The polyene antibiotic filipin inhibits the activities of both photosystem I and photosystem II in maize mesophyll chloroplasts and pea chloroplasts. Maximum inhibition of photosystem II activity was observed at a filipin concentration of about 0.4 mm in maize mesophyll chloroplasts and 1.0 mm in pea chloroplasts. Inhibition of photosystem II activity was temperature dependent, being much less if the antibiotic and chloroplasts were incubated at 0 °C compared to 25 °C. The inhibition of photosystem I activity of both maize mesophyll and pea chloroplasts caused by filipin, could be overcome by the addition of the soluble electron transfer protein, plastocyanin. It is concluded that the inhibition of photochemical activity caused by filipin is a secondary effect resulting from a change in membrane conformation induced by the antibiotic.  相似文献

The pH dependence of the photoreduction of ferricyanide and the photoreduction of NADP from water and photosystem I activity have been compared in isolated chloroplasts from mesophyll and bundle sheath cells of Zea mays. The maximum activity of photoreduction of ferricyanide occurs at pH 8.5 in isolated mesophyll chloroplasts. The addition of methylamine does not cause a marked shift in the pH maximum, but brief sonication lowers the pH maximum to 7.0. In contrast, isolated bundle sheath chloroplasts have a pH maximum at 7.0 and the shape of the pH versus activity curve is similar to that of sonicated mesophyll chloroplasts. When photoreduction of ferricyanide by the isolated chloroplasts is measured at their pH maxima, the values for bundle sheath chloroplasts are about half those of methylamine-treated mesophyll chloroplasts on a chlorophyll basis.  相似文献

Linolenic acid is an inhibitor of electron transport in chloroplasts of higher plants. It has obvious effects on the structure and function of chloroplasts. In the present paper, we investigated the nano-anatase relieving the inhibition of photoreduction activity and oxygen evolution caused by linolenic acid in spinach chloroplasts. The results showed that linolenic acid in various concentrations could obviously reduce the whole chain electron transport and the photoreduction activity of two photosystems, especially on the oxidative reside and reduce reside of photosystem II (PS II). After adding nano-anatase to chloroplasts treated by linolenic acid, the whole chain electron transport rate, the photoreduction activity of two photosystems, and the oxygen evolution rate were increased significantly, indicating that nano-anatase could obviously decrease the inhibition of linolenic acid on the electron transport, photoreduction activity, and oxygen evolution of spinach chloroplasts.  相似文献

Spinach chloroplasts were immobilized with vinyl monomers by radiation-induced polymerization at low temperature and stored in buffer containing bovine serum albumin. The lifetime of O2 evolution activity in photosystem II was prolonged remarkably in immobilized chloroplasts. Thermostability of immobilized chloroplasts stored in buffer containing bovine serum albumin was far better than that of immobilized chloroplasts in pure buffer and that of intact chloroplasts. When immobilized chloroplasts were stored in buffer including polyethylene glycol, the lifetime of O2 evolution activity was longer than for those stored in buffer containing bovine serum albumin.  相似文献

The response of the chloroplastic antioxidant system of the cultivated tomato Lycopersicon esculentum (Lem) and its wild salt-tolerant related species L. pennellii (Lpa) to NaCl stress was studied. An increase in H2O2 level and membrane lipid peroxidation was observed in chloroplasts of salt-stressed Lem. In contrast, a decrease in these indicators of oxidative stress characterized chloroplasts of salt-stressed Lpa plants. This differential response of Lem and Lpa to salinity, correlates with the activities of the antioxidative enzymes in their chloroplasts. Increased activities of total superoxide dismutase (SOD), ascorbate peroxidase (APX), monodehydroascorbate reductase (MDHAR), glutathione-S-transferase (GST), phospholipid hydroperoxide glutathione peroxidase (PHGPX) and several isoforms of non-specific peroxidases (POD) were found in chloroplasts of salt-treated Lpa plants. In these chloroplasts, in contrast, activity of lipoxygenase (LOX) decreased while in those of salt-stressed Lem it increased. Although total SOD activity slightly increased in chloroplasts of salt-treated Lem plants, differentiation between SOD types revealed that only stromal Cu/ZnSOD activity increased. In contrast, in chloroplasts of salt-treated Lpa plants FeSOD activity increased while Cu/ZnSOD activity remained unchanged. These data indicate that salt-dependent oxidative stress and damage, suffered by Lem chloroplasts, was effectively alleviated in Lpa chloroplasts by the selective up-regulation of a set of antioxidative enzymes. Further support for the above idea was supplied by leaf discs experiments in which pre-exposure of Lpa plants to salt-treatment conferred cross-tolerance to paraquat-induced oxidative stress while increased oxidative damage by paraquat-treatment was found in salt-stressed Lem plants.  相似文献

