3H-nicotine binding was performed on intact and solubilized rat brain membranes as well as membranes from the electric organ of the Torpedo fish. The Kd for binding to intact and solubilized rat brain membranes was 5.6 × 10?9 M and 1.1 × 10?8M respectively, and the binding capacity 2.0 × 10?14 and 3.0 × 10?13 moles /mg protein respectively. The Kd for Torpedo membranes was 3.1 × 10?7M and the binding capacity 6.8 × 10?13 moles/mg protein. The binding was stereospecific with the affinity of the (?)-nicotine being about 8 times greater than the (+)-nicotine with all three preparations. The relative affinity for the nicotine binding site of nicotinic cholinergic drugs was considerably less in rat brain than in Torpedo membranes, where the sites are mainly cholinergic. A comparison was made of the ability of a variety of cholinergic drugs and nicotine derivatives to compete with 3H-nicotine binding and their relative pharmacologic potency to produce or inhibit a characteristic prostration syndrome caused by (?)-nicotine administered intraventricularly to rats. From such studies it was concluded that nicotine, in part, may be interacting at noncholinergic sites in rat brain.  相似文献

2-Deoxy-2,3-dehydro-N-acetylneuraminic acid and its methyl ester are competitive inhibitors of Arthrobacter sialophilus neuraminidase with Ki = 1.4 × 10?6M and 4.8 × 10?5M, respectively. The Km for the substrate, N-acetylneuraminlactose, is 1.0 × 10?3M. These data, taken together with the conformation of these compounds, indicate that these compounds are transition-state analogs of the enzyme. These results also suggest that the substrate upon binding to neuraminidase is distorted to a conformation approaching that of a half-chair.  相似文献

Binding of 125I-labelled ovine prolactin to female rat liver membranes underequilibrium conditions showed an apparent Kd of 200 pM, and a Hill coefficient of 1.0. The association rate was second order, with a rate constant K1, of 2.1 × 107, 1.4 × 107, 1.2 × 107 and 4 × 106 M?1. min?1 at 37, 30, 24 and 4° respectively. At 24° there were two components to the dissociation; a faster phase with K?1=1.26 × 10?2. min?1 (T12=55 minutes) and a slower phase with K?1=1.103 × 10?3. min?1. The apparent Kd (from K?1K1) was 1.05 nM for the faster phase and 87.5 pM for the slower phase. These data suggest that there is a conformational change following hormone binding which results in an increased receptor affinity, which effectively prevents release of bound hormone.  相似文献

Membranes from homogenates of corpus striatum bound 3H-dihydromorphine in a saturable fashion with a Km value of 1 × 10?9M. The binding of 3H-dihydromorphine to the membranes was reduced to about 10% by 10?7M levorphanol but not by 10?7M dextrorphan. The binding of 3H-dihydromorphine became less sensitive to 10?7M levorphanol when the concentration of 3H-dihydromorphine was greater than 2 × 10?9M. Other opiate narcotics, e.g. morphine and l-methadone, were as effective as levorphanol in competition for the binding 3H-dihydromorphine with ED50 values of 2–4 × 10?9M. d-Methadone and dextrorphan were about 1/50 and 1/2000 as effective as their respective levo-isomers. The opiate antagonist, naloxone, also competed effectively for the binding sites with an ED50 value of 3.3 × 10?9M. Substances like acetylcholine, choline, serotonin, norepinephrine and dopamine were ineffective. Only ionophores specific for divalent cations stimulated the binding of 3H-dihydromorphine suggesting that some endogenous divalent cations may be inhibitory to the binding of the opiate narcotic. The receptors of 3H-dihydromorphine probably exist in the membranes of nerve endings and have a density of 6 × 1012 sites per g in corpus striatum. We conclude that the described technique can successfully detect the opiate narcotic receptors in the central nervous system without the usual method of displacement.  相似文献

