blood (60.4%) followed by muscle (7.7%) and liver (2.2%).  相似文献

Abstract— —The lipid composition of human, bovine and sheep pineal glands was determined. No characteristic species difference was found in lipid content and composition. The total lipid was 2·9–4·0 per cent of wet weight of which phospholipid comprised 58–71 per cent and cholesterol 13·9–15·8 per cent in the three species. The phospholipid composition was 45 per cent phosphatidyl choline, 22 per cent phosphatidyl ethanolamine, 14 per cent sphingomyelin, 9 per cent phosphatidyl serine, 8 per cent monophosphoinositide, and 2 per cent diphosphatidyl glycerol. The major fatty acids found were C14:0, C16:0, C18:0, C16:1, and C18:1. In contrast to other tissues, pineal sphingomyelin has a low C24:1 content. No significant amounts of polyphosphoinositides or gangliosides were detected. When its lipid composition is compared with that of a number of other tissues, pineal is found to be most similar to testes and unlike pituitary and brain.  相似文献

In a previous study, D-chiroinositol added to a meal (0.5 g/kg) resulted in significantly lower postprandial plasma glucose concentrations without an increase in insulin concentrations in obese insulin-resistant monkeys. The present report describes the effects of another isomer of inositol, myoinositol, on postprandial plasma glucose and insulin concentrations and on urine glucose concentrations in 6 similarly insulin-resistant monkeys. The three 5 day study periods included a control period (liquid diet ad libitum) and 2 experimental periods (liquid diet ad libitum with either 1.5 g/kg/day myoinositol or D-chiroinositol added). Twenty-four hour urine samples were collected during each 5 day period. On the sixth day of each period the monkeys were anesthetized 110 min after completing either the control meal (15 ml/kg) or the experimental meals (1.5 g/kg myoinositol or D-chiroinositol) and plasma samples were obtained at 120, 150,180, 210, 240, 270 and 300 min. The plasma glucose concentration was lower after the meal with myoinositol compared to the control meal at 120, 150 and 180 min (p's<0.05). The plasma insulin concentration was lower after the meal with myoinositol compared to the control meal at 150 and 180 min (p's<0.05). In addition, 24 hour urine glucose concentrations were lower during the myoinositol diet compared to the control diet (p<0.001). The plasma glucose concentration was lower after the meal with D-chiroinositol compared to the control meal at 150, 240, 270 and 300 min (p's≥0.05). In obese insulin-resistant monkeys, myoinositol added to the diet lowers urine glucose concentrations and both myoinositol and D-chiroinositol added to a meal lower postprandial plasma glucose concentrations without increasing postprandial insulin concentrations. Therefore, myoinositol, like D-chiroinositol, may be a useful agent for reducing meal-induced hyperglycemia without inducing hyperinsulinemia in insulin-resistant subjects.  相似文献

The fate of 14C-labeled 2,4,6-trinitrotoluene (TNT) in an activated-sludge system was investigated. No [14C]TNT could be detected in the contents of an aerated reactor after 3 to 5 days of incubation. No significant 14CO2 was formed, and the radioactivity was about equally divided between the floc and the supernatant. The radioactive carbon present in the microflora was mainly associated with the lipid and protein components, but the characteristic constituents of these compounds (e.g., fatty acids and amino acids) were not radioactive. The major part of the 14C present in the lipid and protein fractions was found in precipitates that formed in both fractions. The solubility properties and infrared spectra of these precipitates suggested that they are macromolecular structures of the polyamide type formed by the reaction of TNT biotransformation products with lipids, fatty acids, and protein constituents of the microbial flora. This hypothesis is further supported by the correspondence of the infrared spectrum of the lipid precipitate with that of a model compound synthesized from TNT transformation products and lipid precursors. The resistance of these macromolecules to further biodegradation was paralleled by the reported resistance to microbial attack of polyamides containing similar linkages.  相似文献