Galactolipase activity in chloroplasts of several chill-resistant plants was found to be very low [0.02-0.13 mumol free fatty acid (FFA) liberated min-1 mg protein-1] or not detected. The same phenomenon was observed for soybean and members of the Cucurbitaceae such as cucumber, pumpkin, melon and squash. Since, following cold storage of cucumber leaves, the levels of monogalactosyl-diacylglycerol and digalactosyl-diacylglycerol in chloroplasts decrease while those of FFA accumulate it seems likely that in these typical chill-sensitive plants galactolipase is present but inactivated during isolation procedure. The low galactolipase activity in chloroplasts was accompanied by a relatively low FFA content ranging from 0.05 mumol to 0.30 mumol FFA mg chlorophyll (Chl)-1. However, both pea and horse bean chloroplasts (with low galactolipase activity) exhibit about 0.45 mumol FFA mg Chl-1. Elevated galactolipase activity was observed in chloroplasts of most chill-sensitive species (ranging from 0.31 mumol to 1.32 mumol FFA liberated min-1 mg protein-1) as well as in chloroplasts from broad bean, a member of a chill-resistant species (1.26 mumol FFA liberated min-1 mg protein-1). In addition in this latter group of plants FFA level in chloroplasts often did not fit the galactolipase activity. The results suggest that there exists a tendency for chilling tolerance of plants to decrease both galactolipase activity and FFA level. However, in some plant species with elevated galactolipase activity the chloroplast FFA level does not correlate with enzyme activity.  相似文献

Oxygen-evolution activity of spinach chloroplasts was investigatedby washing chloroplasts with 0.8M Tris buffer containing 20%acetone. This inactivitation was easily removed by two successivetreatments, dark- and light-reactivations. The first treatmentwas dark-reactivation step, rewashing inactivated chloroplastswith reduced DPIP (DPIP treatment). The second one was a light-reactivatedchloroplasts with incubating chloroplasts with Mn2+, Ca2+, dithiothreitoland bovine serum albumin under ilumination. Both light- and dark-reactivation treatments were required toregain oxygen-evolution activity of Tris-acetone-washed chloroplasts,which is characteristic of such chloroplasts. However, in Tris-washedchloroplasts considerable activity was recovered by dark-reactivationalone. Manganese and calcium contents of Tris-acetone-washed chloroplastswere compared with those of chloroplasts obtained by other preparations. Tris-acetone washing was presumed to inhibit the oxygen-evolutionsite of Photo-systetm II by affecting Mn, Ca and other substancesin chloroplasts. The inhibition site was estimated from a changein fluorescence yield of chlorophyll and the effect of artificialelectron donor specific for Photosystem II on NADP photoreductionactivity. Phosphoglucomutase (PGM) activity was measured in spinach (Spinacia oleracea L.) chloroplasts. Initial enzyme activity in a chloroplast lysate was 5 to 10% of total activity measured with 1 micromolar glucose 1,6-bisphosphate (Glc 1,6-P2) in the assay. Initial PGM activity increased 2- to 3-fold when chloroplasts were illuminated for 10 minutes prior to enzyme measurement and then decreased slowly in the dark. Measurements of total enzyme activity were unchanged by prior light treatment. Initial PGM activity from light treated chloroplasts was sufficient to account for in vivo rates of starch synthesis. Changes in PGM activity were affected by stromal pH and orthophosphate concentration. Photosynthetic inhibitors, dl-glyceraldehyde, glycolaldehyde, and glyoxylate, decreased and 3-phosphoglyceric acid increased light induced changes of PGM activity. Dark preincubation of chloroplasts with 10 millimolar dithiothreitol had no effect upon initial PGM activity, suggesting that light effects did not involve a sulfhydryl mechanism. Hexose monophosphate levels increased in illuminated chloroplasts. Activation of PGM in a chloroplast lysate by Glc 1,6-P2 was maximal between pH 7.5 and 8.5. Stromal concentrations of Glc 1,6-P2 were between 20 and 30 micromolar for both light and dark incubated chloroplasts and these levels should saturate PGM activity. Light dependent alterations of enzyme activity may be due to changes of phosphorylated PGM levels in the stroma or are the result of changes in residual activity by the dephosphorylated form of the enzyme. The above results indicate that PGM activity in spinach chloroplasts may be regulated by light, stromal pH, and Glc 1,6-P2 concentration.  相似文献