4′-Iodo-, 4′-bromo-, 4′-chloro- and 4′-fluoro-2,3,4,5-tetrachlorobiphenyl were administered to immature male Wistar rats and the effects of this homologous series of 4′-halo-2,3,4,5-tetrachlorobiphenyls on the microsomal drug-metabolizing enzymes were determined. All the halogenated biphenyls increased microsomal benzo[a]pyrene hydroxylase (or aryl hydrocarbon hydroxylase, AHH), ethoxyresorufin (ER) O-deethylase and dimethylaminoantipyrine (DMAP) N-demethylase. The effects of the 4′-halo-2,3,4,5-tetrachlorobiphenyls on the microsomal enzyme activities and on the relative peak intensities and spectral shifts of the reduced cytochrome P-450:CO and ethylisocyanide (EIC) binding difference spectra were similar to those observed after coadministration of phenobarbitone (PB) and 3-methylcholanthrene (MC). The relative activities of the halogenated biphenyls were determined using two invitro assays; namely cytochrome P-448 associated induction in rat hepatoma H-4-II E cells in culture and competitive binding to the hepatic cytosolic Ah receptor protein from male Wistar rats. Dose-response experiments for the iodo, bromo, chloro and fluoro analogs gave EC50(M) values of 8.5×10?9, 6.6×10?8, 5.7×10?7, and 3.3×10?5, and 1.5×10?6, 2.5×10?6, 4.1×10?6 and 2.5×10?5 for the ER O-deethylase induction and receptor binding assays respectively. The relative potencies of the 4′-halo-2,3,4,5-tetrachlorobiphenyls followed the order I>Br>Cl>F for both assays and differences in the EC50 values for the iodo and fluoro analogs were greater than three orders of magnitude for ER O-deethylase induction in rat hepatoma cells in culture. One possible explanation for these effects may be associated with differences in the polarizability of the laterally substituted halogen groups. However, other differences in the physico-chemical properties of the halogen atoms may also be important.  相似文献

Crystals of alkaline phosphatase (EC 3.1.3.1; Mr 94,000) grown at pH 9.5 from 2.25 m-(NH4)2SO4 with 5 × 10?5 m-Zn and 10?2 m-Mg present were analyzed by X-ray diffraction at pH 7.5 in 2.66 m-(NH4)2SO4 with 10?2 m-Zn and 10?2 m-Mg present. The crystals are orthorhombic with a = 195.5 A?, b = 168.3 A?and c = 76.33 A?, and the space group is I222. X-ray phases were determined by the multiple isomorphous replacement and anomalous dispersion method using K2PtCl4, KAu(CN)2 and K2OsO4 derivatives. The electron density maps and analysis of metal binding sites reveal one molecule per asymmetric unit with an internal, non-crystallographic, 2-fold rotation axis relating the subunits. Each subunit contains a major αβ domain with a seven-stranded β-sheet flanked by helices. The sheets are roughly coplanar but the general direction of the strands in each is at 20 ° to the rotation axis and thus 40 ° from each other. The helical content of the αβ domain is approximately 27% of the 459 residues in the monomer and the β content is approximately 7%. The chains in a smaller domain are more convoluted and less easily characterized than in the αβ domain. In both there is extensive monomer-monomer contact.Removal of the zinc and magnesium from the parent crystal produces a stable apoenzyme crystal and addition of cobalt at 10?2 m or cadmium at 10?2 or 5 × 10?2 m reveals seven metal binding sites per dimer. The active centers are 32 Å apart and each is shown by anomalous dispersion data to contain two metal binding sites, A and B. The cadmium derivative refinement determined the A-B separation to be 4.9 Å. Comparison of the parent and apo structures by means of difference maps reveals the double metal site with Zn at A and probably Mg at B. A prominent, partially resolved peak centered 7 Å away is interpreted as a stabilization of the backbone in this position by the metal ion co-ordination to a side-chain. Several negative peaks within 10 Å of the metals indicate local differences between apo and native structures but no significant differences are seen in the other parts of the molecule. At 5 × 10?2 m-Cd two metal sites (D and D′) are found 25.5 Å from the active center, on the surface of the minor domain. They are related to each other by the molecular 2-fold axis with a D-D′ distance of 25 Å. The seventh Cd site, E, is 20 Å from the active center, on the major domain, near a crystalline contact region, and devoid of any molecular symmetry mate.The apparent dissociation constants for cadmium at the A, B and D sites (and A′, B′, D′) are 3 × 10?3 m, 1.5 × 10?1 m and 1.3 × 10?2 m, respectively. Thus in these conditions cadmium is seen to distribute between A and B sites when the combined stoichiometry is two metal ions per dimer.  相似文献