An improved method of fractionating the myoinositol esters of indoleacetic acid (IAA) from maize kernels by gas-liquid chromatography has been developed. Mass spectrometry was employed as an aid in identification of the esters. Maize kernels contain three groups of esters of IAA: (a) IAA myoinositols, (b) IAA myoinositol arabinosides, and (c) IAA myoinositol galactosides. Each group has three chromatographically distinguishable isomers. The glycosylinositols described are unique in that carbon 1 of the sugar is attached to the hydroxyl at C-5 of the myoinositol.  相似文献

Amphotericin B (AmB) autoxidation resulted in oxygen consumption, superoxide anion formation and production of thiobarbituric acid (TBA)-reactive material (malondialdehyde). Malondialdehyde formation increased after incubation of the drug with ascorbate-ADP-FeCl3. Growth of Trypanosoma cruzi epimastigotes in the presence of AmB induced a decrease in the free fatty acid content of the cells (57% in control cells vs. 7% in AmB-treated cells), and in the proportion of unsaturated fatty acids as well as cell killing. No changes were detected on sterol content. No evidence was found for lipid peroxidation as a mechanism of cell injury by this antibiotic.  相似文献

The effects of diabetes and insulin administration on certain aspects of phosphoinositide metabolism in R3230AC mammary tumors were studied in vivo. Three weeks after diabetes was induced by streptozotocin, [3H]myoinositol incorporation into PI, PIP and PIP2 was increased in R3230AC tumors, whereas the formation of [3H]IP, [3H]IP2 and [3H]IP3 was decreased. Administration of protamine zinc insulin (3IU, twice daily, for 3 days) to diabetic rats decreased [3H]myoinositol incorporation into phosphoinositides and inositol phosphates in these mammary tumors. The R3230AC tumor from insulin-treated diabetic hosts had lower levels of unmetabolized [3H]-myoinositol compared to tumors from diabetic animals. Enzymatically-dissociated tumor cells from insulin-treated animals displayed decreased myoinositol transport in vitro. These findings suggest that the insulin-induced decrease in the turnover of inositol lipids in vivo in R3230AC mammary tumors could have resulted from the decreased level of [3H]myoinositol in these cells.  相似文献

Myoinositol and its phosphorylated derivatives have been quantitatively determined in normal and Rous-sarcoma-virus-transformed quail cells under various growth conditions using [2-(3)H]myoinositol at isotope equilibrium conditions. The following amounts were determined (nmol/mumol phospholipid, as a unit of cell mass): exponentially growing normal and tumor cells contained 25--40 nmol free inositol, 0.40--0.45 nmol myoinositol 1-phosphate, 0.30--0.50 nmol glycerylphosphoinositol, and 0.03--0.04 nmol myoinositol cyclic 1 : 2-phosphate. At high cell populations in the absence of serum, conditions which result in cessation of growth by normal but not by tumor cells, changed levels were found for glycerylphosphoinositol and free inositol. In tumor cells the levels of these two compounds increased to 0.64 nmol and 64 nmol, respectively. In normal cells glycerylphosphoinositol increased to 0.95 nmol and free inositol showed highly elevated levels of 144 nmol. At short pulses the specific activities of inositol 1-phosphate and inositol cyclic 1 : 2-phosphate were found to be higher than that of phosphatidylinositol. Precise structural identification of photosynthetic polar glycerolipids in microalga Tetraselmis chuii has been established using Ultra Performance Liquid Chromatography-Electrospray ionization-Quadrupole-Time of Flight Mass Spectrometry (UPLC-ESI-Q-TOF-MS) by direct analysis of the total lipids extract. The mass spectrometry was performed in reflective time-of-flight using electron spraying ionization in both positive and negative modes. The structural determination was based on the characteristic product ions yielded by different glycerolipids under ESI-MS/MS mode, and confirmed the molecular species by the carboxylate anions produced by glycerolipids in the negative mode. As a result, more than 40 lipid molecular species, including 11 monogalactosyldiacylglycerols (MGDG), 7 digalactosyldiacylglycerols (DGDG), 16 sulfoquinovosyldiacylglycerols (SQDG), and 9 phosphatidylglycerols (PG), were detected in Tetraselmis chuii, which had never been identified before in this microalga. Furthermore, some intact lipid molecules with hydroxylated fatty acids that could not be detected by the traditional GC-MS method were found this time, providing novel information for the photosynthetic lipidome of Tetraselmis chuii. Comparative studies on fatty acids at the sn-2 position showed that SQDG and MGDG are dominantly biosynthesized through the prokaryotic pathway, PG is a typically mixed biosynthetic pathway, while DGDG is somewhat peculiar with C14:0 and C16:0 at its sn-2 position. This method could provide a full structural profile of intact photosynthetic lipid molecular species, which may be applied to study the physiological and ecological functions of lipid by monitoring their individual changes. Divalent cation-induced fusion of large unilamellar vesicles (approx. 0.1 micron diameter) made of phosphatidylserine (PS) or phosphatidylglycerol (PG) has been studied. Intermixing of aqueous contents during fusion was followed by the Tb/dipicolinic acid fluorescence assay, and intermixing of membrane components by resonance energy transfer between fluorescent lipid probes. Both assays gave identical threshold concentrations for Ca2+, which were 2 mM for PS and 15 mM for PG. The dependencies of the initial rate of fusion on the concentration of PG vesicles determined by either assay were identical, the order of this dependence being 1.2 in the concentration range of 5-200 microM lipid. For PS liposomes, this order was found to be 1.5 in the fluorescent lipid assay. No leakage of contents was detected during the fusion of PG vesicles. Mg2+ inhibited the Ca2+-induced fusion of PS vesicles, but did not cause any fusion by itself, consistent with previous results with the Tb/dipicolinic acid assay.  相似文献