Photosynthetic electron transport activity has been measured in chloroplasts isolated from dark-grown seedlings of Pinus silvestris L. and in chloroplasts isolated from seedlings subjected to illumination for periods of up to 48 h. Activities of photosystem 2, photosystem 1 and photosystem 2 plus 1 have been measured. Chloroplasts isolated from dark-grown seedlings showed significant electron transport activity through both photosystems and through the entire electron transport chain from water to NADP. Illumination of the seedlings for only 5 min markedly promoted photosystem 2 activity. The artificial electron donor, diphenylcarbazide. promoted activity in chloroplasts from dark-grown seedlings and in chloroplasts from seedlings illuminated for up to 30 min. In comparison to photosystem 2 and overall electron transport from water to NADP, photosystem 1 activity increased only slightly during illumination. Measurements of electron transport and fluorescence kinetics have confirmed that photosynthetic electron transport capacity is limited on the water splitting side of photosystem 2 in dark-grown seedlings, whereas the primary and secondary electron acceptors of photosystem 2 are fully synthesized and functioning in darkness. Polyethylene glycol must be used as a protective agent when isolating photoactive chloroplasts from secondary needles of conifers. However, the presence of polyethylene glycol, when isolating chloroplasts from dark-grown pine cotyledons, caused a total inhibition of the activity of photosystem 2. The failure of others to show a substantial electron transport activity in chloroplasts from dark-grown Pinus silvestris might depend on their use of polyethylene glycol in the preparation medium and/or on their use of suboptimal reaction conditions for the electron transport measurements.  相似文献

In order to establish differences in the chilling sensitivity of domestic and wild Lycopersicon species, galactolipase (EC 3.1.1.26) activity, free fatty acid (FFA) level and Hill reaction activity were measured in chloroplasts isolated from control and cold treated leaves of L. esculentum Mill., cv. Norton, L. hirsutum Humb. and Bonpl., L. peruvianum var. glandulosum Mill. Galactolipase activity was higher in chloroplasts from Lycopersicon species with high chilling sensitivity than in chloroplasts of more chilling-resistant ones. A similar relationship was observed for FFA level in chloroplasts from both cold-stored and control leaves. Decrease in Hill reaction activity due to cold stress was greater in chloroplasts of more chilling-sensitive species. The changes are accompanied by a decline of photochemical activity. Considering the changes in the three parameters noted above, an increasing order of chilling tolerance was established: L. esculentum < L. hirsutum (700 m) < L. hirsutum (3100 m) < L. peruvianum (3400 m). It is suggested that measurements of galactolipase activity and FFA may be useful in an evaluation of differences in resistance to chilling injury of closely related species.  相似文献

Intact mesophyll and bundle sheath chloroplasts wee isolated from the NADP-malic enzyme type C4 plants maize, sorghum (monocots), and Flaveria trinervia (dicot) using enzymic digestion and mechanical isolation techniques. Bundle sheath chloroplasts of this C4 subgroup tend to be agranal and were previously reported to be deficient in photosystem II activity. However, following injection of intact bundle sheath chloroplasts into hypotonic medium, thylakoids had high Hill reaction activity, similar to that of mesophyll chloroplasts with the Hill oxidants dichlorophenolindophenol, p-benzoquinone, and ferricyanide (approximately 200 to 300 micromoles O2 evolved per mg chlorophyll per hour). In comparison to that of mesophyll chloroplasts, the Hill reaction activity of bundle sheath chloroplasts of maize and sorghum was labile and lost activity during assay. Bundle sheath chloroplasts of maize also exhibited some capacity for 3-phosphoglycerate dependent O2 evolution (29 to 58 micromoles O2 evolved per milligram chlorophyll per hour). Both the mesophyll and bundle sheath chloroplasts were equally effective in light dependent scavenging of hydrogen peroxide. The results suggest that both chloroplast types have noncyclic electron transport and the enzymology to reduce hydrogen peroxide to water. The activities of ascorbate peroxidase from these chloroplast types was consistent with their capacity to scavenge hydrogen peroxide.  相似文献