A heterogeneous particulate fraction of mouse brain homogenates binds NRDC 157 (3-phenoxybenzyl [1R,cis]-3-(2,2-dibromovinyl)-2,2- dimethylcyclopropanecarboxylate), a potent pyrethroid insecticide, stereospecifically and with high affinity. Stereospecific binding is a minor component of total binding (2.8%); the remainder of observed binding is predominantly nonspecific and unsaturable. Stereospecific binding is half-saturated at 4×10?8M and fully saturated at concentrations in excess of 1×10?7M. The stereospecific binding capacity of this preparation was 200–250 pmoles of NRDC 157 per gram equivalent of brain tissue (2.3–2.8 pmol/mg protein). This binding site may represent the neural receptor involved in the stereospecific toxic action of pyrethroids.  相似文献

Maximum levels of binding of α-bungarotoxin to foetal human brain membranes were found to remain essentially constant at 30–50 fmol/mg protein (1.1–1.5 pmol/g wet weight in whole brain) between gestational ages of 10 and 24 weeks. Equilibrium binding of α-bungarotoxin to both membranes and to detergent extracts showed saturable specific binding to a single class of sites with Kd (app) values of 3.5 × 10?9 M and 2.4 × 10?9 M respectively. Association rate constants, determined from time courses of binding of α-bungarotoxin to membranes and detergent extracts, were 2.3 × 105 M?1 sec?1 and 2.6 × 105 M?1 sec?1 respectively. Dissociation of α-bungarotoxin from both membrane and detergent extracts showed a rapid initial rate with T12 approx 15 min which, in the case of the detergent extract, was followed by a slower dissociation accounting for the remaining 20% of the bound ligand. Competition studies with a number of cholinergic ligands indicated that the α-bungarotoxin-binding sites in foetal brain display a predominantly nicotinic profile.  相似文献

It is reported that receptors for epidermal growth factor (EGF) in HeLa S3 cells exist in two forms, which differ in both affinity and capacity. Both the number of receptors and their distribution into low- and high-affinity forms are modulated by glucocorticoids. Scatchard analysis of saturation binding assays performed at 0 °C indicates that there is a low-affinity class of receptors (Kd ? 1.5 nm), which contains approximately 6 × 104 binding sites per cell, and a second, high-affinity class of receptors (Kd ? 0.16 nm) containing approximately 5 × 103 binding sites per cell. Exposure of HeLa S3 cells to 10?7m dexamethasone for 24 h increased EGF binding to whole cells by increasing the numbers of low- and high-affinity receptors by 20 and 114%, respectively. The increase in EGF binding depends upon the dose of dexamethasone, being raised from 10?11 to 10?6m. EGF binding is half-maximal near 2–4 × 10?9m, a concentration equal to the Kd of dexamethasone for the glucocorticoid receptor in these cells. The increase in EGF binding is specific for glucocorticoids, occurring when the HeLa S3 cells are exposed to 10?7m cortisol or dexamethasone for 24 h, but not when the cells are similarly treated with testosterone, 5α-dihydroxytestosterone, 17β-estradiol, or progesterone. The effect on EGF binding appears to be biphasic; the initial rapid increase occurs between 8 and 12 h, is blocked by both 10?6m cyclohexamide and 0.1 μg/ml actinomycin D, and is followed by a more gradual increase thereafter. A reexamination of the equilibrium and the kinetics of 1,25-dihydroxy vitamin D3 binding with its receptor in chick intestinal cytosol was performed because of the recent availability in our laboratory of high specific activity 1,25-dihydroxy[3H-26,27]vitamin D3 (160 Ci/mmol). Under saturating conditions at 25 °C, Scatchard analysis revealed an equilibrium dissociation constant (Kd) of 7.1 × 10?11m which is several fold lower than previously reported for this binding reaction. Furthermore, an estimate of 1.8 × 103 receptor sites per cell was obtained from the intercept of the line with the abscissa of the Scatchard plot. From a kinetic analysis of 1,25-dihydroxy vitamin D3 binding with chick intestinal cytosol, association and dissociation rate constants were determined. Values that were obtained at 25 °C for these processes were 9.5 × 108m? min? and 7.1 × 10?3 min?, respectively. Although these studies, such as for other steroid hormones, were carried out using a crude native cytosol preparation, we have been able to demonstrate unequivocally through the use of high specific activity 1,25-dihydroxy[3H-26,27] vitamin D3 a truly high affinity binding site.  相似文献