The size increase of small unilamellar vesicles composed of binary mixtures either of saturated fatty acid phosphatidylcholines with different chain lengths or of saturated and unsaturated phosphatidylcholines was found to depend on the miscibility properties of the lipid components. No size increase was detected in vesicles formed by two miscible phosphatidylcholines. In vesicles composed of two lipids which are partially immiscible in the gel state, a size increase was observed at temperatures which mainly overlapped the range of temperatures of the lipid phase transition. The rate of size increase of vesicles composed of two lipids which are immiscible in the gel state was faster than that of vesicles composed of two partially immiscible phosphatidylcholines, and the process occurred not only at the temperature ranges of the lipid phase transition, but also when both lipids were in the gel state. The vesicle size increase process occurred without the mixing of the internal content of the vesicles. A model is proposed in which the presence of 'fractures' between membrane regions of different fluidity and/or lipid composition controls the rate of this process.  相似文献

1. The specific activities of erythrocyte membrane-bound acetylcholinesterase (EC 3.1.1.7.) and soluble hexokinase (EC 2.7.1.1.) in vitamin E deficient and vitamin E sufficient rabbits were investigated. 2. Acetylcholinesterase specific activities values of 43.4 in deficient and 57.4 in sufficient vitamin E rabbits were obtained. Hexokinase specific activity was not modified, and values of 3.31 in deficient and 3.6 in controls were found. 3. No peroxidation process was detected by us on vitamin E deficient diets. 4. These observations would suggest that the membrane stabilizing effect of vitamin E may be accomplished by a mode of action not necessarily related to its ability to prevent lipid peroxidation.  相似文献

Short-chained sugar compounds, thought to be involved in the synthesis of callose, were formed in small amounts from UDP-glucose by soluble extracts from hypocotyls of seedlings of Phaseolus aureus. The properties of the glycosides were investigated by treatment with various chemicals and analysis by paper chromatography, g.l.c. and mass spectrometry. The data obtained support the characterization of these compounds as myoinositol-beta-glucoside and diglucosylmyoinositol. The cyclitol moiety was provided by the enzyme extract. Free myoinositol was not the immediate substrate but a compound containing myoinositol, isolated from the enzyme extract, may be involved. The method of synthesis of these glucosides is compared with that of other cyclitol glycosides.  相似文献