Etioplasts capable of incorporating 14C-leucine into protein have been isolated from dark-grown pea and wheat plants. The requirements for leucine incorporation for etioplasts were similar to those for chloroplasts. An ATP-generating system, Mg2+, and GTP were required. The amino-acid-incorporation activity of etioplasts from wheat was comparable to that of chloroplasts on an RNA basis, whereas the activity of pea etioplasts was about 50% of the activity of pea chloroplasts. The incorporation of leucine into protein by etioplasts and chloroplasts from pea and wheat was inhibited by chloramphenicol, and to a slight extent by cycloheximide.  相似文献

Chloroplasts of chinese mustard (Brassica campestris L.) were immobilized in polyacrylamide gel. A 8% polymer concentration was suitable for the immobilization. The activity of the carbon dioxide fixation of immobilized chloroplasts was 65% of that of free chloroplasts. The optimum conditions for the carbon dioxide fixation of immobilized chloroplasts were similar to that of native chloroplasts. However, immobilized chloroplasts were more stable under alkaline conditions and high temperatures than native chloroplasts. Light penetration of the gel was not a limiting parameter of the carbon dioxide fixation. The lifetime of immobilized chloroplasts was three times longer than that of free chloroplasts. 3-Phosphoglyceraldehyde and other compounds were produced continuously by immobilized chloroplasts.  相似文献

Chloroplasts isolated from spinach leaves by the mechanical method were intact and exhibited high rates of CO2-dependent oxygen evolution whereas chloroplasts isolated from sunflower leaves by the same technique were also intact but showed only low rates of oxygen evolution. The rate of uptake of orthophosphate (Pi) from the suspending medium with sunflower chloroplasts was less than 20% of that in spinach chloroplasts. The apparent Km for Pi transport was lower in sunflower chloroplasts but uptake was competitively inhibited by 3-phosphoglycerate in chloroplasts from both species. Uptake of malate (via the dicarboxylate transporter) and of ATP (via the adenine nucleotide transporter) was also reduced in sunflower chloroplasts compared to spinach chloroplasts. The endogenous Pi content and total exchangeable phosphate pool of sunflower chloroplasts were less than half that in spinach chloroplasts.Addition of a number of possible protective agents to the grinding medium failed to prevent the loss of photosynthetic activity during mechanical isolation of sunflower chloroplasts. Grinding mixtures of spinach and sunflower leaves together indicated that spinach chloroplasts were not inhibited by the sunflower leaf extract. Chloroplasts isolated from sunflower leaves via protoplasts had high rates of CO2-dependent oxygen evolution. The Vmax and Km for Pi uptake, endogenous Pi content and total exchangeable phosphate pool of chloroplasts isolated from sunflower protoplasts were all similar to spinach chloroplasts. It is concluded that inner envelope membrane proteins are damaged during mechanical isolation of sunflower chloroplasts. The decrease in activity of the phosphate transporter and loss of endogenous phosphate may contribute to the low rates of photosynthesis observed in chloroplasts isolated by the mechanical method from leaves of sunflower and possibly other species.Abbreviations PGA 3-phosphoglyceric acid  相似文献

The intracellular localization and some properties of monophenol monooxygenase (MPMO) from fresh tea leaves have been studied. It has been demonstrated that MPMO activity is located in cytosole and chloroplasts. These two forms have different properties. Molecular weights of cytosole and chloroplasts MPMO are 41 and 28 kD respectively. The chloroplasts and cytosole forms of MPMO reveal maximum activity at pH 5.3 and 7.1 respectively.  相似文献