The ATPase enzyme system from two-spotted spider mites, Tetranychusurticae (Koch) was sensitive, in vitro, to four acaricides. Tricyclohexylhydroxytin (PlictranR) was an outstanding inhibitor of oligomycin-sensitive (mitochondrial) Mg2+ATPase from fish brain and spider mite homogenates. The I50 values were 6.6×10?11M and 6.2×10?10M, respectively. Less effective were chlorbenside, chlorfenethol and ovotran. Plictran at a higher concentration (2×10?7M) was also more effective on Na+-K+ATPase both in mites and fish brain homogenates as compared to chlorfenethol, chlorbenside and ovotran. Plictran inhibited oligomycin-insensitive Mg2+ATPase at concentrations of 10?8M but stimulated at high concentrations (5×10?6M and higher).  相似文献

Experiments were designed to determine whether or not various antiestrogens in direct competition with estradiol-17β (E2) would inhibit the translocation of the estrogen receptor complex from the cytoplasm to nuclei in rat uterine tissue. Incubation of the antiestrogens CI-628, cis-clomiphene, U-11,100A and MER-25 with rat uteri caused the nuclear uptake of the antiestrogen receptor complex which was greatest for most antiestrogens at concentrations of 1 × 10?6 to 1 × 10?5M. At higher concentrations of CI-628, cis-clomiphene, and U-11,100A the nuclear binding of the antiestrogen receptor complex was greatly decreased. Incubation of the antiestrogens with E2 resulted in a dramatic inhibition of the nuclear uptake of the estrogen receptor. Trans-clomiphene, a weak estrogen, did not inhibit the movement of the uterine cytoplasmic receptor into the nuclear fraction.  相似文献

Hydrocortisone (cortisol) increased the binding of thyrotropin-releasing hormone (TRH) to specific membrane receptors in 4 clonal strains of rat pituitary cells. At the highest effective cortisol concentration (3–5 × 10?6 M), the increase was observed within 6–8 hr and became maximal (140 to 160% of control binding) by 18–24 hr. Half-maximum stimulation occurred in serum-containing medium at 9 × 10?8 M cortisol, and a significant increase in TRH binding was seen at 3 × 10?8 M. Equilibrium binding studies showed that enhanced TRH binding was explained by an increase in receptor number with no change in affinity. Similar effects were seen with Dexamethasone, but no increase in TRH binding was noted when testosterone, methyltestosterone, progesterone, estradiol or the antiestrogen Lilly 88571 were added to the culture medium. Cortisol treatment did not cause the appearance of specific TRH binding sites in cell strains previously shown to lack receptors for the tripeptide (F4C1, GH12C1 and R5 cells). When added cortisol was removed from medium, receptor number decayed to control values with a T12 of about 30 hr. Previous studies have shown that TRH receptors in GH-cells can be down-modulated by TRH and thyroid hormones; the present findings demonstrate that glucocorticoid hormones can increase the number of TRH receptors in GH-cells.  相似文献

A third melanotropin coding fragment named γ-MSH was discovered by Nakanishi et al (Nature 278, 423–427 (1979)) in the cryptic region outside the portion coding for ACTH and β-LPH in the ACTH/β-LPH precursor mRNA isolated from the intermediate lobe of bovine pituitary. Four possible γ-MSH peptides derived from this coding fragment were synthesized by solid-phase methodology and their bioactivity determined in an in vitro MSH assay as well as the anterior pituitary primary culture assay. Relative to α-MSH, the melanotropic activities of Ac-γ1-MSH, γ1-MSH, γ2-MSH and γ3-MSH are 7.3 × 10?4, 3.3 × 10?5, 1.4 × 10?4 and 4.6 × 10?7 respectively. None of these γ-MSH peptides releases LH, FSH, PRL, GH and TSH in the pituitary culture medium at a dose as high as 100 ng per dish.  相似文献

Using a rapid filtration technique we find stereospecific high-affinity d-LSD binding (4 × 10?9M, half-saturation) in brain fractions from a number of subcortical as well as cortical brain regions. Among the putative neurotransmitters tested only serotonin effectively displaces d-LSD from this specific binding site. Moreover, only the serotonin-displaceable component of binding is saturable in a high-affinity range. No change is observed in specific d-LSD binding in forebrain homogenates from rats in which ascending serotonergic pathways are destroyed by lesions in the raphe nucleus. We conclude that a vast majority of the d-LSD binding sites may be associated with a postsynaptic serotonin receptor rather than a presynaptic receptor associated with serotonergic (raphe) inputs.  相似文献

Intact rat blood platelets are shown to possess benzodiazepine binding sites of the peripheral type, binding of [3H]diazepam being strongly inhibited by Ro5-4864 (Ki = 3.6 ± 0.5 nM) but only weakly inhibited by clonazepam (Ki = 35.1 ± 18.2 μM). Binding of [3H]diazepam is specific and saturable. Scatchard analysis reveals a single class of binding sites with KD = 14.7 ± 1.0 nM and Bmax = 564 ± 75 fmoles/108 platelets. The Hill coefficient is 0.94, indicating a lack of binding site heterogeneity or negative cooperativity. Binding reaches equiliibrium at 6 min, with k+1 = 2.9 × 107 M?1 min?1, and is rapidly reversible (t12 = 2.2 min with K?1 = 0.315 min?1. KD derived from the rate constants agrees with that estimated by Scatchard analysis. KD of the crude membrane fraction of platelets is also close to that of intact platelets. Binding of [3H]diazepam is linear with platelet number (between 0.25–2 × 108 platelets), is temperature sensitive with maximum binding at 0°C, and has a broad optimal pH range between pH 5–9.  相似文献