1. Quercetin, a flavonoid which acts as an energy transfer inhibitor in photophosphorylation is shown to inhibit the P-ATP exchange activity of membrane-bound CF1 and the ATPase activity of isolated CF1. Quercetin, affects also the proton uptake in chloroplasts in a manner similar to that of dicyclohexylcarbodiimide. 2. The light-dependent proton uptake in EDTA-treated chloroplasts is stimulated by quercetin. In untreated chloroplasts quercetin has a dual effect: it enhances at pH above 7.5 while at lower pH values it decreases the extent of H+ uptake. Similar effects were obtained with dicyclohexylcarbodiimide. 3. Like quercetin, dicyclohexylcarbodiimide was also found to inhibit the ATPase activity of isolated CF1. 4. Quercetin inhibits uncoupled electron transport induced by either EDTA-treatment of chloroplasts or by addition of uncouplers. Quercetin restores H+ uptake in both types of uncoupled chloroplasts. 5. The mode of action of quercetin and dicyclohexylcarbodiimide in photophosphorylation is discussed, and interaction with both CF1 and F0 is suggested.  相似文献

Effect of 4-thiouridine, which was proved to inhibit selectively and “light-reversibly” the synthesis of chloroplast ribosomal RNAs in radish cotyledons, on the photo-induced development of photosystem I, II and a complete electron transport chain was investigated with plastids obtained from 4-thiouridine treated dark-grown radish cotyledons after various times of development in the light. It was demonstrated that the 4-thioridine treated chloroplasts showed a higher activity of photoreduction than the control untreated chloroplasts in every system on a chlorophyll basis during the development after 24 hr illumination. This specific activity decreased in both chloroplasts, as the chloroplasts matured with the time of illumination. The activity per g of fresh cotyledons treated with 4-thiouridine, especially in the early stage of development, was lower than that of ones untreated with the drug because total chlorophyll content was poor, but the activity of the former was enhanced with the increase of total chlorophyll content upon illumination while the activity of the latter decreased on 24 hr illumination. Moreover, Hill reaction measurements showed that 4-thiouridine treated chloroplasts were saturated at lower light intensity than untreated ones inspite of the same content of chlorophyll in both the chloroplasts: photoreduction of NADP+ was saturated at 3000 lux for the former and at 5000 lux for the latter. Based upon these results, specific development of the chloroplast is discussed.  相似文献

Chloroplasts were prepared from pea seedlings and tested for NAD kinase activity. More than half of a Ca2+, calmodulin-dependent activity and most of a Ca2+, calmodulin-independent activity of the homogenate were associated with chloroplasts. The Ca2+, calmodulin-dependent activity could be detected by adding Ca2+ and calmodulin to the incubation medium containing intact chloroplasts. This activity could not be separated from the chloroplasts by successive washes or by phase partition in aqueous two-polymer phase systems. After chloroplasts fractionation, the Ca2+, calmodulin-dependent NAD kinase activity was localized at the envelope, and the Ca2+, calmodulin-independent activity was recovered from the stroma. In view of these results and of a previous report [Simon (1982) Plant Cell Rep. 1, 119–122] the occurrence and presumed role of calmodulin in the chloroplast are discussed.  相似文献

Isolated chloroplasts of barley seedlings ( Hordeum vulgare L.) when kept in light, released a soluble, thermostable factor that inhibited phenylalanine ammonia-lyase (PAL) in vitro . Highest inhibition was found when chloroplasts were suspended in PAL-containing extract and kept in light. Efficiency of PAL activity inhibition did not depend greatly on the type of medium used for chloroplast isolation, nor on the composition of buffer in which the enzymatic activity was measured. It is proposed that in green tissues chloroplasts may participate in regulation of cytoplasmic PAL activity.  相似文献

The sites of action of several compounds, reported to inhibit CO(2) fixation by chloroplast preparations were located by developing assays in lettuce chloroplasts to test their effect on partial reactions of the carbon cycle and on carbonic anhydrase. The results indicated that: d, l-glyceral-dehyde and 5'-AMP inhibit phosphoribulose kinase or isomerase. 3-Phosphoglyceric acid and 6-phosphogluconate inhibit ribulose diphosphate carboxylase. Azide, Mg(2+), and nitrite inhibit the activity of carbonic anhydrase of lettuce chloroplasts and light-dependent CO(2) fixation by intact chloroplasts with similar sensitivities. None of these inhibited CO(2) fixation in ruptured chloroplasts. It is suggested that the inhibition by azide, nitrite, and magnesium ions of CO(2) fixation by intact chloroplasts is due to their inhibition of the activity of carbonic anhydrase.  相似文献