Insulin binding to human fetal plasma liver membranes was studied in preparations segregated into three pools according to length of gestation: 15–18 weeks (Pool A), 19–25 weeks (Pool B), and 26–31 weeks (Pool C). Receptor numbers, calculated by extrapolation of Scatchard plots to the X axis, increased from 25 × 1010 sites per 100 μg protein in the youngest group (Pool A) to 46 × 1010 sites per 100 μg protein in Pool B. No further increase in receptor number was seen in Pool C. The affinity constant for insulin at tracer concentrations, Ke (“empty site”), was 1.53 × 108M?1 in Pool A and was only slightly higher than Kf (“filled site”). Ke was higher in Pool B, 1.75 × 108M?1, and in Pool C reached a value of 5.63 × 108M?1. In Pool C Kf was 2.3 × 108M?1. Insulin binding of liver plasma membranes from rat fetuses aged 14, 16, 18, and 21 (term) days and adults was also studied. Maximum binding capacity tended to increase with gestational age and was 130 × 1010 sites per 100 μg protein at term, which was in excess of that found in adult rats (89–90 × 1010). In addition, Ke increased from 0.75 × 108M?1 at 14 days to 3.02 × 108M?1 at term, a value higher than that found in pregnant and nonpregnant adults. Dissociation of insulin in the presence of high concentrations of insulin was significantly enhanced in tissues from 18-day and term fetuses and adults, but not in membranes from fetal rats aged 14 and 16 days. These data appear to indicate that site-site interactions are not present in early fetal existence. These changes in insulin binding with increased length of gestation are not ascribable to changes in relative proportions of hematopoietic and parenchymal tissue. Human fetal plasma liver membranes demonstrated elevated insulin binding with increased gestational age, but comparison of fetal and adult liver could not be done. However, newborn human infants have been shown to have a higher capacity for binding insulin to circulating monocytes than adults. Also, human fetuses apparently lack the capability to diminish monocyte receptors in the presence of hyperinsulinemia. These experiments show that an increase in insulin receptor binding capacity and affinity also occurs in the liver of the rat fetus at term as compared to the adult rat. The reasons and mechanisms underlying enhanced capacity for insulin binding by fetal and newborn members of human and rodent species are not known.  相似文献

In vitro incubations of non-histone proteins from rat liver nuclei with labelled L-3, 5, 3′ triiodothyronine demonstrate the existence of high affinity, limited capacity binding sites for the hormone in this protein group; the affinity was found identical for triiodothyroacetic acid and lower for L-thyroxine. Binding ability was highly temperature dependent. At 4°C, the rate constant of association was 0.9 × 107 M?1 h?1 and the rate constant of dissociation was 0.015 h?1. The dissociation constant Kd was calculated from these data or measured by Scatchard analysis and found to be between 1.6 and 5 × 10?9 M. The maximum binding capacity was 10?13 moles of L-3, 5, 3′ triiodothyronine per 100 μg non-histone proteins or 6000 hormone molecules per nucleus. Protein binding had a half-life of 20 hours at 4°C, in the absence of hormone, but was found to be very stable in the presence of hormone.  相似文献

Anthroylcholine was utilized as an extrinsic fluorescent probe in rapid kinetic studies of calcium dissociation from calmodulin (koff = 10 S?1) and the calmodulin-troponin I complex (koff = 6 S?1). At concentrations lower than 70 μM, the mechanism of dye binding agreed with the simple kinetic scheme in which the dye binds exclusively to the respective calcium complexes of calmodulin and calmodulin-troponin I. The sensitivity of anthroylcholine also made possible the estimation of values for the association (1.0 ± 0.8) × 108M?1 S?1) and dissociation rate constants (2 ± 170 S?1) for troponin I binding to the calcium4-calmodulin complex.  相似文献

Particles from rat oviduct homogenates sedimenting between 1,000 × g for 10 min and 48,000 × g for 30 min bound [3H]oxytocin in vitro. The apparent Kd for oxytocin binding to high affinity sites in particles prepared from estrogen-treated rats was 1.8 × 10?9 M. About 215 fmoles of oxytocin were bound per mg of particulate protein. Oviducal preparations from untreated rats had about 25% the affinity for oxytocin of preparations from estrogen-treated rats. Oxytocin analogues were bound to oviducal particles in the same rank order as their uterotonic potencies: (desamino)oxytocin > (4-threonine)oxytocin > oxytocin > (8-lysine)vasopressin ? desaminotocinol. No oxytocin binding could be shown with the particulate fractions from rat ovary. The binding of oxytocin to the oviduct and uterus are similar in affinity, number of binding sites, ligand specificity, and the increase in response to estrogen treatment.  相似文献
